The oviducts of 4 cows were cannulated and oviduct fluid was collected daily from the exteriorized cannulas for a total of 5 oestrous cycles. Daily serum samples were assayed for oestradiol-17 beta and progesterone to monitor the oestrous cycle. Data for each cycle were compared for oviduct fluid collected during the non-luteal phase (serum progesterone less than or equal to 1.5 ng/ml) and the luteal phase (serum progesterone greater than 1.5 ng/ml). During the non-luteal phase oviduct fluid volume was higher and the osmolality was lower than during the luteal phase. Total protein, cholesterol and phospholipid secreted daily was greater during the non-luteal phase. Cholesterol and protein concentrations were generally lower during the non-luteal phase, but phospholipid concentrations were generally higher. About 40% of the phospholipid in oviduct fluid was phosphatidylcholine and lysophosphatidylcholine, while phosphatidylinositol and lysophosphatidylinositol accounted for 20%. The ratio of 1-acyl-phospholipid to diacylphospholipid increased during the non-luteal phase. An increased cholesterol to phospholipid ratio, and a decreased cholesterol to protein ratio in oviduct fluid also were associated with the non-luteal phase. Changes in the lipid composition of oviduct fluid during the oestrous cycle may play a role in the preparation of gametes for fertilization.  相似文献

Mammalian oviduct acts as a reservoir for spermatozoa and provides an environment in which they may compete for the opportunity to fertilize the oocyte. Whilst in the oviduct spermatozoa undergo capacitation essential for fertilization. Sperm-oviduct interaction is essential for sperm capacitation and is a tightly regulated process influenced by the local microenvironment. Previously we reported that the endocannabinoid anandamide (AEA) regulates sperm release from epithelial oviductal cells by promoting sperm capacitation. The aims of this work were to measure the AEA content and to characterize the main AEA metabolic pathway in the bovine oviduct and determine how these change through the oestrous cycle. In this study, the levels of AEA and two other N-acylethanolamines, N-oleoylethanolamine and N-palmitoylethanolamine, were measured in bovine oviduct collected during different stages of oestrous cycle by ultra high performance liquid chromatography tandem mass spectrometry. Results indicated that intracellular oviductal epithelial levels of all three N-acylethanolamines fluctuate during oestrous cycle. Anandamide from oviductal fluid also varied during oestrous cycle, with the highest values detected during the periovulatory period. Endocannabinoid levels from ipsilateral oviduct to ovulation were higher than those detected in the contralateral one, suggesting that levels of oviductal AEA may be regulated by ovarian hormones. The expression and localization of N-acylethanolamines metabolizing enzymes in bovine oviduct were also determined by RT-PCR, Western blot, and immunohistochemistry but no change was found during the oestrous cycle. Furthermore, nanomolar levels of AEA were detected in follicular fluids, suggesting that during ovulation the mature follicle may contribute to oviductal AEA levels to create an endocannabinoid gradient conducive to the regulation of sperm function for successful fertilization.  相似文献

beta-Endorphin was measured by radioimmunoassay in homogenates of ovaries from immature Sprague-Dawley rats (21-29 days of age) and found to be present at levels of about 0.6-0.7 ng/ovary. After administration of PMSG there was approximately a 4-fold increase (2-3 ng/ovary) in total ovarian immunoreactive (ir) beta-endorphin 48 h after injection. Analysis of follicular fluid from similarly treated rats indicated about the same amount of ovarian ir-beta-endorphin (2-3 ng/ovary) as in ovarian homogenates, suggesting that most of the ir-beta-endorphin is localized in follicular fluid of PMSG-primed immature rats. Immature rats were made pseudopregnant by administration of hCG 48 h after PMSG, and at 24 h after injection of hCG there was a slight, but significant and reproducible, increase in the ovarian content of ir-beta-endorphin. The serum concentration of ir-beta-endorphin was in the range of 1-3 ng/ml and was unaffected by PMSG and PMSG/hCG; likewise, the pituitary content of ir-beta-endorphin did not change following administration of gonadotrophins to immature rats. In mature cyclic animals, levels of 2-4 ng ir-beta-endorphin/ovary were found, comparable to those in the ovaries of PMSG-primed immature rats, and there were only small changes during the oestrous cycle. In addition to ir-beta-endorphin, we also obtained evidence for the presence of alpha-N-acetylated opioid peptides (endorphins or enkephalins) in the ovaries of PMSG-primed immature and mature rats. The physiological role of the opioid peptides in reproductive tissue is unknown, but they are presumably acting in an autocrine or paracrine fashion.  相似文献

