Immunologically potent, RNA protein fraction could be isolated from axenic Entamoeba histolytica (NIH: 200) using a phenol extraction method. Immunization of animals with amoebal RNA protein (Eh RNA) fraction evoked an intense cellular sensitization. In immunized animals the percent leucocyte migration inhibition was found to be 73.7 +/- 6.9. In control groups where the animals were treated with Freund's complete adjuvant, yeast RNA and amoebal RNA treated with RNase, the percent leucocyte migration inhibition was 15.8 +/- 2.9, 16.8 +/- 6.2 and 14.0 +/- 4.0 respectively. Anti Eh RNA antibodies (haemagglutinins and precipitins) could not be demonstrated in immunized animals at any stage of observations. The cellular immunity could be maintained at its high level by giving three immunization doses of Eh RNA and also a high degree (95.45%) of protection against experimental challenge with virulent strain of E. histolytica could be obtained in immunized animals. The immunogenic and protective capabilities of Eh RNA were found to be specific since they were abolished by the RNase treatment. Further, the yeast RNA failed to elicit immunity.  相似文献

Single ip injections of 600 mg/kg 5-(3,3-dimethyl-1-triazeno)-imidazole-4-carboxamide (DIC) and 900 mg/kg 5-[3,3-bis(2-chlorethyl)-1-triazeno]-imidazole-4-carboxamide (BIC) were given to pregnant Wistar rats at day 12 and the animals were killed 4 h after injection and at days 13-17 of gestation. Fetal tissues were used to determine total DNA, RNA, and protein and the data used to derive cell number and cell weight, RNA, and protein/cell. Both compounds reduced total fetal body weight, DNA, RNA, and protein but reduction of RNA by BIC was not statistically significant. These effects were observed 4 h after injection, increased with age (days 13-17), and were 3-4 times greater for DIC than BIC. By using the value of 6.2 mumug DNA/cell, cell number and per-cell values for weight, RNA, and protein, and weight: DNA, RNA:DNA, and protein:DNA ratios were computed. The per-cell values and ratios in the DIC-exposed animals were 8-44% greater and in BIC-treated animals 0-11% greater than control animals of the same gestational age. Percentage of body water was the same in the experimental and control animals. The differences in DNA, RNA, and protein are believed to be related to drug-induced growth retardation incident to total fetal DNA reduction resulting in diminished cell number.  相似文献

The labeling of RNA in young and adult rat brain has been studied by measuring in vitro (tissue slices incubation) the incorporation of labeled uridine into RNA of total tissue and of the various subcellular fractions purified from cerebral hemispheres of 1- and 10-month-old rats. Gel electrophoretic analysis of the newly synthesized nuclear and microsomal RNA was also accomplished. An active metabolism of RNA in adult animals was found; moreover, distinct differences in ribosomal RNA processing in cerebral hemispheres of 1- and 10-month-old rats, with a more rapid processing in the brain of adult animals, were obtained.  相似文献

The effects of aging on in vivo DNA and RNA labeling and on RNA content in various brain regions of 4-, 12-, and 24-month-old rats were investigated. No difference in [methyl-14C]thymidine incorporation into DNA of cerebral cortex and cerebelllum during aging was observed.The ratio of RNA/DNA content significantly decreased from 4 to 24 months of age in cerebral cortex, cerebellum and striatum. RNA labeling decreased by 15% in cerebral cortex of 24-month-old animals while in the other brain areas examined (cerebellum, hippocampus, hypothalamus, brainstem, striatum) did not change during aging.In the cerebral cortex, the ratio of the specific radioactivity of microsomal RNA to that of nuclear RNA, determined by in vivo experiments, was not affected by the aging process. A significant decrease of total, poly(A)+ RNA and poly(A)- RNA content was observed in the same brain area of 24-month-old rats compared to 4-month-old ones. Moreover, densitometric and radioactivity patterns obtained by gel electrophoresis of labeled RNA after in vitro experiments (tissue slices of cerebral cortex) showed a different ribosomal RNA processing during aging. In vivo chronic treatment with CDP-choline was able to increase RNA labeling in corpus striatum of 24-month-old animals.  相似文献

