Sphaeropsis pyriputrescens, the causal agent of Sphaeropsis rot of pears and apples, is a recently described species. In this study the effects of culture media, temperature, water potential, pH and light on mycelial growth and pycnidial production of S. pyriputrescens were evaluated. Apple juice agar and pear juice agar were most suitable for mycelial growth of all six isolates tested. Cornmeal agar was not suitable for either mycelial growth or pycnidial production. The fungus grew from -3 to 25 C, with optimum growth at 20 C and no growth at 30 C. The fungus grew at water potential as low as -5.6 MPa on potassium chloride-amended potato-dextrose agar (PDA). Hyphal extension was not observed at -7.3 MPa after 10 d incubation, but growth resumed when the inoculum plugs were placed on PDA. The fungus grew at pH 3.3-6.3 and optimum growth was at pH 3.3-4.2. No mycelial growth was observed at pH above 7.2 after 10 d incubation, but growth resumed when the inoculum plugs were transferred onto PDA. Regardless of medium tested, few pycnidia formed at 20 C in the dark. Pycnidial production was enhanced significantly by fluorescent light, but continuous light appeared to reduce pycnidial production, depending on the medium. Oatmeal agar (OMA) was most suitable for production of pycnidia and conidia. Pycnidia that formed on 3 wk old OMA cultures at 20 C under 12 h light/12 h dark produced abundant conidia, and the technique is recommended for inoculum production.  相似文献

Optimising sporulation and virulence in Drechslera avenacea   总被引:1,自引:0,他引:1  
Studies were conducted on agar media to optimise sporulation of Drechslera avenacea, a fungal pathogen being evaluated as a biological control agent for Avena species (wild oats). Conidium production was affected by nutrition, pH, temperature and light conditions. Of the agar media tested, Czapek Dox agar (CZA) and half-strength oatmeal agar (½OMA) were the only media where sporulation occurred at all temperatures tested under a 12-h light:12-h dark photoperiod (L/D). The optimum temperature for conidium production was 20°C on ½OMA, whereas there was no optimum temperature on CZA. Under a 12-h near-ultraviolet (NUV):12-h dark photoperiod (NUV/D), similar numbers of conidia were produced on CZA at 6.66, 14.56, and 22.78 W m?2, whereas on ½OMA conidium production was the highest at 14.56 W m?2. When NUV/D and L/D conditions were compared, similar numbers of conidia where produced on CZA, whereas ½OMA conidium production was superior under the NUV/D photoperiod. Considerable variation in sporulation and degree of virulence of D. avenacea was detected among isolates from different geographic areas. The most virulent conidia were obtained on ½OMA at 20°C incubated under continuous illumination NUV light. Therefore, the most suitable conditions for conidium production of D. avenacea were growth for 1 week on ½OMA at 20°C under continuous NUV at an intensity of 14.56 W m?2. Under these conditions, 1.1×105 conidia mL?1 were produced which is the highest sporulation yet reported for any Drechslera spp., which are traditionally poor sporulators.  相似文献

The effect of temperature and light conditions (spectral quality, intensity and photoperiod) on germination, development and conidiation of tomato powdery mildew (Oidium neolycopersici) on the highly susceptible tomato cv. Amateur were studied. Conidia germinated across the whole range of tested temperatures (10–35°C); however, at the end‐point temperatures, germination was strongly limited. At temperatures slightly lower than optimum (20–25°C), mycelial development and time of appearance of the first conidiophores was delayed. Conidiation occurred within the range of 15–25°C, however was most intense between 20–25°C. Pathogen development was also markedly influenced by the light conditions. Conidiation and mycelium development was greatest at light intensities of approximately 60 μmol/m2 per second. At lower intensities, pathogen development was delayed, and in the dark, conidiation was completely inhibited. A dark period of 24 h after inoculation had no stimulatory effect on later mycelium development. However, 12 h of light after inoculation, followed by continuous dark, resulted in delayed mycelium development and total restriction of pathogen conidiation (evaluated 8 days postinoculation). When a longer dark period (4 days) was followed by normal photoperiod (12 h/12 h light/dark), mycelium development accelerated and the pathogen sporulated normally. When only inoculated leaf was covered with aluminium foil while whole plant was placed in photoperiod 12 h/12 h, the intensive mycelium development and slight subsequent sporulation on covered leaf was recorded.  相似文献

