Some novel cephalexin-derived furanyl, thiophenyl, pyrrolyl, salicylyl and pyridyl Schiff's bases and their cobalt (II), copper (II), nickel (II) and zinc (II) complexes have been synthesized and studied for their antifungal properties against Trichophyton longifusus, Candida albicans, Aspergillus flavus, Microsporum canis, Fusarium solani and Candida glaberata. The presence of metal ions in the investigated Schiff's base complexes reported here lead to significant antifungal activity, whereas the parent ligands were generally less active. 相似文献

Kefzol (kzl), a β-lactam antibiotic, possesses various donor sites for interaction with transition metal(II) ions [Co(II), Cu(II), Ni(II) and Zn(II)] to form complexes of the type [M(kzl)2]Cl2 and [M(kzl)Cl], with molar ratio of metal: ligand (M:L) of 1:2 and 1:1 respectively. These complexes were prepared and characterized by physicochemical and spectroscopic methods. Their IR and NMR spectra suggest that kefzol potentially acts as a bidentate, tridentate as well as monoanionic tetradentate ligand. The complexes have been screened for antibacterial activity and results were compared with the activity of the uncomplexed antibiotic against Staphylococcus aureus, Klebsiella pneumoniae, Pseudomonas aeruginosa, Escherichia coli and Proteus mirabilis. The metal complexes were found to be more potent against one or more bacterial species than the uncomplexed kefzol. 相似文献

A series of isatins incorporating thiazole, thiadiazole, benzothiazole and p-toluene sulfonyl hydrazide moieties, along with their cobalt(II), copper(II), nickel(II) and zinc(II) metal complexes have been synthesized and characterized by elemental analyses, molar conductances, magnetic moments, IR, NMR and electronic spectral data. These compounds have been screened for antibacterial activity against Escherichia coli, Bacillus subtillis, Shigella flexneri, Staphylococcus aureus, Pseudomonas aeruginosa and Salmonella typhi, and for antifungal activity against Trichophyton longifusus, Candida albicans, Aspergillus flavus, Microsporum canis, Fusarium solani and Candida glaberata using the agar-well diffusion method. All the synthesized compounds have shown good affinity as antibacterial and/or antifungal agents which increased in most of the cases on complexation with the metal ions. 相似文献

Nickel/cobalt transporters (NiCoTs), a family of secondary metal transporters in prokaryotes and fungi, are characterized by an eight-transmembrane-domain (TMD) architecture and mediate high-affinity uptake of cobalt and/or nickel ions into the cells. One of the strongly conserved regions within the NiCoTs is the signature sequence RHA(V/F)DADHI within TMD II. This stretch of amino acid residues plays an important role in the affinity, velocity and specificity of metal transport. Some relatives of the NiCoTs, named HupE, UreJ and UreH, contain a similar signature sequence and are encoded within or adjacent to [NiFe] hydrogenase or urease operons, or elsewhere in the genome of many prokaryotes. HupE and UreH from Rhodopseudomonas palustris CGA009 and UreJ from Cupriavidus necator H16 were shown to mediate Ni2+ transport upon heterologous production in E. coli. Other variants of NiCoTs are found in many marine cyanobacteria and in plants. The cyanobacterial proteins are encoded by a segment adjacent to the genes for [Ni] superoxide dismutase and a corresponding putative maturation peptidase. The plant proteins contain N-terminal sequences resembling bipartite transit peptides of thylakoid lumenal and thylakoid integral membrane precursor proteins; expression of a YFP-fusion protein in transfected leaf cells is consistent with targeting of this protein to the plastid, but the function of the plant gene product has yet to be demonstrated. 相似文献

A new series of antibacterial and antifungal triazine-derived mono- and di-substituted (symmetrical and unsymmetrical) Schiff-bases and their cobalt(II), copper(II), nickel(II) and zinc(II) metal complexes have been synthesized and characterized by their elemental analyses, molar conductances, magnetic moments and IR and electronic spectral measurements. IR spectra indicated the ligands to act as tridentate towards divalent metal ions via a trazine-N, the azomethine-N and, indole-NH and deprotonated-O of salicylaldehyde. The magnetic moments and electronic spectral data suggest octahedral geometry for the Co(II), Ni(II) and Zn(II)complexes and square-pyramid for Cu(II) complexes. NMR spectral data of the ligands and their diamagnetic zinc(II) complexes well-define their proposed structures/geometries. Elemental analyses data of the ligands and metal complexes agree with their proposed structures/geometries. The synthesized ligands, along with their metal complexes were screened for their antibacterial activity against Escherichia coli, Bacillus subtillis, Shigella flexneri, Staphylococcus aureus, Pseudomonas aeruginosa and Salmonella typhi and for antifungal activity against Trichophyton longifusus, Candida albicans, Aspergillus flavus, Microsporum canis, Fusarium solani and Candida glaberata. The results of these studies show the metal complexes to be more antibacterial/antifungal against two or more species as compared to the uncomplexed Schiff-base ligands. 相似文献

