This study describes the spermatozoa of the dendrobatids Epipedobates femoralis and Colostethus sp. using light and transmission electron microscopy. Both species possess a biflagellate spermatozoon, an unusual characteristic only previously reported in two anuran species belonging to the families Leptodactylidae and Racophoridae. The acrosomal complex of both species consists of a conical acrosomal vesicle and a subacrosomal cone, both of which cover the anterior portion of the nucleus, but to differing extents. In the midpiece, the centrioles are disposed parallel to each other and to the cell axis and give rise to two axonemes. Two paraxonemal rods were also seen entering the nuclear fossa. Both flagella are surrounded by a single mitochondrial collar. Each flagellum is formed by an axial fiber connected to the axoneme by an axial sheath; juxta-axonemal fibers are absent. Our data seem to support that Epipedobates femoralis should be placed in a separate clade possibly related to Colosthetus and that these two genera may not be monophyletic.  相似文献

We describe the sperm ultrastructure of three species of frogs in the genus Phyllomedusa . According to micrographs, total size of the spermatozoon of Phyllomedusa hypochondrialis is significantly smaller than those of Phyllomedusa bicolor and Phyllomedusa tarsius . The acrosome complex consists of two conical structures covering the nucleus, the acrosome vesicle and the subacrosomal cone. The subacrosomal cone of P. bicolor and P. tarsius is less electron-dense and appears more granular in transverse section than in P. hypochondrialis . In P. bicolor and P. tarsius , the nuclear space is reduced and the subacrosomal cone fills most of the space between the acrosome vesicle and nucleus. The anterior region of the nucleus in the spermatozoa of P. bicolor and P. tarsius ends abruptly, while in P . hypochondrialis it is sharp-ended. In P. bicolor and P. tarsius , the axial fibre is much larger than in P . hypochondrialis . The sperm ultrastructure of Phyllomedusa appears conservative at the intrageneric level. Future studies on the sperm ultrastructure of hylids can provide new insights on the systematics of the group and a larger database for a cladistic analysis.  相似文献

We make detailed comparisons of the ultrastructure of the spermatozoon among three species of the family Hylidae, Hyla pseudopseudis, Scinax rostratus, and S. squalirostris. The acrosome complex consists of two conical structures covering the nuclear rostrum, the acrosome vesicle, and the subacrosomal cone. The nucleus has a moderately condensed chromatin with a conical shape in longitudinal sections and a circular shape in cross-sections. In H. pseudopseudis, mitochondria are numerous and circular, and in S. rostratus and S. squalirostris there are fewer mitochondria that are more elongate in longitudinal and transverse sections. In H. pseudopseudis, the mitochondrial collar starts adjacent to the distal centriole, occupying the whole midpiece, whereas in both Scinax species the mitochondrial collar starts only at the posterior one-third of the midpiece. In both Scinax species, the presence of juxta-axonemal fiber, axial sheath, and axial fiber in the tail are seemingly plesiomorphic characters, widespread among bufonoid frogs. In H. pseudopseudis, however, the absence of axial fiber and axial sheath seems to be derived from the typical bufonoid condition. The differences between Hyla and Scinax sperm endorse the separation of the two genera and suggest that sperm ultrastructure can be a useful tool to investigate relationships at the intrafamily level.  相似文献

Unlike the primitive type of spermatozoon found in most polychaetes, the spermatozoon of Autolytus has a bilateral symmetry with elongated nucleus, and the mitochondria surround the posterior part of the nucleus. A rather large disk-shaped acrosome is situated along one side of the anterior part of the nucleus. From the anterior margin of the distal centriole emerge long striated rootlets, which run along the nuclear envelope to the anterior part of the nucleus. The spermatozoon of Chitinopoma serrula has an elongated, slightly bent nucleus, a thimble-like acrosome apically on the anterior surface of the nucleus, and an elongated middle piece containing 4 rod-like mitochondria developed from spherical mitochondria surrounding the basal part of the tail flagellum. In the spermatozoon of Capitella capitata, both nucleus and middle piece are elongated compared to the primitive type. The large and conical acrosome is placed asymmetrically at the nucleus and consists of an acrosomal vesicle and subacrosomal substance. The greater part of the middle piece forms a collar around the initial part of the tail flagellum. The cytoplasm of the collar contains granular material. One or two small mitochondria lie around the 2 centrioles at the base of the nucleus.

