Through semi-interpenetration of polymer networks with poloxamer, mechanical properties of chitosan (CS) sponge were increased for wound dressing application. Synthesis of poloxamer macromer was confirmed by proton nuclear magnetic resonance (1H NMR) spectra. Possible interactions between CS and poloxamer in semi-interpenetrating polymer networks (SIPNs), and changes in crystalline structures of both polymers were evaluated by Fourier-transform infrared spectroscopy (FTIR) and X-ray diffraction (XRD), respectively. Swelling behavior, thermal analysis, mechanical properties, and morphology of SIPNs were studied by thermal gravimetric analysis, differential scanning calorimetry (DSC), compressive modulus measurement, and scanning electron microscopy (SEM), respectively. Preparation of poloxamer macromer, and intermolecular hydrogen bonding between CS and poloxamer were confirmed by NMR and FTIR, respectively. Melting temperature of poloxamer in SIPNs decreased due to prevention of crystallization by incorporation of CS. Formation of SIPNs with poloxamer and increasing poloxamer content in CS/poloxamer SIPNs increased mechanical strength of CS sponge compared with CS/poloxamer blend. Formation of SIPNs with poloxamer remarkably increased water content of CS due to hydrophilicity of CS and poloxamer. These results suggest CS/poloxamer sponges prepared by SIPNs method have good possibility for wound dressing application owing to rapid water adsorption, high mechanical strength, and interconnected cross-sectional morphology of SIPNs.  相似文献

γ-Irradiation combined with freeze-thawing, i.e. irradiation followed by freeze-thawing and freeze-thawing followed by irradiation, was applied to prepare poly(vinyl alcohol) (PVA)/water soluble chitosan (ws-chitosan) hydrogels for wound dressing. The properties of these hydrogels were investigated and compared to those prepared by freeze-thawing and by irradiation, respectively. Hydrogels made by irradiation followed by freeze-thawing show larger swelling capacity and mechanical strength, higher thermal stability, lower water evaporation rate, and are less turbid than those made by pure freeze-thawing and freeze-thawing followed by irradiation. Hydrogels made by irradiation alone cannot be used as wound dressing due to their poor mechanical strength. SEM results show that the final structure of hydrogels made by combined irradiation and freeze-thawing is mainly determined by the first processing step. It is found that the appropriate amount of ws-chitosan can endow hydrogels with large swelling capacity and mechanical strength. The presence of ws-chitosan provides the hydrogels with good antibacterial activity against Escherichia coli (E. coli).  相似文献

目的:制备壳聚糖/魔芋葡甘露聚糖复合膜,研究其促创面愈合作用。方法:壳聚糖溶液和魔芋葡甘露聚糖溶液混合后冷冻干燥制成复合膜。扫描电镜观察膜的形态和孔径,并研究比较膜的吸水率,水蒸气透过率,拉伸强度,断裂伸长率和体外降解率。建立大鼠皮肤损伤模型,敷以复合膜治疗,比较创面愈合率,观察创面组织染色结果,评价复合膜的促创面愈合作用。结果:壳聚糖/魔芋葡甘露聚糖复合膜具有三维网状结构,壳聚糖复合魔芋葡甘露聚糖后,膜的吸水率、拉伸强度和断裂伸长率提高,体外降解加速,水蒸气透过率改善。愈合实验表明壳聚糖/魔芋葡甘露聚糖膜具有促进创面愈合作用。结论:壳聚糖/魔芋葡甘露聚糖复合膜制备工艺简单,能有效促进创面愈合,具有成为创伤敷料的潜力。  相似文献

The positive interaction between polysaccharides with active phytochemicals found in medicinal plants may represent a strategy to create active wound dressing materials useful for skin repair. In the present work, blended membranes composed of chitosan (Cht) and aloe vera gel were prepared through the solvent casting, and were crosslinked with genipin to improve their properties. Topography, swelling, wettability, mechanical properties and in vitro cellular response of the membranes were investigated. With the incorporation of aloe vera gel into chitosan solution, the developed chitosan/aloe-based membranes displayed increased roughness and wettability; while the genipin crosslinking promoted the formation of stiffer membranes in comparison to those of the non-modified membranes. Moreover, in vitro cell culture studies evidenced that the L929 cells have high cell viability, confirmed by MTS test and calcein-AM staining. The findings suggested that both blend compositions and crosslinking affected the physico-chemical properties and cellular behavior of the developed membranes.  相似文献

