The effect of hormone-like compounds at different concentrations: 2,4-D (2 × 10?6; 2 × 10?5; and 2 × 10?4M) and 1-NAA (2 × 10?7; 2 × 10?6; 2 × 10?5; 4 × 10?5, and 6 × 10?5 M) on the growth and production of phenolic compounds, including flavans and lignin, was investigated in callus culture of tea plant (Camellia sinensis L., a highly productive strain IFR ChS-2). The growth of the culture was vigorous, and production of phenolic compounds therein was efficient in the medium containing 2 × 10?5 M 2,4-D. Substitution of 1-NAA for 2,4-D in all the cases decelerated the growth of the culture. These changes were more pronounced when 2 × 10?7 and 2 × 10?6 M 1-NAA was used; in this case, biomass accumulation decreased by 1.5–2.0 times as compared with control material growing on the medium with 2 × 10?5 M 2,4-D. In the presence of 1-NAA, the content of total soluble phenolic compounds and flavans in the calli rose by 30% on the average as compared with control material. Accumulation of lignin remained essentially the same. Therefore, the replacement of 2,4-D with 1-NAA in the nutrient medium used for the growing of highly productive strain of tea plant callus did not induce considerable changes in its ability to produce phenolic compounds.  相似文献

The effect of ultraviolet (UV-B) radiation on the accumulation and tissue localization of phenolic compounds in two strains of callus cultures of tea plant (Camellia sinensisL.) were investigated. The strains differed in their morphological and physiological characteristics and biosynthetic capacity. UV-B radiation hampered culture growth, decreased the size of callus-forming cells and promoted the accumulation of soluble and, to a lesser extent, polymeric forms of phenolic compounds, such as lignin. This accumulation was accompanied by an increase in the phenolic compound deposition in cell walls and intercellular space and by deposition of a lignin-like material on the surface of callus cultures. The strain characterized by an increased formation of phenolic compounds was more resistant to UV-B radiation as compared to that with lower phenolic productivity.  相似文献

Cadmium stress in sugar cane callus cultures: Effect on antioxidant enzymes   总被引:5,自引:0,他引:5  
Catalase (CAT) and superoxide dismutase (SOD) are antioxidant enzymes which are important in the metabolism of reactive oxygen species (ROS), and can be induced by environmental stresses including cadmium (Cd), a heavy metal toxic to living organisms. Sugar cane (Saccharum officinarumL.) in vitro callus cultures were exposed to CdCl2 and the activities of CAT and SOD were analysed. Lower concentrations of CdCl2, such as 0.01 and 0.1 mM caused a significant increase in callus growth, whereas 0.5 and 1 mM CdCl2 strongly inhibited growth of the callus cultures, but only after 9 days of CdCl2 treatment. Red-brown patches were also observed in calluses exposed to 0.5 and 1 mM CdCl2. Calluses grown in 0.01 and 0.1 mM CdCl2 did not exhibit any changes in CAT activity even after 15 days of growth in the presence of CdCl2. However, for calluses grown in higher concentrations of CdCl2 (0.5 and 1 mM), a rapid increase in CAT activity was detected, which was 14-fold after 15 days. Furthermore, up to five CAT isoforms were observed in callus tissue. Total SOD activity did not exhibit any major variation. One Mn-SOD and two Cu/Zn-SOD isoenzymes were observed in callus cultures and none exhibited any variation in response to the CdCl2 treatments. The results suggested that in sugar cane callus cultures, CAT may be the main antioxidant enzyme metabolizing H2O2.  相似文献

Electrophoretic patterns for isocitrate dehydrogenase (IDH; EC 1.1.1.42), acid phosphatase (ACP; EC 3.1.3.2), peroxidase (PER; EC 1.11.1.7), and esterase (EST; EC 3.1.1.1) isozymes were determined inCereus peruvianus tissues and used as markers of genetic uniformity of calli and of the plants regenerated from callus cultures. One IDH, six ACP, six PER, and six EST isozymes were induced in cultured callus tissues in medium containing three 2,4-dichlorophenoxyacetic acid and kinetin combinations. Four ACP, two PER, and three EST isozymes were still present in all regenerated plantsin vitro and therefore can be used as markers of theC. peruvianus plants regenerated from callus tissues. The differential patterns of ACP and IDH isozymes and the similar zymograms for PER and EST isozymes presented by callus tissues were used in a comparison of callus tissues cultured for 2 years. The comparative analysis of zymograms within each enzyme system indicated a mean heterogeneity coefficient of 0.33 forC. peruvianus calli cultured for 2 years. Because of the isozyme variations, which developed in culture medium and were transferred to the regenerated plants, the IDH, ACP, PER, and EST enzyme systems can be considered to be good markers for investigating possible genetic variations in plant populations ofC. peruvianus obtainedin vitro from callus culture.This research was supported by the CNPq  相似文献

