Complete dissociation of dimeric plasma sex steroid-binding protein (SBP or SHBG) was obtained in 6 M urea at 10 degrees C. Removal of urea resulted in the refolding of monomers, followed by reformation of dimeric SBP, which migrates with the same mobility as the native protein. Dimerization does not require Ca+2 or steroid. Renatured monomers yield dimers with dissociation constants for 5 alpha-dihydrotesterone (DHT) and 17 beta-estradiol (E2) indistinguishable from those of native human SBP. This phenomenon was also demonstrated by mixing human and rabbit SBPs that, upon renaturation, form a hybrid dimer composed of one human subunit and one rabbit subunit. The hybrid binds both DHT and E2 in contrast to rSBP, which only binds the androgen. Therefore, we conclude that (1) docking of the two subunits creates an asymmetric steroid-binding site located at the interface between the subunits, and (2) only one face of the dimer defines the specificity for binding E2 by encompassing portion of a structural motif that recognizes the flat ring A of E2. The remaining portion, which recognizes the saturated ring A of DHT, is shared by both faces of the dimer. Because native monomers do not exist alone, the often-asked question of whether the SBP monomer binds steroid can be considered meaningless; steroid-binding activity is expressed only in the dimeric state. Finally, formation of the hybrid indicates that SBP dimerization represents a conserved event during the molecular evolution of SBP, suggesting that the structural elements responsible for dimerization will be homologous in SBPs from other species.  相似文献

LHRH and its analogues produce profound antireproductive effects in both sexes of a variety of animal species. Although the LHRH agonists induce gonadotropin release, gonadal steroid secretion, ovulation, and spermatogenesis as an expression of their traditional profertility pharmacologic profile, they paradoxically and characteristically cause predominant antifertility effects which have been extensively evaluated for potential contraceptive purposes. These agonists produce their antireproductive effects in both males and females by common mechanisms, ultimately resulting in disruption of pituitary-gonadal function, depression of steroidogenesis, and inhibition of target organs dependent on such gonadal support. Similar antireproductive effects have been observed with the LHRH antagonists which competitively inhibit LHRH-induced gonadotropin secretion resulting in reduced blood gonadal steroid levels. Use of the inhibitory properties has been extended to cancer therapy based on the ability of the LHRH analogues (particularly the agonists) to inhibit the growth of steroid-dependent (responsive) tumors (e.g., mammary, prostate) similar to that produced by gonadectomy and antisteroid treatments. The use of these peptides for selected hormone-sensitive tumors presents a novel pharmacotherapeutic application for this class of drug.  相似文献

Plasma membranes of decidual tissue cells specifically bind the sex steroid-binding globulin (SBP) complexes with estrogen (estradiol, estriol, estrone) and with the pharmacological agent danazol but do not interact with the SBP-testosterone or SBP-dihydrotestosterone complexes. The selectivity of interaction of the SBP-steroid complexes with decidual tissue cellular membranes provide evidence for the active role of SBP in the realization of steroid effects on the target tissue.  相似文献

Epidemiological evidence implicates dietary isoflavone intake as protective against prostate disease. A putative mechanism is attenuated circulating androgen levels in male populations consuming an isoflavone rich diet. We investigated this hypothesis by collecting plasma from 60 Japanese and 60 New Zealand males aged between 21 and 31 years each consuming their traditional diets. We measured plasma testosterone, dihydrotestosterone (DHT), androstenedione, dehydroepiandrosterone sulfate (DHEAS), the combined levels of androsterone sulfate and epiandrosterone sulfate (AoS/epiAoS), sex hormone-binding globulin, and cortisol and corticosteroid-binding globulin as well as the isoflavones genistein and equol. Plasma genistein and equol levels were several times higher in Japanese males as would be expected from an isoflavone rich diet. However, androstenedione, DHEAS, calculated free testosterone and paradoxically markers of 5alpha-reductase, DHT and AoS/epiAoS were all also significantly higher in Japanese rather than the New Zealand male counterparts. All other comparisons were not significant. Plasma DHT and DHEAS correlated positively with plasma equol and plasma AoS/epiAoS correlated positively with genistein levels. Taken together the results suggest that, rather than reduced levels of steroidogenesis, Japanese males may have increased 5alpha-reductase activity and possibly altered 17beta OH steroid dehydrogenase activity. Significantly the positive association between isoflavones levels and 5alpha-steroids is counter-intuitive to isoflavone intake offering prostate protection, unless this is postulated to occur through other mechanisms.  相似文献