Studies were conducted on agar media to characterize the factors for the optimization of sporulation of Alternaria alternata f. sp. sphenocleae , a fungal pathogen being evaluated as a biological control agent for Sphenoclea zeylanica (gooseweed). A. alternata f. sp. sphenocleae conidiation was affected by nutrition, temperature, light conditions, and moisture. On all agar media tested, except for half-strength potato dextrose agar (½ PDA) and V-8 juice agar (VJA), exposure to different light conditions did not have any significant effect on conidia production. However, when comparing ½ PDA and VJA, sporulation under constant near-ultraviolet (NUV) light at 28 o C increased markedly on VJA, but decreased substantially on ½ PDA. This trend, however, was opposite under dark conditions since ½ PDA produced the greatest number of conidia whereas a 75% reduction in conidia production occurred on VJA in the dark. On all the standard agar media evaluated, the most virulent conidia were obtained on ½ PDA at 28 o C under constant NUV incubated for 4 weeks. Sporulation of A. alternata f. sp. sphenocleae using the sporulation medium (S-medium) technique was rapid. Conidia were produced within 24 h and continuous sporulation was still observed until 120 h. The best primary agar media for conidia production were PDA, ½ PDA and VJA, while water agar was the poorest. Conidia production was optimized with the addition of 20 g l -1 of calcium carbonate (CaCO 3 ) and the addition of 2 ml of sterile distilled water on the medium. The most virulent conidia were produced when the primary agar was ½ PDA, the CaCO 3 concentration was 20 g l -1 , and the cultures were incubated at 18 o C in the dark. Conidiophore induction occurred on nutrient rich media and was stimulated by NUV, while formation of conidia proceeded in darkness after nutrients were depleted under warm dry or cool moist conditions. Culture media, growth conditions, and CaCO 3 affected the inoculum potential of A. alternata f. sp. sphenocleae conidia.  相似文献

A new species of Phacidiopycnis associated with pome fruits is described. The fungus causes fruit rot on apples during storage and is associated with a twig dieback and canker disease of crabapple trees and dead twigs of pear trees. To characterize the biology of the fungus and compare it with Ph. piri, the type species of the genus, effects of nine media and light on mycelial growth and pycnidial production, mycelial growth in response to temperature and mode of conidial germination in response to nutrient were determined. Apple-juice agar, pear-juice agar, prune-juice agar, potato-dextrose agar (PDA) and malt-extract agar, Czapek-Dox agar and oatmeal agar (OMA) favored mycelial growth. Cornmeal agar (CMA) did not favor mycelial growth. Light effect on pycnidial formation was medium dependent. Abundant pycnidia with mature conidia formed in 14 d old PDA and OMA cultures at 20 C, regardless of light, whereas none or very few pycnidia formed on other media in the dark. Fluorescent light stimulated formation of pycnidia except on CMA. The fungus grew at -3-25 C, with optimum growth at 15-20 C. Conidia germinated either by forming germ tubes or less often by budding. Budding of conidia occurred in 1 and 10% pear-juice solutions but not in 100% pear-juice solution. Six isolates of Ph. washingtonensis from different species of pome fruits had identical ITS sequences. The sizes of the ITS region were the same for both Ph. washingtonensis and Ph. piri, and four polymorphic nucleotide sites were found in the ITS region between Ph. washingtonensis and Ph. piri. The similarity in ITS sequences between these two taxa is confirmatory evidence for the erection of the new species of Phacidiopycnis associated with pome fruits we describe here.  相似文献

During 2011–2012, an extensive leaf spot disease caused by Stemphylium lycopersici was observed on vegetable crops including, tomato, eggplant, pepper and lettuce in major vegetable-growing regions of Malaysia. Four isolates of S. lycopersici obtained from each vegetable crop were used to determine cultural and physiological characteristics. The variations were found in colony colour (pale to light grey or light as well as the brown), texture (cottony or mycelium flat), shape (regular with concentric growth rings or irregular) and pigmentation (yellow or deep red) of the cultures. The optimum temperature for the conidial germination and mean radial growth of the isolates was 25?°C, and the radial growth of the isolates was maximal on V-8 juice agar followed by potato carrot agar. The maximum sporulation of S. lycopersici isolates was observed on V-8 juice agar media under 12/12 h light/darkness photoperiod at 25?°C.  相似文献