The DNA-binding and photonuclease activity of newly synthesized tetra-azamacrocyclic ligand L (C32H32N8O4) and its complexes of type [MLCl2] and [ML]Cl2 (where M = Co(II), Fe(II) and Cu(II); L = N,N′-[3-(4-{5-[(2-amino-ethylamino)-methyl]-isoxazol-3yl}-phenyl)-isoxazol-5-yl methyl-ethane-1,2-diamine] are specified. An octahedral geometry has been proposed for Fe(II) and Co(II) complexes, while the Cu(II) complex has a square planar environment. The absorption spectral results indicate that the complexes bind with the base pairs of DNA, with an intrinsic binding constant Kb of Fe(II), Co(II), and Cu(II) complexes found to be 3.2 × 104 M?1, 5.3 × 104 M?1, and 4.2 × 104 M?1, respectively, in 5 mM Tris-HCl/50 mM NaCl buffer at pH 7.2. The large enhancement in the relative viscosity of DNA on binding to the complexes supports the proposed DNA binding modes. The viscosity and thermal denaturation studies sustain the effective intercalation with DNA. The DNA photocleavage studies demonstrated that compounds exhibit significant photonuclease activity by a concentration dependent on singlet oxygen mediated mechanism. 相似文献

The two uncharged compounds 25,26,27,28-(2-N,N-di methyldithiocarbamoylethoxy)calix[4]arene (1) and 25,26,27,28- (2-methylthioethoxy)calix[4]arene (2) are effective extractants for transferring Hg(II), Ag(I), Pd(II) and Au(III) from aqueous solution into chloroform. The electronic absorption spectra of 1 and 2 show additional bands at long wavelength upon complexation with AuCl4−, PdCl42− and PdBr42−, and analogous bands for Hg2+ and Ag+ with 1. For 1 these new bands are considered to be either of the charge transfer type or transitions within the C=S moiety. These new bands for the complexes with 2 are assigned to LMCT transitions of the S → M type. These spectral features are used to obtain information about the solution structures of the complexes that are formed between these metal ions and both 1 and 2. 相似文献

The serum concentrations of copper, zinc, iron, and cobalt and copper/zinc ratio were investigated in horses infected with
equine herpesvirus-1 (EHV-1). Nine horses were naturally infected with the virus and nine healthy horses served as controls.
The concentrations of copper, zinc, iron, and cobalt were determined spectrophotometrically in the blood serum of all horses.
The results were (expressed in micrograms per deciliters) copper 2.80 ± 0.34 vs 1.12 ± 0.44, zinc 3.05 ± 0.18 vs 0.83 ± 0.06,
iron 2.76 ± 0.17 vs 3.71 ± 0.69, cobalt 0.19 ± 0.37 vs 0.22 ± 0.45, and copper/zinc ratio 0.72 ± 0.38 vs 1.41 ± 0.36 for control
vs infected group, respectively. In conclusion, copper and zinc concentrations of the infected group were lower than the control
group (p < 0.001), whereas iron concentration and the copper/zinc ratio of the infected group were higher than the control group (p < 0.05 and p < 0.001). The cobalt concentration was not found to be statistically different between two groups. It might be emphasized
that copper/zinc ratio was significantly affected by the EHV-1 infection, so it could be taken into consideration during the
course of infection.

Abstract: The activities mediated by the N -methyl-D-aspartate (NMDA) receptor were studied in cultured rat cerebellar granule cells. Micromolar concentrations of the metal binding compounds, EDTA, cysteine, and histidine, as well as serum albumin strongly potentiated receptor activity in the presence of millimolar concentrations of Ca2+ and Mg2+ . The findings indicated that these agents remove an endogenous metal, probably Zn2+ , which attenuates NMDA receptor-mediated 45 Ca uptake and toxicity. Several added metal ions were therefore tested at low micromolar concentrations. Zn2+ was found to be the most potent inhibitor of NMDA-induced 45 Ca uptake, followed by Cu2+ and Fe2+ . Co2+ , Cd2+ , Fe3+ , and AI3+ had no significant effect, whereas Ni2+ potentiated the 45 Ca uptake but inhibited at much higher concentrations. The potentiating agents that remove the endogenous metal had a particularly dramatic effect in the presence of Mg2+ , the voltage-dependent suppressor of the NMDA receptor. Mg2+ also played an important role in the inhibitory effect of added Zn2+ . Much lower concentrations of Zn2+ were needed to achieve inhibition of NMDA-induced 45 Ca uptake in the presence of Mg2+ . Under a variety of conditions, a very good correlation was found between NMDA receptor-mediated 45 Ca uptake and the magnitude of acute neurotoxicity. 相似文献