1. The Widnell & Tata (1966) assay method for Mg(2+)-activated DNA-dependent RNA polymerase was used for initial-velocity determinations of rat liver nuclear RNA polymerase. One unit (U) of RNA polymerase was defined as that amount of enzyme required for 1 mmol of [(3)H]GMP incorporation/min at 37 degrees C. 2. Colony fed rats were found to have a mean RNA polymerase activity of 65.9muU/mg of DNA and 18h-starved rats had a mean activity of 53.2muU/mg of DNA. Longer periods of starvation did not significantly decrease RNA polymerase activity further. 3. Rats that had been starved for 18h were used for all feeding experiments. Complete and tryptophan-deficient amino acid mixtures were given by stomach tube and the animals were killed 15-120min later. The response of RNA polymerase to the feeding with the complete amino acid mixture was rapid and almost linear over the first hour of feeding, resulting in a doubling of activity. The activity was still elevated above the starvation value at 120min after feeding. The tryptophan-deficient amino acid mixture produced a much less vigorous response about 45min after the feeding, and the activity had returned to the starvation value by 120min after the feeding. 4. The response of RNA polymerase to the feeding with the complete amino acid mixture was shown to occur within a period of less than 5min to about 10min after the feeding. 5. Pretreatment of the animals with puromycin or cycloheximide was found to abolish the 15min RNA polymerase response to the feeding with the complete amino acid mixture, but the activity of the controls was unaffected. 6. The characteristics of the RNA polymerase from 18h-starved animals and animals fed with the complete or incomplete amino acid mixtures for 1h were examined. The effects of Mg(2+) ions, pH, actinomycin D and nucleoside triphosphate omissions were determined. The [Mg(2+)]- and pH-activity profiles of the RNA polymerase from the animal fed with the complete mixture appeared to differ from those of the enzyme from the other groups, but this difference is probably not significant. 7. [5-(3)H]Orotic acid incorporation by rat liver nuclei in vivo was shown to be affected by the amino acid mixtures in a similar manner to the RNA polymerase. 8. The tryptophan concentrations of plasma and liver were determined up to 120 min after feeding with the amino acid mixtures. Feeding with the complete mixture produced a rapid increase in free tryptophan concentrations in both plasma and liver, but feeding with the incomplete mixture did not alter the plasma concentration. The liver tryptophan concentration increased at about 45min after feeding with the tryptophan-deficient diet. 9. There was a good correlation between the liver tryptophan concentration and RNA polymerase activity in all groups of animals. 10. It was concluded that the rat liver nucleus responded to an increase in amino acid supply by increased synthesis of RNA as a result of synthesis of RNA polymerase de novo. The correlation of tryptophan concentration and RNA polymerase activity appears to reflect the general amino acid concentration required to support hepatic protein synthesis and to produce new RNA polymerase. This new polymerase appears to differ from the basal RNA polymerase by its rapid synthesis and destruction, which may be a means of regulating RNA synthesis by the amino acid concentration in the liver.  相似文献

Cerebellar RNA accumulation, synthesis, and functional capacity was studied in 14-day-old F1 hybrid rats subjected to neonatally induced graft versus host disease (GVHD). There was a decrease in RNA synthetic rate as measured by the uptake of labeled precursors into RNA. The decrease in total cerebellar RNA synthesis was reflected both in a reduced amount of Nissl substance, visible in cresyl violet-stained 10-micron-thick sections of cerebella, and in the total amount of cytoplasmic RNA isolated from individual cerebella from diseased animals compared with control littermates. Analysis of the RNA translational capacity in wheat germ protein synthesizing systems showed that RNA from experimental animals was also biologically less active. Qualitative differences between protein populations in control and diseased animals were analyzed by two-dimensional gel electrophoresis. There were few alterations in the steady state levels of cerebellar protein. However, two-dimensional gel electrophoresis of the peptides synthesized in vitro by RNA from control and diseased animals showed that there were several changes in the relative abundance of some mRNAs between the two RNA populations. These data show that the cerebellar RNA from rats with GVHD differs both qualitatively and quantitatively from that of controls.  相似文献