The Biology of the Acaropathogenic Fungus Hirsutella kirchneri   总被引:1,自引:0,他引:1  
The acaropathogenic fungus Hirsutella kirchneri (Rostrup) Minter , Brady and Hall grew best and produced most mycelia on a medium containing yeast extract , dextrose and agar; conidial production , however , was maximal or potato dextrose agar (PDA) . The best growth on both media was at 25 C and conidial germination was high within a wide range of temperatures (10 - 35 C) . Colony growth , mycelial mass production and conidial yield were best under alternating dark and light regimes . Maximal germination occurred under dark conditions . When grown in continuous light the fungus produced synnemata (compacted conidiophores) which remained viable for 22 weeks . Of six species of phytophagous mites assayed , three spider mites and a rust mite became infected by the fungus , as did , to a limited degree , a parasitic mite . Another four mites , including two pests , a scavenger and a predator , were unaffected . The fungus grew on and sporulated from heat - killed cadavers of a dead mealybug (Homoptera) . Conidial germination and penetration into live mites , under saturation condi tions , were affected little by temperatures . Intra - host growth was temperature dependent , with mite death beginning on the second or third day post - infection . Maximal sporulation from infected mites took place at 25 C . Mortality was quickest at 25 C when mites were held under saturation conditions , but some death also occurred at lower relative humidities . These data are discussed with a view to using H. kirchneri in the biological control of plant mites .  相似文献

The formation of conidia in Phaeocytostroma ambiguum on different media and conditions was investigated in this study. Carnation leaf agar (CLA) and a 12 h photoperiod (24/18 °C) provided excellent conditions for the promotion of rapid formation of both alpha (α) and beta (β) conidia in a number of P. ambiguum isolates. The dimensions of α- and β-conidia amounted to 6.0–19.6 × 3.8–7.5 μm and 6.0–24.9 × 1.1–2.6 μm, respectively. They were produced on short or elongate, simple and branched conidiophores. β-conidia have not been described before in P. ambiguum. Intermediate conidia were rarely found. On agar media the maximum conidia production of Exserohilum monoceras occurred on V-8 juice agar (VA) or centrifuged V-8 juice agar, whereas the optimal radial mycelial growth occurred on Czapek-Dox agar. The optimal temperatures for radial mycelial growth and conidia production were 28 and 27°C respectively. Light prohibited E. monoceras conidia production. The best sporulation occurred under continuous dark conditions. Echinochloa leaf decoction significantly increased conidia production on potato dextrose agar (PDA) and VA, and significantly increased germ tube length on PDA, lima bean agar and VA, but did not affect conidia germination. No conidia were produced in liquid media. Of 22 agricultural-based products evaluated as solid substrates, the most abundant sporulation (1.8 × 106 conidia g-1 of dry weight) occurred on corn leaves. The conidia production of E. monoceras on corn leaves was affected by incubation period, moisture content and substrate quantity. There were no differences in germination rate, germ tube length and virulence of conidia produced on agar media or corn leaves.  相似文献

Light conditions during fungal growth are well known to cause several physiological adaptations in the conidia produced. In this study, conidia of the entomopathogenic fungi Metarhizium robertsii were produced on: 1) potato dextrose agar (PDA) medium in the dark; 2) PDA medium under white light (4.98 W m?2); 3) PDA medium under blue light (4.8 W m?2); 4) PDA medium under red light (2.8 W m?2); and 5) minimum medium (Czapek medium without sucrose) supplemented with 3 % lactose (MML) in the dark. The conidial production, the speed of conidial germination, and the virulence to the insect Tenebrio molitor (Coleoptera: Tenebrionidae) were evaluated. Conidia produced on MML or PDA medium under white or blue light germinated faster than conidia produced on PDA medium in the dark. Conidia produced under red light germinated slower than conidia produced on PDA medium in the dark. Conidia produced on MML were the most virulent, followed by conidia produced on PDA medium under white light. The fungus grown under blue light produced more conidia than the fungus grown in the dark. The quantity of conidia produced for the fungus grown in the dark, under white, and red light was similar. The MML afforded the least conidial production. In conclusion, white light produced conidia that germinated faster and killed the insects faster; in addition, blue light afforded the highest conidial production.  相似文献

