通过农杆菌介导法用含有抗潮霉素和 G U S 基因的双元载体将杀虫结晶蛋白基因cry I A( b) 和cry I A(c) 导入到籼、粳稻幼穗愈伤组织中,然后经过在含有不同浓度潮霉素的培养基上进行数次筛选,获得一批 Bt 转基因株。经 P C R、 Southern 杂交及 Western 印迹分析证实此二基因已整合进水稻中,饲虫试验结果表明,转基因株具有100 % 杀虫率。  相似文献

An insecticidal crystal protein gene of Bacillus thuringiensis was transferred into cabbage genome with the method of Agrobacterium infection. Cotyledons with petioles as explants were cocultivated with Agrobacterial suspension. Calli generated at the basis of petiole were subjected to selection on the MS medium containing 15-30 mg/L kanamycin (Km). About 5% explants produced calli growing continuously on the selective medium. Green shoots appeared on these calli when they were transplanted onto medium with Km and 6-BA for plant differentiation. The shoots were separated and cultivated on medium with kanamycin. About 80% shoots were rooted. Non-transformed control calli could not give normal shoots and roots and brownized and died gradually. Larvae of Pieris rapae showed poisonous symptoms: growth inhibition and mortality when fed with the leaf of the transgenic plants. In this study, six plasmids were constructed to study the effects of the tobacco Rb7 matrix attachment region (MAR) sequence on rice transgene expression. Among them, each of four plasmids contained two identical copies of the MAR sequence flanking two different reporter genes, which encode the green fluorescent protein and -glucuronidase. Two control plasmids contained no MAR sequences. Microprojectile bombardment was used to separately introduce these six plasmids into rice calli. Transgenic rice plants were regenerated, and gene expression was measured in rice leaf extracts of two-month-old transgenic plants. By comparing transgenic plants with the corresponding control plants, transgenic plants harboring each of four plasmids that included the MAR sequence resulted in average gene expression levels enhanced by 3.3-fold, 18-fold, 376-fold and 650-fold. These results suggest that the same MAR sequence can affect the expression of different genes to different extents. One goal of plant scientists and breeders is to maximize expression levels of transgenes, especially in the production of transgene-encoded protein. Therefore, inclusion of the Rb7 MAR sequence would be beneficial.  相似文献

用带有杀虫肽基因的农杆菌(Agrobacterium tum efaciens)LBA4404 转化烟草(Nicotiana tabacum )叶片,共获得30 株抗卡那霉素的再生植株. 用这些再生植株对棉铃虫(Heliothisarm igera)进行毒力测定,有3 株转杀虫肽基因植株对棉铃虫有较强抗性. 与对照相比,这3 株转基因烟草的杀虫率可达30%~45% ,并能显著抑制昆虫蜕皮和生长发育,表现出明显的抗虫作用. 以这3 株为主进行了PCR 扩增及Northern blot实验,结果表明杀虫肽基因已插入到这3 株植株的基因组中并表达出有活性的杀虫肽  相似文献

Tobacco (Nicotiana tabacum)leaves were transformed with Agrobacterium tumefaciens LBA4404 containing the insecticidal peptide gene. Thirty regenerated kanamycin resistant plants were obtained, among which three showed stronger toxicity to Heliothis armigera by feeding experiments. In comparison with feeding of the control plants, mortality of the insects fed on transgenic plants was significantly higher and the growth of the survived insects was remarkably retarded. Results of PCR Southern blot and Northern blot showed that insecticidal peptide gene has been transferred into the genome of these three plants and expressed efficiently to confer the insect resistance of the transgenic plants.  相似文献

Russian wildrye (Psathyrostachys juncea (Fisch.) Nevski) is a cool-season forage species well adapted to semi-arid climates. We are interested in developing biotechnological methods to improve this monocot forage species. Single genotype-derived embryogenic suspension cultures were established from the Russian wildrye cultivar Bozoisky-Select, and were used as target cells for biolistic transformation. A chimeric hygromycin phosphotransferase gene (hph) was used as the selectable marker, and a chimeric -glucuronidase (gusA) gene was co-transformed with hph. Resistant calli were obtained from 29% of the bombarded dishes after selection with 200 mg/l hygromycin. Plants were regenerated from 45% of the hygromycin resistant calli. Thirty-six transgenic Russian wildrye plants were recovered after microprojectile bombardment of suspension cells and subsequent hygromycin selection. The transgenic nature of the regenerated plants was demonstrated by Southern hybridization analysis using undigested and digested genomic DNA samples. When a second gene (gusA) was co-transformed with hph, a reasonably high co-transformation frequency of 78% was observed. Transgenic expression of gusA was confirmed by GUS staining of shoot and leaf tissues. Fertile transgenic plants were obtained after two winters of vernalization under field conditions. This is the first report on the generation of transgenic plants in Russian wildrye.  相似文献

pBinLK carried two insecticidal genes, pea lectin (P-Lec)gene and soybean Kunitz trypsin inhibitor (SKTI) gene, were successfully transferred into 4 upland cotton ( Gossypium hirsutum L. ) cultivars, "Xinluzao-1 ", "Xinluzhong-2", "Jihe-321" and "Liao-9" via Agrobacterium-mediated transformation. Hypocotyl segments from aseptic seedlings were used as receipient. After co-cultivation of hypocotyl segments with A. tumefac/ens (Smith et Townsend) Conn, kanamycin-resistant calli were screened, and somatic embryos and regenerated plants were obtained through various media. Transgenic cotton plants harboring two insecticidal genes were confirmed by NPT-Ⅱ ELISA, PCR and PCR Southern. rllae results of bioassay demonstrated that the transgenic plants showed significant resistance to the larvae of cotton bollwonn (Heliothis armigera Hubner).  相似文献

Robinia pseudoacacia (black locust) plants were regenerated after co-cultivation of stem and leaf segments with Agrobacterium tumefaciens strain GV3101 (pMP90) that harbored a binary vector that included genes for β-glucuronidase (GUS) and hygromycin phosphotransferase. Successful transformation was confirmed by the ability of stem and leaf segments to produce calli in the presence of hygromycin, by histochemical and fluorometric assays of GUS activity in plant tissues, and by Southern blotting analysis. In this transformation system, about 2 months were required for regeneration of transgenic plants from stem and leaf segments. The frequency of transformation from stem segments was approximately 24%, and the morphology of regenerated plants resembled that of the original parental strain. Oryzacystatin (OC) is a proteinaceous cysteine proteinase inhibitor involved in the biodefense of rice seeds. To create transgenic rice plants with increased OC activity, we introduced an OC expressing vector into rice protoplasts and obtained transformed calli. The expression vector contained a bacterial inaA DNA fragment in the 3′-noncoding region as a tag to distinguish the introduced DNA from the intrinsic OC gene. The OC vector and a selection marker gene conferring hygromycin resistance were used together to transfect into rice protoplasts. A number of hygromycin-resistant calli were obtained and studied by polymerase chain reaction and genomic Southern blotting to find if the exogenous OC gene had been integrated. The calli were studied by northern blotting as well to examine mRNA expression. The results showed that integration and expression of the introduced OC gene occurred in 51% and 27%, respectively, of 156 subcultures from 15 hygromycin-resistant calli. As a final step, transgenic rice plants were regenerated from the calli expressing OC. Leaves and seeds from the plants had higher OC activities than those from nontransgenic plants.  相似文献
