【背景】细菌性疾病是林麝规模化养殖的重要制约因素,蜡样芽孢杆菌曾在林麝化脓灶中检出,但是目前对林麝源蜡样芽孢杆菌的研究报道很少。【目的】对分离自病死林麝肝脏中的一株疑似蜡样芽孢杆菌进行分离鉴定和全基因组序列分析,为林麝相关疾病的防治奠定基础。【方法】将病原菌纯化培养后,对病原菌进行生化试验、药敏试验和小鼠致病性试验;并通过第3代单分子测序技术进行全基因组测序,根据测序结果进一步评估物种间的亲缘关系,并进行基因功能注释和遗传进化分析。【结果】该病原菌经平均核苷酸相似度分类和系统发育树分析属于蜡样芽孢杆菌群,生化结果符合蜡样芽孢杆菌的一般特征,将分离菌株命名为SCBCM001。该菌株对小鼠的半数致死量为8.3×107 CFU,对大多数β-内酰胺类、四环素和磺胺异噁唑耐药,对氨基糖苷类、头孢氨苄、头孢哌酮和亚胺培南等药物敏感;全基因组测序结果表明该菌株的染色体大小为5292570bp,GC含量为35.37%,多位点序列分型显示该菌株属于ST427序列类型。在菌株SCBCM001基因组内发现hblA、hblC、hblD、nheA、nheB、clo和cytK等多种毒力因子,同时菌株携带对β-内酰...  相似文献

【背景】鸭源鸡杆菌作为一种条件致病菌能引起家禽卵巢炎、输卵管炎和腹膜炎等疾病,严重威胁养殖业的发展。【目的】四川某养鸡场送检了一批疑似感染鸭源鸡杆菌的病死鸡,为探究其感染机制与防治方法,对该菌进行分离鉴定及全基因组测序分析。【方法】从病料中分离并纯化细菌,再依次进行生化试验、16S rRNA基因序列分析和药敏试验,同时通过全基因组测序对其进行物种分型与毒力、耐药等基因功能注释及遗传进化分析。【结果】该分离菌被鉴定为鸭源鸡杆菌,菌株命名为TS0001,药敏试验显示其仅对硝基呋喃类和少数β-内酰胺类药物敏感,对部分β-内酰胺类、氯霉素、部分氨基糖苷类、大环内酯类、四环素类和磺胺类药物具有耐药性。全基因组序列长度为2 626 722 bp,蛋白质编码基因功能注释显示其有较强的自我加工修饰能力,全基因组注释到83个与毒力因子和耐药性相关的基因,包含4个前噬菌体区域。序列类型分析结果显示,该菌株为ST69型,而且管家基因联合建树表明其与墨西哥普通家鸡分离株7990一致性最高。【结论】本研究为鸭源鸡杆菌的感染机制与防治研究提供了参考,丰富了后续研究的分子生物学背景。  相似文献

【背景】多杀性巴氏杆菌可导致猪肺疫、牛出血性败血症和兔出血性败血症等多种疾病,严重威胁多种动物畜牧养殖业的健康发展。【目的】重庆某兔场送检一批病死兔,为研究其病原和治疗方法,对病原进行了微生物分离和全基因组测序分析。【方法】从2022年重庆某兔场送检兔病料中进行细菌分离纯化、生化试验、16S rRNA基因鉴定、荚膜血清型分型、药敏试验和毒力基因检测,同时通过全基因组测序结果进行毒力、耐药基因注释和遗传进化等分子生物学信息分析。【结果】该菌为兔源A:ST74多杀性巴氏杆菌,命名为LXSS001,基因组序列上传到NCBI数据库(登录号为CP119523.1),药敏试验显示该菌对四环素、杆菌肽、复方新诺明和磺胺异恶唑耐药,对头孢噻肟、头孢哌酮和丁胺卡那等药物敏感。全基因组长度为2 480 671 bp,并注释到了58个毒力基因和9类药物的靶向抗药基因。通过联合建树表明其与3480株一致性最高。【结论】本研究完成了一株A型多杀性巴氏杆菌的分离鉴定和全基因组测序,并揭示了其与国内外其他分离株的进化关系,为多杀性巴氏杆菌的后续研究提供了参考依据。  相似文献