Chitin and chitosan are novel biomaterials. The novel chitosan/gelatin membranes were prepared using the suspension of chitosan hydrogel mixed with gelatin. The prepared chitosan/gelatin membranes were characterized by scanning electron microscopy (SEM), X-ray diffraction (XRD), mechanical, swelling, and thermal studies. The morphology of these chitosan/gelatin membranes was found to be very smooth and homogeneous. The XRD studies showed that the chitosan/gelatin membranes have good compatibility and interaction between the chitosan and gelatin. The stress and elongation of chitosan/gelatin membranes on wet condition showed excellent when the mixture ratio of gelatin was 0.50. The prepared chitosan/gelatin membranes showed good swelling, mechanical and thermal properties. Cell adhesion studies were also carried out using human MG-63 osteoblast-like cells. The cells incubated with chitosan/gelatin membranes for 24 h were capable of forming cell adhesion. Thus the prepared chitosan/gelatin membranes are bioactive and are suitable for cell adhesion suggesting that these membranes can be used for tissue-engineering applications. Therefore, these novel chitosan/gelatin membranes are useful for biomedical applications.  相似文献

Chitosan is a well sought-after polysaccharide in biomedical applications and has been blended with various macromolecules to mitigate undesirable properties. However, the effects of blending on the unique antibacterial activity of chitosan as well as changes in fatigue and degradation properties are not well understood. The aim of this work was to evaluate the anti-bacterial properties and changes in physicochemical properties of chitosan upon blending with synthetic polyester poly(epsilon-caprolactone) (PCL). Chitosan and PCL were homogeneously dissolved in varying mass ratios in a unique 77% acetic acid in water mixture and processed into uniform membranes. When subjected to uniaxial cyclical loading in wet conditions, these membranes sustained 10 cycles of predetermined loads up to 1 MPa without break. Chitosan was anti-adhesive to Gram-positive Streptococcus mutans and Gram-negative Actinobacillus actinomycetemcomitans bacteria. Presence of PCL compromised the antibacterial property of chitosan. Four-week degradation studies in PBS/lysozyme at 37 degrees C showed initial weight loss due to chitosan after which no significant changes were observed. Molecular interactions between chitosan and PCL were investigated using Fourier transform infrared spectroscopy (FTIR) which showed no chemical bond formations in the prepared blends. Investigation by wide-angle X-ray diffraction (WAXD) indicated that the crystal structure of individual polymers was unchanged in the blends. Dynamic mechanical and thermal analysis (DMTA) indicated that the crystallinity of PCL was suppressed and its storage modulus increased with the addition of chitosan. Analysis of surface topography by atomic force microscopy (AFM) showed a significant increase in roughness of all blends relative to chitosan. Observed differences in biological and anti-bacterial properties of blends could be primarily attributed to surface topographical changes.  相似文献

The material behaviour and antimicrobial effect of konjac glucomannan edible film incorporating chitosan and nisin at various ratio or concentrations is discussed. This activity was tested against food pathogenic bacteria namely Escherichia coli, Staphylococcus aureus, Listeria monocytogenes, and Bacillus cereus. Mechanical and physical properties were determined and the results indicated that the blend film KC2 (mixing ratio konjac glucomannan 80/chitosan 20) showed the maximum tensile strength (102.8 ± 3.8 MPa) and a good transparency, water solubility, water vapor transmission ratio. The differential scanning calorimetry (DSC), Fourier transform infrared spectroscopy (FTIR), etc. were used to characterize the structural change of the blend films. The results showed that the strong intermolecular hydrogen bonds took place between chitosan and konjac glucomannan. Incorporation of nisin at 42,000 IU/g of film for the selected blend film KC2 was found to have antimicrobial activity against S. aureus, L. monocytogenes, and B. cereus. The antimicrobial effect of chitosan or KC2 incorporating nisin was much better than that of konjac glucomannan incorporating nisin at each corresponding concentration and existed significant difference (p < 0.05), however, there was no significant difference on the antimicrobial effect between chitosan and KC2 both incorporating nisin. At all these levels, the ternary blend film KC2-nisin had a satisfactory mechanical, physical properties and antimicrobial activity, and could be applied as a potential ‘active’ packaging material.  相似文献

