Neurofilaments (NFs), composed of three distinct subunits NF-L, NF-M, and NF-H, are neuron-specific intermediate filaments present in most mature neurons. Using DNA transfection and mice expressing NF transgenes, we find that despite the ability of NF-L alone to assemble into short filaments in vitro NF-L cannot form filament arrays in vivo after expression either in cultured cells or in transgenic oligodendrocytes that otherwise do not contain a cytoplasmic intermediate filament (IF) array. Instead, NF-L aggregates into punctate or sheet like structures. Similar nonfilamentous structures are also formed when NF-M or NF-H is expressed alone. The competence of NF-L to assemble into filaments is fully restored by coexpression of NF- M or NF-H to a level approximately 10% of that of NF-L. Deletion of the head or tail domain of NF-M or substitution of the NF-H tail onto an NF- L subunit reveals that restoration of in vivo NF-L assembly competence requires an interaction provided by the NF-M or NF-H head domains. We conclude that, contrary to the expectation drawn from earlier in vitro assembly studies, NF-L is not sufficient to assemble an extended filament network in an in vivo context and that neurofilaments are obligate heteropolymers requiring NF-L and NF-M or NF-H.  相似文献

Mutualism is a fundamental building block of ecological communities and an important driver of biotic evolution. Classic theory suggests that a pairwise two‐species obligate mutualism is fragile, with a large perturbation potentially driving both mutualist populations into extinction. In nature, however, there are many cases of pairwise obligate mutualism. Such pairwise obligate mutualisms are occasionally associated with additional interactions with facultative mutualists. Here, we use a mathematical model to show that when a two‐species obligate mutualism has a single additional link to a third facultative mutualist, the obligate mutualism can become permanently persistent. In the model, a facultative mutualist interacts with one of two inter‐dependent obligate mutualists, and the facultative mutualist enhances the persistence not only of its directly interacting obligate mutualist, but also that of the other obligate mutualist indirectly, enabling the permanent coexistence of the three mutualist species. The effect of the facultative mutualist is strong; it can allow a three‐species permanent coexistence even when two obligate mutualists by themselves are not sustainable (i.e. not locally stable). These results suggest that facultative mutualists can play a pivotal role for the persistence of obligate mutualisms, and contribute to a better understanding on the mechanisms maintaining more complex mutualistic networks of multiple species.  相似文献

Obligate psychrophilic, facultative psychrophilic, and mesophilic yeasts were cultured in a carbon-rich medium at different temperatures to investigate whether growth parameters, lipid accumulation, and fatty acid (FA) composition were adaptive and/or acclimatory responses. Acclimation of facultative psychrophiles and mesophiles to a lower temperature decreased their specific growth rate, but did not affect their biomass yield (YX/S). Obligate and facultative psychrophiles exhibited the highest YX/S. Acclimation to lower temperature decreased the lipid yield (YL/X) in mesophilic yeasts, but did not affect YL/X in facultative psychrophilic ones. Similar YL/X were found in both groups of psychrophiles, suggesting that lipid accumulation is not a distinctive characteristic of adaptation to permanently cold environments. The unsaturation of FAs was one major adaptive feature of the yeasts colonizing permanently cold ecosystems. Remarkable amounts of α-linolenic acid were found in obligate psychrophiles at the expense of linoleic acid, whereas it was scarce or absent in all the other strains. Increased unsaturation of FAs was also a general acclimatory response of facultative psychrophiles to a lower temperature. These results improve the knowledge of the responses enabling psychrophilic yeasts to cope with the cold and may be of support for potential biotechnological exploitation of these strains.  相似文献