Many seasonally breeding vertebrate species have an associated reproductive pattern: mating behavior, gonadal activity, and peak circulating androgen levels occur simultaneously. In these species, androgens influence the expression of male mating behavior. Other species have a dissociated reproductive pattern: mating behavior occurs at a different time than peak gonadal activity. In such species, it is hypothesized that mating behavior is not dependent on androgen levels [Crews, D., 1984. Gamete production, sex hormone secretion, and mating behavior uncoupled. Horm. Behav. 18, 22-28]. The salamander Desmognathus ochrophaeus mates in the spring and fall while spermatogenesis occurs during the summer, suggesting that it has a dissociated reproductive pattern and that androgens do not mediate mating behavior. To assess whether mating behavior is regulated by gonadal androgens, we castrated males to reduce endogenous androgens and implanted testosterone propionate (TP) to restore androgen levels. Castrated males mated significantly less than did control males. Castrated males given TP mated as much as control males. Compared to controls, circulating androgen levels (both testosterone (T) and dihydrotestosterone (DHT)) were reduced in castrated males and elevated in castrated males given TP implants. We also found that plasma corticosterone (CORT) levels were strongly and positively correlated with T levels. Together, these data indicate that, although spermatogenesis is dissociated in time from mating behavior, androgens are associated with the expression of mating. Thus, the associated-dissociated dichotomy does not adequately describe the reproductive pattern of D. ochrophaeus. We discuss the limitations of the associated-dissociated framework in clarifying hormone-behavior relationships in reptiles and amphibians.  相似文献

The binding of sex steroids to plasma proteins was examined in post-natal Djungarian and golden hamsters. With dihydrotestosterone or testosterone as ligands, steady-state polyacrylamide gel electrophoresis revealed 2 androgen binding components in the plasma of young Djungarian hamsters of both sexes. The fast-moving component exhibited a low affinity and high capacity for androgens and corresponded to albumin in stained gels. In contrast, the slow moving component was a beta-globulin with high affinity (Ka = 10(9) M-1) and low capacity for androgens, and was identified as a specific sex steroid-binding protein (SBP; also SHBG). This SBP did not bind oestrogens or corticosteroids and was electrophoretically distinct from corticosteroid-binding globulin (CBG). In addition, this protein did not appear to be of testicular origin because it was present in immature females and in immature males that had been castrated for 8 days. Plasma concentrations of SBP in males as measured by a diethyl-aminoethyl-cellulose binding assay were low at birth, became significantly elevated shortly thereafter when plasma androgen values were also elevated, and subsequently fell to low levels during puberty. These changes follow the same general pattern that has been described for other mammals, including humans, during this period of reproductive development. Although the significance of elevated SBP concentrations during prepuberty has yet to be determined, it appears that the increased concentrations of high-affinity androgen binding in the plasma of Djungarian hamsters plays a role in the asynchronous pubertal development of the testes and accessory organs which occurs in this species. The post-natal SBP pattern in females was similar to that observed in males. Plasma SBP levels were low or undetectable in adults of both sexes. As previously described for adult golden hamsters, the plasma of post-natal male and female golden hamsters lacked a specific SBP: androgen binding in this species is apparently limited to albumin and CBG.  相似文献

The male little brown bat is a seasonally reproductive mammal that exhibits dramatic increases in plasma concentrations of sex steroid-binding protein (SBP) in the spring, following arousal from hibernation. Adult male bats, aroused prematurely from hibernation, were found to exhibit increases in plasma SBP titers that were comparable to those observed during normal spring arousal. To evaluate the role of the thyroid gland in the control of SBP in this species, plasma SBP concentrations were determined at weekly intervals in adult male bats that were either thyroparathyroidectomized (TRX) or sham operated (SHAM) after arousal from hibernation. Plasma SBP titers in SHAM males increased markedly within the first week after arousal and by 3 wk had reached levels 20-fold higher than those measured in hibernating controls. In contrast, plasma SBP values in the TRX animals did not increase significantly following arousal but were maintained at low basal levels throughout the experiment. The postarousal rise in SBP, which was blocked by TRX, was completely restored by implantation of either L- or D-thyroxine pellets. In male bats, TRX also hindered the normal postarousal atrophy of the sex accessory glands and resulted in attenuation of the postarousal increases in plasma testosterone concentrations. These effects of TRX were also prevented by treatment with thyroxine. Thus, the thyroid appears to play a significant role in the control of the postarousal rise of SBP in the little brown bat and may be an important factor in the regulation of reproductive function in this species.  相似文献