A major obstacle to native orchid production is difficulty in seed germination. Culture media and light effects on seed germination of Calopogon tuberosus var. tuberosus, a native orchid with horticultural potential, were studied. Culture media included Knudson C, Malmgren modified terrestrial orchid, and PhytoTechnology orchid seed sowing. Effects of 8 weeks continual darkness, 8 weeks 16-h photoperiod, 2 weeks dark followed by 6 weeks 16-h photoperiod, 4 weeks dark followed by 4 weeks 16-h photoperiod, and 6 weeks dark followed by 2 weeks 16-h photoperiod were examined. Percent seed germination was highest on Knudson C after 8 weeks culture; however, seedling development was enhanced on PhytoTechnology seed sowing medium during 8 weeks culture under a 16-h photoperiod. This suggests that while KC and darkness promoted seed germination, P723 and light enhanced further seedling development. Seedlings of C. tuberosus readily acclimated to greenhouse conditions.  相似文献

Ten isolates of Colletotrichum gloeosporioides were collected from different noni growing areas of Tamil Nadu, Karnataka and Kerala in India and their pathogenicity was proved under glass house conditions. Effect of different pH levels, temperature, light intensity and media were tested against the growth of C. gloeosporioides under in vitro. The results indicated that the growth of C. gloeosporioides was maximum in pH range of 6.50–7.00 and temperature range of 25–30°C. Exposure of the fungus to alternate cycles of 12 h light and 12 h darkness resulted in the maximum mycelial growth of C. gloeosporioides compared to the 24 h exposure to continuous light and 24 h exposure to continuous dark. Among the different media tested, host leaf extract medium supported significantly the maximum growth of all the 10 isolates of C. gloeosporioides followed by potato dextrose agar. Further, the strains were found to vary morphologically between the isolates under the study.  相似文献

苹果盘二孢Marssonina coronaria引起的褐斑病是造成我国苹果树早期落叶的主要原因,由于该病菌分离培养困难,阻碍了对其生物学特性的研究,进而影响了对其防治机理和流行规律的研究。本研究应用4种培养基质,探索了3种方法对苹果盘二孢的分离效果。结果表明,3种方法均可分离到病原菌,但组织块分离法和分生孢子团分离法成功率仅有10%左右,而单孢分离法污染少,成功率高达到90%以上,明显优于其他两种方法。不同培养基上菌落形态、大小和产孢情况差异也很大,培养1个月(25℃)后PDA上菌落黑褐色隆起,表面蚯蚓粪状,无气生菌丝,无子实体和基内菌丝;10%V8培养基上菌落中央隆起,黑褐色,表面生少量气生菌丝,边缘放射状,基内菌丝深褐色,有子实体;苹果叶片葡萄糖琼脂培养基(LDA)上菌落平坦,黄褐色,表面生茂密的金黄色气生菌丝,基内菌丝深褐色,有子实体;苹果叶片煎汁葡萄糖琼脂培养基(LEDA)上菌落有明显的不规则隆起,黄褐色至黑褐色,表面生少许气生菌丝,菌落生长缓慢,无基内菌丝,分生孢子盘菌落表面生,菌落直径仅2mm左右,而在其他培养基上的菌落直径可达6-8mm,说明培养基质、分离方法均对苹果盘二孢的分离培养和生长发育有明显的影响。  相似文献

The present study describes the direct regeneration of protocorm-like bodies (PLBs) in leaf explants of the tropical species Oncidium flexuosum. The explants were inoculated in a solid, modified Murashige and Skoog (MS) medium with different concentrations of the growth regulator thidiazuron (TDZ) and with or without 2,4-dichlorophenoxyacetic acid (2,4-D) and naphthalene acetic acid (NAA), and kept away from light or in a 16-h photoperiod. The presence of auxins, 2,4-D, and NAA inhibited the formation of PLBs. The highest frequency of explants that regenerated PLBs (80%) was obtained when they were maintained in a culture medium containing 1.5 μM TDZ under dark conditions. In the same culture medium but under a 16-h photoperiod, 95% of the leaf explants presented necrosis. Therefore, darkness was crucial for the regeneration of PLBs in O. flexuosum leaf explants, which is in disagreement with the literature. PLBs developed from the division of epidermal and subepidermal cells mainly on the adaxial side of the apex region of the explant. Plants with well-developed leaves and roots grew after the PLBs were transferred to growth regulator-free medium under a 16-h photoperiod.  相似文献