Abstract.  1. Host specificity is the degree to which a parasite species occurs in association with a host species.
2. The degree to which obligate ectoparasites are host specific has been debated, but effects of sampling contamination were usually not addressed. Data from a controlled mammal–ectoparasite survey were used to assess host specificity of an obligate group of ectoparasites – streblid bat flies.
3. Host–parasite associations were categorised as primary or non-primary. Non-primary host associations were evaluated against primary associations via proportional comparison.
4. Results indicate that host specificity was high, exceeding previous reports. Natural host transfers were rare.
5. Non-primary host associations were almost completely explained by disturbance transfers during sampling of the host or by contamination upon sampling the parasite. These conclusions likely hold for other taxa of obligate parasites.  相似文献

The small genomes of obligate intracellular bacteria are often presumed to be impervious to mobile DNA and the fluid genetic processes that drive diversification in free-living bacteria. Categorized by reductive evolution and streamlining, the genomes of some obligate intracellular bacteria manifest striking degrees of stability and gene synteny. Nitrosomonas europaea, Nitrobacter agilis, Thiobacillus denitrificans, T. neapolitanus, and T. thioparus (all obligate autotrophic bacteria) have been grown in dialysis culture, on glucose salts media, in the absence of their specific inorganic energy source. Metabolic products for N. agilis grown on nitrite salts medium were identified as keto acids. Pyruvic acid inhibited this organism at 5 x 10(-5)m. Keto acids were not inhibitory for the thiobacilli grown on thiosulfate medium. However, when T. denitrificans was grown on glucose with dialysis, addition of 5 x 10(-4)m pyruvate inhibited growth. Thus, it appears pyruvate may be inhibitory for other autotrophs, as has been reported for T. thiooxidans.  相似文献

Aim To investigate if the current theories associated with recruitment strategies are sufficient to explain differences in the proportions of obligate seeders and resprouters on inselbergs and in the matrix. Location The New England Batholith of eastern Australia. Methods The importance of life form and regenerative response of flora species are compared for twenty‐four archipelagos of inselbergs habitat islands and for their adjacent matrix. Forward stepwise multiple linear regression is used to correlate measured and modelled variables with differences in the proportional abundance of obligate seeders on inselbergs at three levels of sampling. Results A total of 216 inselbergs were surveyed and information on 826 vascular plant species was collated. Herbs were the most common life form both on and off inselbergs. There were generally more shrubs on inselbergs than in the matrix. Resprouters were more common everywhere. However, obligate seeders were proportionately more common on inselbergs. The degree of insularity of outcrop floras accounted for 23–37% of the variance in the proportional abundance of obligate seeders at three sampling scales. Main conclusions A model is proposed whereby the mechanisms that influence the abundances of obligate seeders and resprouters are different on inselbergs and in the matrix. Inselberg environments are harsh, arid, have limited safe recruitment sites and below ground resources, but are not light limited. In such habitats, obligate seeders with persistence life‐history traits are favoured. Within the matrix in eastern Australia frequent biomass removal disturbances occur (such as fire) and resprouting is advantageous. High turnover obligate seeders with no persistence life‐history traits can be favoured in the matrix with a reduced disturbance regime and where competition for light still occurs. Some of these matrix obligate seeders occur on inselbergs due to their infrequent disturbance regimes.  相似文献

在高度专性传粉的榕树-榕小蜂互惠共生系统中普遍存在着一些非传粉小蜂,它们中的一些种类进入果腔后也能为榕树传粉,且在形态和物候上已与传粉榕小蜂发生了趋同进化。但其寄主识别行为是否也与传粉榕小蜂发生了趋同进化还不得而知。我们在西双版纳选择了钝叶榕(Ficuscurtipes)及其3种进果繁殖小蜂开展了相关的行为实验。3种小蜂中,1种是钝叶榕的专性传粉榕小蜂(Eupristina sp.),另外2种是寄居性非传粉小蜂(杨氏金小蜂Diaziellayangi和Lipothymus sp.),这2种非传粉小蜂进入果腔后也像传粉榕小蜂那样为钝叶榕传粉。我们以钝叶榕不同发育时期的榕果及这3种小蜂为材料,采用Y型嗅觉仪观察了这3种小蜂对各发育时期榕果和信息化学物质6-甲基-5-庚烯-2-醇、6-甲基-5-庚烯-2-酮及这2种化合物的混合物的选择行为。结果表明,当提供雌花期榕果与其他发育时期榕果和空气对照供这3种小蜂选择时,它们均显著地偏向于选择雌花期榕果;当提供雄花期榕果与其他发育时期榕果和空气对照供这3种小蜂选择时,它们均显著地偏向于选择其他发育时期榕果和空气对照,即都会避开雄花期榕果;此外,这3种小蜂均对钝叶榕雌花期果释放的一种主要化合物6-甲基-5-庚烯-2-醇的同一剂量(1μL)表现出显著的偏好。这一结果为传粉榕小蜂与非传粉小蜂间的寄主识别行为趋同进化的假说提供了证据。  相似文献

