Summary Vasopressin-induced transformation of ridges to microvilli on the surface of granular cells of toad urinary bladder occurs in conjunction with induced alterations in the water permeability of the luminal membrane. This study was designed to establish the relationship between the time course for induction of microvilli and the time course for induction of increased water permeability after vasopressin stimulation. Hemibladders were examined at 2.5, 5, 10, 20 and 30 min following exposure to 20 mU/ml of vasopressin and at 5, 10, 20, 30, 40, 50 and 60 min after washout of vasopressin. Within 2.5 min, vasopressin initiated complete transformation of ridges to microvilli on approximately 13% of the granular cells, while osmotic water flow (Jv) was 0.31±0.10 l·min–1·cm–2. Five minutes following vasopressin stimulation, microvilli were present on approximately 30% of granular cells andJv was 2.27±0.13 l·min–1·cm–2. At 10 minJv was maximum at 4.03±0.15 l·min–1·cm–2 and 50% of the granular cells were covered with microvilli. This percentage increased to 70% at 20 min and was maintained at 30 min, althoughJv decreased to 3.9±0.35 l·min–1·cm–2 at 30 min. Five minutes following vasopressin washout, ridges interspersed with microvilli reappeared asJv fell to 1.10±0.30 l·min–1·cm–2. At 10 min after vasopressin washout,Jv approached basal levels, but the reversal of microvilli to ridges remained incomplete. At 60 min after vasopressin washout, the granular cells had regained their original ridgelike surface structures. Thus, these studies establish a temporal relationship between the induction and reversibility of vasopressin-induced microvillous formation and alterations in the osmotic water permeability of the apical plasmalemma.  相似文献

Summary Carausius morosus starved and deprived of water lose about 30% of their body weight in 4 days, mainly due to water loss. Isolated inferior tubules from starved dehydrated insects secrete urine at 0.041 nl·mm–1·min–1 compared with 0.118 nl·mm–1·min–1 in those from fed hydrated insects. This difference is due partly to the level of a diuretic (and perhaps also an antidiuretic) hormone in the haemolymph acting directly on the urine-secreting mechanism and partly to changes in the intrinsic capacity of the tubule cells for urine secretion. This latter change is accompanied by structural changes in the tubules. During starvation and dehydration the lumen becomes packed with white granules, the height of the type 1 cells is reduced, their basal infoldings and brush border become shorter and their mitochondrial volume is reduced.  相似文献

Summary Cl transport in apical membrane vesicles derived from bovine tracheal epithelial cells was studied using the Cl-sensitive fluorescent indicator 6-methoxy-N-(3-sulfopropyl) quinolinium. With an inwardly directed 50 mM Cl gradient at 23°C, the initial rate of Cl entry (J Cl) was increased significantly from 0.32±0.12 nmol · sec–1 · mg protein–1 (mean±sem) to 0.50±0.07 nmol · sec–1 · mg protein–1 when membrane potential was changed from 0 to +60 mV with K/valinomycin. At 37°C, with membrane potential clamped at 0 mV, there was a 34±7% (n=5) decrease inJ Cl from a control value of 0.37±0.03 nmol · sec–1 · mg protein–1 upon addition of 0.2mm diphenylamine-2-carboxylate. The following did not alterJ Cl significantly (J Cl values gives as percent change from control): 50mm cis Na (–1±5%), 0.1mm furosemide (–3±4%), 0.1mm furosemide in the presence of 50mm cis Na (–5±2%), 0.1mm H2DIDS (–18±9%), a 1.5 pH unit inwardly directed H gradient (–7±7%), and 0.1mm H2DIDS in the presence of a 1.5 unit pH gradient (4±18%). With inward 50mm anion gradients, the initial rates of Br and I entry (J Br andJ 1, respectively) were not significantly different fromJ Cl.J Cl was a saturable function of Cl concentration with apparentK d of 24mm and apparentV max of 0.54 nmol · sec–1 · mg protein–1. Measurement of the temperature dependence ofJ Cl yielded an activation energy of 5.0 kcal/mol (16–37°C). These results demonstrate that Cl transport in tracheal apical membrane vesicles is voltage-dependent and inhibited by diphenylamine-2-carboxylate. There is no significant contribution from the Na/K/2Cl, Na/Cl, or Cl/OH(H) transporters. The conductive pathway does not discriminate between Cl, Br, and I and is saturable. The low activation energy supports a pore-type mechanism for the conductance.  相似文献