These types of spermatozoa represent early steps in the evolution of modified spermatozoa combined with changed biology of reproduction. The modified spermatozoa are larger than the primitive ones.  相似文献

应用透射电镜对北草蜥精子的超微结构研究结果表明,北草蜥精子头部顶体囊始终呈圆形,由皮质和髓质组成;顶体囊单侧脊的皮质与髓质问具电子透亮区;穿孔器1个,无穿孔器基板;具顶体下腔;细胞核长形,核内小管缺,核前电子透亮区缺,核肩圆。尾部颈段具片层结构。中段短,多层膜结构缺;纵切面上具2层线粒体;横切面上每圈线粒体6个;2组致密体,具连续的环状结构;线粒体与环状结构的排列模式:rs1/mi1、rs2/mi2;纤维鞘伸人中段,具终环。主段前面部分具薄的细胞质颗粒区;纤维3和8至主段前端消失;轴丝呈“9+2”型。蜥蜴科内不同种类的线粒体数目不同,但都具有2组致密体。不同类群蜥蜴的顶体囊、顶体下腔、核前电子透亮区、穿孔器基板、核肩,以及线粒体与致密体的数目和排列方式等精子超微结构特征都为研究蜥蜴的系统发生提供了辅助信息。  相似文献

The spermatozoa of Gymnophiona show the following autapomorphies: 1) penetration of the distal centriole by the axial fiber; 2) presence of an acrosomal baseplate; 3) presence of an acrosome seat (flattened apical end of nucleus); and 4) absence of juxta-axonemal fibers. The wide separation of the plasma membrane bounding the undulating membrane is here also considered to be apomorphic. Three plesiomorphic spermatozoal characters are recognized that are not seen in other Amphibia but occur in basal amniotes: 1) presence of mitochondria with a delicate array of concentric cristae (concentric cristae of salamander spermatozoa differ in lacking the delicate array); 2) presence of peripheral dense fibers associated with the triplets of the distal centriole; and 3) presence of a simple annulus (a highly modified, elongate annulus is present in salamander sperm). The presence of an endonuclear canal containing a perforatorium is a plesiomorphic feature of caecilian spermatozoa that is shared with urodeles, some basal anurans, sarcopterygian fish, and some amniotes. Spermatozoal synapomorphies are identified for 1) the Uraeotyphlidae and Ichthyophiidae, and 2) the Caeciliidae and Typhlonectidae, suggesting that the members of each pair of families are more closely related to each other than to other caecilians. Although caecilian spermatozoa exhibit the clear amphibian synapomorphy of the unilateral location of the undulating membrane and its axial fiber, they have no apomorphic characters that suggest a closer relationship to either the Urodela or Anura.  相似文献

The mature spermatozoon of Admetus pomilio is a spherical cell containing nucleus and tightly coiled flagellum. In early spermatids the Golgi apparatus forms the acrosomal vesicle and at the opposite side the distal centriole gives rise to the axonemal complex of the sperm tail. As the nucleus elongates, chromatin forms twisted filaments and the spermatid nucleus takes on a helical form. Microtubules are juxtaposed with the nucleus envelope, which is separated from a central chromatin mass by an electron lucid region. A long perforatorium, located on the border of the chromatin mass, runs helically in the nucleus from the centriolar region to subacrosomal space. During tail elongation, the anterior part of the axoneme is surrounded by a long, spiral mitochondrial sheath. In the late spermatid, chromatin filaments appear twisted and become aggregated. The nucleus and flagellum undergo further contortions in which the nucleus coils and the flagellum winds up into the body of the cell and coils in a regular fashion. The mitochondrial sheath surrounds about 2/3 of the 9 + 3 axoneme. These features of spermatid ultrastructure resemble those in the primitive Liphistiomorpha.  相似文献

Spermiogenesis, known as spermateleosis in lower vertebrates, is the transformation of the round spermatid into a highly specialized spermatozoon with a species-specific structure. Spermateleosis and sperm morphology of two species of caecilians, Ichthyophis tricolor and Uraeotyphlus cf. narayani, from the Western Ghats of Kerala, India, were studied using light and transmission electron microscopy. Spermateleosis is described in early, mid-, and late phases. During the early phase, the spermatid nucleus does not elongate, but the acrosome vesicle is Golgi-derived and its material is produced as a homogeneous substance rather than as discrete granules. In development of the acrosome, the centrioles shift in position to the lower half of the cell. The acrosomal vesicles take the full shape of the acrosome with the establishment of the perforatorium in midphase. An endonuclear canal develops and accommodates the perforatorium. The incipient flagellum is laid down when the proximal centriole attaches to the posterior side of the nucleus and the distal centriole connects to the proximal centriole, which forms the basal granule of the acrosome. The axial fiber also appears during midphase. The mitochondria shift in position to the posterior pole of the cell to commence establishment of the midphase. Late phase is characterized by nuclear condensation and elongation. Consequently, the final organization of the sperm is established with the head containing the nucleus and the acrosome. The undulating membrane separates the axoneme and axial fiber. Most of the cytoplasm is lost as residual bodies.  相似文献

