Seasonal dynamics in the polyphenolic composition, antioxidant activity, and their relationships during plant development were evaluated for eastern teaberry (Gaultheria procumbens L.) leaves, a traditional herbal medicine of North American natives. With the complementary UHPLC-PDA-ESI-MS3, HPLC-PDA-fingerprint, Folin-Ciocalteau, and n-butanol/HCl assays of methanol-water (75:25, v/v) extracts, the dried leaf samples harvested monthly across the growing season under Polish climate conditions were found rich in structurally diverse polyphenols (149.2–210.7 mg/g DW) including the dominating salicylates (64.6–107.5 mg/g DW), proanthocyanidins (53.0–66.8 mg/g DW), and flavonoids (17.3–25.3 mg/g DW), and the accompanying chlorogenic acid isomers (2.4–4.4 mg/g DW) and simple phenolic acids (0.9–1.1 mg/g DW). Among 28 detected analytes, gaultherin (64.6–107.5 mg/g DW), miquelianin (14.6–21.1 mg/g DW), procyanidin A-type trimer (5.5–9.5 mg/g DW), and (–)-epicatechin (5.8–7.8 mg/g DW) were the most abundant. The phenolic levels and antioxidant activity parameters in the DPPH (EC50, 15.0–18.2 μg DW/mL; 0.95–1.16 mmol Trolox equivalents/g DW) and FRAP (2.3–3.4 mmol Fe 2+/g DW; 0.86–1.26 mmol Trolox equivalents/g DW) assays showed parallel seasonal trends with maxima in September and October. As the subsequent correlation studies confirmed the determinative impact of polyphenols on the leaf antioxidant activity and its seasonal fluctuations, the Fall season could be recommended as optimal for harvesting the plant material for medicinal purposes and cost-effective production of natural health products.  相似文献

Momordica dioica Roxb. Commonly known as “Kakora” in Telugu, is used in the Indian traditional system of medicine for the treatment of diabetes. The aim of this study was to investigate the antidiabetic activity of methanolic extract of M. dioica seeds (MEMD) in streptozotocin (STZ) induced diabetic rats. The in vitro α-amylase inhibitory activity of the MEMD was done by spectrophotometric method. Diabetes was induced by STZ (45 mg/kg; i.p), MEMD (100 & 200 mg/kg; b.wt) and standard drug metformin (50 mg/kg; b.wt) were administered to the diabetic rats. Blood glucose was estimated on the 11th day and the level of MDA, SOD and CAT was estimated in the liver tissue homogenate after the 15 days of experimental period. MEMD showed significant inhibition of alpha amylase activity and the IC50 was found to be 48 μg/ml. Oral administration of MEMD significantly reduced blood glucose level (P < 0.05), diminished the MDA level and refurbished depleted antioxidant enzymes and Insulin level to normalcy. These findings revealed that M. dioica seeds possess antihyperglycemic, antioxidant and anti lipid peroxidative activity and thus mitigate STZ-induced oxidative damage.  相似文献