In a screening program, a number of different bacterial strains with the ability to utilize methanol as a sole carbon and energy source were isolated and described. They are well known methanol utilizing genera Pseudomonas, Klebsiella, Micrococcus, Methylomonas or, on the contrary, the new, unknown genera and species of methylotrophic bacteria. In the last category, Acinetobacter and Alcaligenes are the new reported genera of organisms able to use methanol as a sole carbon and energy source. The present paper reports the very complex physiological and biochemical modifications when very versatile bacteria such as Pseudomonas aeruginosa and Acinetobacter calcoaceticus are cultured on methanol and when the obligate methylotrophic state is compared with the facultative methylotrophic state of the same bacterial strain. Based on experiments and comparisons with literature data, it seems that Methylomonas methanica is the obligate methylotrophic state of Pseudomonas aeruginosa and that Acinetobacter calcoaceticus is the facultative methylotrophic state of Methylococcus capsulatus, an obligate methylotroph. The relationship of the obligate to the facultative and of the facultative to the obligate methylotrophy were established. These new methylotrophic genera and species, the profound physiological and biochemical modifications as well as the new data concerning nature, nomenclature and taxonomy of methanol utilizing bateria were reported for the first time in 1983.  相似文献

The growth patterns of the hydroids Campanularia asymmetrica, Sertularia perpusilla and Algaophenia harpago on the Mediterranean seagrass Posidonia oceanica, and of Laomedea angulata on the temperate seagrass Zostera marina, were examined. The seagrass leaves grow from a basal meristem and have a short life which presents problems for these long-lived obligate epiphytes. Downward growth onto younger leaf tissue, shown by S. perpusilla and C. asymmetrica, reduces the risk of interspecific competition with other epiphytes, and places the hydroids on substrates that survive longer than older distal leaf tissue, close to short young leaves which they can colonize by stolon transfer. S. perpusilla was found only on the basal areas of leaves, possibly because of the progressive degeneration of their most apical tissue. C. asymmetrica showed no such degeneration and was found over most of the length of the leaves. The hydrorhizae of most small A. harpago and L. angulata grew both upward and downward along the leaves. Both species were most abundant on the apical halves of older leaves. This distal distribution is considered to be advantageous for these large branched hydroids which are suited for capturing planktonic prey, and to enhance their probability of colonizing neighbouring plants. Its disadvantages include the reduced probability of maintaining the hydroids on the same plant and an increased risk of being eaten by seagrass grazers.  相似文献