【背景】沙门氏菌(Salmonella spp.)是重要的人畜共患病原菌,其毒力和耐药性的不断增强引起广泛关注。【目的】了解从通辽市一犊牛死亡病例中所分离牛源都柏林沙门氏菌的毒力及耐药性情况。【方法】以病死犊牛肺脏为材料,经细菌分离纯化及16S rRNA基因测序,鉴定病原为沙门氏菌。采用动物试验、药敏试验和PCR方法对分离菌进行毒力、耐药性,以及毒力基因和耐药基因检测,并对其进行全基因组测序分析。【结果】分离菌具有较强毒力,对小鼠半数致死量为2.8×106 CFU/mL。分离菌为多重耐药菌,仅对多粘菌素B和噻孢霉素敏感,对强力霉素和恩诺沙星中度敏感。检测13种沙门氏菌常见毒力基因,检出率为92.3%。对分离菌进行全基因组测序分析,该菌株为都柏林沙门氏菌,基因组大小为4 965 370 bp,GC含量为52.12%,同时携带2个质粒,大小分别为79 524 bp (pTLS-1)和45 301 bp (pTLS-2)。分离菌中共携带996个毒力基因和24个毒力岛;共携带42个耐药基因,其中4个为可水平转移基因,基因组中存在9个可移动遗传元件,包括插入序列和转座子等。【结论】分离牛源都柏林沙门氏菌菌株具有较强毒力且为多重耐药株,携带大量毒力基因及耐药基因。  相似文献

【目的】构建猪支气管败血波氏杆菌(Bordetella bronchiseptica,Bb)Ⅵ型分泌系统(T6SS)溶血素共调节蛋白hcp基因缺失株,并对其基本生物学特性进行初步的研究。【方法】使用自杀性质粒介导同源重组的方法敲除猪支气管败血波氏杆菌QH0814菌株hcp基因,并比较hcp基因缺失前后,菌体对细胞的黏附入侵、小鼠毒力及组织载菌量上的差异。【结果】成功构建支气管败血波氏杆菌hcp基因缺失株QH0814Δhcp,连续传50代且遗传稳定;缺失株与亲本株生长无明显差异;缺失株的黏附能力与亲本株差异不显著,但入侵能力显著降低(P0.05);与亲本株相比,半数致死量提高,同时,缺失株对昆明鼠的感染能力也显著降低(P0.05)。【结论】hcp基因的缺失对支气管败血波氏杆菌增殖无影响,但缺失后其入侵能力和定殖能力显著降低,由此推测hcp基因与支气管败血波氏杆菌的入侵和定殖相关。  相似文献

目的对1例脓疱疮患者脓液中分离的1株社区获得性耐甲氧西林金黄色葡萄球菌(CA-MRSA)HZW450菌株进行全基因组测序并对序列信息解析。方法使用三代测序PacBio技术对HZW450菌株进行全基因组测序,且使用SMRTanalysis v.R2.3.0对该序列进行序列拼接,上传NCBI进行基因功能注释,用RPSBLAST program进行直系同源簇(COG)注释以及毒力因子相关基因和耐药基因分析。同时比较国内其他地区发现的同一序列型(ST)的菌株毒力因子相关基因及耐药相关基因的差异,并比较HZW450菌株耐药基因型与表型的一致性。结果 HZW450菌株的基因组大小为2 831 958bp,GC占比32.9%,其基因组完成图序列已提交至NCBI GenBank数据库,登录号为CP020741。同时经过分析发现该菌株为ST59型,其基因组中含有许多与致病相关的毒力因子,以及含有耐药基因aph(3′)-III、ant(6)-Ia、mecA、norA、erm(B)和cat(pC233),毒力及耐药基因与ST59型其他菌株比较有差异。该菌株临床药敏结果与耐药基因比较分析发现,耐药表型与基因型存在差异。结论本研究报道了1株CA-MRSA(HZW450)菌株的全基因组序列。基因序列分析显示该菌株携带大量毒力基因,包括lukS-PV、lukF-PV、eta、fnbA、fnbB、sspB、sspC等,编码毒素、粘附、免疫逃逸等相关毒力因子,其毒力较强,致病性较高。  相似文献

