Five serially sectioned tissue slices (400 microns) from the preoptic area/hypothalamus of postnatal day 4 rats were cultured using a slice explant roller culture technique. After 18 days in culture, these slices thinned sufficiently to allow immunocytochemical and in situ hybridization histochemical assays for LHRH peptide and LHRH mRNA, respectively. Large numbers of neurons containing mRNA encoding LHRH were detected in these slices using in situ hybridization histochemistry (ISHH). These 35S-labeled cells were distributed in the cultured slices in a pattern similar to that found with LHRH immunocytochemistry and ISHH in vivo, indicating that LHRH neurons were maintained in these cultures in an organotypic manner. Densitometric single cell analyses after ISHH of the culture slices were performed using a Loats image analysis system, so as to provide a density value per cell (density/cell). Comparisons of these density values from the slice explants cultured in presence or absence of 10(-7) M estradiol found that: 1) under basal (control) culture conditions there were no consistent differences in the frequency distributions of the density/cell values between all the five slices derived from either male or female rats, 2) mean density/culture values under control conditions did not differ significantly between slices and sexes, 3) the presence of estradiol in the culture media resulted in an overall decrease in density/cell values, with the most significant decrease occurring in slice 3 which is comparable to the level of the organum vasculosum lamina terminalis/rostral preoptic area (OVLT/rPOA) in vivo, and 4) this decrease in density/cell values in slice 3 due to estradiol treatment, was greater in cultures derived from female vs. male tissues.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Starch is the main reserve compound in woody plant species. Changes in starch content are clear indicators of a variety of plant developmental processes. Thus, carbohydrate extraction and other analytical methods have been widely used to measure changes in starch content. However, the use of these methods can be limited by the fact that starch is often compartmentalized in very small portions of tissue. While changes in these small structures can be histochemically characterized and localized under the microscope, they cannot be quantified. As an alternative, an image analysis system attached to a microscope has been developed to detect quantitative variations in starch in particular tissues or cells. The system has been successfully used to study the differences in starch content of sections from pistillar structures in apricot ( Prunus armeniaca L.). The procedure is based on the measurement of the optical density of black and white images obtained from the microscope. Two staining methods, I2KI (potassium iodide-iodine) and PAS (periodic acid Schiff's reagent), and two embedding techniques, paraffin and JB4 plastic resin, were compared. The best results were obtained using I2KI-stained sections of paraffin-embedded material. Since the procedures used are non-destructive for the tissues studied, additional information can be obtained, on the same section, by the subsequent use of additional stains. The method described here can be used to detect quantitative variations in starch content under the microscope in different plant tissues and thus to follow changes in starch reserves in small structures.  相似文献

Three experiments were conducted to study effects of amylose/amylopectin ratios and starch particle size on ruminal digestion characteristics of barley starch using an automated in vitro gas production system. In Experiment 1, starch digestion characteristics were measured in 12 barley cultivars with different amylose/amylopectin ratios, both as milled grain and as purified starch isolated from the original grain samples. The same 12 cultivars, harvested 1 year later from the same locations, were used in Experiment 2. Gas production was measured in milled samples, and in neutral detergent fibre (NDF) extracted from the same samples. The objective of this approach was to estimate gas production from neutral detergent solubles (NDS) as an approximation of starch. This was done by subtracting the NDF gas curve from the total gas production curve. In Experiment 3, starch digestion characteristics were measured for large and small starch granules from nine of the original cultivars used in Experiment 1. The gas curves obtained were fitted to a three-pool Gompertz model, and the effective rate of digestion (kd) was estimated with a two-compartmental rumen model. In Experiment 1, the effective starch kd for milled barley and purified starch were 0.122 and 0.118/h, respectively. Barley cultivars with low amylose (LA) had a higher effective kd (0.148/h) compared with cultivars with normal amylose (NA) (0.115/h) and high amylose (HA) (0.102/h) (P=0.010). Results obtained with milled barley were supported by the purified barley starch sample results, but differences were smaller and only numerically different. In Experiment 2, the ranking of the amylose groups was consistent with those in Experiment 1 (i.e., LA > NA > HA) (P=0.096). However, these differences were not reflected in the effective kd for the NDS fraction (P=0.366). Thus, factors other than those related to starch per se, or other structural features, are apparently important. Barley cultivars in the LA group had a higher effective kd for aNDF (0.098/h) than did NA and HA barley (0.060 and 0.055/h, respectively). Thus, the effect of the amylose group on the effective kd for aNDF corresponded well with the milled barley results. The NDF fraction, directly or indirectly, has a clear impact on the ruminal digestion rate of barley starch. There was no difference in the effective kd for starch between the small (0.126/h) and large (0.129/h) starch granules.  相似文献