Mucuna pruriens seeds have relatively high crude protein (CP) concentrations, but little is known about their potential to replace commonly used CP supplements in ruminant rations. The aim of this experiment was to determine effects of replacing soybean meal (SB) with Mucuna on the performance of lambs. Forty Rambouillet lambs (33.2 ± 5.73 kg) fed a basal diet of maize grain, cottonseed hulls and urea were randomly assigned to one of four supplements formulated by substituting 0 (SB), 330 (Lo), 670 (Med) or 1000 g/kg (Hi) of soybean meal with rolled Mucuna seeds. Lambs were housed individually in metabolic crates and allowed ad libitum access to isocaloric (metabolizable energy=11.7 MJ/kg dry matter, DM) and isonitrogenous (CP = 146 g/kg, DM) diets for 14 d of adaptation and 7 d of total fecal collection. Fecal egg counts and coccidian oocyst scores were determined on d 14. Dry matter intake (1.7 kg/d versus 1.5 kg/d; P<0.05), CP digestibility (774 g/kg versus 714 g/kg DM; P<0.05) and N retention (28.0 g/d versus 20.4 g/d; P<0.01) were higher and amylase-pretreated neutral detergent fiber digestibility (617 g/kg versus 686 g/kg DM) was lower (P<0.05) in sheep fed SB versus Mucuna diets. However, supplementary protein source did not affect rumen pH, blood urea N or glucose concentration, or fecal egg counts. Increasing the level of Mucuna supplementation increased (P<0.05) level and efficiency of microbial protein synthesis, ruminal fluid acidity, total volatile fatty acid concentration, decreased (P<0.05) coccidian oocyst scores, and tended (P<0.10) to increase N retention. Therefore, SB is a better supplement than Mucuna to support performance of lambs. Nevertheless, Mucuna seeds are a promising CP supplement for situations where cost or availability precludes use of SB in ruminant rations.  相似文献

Response surface methodology was used to evaluate the quantitative effects of three independent variables: rapeseed moisture content, concentration of the added enzymes and conditioning temperature, on the antioxidant capacity and total phenolic, tocopherol, and phospholipid contents in the enzyme-treated rapeseed oils. The highest antioxidant capacity (1220.0, 964.8 μmol TE/100 g) total phenolic (83.3, 74.0 mg SA/100 g) and phospholipid (12,532, 12,376 mg/kg) contents reveal two rapeseed oils extruded from seeds contained 11% moisture, treated with cellulolytic and pectolytic enzymes (0.05%), respectively, and heated at 120 °C. However, the highest content of total tocopherols was determined in rapeseed oils pressed from seeds with 7% moisture, after addition of cellulolytic (0.05%) and pectolytic (0.1%) enzymes, heated at 90 and 105 °C, respectively. Total phenolic and phospholipid contents in the enzyme-treated rapeseed oils correlated significantly (p < 0.0000001) with antioxidant capacities of oils (R2 = 0.8710 and 0.6581, respectively). Experimental results of the antioxidant capacity, total phenolic, tocopherol and phospholipid contents were close to the predicted values calculated from the polynomial response surface models equations (R2 = 0.9727, 0.9870, 0.8390 and 0.9706 for the cellulolytic enzyme-assisted rapeseed oils and R2 = 0.9148, 0.9489, 0.9426 and 0.9479 for the pectolytic enzyme-assisted rapeseed oils). The optimum rapeseed moisture content, enzyme concentration and conditioning temperature for the cellulolytic and pectolytic enzyme-treated rapeseed oils were 11% and 9.7%, 0.08% and 0.1%, and 120 °C, respectively.  相似文献

A series of naphthopyrans was synthesized employing silica supported fluoroboric acid under solvent free conditions in a microwave reactor. The catalytic influence of HBF4–SiO2 was investigated in detail to optimize the reaction conditions. The synthesised compounds were evaluated for in vitro xanthine oxidase inhibitory activity for the first time. Structure–activity relationship analyses have also been presented. Among the synthesised compounds, NP-17, NP-19, NP-20, NP-23, NP-24, NP-25 and NP-26 were the active inhibitors with an IC50 ranging from 4 to 17 μM. Compound NP-19 with a thiophenyl ring at position 1 emerged as the most potent xanthine oxidase inhibitor (IC50 = 4 μM) in comparison to allopurinol (IC50 = 11.10 μM) and febuxostat (IC50 = 0.025 μM). The basis of significant inhibition of xanthine oxidase by NP-19 was rationalized by its molecular docking at MTE binding site of xanthine oxidase.  相似文献