Collagen (Col)–chitosan (Chi) membrane was modified by a hot dehydrogenation cross-linking method. Carbodiimide was added for further crossing modification. Chondroitin sulfate (CS) was added so that Col–Chi sulfate composite membranes were prepared. The structure of the composite membranes was characterized by Fourier transform infrared spectroscopy and X-ray photoelectron spectroscopy, and its mechanical properties, degradation, and cytotoxicity were characterized. The composite membrane was applied to a full-thickness skin injury in animal experiments performed in rabbits. Strong interactions and good compatibility among Col, Chi, and CS in the composite membrane were present. The good mechanical properties, biocompatibility, digestion resistance, and wound healing promotion of the composite membrane make it a potential wound dressing or skin scaffold for tissue engineering.  相似文献

A novel antibacterial film was prepared by blending konjac glucomannan (KGM) and poly(diallydimethylammonium chloride) (PDADMAC) in an aqueous system. The antibacterial activity of the films against Staphylococcus aureus, Bacillus subtilis, Escherichia coli, Pseudomonas aeruginosa, and Saccharomyces were measured by the halo zone test and the double plate method. The films exhibited an excellent antibacterial activity against B. subtilis and S. aureus but not against E. coli, P. aeruginosa or Saccharomyces. The miscibility, morphology, thermal stability, water vapour permeability and mechanical properties of the blend films were investigated by density determination, SEM, ATR-IR, XRD, DSC, TGA, WVA and tensile tests. The results of density determination predicted that the blends of KGM and PDADMAC were miscible when the PDADMAC content was less than 70 wt%. Moreover, SEM and XRD confirmed the result. ATR-IR showed that strong intermolecular hydrogen bonds and electrostatic interactions occurred between KGM and PDADMAC in the blends. The tensile strength and the break elongation of the blends were improved largely to 106.5 MPa and 32.04% and the water vapour permeability decreased when the PDADMAC content was 20 wt%. The thermal stability of the blends was higher than pure KGM. The blends should be good antibacterial materials.  相似文献

Material barrier properties to microbes are an important issue in many pharmaceutical applications like wound dressings. A wide range of biomaterials has been used to manage the chronic inflamed wounds. Eight hydrogel membranes of poly vinyl alcohol (PVA) with κ-carrageenan (KC) and Lactobacillus bulgaricus extract (LAB) have been prepared by using freeze–thawing technique. To evaluate the membranes efficiency as wound dressing agents, various tests have been done like gel fraction, swelling behavior, mechanical properties, etc. The antibacterial activities of the prepared membranes were tested against the antibiotic-resistant bacterial isolates. In addition, the safety usage of the prepared hydrogel was checked on human dermal fibroblast cells. The anti-inflammatory properties of the prepared hydrogel on LPS-PBMC cell inflammatory model were quantified using enzyme-linked immunosorbent assay (ELISA) and real-time polymerase chain reaction (RT-qPCR). The analysis data of TGA, SEM, gel fraction, and swelling behavior showed changes in properties of prepared PVA\KC\LAB hydrogel membrane than pure PVA hydrogel membrane. The antibacterial activities of the prepared membranes augmented in LAB extract-prepared membranes. Out of the eight used hydrogel membranes, the PVAKC4 hydrogel membrane is the safest one on fibroblast cellular proliferation with a maximum proliferation percentage 97.3%. Also, all the used hydrogel membrane showed abilities to reduce the concentration of IL-2 and IL-8 compared with both negative and positive control. In addition, almost all the prepared hydrogel membrane showed variable abilities to downregulate the expression of TNF-α gene with superior effect of hydrogel membrane KC1. PVA/KC/LAB extract hydrogel membrane may be a promising material for wound dressing application and could accelerate the healing process of the chronic wound because of its antimicrobial and anti-inflammatory properties.  相似文献

Biocompatible carboxyethyl chitosan/poly(vinyl alcohol) (CECS/PVA) nanofibers were successfully prepared by electrospinning of aqueous CECS/PVA solution. The composite nanofibrous membranes were subjected to detailed analysis by scanning electron microscopy (SEM), differential scanning calorimetry (DSC), and X-ray diffraction (XRD). SEM images showed that the morphology and diameter of the nanofibers were mainly affected by the weight ratio of CECS/PVA. XRD and DSC demonstrated that there was strong intermolecular hydrogen bonding between the molecules of CECS and PVA. The crystalline microstructure of the electrospun fibers was not well developed. The potential use of the CECS/PVA electrospun fiber mats as scaffolding materials for skin regeneration was evaluated in vitro using mouse fibroblasts (L929) as reference cell lines. Indirect cytotoxicity assessment of the fiber mats indicated that the CECS/PVA electrospun mat was nontoxic to the L929 cell. Cell culture results showed that fibrous mats were good in promoting the cell attachment and proliferation. This novel electrospun matrix would be used as potential wound dressing for skin regeneration.  相似文献