在聚类分析的基础上,研究云南普洱季风常绿阔叶林主要物种非结构性碳水化合物(NSC)及其组分浓度、分配和季节性动态在林冠层、亚冠层和林下层间的变化特征.结果表明:亚冠层中可溶性糖及NSC浓度最高,分别为3.9%和13.3%,可溶性糖淀粉比在林下层最低,为0.76,而淀粉浓度则在各林层间无显著性差异.3个林层的可溶性糖均主要分配在叶片中,淀粉和NSC主要分配在根中.亚冠层中叶片和树干的可溶性糖浓度显著高于林冠层和林下层,枝和根的可溶性糖浓度在3个林层间无显著性差异;叶片的淀粉浓度则随林层高度降低而增加,但根淀粉浓度则是在林下层最低,为10.7%,枝和树干的淀粉浓度在3个林层间无显著差异;叶片NSC浓度为林冠层(10.7%)显著低于亚冠层(12.3%)和林下层(12.0%),但根的NSC浓度在林下层中最低,为14.2%;林下层叶片、枝、树干中可溶性糖淀粉比值均最低,但根的可溶性糖淀粉比值最低值出现在林冠层(0.79).3个林层NSC及其组分均存在显著的季节性变化,可溶性糖及可溶性糖淀粉比均为雨季显著高于旱季,而淀粉和NSC浓度则均在旱季中较高.不同林层NSC及其组分浓度的差异反映了不同高度树种碳利用策略的差异,部分地解释了物种的共存机制.  相似文献

The physico-chemical, morphological, thermal, pasting, textural, and retrogradation properties of the starches isolated from four traditional Taewa (Maori potato) cultivars (Karuparera, Tutaekuri, Huakaroro, Moemoe) of New Zealand were studied and compared with starch properties of a modern potato cultivar (Nadine). The relationships between the different starch characteristics were quantified using Pearson correlation and principal component analysis. Significant differences were observed among physico-chemical properties such as phosphorus content, amylose content, swelling power, solubility and light transmittance of starches from the different potato cultivars. The starch granule morphology (size and shape) for all the potato cultivars showed considerable variation when studied by scanning electron microscopy and particle size analysis. Starch granules from Nadine and Moemoe cultivars showed the presence of large and irregular or cuboid granules in fairly high number compared with the starches from the other cultivars. The transition temperatures (To; Tp; Tc) and the enthalpies (ΔHgel) associated with gelatinization suggested differences in the stability of the crystalline structures among these potato starches. The Moemoe starch showed the lowest To, while it was higher for Tutaekuri and Karuparera starches. Pasting properties such as peak, final and breakdown viscosity and texture profile analysis (TPA) parameters of starch gels such as hardness and fracturability were found to be higher for Nadine and Huakaroro starches. The Nadine and Huakaroro starch gels also had lower tendency towards retrogradation as evidenced by their lower syneresis (%) during storage at 4 °C. Principal component analysis showed that the Tutaekuri and Nadine cultivars differed to the greatest degree in terms of the properties of their starches.  相似文献

In avocado, only a very small fraction of the flowers are able to set fruit. Previous work in other woody perennial plant species has shown the importance of carbohydrates accumulated in the flower in the reproductive process. Thus, in order to explore the implications of the nutritive status of the flower in the reproductive process in avocado, the starch content in the pistil has been examined in individual pollinated and non‐pollinated flowers at anthesis and during the days following anthesis. Starch content in different pistilar tissues in each flower was quantified with the help of an image analysis system attached to a microscope. Flowers at anthesis were rich in highly compartmentalized starch. Although no external morphological differences could be observed among flowers, the starch content varied widely at flower opening. Starch content in the ovary is largely independent of flower size because these differences were not correlated with ovary size. Differences in the progress of starch accumulation within the ovule integuments between pollinated and non‐pollinated flowers occurred concomitantly with the triggering of the progamic phase. The results suggest that starch reserves in the ovary could play a significant role in the reproductive process in avocado.  相似文献