Aims: To investigate the effect of phenolic compounds on the growth of Oenococcus oeni. Methods and Results: Oenococci are usually grown in media often supplemented with complex additives such as tomato juice. In order to improve our knowledge about the growth requirements of oenococci, we added several juices and leaf extracts such as green tea to the culture media and screened them for growth‐stimulating substances to substitute complex supplements such as juices by more defined components. We found that also green tea could cause a growth stimulation of Oenococcus oeni strain B2. Conclusions: Further experiments showed that the stimulating effect was as a result of the phenolic compounds of green tea, especially epigallocatechin gallate (EGCG). On the other hand, EGCG could also inhibit the growth of O. oeni strain B2 just depending on its concentration. Significance and Impact of the Study: Individual catechins should have a minor influence on the growth of oenococci during wine making as their concentration in grapes is <30 mg kg?1 grape. Whether there is a synergistic effect of the different catechins in wine has to be investigated.  相似文献

A simple method for the Agrobacterium-mediated transformation of callus cultures of nine plant species, Lycopersicum esculentum Mill, Petunia hybrida Vilm, Pimpinella anisum L., Solanum melongena L., S. tuberosum L., Nicotiana glauca Graham, N. glutinosa L., N. plumbaginifolia Viviani and N. tabacum L., is described. Plant calli were resuspended in liquid media, co-cultivated with A. tumefaciens, and plated on restrictive media. The combination of a gene for kanamycin resistance and a gene for firefly luciferase was convenient in the selection and confirmation of hundreds of transformants. Four strains of A. tumefaciens, A208, A348, A281, and PC2760, were employed. All of the callus cultures were successfully transformed with at least one strain of A. tumefaciens, and A281 was the most effective of the four strains. N. glutinosa, N. plumbaginifolia, N. tabacum, P. hybrida and L. esculentum were transformed more efficiently than the other species tested.  相似文献

Summary Callus was initiated from in vitro-grown plants of Gladiolus cultivars ‘Jenny Lee’ and ‘Florida Flame.’ The age of callus used for regeneration of plants was either 9 mo. old or 8 yr old from ‘Jenny Lee,’ and 4 mo. old from ‘Florida Flame.’ Regeneration medium consisted of Murashige and Skoog’s basal salts medium supplemented with 2.0 mg/l (9.3 μM) kinetin. This medium was supplemented with various concentrations of either bialaphos (Meiji Seika, Tokyo, Japan) or phosphinothricin (Hoechst-Roussell, Frankfurt, Germany). Bialaphos was more effective than phosphinothricin at stimulating plant regeneration. Plants regenerated from 8-yr-old callus of ‘Jenny Lee’ only when the regeneration medium was supplemented with 0.10 mg/l bialaphos. A bialaphos concentration of 0.01 mg/l stimulated regeneration from 9-mo.-old callus of cultivar ‘Jenny Lee’ and 4-mo.-old callus of ‘Florida Flame.’  相似文献

Effector studies with two isoenzymes (I and IV) of glucose-6-phosphate dehydrogenase (G6PDH) from tobacco suspension culture WR-132 revealed that chlorogenic acid, at 0.4 mM, inhibited both isoenzymes almost 100%, with the inhibition decreasing as the concentration of the acid was reduced. At 0.3 and 0.4 mM, the coumarin glucosides scopolin and esculin were inhibitory, whereas their aglucones scopoletin and esculetin were less inhibitory, and at low concentrations of glucose-6-phosphate (G6P), the latter two were actually stimulatory for G6PDH I. Of the possible effectors studied, only scopoletin and esculetin exhibited a significant activation of G6PDH I under these conditions. However, with G6PDH IV these two effectors do not show the same marked activation at the low G6P concentrations. The phenolic acids, caffeic and ferulic, were less inhibitory than the coumarins tested. The activation of G6PDH I by scopoletin, a compound which accumulates in tobacco under certain stress conditions, gives a possible clue as to the resulting enhanced activity of the hexose monophosphate pathway that has been reported for some plants subjected to stress conditions.  相似文献