Concentration of ascorbic acid was determined in different parts of buffalo ovary at four different stages of oestrous cycle viz. early luteal, mid luteal, late luteal and follicular. The stages were decided from the physical and morphological examinations of corpora lutea. The ovary was dissected in three components viz. corpus luteum, follicular fluid and ovarian stromal tissue for ascorbic acid assay. Corpus luteum showed significant change in concentration of ascorbic acid with the advancement of oestrous cycle, value being highest in late- luteal stage. Follicular fluid and ovarian stromal tissue did not show significant changes in ascorbic acid at any stage of the oestrous cycle. Small follicles, irrespective of the stage of oestrous cycle had, however, significantly higher ascorbic acid content than large follicles.  相似文献

The affinity and the density of peripheral-type benzodiazepine binding sites (PBzS) in tissues from the genital organs of female rats were studied during the oestrous cycle. When comparing PBzS density on the day of oestrus to PBzS density on the day of pro-oestrus, a significant increase was observed in the ovary (1.9-fold), oviduct (2.4-fold) and uterus (1.7-fold), but not in the kidney. Serum oestradiol also increased to a maximum on the day of pro-oestrus. The ovarian and uterine PBzS density and serum concentrations of oestradiol and progesterone were measured every 8 h between the days of dioestrus and pro-oestrus. Ovarian and uterine PBzS density increased to a maximal value at 01:00 and 09:00 h, respectively, on the day of pro-oestrus. However, a significant increase in PBzS density occurred in the ovary (P less than 0.02) and uterus (P less than 0.001) at 09:00 h on the day of pro-oestrus as compared to 09:00 h on the day of dioestrus. These changes were associated with an increase in serum oestradiol and progesterone concentrations. The affinity of PBzS in all tissues examined remained unaltered during the oestrous cycle. This study demonstrates that changes associated with the oestrous cycle occur in the density of PBzS in various genital organs.  相似文献

Oviducal secretions were obtained from conscious unrestrained ewes throughout the oestrous cycle via indwelling cannulae and the content of prostaglandin F (PGF) was determined by radioimmunoassay. Levels of PGF of up to 230 ng/ml were found in oviducal fluids obtained from ewes showing regular patterns of secretion and normal cyclical ovarian function as indicated by plasma progesterone measurement. Relatively large day to day fluctuations in content were evident, but there was no consistent relationship between concentration and stage of the oestrous cycle. Concentrations of PGF in excess of 100 ng/ml were common in preparations where autopsy later revealed infection or tissue irritation, and the concentration of PGF invariably exceeded 75 ng/ml when the concentration of protein in the oviducal fluid was abnormally high.  相似文献

Harderian glands of Syrian hamsters contained measurable levels of immunoreactive somatostatin. After an extraction procedure, serial dilutions of tissue were assayed and showed parallelism in the displacement curve with dilutions of purified somatostatin standard in the radioimmunoassay. Somatostatin concentrations were higher in female hamsters (10.0 +/- 2.1 ng/mg protein) than in males (2.6 +/- 0.4 ng/mg protein). Castrated males had somatostatin values in the range of females (12.4 +/- 2.3 ng/mg protein) at 1 month after gonadectomy. Testosterone implants prevented the rise of Harderian gland somatostatin in castrated males. Gonadectomized males had lower somatostatin content in the gland than did control males (1.0 +/- 0.2 ng/mg protein) at 2 months after castration. Somatostatin values in females were unaffected by gonadectomy, but there were variations during the oestrous cycle, with a nadir detected at dioestrus-1 and maximal values coincident with the day of the ovulation.  相似文献

Thioredoxin expression within the reproductive tissues of the female mouse was analysed during the oestrous cycle stages of dioestrus, oestrus and metoestrus by Western blot analyses and immunocytochemistry. From Western blot analyses the expression of thioredoxin was found to be increased in oestrus compared to dioestrus and metoestrus. Localisation of thioredoxin within the reproductive organs of the mouse during the oestrous cycle has shown that the expression of thioredoxin is specific for distinct areas within the reproductive organs. These areas are the stratified squamous epithelium of the vagina, the simple columnar epithelium and the uterine glands of the uterus, the ciliated columnar epithelium of the oviduct, the corpus lutea, the interstitial cells and the secondary follicles of the ovary. The discrete cellular localisation and oestrous dependence of thioredoxin expression are suggestive of specific roles in various reproductive processes.  相似文献

