The Fab arms of immunoglobulin G (IgG) have long been known to hinge about their joint with the Fc subunit. Using monoclonal antibodies bound to influenza haemagglutinin (HA) as position markers, we now show that these arms can also rotate about their long axis with respect to Fc. We also show that when two IgGs are bound cyclically with two HA molecules, the arms can bend between the variable and constant domains to accommodate bond angle constraint.  相似文献

The physical properties of a triple-helical DNA four-way junction J(T2T4) have been characterized by means of UV spectroscopy, CD spectroscopy, and differential scanning calorimetry (DSC). J(T2T4) is another four-way junction that was designed in addition to J(T1T3) (N. Makube and H. H. Klump (2000) Arch. Biochem. Biophys. 377, 31-42) to study the effects of third strands on the stability of the four-way junction with triple-helical arms. The pH titration curves illustrate the sequential folding of single strands to double-helical four-way junctions and finally the binding of third strands to their respective W-C duplexes. CD measurements confirm triplex formation under appropriate pH and ionic strength conditions. The CD spectra also suggest different melting patterns for the triple-helical arms of J(T2T4). The melting temperature as a function of pH or ionic strength characterizes the effect of the third strands on the structural stability. Increased sodium concentration and low pH conditions enhances and stabilizes the overall structure of the junction. The results also indicate that all triplexes in J(T2T4) are formed in the absence of salt and at low pH; however, the junction may, under these conditions, assume a conformation different from the one assumed in the presence of salt. Through the deconvolution of DSC data, the calorimetric enthalpies associated with melting of arms of the junctions were determined. The loops are designed to have the same enthalpic effect on the different arms. The stabilizing effect of the loops is more pronounced when those loops are shifted from arms 1 and 3 in J(T1T3) to arms 2 and 4 in J(T2T4) without changing any of the sequences. Overall, J(T2T4) is slightly more stable than J(T1T3). The differences can be attributed to sequence effects rather than structural effects. All the results illustrate that binding of the third strand in either of the two orientations 5'5'3' (J(T2T4)) or 5'3'3' (J(T1T3)) stabilizes the underlying double-helical four-way junction and its triple-helical arms.  相似文献

The contractile vacuole complex is a membrane-bound osmoregulatory organelle of fresh water protozoa such as Paramecium. In Paramecium it consists of a central vacuole (the contractile vacuole) and 5-10 arms that radially extend from the vacuole into the cytosol (the radial arms). Excess cytosolic water, acquired osmotically, is segregated by the radial arms and enters the vacuole, so that the vacuole swells (the fluid-filling phase). The vacuole then rounds (the rounding phase) and the radial arms sever from the vacuole. The vacuole membrane then fuses with the plasma membrane at the pore region and the pore opens. The vacuole shrinks as its fluid is discharged through the pore (the fluid-discharging phase). The pore closes when the fluid has been discharged. The radial arms then reattach to the vacuole, so that the vacuole swells again as the fluid enters from the arms (the next fluid-filling phase). We found that the vacuole continued to show rounding and slackening even after it together with a small amount of cytosol had been isolated from the cell. Using a microcantilever placed on the surface of the vacuole the tension of the in vitro vacuole increased to 5 x 10(-3)N m(-1) as the vacuole rounds, and its lowest value was 1 x 10(-4)N m(-1) during slackening. We propose a hypothesis that an increase in the spontaneous curvature of the organelle's membrane leads to an increase in membrane tension and thus to the vacuole's rounding, severing of the radial arms from the vacuole, and opening of the pore. Conversely, a decrease in the spontaneous curvature accompanied by a decrease in membrane tension could lead to the closing of the pore and reattachment of the radial arm at the start of the fluid-filling phase.  相似文献

Transverse sections of flagellar axonemes from a variety of lower plant spermatozoids were examined by electron microscopy. Motile sperm of four ferns (Marsilea, Pteridium, Lygodium and Aneimia), a horsetail (Equisetum) and a liverwort (Marchantia) were fixed in the presence of tannic acid to visualise the dynein arms. In all cases the inner dynein arms were clearly resolved but the outer arms were absent. Absence of outer arms therefore appears to be a common feature of the archegoniate plants. The implications of these findings to our understanding of the evolution of the land plants and the role of the dynein arms in flagellar beating is discussed.  相似文献