Root, hypocotyl, cotyledon, stem and leaf of Cucumis melo var. utilissimus seedlings were used for callus induction. Comparison was made between these parts, between callus tissues originating from all the parts and between each part and its callus, with respect to the fatty acid composition of total lipids. In all the parts there was a greater proportion of unsaturated fatty acids, the predominant fatty acid in root, stem and leaf being linolenic acid whilst in the cotyledon linoleic predominated. In the hypocotyl these two acids were present in equal amounts. In callus cultures the proportion of saturated acids was greater and the predominant fatty acid was palmitic. The major unsaturated fatty acid in callus cultures was linolenic. The analysis showed that callus tissue and its respective plant part had different fatty acid patterns and that all the callus cultures had very similar patterns irrespective of their origin.  相似文献

The phyto-constituents of Gymnema sylvestre are used in the treatment of diabetes and obesity. The present work reports on the extraction of gymnemic acid through gymnemagenin from callus cultures of G. sylvestre. Components were separated on pre-coated silica gel 60 GF254 plates with chloroform:methanol (8:2) and scanned using a densitometric scanner at 205 nm in the near-UV region. Linearity of determination of gymnemagenin was observed in the range 2-10 microg. The average percentage recovery of gymnemagenin from leaf callus extracts was 98.9+/-0.3.  相似文献

Cigarette beetle, Lasioderma serricorne (F.), is a common food-product pest widely found in homes. This paper investigated the relationship between the number of infesting cigarette beetles and the phenolic content of pennywort tea made from young and mature leaves during one-year storage. It was found that cigarette beetles started to appear in tea bags after six months of storage. More adults were found than larvae and pupae. In addition, the number of cigarette beetles infesting pennywort tea made from mature leaves was higher than when it was made from young leaves. The amount of phenolic compounds in pennywort tea was analyzed by Folin–Ciocalteau assay. The results of a regression analysis indicated that the number of adult cigarette beetles correlated negatively with the amount of phenolic compounds in pennywort tea made from young leaves – when the total phenolic content decreased, the number of infesting cigarette beetles increased (r2 = 0.80). The number of larvae and pupae showed a weak negative correlation with the amount of phenolic compounds (r2 = 0.40 and r2 = 0.56, respectively). Similar correlations were found in pennywort tea made from mature leaves, but the number of infesting beetles was higher due to the lower phenolic content of the mature leaves.  相似文献

Callus cultures of Siberian larch (Larix sibiricaLedeb.) and Siberian spruce (Picea obovataLedeb.) were used to demonstrate the elicitor activity of two ophiostomatoid fungal species, Ceratocystis laricicolaand Ceratocystis polonica, as the pioneer settlers on larch and spruce, respectively. The extract from C. laricicolamycelium stimulated the accumulation of lignin in larch cells by 37% and that of bound proanthocyanidins by 25%. In spruce callus cultures, C. laricicolaand C. polonicaincreased the bound PA content by 25 and 46%, respectively. In the callus cultures of larch and spruce, the addition of extract of C. laricicolaincreased the concentration of p-hydroxybenzoic acid 13-fold and nearly 4-fold, respectively. The metabolic characteristics of accumulation of phenolic compounds in conifer cells are discussed.  相似文献

The present study was conducted in the hill station of G B Pant University of Agriculture and Technology, Ranichauri, Uttarakhand, which is known for its diversity in medicinal and aromatic plants. The plant geranium, also known as rose or lemon geranium is highly valued for its essential oil which is used in the cosmetic and preferring industries. The plant suffers due to stem and root rot both during summer and winter seasons. In the present study, geranium plants showed maximum stem rot incidence (60.55%) and plant mortality (17.20%) during the winter season, whereas in summer the crops showed maximum root rot and wilt complex incidence (38.33%) with plant mortality up to (12.22%). The average stem rot mortality was comparatively low (16–20%) in summer but increased to 18–26% in winter crop. Several phenolic acids such as as tannic, gallic, caffeic, ferulic and benzoic were detected by using high performance liquid chromatography (HPLC) in variable amounts in these plants. The disease intensity varied from 97.77 to 92.46% from season to season. The significance of the phenolic compounds is discussed in relation to disease prevalence.  相似文献