The ovarian bursa was peeled from around one ovary of each rat and the rats were killed 1, 2, 3, 4 and 5 weeks later. The proportion of rats that maintained a bursa-free ovary did not change over the 5-week period (80-89%). Ovulation from the peeled ovary occurred in all rats but oocytes (1-4) were found in the ipsilateral oviduct in only 18% of the rats. The presence of oocytes in the oviduct was normally associated with some degree of re-encapsulation of the ovary. In another experiment rats were mated within 1 week of removal of the bursa from around the ovary. Unilateral pregnancy resulted in 92% of the rats. In a third experiment fertilized oocytes from mated donor rats were transferred into the oviduct next to the peeled ovary in 15 mated recipients. Of 85 zygotes transferred, 51 survived to be viable fetuses on Day 20. A single fetus developing from an endogenous oocyte was found in the transfer uterine horn in only one rat. This preparation may be useful in studies which attempt to determine the viability of oocytes that have undergone various manipulations in vivo or in vitro.  相似文献

Basal adenylate cyclase values for corpora lutea (CL) removed from cyclic gilts on Days 3, 8, 13 and 18 were 178 +/- 61, 450 +/- 46, 220 +/- 25 and 208 +/- 18 pmol cAMP formed/min/mg protein, respectively. Basal activity was significantly elevated on Day 8 (P less than 0.001). LH-stimulatable adenylate cyclase values for CL from Days 3, 8, 13 and 18 were 242 +/- 83, 598 +/- 84, 261 +/- 27 and 205 +/- 17 pmol cAMP formed/min/mg protein respectively. Serum progesterone concentrations of 12 gilts bled every 2 days through one complete oestrous cycle ranged from 1.1 to 26.9 ng/ml with highest values between Days 8 and 12. The decline in serum progesterone concentrations was coincident with the decrease in basal adenylate cyclase activity. There was no LH-stimulatable adenylate cyclase activity present in the CL at the specific times of the oestrous cycle examined. We conclude that progesterone secretion by the pig CL is apparently dependent on basal activity of adenylate cyclase.  相似文献

In vivo, bull sperm capacitation seems to occur mainly in the oviduct. Capacitation of bull spermatozoa can be triggered in vitro by exposure to heparin, a heavily sulphated glycosaminoglycan (S-GAG). We determined the concentration of S-GAGs in oviductal fluid from dairy heifers, collected over the course of several oestrous cycles via surgically implanted intraluminal catheters. We also investigated the presence of syndecans, i.e. heparan sulphate proteoglycans, in the bovine oviductal epithelium of Swedish dairy cattle during standing oestrus and the luteal phase of the oestrous cycle, using immunohistochemistry for three different polyclonal antibodies raised against human syndecan-2 and rat syndecan-1 and syndecan-2, respectively. The concentration of S-GAGs in oviductal fluid obtained from the ampullar segment of the oviduct was significantly higher (P=0.0026) than it was in fluid from the isthmic segment during the functional period, i.e. from prooestrus to metaoestrus (73.5+/-10.49 mg/L in ampullar ODF, compared to 43.2+/-10.74 mg/L in isthmic ODF); least square mean (L.S.M.)+/-standard error of the mean (S.E.M.). There was also a significantly higher concentration of S-GAGs in the fluid from the oviduct ipsilateral to the ovulation side 73.5+/-10.54 mg/L on the ovulation side, compared to 43.1+/-10.71 mg/L in the oviduct on the contralateral side (L.S.M.+/-S.E.M., P=0.0026) during this period. Both syndecan-1 and syndecan-2 were present in the epithelial cells lining all studied segments of the bovine oviduct, i.e. the UTJ, isthmus and ampulla, during both standing oestrus and dioestrus. The syndecans and S-GAGs found may influence the gametes, while they reside in the oviduct; the amounts of S-GAGs found in the bovine oviduct seem sufficient to act as capacitating factors in vivo.  相似文献

Oestrus and anoestrous mongrel dogs were anaesthetized with chloralose-urethane. In one group, the ovaries were isolated in situ and the effects of a 15-min infusion of PGF-2alpha or PGE-2 on perfusion pressure were measured. In the other group, heated thermocouples were introduced into the stroma of each ovary to measure the changes of local blood flow in response to PGF-2 alpha and PGE-2 infused into the ovarian bursa for 15 min. Intra-arterial infusion of 25.50, 100 or 200 ng PGF-2alpha/kg/min did not affect perfusion pressure; PGE-2 doses of 3.1, 6.2, 12.5 or 25 ng/kg/min caused reductions in proportion to the dose. All doses of PGE-2alpha (50.0, 100 or 200 ng/kg/min) or PGE-2 (25, 50 or 100 ng/kg/min) increased blood flow in the ovarian stroma in proportion to the dose when administered by infusion into the ovarian bursa. There were no differences in the results from oestrous and anoestrous dogs. It is concluded that PGF-2alpha changes intraovarian blood distribution without interfering with the total blood flow while PGE-2 increases both the total and local ovarian blood flow.  相似文献