【背景】弯曲菌(Campylobacter)是一种世界范围内能引起胃肠炎的最常见食源性病原菌,对临床上重要的抗菌药物耐药越来越严重,对食品安全和公共健康造成重大威胁。【目的】研究一株同时携带optr A和cfr C基因的猪源结肠弯曲菌(Campylobactercoli)耐药表型和耐药基因,同时对该菌全基因组特征、毒力基因分布情况以及optr A和cfr C基因环境进行分析。【方法】采用琼脂平板稀释法进行最低抑菌浓度(minimal inhibitory concentration, MIC)测定,并且对该菌进行全基因组测序。【结果】该菌株对四环素、克林霉素、阿奇霉素、氟苯尼考和利奈唑胺呈现高度耐药,对环丙沙星和庆大霉素敏感。全基因组测序得到一条大小为1 436 486 bp的环状DNA (GC含量为31.63%),携带四大类抗生素中共计12种耐药基因,均定位于染色体上,其中氨基糖苷类耐药基因数量最多。此外,携带包含黏附、侵袭和移动等相关毒力基因83个,其中与移动相关的毒力基因数量最多。对4个基因岛分析发现,基因岛GIs002和GIs003中含有耐药基因序列,cfr C位于基因岛GIs...  相似文献

【背景】2021年6月,广东省茂名市某散养户送检了一头发病仔猪,猪身上长有脓疱,四肢关节肿大,关节内可见脓液。【目的】确定引起仔猪发病的病原菌,分析其药物敏感性,为临床用药提供指导;对分离菌株进行全基因组序列分析,挖掘其毒力因子和耐药基因,揭示该菌致病和耐药的分子机制。【方法】取关节脓液分离细菌;通过革兰氏染色、16S rRNA基因和全基因组测序分析,鉴定细菌种类;通过溶血试验、血浆凝固酶试验和生长曲线测定,确定分离菌株的溶血活性、血浆凝固酶活性和生长特性;用小鼠感染模型评估分离菌株的致病性;用纸片扩散法测定分离菌株的药物敏感性;通过全基因组序列分析挖掘分离菌株的毒力因子和耐药基因。【结果】分离菌株被鉴定为猪葡萄球菌(Staphylococcus hyicus);该菌不溶血,无血浆凝固酶活性,在胰蛋白胨大豆肉汤培养基中于37℃、120r/min条件下生长良好;小鼠感染试验结果显示,该菌具有高致病性;药敏试验结果显示,该菌对苯唑西林、大观霉素等7种药物敏感,对青霉素G、红霉素等9种药物耐药;全基因组序列分析结果显示,该菌携带多个毒力因子和耐药基因。【结论】从发病仔猪的关节脓液中分离到一株猪葡萄球菌,可用苯唑西林、大观霉素等药物防控该菌感染;解析了该菌的基因组信息,为后续深入研究该菌致病和耐药的分子机制奠定了基础。  相似文献

【目的】创伤弧菌是致死率最高的弧菌物种,但目前尚无在全基因组层面挖掘毒力相关因子的研究。本研究以创伤弧菌分离来源(临床和环境)作为不同表型,通过与260株基因组序列进行关联分析,挖掘毒力相关因子,从而进一步了解创伤弧菌致病因素。【方法】对139株创伤弧菌分离株进行高通量测序,获取其全基因组序列;与公共数据库已公开发表的121株基因组整合,使用pyseer软件进行全基因组关联分析,对与不同分离来源显著相关的基因进行注释和解读。【结果】共发现11个基因与临床分离株显著相关,其中9个是本研究新发现的创伤弧菌潜在毒力相关因子。【结论】本研究使用群体基因组学和统计遗传学方法,在全基因组范围扫描挖掘了创伤弧菌毒力相关因子,为深入揭示该物种致病机制、设计新的疫苗和治疗靶点提供了重要依据。  相似文献

【目的】探讨鼠衣原体(Chlamydia muridarum,Cm)标准株与减毒株全基因组序列中存在的差异,筛选毒力基因并建立不同基因型的单克隆菌株,为后续致病机制的研究奠定基础。【方法】将Cm标准株G0和减毒株G28以高通量测序法进行全基因组测序,通过全基因组序列比对分析并筛选潜在的毒力相关基因;经空斑形成实验(Plaque assay)在混合菌G0和G28中大量挑取空斑,以毒力靶基因的PCR测序鉴定从G0和G28中筛选含有不同毒力基因型的单克隆菌株。【结果】全基因测序结果显示Cm G0与G28的TC0412、TC0237和TC0668基因明显不同。通过挑取空斑初步筛选了111个空斑样品,通过3个毒力基因的PCR测序鉴定最终获得了G0和G28来源的56个单克隆菌株,并根据TC0412蛋白型分成B3、D1和E1三组,每组中含有TC0237和TC0668基因差异性菌株。【结论】Cm的致病能力可能与TC0412、TC0237和TC0668基因有关,筛选获得的单克隆菌株通过分组匹配后可用于后续的毒力基因功能研究。  相似文献