Summary We examined the direct effects of isoproterenol (ISO) andl-norepinephrine (NE) on electrolyte transport in isolated rabbit cortical collecting tubules (CCT) perfusedin vitro. The addition of either ISO (10–6 m) or NE (10–6 m) to the bath decreased transepithelial potential difference (PD), on average by 51 and 25%, respectively. These effects of ISO and NE were abolished by prior addition of the -adrenergic blocker,l-propranolol. ISO (10–5 m) had no effect from lumen. Also, osmotic water permeability was not influenced by ISO. Ouabain and ISO had additive effects on PD. Elimination of chloride from both perfusate and bath, or addition of acetazolamide, abolished the effect of ISO on PD. Although isotopic sodium flux from lumen to bath was not influenced by ISO, chemical net chloride absorption increased from 1.1±0.4 to 2.7±0.6 peq·cm–1·sec–1 (n=8,p<0.005). In conclusion, both ISO and NE are capable of decreasing PD in rabbit CCT perfusedin vitro. This effect is mediated by -adrenergic receptors and is accompanied by the increase in net chloride absorption. Although the mechanism responsible for this decrease in PD with ISO is unclear, active chloride absorption, active hydrogen secretion, or membrane chloride permeability changes may account for the effects of ISO.  相似文献

Summary Na+–H+ exchange activity in renal brush border membrane vesicles isolated from hyperthyroid rats was increased. When examined as a function of [Na+], treatment altered the initial rate of Na+ uptake by increasingV m (hyperthyroid, 18.9±1.1 nmol Na+ · mg–1 · 2 sec–1; normal, 8.9±0.3 nmol Na+ · mg–1 · 2 sec–1), and not the apparent affinityK Na + (hyperthyroid, 7.3±1.7mm; normal, 6.5±0.9mm). When examined as a function of [H+] and at a subsaturating [Na+] (1mm), hyperthyroidism resulted in the proportional increase in Na+ uptake at every intravesicular pH measured. A positive cooperative effect on Na+ uptake was found with increased intravesicular acidity in vesicles from both normal and hyperthyroid rats. When the data were analyzed by the Hill equation, it was found that hyperthyroidism did not change then (hyperthyroid, 1.2±0.06; normal, 1.2±0.07) or the [H+]0.5 (hyperthyroid, 0.39±0.08 m; normal, 0.44±0.07 m) but increased the apparentV m (hyperthyroid, 1.68±0.14 nmol Na+ · mg–1 · 2 sec–1; normal 0.96±0.10 nmol Na+ · mg–1 · 2 sec–1). The uptake of Na+ in exchange for H+ in membrane vesicles from normal and hyperthyroid animals was not influenced by membrane potential. H+ translocation or debinding was rate limiting for Na+–H+ exchange since Na+–Na+ exchange activity was greater than Na+–H+ exchange activity. Hyperthyroidism caused a proportional increase and hypothyroidism caused a proportional decrease in Na+–Na+ and Na+–H+ exchange. We conclude that hyperthyroidism leads to either an increase in the number of functional exchangers in the membrane or exactly proportional increases in the rate-limiting steps for Na+–Na+ and Na+–H+ exchange activity.  相似文献