An enzymatic microassay method for glutamate decarboxylase (GAD) and gamma-aminobutyric acid (GABA) was improved to yield a high sensitivity and a low blank. The 20-microns thick freeze-dried sections (0.2-1.5 micrograms dry weight) were prepared from the oviduct and ovary of rat. The analysis of these microsamples by the improved method showed that, contrary to the previous observations, the rat ovary is devoid of GAD activity and contains a trace amount of GABA. Both are present abundantly in the oviduct. In the oviduct mucosa, significant GAD activity was found in the estrous phase, whereas the activity was nearly null during other phases of the estrous cycle. GABA concentration in the oviduct mucosa was 10-fold higher than in the cerebral cortex; its variation during the estrous cycle was not remarkable. In the muscle layer of oviduct, GAD activity had a low peak in the estrous phase and GABA concentration was almost constant during the estrous cycle. The denervation experiment showed that GAD is present in the nerve terminals innervating the oviduct.  相似文献

【目的】明确斑衣蜡蝉Lycorma delicatula雌成虫生殖系统整体形态及超微结构特征,为蜡蝉总科昆虫分类及系统发育探讨提供更多形态学证据。【方法】采用光学显微镜与透射电子显微镜,观察斑衣蜡蝉雌成虫生殖系统整体形态和各主要器官的超微结构。【结果】斑衣蜡蝉雌成虫生殖系统主要包括1对卵巢、1个中输卵管、1个交配囊、1个交配囊管、1个前阴道、1个后阴道、1个受精囊、1个受精管和2根受精囊附腺。卵巢为端滋式,由14根卵巢小管组成,卵室由固有膜、滤泡细胞和卵细胞组成,卵巢小管中的滋养细胞清晰可见;中输卵管位于前阴道基部,由中输卵管腔、上皮细胞、肌肉鞘和基膜组成;交配囊膨大呈圆球状,囊壁由上皮细胞、肌肉层和基膜组成;交配囊管呈圆柱状,连接交配囊和后阴道,由肌肉鞘、上皮细胞层和管腔组成;前、后阴道超微结构相似,主要由肌肉鞘、基膜、上皮细胞和管腔组成,但后阴道上皮细胞细胞核周围存在分泌颗粒,且管腔内有大量微绒毛,而前阴道壁内包含有大量囊泡结构;受精管从中输卵管末端延伸至受精囊,由基膜、厚层肌肉鞘和管腔组成;受精囊为受精管近末端略膨大的囊状结构,由肌肉鞘、基膜、上皮细胞和囊腔构成;雌性受精囊附腺着生于受精囊末端,为均匀的螺旋管状,主要由肌肉层、上皮细胞层和附腺中心管腔组成。【结论】斑衣蜡蝉雌性生殖系统与已报道的蜡蝉总科其他类群的雌性生殖系统结构相似,但卵巢小管数目有差异;蝉亚目中不同总科雌成虫雌性附腺与受精囊附腺的形态特征存在明显区别;斑衣蜡蝉雌性生殖系统超微结构与叶蝉总科和沫蝉总科昆虫也存在部分差异。这些差异是否可以作为头喙亚目高级阶元的划分依据仍有待于进一步研究。  相似文献

The origin of gamma-aminobutyric acid (GABA) in the rat oviduct was investigated by measuring GABA levels in the oviduct after selective ablation of the extrinsic oviductal innervation. Rats killed 20 days after ablation of nerves connected with the left oviduct showed no differences in GABA levels in the left vs the right oviduct. Rats killed 50 days after ablation of the left ovarian vascular nerve bundle showed a decreased GABA content in the left vs the right (intact) oviduct which was more pronounced in rats killed 90 days after ablation. In contrast, GABA levels were unchanged 50 days after ligation of either the suspensory ligament of the ovary or the uterine artery. Our results indicate the involvement of a GABAergic component in the extrinsic innervation of the rat oviduct.  相似文献

Blood flow in the ovary and oviduct of rats after sympathetic denervation   总被引:1,自引:0,他引:1  
The effect of sympathetic denervation on blood flow in the ovary and oviduct was studied in rats undergoing oestrous cycles or at Day 14 of pregnancy. The ovary and oviduct on one side were denervated by briefly freezing the ovarian vascular pedicle and the ovarian suspensory ligament. Blood flow was measured using 15 microns 57Co-labelled microspheres while the rats were under barbiturate anaesthesia. In cyclic rats denervation raised blood flow to the oviduct by 90% the next day (P less than 0.01) and 39% at 4-10 days (0.05 less than P less than 0.1). Blood flow to the ovary was not affected. Denervation on Day 13 of pregnancy raised blood flow in the oviduct 5-fold at Day 14 (P less than 0.01) and denervation on Day 7 raised blood flow 3-fold on Day 14 (P less than 0.05). Blood flow to the luteal and non-luteal components of the ovary was not affected. Sham-operation did not affect blood flow in the oviduct or ovary. It is concluded that sympathetic nerves exert tonic vasoconstrictor control on the vasculature of the oviduct but not on that of the ovary, and that these nerves do not regulate the major changes in blood flow that occur in ovaries in various physiological states.  相似文献