A xanthine oxidase (XOD) was expressed, purified and partially characterized from Arthrobacter sp. with a negative immune protocol. To determine the optimal inducer for XOD, xanthine, hypoxanthine and uric acid were added into the medium of cultivation. The results revealed that with the inducement of about 14 mM xanthine, the highest XOD activity could be detected. To separate XOD from Arthrobacter sp., the cells were first cultured without any inducement; then the total proteins of the collected cells were extracted and immunized to rabbits for the polyclonal antibodies. These antibodies were then coupled with sepharose CL 6 B, and the medium was further employed to deplete most of the cells’ back ground proteins. Began with ~20 mg crude protein from disrupted cells was subjected to the antibody medium, and ~1.45 mg protein was detected in unbinding fractions with ~92.0% of activity. The extracted xanthine oxidase was ~85% pure with native-PAGE analysis, and ~90% pure with SDS-PAGE analysis, the yield of protein was ~7.4%. The specific activity of the enzyme was 36.0 U/mg. The native enzyme should be a dimer (~280 kDa) of a protein composed with two different peptides with the mass of approximately 55.5 and 85.5 kDa, respectively. The optimal pH and temperature of this enzyme were determined at about pH 7 and 50 °C. Furthermore, EDTA revealed almost no influences on the activity.  相似文献

Natural extracts or compounds that possess both antioxidant and antiglycation activities might have great therapeutic potential for treating diabetic complications. The main purpose of this study was to evaluate the antioxidant and antiglycation properties of aqueous and EtOH (70%) extracts as well as three isolated compounds (protocatechuic acid, catechin and vanillin) from Hydnora johannis (Hydnoraceae) roots. The antioxidant activity of both extracts and isolated compounds was tested by measuring their capacity to scavenge DPPH and ABTS radicals. The glucose-induced advanced glycation end-product (AGE) formation of the extracts and isolated compounds was also carried out using in vitro glucose-bovine serum albumin (BSA-glucose) assay. Results showed that the ethanolic (70%) extract as well as isolated protocatechuic acid and catechin exhibited strong antioxidant and inhibitory effect of AGE formation. Thus, H. johannis roots with its high amount of protocatechuic acid (≈ 3.75 mg/100 g) and catechin (≈ 26.9 mg/100 g) could be a natural candidate for studies of herbal complement to diabetes treatment since it combines antioxidant and anti-AGE formation activities.  相似文献

Folic acid is antidepressant, either alone or combined with several antidepressant drugs. However, the antidepressant-like actions of folic acid combined with intra-lateral septal (LSN) infusions of neuropeptide Y (NPY) in the forced swimming test (FST) have not been tested before. Thus, systemic injections of fluoxetine (20.0 mg/kg, P < 0.05; s.c.) or 17-β estradiol (10.0 μg/rat, P < 0.05; s.c.) or oral administrations of folic acid (50.0 mg/kg, P < 0.05; 75.0 mg/kg, P < 0.05) or NPY intra-LSN (3.0 μg, P < 0.05; 3.5 μg, P < 0.05) reduced immobility of ovariectomized Wistar rats. Subthreshold doses of: folic acid (25.0 mg/kg) or 17-β estradiol (5.0 μg/rat, P < 0.05) or fluoxetine (15.0 mg/kg, P < 0.05; s.c.) combined with subthreshold doses of NPY (2.5 μg/rat, P < 0.05; intra-LSN) and these combinations produced antidepressant-like actions; which were canceled by BIBP 3226 (a NPY-Y1 receptor antagonist). It is concluded that folic acid produced antidepressant-like effects probably through the participation of the NPY Y1 receptors found in the lateral septal nuclei.  相似文献

The sap from the succulent, Desmidorchis flava (N.E.Br) Meve & Liede (Apocyanaceae), provided two new pregnane glycosides named desmiflavasides A (1) and B (2) whose structures were established from 1D and 2D NMR spectroscopic techniques and mass spectromentry (ESIMS) measurements. Desmiflavaside B (2) was tested for anticancer activity and induced a 27.4% and 33.1% growth inhibition of the breast cancer cell line (MDA MB231) at a concentration of 75 μg/ml and 100 μg/ml, respectively. Desmiflavasides A (1) and B (2) were additionally evaluated for: DPPH antioxidant activity, urease enzyme inhibition as well as for xanthine oxidase enzyme, α-glucosidase enzyme and acetylcholinesterase inhibition activities. They displayed weak antioxidant and urease enzyme inhibition activities with 15% inhibition.  相似文献