Summary To study Cl conductive and cotransport mechanisms, primary cultures of canine tracheal cells were grown to confluency on thin glass cover slips and on porous filters. Transepithelial resistance was >100 ·cm2, and short circuit current (I sc=2–20 A/cm2), representing active secretion of Cl, increased >threefold with addition of 10 m isoproterenol to the serosal solution. Cells made transiently permeable in hypotonic solution were loaded with the Cl-sensitive fluorophore 6-methoxy-N-(3-sulfopropyl) quinolinium (SPQ) (5mm, 4 min, 150 mOsm). The electrical properties of the cell monolayers were not altered by the loading procedure. Intracellular SPQ fluorescence was monitored continuously by epifluorescence microscopy (excitation 360±5 nm, emission>410 nm). SPQ leakage from the cells was <10% in 60 min at 37°C. Intracellular calibration of SPQ fluorescencevs. [Cl] (0–90mm) was carried out using high-K buffers containing the ionophores nigericin (5 m) and tributyltin (10 m); SPQ fluorescence was quenched with a Stern-Volmer constant of 13m –1. Intracellular Cl activity was 43±4mm. Cl flux was measured in response to addition and removal of 114mm Cl from the bathing solution. Addition of 10 m isoproterenol increased Cl efflux from 0.10 to 0.27mm/sec. The increase was inhibited by the Cl-channel blocker diphenylamine-2-carboxylic acid (1mm). In the absence of isoproterenol, removal of external Na or addition of 0.5mm furosemide, reduced Cl influx by >fourfold. In ouabain-treated monolayers, removal of external K in the presence of 5mm barium diminished Cl influx by >twofold, suggesting that Cl entry is in part K dependent. These results establish an accurate optical method for the realtime measurement of intracellular Cl activity in tracheal cells that does not require an electrically tight cell monolayer. The data demonstrate the presence of an isoproterenol-regulated Cl channel and a furosemide-sensitive cation-coupled transport mechanism.  相似文献

Summary Isolated epithelial cells were obtained from abdominal skin of the frogLeptodactylus ocellatus by a trypsination-dissection method. As estimated by nigrosin staining, the amount of damaged cells is only 6.6±0.7 per cent. When washed briefly after incubation the ionic concentrations in these cells were (mm): K+ 14.20±4.0; Na+ 15.8±1.8; and Cl 57.2±5.3. If they are not washed, the concentration of K+ remains essentially the same (131.2±1.4mm) but the Na+ concentration is much higher (38.5±0.9mm). It is shown that a large fraction of Na+ is contained in a compartment that is freely connected with the bathing solution. Ouabain (10–4 m) elicits a marked decrease of K+, a slight decrease of Cl, and an increase of Na+ content. In an equal period, low temperature (3°C) produces a similar effect, although less marked than ouabain.  相似文献

Summary We have measured the intracellular potassium activity, [K+]i and the mechanisms of transcellular K+ transport in reabsorptive sweat duct (RSD) using intracellular ion-sensitive microelectrodes (ISMEs). The mean value of [K+]i in RSD is 79.8±4.1mm (n=39). Under conditions of microperfusion, the [K+]i is above equilibrium across both the basolateral membrane, BLM (5.5 times) and the apical membrane, APM (7.8 times). The Na+/K+ pump inhibitor ouabain reduced [K+]i towards passive distribution across the BLM. However, the [K+]i is insensitive to the Na+/K+/2 Cl cotransport inhibitor bumetanide in the bath. Cl substitution in the lumen had no effect on [K+]i. In contrast, Cl substitution in the bath (basolateral side) depolarized BLM from –26.0±2.6 mV to –4.7*±2.4 mV (n=3;* indicates significant difference) and decreased [K+]i from 76.0±15.2mm to 57.7* ±12.7mm (n=3). Removal of K+ in the bath decreased [K+]i from 76.3±15.0mm to 32.3*±7.6mm (n=4) while depolarizing the BLM from –32.5±4.1 mV to –28.3*±3.0 mV (n=4). Raising the [K+] in the bath by 10-fold increased [K+]i from 81.7±9.0mm to 95.0*±13.5mm and depolarized the BLM from –25.7±2.4 mV to –21.3*±2.9 mV (n=4). The K+ conductance inhibitor, Ba2+, in the bath also increased [K+]i from 85.8±6.7mm to 107.0*±11.5mm (n=4) and depolarized BLM from –25.8±2.2 mV to –17.0*±3.1 mV (n=4). Amiloride at 10–6 m increased [K+]i from 77.5±18.8mm to 98.8*±21.6mm (n=4) and hyperpolarized both the BLM (from –35.5±2.6 mV to –47.8*±4.3 mV) and the APM (from –27.5±1.4 mV to –46.0* ±3.5 mV,n=4). However, amiloride at 10–4 m decreased [K+]i from 64.5±0.9mm to 36.0*±9.9mm and hyperpolarized both the BLM (from –24.7±1.4 mV to –43.5*±4.2 mV) and APM (from –18.3±0.9 mV to –43.5*±4.2 mV,n=6). In contrast to the observations at the BLM, substitution of K+ or application of Ba2+ in the lumen had no effect on the [K+]i or the electrical properties of RSD, indicating the absence of a K+ conductance in the APM. Our results indicate that (i) [K+]i is above equilibrium due to the Na+/K+ pump; (ii) only the BLM has a K+ conductance; (iii) [K+]i is subject to modulation by transport status; (iv) K+ is probably not involved in carrier-mediated ion transport across the cell membranes; and (v) the RSD does not secrete K+ into the lumen.  相似文献