In order to dissect the genetic regulation of leafblade morphogenesis, 16 genotypes of pea, constructed by combining the wild-type and mutant alleles of MFP, AF, TL and UNI genes, were quantitatively phenotyped. The morphological features of the three domains of leafblades of four genotypes, unknown earlier, were described. All the genotypes were found to differ in leafblade morphology. It was evident that MFP and TL functions acted as repressor of pinna ramification, in the distal domain. These functions, with and without interaction with UNI, also repressed the ramification of proximal pinnae in the absence of AF function. The expression of MFP and TL required UNI function. AF function was found to control leafblade architecture multifariously. The earlier identified role of AF as a repressor of UNI in the proximal domain was confirmed. Negative control of AF on the UNI-dependent pinna ramification in the distal domain was revealed. It was found that AF establishes a boundary between proximal and distal domains and activates formation of leaflet pinnae in the proximal domain.  相似文献

Seven bean rhizobial strains EBRI 2, 3, 21, 24, 26, 27 and 29 identified as Rhizobium etli, and EBRI 32 identified as Rhizobium gallicum, isolated from Egyptian soils and which nodulated Phaseolus vulgaris efficiently, were subjected to hybridization with a nifH probe in order to estimate the copy number of this gene. Seven strains (EBRI 2, 3, 21, 24, 26, 27 and 29) which were only able to nodulate Phaseolus vulgaris, contained three copies of the nifH gene, consistent with their identification as Rhizobium etli bv. phaseoli. Only one strain (EBRI 32) which nodulated both Phaseolus vulgaris and Leucaena leucocephala, had one copy of nifH gene. This confirmed the classification of this strain as Rhizobium gallicum bv. gallicum.  相似文献

Data from microscopic morphology, single-spore cultures, and DNA analyses of teleomorphs and anamorphs support the recognition of five species of Prosthecium with Stegonsporium anamorphs on Acer: P. acerinum sp. nov., the teleomorph of S. acerinum; P. acerophilum comb. nov., formerly known as Dictyoporthe acerophila; P. galeatum comb. nov., originally described as Massaria galeata; P. opalus sp. nov.; and P. pyriforme sp. nov., the teleomorph of S. pyriforme s. str. The morphology of both type specimens and freshly collected material was investigated. The teleomorphs have brown ellipsoidal ascospores with five distosepta and often a longitudinal distoseptum. The anamorphs of all species described here belong to Stegonsporium; their connection to the Prosthecium teleomorphs was demonstrated by morphology and DNA sequences of single spore cultures derived from both ascospores and conidia. The anamorphs and teleomorphs of all five Prosthecium species are described and illustrated by LM images, and a key to these species is provided. As perceived from this work, S. pyriforme is restricted to Europe and does not occur in North America, whereas S. acerinum is restricted to North America, not found in Europe. The host associations given in the literature are revised and evidence is provided that only A. opalus, A. pseudoplatanus, and A. saccharum are confirmed hosts of Prosthecium with Stegonsporium anamorphs. Molecular phylogenetic analyses of tef1, ITS rDNA, and partial nuLSU rDNA sequences confirm that the species with Stegonsporium anamorphs are closely related to P. ellipsosporum, the generic type species. Stilbospora macrosperma is confirmed as the anamorph of P. ellipsosporum by DNA data of single spore isolates obtained from both ascospores and conidia.  相似文献

Many Cola plant species are endemic to West and Central Africa. Cola acuminata and Cola nitida are used as masticatory when fresh, while the dried nuts are used for beverages and pharmaceutical purposes in Europe and North America. Garcinia kola seeds, that serve as a substitute for the true kola nuts, are used in African traditional medicine for the treatment of various diseases, including colic, headache and liver cirrhosis. Seeds extracts of G. kola are also known for their anti-inflammatory, antimicrobial and antiviral properties. To gain information on the chemical properties of the kolas, we have isolated and analyzed cell wall polysaccharides, arabinogalactan-proteins and phenolic substances from the seeds of the three kola species. The sugar composition of cell wall material of C. acuminata, C. nitida and G. kola revealed that Gal (up to 30%), Ara, GalA and Glc as the predominant monosaccharides, representing approximately 90% by mol of the total hydrolysable sugar present in this material. In Ammonium oxalate cell wall fraction, GalA was found to be the major sugar present in all kola species. In the alkali-soluble fraction, there were significant differences in the level of Glc and Gal. The level of Glc was high in C. acuminata and C. nitida while the level of Gal and Xyl were high in C. nitida and G. cola. Isolation and quantification of arabinogalactan-proteins demonstrate that G. kola seeds contained four to eight times more of these proteoglycans than the seeds of the other two species. Finally, analysis of soluble phenolic substances shows that caffeine and catechin were largely represented in C. acumina and C. nitida seeds, with caffeine accounting for 50% of all soluble phenolics. These findings indicate that the three Kola seeds are highly enriched in pectins and proteoglycans and that C. acuminata and C. nitida can be used as a possible source of caffeine and catechin.  相似文献