A complete randomised block design experiment was conducted to investigate the effects of benzoic acid inclusion level on nitrogen (N) metabolism, and manure ammonia (NH3) and odour emissions in finishing pigs. Sixteen boars (64 kg live weight ± 1.5 kg) were assigned to one of four dietary treatments (T) varying in benzoic acid concentration: (T1) 0 g benzoic acid/kg (as fed); (T2) 10 g benzoic acid/kg; (T3) 20 g benzoic acid/kg; (T4) 30 g benzoic acid/kg. Animals were housed in individual metabolism crates and feed was provided ad libitum. All diets were formulated to have similar concentration of digestible energy and ileal digestible lysine with benzoic acid replacing wheat in the diet. There was a linear decrease in NH3 emission (P<0.001), as the dietary benzoic acid concentration increased (141.4 mg/g versus 40.5 mg/g N intake (S.E.M. 12.1) over the 240-h storage period). However, there was no effect (P>0.05) of benzoic acid on odour concentration. Urinary nitrogen (N) excretion, total N excretion and the urinary:faecal N ratio were linearly reduced (P<0.05) with increasing benzoic acid inclusion. Furthermore, N retention increased linearly (P<0.05) as benzoic acid concentration increased from 0 g/kg to 30 g/kg in the diet. In conclusion, the inclusion of benzoic acid in the diet of finishing pigs has the potential to reduce total and urinary N excretion and the urinary to faecal N ratio. This was mirrored by reductions in manure NH3 emissions in the benzoic acid supplemented treatments.  相似文献

A method is described for construction of an amperometric xanthine biosensor based on covalent Immobilization of xanthine oxidase (XOD) onto citrate capped silver nanoparticles deposited on Au electrode surface through cysteine self assembled monolayers (SAM). The biosensor showed optimum response within 5 s at pH 7.0 and 35 °C, when polarized at 0.5 V vs. Ag/AgCl. The linear working range of biosensor for xanthine was from 2 to 16 μM, with a detection limit of 0.15 μM and sensitivity of 0.17 mA/μM/cm2. The mean analytical recovery of exogenously added xanthine in fish meat extract (5 g/l and 10 g/l) was 96.2 ± 2.3% and 95.2 ± 3.4%, respectively. Within and between batches coefficients of variation were <2.6% and <3.4%, respectively. The biosensor measured xanthine in fish, chicken, pork, and beef meat. The enzyme electrode lost 20% of its initial activity after its regular 180 uses over a period of 60 days, when stored at 4 °C in dry state.  相似文献

Cinnamon bark has been reported to be effective in the alleviation of diabetes through its antioxidant and insulin-potentiating activities. The water-soluble polyphenolic oligomers found in cinnamon are thought to be responsible for this biological activity. In this study, the hypoglycemic activity of a polyphenolic oligomer-rich extract from the barks of Cinnamomum parthenoxylon (Jack) Nees was studied in normal, transiently hyperglycemic, and streptozotocin (STZ)-induced diabetic rats. Oral administration of the extract at doses of 100, 200, and 300 mg/kg body wt. caused significant changes in body weight loss and fasting blood glucose levels of normal rats. In STZ-induced diabetic rats, after administration of the extract at doses of 100, 200, and 300 mg/kg body wt. over 14 days, the blood glucose levels were decreased by 11.1%, 22.5%, and 38.7%, respectively, and the plasma insulin levels were significantly increased over pre-treatment levels. In an oral glucose tolerance test, the extract produced a significant decrease in glycemia 90 min after the glucose pulse. These results suggest that Cinnamomum parthenoxylon polyphenolic oligomer-rich extract could be potentially useful for post-prandial hyperglycemia treatment.  相似文献

