Summary Two new nitrite oxidizing bacteria for which the names Nitrococcus mobilis and Nitrospina gracilis are proposed were isolated from the marine environment. Nitrococcus mobilis was cultured from South Pacific waters and it is a large motile coccus with unique tubular cytomembranes. Nitrospina gracilis was isolated from South Atlantic waters and it is a long slender rod which lacks an extensive cytomembrane system. Both are obligate marine organisms and both are obligate chemoautotrophs. The Irish plant which, following Mitten, has commonly been referred to as Plagiochila ambagiosa Mitt. is specifically different from the type of P. ambagiosa, and is therefore described as Plagiochila atlantica F. The inhibition of cytochrome c oxidase by cyanide, starting either with the resting or the pulsed enzyme, was studied by rapid-freeze quenching followed by quantitative e.p.r. It is found that a partial reduction of cytochrome oxidase by transfer of 2 electron equivalents from ferrocytochrome c to cytochrome a and CuA will induce a transition from a closed to an open enzyme conformation, rendering the cytochrome a3-CuB site accessible for cyanide binding, possibly as a bridging ligand. A heterogeneity in the enzyme is observed in that an e.p.r. signal from the cytochrome a3 3+-HCN complex is only found in 20% of the molecules, whereas the remaining cyanide-bound a3-CuB sites are e.p.r.-silent.  相似文献

Fumarase from chicken heart is purified 400 times from the crude muscle extract. The isolation procedure includes ammonium sulfate fractionations, Bio-Gel P-300 column chromatography and electrofocusings on pH-gradients from pH 3 to 10 and from pH 7 to 9. Chicken fumarase behaves as an homogeneous protein in sedimentation, diffusion and electrofocusing studies; the protein possesses a single amino-terminal residue: lysine. The analysis of the CD and ORD spectra suggests the presence of 60-65 p. cent of alpha-helix, 0 - 5 p. cent of beta-structure with the remaining portions of the protein in an unordered conformation. Chicken fumarase is found to be composed of 4 subunits of identical molecular weight (51.000) and devoid of disulfide bridges. Finally, the physicochemical properties of chicken fumarase are compared with those of the porcine enzyme.  相似文献

Monogeneans from three species of Cephalopholis, namely C. argus, C. sonnerati and C. boenak, are described from fish caught off New Caledonia, South Pacific, with comparisons with material from off Queensland, Australia. Pseudorhabdosynochus argus n. sp. from C. argus is present off New Caledonia and Australia; it is characterised by its male quadriloculate organ with very elongate cone, and its sclerotised vagina with anterior trumpet, coiled primary canal and distal part with two chambers and an accessory part. C. boenak has no monogeneans off New Caledonia, but off Australia it harbours Pseudorhabdosynochus sp., a new species which is morphologically related to P. argus. P. minutus n. sp. from C. sonnerati is characterised by its minute body and a sclerotised vagina with two spherical chambers. Diplectanum nanus n. sp. from C. sonnerati is characterised by its very small funnel-shaped male copulatory organ and minute body. A new species, Haliotrema sp. from C. sonnerati is characterised by a very elongate tubular penis; it is distinct from H. cromileptis Young, 1968 (redescribed herein from specimens collected from Cromileptes altivelis off New Caledonia). The species described here include the first members of Pseudorhabdosynochus and the first diplectanids described from species of Cephalopholis. There is no evidence for a clade of Pseudorhabdosynochus species specific to members of Cephalopholis, since the species described here share similarities with other species from Epinephelus. However, it is suggested that the gill structure of Cephalopholis spp. imposes selection toward small body sizes for monogeneans.  相似文献

The role and historical progress of n.m.r. applications in biochemistry are briefly outlined. Technical advances over the years have made n.m.r., at last, a technique which can give valuable information about a wide range of biochemical topics, from enzyme kinetics in vivo to the structure of protein-DNA complexes. Emphasis here is placed on studies of proteins, especially those made up from mosaics of modules. It is shown that n.m.r. can readily give detailed structural information about individual protein modules and that valuable information about the structure and function of the intact mosaic protein can be inferred.  相似文献

Summary Nitrosolobus multiformis is a lobular shaped, previously undescribed ammonia-oxidizing bacterium. The organism is ubiquitous and was isolated from soil samples obtained in various parts of the world. Its lobular nature and its internal, partially compartmentalized cytoplasm makes it morphologically unique and easily distinguishable from all other microorganisms. Physiological compartmentalization also occurs and is characterized by glycogen deposition in the peripheral compartments of the cell. The cells are obligate chemoautotrophs using CO2 and ammonia as primary carbon and energy sources. Its obligate chemoautotrophic nature stems primarily from a metabolic deficiency. Even though the cells cannot be grown on an organic medium the cells still have a slight heterotrophic potential and are able to oxidize and assimilate minute amounts of acetate in the absence of an inorganic energy source.Contribution No. 2606 from the Woods Hole Oceanographic Institution, Woods Hole, Massachusetts 02543.  相似文献