The opponent-independent cost game is an animal contest in which the bigger and more heavily-armed opponent wins a disputed resource without significant fighting costs. A strategy is a choice of investment level in armament. Increasing armament is assumed to have fitness costs that are unrelated to contests; i.e. the cost of an individual's investment in arms is independent of the strategy played by an opponent.Previous work with this model showed that no ESS exists if a strategy prescribes an arms level exactly. This is equivalent to the notion that there is no environmental variation in the arms level attained by a given strategy. If environmental variation is introduced, a pure ESS can generally exist. A strategy is assumed to prescribe an exact investment cost, but this is translated into a probability distribution of arms levels attained, rather than an exact arms level. Increasing investment increases the mean of the arms level distribution. The ESS investment level depends both on how environmental factors distribute arms levels, and on the shape of the cost function (i.e. on the way that costs increase with investment); in some instances there is no ESS. Two types of model are investigated; in one fitness is additive (benefit-cost), in the other it is multiplicative (benefit × survivorship). The multiplicative model is likely to apply to the case where contests are between males for access to females. Here the ESS investment level (an ESS degree of risk that a male sustains as a result of armament) increases as fewer individuals guard the available resources. Thus sexual size dimorphism (male/female size) and relative male armament should increase as harem size increases. The ESS investment level will also be highest if most individuals are small and poorly armed, as would often be the case where size increases throughout life.The model can be applied to coevolutionary arms races between two classes of opponent, such as prey and predator, or parent and offspring. Here the ESS is likely to be a pair of ESS arms levels, one for each class of opponent.  相似文献

Genetic and cytogenetic investigations of two unstable Y-linked translocations in Anopheles culicifacies indicate that crossing over can occur between the X and Y chromosomes in this species. Moreover, these observations suggest that the long arms of the X and Y chromosomes are homologous and that the sex determining mechanism(s) is located in the short arms.  相似文献

Our goal was to determine the direction of force generation of the inner dynein arms in flagellar axonemes. We developed an efficient means of extracting the outer row of dynein arms in demembranated sperm tail axonemes, leaving the inner row of dynein arms structurally and functionally intact. Sperm tail axonemes depleted of outer arms beat at half the beat frequency of sperm tails with intact arms over a wide range of ATP concentrations. The isolated, outer arm-depleted axonemes were induced to undergo microtubule sliding in the presence of ATP and trypsin. Electron microscopic analysis of the relative direction of microtubule sliding (see Sale, W. S. and P. Satir, 1977, Proc. Natl. Acad. Sci. USA, 74:2045-2049) revealed that the doublet microtubule with the row of inner dynein arms, doublet N, always moved by sliding toward the proximal end of the axoneme relative to doublet N + 1. Therefore, the inner arms generate force such that doublet N pushes doublet N + 1 tipward. This is the same direction of microtubule sliding induced by ATP and trypsin in axonemes having both inner and outer dynein arms. The implications of this result for the mechanism of ciliary bending and utility in functional definition of cytoplasmic dyneins are discussed.  相似文献

Treatment of porcine immunoglobulin M (IgM) with pepsin at pH 4.6 and 37 degrees C was found to gradually remove Fab arms and Cmicro2 domains over a period of 18h. Structural studies failed to find any other change. The main products can therefore be regarded as IgM-like molecules with limited numbers of Fab arms and Cmicro2 domains. Results indicated that this removal of Fab arms is probably a random process. As the average number of Fab arms per molecule was decreased the ability to agglutinate Salmonella oranienburg (mt-H) gradually diminished. Complement fixation by the complexes however, decreased rapidly, and became negligible when the average number of Fab arms was four. This was confirmed by using a preparation containing mainly molecules with three or four Fab arms. The overall results showed that molecules with three or four Fab arms can agglutinate Salmonella but that these complexes do not fix complement. Molecules with five arms probably behave like those with four. Complexes formed by molecules with six arms fix complement quite efficiently. Possible explanations for these results are discussed.  相似文献

Recommendations vary on whether blood pressures should be measured in the right or in the left arm because no frequency distributions for a pressure difference between the arms exist. We took a total of 12 blood pressure determinations in both arms of 174 elderly persons and analyzed the data by a least-squares components of variance method. The mean difference between the arms (right minus left) was 0.93 mm of mercury for systole and 0.70 mm of mercury for diastole. For systole the proportion of persons having arm pressure differences exceeding 10 mm of mercury is 1.4% and that exceeding 7.5 mm of mercury is 6.5%. For most people, the pressure difference between the arms is small.  相似文献

