In this paper, the detailed morphology of Prorocentrum donghaiense Lu from both field samples and cultures was examined, and a taxonomic comparison was made between P. donghaiense and some related Prorocentrum spp. using morphological and molecular data and other published information. There were distinct differences among these species in morphological characteristics that historically have been presented as conservative features. The discrepancies extended beyond that of individual variations within the same species due to environmental factors. Therefore, these morphological features may not be conservative but, rather, polymorphic depending on environmental conditions. Based on this analysis, we suggest that the high-biomass bloom-forming species in the East China Sea, previously reported as Prorocentrum dentatum Stein, is P. donghaiense Lu. The species reported from the East China Sea and Japanese and Korean waters appear to be the same species. Molecular data also suggest that P. dentatum (CCMP1517) and P. donghaiense are genetically identical. Therefore, the geographic distribution of P. donghaiense may be much wider than expected.  相似文献

Difficulties often occur in separating closely related dinoflagellate species. In this study, the potential utility of mitochondrial cytochrome b (cob) gene sequence and mRNA editing characteristics was assessed using Prorocentrum Ehrenberg as a model. The cob sequences and the patterns of their mRNA editing were analyzed for several Prorocentrum taxa. Results revealed little difference in cob sequence and mRNA editing characteristics between geographic populations of P. minimum (Pavillard) Schiller, while a notable difference was detected between different species (P. minimum and P. micans Ehrenberg). Furthermore, these P. minimum populations consistently formed a tight cluster on phylogenetic trees inferred from cob sequences as well as mRNA editing characteristics, whereas different Prorocentrum species were well separated, with a genetic distance of 0.0042±0.0024 for the former and 0.0141±0.0012 for the latter (P<0.01; two‐tailed t‐test). When the analysis was applied to the case of P. donghaiense Lu et Goebel and CCMP1517 strain of P. dentatum Stein, no differences were detected between these two taxa with respect to cob mRNA editing pattern and only small differences equivalent to those between P. minimum populations were detected in terms of cob sequence. On the cob sequence‐ and editing‐based phylogenetic trees, P. donghaiense and P. dentatum CCMP1517 consistently clustered together at a position sister to P. minimum. The results suggest that cob, combined with its mRNA editing, can potentially be a useful delineator of Prorocentrum species, and that P. donghaiense and P. dentatum CCMP1517 are most likely the same species and both are closely related to P. minimum.  相似文献

Enhanced solar ultraviolet‐B (UV‐B) irradiation resulted from the stratospheric ozone depletion has adverse impacts on the primary productivity of marine algae. The effects of UV‐B enhancement on marine algae include reduction in photosynthesis and biomass, alteration in species competition, impaired chloroplast function, and damage to DNA. Harmful algal blooms (HAB) are an ubiquitous natural phenomenon caused by the excessive growth of phytoplankton. Many studies have examined the influence of supplemental UV‐B irradiation on different algae, but the effects of UV‐B irradiation on the interspecific growth interaction of HAB species has received little attention. In this study, Heterosigma akashiwo and Prorocentrum donghaiense were chosen to investigate the effect of UV‐B irradiation on the growth interaction between two HAB species by a co‐culture method. In a mixed culture experiment, H. akashiwo inhibited the growth of P. donghaiense in an inoculum dependent manner. In the absence of UV‐B irradiation, H. akashiwo dominated the competition with P. donghaiense under different inoculation proportions. Surprisingly, supplemental UV‐B irradiation alters the growth interaction between the two HAB species. P. donghaiense dominated the competition at the inoculation proportion of H: P = 1: 4. While several DNA-based methods have been developed for the putatively toxic dinoflagellate Pfiesteria piscicida Burkholder et Steidinger, an independent detection method such as immunofluorescence can be a useful alternative. In this study, P. piscicida-specific antisera were developed, and an immunofluorescence (IF) procedure was optimized. A total of six antisera were raised using whole cells (WCA) and the insoluble cellular fraction (ICF) as antigens, respectively, and their titer and specificity were examined using dot blot analysis and whole cell IF. Results showed that the two antisera produced from the ICF antigen had a markedly higher titer (1500) than the other four yielded from the WCA (200). In addition, the two ICF-derived antisera exhibited much higher species specificity, showing no cross-reaction with P. shumwayae, Cryptoperidiniopsis sp., Karlodinium micrum, and other more distant algae tested, and very low background for field collected samples. In evaluation of the IF technique using a P. piscicida-specific polymerase chain reaction (PCR) technique, results from both methods generally agreed well for both field samples (from eastern Long Island Sound) spiked with cultured P. piscicida and those containing naturally occurring P. piscicida (from Chesapeake Bay tributaries).  相似文献