Summary Pulmonary CO-diffusing capacity (D l CO), lung volume, pulmonary perfusion and O2-uptake were measured by non-invasive techniques in the lizardsVaranus exanthematicus andTupinambis teguixin (mean body weight 2.2 kg for both species).The CO-diffusing capacity was at 25–27°C 0.059 mlstpd·kg–1·min–1·Torr–1 inVaranus, which is 47% greater than the value of 0.040 mlstpd·kg–1·min–1·Torr–1 inTupinambis. The lung volume ofVaranus was 36 ml·kg–1 and that ofTupinambis 20 ml·kg–1. At 35–37°C the diffusing capacity of lizard lungs are about 25% of those for mammals of comparable size.InVaranus pulmonary CO-diffusing capacity increased with temperature from 0.027 mlstpd·kg–1·min–1·Torr–1 at 17–19 °C to 0.075 mlstpd·kg–1·min–1·Torr–1 at 35–37 °C. This change closely matched a concomitant increase of O2-uptake. Pulmonary perfusion increased from 27 ml·kg–1·min–1 to 55 ml·kg–1·min–1 within this temperature range.The study emphasizes that pulmonary diffusing capacity cannot be fully evaluated without information on pulmonary perfusion and O2-uptake. In reptiles and other ectotherms diffusing capacity must be reported at specified body temperature.  相似文献

Gallbladder Na+ absorption is linked to gallstone formation in prairie dogs. We previously reported Na+/H+ exchanger (NHE1-3) expression in native gallbladder tissues. Here we report the functional characterization of NHE1, NHE2 and NHE3 in primary cultures of prairie dog gallbladder epithelial cells (GBECs). Immunohistochemical studies showed that GBECs grown to confluency are homogeneous epithelial cells of gastrointestinal origin. Electron microscopic analysis of GBECs demonstrated that the cells form polarized monolayers characterized by tight junctions and apical microvilli. GBECs grown on Snapwells exhibited polarity and developed transepithelial short-circuit current, Isc, (11.6 ± 0.5 µA · cm–2), potential differences, Vt (2.1 ± 0.2 mV), and resistance, Rt (169 ± 12 · cm2). NHE activity in GBECs assessed by measuring dimethylamiloride-inhibitable 22Na+ uptake under a H+ gradient was the same whether grown on permeable Snapwells or plastic wells. The basal rate of 22Na+ uptake was 21.4 ± 1.3 nmol · mg prot–1 · min–1, of which 9.5 ± 0.7 (~45%) was mediated through apically-restricted NHE. Selective inhibition with HOE-694 revealed that NHE1, NHE2 and NHE3 accounted for ~6%, ~66% and ~28% of GBECs total NHE activity, respectively. GBECs exhibited saturable NHE kinetics (Vmax 9.2 ± 0.3 nmol · mg prot–1 · min–1; Km 11.4 ± 1.4 mM Na+). Expression of NHE1, NHE2 and NHE3 mRNAs was confirmed by RT-PCR analysis. These results demonstrate that the primary cultures of GBECs exhibit Na+ transport characteristics similar to native gallbladder tissues, suggesting that these cells can be used as a tool for studying the mechanisms of gallbladder ion transport both under physiologic conditions and during gallstone formation.  相似文献