Moment arms are important for understanding muscular behavior and for calculating internal muscle forces in musculoskeletal simulations. Biarticular muscles cross two joints and have moment arms that depend on the angle of both joints the muscles cross. The tendon excursion method was used to measure the joint angle-dependence of hamstring (biceps femoris, semimembranosus and semitendinosus) moment arm magnitudes of the feline hindlimb at the knee and hip joints. Knee angle influenced hamstring moment arm magnitudes at the hip joint; compared to a flexed knee joint, the moment arm for semimembranosus posterior at the hip was at most 7.4 mm (25%) larger when the knee was extended. On average, hamstring moment arms at the hip increased by 4.9 mm when the knee was more extended. In contrast, moment arm magnitudes at the knee varied by less than 2.8 mm (mean=1.6 mm) for all hamstring muscles at the two hip joint angles tested. Thus, hamstring moment arms at the hip were dependent on knee position, while hamstring moment arms at the knee were not as strongly associated with relative hip position. Additionally, the feline hamstring muscle group had a larger mechanical advantage at the hip than at the knee joint.  相似文献

The upper arms of adult newts (Notophthalmus viridescens) were surgically manipulated to create double-half dorsal, double-half ventral, double-half anterior, and double-half posterior upper arms, and longitudinal half-dorsal, half-ventral, half-anterior, and half-posterior upper arms. Amputation through the double-half upper arms usually failed to elicit normal distal regeneration, despite the fact that an apparently normal regeneration blastema was initially formed. Instead, regeneration in these cases was limited to the formation of a variable number of small cartilage elements. On the basis of these results it is concluded that a complete limb circumference is required for distal transformation in newts, in addition to the well-established requirements for a wound epidermis, adequate innervation and dedifferentiation leading to blastema formation. A model for the sequential generation of new parts of the limb pattern during distal transformation from a complete circumference is presented. This model can also account for the occurrence of normal early stages of regeneration in double-half upper arms. Half upper arms which were amputated immediately were shown to develop single, complete regenerates. If amputation of half upper arms was delayed three or more weeks to permit complete wound healing, a supernumerary limb from the lateral wound surface sometimes developed in addition to a complete, single limb from the distal amputation surface.  相似文献

We have examined the effects of ATP and deoxycholate (DOC) on the cytoskeletal organization of Triton-demembranated microvilli (MV) isolated from chicken intestine brush borders. Isolated MV are composed of a core of tightly bundled microfilaments from which arms project laterally to the plasma membrane with a 33-nm periodicity. These lateral arms spiral around the core microfilaments as a helix with a 25 degrees pitch. Demembranated MV consist of four polypeptides with mol wt of 110,000, 95,000, 68,000, and 42,000, present in molar ratios of 1.1:1.6:1.3:10.0. After addition of 50 microM ATP and 0.1 mM Mg++, the cytoskeletons are organized as a tight bundle of microfilaments from which lateral arms are missing. In these ATP-treated cytoskeletons, the 110-kdalton polypeptide is reduced in amount and the 95,000, 68,000, and 42,000 polypeptides are present in a 1.3:1.2:10.0 ratio. In contrast, after incubation with 0.5% DOC, the core microfilaments are no longer tightly bundled yet the lateral arms remain attached with a distinct 33-nm periodicity. These DOC-treated cytoskeletons are depleted of the 95,000 and 68,000 polypeptides and are composed of the 110,000 and 42,000 polypeptides in a 2:10 molar ratio. These results suggest that the microfilaments are associated into a core bundle by the 95- and 68-kdalton polypeptides and from this core bundle project the lateral arms composed of the 110-kdalton polypeptide.  相似文献

