Summary The influence of culture pH on the metabolism of Gluconobacter oxydans was determined. An acidic milieu during growth of the organism enhances the oxidation rate. The CO2 evolution rate representing the assimilation of the product is inhibited by a low pH value. Growth of the bacteria is possible both on glycerol and DHA in separate phases, which is not a controlled as diauxic growth. Summary The inhibitory effects of DHA on Gluconobacter oxydons were measured to forrmulate a fermentation model. Growth of Gluconobacter oxydons is inhibited by DHA which can be modelled by a linear Term. The inhibition of product formation by DHA was measured and described by a classical feedback inhibition kinetic. Additionally, an irreversible destruction of Gluconobacter cells by DHA was discovered. This toxic effect of DHA could be modelled by a death rate kinetic and introduction of a damaged cell type. DHA also inhibits the activity of the pentose cycle as can be measured via the CO2 evolution rate.  相似文献

Abstract: Transport of ascorbate (AA) and dehydroascorbate (DHA) through the petiole into detached leaves of Lepidium sativum and other plant species via the transpiration stream, and energized uptake into leaf tissue, were measured indirectly by recording changes in membrane potential and apoplastic pH simultaneously with substrate‐stimulated respiration and transpiratory water loss. When 25 mM AA or DHA was fed to the leaves, steady state respiration at 25 °C was transiently increased by more than 50 % with AA and 70 % with DHA. Stimulation of respiration was accompanied by a transient breakdown of membrane potential followed by alkalinization of the leaf apoplast suggesting energized uptake at the expense of the transmembrane proton motive force. The average CO2/AA ratio calculated from stimulated respiration during ascorbate uptake was 0.76 ± 0.26 (n = 17). The corresponding ratio for DHA was 1.38 ± 0.28 (n = 11). Far lower CO2/substrate ratios were observed when NaCl or KCl were fed to leaves. The differences indicate either partial metabolism of AA and DHA in addition to energized transport, or less likely, higher energy requirement for transport of AA and DHA than for the inorganic salts. Maximum rates of energized AA transport into leaf tissue (deduced from maxima of extra respiration and calculated on the basis of CO2/AA = 0.76) were close to 650 nmol m‐2 leaf area s‐1, i.e. far higher than most previously reported rates of transport. When the apoplastic concentration of AA was decreased below steady state levels during infiltration/centrifugation experiments, AA was released from leaf cells into the apoplast. This suggests that AA oxidation to DHA in the apoplast (as occurs during extracellular ozone detoxification) triggers energized transport of the DHA into the symplast and simultaneously AA release from the symplast into the apoplast, perhaps together with protons in a reversal of the energized uptake process.  相似文献

Microalgae biomass can be a feasible source of ω‐3 fatty acids due to its stable and reliable composition. In the present study, the Crypthecodinium cohnii growth and docosahexaenoic acid (DHA, 22:6ω3) production in a 100 L glucose‐fed batch fermentation was evaluated. The lipid compounds were extracted by supercritical carbon dioxide (SC‐CO2) from C. cohnii CCMP 316 biomas, was and their fatty acid composition was analysed. Supercritical fluid extraction runs were performed at temperatures of 313 and 323 K and pressures of 20.0, 25.0 and 30.0 MPa. The optimum extraction conditions were found to be 30.0 MPa and 323 K. Under those conditions, almost 50% of the total oil contained in the raw material was extracted after 3 h and the DHA composition attained 72% w/w of total fatty acids. The high DHA percentage of total fatty acids obtained by SC‐CO2 suggested that this extraction method may be suitable for the production of C. cohnii value added products directed towards pharmaceutical purposes. Furthermore, the fatty acid composition of the remaining lipid fraction from the residual biomass with lower content in polyunsaturated fatty acids could be adequate for further uses as feedstock for biodiesel, contributing to the economy of the overall process suggesting an integrated biorefinery approach.  相似文献