An enhanced RNA-transport activity was observed in vitro from nuclei obtained from animals with cardiac hypertrophy as compared with that of sham-operated controls. The 100 000 g supernatants obtained from hypertrophic hearts stimulated the RNA transport from nuclei of sham-operated controls, and this stimulation was maximum with 40% supernatant. Ca2+- and nucleic acid-dependent ATPase and alkaline phosphatase activities, which may be involved in an energy-dependent transport, were high in nuclei from hypertrophic hearts, and the nuclei of sham-operated animals showed higher activities of these enzymes after incubation with supernatant from hypertrophic hearts, which stimulates the RNA transport in vitro from nuclei of sham-operated animals.  相似文献

We have previously shown that the content of glial fibrillary acidic protein (GFAP) gradually increases in the spinal cord of Lewis rats with acute experimental autoimmune encephalomyelitis (EAE), reaching a level 1.5-2 times greater than that in controls by 35 days postimmunization (dpi). We report here that the increase in GFAP mRNA level followed a completely different time course and reached higher levels relative to controls than did that of the protein. Total RNA was isolated using a modified version of current methods using phenol/chloroform extractions to ensure optimal recovery from spinal cord. Control animals yielded 323 +/- 35 micrograms (mean +/- SD; n = 34) of total RNA/spinal cord throughout the experimental period. EAE animals contained up to three times as much total RNA during 11-14 dpi, a finding largely reflecting the infiltration of inflammatory cells. By 65 dpi, total RNA levels closely approached control values. As early as 10 dpi, increased amounts of GFAP mRNA were detected in EAE animals relative to controls. During 11-14 dpi, GFAP mRNA levels reached six- to eightfold greater than values in controls and then slowly declined throughout the remainder of the time course, with a fourfold increase still detected at 65 dpi. However, coinciding with the height of inflammation and clinical signs at 12 dpi, the GFAP mRNA content dropped to approximately 50% of the level at 11 dpi but rose again at 13 dpi. This dip was mirrored by a similar decrease in neurofilament mRNA content, but otherwise the level of this message remained relatively constant and equal to that in controls.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Agarose-gel electrophoresis of polyadenylated RNA from livers of oestrogen-treated male rainbow trout revealed a major high-Mr species (7200 nucleotides), which is absent from the polyadenylated RNA isolated from hormonally unstimulated male trout liver. Translation in vitro of the RNA from oestrogen-treated males in a mRNA-dependent rabbit reticulocyte lysate produced a protein (Mr 200 000) that could be immunoprecipitated with antibodies against trout serum vitellogenin, but no immunoprecipitable protein was synthesized with RNA from control animals. DNA complementary to the RNA from oestrogen-stimulated and control male trout liver was synthesized and back-hybridized, with R0t1/2 of 3.8 X 10(-2) and 1 X 10(-1) mol X litre-1 X s for RNA from hormone-treated and control animals respectively. The 9% increase in the abundant mRNA after oestrogen stimulation is due to the induction of vitellogenin mRNA.  相似文献

The effect of putrescine on oxyntic gland and colonic mucosal growth was studied by measuring the rate of [3H]-thymidine incorporation into mucosal DNA in vitro (DNA synthesis) and DNA, RNA and protein content of the mucosa following intramuscular injections of the compound (50 mumoles/100g). Saline injected animals served as controls. Multiple injections of putrescine during a 2-day fast produced a significant enhancement of mucosal DNA synthesis in oxyntic gland and colonic mucosa, with no apparent change in DNA, RNA or protein content in either of the tissues, compared to the corresponding saline-controls, when measurements were made 12-24 h after the last injection. However, when the animals were killed after 4 days, DNA, RNA and protein content of oxyntic gland mucosa, and DNA and protein content of colonic mucosa were found to be significantly higher than in the respective saline-controls. We conclude that putrescine, taken up from the blood, can stimulate growth of gastrointestinal mucosa.  相似文献

The effect of whole-body irradiation on RNA synthesis in regenerating and non-growing livers was studied in intact and adrenalectomized rats. The animals were divided into four sub-groups: (1) control; (2) irradiation only; (3) partially-hepatectomized; and (4) irradiated partially-hepatectomized. Newly-synthesized RNA was determined by 30 or 40 min uptake of (6-14C) orotic acid. Both nuclear and polyribosomal RNA synthesis in regenerating livers of adrenalectomized rats were depressed below their control levels, regardless of whether irradiation was 2 hours before or 2 hours after partial hepatectomy. Specific radioactivity values of regenerating livers of adrenal intact and adrenalectomized rats were elevated above those of the non-growing livers from irradiated and unirradiated rats.  相似文献