Species-specific extracellular phosphatase activity (PA) was studied in natural phytoplankton with an emphasis on short-term variation in PA and its consequences for the diagnosis of inorganic phosphorus (P) deficiency. Extracellular PA was measured over 3 days at 4-h intervals to compare variations in algal PA in two distinct aquatic environments: in the Římov reservoir and acidified Plešné Lake. PA was directly detected in an epifluorescence microscope using fluorescently labelled enzyme activity; the fluorescence signal was quantified using image analysis. High short-term variation was found both in the total PA of the samples and specific PA of Ankyra ancora (G. M. Smith) Fott, Monoraphidium dybowski (Wolosz.) Hind. and Kom.-Legn., and Chlorogonium fusiforme Matwienko. The specific PA of these species varied between zero and 157 fmol cell−1 h−1. The PA of Chlorogonium fusiforme showed a significant relationship with the time of day. During the study period, populations of the certain species were present that differed in physiological status and activity due to diurnal or random changes caused by mixing in the reservoir or lake, or both. Therefore, species-specific extracellular PA should be interpreted carefully, as an indicator of P deficiency in freshwater phytoplankton if based on limited number of samplings.  相似文献

Proliferating cell nuclear antigen (PCNA) was detected in Prorocentrum donghaiense Lu and Dunaliella salina Teodoresco by enhanced chemiluminescence techniques with one mono-antibody. The observed band detected on western blots of P. donghaiense and D. salina had a molecular weight of 36–33 kDa rather than a PCNA-like protein with a size of 55 kDa reported in the dinoflagellates Crypthecodinium cohnii Biecheler and Gymnodinium catenatum Brav. The abundance of PCNA proteins was growth-stage dependent. Whole-cell immunoflurescence labeling showed that the PCNA antibody specifically stained the target proteins in P. donghaiense and D. salina, and PCNA is only present in the nucleus during the cell cycle. Synchronized cells of P. donghaiense show a cell cycle specific expression pattern with the highest expression in S phase and little expression in the G1 and G2/M phases. The results demonstrated that the PCNA-like proteins could be a marker for the estimation of marine phytoplankton growth rates. The different sizes of the PCNA-like proteins observed in dinoflagellates could be related to the variety of dinoflagellate chromosomal structure.  相似文献

在东海赤潮爆发区域运用围隔生态系实验方法,研究了不同营养盐条件下围隔生态系内多胺浓度变化。结果表明:2010年选用东海原甲藻赤潮爆发处海水,东海原甲藻是各围隔生态系内主要优势种,没有种群演替现象发生。两种营养盐添加方式下各围隔内精胺浓度维持较高水平,都呈现先波折下降后波折上升的趋势,与东海原甲藻的生长变化正好相反;各围隔内腐胺浓度水平较高,变化起伏较大,其中有两个实验组腐胺整体变化趋势与东海原甲藻生长趋势类似;所有围隔内亚精胺浓度最低,波动较小。2011年取用中肋骨条藻赤潮爆发处海水,所有围隔生态系内优势种都发生了从中肋骨条藻到东海原甲藻的演替。各围隔生态系内腐胺浓度最高,在中肋骨条藻生长初期腐胺浓度下降,随着中肋骨条藻的生长有所上升,实验后期随着东海原甲藻的生长又整体呈现出下降趋势;各实验组精胺浓度较低,在中肋骨条藻消亡东海原甲藻出现的种群演替期间,都呈现出较大波动;各围隔内亚精胺浓度较低,在整个种群演替过程中没有明显的变化。围隔生态系中补充营养盐,通过对浮游植物生长的影响,间接影响围隔生态系内的多胺变化。  相似文献

Extracellular human granulocyte-macrophage colony stimulating factor (hGM-CSF) expression was studied under the control of the GAP promoter in recombinant Pichia pastoris in a series of continuous culture runs (dilution rates from 0.025 to 0.2 h−1). The inlet feed concentration was also varied and the steady state biomass concentration increased proportionally demonstrating efficient substrate utilization and constancy of the biomass yield coefficient (Yx/s) for a given dilution rate. The specific product formation rate (qP) showed a strong correlation with dilution rates demonstrating growth associated product formation of hGM-CSF. The volumetric product concentration achieved at the highest feed concentration (4×) and a dilution rate of 0.2 h−1 was 82 mg l−1 which was 5-fold higher compared to the continuous culture run with 1× feed concentration at the lowest dilution rate thus translating to a 40 fold increase in the volumetric productivity. The specific product yield (YP/X) increased slightly from 2 to 2.5 mg g−1, with increasing dilution rates, while it remained fairly invariant, for all feed concentrations demonstrating negligible product degradation or feed back inhibition. The robust nature of this expression system would make it easily amenable to scale up for industrial production.  相似文献