We evaluated differences in composition of Iuka gamagrass (Tripsacum dactyloides L.) hay harvested at 06:00 (AM harvest) or 18:00 h (PM harvest), and measured how protein supplementation and time of harvest interact to affect the voluntary intake, digestibility, and N balance of goats. Boer cross wethers (n = 28; 24 kg) were randomly assigned to be fed supplement (310 g/kg of crude protein (CP), fed at 110 g/kg of dry matter (DM) intake, 14 goats) or no supplement (14 goats). Within supplemented or not supplemented groups, goats were randomly assigned to a crossover design of AM harvest (seven goats) or PM harvest (seven goats), and housed individually in metabolism crates with free access to water and mineral blocks. They were fed twice daily, with supplement offered 30 min prior to the morning feedings. After a 7-d adaptation, voluntary intake (goats were offered 1100 g/kg of previous day's intake) was measured for 14 d, followed by a 4-d adjustment phase to equalize DM offered between periods, and finally a 5-d digestion and balance phase. After Period 1, goats were switched to their new hay harvest times, and the protocol was repeated. Compared to the AM harvest, the PM harvest had higher (P<0.03) proportions of total nonstructural carbohydrates (TNC, 70.8 g/kg DM versus 59.0 g/kg DM), monosaccharides (37.0 g/kg DM versus 28.6 g/kg DM), di- and polysaccharides (18.5 g/kg DM versus 15.4 g/kg DM) and less neutral detergent fiber (NDF, 700 g/kg versus 710 g/kg). Crude protein (79 g/kg DM) and starch (15.2 g/kg DM) were similar for the PM and AM harvest. Dry matter digestibility was higher (P<0.03) for the PM versus AM harvests (555 g/kg DM versus 531 g/kg DM) and for supplemented versus not supplemented (563 g/kg DM versus 522 g/kg DM). Voluntary gamagrass DM intake (550 g/d versus 548 g/d) and calculated total digestible DM intake (327 g/d versus 313 g/d) were similar for the PM and AM harvest. However, total digestible DM intake during the digestion and balance phase was higher (P<0.01) for the PM versus AM harvest (317 g/d versus 299 g/d). Time of harvest did not affect N intake, digestion, or calculated retention. Compared to no supplementation, the supplement improved (P<0.01) N digestion (6.1 g/d versus 3.7 g/d) and retention (2.2 g/d versus 1.1 g/d). The PM harvest increased DM digested, largely TNC and digestible DM intake by goats due to increased TNC and not because of a 2% increase in DM intake. Providing a protein supplement had very limited effects on intake and digestibility of gamagrass.  相似文献

The aim of present work was to assess the total phenolic content (TPC), total flavonoid content (TFC), phenolic compounds and antioxidant properties of various extracts of three Ceropegia spp.: Ceropegia spiralis, Ceropegia panchganiensis and Ceropegia evansii from Western Ghats of India. TPC of the samples varied from 0.3 ± 0.2 to 28.5 ± 0.3 mg TAE/g FW, whereas, TFC of the samples ranged between 0.1 ± 0.1 and 15.3 ± 0.3 mg RE/g FW. The major phenolic compounds identified were gallic acid, vanillin, cathechol and ferulic acid. All the extracts possess 1,1-diphenyl-2-picryl hydrazyl (DPPH) radical scavenging activity, ferric reducing antioxidant power (FRAP) as well as metal chelating ability and this was also supported by significant correlation with TPC and TFC. To the best of our knowledge, this is the first paper presenting comprehensive data on TPC, TFC, phenolic profile and antioxidant properties of the Ceropegia spp.  相似文献