Excellent estimations of initial rates can be obtained from plots of delta P/t versus product formed (where P is the instantaneous concentration of the product). delta P/t is the chord from P0,t0 to P,t on an ordinary P-versus-t plot. When the chord is plotted as a function of product, the intercept at P0 of the resulting curve is necessarily dP/dt0. This curve approximates to a straight line extremely closely in all cases tested thus far. If delta P/t versus product is calculated from the integrated rate equation for a first-order reaction, and if a straight line is fitted through points representing the first 50% of the reaction, the discrepancy between the true initial rate and dP/dt0 estimated from the plot is 0.68%. For the most common form of the integrated rate equation for catalysed reactions the discrepancy varies between 0 and 0.90%. Because of the complexities of the integrated rate equations, catalysed second-order reactions have not been evaluated directly; uncatalysed reactions have been done instead. For a reaction with one reactant and two products, the discrepancy varies from 0.68 to 2.02%. For two reactants and one product, it varies from 0 to 0.68%; for two and two, 0 to 2.02%. The larger discrepancies occur only when unfavourable equilibrium constants are being overcome by the initial conditions.  相似文献

Two major subspecies of Staphylococcus cohnii, namely S. cohnii subsp. cohnii, from humans, and S. cohnii subsp. urealyticum, from humans and other primates, are described on the basis of a study of 14 to 25 strains and 18 to 33 strains, respectively. DNA-DNA hybridization studies conducted in our laboratory in 1983 (W. E. Kloos and J. F. Wolfshohl, Curr. Microbiol. 8:115-121, 1983) demonstrated that strains representing the different subspecies were significantly divergent. S. cohnii subsp. urealyticum can be distinguished from S. cohnii subsp. cohnii on the basis of its greater colony size; pigmentation; positive urease, beta-glucuronidase, and beta-galactosidase activities; delayed alkaline phosphatase activity; ability to produce acid aerobically from alpha-lactose; and fatty acid profile. The type strain of S. cohnii subsp. cohnii is ATCC 29974, the designated type strain of S. cohnii Schleifer and Kloos 1975b, 55. The type strain of S. cohnii subsp. urealyticum is ATCC 49330.  相似文献

Summary A vigorously nitrogen-fixing bacterium was isolated from a West Bengal soil. Cells from young cultures on nitrogen-deficient agar medium appear as rods measuring approx. 3–6x1.0–1.2 ., with rounded ends and homogeneous cytoplasm. Older cells contain big refractile bodies representing vacuoles and fat deposits and show a wide variation in shape. Short motile cells with a single polar flagellum arise in liquid medium with combined (ammonia) nitrogen. The Gram reation is negative and endospores are not formed.The organism is aerobic but catalase-negative. It grows rather feebly in sugar-free protein media, but in nitrogen-deficient media it produces ah heavy and extremely tenacious growth, of a characteristic uneven appearance on agar and changing from white to dark red-brown. Glucose, fructose, ethanol, glycerol, mannitol and sorbitol are the best sources of carbon. Acid but no gas is formed from several carbohydrates. Nitrogen fixation is slow but may amount to 20–25 mg. per gram glucose or mannitol supplied. Molybdenum stimulates nitrogen fixation strongly and cannot be replaced by vanadium.Various forms of combined nitrogen, especially glutamic acid, are assimilated more readily than free nitrogen. Growth can be initiated within aPh-interval from approx. 5 to 9; growing cultures may be acidified to Ph 4.5 or less.The organism is morphologically, culturally and serologically different from Azotobacter and Beijerinckia and is regarded as representing a new genus.  相似文献

Conlarium duplumascospora gen. et. sp. nov. and Jobellisia guangdongensis sp. nov. are described and illustrated from submerged wood collected from Guangdong Province, China. Conlarium duplumascospora is characterized by gregarious, coriaceous and beaked ascomata; cylindrical, unitunicate asci with a bipartite apical ring; biseriate, fusiform, hyaline, 0-5-septate ascospores with or without appendages; and anamorph with muriform conidia. Jobellisia guangdongensis is characterized by globose to subglobose, gregarious and papillate ascomata; three-layered peridium; cylindrical, unitunicate asci with a refractive apical ring; and one-septate, fusiform, greenish brown ascospores. Sequence analyses of partial nuclear large subunit ribosomal DNA (LSU rDNA) were performed to infer the phylogenetic affinities of these new taxa. A four-step procedure for purification of a nuclease from the keratinous sponge Verongia aerophoba is described. The extracted material is lyophilized, acidified, and subjected to chromatography on Sephadex, hydroxyapatite, and phosphocellulose. The nuclease is purified about 1,000-fold from the crude extract and approximately 1,600-fold from concomitant acid RNase. Phosphodiesterase is lost after chromatography on Sephadex. The purified enzyme solution contains one single activity as determined by polyacrylamide gel electrophoresis.  相似文献