Regular (25% amylose) and waxy corn starches blended to various ratios with Eastar Bio Copolyester 14766 (EBC) were extruded into loose-fill foams using a twin screw extruder. Included in this study were two types of corn starch, three levels of EBC content, and three levels of starch moisture content. Waxy starch produced foams with greater radial expansions and lower unit and bulk densities than regular starch. Regular starch foams had lower water solubility indices (WSIs) than waxy starch foams. Foams made of both types of starch possessed similar mechanical properties. No differences were observed in compressibilities and spring indices of either waxy or regular starch foams. Higher levels of EBC addition resulted in less radial expansion and higher unit and bulk densities. Foams made with 10% EBC had higher compressibility than foams containing 25% EBC. Spring indices of single-piece samples and compressibilities and spring indices of bulk samples were not affected by the differences in the level of EBC addition. At 19% and 22% of moisture contents, foams had greater radial expansion than at 25% moisture content. Unit and bulk densities were not affected by variations in moisture content. At 22% moisture content, a lower WSI of 18.5% was obtained. At 22% moisture content, softer foams were produced. At all three levels of moisture content, no differences were detected in the spring indices.  相似文献

Root nodule ontogeny was followed in different parts of the root system of field peas (Pisum sativum L. cv. Century) to investigate the contribution to total nitrogen fixation by different nodule subpopulations. Seed-inoculated plants were grown to maturity in controlled-environment growth chambers. In a flow-through system nitrogenase activity (H2-evolution in air) and nodulated-root respiration (net CO2-evolution) were measured weekly or biweekly in different parts (top and mid) of the root system. Root nodule extracts were assayed for total soluble cytosolic protein, total heme, proteolytic capacity (at pH 7.0), soluble carbohydrates and starch. Total nitrogenase activity and nodule respiration were higher in the top zone, which was explained by differences in root and nodule mass. Nodule specific nitrogenase activity was similar in both zones, and gradually declined throughout the experiment. No differences were found between nodule subpopulations in the dry-matter specific concentrations of glucose, fructose, sucrose or starch. Neither did nodule concentrations of protein or leghemoglobin differ between the zones. Throughout reproductive growth, no decline was found in total or nodule specific nitrogenase activity, in any of the nodule subpopulations. Growth of the root nodules continued throughout the experiment, though growth of shoot and roots had ceased. The data gives no support for carbohydrate limitation in root nodules during pod-filling, since nodule respiration remained high, the concentration of soluble carbohydrates increased significantly, and the amount of starch was not reduced. We conclude that when this symbiosis is grown under controlled conditions, nitrogenase activity in nodules sub-sampled from the crown part of the root system is representative for the whole nodule population.  相似文献

实验以鱼粉和酪蛋白为主要蛋白源、以鱼油和豆油为主要脂肪源(鱼油/豆油, 1/1), 设计了7组不同玉米淀粉水平(6%、12%、18%、24%、30%、36%和42%)的等氮(35%)等脂(8%)饲料来饲喂芙蓉鲤鲫(Carassiusauratus var Furong carp ♀Cyprinus carpio red crucian carp ♂)幼鱼(8.940.59) g以确定其最适饲料淀粉水平。实验周期为61d。结果表明, 随饲料玉米淀粉水平的增加实验鱼增重率、特定生长率和饲料效率先上升(P0.05)而后趋于稳定, 18%42%饲料玉米淀粉组之间差异不显著(P0.05)。鱼体蛋白沉积率随饲料玉米淀粉水平的增加而增加, 30%和42%处理组显著高于6%和12%处理组(P0.05)。血浆葡萄糖没有受饲料玉米淀粉水平的影响(P0.05), 血浆总胆固醇含量在30%处理组最高(P0.05), 血浆甘油三酯随饲料玉米淀粉水平的增加而上升(P0.05)。肝糖原随饲料玉米淀粉水平的增加而上升, 36%处理组显著高于6%和12%处理组(P0.05)。肌糖原和肠道淀粉酶活性随饲料玉米淀粉水平增加而显著上升(P0.05)。通过特定生长率和饲料淀粉水平的折线法分析表明, 芙蓉鲤鲫幼鱼达到最大生长时饲料淀粉水平为27.47%。  相似文献

