Conventional native "green gel" systems resolve at most 10 chlorophyll-protein complexes from thylakoid membranes of higher plants and green algae. Such analyses suggest a simplicity of the thylakoid membrane that is not supported by a growing body of evidence on the heterogeneity of photosystems I and II (PSI and PSII) and their associated antennae (LHCI and LHCII). We report here the development and characterization of a low ionic strength native "green gel" system that resolves from 16 to 20, mostly large chlorophyll-protein complexes from a variety of higher plant and green algal species with very little release of free pigment. In Chlamydomonas, this system resolves multiple PSI-LHCI complexes, multiple PSII-LHCII complexes, four oligomeric LHCII complexes, as well as several low electrophoretic mobility reaction center complexes, and a number of small complexes. We have obtained similar resolution with a large number of higher plant and green algal species. We also demonstrate how this system can be used as a sort of "fingerprinting" technique to distinguish thylakoids of different species, and for the analysis of photosynthetic mutants, using the chlorophyll b-less chlorina f2 mutant of barley as an example.  相似文献

Chemostat-grown cells of the chlorophyte Dunaliella tertiolecta (Butcher) exposed to triphenyltin were examined using transmission electron microscopy. Following a 1-h exposure to 21 and 84μM triphenyltin, mitochondria underwent structural damage and the thylakoid membranes of a small proportion of cells spread from the usual compact arrangement. Prolonging the exposure time resulted in significant cell lysis in cultures exposed to 84μM triphenyltin. Received 05 May 1997/ Accepted in revised form 28 January 1998  相似文献

The kinetics of a conformational change which occurs in nucleosome core particles at about 1 mM ionic strength have been studied by observing changes in the fluorescence of labeled histone H3. The unfolding reaction is intramolecular since no concentration dependence is observed. However, the kinetics are unexpectedly complicated and reveal evidence of at least three relaxation times. It is possible to fit the kinetics observed under several conditions to a consistent four-state cyclic mechanism in which folded and unfolded forms can inter-convert by two parallel pathways, each involving a distinct intermediate. While the data are not sufficient to establish this mechanism as a unique choice, they exclude many simpler possibilities. The cyclic mechanism is quite reasonable in view of what is currently known about the structures of the folded and unfolded forms.  相似文献

Thyroglobulin molecules were studied in the electron microscope with negative staining technique. In a first series of experiments samples of thyroglobulin varying in iodine content from 0.5 to 0.03% were prepared from the thyroids of mice and rats kept on iodine-poor diets. All samples contained thyroglobulin molecules of the normal ovoid shape, not deviating in size or shape from molecules obtained from normal thyroids. However, in addition, another type of molecule having a cylindrical shape was observed in all samples. The proportion of these cylindrical molecules increased from a few per cent in the moderately iodine-poor thyroglobulin samples to more than 80% in the highly iodine-deficient thyroglobulin (0.03%). In a second series of experiments extremely iodine-poor thyroglobulin (smaller than 0.005%) was obtained from propylthiouracil-treated rats. In these preparations practically all molecules had a cylindrical shape. These samples also contained smaller particles interpreted to be dissociation products. The cylindrical molecules were of two types, one appearing compact and measuring 250 times 135 A (length times diameter) and the other appearing porous and having a length of 145 and a diameter of 205 A. It is concluded that the cylindrical molecules represent non- or low-iodinated thyroglobulin and it is suggested that the porous cylindrical molecule is an unfolded form of the compact cylinder.  相似文献

Previous crystallographic studies of tuna ferricytochrome c have employed crystals grown from solutions of ammonium sulfate, corresponding to an ionic strength of 9.5 M (Takano, T., and Dickerson, R. E. (1981) J. Mol. Biol. 153, 95-115). To obtain a structure at a lower ionic strength, the ferric tuna protein was crystallized at neutral pH with polyethylene glycol at an ionic strength of 45 mM, These crystals (space group P2(1), a = 37.11 A, b = 107.66 A, c = 55.75 A, beta = 105.3 degrees) contain four molecules/asymmetric unit and grow to dimensions of 0.2 X 0.4 X 1.0 mm in 2-4 weeks. They diffract to beyond 1.8 A and are stable in the x-ray beam. We have recorded 28,198 unique Bragg reflections (83% of those possible) to a resolution of 1.89 A from a native crystal. We are undertaking a solution of this structure by the molecular replacement method.  相似文献