Summary The effects of stepwise concentration changes of K+ and HCO 3 in the basolateral solution on the basolateral membrane potential (V bl) of proximal tubule cells of the doubly-perfusedNecturus kidney were examined using conventional microelectrodes. Apparent transference numbers were calculated from changes inV bl after alterations in external K+ concentration from 1.0 to 2.5mm (t K, 1.0–2.5), 2.5 to 10, and in external HCO 3 concentration (at constant pH) from 5 to 10mm (t HCO3, 5–10), 10 to 20, or 10 to 50.t K, 2.5–10 was 0.38±0.02 under control conditions but was sharply reduced to 0.08±0.03 (P>0.001) by 4mm Ba++. This concentration of Ba++ reducedV bl by 9±1 mV (at 2.5 external K+). Perfusion with SITS (5×10–4 m) for 1 hr hyperpolarizedV bl by 10±3 mV and increasedt K, 2.5–10 significantly to 0.52±0.01 (P<0.001). Ba++ application in the presence of SITS depolarizedV bl by 22±3 mV. In control conditionst HCO3, 10–50 was 0.63±0.05 and was increased to 0.89±0.07 (P<0.01) by Ba++ but was decreased to 0.14±0.02 (P<0.001) by SITS. In the absence of apical and basolateral chloride, the response ofV bl to bicarbonate was diminished but still present (t HCO3, 10–20 was 0.35±0.03). Intracellular pH, measured with liquid ion-exchange microelectrodes, increased from 7.42±0.19 to 7.57±0.17 (P<0.02) when basolateral bicarbonate was increased from 10 to 20mm at constant pH. These data show that the effects of bicarbonate onV bl are largely independent of effects on the K+ conductance and that there is a significant current-carrying bicarbonate pathway in the basolateral membrane. Hence, both K+ and HCO 3 gradients are important in the generation ofV bl, and their relative effects vary reciprocally.  相似文献

Summary Efflux of36Cl from frog sartorius muscles equilibrated in two depolarizing solutions was measured. Cl efflux consists of a component present at low pH and a pH-dependent component which increases as external pH increases.For temperatures between 0 and 20°C, the measured activation energy is 7.5 kcal/mol for Cl efflux at pH 5 and 12.6 kcal/mol for the pH-dependent Cl efflux. The pH-dependent Cl efflux can be described by the relationu=1/(1+10n(pK a -pH)), whereu is the Cl efflux increment obtained on stepping from pH 5 to the test pH, normalized with respect to the increment obtained on stepping from pH 5 to 8.5 or 9.0. For muscles equilibrated in solutions containing 150mm KCl plus 120mm NaCl (internal potential about –15 mV), the apparent pK a is 6.5 at both 0 and 20°C, andn=2.5 for 0°C and 1.5 for 20°C. For muscles equilibrated in solutions containing 7.5mm KCl plus 120mm NaCl (internal potential about –65 mV), the apparent pK a at 0°C is 6.9 andn is 1.5. The voltage dependence of the apparent pK a suggests that the critical pH-sensitive moiety producing the pH-dependent Cl efflux is sensitive to the membrane electric field, while the insensitivity to temperature suggests that the apparent heat of ionization of this moiety is zero. The fact thatn is greater than 1 suggests that cooperativity between pH-sensitive moieties is involved in determining the Cl efflux increment on raising external pH.The histidine-modifying reagent diethylpyrocarbonate (DEPC) applied at pH 6 reduces the pH-dependent Cl efflux according to the relation, efflux=exp(–k·[DEPC]·t), wheret is the exposure time (min) to DEPC at a prepared initial concentration of [DEPC] (mm). At 17°C,k –1=188mm·min. For temperatures between 10 and 23°C,k has an apparent Q10 of 2.5. The Cl efflux inhibitor SCN at a concentration of 20mm substantially retards the reduction of the pH-dependent Cl efflux by DEPC. The findings that the apparent pK a is 6.5 in depolarized muscles, that DEPC eliminates the pH-dependent Cl efflux, and that this action is retarded by SCN supports the notion that protonation of histidine groups associated with Cl channels is the controlling reaction for the pH-dependent Cl efflux.  相似文献