We previously proposed a mechanism for the structural formation of cellulose from its solution using a molecular dynamics (MD) simulation and suggested that the initial structure from its solution plays a critical role in determining its final structure. Structural changes in the van der Waals-associated cellulose molecular sheet as the initial structure were examined by MD simulation; the molecular sheet was found to be disordered due to maltohexaoses as an amylose model in terms of the hydrogen bonding system of cellulose. The structure and properties of cellulose/amylose blends prepared from an aqueous NaOH solution were examined experimentally by wide-angle X-ray diffraction and dynamic viscoelasticity measurements. The crystallinity of cellulose in the cellulose/amylose blend films was lower than that of cellulose film. The diffraction peaks of the cellulose/amylose blends were slightly shifted; specifically, () was shifted to a higher angle, and (1 1 0) and (0 2 0) were shifted to lower angles. These experimental results probably resulted from the disordered molecular sheet, as revealed by MD simulations.  相似文献

Nonwoven fiber mats of chitosan with potential applications in air and water filtration were successfully made by electrospinning of chitosan and poly(ethyleneoxide) (PEO) blend solutions. Electrospinning of pure chitosan was hindered by its limited solubility in aqueous acids and high degree of inter- and intrachain hydrogen bonding. Nanometer-sized fibers with fiber diameter as low as 80 +/- 35 nm without bead defects were made by electrospinning high molecular weight chitosan/PEO (95:5) blends. Fiber formation was characterized by fiber shape and size and was found to be strongly governed by the polymer molecular weight, blend ratios, polymer concentration, choice of solvent, and degree of deacetylation of chitosan. Weight fractions of polymers in the electrospun nonwoven fibers mats were determined by thermal gravimetric analysis and were similar to ratio of polymers in the blend solution. Surface properties of fiber mats were determined by measuring the binding efficiency of toxic heavy metal ions like chromium, and they were found to be related with fiber composition and structure.  相似文献

为了进一步提高伤口敷料的止血性能,文中在生物相容性良好的壳聚糖溶液中引入含有多种生长因子的人源性富血小板血浆(Humanplatelet-richplasma,hPRP),并加入不同体积比例(1∶1、1∶3、3∶1、1∶0)的丝素蛋白溶液以提高材料的多孔性与止血性,通过冷冻干燥法制备不同配比的hPRP-壳聚糖/丝素蛋白敷料,并将纯壳聚糖敷料作为对照组,研究hPRP和丝素蛋白对敷料的止血性能的影响以及丝素蛋白对PRP中生长因子控制释放的影响。结果表明,在壳聚糖敷料中引入hPRP对敷料的止血性有所提高,但对敷料的多孔结构及吸水率无明显改善,若在hPRP-壳聚糖溶液中按照体积比为1∶1的比例加入丝素蛋白溶液,会得到具有较为均匀的多孔结构的敷料,敷料的孔隙率与吸水率分别可达到86.83%±3.84%与1 474%±114%,且该比例的敷料在快速止血性能上表现优异。此外,加入丝素蛋白与壳聚糖比例为1∶1的PRP敷料能有效减少PRP中生长因子在初始阶段的爆裂释放。因此,含hPRP的壳聚糖/丝素蛋白复合敷料有望成为一种能快速止血且能促进伤口愈合的新型伤口敷料。  相似文献

Crystallization behavior and environmental biodegradability were investigated for the films of bacterial poly(3-hydroxybutyric acid) (PHB) blends with chitin and chitosan. The blend films showed X-ray diffractive peaks that arose from the PHB crystalline component. It was suggested that the lamellar thickness of the PHB crystalline component in the blends was large enough to show detectable X-ray diffractive peaks, but this was too small to show observable melting endotherm in the DSC thermogram and the crystalline band absorption in the FT-IR spectrum. In the PHB/chitin and PHB/chitosan blends, thermal transition temperatures of PHB amorphous region observed by dynamic mechanical thermal analysis were almost the same as that of neat PHB. Both the PHB/chitin and the PHB/chitosan blend films biodegraded in an environmental medium. Several blend films showed faster biodegradation than the pure-state component polymers.  相似文献