Plasma gonadotropin II (GTH II) concentrations were significantly higher (approx. 15-20-fold) in estradiol-17beta (E(2)) treated (1.0 microg or 2.5 microg g(-1) body weight) female black porgy from days 4 to 12 compared with the control. E(2) (1 microg g(-1) wt.) had a stronger stimulation on plasma GTH II in early recrudescent phase (low GSI) males (11-fold) than in high GSI and late spermiating males (2.6-fold, P< 0.05). No effect of androgens (testosterone, T; 5alpha-dihydrotestosterone, DHT) on plasma GTH II levels was observed either sex. The levels of plasma GTH II were stimulated in 1,4,6-androstatriene-3,17-dione (ATD, 1 microg g(-1), 2 microg g(-1) body wt.) and fadrozole-treated (1 microg g(-1), 3 microg g(-1) body wt.) groups compared to control. Tamoxifen (1 microg g(-1), 3 microg g(-1) body wt.) but not enclomiphene could stimulate high GTH II levels in plasma. In another experiment of ATD in combination with T, T treatment further attenuated the ATD stimulation of plasma GTH II levels. We concluded that GTH II secretion is positively regulated by an estrogen-specific effect in female and male black porgy. Gonadal stage had significant effects on the responsiveness of GTH II to E(2) stimulation in males. A negative aromatase-dependent feedback control of plasma GTH II levels was also suggested in the protandrous black porgy, Acanthopagrus schlegeli.  相似文献

Plasma steroid binding was examined in samples obtained from seven species of bats representing four different families. A specific sex steroid-binding protein (SBP) was identified by steady-state polyacrylamide gel electrophoresis in representatives of two families, the phyllostomids and the vespertilionids. In these species, as in primates, SBP not only exhibited high affinity for the androgens testosterone and dihydrotestosterone (DHT), but also for estradiol. A specific SBP was not identified in the tropical American vampire bat or in the two species of pteropodids examined. In all species examined, except for the vampire bat, a specific corticosteroid-binding globulin (CBG) was also identified. In addition to binding glucocorticoids, CBG in these species appeared to bind androgens as well.  相似文献

The role of the carbohydrate component of sex steroid-binding globulin (SBP) from human blood in the glycoprotein interaction with the recognition system for SBP-estrogen complexes in human decidual endometrium plasma membrane was studied. It was shown that the removal of N-acetylneuraminic acid residues from the oligosaccharide chains of SBP did not affect the steroid-binding or immunochemical properties of the glycoprotein. At the same time, the above modification of the glycoprotein resulted in a loss by SBP of its ability to specifically interact with the membrane recognition system. It is concluded that the oligosaccharide chains of SBP are involved in the formation of determinants needed for recognition of the SBP-estrogen complexes by endometrium cell plasma membranes.  相似文献

A full-length 1,209 bp cDNA encoding the human sex steroid-binding protein of plasma (SBP or SHBG) and testis (ABP) was constructed and expressed in BHK-21 cells. The sequence agrees with the published gene and protein sequences. The cells were found to secrete SBP following transfection and G418r selection. The recombinant protein binds 5 alpha-dihydrotestosterone with a Kd of 0.28 nM. It also binds testosterone and 17 beta-estradiol but not progesterone, estrone or cortisol revealing a steroid-binding specificity identical to that of human SBP. SDS-PAGE patterns are less complex than human SBP and show a monomeric molecular weight of about 43 kDa.  相似文献

Significant developments have taken place within the past five years on the characterization, molecular biology and function of the plasma sex steroid-binding protein, SBP (or sex hormone binding globulin, SHBG). During the span of that time, amino acid sequences of two SBPs have been established, amino acid residues in the steroid-binding site have been identified, the structure of the human SBP gene has been deduced and evidence for the possible existence of a SBP membrane receptor has been presented. This review covers the salient aspects of these and other developments including a critical analysis of the various proposed models and interpretations with regards to the structure, evolution, molecular biology and function of SBP.  相似文献