Summary Forskolin (i.e, cAMP)-modulation of ion transport pathways in filter-grown monolayers of the Cl-secreting subclone (19A) of the human colon carcinoma cell line HT29 was studied by combined Ussing chamber and microimpalement experiments.Changes in electrophysiological parameters provoked by serosal addition of 10–5 m forskolin included: (i) a sustained increase in the transepithelial potential difference (3.9±0.4 mV). (ii) a transient decrease in transepithelial resistance with 26±3 · cm2 from a mean value of 138±13 · cm2 before forskolin addition, (iii) a depolarization of the cell membrane potential by 24±1 mV from a resting value of –50±1 mV and (iv) a decrease in the fractional resistance of the apical membrane from 0.80±0.02 to 0.22±0.01. Both, the changes in cell potential and the fractional resistance, persisted for at least 10 min and were dependent on the presence of Cl in the medium. Subsequent addition of bumetanide (10–4 m), an inhibitor of Na/K/2Cl cotransport, reduced the transepithelial potential, induced a repolarization of the cell potential and provoked a small increase of the transepithelial resistance and fractional apical resistance. Serosal Ba2+ (1mm), a known inhibitor of basolateral K+ conductance, strongly reduced the electrical effects of forskolin. No evidence was found for a forskolin (cAMP)-induced modulation of basolateral K+ conductance.The results suggest that forskolin-induced Cl secretion in the HT-29 cl.19A colonic cell line results mainly from a cAMP-provoked increase in the Cl conductance of the apical membrane but does not affect K+ or Cl conductance pathways at the basolateral pole of the cell. The sustained potential changes indicate that the capacity of the basolateral transport mechanism for Cl and the basal Ba2+-sensitive K+ conductance are sufficiently large to maintain the Cl efflux across the apical membrane. Furthermore, evidence is presented for an anomalous inhibitory action of the putative Cl channel blockers NPPB and DPC on basolateral conductance rather than apical Cl conductance.  相似文献

Summary Measurements of unidirectional calcium fluxes in stripped intestinal epithelium of the tilapia,Oreochromis mossambicus, in the presence of ouabain or in the absence of sodium indicated that calcium absorption via the fish intestine is sodium dependent. Active Ca2+ transport mechanisms in the enterocyte plasma membrane were analyzed. The maximum capacity of the ATP-dependent Ca2+ pump (V m :0.63 nmol·min–1 mg–1,K m : 27nm Ca2+) is calculated to be 2.17 nmol·min–1·mg–1, correcting for 29% inside-out oriented vesicles in the membrane preparation. The maximum capacity of the Na+/Ca2+ exchanger with high affinity for Ca2+ (V m :7.2 nmol·min–1·mg–1,K m : 181nm Ca2+) is calculated to be 13.6 nmol·min–1·mg–1, correcting for 53% resealed vesicles and assuming symmetrical behavior of the Na+/Ca2+ exchanger. The high affinity for Ca2+ and the sixfold higher capacity of the exchanger compared to the ATPase suggest strongly that the Na+/Ca2+ exchanger will contribute substantially to Ca2+ extrusion in the fish enterocyte. Further evidence for an important contribution of Na+/Ca2+ exchange to Ca2+ extrusion was obtained from studies in which the simultaneous operation of ATP-and Na+-gradient-driven Ca2+ pumps in inside-out vesicles was evaluated. The fish enterocyte appears to present a model for a Ca2+ transporting cell, in which Na+/Ca2+ exchange activity with high affinity for Ca2+ extrudes Ca2+ from the cell.  相似文献