Exposure to low but nonfreezing temperatures induces the breakdown of starch and the accumulation of sucrose, glucose and fructose in potato tubers, a complex phenomenon known as low-temperature sweetening (LTS). A kinetic model for the degradation of starch to sucrose, fructose, glucose, hexose phosphates and carbon dioxide in 2 degrees C-stored mature Solanum tuberosum cv. Norchip (LTS-sensitive) and Solanum tuberosum seedlling ND860-2 (LTS-tolerant) tubers is presented in this work. Analysis of sugar accumulation data in tubers grown in 1993 and 1994 showed no significant differences in the rates of conversion of starch to hexose phosphates and hexose phosphates to sucrose for both cultivars (P > 0.05). The rate constant corresponding to invertase activity was 2.3 day(-1) for Norchip tubers and 1.1 day(-1) for ND860-2 tubers grown in 1993 (P < or = 0.05); however, no significant differences were observed in invertase activity for 1994-grown tubers (P > 0.05). The accumulation of the reducing sugars fructose and glucose was found to be dependent on the relative difference in rate constants corresponding to invertase activity and glycolytic/respiratory capacity. This difference was 3-4 fold greater for Norchip in 1993, and 4-6 fold greater for Norchip in 1994, than for ND860-2 (P < or = 0.05). Results from the analysis also suggest that the amount of available starch for degradation was greater in Norchip tubers than ND860-2 tubers (P < or = 0.05). Our analysis suggests that tubers with decreased invertase activity coupled to increased glycolytic/respiratory capacity should be more tolerant to low-temperature stress.  相似文献

The nature of the cytoplasmic pathway of starch biosynthesis was investigated in the model heterotrophic dinoflagellate Crypthecodinium cohnii. The storage polysaccharide granules were shown to be composed of both amylose and amylopectin fractions with a chain length distribution and crystalline organization very similar to those of green algae and land plant starch. Preliminary characterization of the starch pathway demonstrated that C. cohnii contains multiple forms of soluble starch synthases and one major 110-kDa granule-bound starch synthase. All purified enzymes displayed a marked substrate preference for UDP-glucose. At variance with most other microorganisms, the accumulation of starch in the dinoflagellate occurs during early and mid-log phase, with little or no synthesis witnessed when approaching stationary phase. In order to establish a genetic system allowing the study of cytoplasmic starch metabolism in eukaryotes, we describe the isolation of marker mutations and the successful selection of random recombinant populations after homothallic crosses.  相似文献

PurposeTo define a method and investigate how the adjustment of scan parameters affected the image quality and Hounsfield units (HUs) on a CT scanner used for radiotherapy treatment planning. A lack of similar investigations in the literature may be a contributing factor in the apparent reluctance to optimise radiotherapy CT protocols.MethodA Catphan phantom was used to assess how image quality on a Toshiba Aquilion LB scanner changed with scan parameters. Acquisition and reconstruction field-of-view (FOV), collimation, image slice thickness, effective mAs per rotation and reconstruction algorithm were varied. Changes were assessed for HUs of different materials, high contrast spatial resolution (HCSR), contrast-noise ratio (CNR), HU uniformity, scan direction low contrast and CT dose-index.ResultsCNR and HCSR varied most with reconstruction algorithm, reconstruction FOV and effective mAs. Collimation, but not image slice width, had a significant effect on CT dose-index with narrower collimation giving higher doses. Dose increased with effective mAs. Highest HU differences were seen when changing reconstruction algorithm: 56 HU for densities close to water and 117 HU for bone-like materials. Acquisition FOV affected the HUs but reconstruction FOV and effective mAs did not.ConclusionsAll the scan parameters investigated affected the image quality metrics. Reconstruction algorithm, reconstruction FOV, collimation and effective mAs were most important. Reconstruction algorithm and acquisition FOV had significant effect on HU. The methodology is applicable to radiotherapy CT scanners when investigating image quality optimisation, prior to assessing the impact of scan protocol changes on clinical CT images and treatment plans.  相似文献