Temporal relationships between concentrations of sex steroid-binding protein (SBP), corticosteroid-binding globulin (CBG), total and free estradiol, total and free testosterone, cortisol, and progesterone were studied in plasma obtained at 1- to 3-day intervals throughout gestation in six rhesus macaques. Concentrations of SBP and CBG were measured by diethylaminoethyl cellulose filter assays. Total and free steroids were estimated by radioimmunoassay and ultrafiltration dialysis, respectively. We found that SBP was elevated between days 30 and 50 and CBG between days 60 and 140; both then declined until term (167 days). Estradiol increased gradually throughout gestation. Testosterone was elevated between days 10 and 40, then declined, and rose slightly in late gestation until approximately 15 days before delivery, when it increased markedly. Free estradiol and testosterone increased dramatically before parturition. Progesterone was elevated between days 25 and 45 and declined to relatively constant levels thereafter. Cortisol was essentially unchanged throughout gestation. Our data show that in the pregnant rhesus, levels of SBP and CBG vary independently of one another, but both decline before term; concentrations of both total and free estradiol and testosterone increase markedly before parturition; in late gestation, elevated estrogen is not associated with increased levels of SBP or CBG (as it is in human females).  相似文献

We investigated the capacity of ovaries of yellow perch to produce steroid hormones in vitro and the ovarian response to gonadotropin and phorbol ester during the annual reproductive cycle. The effects of photothermal manipulation on perch gonadal steroidogenesis and its regulation have also been examined. Initially, all females kept indoors were exposed to the same water temperature and photoperiod. By the end of August, following the first sampling, fish were submitted to different photothermal regimes. Group A was maintained under photothermal conditions characteristic for southern Ohio. Group B was submitted to a condensed light/temperature regime designed to accelerate physiological changes that depend on photothermal stimuli. In group A, basal in vitro production of ovarian estradiol (E2) was the highest in October and November, and hCG significantly stimulated E2 secretion during the entire period of vitellogenesis (October-January). In this group, the highest production of basal testosterone (T) was observed before spawning. hCG-stimulated production of T was highest at the beginning of vitellogenesis. Gonadotropin stimulated T production before spawning, a time when gonadotropin was unable to stimulate E2 production. Phorbol ester (PDBu) stimulated E2 and T production during vitellogenesis at the same time points as hCG did (E2: December, January; T: December). hCG-stimulated T production was not mimicked by PDBu in April. Condensing of the photothermal cycle resulted in diminished ovarian production of E2 during vitellogenesis. Moreover, the fish submitted to a condensed photothermal cycle demonstrated augmented T production during the postvitellogenic stage of ovarian development. Ovaries of group B did not respond to PDBu. Generally, the seasonal fluctuations in ovarian capacity to produce E2 and T as well as in gonadal responsiveness to gonadotropin observed in female yellow perch illustrate the dynamic nature of ovarian endocrine function. The lack of response to gonadotropin with regard to E2 production prior to spawning is not due to insensitivity to gonadotropin, but rather due to some deficiency in steroidogenesis (e.g. reduced aromatase activity). It appears also that ovarian steroidogenesis and its regulation are dependent on annual changes of photothermal conditions.  相似文献

The chronic administration of superactive agonists of gonadotropin releasing hormone (GnRH-A) have been reported to have a direct inhibitory effect on the sex tissues of the male rat. In an attempt to confirm or refute this statement, adult male rats were either left intact or were castrated and then treated daily for 14 days with either testosterone (T), dihydrotestosterone (DHT) or sesame oil (vehicle). Half of the intact and castrate animals also received daily injections of 200 ng of the GnRH agonist, D-Leu6, des-Gly10-GnRH ethylamide for 14 days. Twenty-four hours after completing treatment, blood levels of follicle-stimulating hormone (FSH), luteinizing hormone (LH) and T were measured by radioimmunoassay and the ventral prostate gland (VP), seminal vesicle (SV) and penis were weighed. After 2 weeks of GnRH-A treatment, the plasma T level was reduced from 2506 +/- 170 (pg/ml +/- SEM) in the intact, nontreated animals to 907 +/- 69 in the intact, GnRH-A-treated group, indicating that the dosage of GnRH-A used in this study had an inhibiting effect on T secretion. No differences were observed in the VP, SV and penile weights between the castrate, GnRH-A and the castrate, nontreated groups. When exogenous T or DHT was given for 14 days to these castrated animals, the concomitant administration of GnRH-A did not appear to have any effect on the plasma T levels or the sex accessory tissue weights. These data suggest that GnRH-A itself does not appear to have a direct inhibitory or stimulatory effect on the sex tissues of the adult male rat.  相似文献