The nucleolus organizing regions (NORs) on the short arms of chromosomes 1A(m) and 5A(m) of diploid wheat, Triticum monococcum L., are at the most distal loci in the linkage maps of these two chromosome arms. This distal location differs from the interstitial location of the Nor loci on chromosome arms 1BS of tetraploid Triticum turgidum L. and hexaploid T. aestivum L., 5DS of T. aestivum and diploid Ae. tauschii Coss., and 5HS of barley. Moreover, the barley 5HS locus is at a different location than the 5DS locus. However, other markers, including the centromeres, are colinear. These findings showed that the major Nor loci have repeatedly changed position in the chromosome arms during the radiation of species in the tribe Triticeae without rearrangements of the linkage groups. It is suggested that Nor loci may change position via dispersion of minor loci, that are shown here to exist in the T. monococcum genome, magnification of gene copy numbers in these minor loci, and subsequent deletion of the original major loci. Implications of these findings for the use of rRNA nucleotide sequences in phylogenetic reconstructions are pointed out.  相似文献

Abstract

Biomechanical investigations examining shoulder function commonly observe a high degree of inter-individual variability in muscle activity and kinematic patterns during static and dynamic upper extremity exertions. Substantial differences in musculoskeletal geometry between individuals can alter muscle moment arms and lines of action that, theoretically, alter muscle activity and shoulder kinematics. The purposes of this research were to: (i) quantify model-predicted functional roles (moment arms, lines of action) of the scapulohumeral muscles, (ii) compare model predictions to experimental data in the literature, and (iii) evaluate sensitivity of muscle functional roles due to changes in muscle attachment locations using probabilistic modeling. Monte Carlo simulations were performed to iteratively adjust muscle attachment locations at the clavicle, scapula, and humerus of the Delft Shoulder and Elbow Model in OpenSim. Muscle moment arms and lines of action were quantified throughout arm elevation in the scapular plane. In general, model-predicted moment arms agreed well with the reviewed literature; however, notable inconsistencies were observed when comparing lines of action. Variability in moment arms and lines of action were muscle-specific, with 2 standard deviations in moment arm and line of actions as high as 25.8?mm and 18.8° for some muscles, respectively. Moment arms were particularly sensitive to changes in attachment site closest to the joint centre. Variations in muscle functional roles due to differences in musculoskeletal geometry are expected to require different muscle activity and movement patterns for upper extremity exertions.  相似文献

Comatulids are able to perform quick and complex movements of the arms which are used to swim, creep, walk, and also form a rigid, feeding-filtration fan. To perform such versatile movements, the arms of these animals are equipped with a classical endoskeletal system, with joints, muscles, ligaments, and a hydroskeleton of three different coelomic channels. Light microscopic study of the detailed anatomical organization of the arm clarifies both the complex relations between the parts involved in the movements and their functional responsibilities. In particular, (1) the ventral muscle bundles show a heterogeneous structure that consists of different and variously arranged populations of fibers, which allows the different flexing movements of the arms (i.e., flexion and maintaining the flexed state); (2) the ligaments (both dorsal and interarticular) consist only of collagen fibrils and, therefore, have a passive function in binding the skeletal pieces together: their possible active engagement in the extending movements of the arms is thus excluded; (3) owing to the absence of other suitable antagonists to the flexor muscles, the only efficient antagonist system seems to be the coelomic cavities, which are well separated from each other and are also provided with muscular valves. They thus function as typical hydraulic systems, which allows the arm to perform both simple extensions and very complex combined movements and to maintain some rigid straight or twisted positions.  相似文献

The elevated plus-maze test is usually run with a short edge surrounding the open arms in order to prevent the rats from falling. The present experiment investigated the role of transparent edges differing in heights: 1 (used as control), 5, 10, 20 and 40 cm, the latter the same height as the closed arm walls. Additionally, this 40-cm high transparent edge was also studied covered by white translucent or black opaque paper. The data show that the time spent in the open arms was significantly greater when the edge height was 5, 10 or 40 cm covered by the white or black paper. However, there were no differences from the 1-cm control edge when the height was 40 cm transparent. A similar effect was observed when entries in the open arms and total entries were analyzed. The facts that there were no differences when the open arms were surrounded by 1- or 40-cm transparent edges (which allow thigmotaxis) and that the same 40-cm edge caused increases in exploratory behavior when covered by papers indicate that vision triggers aversion to open spaces.  相似文献

Recombination of chromosomes 3A(m) and 5A(m) of Triticum monococcum with closely homeologous chromosomes 3A and 5A of T. aestivum was compared with recombination across corresponding homologous chromosome pairs. Differentiation between the homeologues impacted recombination in the proximal regions of the long arms the most and in the distal regions of the long arms the least. It is concluded that this variation principally reflects allocation of multiple crossovers across an arm and positive crossover interference across chromosome arms. Recombination rates between homeologous chromosomes 5A(m) and 5A differed in the opposite sexes.  相似文献