The cell sap of the internode ofNitella flexilis was replaced with the isotonic artificial pond water of high Ca2+-concentration (0.1 mM KCl, 0.1 mM NaCl, 10 mM CaCl2 and 275 mM mannitol) and changes in osmotic value and concentrations of K+, Na+ and Cl of the cells were followed. When the operated cells were incubated in the artificial pond water containing 0.1 mM each of KCl, NaCl, CaCl2, they survived for only a short period of time (<10 hr). The cells did not absorb ions from the artificial pond water and showed a conspicuous decrease in the rate of cytoplasmic streaming. In such cell the concentration of K+ in the protoplasm decreased significantly. In order to reverse normal concentration gradients of K+ and Na+ across the protoplasmic layer, the cells of low vacuolar ionic concentrations were incubated in the artificial cell sap (90 mM KCl, 40 mM NaCl, 15 mM CaCl2, 10 mM MgCl2). It was found that the cells rapidly absorbed much K+, Na+ and Cl and survived for a longer period (1–2 days). During this period the rate of cytoplasmic streaming was nearly normal. Furthermore, the cell lost much mannitol, indicating an enormous increase in permeability to it. Since both absorption of ions and leakage of mannitol at 1 C occurred at nearly the same rates as at 22 C, the processes are assumed to be passive.  相似文献

This study shows that xanthan molecules at room temperature may assume at least three different conformations in 0.1 m NaCl aqueous solutions in which the local structure is ordered: (1) the native compact double helix, (2) the extended double helix, and (3) the extended single helix. Experiments including viscosity, low-angle light scattering and optical rotation measurements have been carried out with a fully pyruvated and fully acetylated native laboratory sample supplied as fermentation broth. Two major conformation changes of the native double helix which were found irreversible in our experimental conditions can be induced by treatments at low ionic strength. After treatment in 10−4m NaCl, xanthan is still a double helix in 10−1m NaCl, but the backbone of each strand has been extended. After the sample has been in 10−5m NaCl, the double helix has been dissociated and a single helix sample is obtained. Thus, the denaturing of xanthan is a two-step process. The first step consists of the extension of the two chains inside the double helix, and the second is a dissociation of the native double strand.  相似文献

The method of Na-dodecylsulfate electrophoresis in polyacrylamide gel demonstrated that the so-called tonoactomyosin of smooth muscles extracted from the muscle homogenate with salt media of low ionic power represented a complicated protein system into whose composition there was included a heavy myosin chain with the mol wt of 210000, premyosin subunit with the mol wt of 230000, actin, and, possibly, a number of other proteins. The extracts of low ionic power possessed Mg2+ and Ca2+ activated by ATP-ase activity. The premyosin subunit was also revealed in the extracts of low ionic power from the skeletal muscle homogenates. It is supposed that premyosin subunit was included into the enzymatic system responsible for the ATP-asic properties of the extracts of low ionic power from the homogenates of different types of muscles.  相似文献

Neutron scattering studies of nucleosome structure at low ionic strength   总被引:1,自引:0,他引:1  
Ionic strength studies using homogeneous preparations of chicken erythrocyte nucleosomes containing either 146 or 175 base pairs of DNA show a single unfolding transition at about 1.5 mM ionic strength as determined by small-angle neutron scattering. The transition seen by some investigators at between 2.9 and 7.5 mM ionic strength is not observed by small-angle neutron scattering in either type of nucleosome particle. The two contrasts measured (H2O and D2O) indicate that only small conformational changes occur in the protein core, but the DNA is partially unfolded below the transition point. Patterson inversion of the data and analysis of models indicate that the DNA in both types of particle is unwinding from the ends, leaving about one turn of supercoiled DNA bound to the histone core in approximately its normal (compact) conformation. The mechanism of unfolding appears to be similar for both types of particles and in both cases occurs at the same ionic strength. The unfolding observed for nucleosomes in this study is in definite disagreement with extended superhelical models for the DNA and also disagrees with models incorporating an unfolded histone core.  相似文献