Comparison of the spermatozoa of three genera of limnodynastines (Myobatrachidae: Anura) with those of 42 other species of frogs (in 11 families) previously examined allows the following phylogenetic inferences. The bufonoid neobatrachians (corresponding with the Arcifera of some classifications) form a monophyletic assemblage which is characterized by the possession, unique for the Anura, of a conical perforatorium; the rod-like perforatorium, which is plesiomorphic for tetrapods, is absent. Of the taxa investigated, the myobatrachids appear to be the sister-group of the remaining bufonoids, here termed eubufonoids (leptodactylids, rhinodermatids, hylids, and bufonids). The spermatozoa of the myobatrachids Limnodynastes, Neobatrachus, and Mixophyes are very similar to each other despite extremely varied fertilization biology. Symplesiomorphies for the Anura which they exhibit include the conical acrosome with subacrosomal material, the elongate, cylindrical nucleus, single flagellum and, paralleling this, an undulating membrane which is supported by a longitudinal element, the axial (major) fibre, the latter being accompanied in the midpiece by mitochondria. A myobatrachid synapomorphy appears to be the presence of a periaxial sheath enclosing the axial fibre of the flagellum. Forward extension of the axial fibre into the centriolar fossa in myobatrachids is seen elsewhere only exceptionally (in the bufonid Nectophrynoides). Bufonids, and most other eubufonoids including leptodactylids, differ apomorphically from Limnodynastes, and Neobatrachus in location of the mitochondria at the axonemal end (and in a collar) rather than the axial fibre end of the undulating membrane. In Mixophyes, mitochondria surround the nuclear-axonemal junction in a cytoplasmic droplet and are probably shed with this at maturity.  相似文献

蓝尾石龙子(Eumeces elegans)附睾以2.5%戊二醛和1%锇酸双重固定,按常规制作超薄切片,用H-600透射电镜研究观察精子的超微结构。精子由头部和尾组成,头部由顶体复合体和核组成,尾由颈段、中段、主段和末段组成。头部的顶体囊前部扁平,分为皮质和髓质,顶体下锥由类结晶状的顶体下物质组成,穿孔器顶端尖,、穿孔器基板塞子状,细胞核延长,核内小管缺,核伸展部前端具一电子透明区,核肩圆,核陷窝锥形。颈段具片层结构,近端中心粒和远端中心粒的长轴呈直角,9束外周致密纤维与远端中心粒相应的9束三联微管相联,向后与轴丝相应的9束双联微管相联,中央纤维与2个中央单微管相联。中段短,含有线状嵴的柱状线粒体,由连续的规则小卵状或小梯形致密体组成线粒体间的环状结构,纤维鞘伸入中段,终环紧贴于细胞膜的内表面。线粒体与环状结构的模式为:rs1/mi1,rs2/mi2,rs3/mi3,rs4/mi4,横切面上每圈线粒体数目为10个。主段前面部分具薄的细胞质颗粒区。纤维3和8至主段前端消失。轴丝复合体呈“9 2”型。蓝尾石龙子精子超微结构与已描述的石龙子科种类比较发现,与蜓蜥群和胎生群的石龙子相似;但没有发现石龙子科精子的独征。  相似文献

We describe, for the first time, the sperm ultrastructure of the two genera of Pseudinae. Based on sperm ultrastructure, the five species herein examined can be separated into three groups: one containing Pseudis paradoxa, P. bolbodactyla, and P. tocantins, the second containing P. minuta, and the third containing Lysapsus laevis. The midpiece is similar in all species and auxiliary fibers and the undulating membrane are absent. In Pseudis a subacrosomal cone and a multilaminar structure (P. minuta) or a granular material (P. paradoxa group) are seen above the nucleus. Lysapsus laevis has only remnants of the subacrosomal cone. All species have peripheral fibers associated with the outer doublets of the axoneme. We tested the hypothesis of correlation between the presence of an undulating membrane and fertilization environments in anurans using a concentrated changes test (CCT) based on the Hay et al. (Mol Biol Evol 1995;12:928-937) hypothesis of phylogenetic relationships among anuran families. Only a subset of the resolved topologies derived from the Hay et al. (1995) cladogram, where Ranoidea is the sister-group of Sooglossidae, produced significant probabilities of the CCT. Therefore, support for the correlation between sperm ultrastructure and fertilization environments in anurans is, at best, equivocal.  相似文献

用光镜和电镜观察了北方山溪鲵(Batrachuperus tibetanus)精子发生过程中各种类型生精细胞的显微与超微结构变化。结果显示,北方山溪鲵在4~8月时处于精子发生期,精子形成在7~8月。成熟精子的结构具有小鲵科精子的一些共同特征,如顶体前端呈三叶草状,尾部由轴纤维、波动膜、轴丝及轴丝旁纤维构成,轴纤维粗大呈圆柱形,尾部无线粒体等。比较分析认为,在两栖类的系统发育中,轴纤维、波动膜和轴丝旁纤维的消失为近裔性状。  相似文献