在研究四川省玉米纹枯病菌系时,获得23个RhizoctoniazeaeVoorhees菌株。该菌菌丝形态与R.solaniKuhn相似;菌丝细胞含3个以上椭圆形细胞核;隔膜具桶孔,桶孔周围的隔膜肿体(septalswelling)与隔膜板垂直,两端不再膨大外延;菌丝生长较快,在PDA平板26℃下培养,每24小时菌落直径增长33.2~39.1mm;菌丝与苯酚相互作用后,着色较深,呈深褐色。菌核初期为白色或奶油色,成熟后为红褐色;常散生于基质内,少数着生在基质表面和培养皿的内壁上;球形或近球形,直径0.25~1.0mm;结构致密,无菌环和菌髓分化,表面较光滑。223个R.zeae菌株均与Oniki等建立的WaiteacircinataWAG-Z标准菌株发生融合,但与R.solani的融合群标准菌株均无融合反应。R.zeae对玉米的致病性较强,稍弱于玉米纹枯病主菌系R.solaniAG-1-IA,其病斑多半不连续,颜色较深,也可0侵染果穗引致穗腐并在苞叶上产生大量红褐至黑褐色的小菌核。  相似文献

Seven strains of edible mushrooms were studied with regard to mycelial growth on different growth media and culture conditions. Medium WDA (wheat/dextrose/agar) promoted higher rates of mycelial growth for all the mushrooms investigated. The majority of the strains presented higher growth rates at 30°C, but only Lentinus edodes kept maximum rates at low pH (pH 4.0), followed by Stropharia rugosoannulata and Pleurotus ostreatus (pH 5.0). Absence of light favoured rapid mycelium development in all the strains tested.  相似文献

Light conditions during mycelial growth are known to influence fungi in many ways. The effect of visible-light exposure during mycelial growth was investigated on conidial tolerance to UVB irradiation and wet heat of Metarhizium robertsii, an insect-pathogenic fungus. Two nutrient media and two light regimens were compared. Conidia were produced on (A) potato dextrose agar plus yeast extract medium (PDAY) (A1) under dark conditions or (A2) under continuous visible light (provided by two fluorescent lamps with intensity 5.4 W m(-2)). For comparison, the fungus was also produced on (B) minimal medium (MM) under continuous-dark incubation, which is known to produce conidia with increased tolerance to heat and UVB radiation. The UVB tolerances of conidia produced on PDAY under continuous visible light were twofold higher than conidia produced on PDAY medium under dark conditions, and this elevated UVB tolerance was similar to that of conidia produced on MM in the dark. The heat tolerance of conidia produced under continuous light was, however, similar to that of conidia produced on MM or PDAY in the dark. Conidial yield on PDAY medium was equivalent when the fungus was grown either under continuous-dark or under continuous-light conditions.  相似文献

The effect of inhibitors of DNA methylation on light-sensitive developmental stages of the filamentous fungus Neurospora crassa was studied. Under conditions of nitrogen starvation, when blue light induced protoperithecia development and inhibited conidia formation, 5-azacytidine (3-300 microM) inhibited protoperithecia formation and stimulated conidia formation (a 700-fold increase after light induction). After treatment of the mycelium with 5-azacytidine, the protoperithecia formation was accompanied by inversely proportional changes in the formation of conidia, both in the dark and after illumination. In the mycelium cultivated on the Vogel's medium, 5-azacytidine (up to 30 microM) and methotrexate (up to 3 microM) stimulated the light-induced carotenoid synthesis by 30%, whereas higher concentrations of these agents were toxic to carotenoid synthesis and growth.  相似文献

选择新疆阿魏植物内生真菌中一株药理活性较强的菌,研究其生长特性,分别考察不同培养基、温度、光照及起始pH值4个单因素对菌种生长的影响,通过响应面试验优化,确定菌种最佳生长条件。结果表明,在黑暗条件下,采用土豆培养基(PDA),pH值为7,温度25℃环境中,该菌种可正常生长和繁衍,菌株平均生长速率为10.25mm·d~(-1)。  相似文献