The formation of the four-way junction containing four triple-helical arms has been demonstrated using chemical methods (polyacrylamide gel electrophoresis and chemical footprinting using OsO(4) as a probe) and physical methods (UV absorbance melting and DSC). The junction J(T1T3) was assembled from two 20-mer purine strands and two 44-mer pyrimidine strands. To determine the contribution of the different arms to the stability of the complete structure of J(T1T3), the junction was compared to two simplified substructures, J(T1) and J(T3), respectively. Common to these complexes is the underlying double-helical four-way junction Js. Addition of Na(+) had a profound effect on stabilizing and subsequently folding the junctions into the stacked X-structures. The following results support the structure present: (i) The native polyacrylamide electrophoresis exhibits only a single band(s) corresponding to one species present when all four single strands are mixed in equal amounts. (ii) OsO(4) modifications were investigated at pH 5.0 and in the presence of 10 mM Mg(2+) and 100 mM Na(+). There is no cleavage of thymine residues at the branch point and throughout the structure. (iii) The thermal unfolding of J(T1) and J(T3) illustrates that the triple-helical arms are more stable than the double-helical arms which are contained in these junctions and that J(T1T3) with four triple-helical arms is slightly more stable than J(T1) and J(T3). (iv) The calorimetric transition enthalpies determined for the arms of J(T1T3) are comparable to those associated with the unfolding of its corresponding arms in J(T1) and J(T3). The results also illustrate that the formation of the junctions is not restricted by the pH, [Na(+)], sequence composition of the arms, and/or the loop position.  相似文献

The complete larval development of an echinoid in the family Aspidodiadematidae is described for the first time from in vitro cultures of Aspidodiadema jacobyi, a bathyal species from the Bahamian Slope. Over a period of 5 months, embryos grew from small (98-micron) eggs to very large (3071-micron) and complex planktotrophic echinopluteus larvae. The fully developed larva has five pairs of red-pigmented arms (preoral, anterolateral, postoral, posterodorsal, and posterolateral); fenestrated triangular plates at the bases of fenestrated postoral and posterodorsal arms; a complex dorsal arch; posterodorsal vibratile lobes; a ring of cilia around the region of the preoral and anterolateral arms; and a long, unpaired posterior process containing a fenestrated rod. The presence of a posterior process and posterodorsal arms makes the larva of Aspidodiadema jacobyi much more similar to larvae of irregular urchins in the order Spatangoidea than to other families of the order Diadematoida, to which the family is normally assigned. This unexpected larval form lends support to a recommendation that the Aspidodiadematidae should be either elevated to ordinal status as a sister group of the order Diadematoida, or split off as a sister group of the other families within the order. In either case, if we accept the parsimonious hypothesis that the aboral process and posterodorsal arms were derived only once in the evolutionary history of euechinoids, then the larval data suggest that the Aspidodiadematidae may be very near the node where the irregular and regular euechinoids first diverged.  相似文献

The elevated plus-maze is an apparatus composed of enclosed and open (elevated) arms and time spent in the open arms by a rat can be increased/decreased by anxiolytic/anxiogenic agents. In the forced swim test, floating behavior is used as an index of behavioral despair and can be decreased by antidepressant agents. As the comorbidity between anxiety and depression is a remarkable issue in human behavioral disorders, a possible relationship between the behaviors seen in the cited tests is of great relevance. In the present study, fifty-four male rats (Rattus norvegicus) were submitted to a plus-maze session and to a 2-day forced swim protocol. According to their time in the open arms, they were divided into three groups: Low Open, Medium Open and High Open. Some plus-maze measures were found to be coherent with time in the open arms and are suggested to also be reliable anxiety indexes. In the forced swim test, the Low Open group showed decreases in floating duration from forced swim Session 1 to Session 2, an alteration opposite to that observed in the other groups (particularly, the Medium Open group). The Low Open group also showed increases in floating latency, again in sharp contrast with the alteration found in the other groups. Accordingly, positive and negative correlation were found between time in the open arms and floating duration and latency, respectively. Results are compared to previous studies and mediation of the effect by reactivity to aversive stimulation or alterations induced by open arm exposure is discussed.  相似文献