The objective of the present study was to characterize the metabolism of Clostridium thermolacticum, a thermophilic anaerobic bacterium, growing continuously on lactose (10 g l−1) and to determine the enzymes involved in the pathways leading to the formation of the fermentation products. Biomass and metabolites concentration were measured at steady-state for different dilution rates, from 0.013 to 0.19 h−1. Acetate, ethanol, hydrogen and carbon dioxide were produced at all dilution rates, whereas lactate was detected only for dilution rates below 0.06 h−1. The presence of several key enzymes involved in lactose metabolism, including beta-galactosidase, glyceraldehyde-3-phosphate dehydrogenase, pyruvate:ferredoxin oxidoreductase, acetate kinase, ethanol dehydrogenase and lactate dehydrogenase, was demonstrated. Finally, the intracellular level of NADH, NAD+, ATP and ADP was also measured for different dilution rates. The production of ethanol and lactate appeared to be linked with the re-oxidation of NADH produced during glycolysis, whereas hydrogen produced should come from reduced ferredoxin generated during pyruvate decarboxylation. To produce more hydrogen or more acetate from lactose, it thus appears that an efficient H2 removal system should be used, based on a physical (membrane) or a biological approach, respectively, by cultivating C. thermolacticum with efficient H2 scavenging and acetate producing microorganisms.  相似文献

Interactions between Prorocentrum donghaiense Lu and Scrippsiella trochoidea (Stein) Loeblich III, two species of causative bloom dinoflagellates in China, were investigated using bi-algal cultures under controlled laboratory conditions. The growth of P. donghaiense and S. trochoidea were significantly suppressed when the initial cell densities were set at 1.9 × 104 cells mL−1 or 1.9 × 105 cells mL−1 for P. donghaiense and 1.0 × 104 cells mL−1 for S. trochoidea when the initial size/density ratio was 1:1 or 10:1, respectively, but no out-competement was observed in either bi-algal culture by the end. The simultaneous assay on the culture filtrate showed that P. donghaiense filtrate prepared at a lower initial density (1.9 × 104 cells mL−1) stimulated the co-cultured S. trochoidea at a density of 1.0 × 104 cells mL−1, but filtrate at a higher density (1.9 × 105 cells mL−1) depressed its growth. Differently, the filtrate of S. trochoidea at a density of 1.0 × 104 cells mL−1 significantly suppressed the growth of P. donghaiense at a density of 1.9 × 104 cells mL−1, but had little stimulatory effect on P. donghaiense at a density of 1.9 × 105 cells mL−1compared to the control (P > 0.05). It is likely that these two species of microalgae interact with each other mainly by releasing allelochemical substance(s) into the culture medium, and a direct cell-to-cell contact was not necessary for their mutual interaction. We then quantify their interactions in the bi-algal culture by using a mathematical model. The estimated parameters from the model showed that the inhibition exerted by S. trochoidea on P. donghaiense was about 43 and 24 times stronger than the inhibitory effect that P. donghaiense exerted on S. trochoidea when the initial size/density were 1:1 and 10:1, respectively. S. trochoidea seemed to have a survival strategy that was superior to P. donghaiense in the bi-algal culture under controlled laboratory conditions. We also observed a closely positive relationship between the initial cell density and its effect on the co-cultured microalga by measuring the fluorenscence: filtrate prepared from higher initial cell density had stronger interference on the co-cultured microalga. Moreover, pre-treated under different temperature conditions (30 °C, 60 °C and 100 °C) would significantly changed the effect of culture filtrate on the co-cultured microalga. Result inferred that P. donghaiense or S. trochoidea would release allelochemicals into the bi-algal culture medium and the allelochemicals might be a mixture with temperature-sensitive components in it.  相似文献

Employingin situ enclosures containing inocula of the lake zooplankton (mainlyDaphnia galeata, Daphnia cucullata andBosmina spp.) from a moderately eutrophic Lake Ros (Northern Poland) or large-bodiedDaphina magna, the following observations on succession of phytoplankton were made: 1) whereasD. magna could control the density of all the photoplankton size classes, the lake zooplankton could not suppress the large-sized phytoplankters or net phytoplankton; 2) the lake zooplankton was able to control the density of small algae (< 50μm), but its effect on large algae may be opposite: a promotion of net phytoplankton growth by removing small-sized algae which can out-compete net phytoplankton for limited PO4-P resources (<5μg P l−1). Since efficiency of phytoplankton density control byD. magna decreased with an increase in net phytoplankton abundance, biomanipulation could not be successful without introducing or maintaining a high population of large-bodied cladoceran species before high densities of large algae would make the control of phytoplankton inefficient.  相似文献