Weight, RNA, DNA and protein content of the oviduct, vaginal cul-de-sac, lateral vagina and urogenital sinus and oestradiol and progesterone cytosol receptor concentrations in vaginal cul-de-sac, lateral vagina and urogenital sinus were examined after administration of oestradiol to ovariectomized animals and on days 0, 5, 9 and 13 of the non-pregnant cycle and on day 13 of the pregnant cycle. In ovariectomized animals, oestradiol induced an increase in weight, RNA:DNA and protein:DNA ratios and a decrease in DNA:tissue weight ratio for each organ and in addition an increase in total DNA in vaginal cul-de-sac and urogenital sinus. There was no effect of oestradiol on oestradiol cytosol receptor concentration but there was a significant increase in progesterone cytosol receptor concentration in all organs that were examined. During the oestrous cycle, changes in the wet weight of each organ showed a common pattern with maximum weight at day 0 followed by variable rates of decline until day 13. In oviduct and vaginal cul-de-sac, the decrease in weight was paralleled by a decrease in RNA:DNA and protein:DNA ratios whereas the DNA: tissue weight ratio showed the opposite pattern and total DNA remained unchanged. The changes in the lateral vagina and urogenital sinus were similar except that a significant decline in total DNA was also seen after day 0 and the DNA:tissue weight and protein:DNA ratios in the urogenital sinus and the lateral vagina respectively showed no significant changes. Progesterone cytosol receptor concentration in the lateral vagina and urogenital sinus were high on day 0 and then declined until day 13. In contrast, there were no consistent effects on oestradiol receptor concentration.  相似文献

Two experiments were carried out on ewes with ovarian autotransplants to estimate the ovarian uptake of glucose and production of lactate. The first was carried out in the luteal phase of the oestrous cycle. Samples of carotid arterial, ovarian venous and jugular venous blood were collected simultaneously for glucose analysis. The arterial concentration of glucose (58.0 ± 5.0 mg/dL; Mean ± SEM) was significantly higher than the ovarian venous concentration (42.3 ± 2.4 mg/dL; P < 0.001). Next, a second more complete experiment was carried out in the luteal and follicular phases of the oestrous cycle. The oestrous cycle was synchronised and samples of carotid arterial, ovarian venous and jugular venous blood were collected simultaneously for glucose and lactate analysis. There were significant positive arterio-venous differences in the concentration of glucose in the luteal (5.6 ± 1.2 mg/dL, mean ± SEM; P = 0.001), early (3.1 ± 0.82 mg/d; P = 0.003) and late follicular (6.4 ± 1.3 mg/dL; P = 0.001) phases of the oestrous cycle. There was a significant negative arterio-ovarian venous difference in the concentration of lactate in only the luteal phase (-2.2 ± 0.96 mg/dL; P = 0.043).The results show significant removal of glucose from the arterial circulation during its passage through the ovary in the luteal, early follicular and late follicular phases of the oestrous cycle. Furthermore, there was lactate production in the luteal phase but not in the follicular phase suggesting that in the luteal phase of the oestrous cycle, ovarian metabolism can be anaerobic.  相似文献

Summary The distribution of L-glutamate decarboxylase (GAD), a major biosynthetic enzyme for gamma-aminobutyric acid (GABA), was examined in the oviduct and ovary of the rat by means of an immunohistochemical technique. The polyclonal antiserum raised against brain GAD showed specific immunoreaction in some non-neuronal elements of the sex organs. In the oviduct, the inner layer of the mucosa was predominantly labelled. The selective distribution of GAD immunoreactivity in epithelial cells of the oviduct is consistent with former findings for GABA-like immunoreactivity in the same organ, indicating that the GAD-catalyzed reaction may be a major biosynthetic pathway for GABA even in these cells. In the ovary, vacuole-like formations within the follicular fluid and oocytes showed intense, specific staining. The occurrence of GAD immunoreactivity inside developing ovarian follicles including the oocyte may suggest a role for GABA related to follicular development and certain functions concerning the ovum.  相似文献