黑粉菌属是Roussel 1806年建立的,全世界记载有三百余种,主要寄生于禾本科,是经济作物及牧草的重要致病菌·长期以来,对黑粉菌的邢子使用过各种名称,如厚垣孢子,冬孢子及黑粉孢子等.本文采用黑粉孢子以区别锈菌的冬孢子. 芳’(1979)在《中国真菌总汇》中列出黑粉菌属五十种及一个变型.作者经过显微结构和超显微结构的研究,承认其中二十九种为正确名称,八种及一变型为异名,顶黑粉菌(Ustilago acrearus Berk.)由于错拼而被废弃.埃地黑粉菌(Ustilago emodensis Berk.)被转移至利罗粉菌属(Liroa).另有十一种黑粉菌因缺少标本留待今后订正.自1979年以后,杨信东(1983)增加黑粉菌属二种我国新纪录,K.范基和郭林(1986)描述一新种,四种新纪录.在本文中,作者描述一新种:鸢尾蒜黑粉(Ustilago ixiolirii Guo L) ,孢子堆生在蒴果内,不开裂,黑色,粉末状.黑粉孢子球形,近球形,稀椭圆形, 12.5-21×10-21μm,黑褐色,壁厚1-1.Sμm,纹饰脑状.是迄今生在石蒜科植物上唯一黑粉菌的种,其它几种黑粉菌均属条黑粉菌属.本文增加七种我国新纪录.共计四十九种,寄生于六科四十四属植物,主要是禾本科和蓼科.这仅是黑粉菌属研究的初步报告,在全国范围内大量采集黑粉菌标本后,作者相信会有更多新种和我国新纪录被发现.利罗黑粉菌属(Liroa)是从黑粉菌属(Ustaligo)分出的,此属为单种属.  相似文献

The present work investigates some probiotic properties of four different microorganisms (Bifidobacterium animalis var. lactis BB-12, Escherichia coli EMO, Lactobacillus casei and Saccharomyces boulardii). In vitro and in vivo tests were carried out to compare cell wall hydrophobicity, production of antagonistic substances, survival capacity in the gastrointestinal tract of germ-free mice without pathological consequence, and immune modulation by stimulation of Küpffer cells, intestinal sIgA and IL-10 levels. In vitro antagonism against pathogenic bacteria and yeast was only observed for the probiotic bacteria B. animalis and L. casei. The hydrophobic property of the cell wall was higher for B. animalis and E. coli EMO, and this property could be responsible for a better ability to colonize the gastrointestinal tract of germ-free mice. Higher levels of sIgA were observed mainly for S. boulardii, followed by E. coli EMO and B. animalis, and only S. boulardii induced a significant higher level of IL-10. In conclusion, for a probiotic use, S. boulardii presented better characteristics in terms of immunomodulation, and B. animalis and L. casei for antagonistic substance production. The knowledge of the different probiotic properties could be used to choice the better microorganism depending on the therapeutic or prophylactic application.  相似文献

The Pax6 genes eyeless (ey) and twin of eyeless (toy) are upstream regulators in the retinal determination gene network (RDGN), which instructs the formation of the adult eye primordium in Drosophila. Most animals possess a singleton Pax6 ortholog, but the dependence of eye development on Pax6 is widely conserved. A rare exception is given by the larval eyes of Drosophila, which develop independently of ey and toy. To obtain insight into the origin of differential larval and adult eye regulation, we studied the function of toy and ey in the red flour beetle Tribolium castaneum. We find that single and combinatorial knockdown of toy and ey affect larval eye development strongly but adult eye development only mildly in this primitive hemimetabolous species. Compound eye-loss, however, was provoked when ey and toy were RNAi-silenced in combination with the early retinal gene dachshund (dac). We propose that these data reflect a role of Pax6 during regional specification in the developing head and that the subsequent maintenance and growth of the adult eye primordium is regulated partly by redundant and partly by specific functions of toy, ey and dac in Tribolium. The results from embryonic knockdown and comparative protein sequence analysis lead us further to conclude that Tribolium represents an ancestral state of redundant control by ey and toy.  相似文献