This study examines factors affecting oral bioaccessibility of metals in household dust, in particular metal speciation, organic carbon content, and particle size, with the goal of addressing risk assessment information requirements. Investigation of copper (Cu) and zinc (Zn) speciation in two size fractions of dust (< 36 μ m and 80–150 μ m) using synchrotron X-ray absorption spectroscopy (XAS) indicates that the two metals are bound to different components of the dust: Cu is predominately associated with the organic phase of the dust, while Zn is predominately associated with the mineral fraction. Total and bioaccessible Cu, nickel (Ni), and Zn were determined (on dry weight basis) in the < 150 μ m size fraction of a set of archived indoor dust samples (n = 63) and corresponding garden soil samples (n = 66) from the City of Ottawa, Canada. The median bioaccessible Cu content is 66 μ g g?1 in dust compared to 5 μ g g?1 in soil; the median bioaccessible Ni content is 16 μ g g?1 in dust compared to 2 μ g g?1 in soil; and the median bioaccessible Zn content is 410 μ g g?1 in dust compared to 18 μ g g?1 in soil. For the same data set, the median total Cu content is 152 μ g g?1 in dust compared to 17 μ g g?1 in soil; the median total Ni content is 41 μ g g?1 in dust compared to 13 μ g g?1 in soil; and the median total Zn content is 626 μ g g?1 in dust compared to 84 μ g g?1 in soil. Organic carbon is elevated in indoor dust (median 28%) compared to soil (median 5%), and is a key factor controlling metal partitioning and therefore bioaccessibility. The results show that house dust and soil have distinct geochemical signatures and should not be treated as identical media in exposure and risk assessments. Separate measurements of the indoor and outdoor environment are essential to improve the accuracy of residential risk assessments. 相似文献

Biosorption of chromium, copper, and nickel from aqueous solution by Tamarindus indica fruit nut testa (TFNT) in its pristine and acid-treated forms was studied under equilibrium and column-flow conditions. TFNT, a tannin-containing material, was characterized by energy dispersion x-ray fluorescence (EDXRF) and Fourier transform infrared (FTIR) spectral techniques and surface analysis. The effect of experimental variable parameters such as pH, concentration of metal ions, amount of adsorbent, and contact time on adsorption was investigated. Batch isothermal equilibrium data were analyzed on the basis of Langmuir and Freundlich adsorption isotherms. The kinetics of the adsorption process were studied in terms of Lagergren first-order kinetic model. The monolayer adsorption capacities of pristine and acid-treated forms of tamarind seed coat were found to be 44.8 and 77.5 mg/g for Cr(VI), 55.8 and 99.0 mg/g for Ni(II), and 84.7 and 85.4 mg/g for Cu(II) ions, respectively. The column-flow adsorption data were used to obtain breakthrough curves. The biosorbent loaded with the metal ions was regenerated using 1.0 M HCl and the regenerated bed was used for subsequent adsorption-desorption cycle. 相似文献

The synthesis and characterization of a ferrocenyl-derived tridentate ligand, ferrocenyltris((methylthio)methyl)borate (FcTtP−), and its representative metal complexes, [(FcTt)Cu]4 and [FcTt]2M (M = Fe, Co and Ni), are reported. The M = Fe complex exhibits spin-crossover behavior with a μeff = 1.19 μB at 25°C. The low-spin Co(II) derivative (1.88 μB) exhibits a characteristic axial electron paramagnetic resonance (EPR) spectrum, gav = 2.13, A = 53 G and A¦ = 43 G. The [FcTt]2M complexes display reversible two-electron redox processes assigned to ligand-centered events about 200 mV negative of the ferrocene-ferrocenium couple. [(FcTt)Cu]4 and [FcTt]2Ni have been characterized by X-ray diffraction. The copper(II) complexes with ciprofloxacin (CFLH), levofloxacin (LFLH), norfloxacin (NFLH), and neutral bidentate ligands have been synthesized and characterized. The complexes have been evaluated for their antibacterial activity against selective species. Complexes have been also checked for their interacting behavior with DNA, and were found to have two different modes of interaction, classical and partial intercalation. DFT calculations for M@C12H12N6 and M@C12H24O6 (where M = Rb+, Cs+, Sr2+, and Ba2+, C12H12N6 = hexaaza[18]annulene, and C12H24O6 = 18-crown-6) were performed using the recently developed model core potential parametrization.