Pseudomonas fluorescens strains are used in agriculture as plant growth-promoting rhizobacteria (PGPR). Nontarget effects of released organisms should be analyzed prior to their large-scale use, and methods should be available to sensitively detect possible changes in the environments the organism is released to. According to ecological theory, microbial communities with a greater diversity should be less susceptible to disturbance by invading organisms. Based on this principle, we laid out a pot experiment with field-derived soils different in their microbial biomass and activity due to long-term management on similar parent geological material (loess). We investigated the survival of P. fluorescens CHA0 that carried a resistance toward rifampicine and the duration of potential changes of the soil microflora caused by the inoculation with the bacterium at the sowing date of spring wheat. Soil microbial biomass (C mic, N mic) basal soil respiration (BR), qCO2, dehydrogenase activity (DHA), bacterial plate counts, mycorrhiza root colonization, and community level substrate utilization were analyzed after 18 and 60 days. At the initial stage, soils were clearly different with respect to most of the parameters measured, and a time-dependent effect between the first and the second set point were attributable to wheat growth and the influence of roots. The effect of the inoculum was small and merely transient, though significant long-term changes were found in soils with a relatively low level of microbial biomass. Community level substrate utilization as an indicator of changes in microbial community structure was mainly changed by the growth of wheat, while other experimental factors were negligible. The sensitivity of the applied methods to distinguish the experimental soils was in decreasing order N mic, DHA, C mic, and qCO2. Besides the selective enumeration of P. fluorescens CHA0 rif+, which was only found in amended soils, methods to distinguish the inoculum effect were DHA, C mic, and the ratio of C mic to N mic. The sampling time was most sensitively indicated by N mic, DHA, C mic, and qCO2. Our data support the hypothesis—based on ecosystem theory—that a rich microflora is buffering changes due to invading species. In other words, a soil-derived bacterium was more effective in a relatively poor soil than in soils that are rich in microorganisms.  相似文献

Marine pCO2 enrichment via ocean acidification (OA), upwelling and release from carbon capture and storage (CCS) facilities is projected to have devastating impacts on marine biomineralisers and the services they provide. However, empirical studies using stable endpoint pCO2 concentrations find species exhibit variable biological and geochemical responses rather than the expected negative patterns. In addition, the carbonate chemistry of many marine systems is now being observed to be more variable than previously thought. To underpin more robust projections of future OA impacts on marine biomineralisers and their role in ecosystem service provision, we investigate coralline algal responses to realistically variable scenarios of marine pCO2 enrichment. Coralline algae are important in ecosystem function; providing habitats and nursery areas, hosting high biodiversity, stabilizing reef structures and contributing to the carbon cycle. Red coralline marine algae were exposed for 80 days to one of three pH treatments: (i) current pH (control); (ii) low pH (7.7) representing OA change; and (iii) an abrupt drop to low pH (7.7) representing the higher rates of pH change observed at natural vent systems, in areas of upwelling and during CCS releases. We demonstrate that red coralline algae respond differently to the rate and the magnitude of pH change induced by pCO2 enrichment. At low pH, coralline algae survived by increasing their calcification rates. However, when the change to low pH occurred at a fast rate we detected, using Raman spectroscopy, weaknesses in the calcite skeleton, with evidence of dissolution and molecular positional disorder. This suggests that, while coralline algae will continue to calcify, they may be structurally weakened, putting at risk the ecosystem services they provide. Notwithstanding evolutionary adaptation, the ability of coralline algae to cope with OA may thus be determined primarily by the rate, rather than magnitude, at which pCO2 enrichment occurs.  相似文献

Naturally grown trees of Mediterranean evergreen oak (Quercus ilex L.), representing the climax species of the region, were enclosed in six large open-top chambers and exposed to ambient and elevated CO2 concentrations during a 3 year period. Maximum daily net photosynthetic rates measured at the two different CO2 concentrations were from 30 to 100% higher in elevated than in ambient [CO2] throughout the experimental period. The increase in maximum daily photosynthesis was also accompanied by a 93% rise in the apparent quantum yield of CO2 assimilation, measured during periods of optimum soil moisture conditions. Hence, no clear evidence of down-regulation of net photosynthetic activity was found. Interactions between atmospheric CO2 concentration and plant water stress were studied by following the natural evolution of drought in different seasons and years. At each level of water stress, the maximum rate of carbon assimilation was higher in elevated than in ambient [CO2] by up to 100%. Analysis of in vivo chlorophyll fluorescence parameters in normal (21%) and low (2%) oxygen concentrations provided useful insights into the functioning and stability of the photosynthetic processes. The photochemical efficiency of PSII (Fv/Fm) progressively decreased as drought conditions became more evident; this trend was accentuated under elevated [CO2]. Thermal de-excitation processes were possibly more significant under elevated than under ambient [CO2], in a combination of environmental stresses. This research suggests two possible conclusions: (i) a ‘positive’ interaction between elevated [CO2] and carbon metabolism can be obtained through relief of water stress limitation in the summer months, and (ii) elevated [CO2], under drought conditions, may also enhance the significance of slow-relaxing quenching.  相似文献