The harmful effects of the 3,4-dihydroxy-l-phenylalanine (l-dopa) in Mucuna pruriens (Velvet bean) seeds have limited its use as a protein supplement for monogastrics and humans. Little is known about the extent of metabolism of Mucuna l-dopa in ruminants or its accumulation in ruminant tissues consumed as food by humans. This study aimed to determine if replacing soybean meal (SB) with Mucuna increases concentrations of l-dopa in rumen fluid, blood and muscle tissue of lambs. Twenty-seven RM lambs (RM; initial body weight, BW = 33.8 ± 5.44 kg) and 12 Florida Native (FN; initial BW = 24.9 ± 8.63 kg) lambs were assigned to four treatments and fed a basal diet of coastal bermudagrass hay, corn grain, and liquid molasses for 42 (FN) or 49 days (RM) and then slaughtered. Dietary supplements were formulated by substituting 0 (SB), 330 (Lo), 670 (Med) or at least 1000 (Hi) g/kg of SB with rolled Mucuna seeds (l-dopa, 24 g/kg dry matter, DM). Body weight was measured weekly and carcass characteristics and concentrations of l-dopa in rumen fluid, blood and the sterno-mandibularis muscle were measured at slaughter when concentrations of blood urea N, glucose, haptoglobin and cerruloplasmin were also measured. Lambs fed SB had higher (P<0.05) average daily gain than those fed Mucuna (0.20 versus 0.15 kg/day for RM; 0.21 versus 0.14 kg/day for FN). However, concentrate protein source did not affect dressing and concentrations of blood urea N, or blood glucose. Feeding the Hi diet versus the SB diet did not increase concentrations of blood cerruloplasmin, or l-dopa or concentrations of l-dopa metabolites in blood. No l-dopa was found in the ruminal fluid of the lambs and l-dopa concentrations in the sterno-mandibularis muscle were low (i.e., <5 ng l-dopa/g), and unaffected (P>0.05) by diet. Ingested Mucuna l-dopa was extensively metabolized in lambs, and did not accumulate to toxic levels in muscle tissues.  相似文献

The antioxidant activities of isoorientin-6″-O-glucoside were studied using various models. Isoorientin-6″-O-glucoside was more potent than Trolox, probucol and butylated hydroxytoluene (BHT) in reducing the stable free radical 1,1-diphenyl-2-picrylhydrazyl (DPPH). It also scavenged superoxide anion, peroxyl and hydroxyl radicals that were generated by xanthine/xanthine oxidase, 2,2′-azobis(2-amidinopropane) dihydrochloride (AAPH) and Fe3+–ascorbate–EDTA–H2O2 system, respectively. The IC50 value, stoichiometry factor and second-order rate constant were 9.0 ± 0.8 μM, 1.8 ± 0.1 and 2.6 × 1010 M−1 s−1 for superoxide generation, peroxyl and hydroxyl radicals. However, isoorientin-6″-O-glucoside did not inhibit xanthine oxidase activity or scavenge hydrogen peroxide (H2O2), carbon radical or 2,2′-azobis(2,4-dimethylvaleronitrile) (AMVN)-derived peroxyl radical in hexane. Isoorientin-6″-O-glucoside inhibited Cu2+-induced oxidation of human low-density lipoprotein (LDL) as measured by fluorescence intensity, thiobarbituric acid-reactive substance formation and electrophoretic mobility. Since isoorientin-6″-O-glucoside did not possess pro-oxidant activity, it may be an effective water-soluble antioxidant that can prevent LDL against oxidation.  相似文献