Porphyra is one of the world’s most valued maricultured seaweeds and has been cultivated for several hundred years in Asia. The objective of this study was to produce critical information as a guide for the selection of an appropriate Porphyra species from coastal New England for the development of a land-based aquaculture system. Four Northwest Atlantic Porphyra species: P. leucosticta, P. amplissima, P. linearis and P. umbilicalis, were cultivated for 1 and 2 weeks at saturated light intensities (100–150 μmol photons m−2s−1) and six combinations of ammonium (25 and 250 μmoles L−1) and temperature (10, 15 and 20°C). Specific growth rate (SGR) increased with decreasing temperature in P. leucosticta, P. linearis and P. umbilicalis and increased with increasing temperature in P. amplissima. The SGR of all species was greater at the higher ammonium concentration. Porphyra linearis had the highest SGR, increasing in biomass by approximately 16% day−1. Phycoerythrin (PE) content was higher at 10°C and 250 μmoles L−1 in all species except P. amplissima. The PE content, measured as fresh weight (FW), of P. linearis (29 mg g−1 FW−1) and P. umbilicalis (26 mg g−1 FW−1) was significantly higher than the other two species. Tissue nitrogen content of all species measured in dry weight was on average 1.45% higher at 250 μmoles L−1 than at 25 μmoles L−1 ammonium concentration. Porphyra umbilicalis had the highest tissue nitrogen contents (6.76%) at 10°C and 250 μmoles L−1 ammonium. Based on these results, P. linearis and P. umbilicalis should be considered as potential candidates for bioremediation with finfish and shellfish mariculture.  相似文献

Sweet sorghum juice supplemented with 0.5% ammonium sulphate was used as a substrate for ethanol production by Saccharomyces cerevisiae TISTR 5048. In batch fermentation, kinetic parameters for ethanol production depended on initial cell and sugar concentrations. The optimum initial cell and sugar concentrations in the batch fermentation were 1 × 108 cells ml−1 and 24 °Bx respectively. At these conditions, ethanol concentration produced (P), yield (Y ps) and productivity (Q p ) were 100 g l−1, 0.42 g g−1 and 1.67 g l−1 h−1 respectively. In fed-batch fermentation, the optimum substrate feeding strategy for ethanol production at the initial sugar concentration of 24 °Bx was one-time substrate feeding, where P, Y ps and Q p were 120 g l−1, 0.48 g g−1 and 1.11 g l−1 h−1 respectively. These findings suggest that fed-batch fermentation improves the efficiency of ethanol production in terms of ethanol concentration and product yield.  相似文献

We isolated and characterized seven microsatellite loci for the perennial herb Ixeridium dentatum ssp. dentatum, an apomictic triploid distributed throughout the lowland areas of East Asia. The number of alleles ranged from two to seven in 32 screened individuals of I. dentatum ssp. dentatum from Japan. The observed and expected heterozygosities were 0.000–0.950 and 0.000–0.891, respectively, calculated using genotypes of 20 individuals of I. dentatum ssp. nipponicum. One locus (msid4) deviated significantly from Hardy–Weinberg equilibrium (P = 0.0001). These microsatellites were also tested for cross-amplification in 11 other taxa of Lactuceae, including five endangered taxa. These primers should be useful genetic tools not only for Ixeridium but also for other Lactuceae taxa.  相似文献

Previously, a Saccharomyces cerevisiae strain was engineered for xylose assimilation by the constitutive overexpression of the Orpinomyces xylose isomerase, the S. cerevisiae xylulokinase, and the Pichia stipitis SUT1 sugar transporter genes. The recombinant strain exhibited growth on xylose, under aerobic conditions, with a specific growth rate of 0.025 h−1, while ethanol production from xylose was achieved anaerobically. In the present study, the developed recombinant yeast was adapted for enhanced growth on xylose by serial transfer in xylose-containing minimal medium under aerobic conditions. After repeated batch cultivations, a strain was isolated which grew with a specific growth rate of 0.133 h−1. The adapted strain could ferment 20 g l−1 of xylose to ethanol with a yield of 0.37 g g−1 and production rate of 0.026 g l−1 h−1. Raising the fermentation temperature from 30°C to 35°C resulted in a substantial increase in the ethanol yield (0.43 g g−1) and production rate (0.07 g l−1 h−1) as well as a significant reduction in the xylitol yield. By the addition of a sugar complexing agent, such as sodium tetraborate, significant improvement in ethanol production and reduction in xylitol accumulation was achieved. Furthermore, ethanol production from xylose and a mixture of glucose and xylose was also demonstrated in complex medium containing yeast extract, peptone, and borate with a considerably high yield of 0.48 g g−1.  相似文献