应用透射电镜、扫描电镜和光学显微镜对峨眉髭蟾(Vibrissaphoraboringii)精子的形态和超微结构研究的结果表明:峨眉髭蟾的精子具角蟾科物种精子基本的形态和结构特征,即精子头部呈螺旋状,尾部呈弯曲状;精子具锥形的顶体、纤维束构成的穿孔器、平行排列的中心粒和双轴丝;线粒体位于尾部;精子核窝不明显、无轴纤维和波动膜等特征。此外,对已有报道的角蟾科和无尾类物种精子的特征进行分析比较表明:(1)角蟾科精子细胞核呈螺旋状,中心粒平行排列,尾部具双轴丝等结构不同于无尾类其他科精子的结构,具有明显的科间差别;(2)角蟾科精子各部的量度,尾部线粒体的分布和数量,以及轴丝的排列等特征在属间和种间表现出明显的差异;(3)峨眉髭蟾和东南亚拟髭蟾指名亚种精子的形态和超微结构存在明显的差异。  相似文献

Summary Early spermatids of the crabUca tangeri consists of the nucleus of granular chromatin and the cytoplasm, which contains a proacrosomal vesicle in close association with membrane lamellae. In the mid spermatids an invagination of the acrosomal vesicle membrane gives rise to the formation of the perforatorium, a spindle-shaped tubule which encloses tubular membranous structures. The pair of centrioles located at the base of the acrosome is not directly involved in perforatorial differentiation. The acrosomal vesicle shows a heterogeneous content composed of the operculum, the thickened ring, and three layers of different materials concentrically arranged around the perforatorium. During the late spermatid stage the nuclear profile differentiates numerous slender arms and the chromatin arranges into fibers. Membranous tubules from the cytoplasm become incorporated into the tubular structures of the perforatorium. The mature spermatozoon has the typical structure of the branchyuran sperm, with a complex acrosome, cupped by the nucleus, and a thin cytoplasmic band intervening between the former main elements. The centrioles are degenerate. The nuclear arms are unusually numerous (more than 20) and lack microtubules or microtubular derivatives.  相似文献

The identification of Diopatra species lacks of clear diagnostic features of taxonomic importance and the knowledge of their reproductive characters is scant. The spermatozoa of Diopatra neapolitana were ultrastructurally investigated by electron microscopy in order to correlate the mode of reproduction with sperm cells morphology. The mature male gamete has a depressed subspherical nucleus, a cone-like acrosome, and a long flagellum. The acrosome is conical in shape and radially symmetrical, with a base diameter twice the height. Within the acrosome vesicle, the basal region includes a very electron-dense thickened ring composed of paracrystalline substances. The subacrosomal space is filled with a poorly electron-dense material, with straight filaments axially arranged to form a perforatorium. The nucleus contains the complete axial canal, holding the hind perforatorium region. The middle piece consists of five mitochondria with well-distinct membranes and tubulo-vesicular cristae. Two centrioles are located perpendicularly to each other. The proximal one lies in the central fossa and the distal one, slightly eccentric to the sperm axis, anchors to the plasma membrane by nine satellite rays of the pericentriolar complex. The axoneme has a 9+2 arrangement of microtubules. In general, the spermatozoon of D. neapolitana conforms exteriorly to the typical ect-aquasperm; the acrosome complex ultrastructure, however, shows noticeable modifications from the basic form. This finding agrees with the previously observed reproductive pattern (broadcast spawning—free-swimming larvae) of D. neapolitana belonging to Santa Gilla population, and may be helpful to solve the taxonomic problems of the D. neapolitana complex as well.  相似文献

The structural organisation of the spermatozoon from two species of bandicoot rats Bandicota bengalensis and Bandicota indica was investigated by light and electron microscopy together with the effect of incubation in Triton-X 100 and sodium dodecyl sulphate. The sperm head of B. bengalensis is invariably falciform, has a uniform electron-dense nucleus capped by an acrosome with a posteriolateral equatorial segment, a subacrosomal cytoskeleton with a large rostral perforatorium, and a sperm tail, attached to the lower concave surface of the sperm head, with typical coarse fibres and fibrous sheath. By contrast, the sperm head shapes of B. indica are generally conical or bulbous, the nucleus contains a few large vacuoles, the acrosome lacks an equatorial segment, no recognisable perforatorium occurs, and the sperm tail, which is attached basally, is very short with only modest development of coarse fibres and fibrous sheath. These results indicate that, within the genus Bandicota, huge interspecific differences in morphology of the spermatozoon have evolved. The spermatozoa of B. bengalensis are similar to those of Rattus and many other murids and thus presumably represent the ancestral condition, whereas those of B. indica (and B. savilei) are unlike spermatozoa from any other eutherian mammal so far described. © 1998 The Royal Swedish Academy of Sciences.