Milk fatty acid (FA) composition of dairy cows from two grazing studies was examined. In the first study, effects of concentrate supplementation and pasture allowance were evaluated using 20 multiparous Holstein cows in five 4 × 4 Latin squares. The four treatments resulted from the combination of two pasture allowances (i.e., low, 25 versus high, 40 kg dry matter/cow/day) and two concentrate supplementation levels (i.e., 0 versus 1 kg concentrate/4 kg milk). No interactions occurred between concentrate supplementation and pasture allowance for milk FA composition. Concentrate supplementation increased short-chain FA content, and reduced the content of long-chain FA, trans11 C18:1, and cis9, trans11 conjugated linoleic acid (CLA) (1.36 versus 1.18 g/100 g). Concentrate supplementation increased saturated FA (58.6 versus 54.0 g/100 g) and reduced unsaturated FA content (39.9 versus 43.8 g/100 g). Grazing at high pasture allowance increased short-, medium-, and long-chain FA content, without affecting cis9, trans11 CLA content. Saturated FA content was higher (57.1 versus 55.6 g/100 g), and unsaturated FA content was lower (41.3 versus 42.5 g/100 g), when cows grazed at high pasture allowance. Concentrate supplementation reduced unsaturated FA and cis9, trans11 CLA in milk of dairy cows grazing at two pasture allowances. In the second study, two experiments evaluated effects of different energy supplements in grazing dairy cows. In Experiment 1, 25 multiparous Holstein cows were assigned to a cracked corn (CC) or a steam flaked corn (SFC) supplement containing 667 g/kg of corn grain plus a pelleted protein/mineral supplement. In Experiment 2, 22 multiparous Holstein cows were assigned to a ground corn grain (GC) or a non-forage fiber (NFF) supplement. The GC supplement contained 850 g/kg of corn grain, and the NFF supplement contained 440 g/kg of non-forage fiber sources (i.e., beet pulp, soyhulls, wheat middlings) that partially replaced corn grain. Milk FA composition was not affected by corn processing or carbohydrate source. The content of short-, medium-, long-chain FA, and cis9, trans11 CLA (2.5 g/100 g in Experiment 1; 2.1 g/100 g in Experiment 2) was similar between supplements in both experiments. The type of supplement did not affect the content of saturated (62.6 g/100 g in Experiment 1; 65.4 g/100 g in Experiment 2) and unsaturated FA (37.5 g/100 g in Experiment 1; 34.6 g/100 g in Experiment 2). Species of the family Combretaceae are used extensively in traditional medicine against inflammation and infections, and although antibacterial activity has been reported in non-polar extracts, further rationale for the widespread use of the Combretaceae is expected to exist. Methanol extracts of leaves of ten different Combretum species were evaluated for antioxidant activity by spraying TLC chromatograms of each leaf extract with 2,2-diphenyl-1-picrylhydrazyl (DPPH). Compounds with antioxidant activity were detected by bleaching of the purple DPPH colour. Leaf extracts of Combretum apiculatum subsp. apiculatum had the most antioxidant compounds. This species was consequently selected for phytochemical investigation. A DPPH assay-directed fractionation of the leaf extracts of C. apiculatum led to the isolation of four antioxidant compounds from the ethyl acetate and butanol soluble fractions. The structures of the compounds were determined by spectroscopic analyses (1H-NMR, 13C-NMR and MS) and identified as: cardamonin (1), pinocembrin (2), quercetrin (3) and kaempferol (4). In a quantitative antioxidant assay, the more polar fractions (ethyl acetate and butanol) obtained by solvent–solvent fractionation had the highest antioxidant activity among the solvent fractions obtained from C. apiculatum, with EC50 values of 3.91 ± 0.02 and 2.44 ± 0.02 μg/ml respectively. Of the four isolated compounds, quercetrin (4) and kaempferol (3) had the strongest antioxidant activity, with EC50 values of 11.81 ± 85 and 47.36 ± 0.03 μM respectively. Cardamonin (1) and pinocembrin (2) did not demonstrate strong activity. L-ascorbic acid was used as standard antioxidant agent (EC50 = 13.37 ± 0.20 μM or 2.35 μg/ml). The cytotoxicity of cardamonin and pinocembrin was evaluated on Vero kidney cells using the MTT (3-(4,5-dimethylthiazol)-2,5-diphenyl tetrazolium bromide) assay with berberine as positive control. At concentrations higher than 50 μg/ml of cardamonin or pinocembrin, the cells were not viable. Cardamonin was more toxic (LC50 = 1.97 μg/ml) than pinocembrin (LC50 = 29.47 μg/ml) and even the positive control, berberine (LC50 = 12.35 μg/ml).  相似文献