An effective, simple, and convenient method to improve yeast’s multiple-stress tolerance, and ethanol production was developed. After an ethanologenic Saccharomyces cerevisiae strain SC521 was treated by nine cycles of freeze-thaw, a mutant FT9-11 strain with higher multiple-stress tolerance was isolated, whose viabilities under acetic acid, ethanol, freeze-thaw, H2O2, and heat-shock stresses were, respectively, 23-, 26-, 10- and 7-fold more than the parent strain at an initial value 2 × 107 c.f.u. per ml. Ethanol production of FT9-11 was similar (91.5 g ethanol l−1) to SC521 at 30°C with 200 g glucose l−1, and was better than the parent strain at 37°C (72.5 g ethanol l−1), with 300 (111 g ethanol l−1) or with 400 (85 g ethanol l−1) g glucose l−1.  相似文献

In the framework of the EU Project STRATEGY, a short-term study was carried out in the Marinello ecosystem, a small brackish area located on the Tyrrhenian coast of Sicily (Italy). The investigation was aimed at understanding the dynamics of phytoplankton toxic blooms in relation to other planktonic species and environmental conditions. The study started on 10 March 2003, in coincidence with the first detection of Alexandrium minutum, a dinoflagellate known as a producer of Paralyzing Shellfish Toxins (PST) and lasted until 4 June 2003, when the bloom collapsed. The specific identity of A. minutum was confirmed on field mixed samples, through the use of species-specific PCR-primers targeting the 5.8S rDNA-ITS regions. Water samples and phytoplankton net hauls were taken approximately at 10 days intervals in the Verde Pond, one of the five basins of the Marinello ecosystem, in order to evaluate the incidence of toxic and non-toxic dinoflagellate species over the whole planktonic community. The evolution of the main environmental and trophic parameters (temperature, salinity, dissolved oxygen, POC, C/N, DIN, PO4–P) was simultaneously investigated. Alexandrium blooms were mostly characterized by A. minutum (max. 6 × 105 cells l−1 on April 11) and Alexandrium tamarense as an associated species (max. 2.5 × 104 cells l−1 on March 25). During the bloom, dinoflagellates or small flagellates dominated over the other taxa, with a minimum incidence of diatoms. The load of dissolved inorganic nitrogen was maximum in the pre-bloom phase (29 μM on March 19), after which it decreased sharply. An oxygen supersaturation event was registered in coincidence with the A. minutum bloom. The amounts of POC ranged between 266 and 658 μg l−1 showing a discontinuous temporal trend. A recent introduction of A. minutum into the Verde Pond is suggested on the basis of the absence of this species in past years.  相似文献

Spiny-surfaced species of Prorocentrum form harmful algal blooms, and its taxonomic identity is obscure due to the size and shape variability. Molecular phylogenies reveal two major clades: one for P. cordatum with sequences mainly retrieved as P. minimum, and the other for P. shikokuense with sequences also retrieved as P. dentatum and P. donghaiense. Several closely related clades still need to be characterized. Here, we provide nuclear SSU and LSU rRNA genes, and nuclear ITS region (ITS1-5.8S gene-ITS2) sequences of the strain CCMP3122 isolated from Florida (initially named P. donghaiense) and strains Prorocentrum sp. RCC6871–2 from the Ross Sea, Antarctica. We describe Prorocentrum thermophilum sp. nov. based on the strain CCMP3122, a species also distributed in the open waters of the Gulf of Mexico, New Zealand, and the Arabian Gulf; and Prorocentrum criophilum sp. nov. based on the strain RCC6872, which is distributed in the Antarctic Ocean and Arctic Sea. Prorocentrum thermophilum is roundish (~14 μm long, ~12 μm wide), with an inconspicuous anterior spine-like prolongation under light microscopy, valves with tiny, short knobs (5–7 per μm2), and several (<7) large trichocyst pores (~0.3 μm) in the right valve, as well as smaller pores (~0.15 μm). Prorocentrum criophilum is round in valve view (~11 μm long, 10 μm wide) and asymmetrically roundish in lateral view, the periflagellar area was not discernible under light microscopy, valves with very tiny, short knobs (6–10 per μm2), and at least 12 large pores in the right valve. Other potentially undescribed species of spiny-surfaced Prorocentrum are discussed.  相似文献