Summary We have investigated the effect of a purified preparation of Charybdotoxin (CTX) on the Ca-activated K+ (Ca–K) channel of human red cells (RBC). Cytosolic Ca2+ was increased either by ATP depletion or by the Ca ionophore A23187 and incubation in Na+ media containing CaCl2. The Ca–K efflux activated by metabolic depletion was partially (77%) inhibited from 15.8±2.4 mmol/liter cell · hr, to 3.7±1.0 mmol/liter cell · hr by 6nm CTX (n=3). The kinetic of Ca–K efflux was studied by increasing cell ionized Ca2+ using A23187 (60 mol/liter cell), and buffering with EGTA or citrate; initial rates of net K+ efflux (90 mmol/liter cell K+) into Na+ medium containing glucose, ouabain, bumetanide at pH 7.4 were measured. Ca–K efflux increased in a sigmoidal fashion (n of Hill 1.8) when Ca2+ was raised, with aK m of 0.37 m and saturating between 2 and 10 m Ca2+. Ca–K efflux was partially blocked (71±7.8%, mean ±sd,n=17) by CTX with high affinity (IC500.8nm), a finding suggesting that is a high affinity ligand of Ca–K channels. CTX also blocked 72% of the Ca-activated K+ efflux into 75mm K+ medium, which counteracted membrane hyperpolarization, cell acidification and cell shrinkage produced by opening of the K+ channel in Na+ media. CTX did not block Valinomycin-activated K+ efflux into Na+ or K+ medium and therefore it does not inhibit K+ movement coupled to anion conductive permeability.TheV max, but not theK m–Ca of Ca–K efflux showed large individual differences varying between 4.8 and 15.8 mmol/liter cell · min (FU). In red cells with Hb A,V max was 9.36±3.0 FU (mean ±sd,n=17). TheV max of the CTX-sensitive, Ca–K efflux was 6.27±2.5 FU (range 3.4 to 16.4 FU) in Hb A red cells and it was not significantly different in Hb S (6.75±3.2 FU,n=8). Since there is larger fraction of reticulocytes in Hb S red cells, this finding indicates that cell age might not be an important determinant of theV max of Ca–K+ efflux.Estimation of the number of CTX-sensitive Ca-activated K+ channels per cell indicate that there are 1 to 3 channels/per cell either in Hb A or Hb S red cells. The CTX-insensitive K+ efflux (2.7±0.9 FU) may reflect the activity of a different channel, nonspecific changes in permeability or coupling to an anion conductive pathway.  相似文献

Summary The effect of chloride on 4,4-dibenzamido-2,2-disulfonic stilbene (DBDS) binding to band 3 in unsealed red cell ghost membranes was studied in buffer [NaCl (0 to 500mm) + Na citrate] at constant ionic strength (160 or 600mm). pH 7.4, 25°C. In the presence of chloride, DBDS binds to a single class of sites on band 3. At 160mm ionic strength, the dissociation constant of DBDS increases linearly with chloride concentration in the range [Cl]=450mm. The observed rate of DBDS binding to ghost membranes, as measured by fluorescence stopped-flow kinetic experiments, increases with chloride concentration at both 160 and 600mm ionic strength. The equilibrium and kinetic results have been incorporated into the following model of the DBDS-band 3 interaction: The equilibrium and rate constants of the model at 600mm ionic strength areK 1=0.67±0.16 m,k 2=1.6±0.7 sec–1,k –2=0.17±0.09 sec–1,K 1=6.3±1.7 m,k 2=9±4 sec–1 andk –2=7±3 sec–1. The apparent dissociation constants of chloride from band 3,K Cl, are 40±4mm (160mm ionic strength) and 11±3mm (600mm ionic strength). In mammals, D-fructose transport takes place across the brush-border membrane of the small intestine through GLUT5, a member of the facilitative glucose transporter family. In the present paper, we describe and characterize for the first time the apical transport of D-fructose in chicken intestine. Brush-border membrane vesicles (BBMV) were obtained from jejunum of 5- to 6-wk-old chickens. D-Fructose uptake by BBMV from chicken jejunum comprises a saturable component and a simple diffusion process. The maximal rate of transport (Vmax) for D-fructose was 2.49 nmol·(mg prot)–1·s–1, the Michaelis constant (Km) was 29 mM, and the diffusion constant (Kd) was 25 nl·(mg prot)–1·s–1. The apical transport of D-fructose was Na+-independent, phlorizin-, phloretin-, and cytochalasin B-insensitive, and did not show cis-inhibition by D-glucose or D-galactose. These properties, together with the detection of specific GLUT5 mRNA, indicate the presence of a low-affinity high-capacity GLUT5-type carrier in the chicken jejunum, responsible for the entry of D-fructose across the brush-border membrane of enterocytes.  相似文献