Respiration rates predict differences in growth of coast redwood   总被引:1,自引:0,他引:1  
The relation between growth rate traits (height, basal diameter, stem volume and branch diameter) and two measures of respiration rate [metabolic heat rate (q) and CO2 production rate (Rco2)] and their ratio (q/Rco2) was examined on a collection of 192 different genotypes of coast redwoods [Sequoia sempervirens (D.Don) Endl.]. Branch diameter was not correlated with any of the respiratory measures, but the other three growth traits gave highly significant (P < 0.001) correlations with positive slopes. Combining the four growth traits and the three respiratory variables (q, RCo2 and q/Rco2) to give two canonical variates, one representing growth and one representing respiration, gives an even stronger linear correlation (r= 0–85). These data suggest that simultaneous assay of multiple respiratory measures on juvenile trees can be used to predict their longer-term growth rates.  相似文献

Modeling analyses suggest that an increase in growth rate of atmospheric CO2 concentrations during an anomalously warm year may be caused by a decrease in net ecosystem production (NEP) in response to increased heterotrophic respiration (Rh). To test this hypothesis, 12 intact soil monoliths were excavated from a tallgrass prairie site near Purcell, Oklahoma, USA and divided among four large dynamic flux chambers (Ecologically Controlled Enclosed Lysimeter Laboratories (EcoCELLs)). During the first year, all four EcoCELLs were subjected to Oklahoma air temperatures. During the second year, air temperature in two EcoCELLs was increased by 4°C throughout the year to simulate anomalously warm conditions. This paper reports on the effect of warming on soil CO2 efflux, representing the sum of autotrophic respiration (Ra) and Rh. During the pretreatment year, weekly average soil CO2 efflux was similar in all EcoCELLs. During the late spring, summer and early fall of the treatment year, however, soil CO2 efflux was significantly lower in the warmed EcoCELLs. In general, soil CO2 efflux was correlated with soil temperature and to a lesser extent with moisture. A combined temperature and moisture regression explained 64% of the observed variation in soil CO2 efflux. Soil CO2 efflux correlated well with a net primary production (NPP) weighted greenness index derived from digital photographs. Although separate relationships for control and warmed EcoCELLs showed better correlations, one single relationship explained close to 70% of the variation in soil CO2 efflux across treatments and years. A strong correlation between soil CO2 efflux and canopy development and the lack of initial response to warming indicate that soil CO2 efflux is dominated by Ra. This study showed that a decrease in soil CO2 efflux in response to a warm year was most likely dominated by a decrease in Ra instead of an increase in Rh.  相似文献

The potential use of n-dodecane as an oxygen vector for enhancement of Crypthecodinium cohnii growth and docosahexaenoic acid (DHA) production was studied. The volumetric fraction of oxygen vector influenced the gas–liquid volumetric mass transfer coefficient k L a positively. The k L a increased almost linearly with the increase of volumetric fraction of n-dodecane up to 1%. The stirring rate showed a higher influence on the k L a than the aeration rate. The effects of this hydrocarbon on C. cohnii growth and DHA production were then investigated. A control batch fermentation without n-dodecane addition (CF) and a batch fermentation where n-dodecane 1% (v/v) was added (DF) were carried out simultaneously under the same experimental conditions. It was found that, before 86.7 h of fermentation, the biomass concentration, the specific growth rate, the DHA, and total fatty acids (TFA) production were higher in the CF. After this fermentation time, the biomass concentration, the DHA and TFA production were higher in the DF. The highest DHA content of biomass (6.14%), DHA percentage of TFA (51%), and DHA production volumetric rate r DHA (9.75 mg l−1 h−1) were obtained at the end of the fermentation with n-dodecane (135.2 h). The dissolved oxygen tension (DOT) was always higher in the DF, indicating a better oxygen transfer due to the oxygen vector presence. However, since the other C. cohnii unsaturated fatty acids percentages did not increase with the oxygen availability increase due to the n-dodecane presence, a desaturase oxygen-dependent mechanism involved in the C. cohnii DHA biosynthesis was not considered to explain the DHA production increase. A selective extraction through the n-dodecane was suggested.  相似文献