Two experiments examined the effects of the free forms of testosterone (T), dihydrotestosterone (DHT), and estradiol (E2) upon male mouse (Mus musculus) courtship vocalizations and seminal vesicle weight. In the first experiment, administration of T to castrated males was associated with a large number of vocalizations and large seminal vesicle weights, DHT was associated with large seminal vesicle weights but very few vocalizations, whereas E2 was associated with low measures of both vocalization and seminal vesicle weight. In the second experiment, T and DHT had effects highly similar to those of the previous experiment; however, in contrast to the previous experiment, both low and high dosages of E2 were associated with large numbers of ultrasonic vocalizations but small seminal vesicles. A mixture of E2 and DHT was very similar to T in its effect upon both measures. We suggest from these results that hormonal mechanisms similar to those reported for rat sex behavior may interact with situational variables to determine the expression of male mouse courtship.  相似文献

Male and female Carassius auratus were exposed to safe (SC; 0.01 ppm) and sublethal (SL; 0·1 ppm) concentrations of an organochlorine pesticide γ-hexachlorocyclohexane (γ-HCH) for 4 weeks during the pre-spawning phase (June) of the annual reproductive cycle. Gonadosomatic index and gonadotropin levels were significantly lower after exposure to both γ-HCH concentrations than in control fish. After 4 weeks exposure, gonadal tissue from control, SC and SL exposed fish was incubated with carp hypophyseal homogenate (chh). The chh stimulated production of testosterone, 17,20β-dihydroxy-4-pregnen-3-one (17,20βP), 11-deoxycortisol, and 11-ketotesterone (11-KT) was estimated in the unconjugated (free) and conjugated (glucuronide) fractions by radioimmunoassay. In both sexes, testosterone production was greatly decreased in γ-HCH exposed fish compared to controls. 17,20βP production was low in all fish and was unaffected by γ-HCH. Free 11-deoxycortisol production by testicular fragments was higher in SL and SC compared with controls, while conjugated 11-deoxycortisol was increased only higher in the SL. In ovarian fragments from exposed fish, free 11-deoxycortisol decreased while glucuronide concentrations increased compared with controls. 11-KT production was significantly decreased in testicular fragments of exposed fish. The results indicate that γ-HCH inhibits gonadal recrudescence by decreasing both gonadotropin secretion and its potential for stimulation of steroidogenesis.  相似文献

The effect of unsaturated and saturated nonesterified fatty acids (NEFAs) on the electrophoretic, immunological, and steroid-binding properties of human sex hormone-binding protein (SBP) were investigated. Tests were carried out on whole serum from pregnant women and on purified SBP using polyacrylamide gel electrophoresis, crossed immunoelectrophoresis with autoradiography, and equilibrium dialysis. All three methods showed that NEFAs influence the binding of sex steroids to SBP both in whole serum and with the purified protein. Saturated NEFAs caused a 1.5-2-fold increase in binding of dehydrotestosterone, testosterone, and estradiol to SBP, while unsaturated NEFAs, such as oleic (18:1) and docosahexaenoic (22:6) acids inhibited the binding of these steroids to SBP. Thus, unsaturated NEFAs in the concentration range 1-100 microM are more inhibitory for estradiol binding than for testosterone or dehydrotestosterone binding. In addition to these binding changes, polyacrylamide gel electrophoresis and immunoelectrophoretic studies revealed a shift in SBP from the slow-moving active native form to a fast-moving inactive one. There was also a reduction in the apparent SBP concentration by Laurell immunoelectrophoresis in the presence of unsaturated NEFA (5.5 nmol of NEFA/pmol of protein). These studies indicate that unsaturated NEFAs induce conformational changes in human SBP which are reflected in its electrophoretic, immunological, and steroid-binding properties. They suggest that the fatty acid content of the SBP environment may result in lower steroid hormone binding and thus increased free hormone levels.  相似文献

Plasma testosterone (T), dihydrotestosterone (DHT), 17-hydroxyprogesterone (17OHP), androstenedione (A), estradiol (E2), and dehydroepiandrosterone sulfate (DHAS) were measured by radioimmunoassay after celite chromatography prior to and after a 3-hour infusion of the synthetic gonadotropin releasing factor, GnRH, in normal prepubertal and pubertal boys. Plasma T levels rose (p less than 0.001) in the pubertal but not prepubertal boys. 17OHP concentrations increased in those boys who had an increment of T. A, DHT, E2 or DHAS levels did not increase after GnRH. Basal levels of T, DHT, A and DHAS correlated with the peak and mean serum LH levels attained during the GnRH infusion. These data confirm the greater Leydig cell responsivity to transient rises of endogenous gonadotropin in pubertal males and also suggest that there may be a relationship between adrenal androgen production and maturation of the hypothalamic-pituitary-gonadal system.  相似文献