A comparative study has been carried out with FDP aldolases fromEscherichia coli 518 andLactobacillus casei ATCC 7469, which had been purified 17.6- and 65-fold, respectively. The aldolase ofL.casei was stable only in the presence of mercaptoethanol, whereas that ofE.coli was strongly inhibited at low (1.0×10–4 m) and activated at high concentrations (2.0×10–1 m) of the same compound.p-Chloromercuric benzoic acid inhibited both aldolases, with 40% inhibition at 2×10–5 m withE.coli aldolase against at 2×10–4 m withL.casei aldolase. Significant differences were also observed in pH optima and Km values.E.coli aldolase exhibited a maximal activity at pH 9.0 and gave a Km value of 1.76×10–3 m FDP with strong substrate inhibition above 7×10–3 m, against pH 6.8–7.0 and a Km of 7.04×10–3 m FDP forL.casei aldolase. Strong resistance ofL.casei aldolase against inhibition by EDTA, Ca2+ and Mn2+ was observed compared with complete inhibition at concentrations of 20mm, 40mm and 20mm, respectively, withE. coli aldolase. Polyacrylamide gel electrophoresis did not reveal any differences between the two enzyme preparations.The differences of the properties of FDP aldolases from different bacterial genera are discussed in relation to other Class II aldolases.  相似文献

Summary The steady-state, current-voltage (I–V) characteristics of stomatal guard cells fromVicia faba L. were explored by voltage clamp using conventional electrophysiological techniques, but with double-barrelled microelectrodes containing 50mm K+-acetate. Attention was focused, primarily, on guard cell response to metabolic blockade. Exposures to 0.3–1.0mm NaCN and 0.4mm salicylhydroxamic acid (SHAM) lead consistently to depolarizing (positive-going) shifts in guard cell potentials (V m ), as large as +103 mV, which were generally complete within 60–90 sec (mean response half-time, 10.3±1.7 sec); values forV m in NaCN plus SHAM were close or positive to –100 mV and well removed from the K+ equilibrium potential. Guard cell ATP content, which was followed in parallel experiments, showed a mean half-time for decay of 10.8±1.9 ([ATP] t=0, 1.32±0.28mm; [ATP] t=60–180sec, 0.29±0.40mm). In respiring cells, theI–V relations were commonly sigmoid aboutV m or gently concave to the voltage axis positive toV m . Inward- and outward-rectifying currents were also observed, especially near the voltage extremes (nominally –350 and +50 mV). Short-circuit currents (atV=0 mV) were typically about 200–500 mA m–2. The principal effect of cyanide early on was to linearize theI–V characteristic while shifting it to the right along the voltage axis, to decrease the membrane conductance, and to reduce the short-circuit current by approx. 50–75%. The resulting difference-current-voltage (dI–V) curves (±cyanide) showed a marked sensitivity to voltages negative from –100 mV and, when clamp scans had been extended sufficiently, they revealed a distinct minimum near –300 mV before rising at still more negative potentials. The difference currents, along with changes in guard cell potential, conductance and ATP content are interpreted in context of a primary, ATP-consuming ion pump. FittingdI–V curves to reaction kinetic model for the pump [Hansen, U.-P., et al. (1981)J. Membrane Biol. 63:165; Blatt, M.R. (1986)J. Membrane Biol. 92:91] implicates a stoichiometry of one (+) charge transported outward for each ATP hydrolyzed, with pump currents as high as 200 mA m–2 at the free-running potential. The analysis indicates that the pump can comprise more than half of the total membrane conductance and argues against modulations of pump activity alone, as an effective means to controlling K+ transport for stomatal movements.  相似文献

Summary Addition of glucose or the nonmetabolizable analogue -methyl-d-glucoside to rabbit proximal tubules suspended in a glucoseand alanine-free buffer caused a sustained increase in intracellular Na+ content (+43±7 nmol · (mg protein)–1) and a concomitant but larger decrease in K+ content (–72±11 nmol· (mg protein)–1). A component of the net K+ efflux was Ba2+ insensitive, and was inhibited by high (1mm) but not low (10 m) concentrations of the diuretics, furosemide and bumetanide. The increase in intracellular Na+ content is consistent with the view that the increased rates of Na+ and water transport seen in the proximal tubule in the presence of glucose can be attributed (at least in part) to a stimulation of basolateral pump activity by an increased [Na+] i .  相似文献