In this paper, hydrophilic polymer membranes based on macromolecular chitosan networks have been synthesized and characterized. The structure of the membrane has been altered in several ways during the formation to adjust the properties, particularly with regard to the elasticity, tensile strength, permeability, and surface structure. An alteration of the network structure was achieved by addition of flexibilizer, cross-linking with dialdehydes, symplex formation of the chitosan with the polyanion sulfoethyl cellulose, and the introduction of artificial pores on the micro- and nanometer scale into the chitosan matrix with silica particles or poly(ethylene glycol). The resulting network structures and morphologies of these unique membranes that combine the novel alteration techniques have been characterized in detail and correlated with molecular parameters of the chitosan as degree of deacetylation, molar mass, and charge density. Finally, we report on the impact of the new network structures on physical properties of the membranes, the water vapor and gas permeability and the tensile strength, to evaluate possible application of the membranes as a wet wound dressing material with microbial barrier function that actively assists the healing process of problematic wounds. Wound dressing is one of the most promising medical applications for chitin and chitosan. The adhesive nature of chitin and chitosan, together with their antifungal and bactericidal character, and their permeability to oxygen, is a very important property associated with the treatment of wounds and burns. Different derivatives of chitin and chitosan have been prepared for this purpose in the form of hydrogels, fibers, membranes, scaffolds and sponges. The purpose of this review is to take a closer look on the wound dressing applications of biomaterials based on chitin, chitosan and their derivatives in various forms in detail.  相似文献

Different molecular weight, 90% deacetylated chitosans were obtained by ultrasonic degradation on 90% deacetylated chitosan at 80 °C for various times.

Ninety percent deacetylated chitosan was prepared from alkali treatment of chitin that was obtained from red shrimp waste. Number average-, viscosity average- molecular weights were measured by gel permeation chromatography and the viscometric method, respectively. Degree of deacetylation was measured by the titration method. Enthalpy, maximum melting temperature, tensile strength and elongation of the membranes, flow rate of permeates and water are properties measured to elucidate the effect of molecular weight of chitosan on the above thermal, mechanical, and permeation properties, respectively of the prepared membranes. Results show tensile strength, tensile elongation, and enthalpy of the membrane prepared from high molecular weight chitosans were higher than those from low molecular weight. However, the permeability show membranes prepared from high molecular weight chitosans are lower than that from those of low molecular weight.  相似文献

Ideal wound dressing materials should create a good healing environment, with immediate hemostatic effects and antimicrobial activity. In this study, chitosan/konjac glucomannan (CS/KGM) films embedded with gentamicin-loaded poly(dex-GMA/AAc) nanoparticles (giving GNP-CS/KGM films) were prepared as novel wound dressings. The results revealed that the modified CS/KGM films could be used as effective wound dressings and had significant hemostatic effects. With their microporous structure, the films could effectively absorb water from blood and trap blood cells. The gentamicinloaded poly(dex-GMA/AAc) nanoparticles (GNPs) also further promoted blood clotting, with their favorable water uptake capacity. Thus, the GNP-CS/KGM films had wound healing and synergistic effects that helped to stop bleeding from injuries, and also showed good antibiotic abilities by addition of gentamicin to the NPs. These GNPCS/KGM films can be considered as promising novel biodegradable and biocompatible wound dressings with hemostatic capabilities and antibiotic effects for treatment of external bleeding injuries.  相似文献

Chitosan cross-linked cellulose fibers were prepared using non-toxic procedures in order to confer antimicrobial properties to cellulose fibers. Citric acid was used as the cross-linker and NaH2PO4 as catalyst in previously UV-irradiated cellulose fibers. Further heat dried-cure process and washing with detergent, water and acetic acid (0.1 M) gave a maximum incorporation of chitosan of 27 mg per gram of functionalized textile. The thermogravimetric analysis of the material with the highest chitosan content showed an increased thermal stability compared to cellulose and chitosan. The UV-irradiation induced morphological changes, such as less entangled cellulose fibers, as observed by scanning electron microscopy, which was prompted to enhance the chitosan incorporation. The biomass and spore germination percentage of Penicillium chrysogenum and colony forming units per millilitre for Escherichia coli decreased significantly on the composed materials as compared to raw cellulose fiber and it was similar to that obtained with a commercial antimicrobial cellulose fiber.  相似文献