Polyphenols are the predominant ingredients in apple seeds. However, few data are available on the phenolic profile or antioxidant activity in apple seeds in previous researches. In this study, low-molecular-weight phenolic compounds and antioxidant activity in seeds, peels, and flesh of seven apple cultivars grown in northwest China were measured and analyzed using HPLC and FRAP, DPPH, ABTS assays, respectively. HPLC analysis revealed phloridzin as the dominant phenolic compound in the seeds with its contents being 240.45–864.42 mg/100 gDW. Total phenolic content (TPC) measured by the Folin–Ciocalteu assay in apple seed extracts of seven cultivars ranged from 5.74 (Golden Delicious) to 17.44 (Honeycrisp) mgGAE/gDW. Apple seeds showed higher antioxidant activity than peels or flesh; antioxidant activity in seeds varied from 57.59 to 397.70 μM Trolox equivalents (TE)/g FW for FRAP, from 37.56 to 64.31 μM TE/g FW for DPPH, and from 220.52 to 708.02 μM TE/g FW for ABTS. TPC in apple seeds was significantly correlated with all three assays. Principal component analysis (PCA) indicated that Honeycrisp was characterized with high contents of total polyphenols and phloridzin. Our findings suggest that phenolic extracts from apple seeds have good commercial potential as a promising antioxidant for use in food or cosmetics.  相似文献

The objective was to identify legume shrub species for development of agroforestry technologies based on seed and forage (leaves and twigs < 10 mm diameter) yield, and determinants of forage quality. Ten individual plants of Bituminaria bituminosa ‘Ecotypes 1’, B. bituminosa ‘Ecotypes 2’, Medicago citrina, and M. arborea from Spain; Colutea istria and Onobrychis aurantiaca from Syria; C. istria from Jordan; Chamaecytisus mollis from Morocco; and Coronilla glauca from France were randomly selected from plots established in a non-tropical dryland environment in northwest Syria in 2000. Five individual plants of each species were cut back to 0.5 m above ground in March 2004. Coppice regrowths were pruned in December 2004 and April 2005 to determine forage yield and proportion of forage in the total above ground biomass (PEFB). Forage samples were analyzed for concentrations of crude protein (CP), lignin(sa), acid detergent fibre (ADFom), neutral detergent fibre (aNDFom), in vitro organic matter (OM) digestibility (IVOMD), and in vitro 24 h gas production (IVGP24h). Matured seeds were hand harvested from the remaining five plants of each species to estimate seed yield. Forage (21–250 kg DM/ha) and seed (0–200 kg DM/ha) yields; PEFB (0.22–0.96); and concentrations of CP (85–115 g/kg DM), lignin(sa) (14–42 g/kg DM), ADFom (94–170 g/kg DM), aNDFom (122–217 g/kg DM), IVOMD (456–617 g/kg OM), and IVGP24h (27–42 ml 200 mg/DM) varied (P<0.05) among shrub species. The IVOMD and IVGP24h were positively correlated (r = 0.75, P<0.032), whereas IVOMD and IVGP24h were negatively correlated with ADFom, lignin(sa) and aNDFom. In terms of forage and seed yields and determinants of forage quality, C. istria from Jordan, M. arborea, B. bituminosa ‘Ecotype-2’, C. istria and O. aurantiaca have higher potential than C. mollis, C. glauca and B. Bituminosa ‘Ecotype-1’ for the development of agroforestry technologies in non-tropical dry areas.  相似文献