The metabolic function of the plastidic ATP/ADP transporter (AATP) in heterotrophic plastids was examined in transgenic potato plants that exhibited increased or decreased amounts of the protein. Altered mRNA levels correlated with activities of the plastidic ATP/ADP transporter. Potato tubers with decreased plastidic ATP/ADP transporter activities exhibited reduced starch contents whereas sense lines accumulated increased amounts of tuber starch. Starch from wild-type tubers had an amylose content of 18.8%, starch from antisense plants contained 11.5–18.0% amylose, whereas starch from sense plants had levels of 22.7–27.0%. The differences in physiological parameters were accompanied with altered tuber morphology. These changes are discussed with respect to the stromal ATP supply during starch biosynthesis.  相似文献

对64份淮山种质资源品质性状进行了分析,结果表明:铁、氨基酸、总皂甙、锌含量的变异系数较大,分别为66.2%、59.9%、44.09%、42.77%;3种类型的淮山种质资源,其淀粉含量差异较大,褐苞薯蓣(23.52%)参薯(20.48%)薯蓣(12.93%);因子分析结果显示,淀粉和蛋白质含量累计贡献率达66.786%,可用这2个主成分较好地代替6个品质特性来评价与评判淮山品质;相关性分析表明,淀粉含量与总皂甙含量呈显著正相关,蛋白质含量与总皂甙含量呈极显著正相关,淀粉、蛋白质含量这2个主成分与总皂甙含量之间存在相互促进的效用;聚类分析表明,整个淮山资源可分为3大类,分别为高淀粉型、低淀粉型、高铁型。淮山种质资源品质性状分析,可为日后进行淮山种质资源的创新利用、新品种的选育提供可靠的依据。  相似文献

植物光合作用产生的非结构性碳水化合物(NSCs)水平可以反映植物和生态系统对环境变化的响应程度。近年来, 草原极端干旱事件的发生频率和持续时间增加趋势明显, 对生态系统结构和功能产生深远影响。该研究以内蒙古呼伦贝尔草甸草原为研究对象, 通过连续4年减少66%生长季降水量的控制实验来模拟极端干旱事件, 分析草原6种优势物种和植物功能群NSCs各组分对极端干旱的响应规律与机制。结果显示, 由于植物生物学、光合特性以及生理生态等特性的差异, 不同物种对干旱胁迫的响应具有明显差异。这表明草地植物NSCs组分及其利用策略对干旱胁迫的响应具有物种特异性, 从而导致其生物量的不同响应。将6种植物分为禾草和非禾草两类, 发现干旱显著增加了禾草的淀粉含量, 但对其可溶性糖含量无显著影响; 相反, 干旱显著增加了非禾草功能群的可溶性糖含量, 对其淀粉含量无显著影响, 表明不同功能群采取了不同的干旱应对策略。禾草选择将光合作用固定的能量进行储存以应对干旱胁迫, 其生物量对干旱响应不敏感; 而非禾草选择将能量以可溶性糖的形式直接供植物生长利用以及抵御干旱胁迫, 其生物量对干旱响应较为敏感。这一发现可为预测在全球气候变化背景下草甸草原生态系统结构与功能对极端干旱的响应提供科学参考。  相似文献

To examine the effect of two types of resistant starch on blood glucose and insulin levels, colonic events, hypolipidemic actions and humoral immune responses, Sprague-Dawley streptozotocin-induced diabetic rats were fed diet containing resistant starch from corn or rice. The marked body weight loss by inducing diabetes was not recovered by feeding resistant starch, even though there are no differences in food intakes compared to the non-diabetic control rats. No significant effect of resistant starch feeding on blood glucose and insulin was found. Even though the length of small intestines, and cecum, colon and rectum together with the tissue weight of cecum were not affected by feeding resistant starch, the intestinal transit time was markedly shortened by both types of resistant starch and resistant starch from corn had a more pronounced effect. The short chain fatty acids in the intestinal contents did not appear to be different among the groups. Nonetheless, both of resistant starch from corn and rice significantly lowered plasma total lipid and cholesterol concentrations compared to the diabetic control. The total liver cholesterol lowering effect was observed with resistant starch from rice. Neither immunoglobulin G nor C(3) were influenced by resistant starch.  相似文献