Callus cultures were established from hypocotyl explants of R. bracteosa, R. chalepensis and R. macrophylla. Calli were maintained for more than three years on MS-medium supplemented with 1 mg l-1 of each 2,4-D and kinetin. Acridone and furoquinoline alkaloids and coumarins have been isolated from four week old calli grown on a hormone containing and hormone-free medium. AIMS: The goal of this study was to examine the growth of Oenococcus oeni in the presence of phenolic compounds under wine conditions and to see how these compounds affect bacterial metabolism. METHODS AND RESULTS: Phenolic compounds have been added to a basal medium that simulates the composition of wine. Fifty milligrams per litre or more of phenolic compounds stimulated bacterial growth. Oenococcus oeni seemed to use citric acid and trehalose, if they were present, before glucose and fructose. Citrate was completely exhausted in three days and the yield of acetate was higher when phenolic compounds were present. CONCLUSIONS: Phenolic compounds reduced the rate of sugar consumption and enhanced citric acid consumption, increasing the yield of acetic acid. SIGNIFICANCE AND IMPACT OF THE STUDY: This study allows a better knowledge of co-metabolism of citric acid and sugars by O. oeni in the presence of phenolic compounds of wine.  相似文献

Abstract

Several 7-chloro-imidazo(1,2-c)pyrimidines were tested on tobacco callus cultures in order to verify a possible cytokinin and anticytokinin-like activity. These compounds were used alone and in combination with kinetin or zeatin.

The aminofurfuryl derivatives showed a strong inhibitory action on cell multiplication and this effect was enhanced when they were mixed with kinetin or zeatin.

The isopentenyl derivatives on the contrary were able to induce cell division in tobacco callus.  相似文献

The mutagenicities of 8 indole compounds (indole-3-acetonitrile, indole-3-carbinol, indole-3-acetamide, indole-3-acetic acid, 3-methylindole, indole-3-aldehyde, indole-3-carboxylic acid and indole) derived from indole glucosinolate were studied by mutation tests on Salmonella typhimurium TA98 and TA100 and Escherichia coli WP2 uvrA/pKM101 with and without S9 mix. None of the 8 indole compounds were mutagenic, but they became mutagenic on these 3 tester strains when treated with nitrite at pH 3. The nitrite-treated indole compounds were mutagenic without metabolic activation system (S9 mix), and their mutagenicities were decreased by the addition of S9 mix.  相似文献

The influence of culture medium and explant on callus and shoot formation of lentil (Lens culinaris Medik.) has been studied. Three different explants (shoot-tip, first node and first pair of leaves) from three Spanish lentil cultivars were cultivated on two basal media: Murashige and Skoog medium (MS) and medium with mineral salts of MS medium plus vitamins of Gamborg's B5 medium (MSB), supplemented with growth regulators. Media with 2,4-D induced the formation of calli in all explants, but no organ regeneration was obtained from these calli. Multiple shoot formation was obtained from 33% to 92% of the explants in media supplemented with 2.25 mg l–1 of BA and 0.186 mg l–1 NAA+2.25 mg l–1 BA; in the other media one to two shoots per explant were formed in 10 to 98% of the explants. Root formation from explants was achieved only in media with NAA or IAA. Of the explants tested, the best morphogenetic responses were obtained from nodes and the poorest from leaves.  相似文献

The leaves of Cyclosorus extensa are used in the preparation of rice beer in Assam, India. The optimal conditions of time and temperature of fermentation for extraction of bioactive compounds from the dried leaves were obtained using response surface methodology. The central composite rotatable design was used and 13 experimental runs based on two-factor-five-level design were generated and performed for each of the solvents. The independent variables were extraction time (12 and 48?h) and temperature (25 and 55°C). The responses studied were total polyphenol content, radical scavenging activity, antibacterial activity, and antifungal activity. The analysis of variance of the test data was performed and the sequential sum of squares, F-value, R2, and adjusted R2 were deduced. The predicted models for all the response variables were adequately fitted to the observed experimental data (p?≤?0.001). The maximum extraction of bioactive compounds under the optimum conditions of extraction temperature and time for hexane, ethyl acetate, methanol, and distilled water were found to be 25°C for 29.43?h, 28.28°C for 41.27?h, 43.95°C for 29.61?h, and 55.00°C for 48.00?h, respectively. It was also observed that the solubility of the polyphenols was higher in methanol, followed by ethyl acetate, and the highest antibacterial activity against Escherichia coli was shown by the ethyl acetate extracts.  相似文献