These mechanisms involve adaptations for survival in distinct intracellular compartments. We evaluated the possibility of storing the cultures of obligate anaerobic microorganisms (clostridia. acetogenic and sulfate-reducing bacteria, and methanogenic archaea) in 25% glycerol at -70 degrees C for a long time (up to 3 years). Obligate cooperative breeding species demonstrate a high rate of group extinction, which may be due to the existence of a critical number of helpers below which the group cannot subsist. Through a simple model, we study the population dynamics of obligate cooperative breeding species, taking into account the existence of a lower threshold below which the instantaneous growth rate becomes negative. The model successively incorporates (i) a distinction between species that need helpers for reproduction, survival or both, (ii) the existence of a migration rate accounting for dispersal, and (iii) stochastic mortality to simulate the effects of random catastrophic events. Our results suggest that the need for a minimum number of helpers increases the risk of extinction for obligate cooperative breeding species. The constraint imposed by this threshold is higher when helpers are needed for reproduction only or for both reproduction and survival. By driving them below this lower threshold, stochastic mortality of lower amplitude and/or lower frequency than for non-cooperative breeders may be sufficient to cause the extinction of obligate cooperative breeding groups. Migration may have a buffering effect only for groups where immigration is higher than emigration; otherwise (when immigrants from nearby groups are not available) it lowers the difference between actual group size and critical threshold, thereby constituting a higher constraint.  相似文献

Programmed cell death by apoptosis of unnecessary or potentially harmful cells is clearly beneficial to multicellular organisms. Proper functioning of such a program demands that the removal of dying cells proceed without an inflammatory reaction. Phosphatidylserine (PS) is one of the ligands displayed by apoptotic cells that participates in their noninflammatory removal when recognized by neighboring phagocytes. PS ligation induces the release of transforming growth factor-beta (TGF-beta), an antiinflammatory cytokine that mediates the suppression of macrophage-mediated inflammation. In Hydra vulgaris, an organism that stands at the base of metazoan evolution, the selective advantage provided by apoptosis lies in the fact that Hydra can survive recycling apoptotic cells by phagocytosis. In unicellular organisms, it has been proposed that altruistic death benefits clonal populations of yeasts and trypanosomatids. Now we show that advantageous features of the apoptotic process can operate without death as the necessary outcome. Leishmania spp are able to evade the killing activity of phagocytes and establish themselves as obligate intracellular parasites. Amastigotes, responsible for disease propagation, similar to apoptotic cells, inhibit macrophage activity by exposing PS. Exposed PS participates in amastigote internalization. Recognition of this moiety by macrophages induces TGF-beta secretion and IL-10 synthesis, inhibits NO production, and increases susceptibility to intracellular leishmanial growth.  相似文献

Peptidoglycan is the predominant stress‐bearing structure in the cell envelope of most bacteria, and also a potent stimulator of the eukaryotic immune system. Obligate intracellular bacteria replicate exclusively within the interior of living cells, an osmotically protected niche. Under these conditions peptidoglycan is not necessarily needed to maintain the integrity of the bacterial cell. Moreover, the presence of peptidoglycan puts bacteria at risk of detection and destruction by host peptidoglycan recognition factors and downstream effectors. This has resulted in a selective pressure and opportunity to reduce the levels of peptidoglycan. In this review we have analysed the occurrence of genes involved in peptidoglycan metabolism across the major obligate intracellular bacterial species. From this comparative analysis, we have identified a group of predicted ‘peptidoglycan‐intermediate’ organisms that includes the Chlamydiae, Orientia tsutsugamushi, Wolbachia and Anaplasma marginale. This grouping is likely to reflect biological differences in their infection cycle compared with peptidoglycan‐negative obligate intracellular bacteria such as Ehrlichia and Anaplasma phagocytophilum, as well as obligate intracellular bacteria with classical peptidoglycan such as Coxiella, Buchnera and members of the Rickettsia genus. The signature gene set of the peptidoglycan‐intermediate group reveals insights into minimal enzymatic requirements for building a peptidoglycan‐like sacculus and/or division septum.  相似文献

In Pseudomonas C, an obligate RMP-methylotroph, externally added formic acid and multi-carbon intermediates are co-metabolized in the obligate presence of methanol. Cell yield and methanol-carbon conversion to cell mass are optimal in a chemostat culture growing on a mixture of energy sources and methanol as the sole limiting carbon source. Growth on methanol as the sole, obligate energy source and an additional carbon source (e.g. pyruvate) is less efficient. Under these energy limited conditions, the cyclic oxidation pathway is predominantly operated.  相似文献