The regulation of photosystem II (PSII) by light-, CO2-, and O2-dependent changes in the capacity for carbon metabolism was studied. Estimates of the rate of electron transport through PSII were made from gas-exchange data and from measurements of chlorophyll fluorescence. At subsaturating photon-flux density (PFD), the rate of electron transport was independent of O2 and CO2. Feedback on electron transport was observed under two conditions. At saturating PFD and low partial pressure of CO2, p(CO2), the rate of electron transport increased with p(CO2). However, at high p(CO2), switching from normal to low p(O2) did not affect the net rate of photosynthetic CO2 assimilation but the rate of electron-transport decreased by an amount related to the change in the rate of photorespiration. We interpret these effects as 1) regulation of ribulose-1,5-bisphosphatecarboxylase (RuBPCase, EC 4.1.1.39) activity to match the rate of electron transport at limiting PFD, 2) regulation of electron-transport rate to match the rate of RuBPCase at low p(CO2), and 3) regulation of the electron-transport rate to match the capacity for starch and sucrose synthesis at high p(CO2) and PFD. These studies provide evidence that PSII is regulated so that the capacity for electron transport is matched to the capacity for other processes required by photosynthesis, such as ribulose-bisphosphate carboxylation and starch and sucrose synthesis. We show that at least two mechanisms contribute to the regulation of PSII activity and that the relative engagement of these mechanisms varies with time following a step change in the capacity for ribulose-bisphosphate carboxylation and starch and sucrose synthesis. Finally, we take advantage of the relatively slow activation of deactivated RuBPCase in vivo to show that the activation level of this enzyme can limit the rate of electron transport as evidenced by increased feedback on PSII following a step change in p(CO2). Abstract

The objective of this study was to investigate the effect of increasing CO2 concentration on the growth and the capability of Tetraselmis chui. in removal of nitrate, ammonium and phosphate from shrimp pond wastewater (SPWW). The factorial experimental design was used with the treatment of SPWW percentage in culture medium, namely: 100% SPWW, 75% SPWW + 25% Sea Water (SW) and 75% SW + 25% SPWW coupled with three CO2 concentration treatments: 390?ppm, 550?ppm and 1000?ppm using CO2 system. Growth of T. chui. for lengh of cultivation period tended to be higher at treatments of 390?ppm CO2 and 100% SPWW, however there was a declining growth over period of cultivation for both treatments. The growth rate of T. chui was higher for all percentage of SPWW treatments in culture medium at 390?ppm CO2 concentration compared to other percentage of SPWW treatments and CO2 concentration treatments. There was a decreasing of growth rate with increasing CO2 concentration at 100% SPWW and 75% SPWW + 25% SW in culture medium. Nitrogen removal efficiency and removal rate by T. chui. were strongly affected by CO2 concentration. However, there was no significant effect of increasing CO2 concentration to removal efficiency and rate of PO4 by T. chui.  相似文献