This paper,third in the series of an account of the genus Athyrium in China, presents a revision of the series Epiraches Ching et Y.T.Hsieh ex Y.T.Hsieh.A total of 11 species are recognized from China,and more than 20 names are reduced to synonyms in the present paper.Ser.Epiraches is anatural group in Athyrium,it is distinguished from all the other groups of the genus by its Asplenioid sori and indusia,The series is entirely Asian tropical,subtropical and primarily montane.Its northern limits in central and eastern China are not beyond the Yangtze River.Southwestern part and Taiwan of China and Japan Islands are centers of diversity for the series,each with more than five species.Some speciesare disjunctly distributed in southwest China-Taiwan-Japan;one species(Athyrium roseum) occurs discontinuously between Yunnan and Taiwan;one endemic to Hainan(A. hainanense)is the only representative of Athyrium on the island.All members of this series grow in evergreen broad-leaf forests,generally from 500 to 1800 m above sea level.A key to the species,and illustrations of the middle pinnae from different specimens are provided.  相似文献

The cell membrane complex of wool has been examined by electron microscopy of stained cross sections after immersion of the wool in formic acid. The cell membrane complex of the cortex is considerably modified by the treatment, but that of the cuticle appears unchanged. Resistant membranes from cuticle cells, cortical cells and wool have been prepared by treatment with performic acid-ammonia. Amino acid analyses show that the resistant membranes from the cuticle contain citrulline but those from cortical cells do not. It is concluded that the cell membrane complex of the cuticle differs from that of the cortex. Because of the high lysine content of the resistant membranes, their resistance to chemical attack, the hydrophobicity of epicuticle and the observation of a small amount of epsilon-(gamma-glutamyl)lysine, it is postulated that the resistant membranes may contain an appreciable amount of epsilon-(gamma-glutamyl)lysine cross links.  相似文献

A well-marked hierarchy of centres can be recognized within the suboesophageal lobes and ganglia of the arms. The inputs and outputs of each lobe are described. There are sets of motoneurons and intermediate motor centres, which can be activated either from the periphery or from above. They mostly do not send fibres up to the optic or higher motor centres. However, there is a large set of fibres running from the magnocellular lobe to all the basal supraoesophageal lobes. The centre for control of the four eye-muscle nerves in the anterior lateral pedal lobe receives many fibres direct from the statocyst and from the peduncle and basal lobes, but none direct from the optic lobe. The posterior lateral pedal is a backward continuation of the oculomotor centre, containing large cells that may be concerned in initiating attacks by the tentacles. An intermediate motor centre in the posterior pedal lobe probably controls steering. It sends fibres to the funned and head retractors, and by both direct and interrupted pathways to the fin lobe. It receives fibres from the crista nerve and basal lobes, but none direct from the optic lobe. The jet control centre of the ventral magnocellular lobe receives fibres from the statocyst and skin and also from the optic and basal lobes. Some of these last also give extensive branches throughout the palliovisceral lobes. The branching patterns of the dendritic collaterals differ in the various lobes. Some estimates are given of the numbers of synaptic points. The dendritic collaterals of the motoneurons spread through large volumes of neuropil and they overlap. The incoming fibres spread widely and each presumably activates many motoneurons either together or serially. Many of the lobes contain numerous microneurons with short trunks restricted to the lobe, but there are none of these cells in the chromatophore lobes or fin lobes. The microneurons have only few dendritic collaterals, in contrast to the numerous ones on the nearby motoneurons.  相似文献

A new subspecies, Staphylococcus capitis subsp. ureolyticus, was isolated from human skin and is described on the basis of studies of 15 to 26 strains. DNA-DNA reassociation reactions demonstrated that these strains were closely related to Staphylococcus capitis but were significantly divergent. The strains of S. capitis subsp. ureolyticus can be distinguished from S. capitis by their positive urease activity, their ability to produce acid from maltose under aerobic conditions, their fatty acid profile, and their colony morphology. The type strain of the new subspecies is strain ATCC 49326.  相似文献