The influence of partial replacement of starch by sucrose on dietary cholesterol-induced serum lipoprotein responses was examined in 10 male cynomolgus monkeys (Macaca fascicularis). In a crossover design two semipurified diets provided either starch or starch and sucrose (1:1) as carbohydrate (49% by calories) with 0.4 mg cholesterol/kcal. Six weeks of starch + sucrose diet resulted in significantly reduced levels (mean +/- SE, mg/dl) of serum total cholesterol (264 +/- 9 vs 244 +/- 8) and apo B (110 +/- 6 vs 96 +/- 6) when compared with starch diet, whereas serum triglyceride levels remained similar between diets. With respect to changes in lipids and apolipoproteins (A-I or B) of very low (VLDL), low (LDL), intermediate (IDL), and high (HDL) density lipoproteins, starch + sucrose diet significantly increased VLDL-apo B (+34%), and decreased LDL-cholesterol (-18%) and LDL-apo B (-15%) as compared with starch alone; no differences were found in IDL and HDL between diets. The relative proportion of starch to sucrose in a diet appears to influence the magnitude of response of lipoproteins to dietary cholesterol.  相似文献

Rice ratooning, or the production of a second rice crop from stubble after the harvest of the main crop, is considered to be a green and resource-efficient rice production system. The present study was conducted to examine variance in amylose content (AC), grain morphology, crystal structure, and thermal properties of starch between main- and ratoon-season rice of seven varieties. Ratoon-season rice grains had higher ACs and significantly lower transition gelatinization temperatures (To, Tp, and Tc) than did main-season rice grains. The relative crystallinity and lamellar peak intensity of ratoon-season rice starch were 7.89% and 20.38% lower, respectively, than those of main-season rice starch. In addition, smaller granules with smoother surfaces and lower thermal parameters were observed in the starch of ratoon-season rice. The relative crystallinity and lamellar peak intensity of starch correlated negatively with the AC and positively with transition gelatinization temperatures. These results suggest that the superior cooking quality of ratoon-season rice is attributable to the moderate increase of grain AC, which reduces the relative crystallinity, weakens the crystal structure, and lead to a decrease in the gelatinization temperature.  相似文献

以‘津优35号’黄瓜为试材,采用裂区-再裂区设计,研究了CO2加富下水氮耦合对黄瓜叶片光合作用和超微结构的影响.主区设大气CO2浓度(400 μmol·mol-1,A)和加富CO2浓度(800±20 μmol·mol-1,E)2个CO2浓度处理,裂区设无干旱胁迫(田间持水量的95%,W)和干旱胁迫(田间持水量的75%,D)2个水分处理,再裂区设施氮量450 kg·hm-2(低氮,N1)和900 kg·hm-2(高氮,N2)2个氮素处理.结果表明: 在干旱和高氮条件下,CO2加富提高了黄瓜的株高,且使高氮下的叶面积显著增加.正常灌溉条件下,高氮处理的光合速率、气孔导度和蒸腾速率高于低氮处理,而干旱条件下则相反;CO2加富提高了黄瓜叶片的水分利用效率,并且随着施氮量的增加,其水分利用效率提高.干旱胁迫下,黄瓜近轴面气孔密度增加,而CO2加富和高氮却显著降低了气孔密度.高氮处理增加了黄瓜叶片叶绿体数量而减少了淀粉粒数量,干旱胁迫使叶绿体数量减少,但使淀粉粒数量呈上升趋势.干旱胁迫增加了叶绿体长度和宽度,显著增加了淀粉粒的大小,而高氮降低了叶绿体和淀粉粒的长度和宽度.CO2加富和高氮均使基粒厚度和片层数增加(ADN2除外),并且EDN2处理的片层数显著高于ADN2.综上所述,CO2加富和适宜的水、氮条件能促进黄瓜叶片叶绿体类囊体膜系的发育,显著增加基粒厚度和基粒片层数,有效改善黄瓜的叶绿体结构,增强光合性能,提高黄瓜植株对CO2和水、氮的吸收利用能力.  相似文献