Membrane inlet mass spectrometry indicated massive light-dependent cycling of inorganic carbon between the medium and the cells of various phytoplankton species representing the main groups of aquatic primary producers. These included diatoms, symbiotic and free living dinoflagellates, a coccolithophorid, a green alga and filamentous and single cell cyanobacteria. These organisms could maintain an ambient CO2 concentration substantially above or below that expected at chemical equilibrium with HCO3 . The coccolithophorid Emiliania huxleyishifted from net CO2 uptake to net CO2 efflux with rising light intensity. Differing responses of CO2 uptake and CO2 fixation to changing light intensity supported the notion that these two processes are not compulsorily linked. Simultaneous measurements of CO2 and O2 exchange and of the fluorescence parameters in Synechococcus sp. strain PCC 7942, showed that CO2 uptake can serve as a sensitive probe of the energy status of the photosynthetic reaction centers. However, during transitions in light intensity, changes in CO2 uptake did not accord with those expected from fluorescence change. Quantification of the net fluxes of CO2, HCO3 and of photosynthesis at steady-state revealed that substantial HCO3 efflux accompanied CO2 uptake and fixation in the case of `CO2 users'. On the other hand, `HCO3 users' were characterized by a rate of net CO2 uptake below that of CO2 fixation. The results support the notion that entities associated with the CCM function not only in raising the CO2 concentration at the site of Rubisco; they may also serve as a means of diminishing photodynamic damage by dissipating excess light energy. Algal cultivation is a potential candidate for CO2 mitigation. CO2 plays important roles in mass cultivation of algae, including supplying carbon source and adjusting medium pH. To assess the possibility of using edible cyanobacterium Nostoc flagelliforme as carbon storage device, the growth characteristics of N. flagelliforme batch cultured under elevated CO2 concentrations (0, 2.5, 5, 20, and 40%) were investigated in this study. Results showed that the net photosynthetic rate, efficiency and carbon sequestration rate at 20% CO2 were increased at a maximum of 121 μmol O2 (mg chla)?1 h?1 8.40% and 0.17 g CO2 L?1 day?1, and increased by 0.42, 1.03 and 1.13 folds compared with that of the control, respectively. Higher CO2 concentration resulted in the declines in photosynthetic rate, efficiency and carbon sequestration rate because of medium pH reduction. Accordingly, the dry cell weight, amount of exopolysaccharides and protein content of N. flagelliforme cells at 20% CO2 were obtained at a maximum of 1.45 g L?1, 54.98 mg L?1 and 57.75%, increased by 0.93, 0.29 and 0.8 folds compared with that of the control, respectively. These results provided important information for CO2 mitigation by N. flagelliforme and would shed more light on elucidating the mechanisms of CO2 tolerance in cyanobacterium.  相似文献

Summary Eucalyptus camaldulensis shoots were cultured photoautotrophicallyin vitro for 6 wk with four different types of supporting materials (agar matrix, Gelrite matrix, plastic net, or vermiculite) under CO2-nonenriched or CO2-enriched conditions. Plantlets from each treatmentin vitro were then grownex vitro in a greenhouse for 4 wk. The growth and net photosynthetic rate of plantletsin vitro, as well as subsequent growth, survival percentage, transpiration rate, and net photosynthetic rate of plantletsex vitro were evaluated. CO2 enrichment significantly increased growth (total dry weight and number of primary roots) and net photosynthetic rate of plantletsin vitro, as well as the growth and survival percentage of plantletsex vitro regardless of the type of supporting materials. The growthin vitro was greatest in the vermiculite, followed by the plastic net, Gelrite matrix, and agar matrix (in descending order) under either the CO2-nonenriched or CO2-enriched conditions. The growth and survival percentage of plantletsex vitro were highest in the vermicultie under the CO2-enriched condition. The extensive root system producedin vitro was necessary for growth and survival of plantletsex vitro.  相似文献

The freshwater microalga, Chlamydomonas reinhardtii Dangeard, was cultured under 350 and 700 ppmv CO2 to determine the impact of doubled atmospheric CO2 concentration on its growth and photosynthesis. No significant difference was observed in the specific growth rate, photosynthetic efficiency, maximal net photo‐synthetic rate and light‐saturating point between the low and high CO2 cultures. Both the low‐ and high‐CO2‐grown cells showed reduced light‐dependent O2 evolution rate and photochemical efficiency (Fv/Fm) owing to photoinhibition when exposed to high photon flux density. However, high‐CO2‐grown cells were less photoinhibited, and showed better recovery in dim light or darkness during the initial period of the recovery process.  相似文献