Between postnatal days 10 and 16 the cerebral cortex of rats displayed a rise in protein content and a tendency towards RNA increase, whereas changes in DNA content (cell density) were absent. Young rats whose eyelids had been sutured and covered with black collodion at the age of 7 or 8 days exhibited some developmental retardation already by day 16. At an age of 5 weeks the brains of control, eyelid-sutured and dark-reared rats did not differ in their weight. However, dark-reared rats had a highly significantly increased RNA concentration in the occipital cortex (not differing significantly from controls) than eyelid-sutured animals (p less than 0.005). In the remainder of cortex, eyelid-sutured animals displayed an increase (p less than 0.10) of RNA concentration as compared with controls. The most pronounced intergroup differences were found on comparing RNA, DNA and protein concentrations between occipital and residual cortices within the individual animal groups. The difference was practically nil in control animals; all components were highly significantly raised (RNA: p less than 0.001; DNA: p less than 0.01; protein: p less than 0.001) in the residual cortex in eyelid-sutured animals; dark-reared animals displayed a similar tendency as eyelid-sutured animals that, nevertheless, did not reach statistical significance.  相似文献

The Authors demonstrate that the in vitro stimulation of mitochondrial RNA synthesis produced by thyroid hormones takes place also at physiological levels, equal to those held in the liver cells of experimental animals. Two groups of male rats have been used: normal control animals (N) and animals surgically thyroidectomized on the 25th day of life (T). The animals were fed and kept in standard conditions and killed on the 85th day of life. The purification of mitochondrial samples and the determination of the mitochondrial RNA synthesis were carried out as previously described. The results suggest that the in vitro stimulation of mitocondrial RNA synthesis is already significant at the concentration of lnM. The trends are qualitatively comparable for either N or T animals. The structural analogues TRIAC (3,5,3'-triiodothyroacetic acid) and TRIPROP (3,5,3'-triiodothyropropionica acid) exhibit a clearly stimulatory effect on samples of N animals, while on samples of T animals is significant only for the first analogue. 1. The influences of age and weaning on muscle protein synthesis were studied in vivo, by injecting a large dose of [3H]valine into 1-, 5- and 8-week-old suckling or 8-week-old weaned lambs. 2. The fractional rates of protein synthesis, in red- and white-fibre-type skeletal muscles or striated and smooth visceral muscles, were in 8-week-old suckling animals 24-37% of their values at 1 week of age. This developmental decline was related to decreased capacities for protein synthesis, i.e. RNA/protein ratios. 3. At 8 weeks of age, suckling and weaned lambs had similar fractional synthesis rates, capacities for protein synthesis and efficiencies of protein synthesis (i.e. rates of protein synthesis relative to RNA) in skeletal muscles. 4. In contrast, visceral-muscle fractional synthesis rates were lower in 8-week-old suckling lambs than in weaned animals, owing to decreased efficiencies of protein synthesis. It was concluded that developmental factors and the change to a solid diet, or weaning in itself, or both, affect differently skeletal and visceral muscle protein synthesis in the immature lamb.  相似文献

Experiments were conducted on male rats, 250-300 g in weight. Adaptation to high altitude hypoxia was created by placing the animals daily for 5 hours, into an altitude chamber, at an "altitude" of 6000 m. The degree of hypertrophy of the right ventricle and its RNA content was studied after 20 days of adaptation, as well as 2, 10, 20 and 40 days after cessation of hypoxia. Twenty days after the beginning of adaptation the muscle mass of the right ventricle the RNA concentration and amount in it was found to increase considerably. After cessation of hypoxia half of the acquired increase in the ventricle muscle mass was lost in 10 days, and half of the acquired increase in the RNA--as soon as in 2 days. Forty days after cessation of hypoxia the right ventricle mass and its RNA content in the adapted animals did not differ from the same indices in control rats.  相似文献