Saplings of four clones of Sitka spruce and cherry were grown for three and two growing seasons, respectively, in open top chambers at two CO2 concentrations (≈ 350 and ≈ 700 μmol mol–1) to determine whether the increase in total biomass brought about by enhanced [CO2] is a result of a transient or persistent effect in nonlimiting conditions. Classical growth analysis was applied to both species and mean current relative growth rate of total dry mass (RT) and leaf dry mass (RL), and period relative growth rate of total dry mass ( ) and leaf dry mass ( ) were calculated. Sitka spruce saplings and cherry seedlings showed a positive growth response to elevated [CO2], and at the end of the experiments both species were ≈ 40% larger in elevated [CO2] than in ambient [CO2]. As a result, the period mean and were significantly higher in elevated [CO2]. The differences in plant dry mass at the end of the experiments were a consequence of the more rapid growth in the early phase of exposure to elevated [CO2]. After this initial phase mean RT and RL were similar or even lower in elevated [CO2] than in ambient [CO2]. NAR of both species was much higher in elevated [CO2], whereas both LAR, SLA, and LMR showed the opposite trend. The higher LAR and SLA of plants in ambient [CO2] contributed to a compensation by which they maintained RT similar to that of elevated [CO2] saplings despite lower NAR and photosynthetic rate. However, when the same size the trees were similar amongst the [CO2] treatments, indicating that one of the main effect of elevated [CO2] on tree growth is to speed-up early development in all aspects.  相似文献

The role of carbon dioxide in glucose metabolism of Bacteroides fragilis   总被引:2,自引:0,他引:2  
The effect of CO2 concentration on growth and glucose fermentation of Bacteroides fragilis was studied in a defined mineral medium. Batch culture experiments were done in closed tubes containing CO2 concentrations ranging from 10% to 100% (with appropriate amounts of bicarbonate added to maintain the pH at 6.7). These experiments revealed that CO2 had no influence on growth rate or cell yield when the CO2 concentration was above 30% CO2 (minimum available CO2–HCO 3 - , 25.5 mM), whereas a slight decrease in these parameters was observed at 20% and 10% CO2 (available CO2–HCO 3 - , 17 and 8.5 mM, respectively). If CO2–HCO 3 - concentrations were below 10 mM, the lag phase lengthened and a decrease in maximal growth rate and cell yield were observed. The amount of acetate made decreased, while d-lactate concentration increased. A net production of CO2 allowed growth under conditions of extremely low concentrations of added CO2.When B. fragilis was grown in continuous culture with 100% CO2 or 100% N2, the dilution rate influenced the concentrations of acetate, succinate, propionate, d-lactate, l-malate and formate formed. Decreasing the dilution rate favored propionate and acetate production under both conditions. When the organism was grown with 100% N2, the amount of propionate formed was greater than the amount of succinate formed at all dilution rates. Except at slow dilution rates the reverse was true when 100% CO2 was used. B. fragilis was unable to grow at dilution rates faster than 0.154 h-1 when grown with 100% N2; the Y glc max was 67.9 g DW cells/mol glucose and m s was 0.064 mmol glucose/g DW·h. If the gas atmosphere was 100% CO2 the organism was washed out of the culture when the dilution rate exceeded 0.38 h-1; the Y glc max was 59.4 g DW cells/mol glucose and m s was 0.094 mmol glucose/g DW·h.Measurement of the phosphoenolpyruvate (PEP) carboxykinase (E.C. 4.1.1.49) with whole, permeabilized cells of B. fragilis showed an increase of specific enzyme activity with decreasing CO2 concentrations. The mechanisms used by B. fragilis to adjust to low levels of CO2 are discussed.  相似文献

Genetic variation in plant response to atmospheric carbon dioxide (CO2) may have influenced paleo‐vegetation dynamics and could determine how future elevated CO2 drives plant evolution and ecosystem productivity. We established how levels of relatedness – the maternal family, population, and provenance – affect variation in the CO2 response of a species. This 2‐year growth chamber experiment focused on the germination, growth, biomass allocation, and survivorship responses of Acer rubrum to four concentrations of CO2: 180, 270, 360, and 600 μL L?1– representing Pleistocene through potential future conditions. We found that all levels of relatedness interacted with CO2 to contribute to variation in response. Germination responses to CO2 varied among families and populations, growth responses depended on families and regions of origin, and survivorship responses to CO2 were particularly affected by regional identities. Differences among geographic regions accounted for 23% of the variation in biomass response to CO2. If seeds produced under subambient CO2 conditions behave similarly, our results suggest that A. rubrum may have experienced strong selection on seedling survivorship at Pleistocene CO2 levels. Further, this species may evolve in response to globally rising CO2 so as to increase productivity above that experimentally observed today. Species responses to future atmospheric CO2 and the accompanying biotic effects on the global carbon cycle will vary among families, populations, and provenances.  相似文献
