Efficient and rapid tissue culture systems were developed for Piper longum, an important medicinal plant, through shoot tip multiplication and direct regeneration. Multiple shoots were induced from shoot tips cultured on agar-based Murashige and Skoog (MS) medium containing 4.44–22.19 M benzyladenine (BA) and 4.64–13.9 M kinetin (K). Maximum number of shoots were induced with 8.9 M BA and 4.64 M K. Adventitious shoot regeneration from leaf segments was achieved on MS containing 3.6–22.19 M BA along with 3.31–12.4 M picloram (P). Shoot differentiation occurred directly from the leaf bases without intermediale callus formation. Maximum shoot buds were obtained on MS medium with 17.76 M BA and 8.28 M P. Elongated shoots were separated and rooted in MS supplemented with 2.46 M indole butyric acid (IBA). Plantlets, thus developed were established in soil. 相似文献

MMS2 (half strength of both major salts and minor salts and full strength of vitamins) medium containing 1.0 μM IBA in the dark
for one initial week at 30°C, followed by subculturing them in a plant-growth regulator (PGR)-free medium in the light. The

Different vegetative parts of Fagraea fragrans Roxb., a valuable timber tree of South East Asia, were used as explants in in vitro studies.Nodal segments showed the best growth response as numerous adventitious shoots were regenerated in MS medium supplemented with Benzyl adenine (BA, 8.8 um) and 2,4-dichlorophenoxyacetic acid (2,4-D, 0.5 um). Shoot buds also developed from the leaf and root segments that were subcultured. The detailed process of callus growth and differentiation leading to the formation of whole plantlets is described. An efficient in vitro multiplication system via multiple shoot bud induction and regeneration has been developed in Chlorophytum arundinaceum using shoot crown explants. Optimum regeneration frequency (87%) and desirable organogenetic response in the form of de novo organized multiple shoot buds without an intervening callus phase was obtained on Murashige and Skoog's (MS) minimal organics medium containing 3% sucrose (w/v) supplemented with 4×10−6 M Kn and 2×10−6 MIBA. Axenic secondary explants with multiple shoot buds on subculturing elicited best response with 1×10−5 M Kinetin (Kn) and 5×10−6 M indole-3-butyric acid (IBA) giving rise to an average of 18.74 shoots per culture with mean shoot length of 7.6 cm ± 1.73. Varying molar ratios of either Kn/IBA or Kn/NAA revealed statistically significant differences in the regeneration frequencies among the phytohormone treatments. It was observed that the shoot bud differentiation and regeneration was influenced by the molar ratios of cytokinins/auxin rather than their relative concentrations. Healthy regenerated shoots were rooted in half strength MS basal medium containing 3% sucrose (w/v) supplemented with 5×10−6 M IBA. Following simple hardening procedures, rooted plantlets, were transferred to soil-sand (1:1; v/v) with more than 90% success. Genetic fidelity was assessed using random amplified polymorphic DNA (RAPD), karyotype analysis and meiotic behaviour of in vitro and in vivo plants. Five arbitrary decamers displayed same banding profile within all the micropropagated plants and in vivo explant donor. The cytological and molecular analysis complemented and compared well and showed no genomic alterations in the plants regenerated through shoot bud differentiation. High multiplication frequency, molecular, cytological and phenotypic stability ensures the efficacy of the protocol developed for the production and conservation of this important endangered medicinal herb. 相似文献

Micropropagation offers a great potential to produce millions of clonal individuals through tissue culture via induction of morphogenesis. The aim of this work was to obtain an efficient protocol for callus regeneration for Gentiana kurroo Royle. The morphogenic response of different explants (leaves, petioles, roots) varied and responded differently for regeneration according to combinations of growth regulators. The petiole explants were best responding for callus induction and subsequently for indirect and direct regeneration. The callus induction was achieved on MS basal + 1.0 mg/l benzyladenine (BA) and 3.00 mg/l naphthalene acetic acid (NAA). MS medium supplemented with 0.10 mg/l NAA and 1.0 mg/l thidiazuron (TDZ) was recorded as the best medium for indirect regeneration. However, for direct regeneration the maximum number of shoot emergence was observed on MS basal fortified with 0.10 mg/l NAA + 0.75 mg/l TDZ. Half strength MS basal supplemented with indole-3-butyric acid (IBA) 1.00 mg/l gave best response for root induction. Subsequently, the plantlets were transferred and 100 % survival rate was recorded only on autoclaved cocopeat. No morphological variations were recorded in the callus regenerated plantlets. 相似文献

plant in rough environment. In this study, we developed 12 microsatellite loci from P. harmala by the combining biotin capture method for the first time. A total of 31 microsatellite sequences were recovered through
screening the library and 12 of them are polymorphic. The number of alleles per locus in 36 sampled individuals ranged from
3 to 8, expected heterozygosity and observed heterozygosity ranged from 0.1381 to 0.6821 and from 0.3573 to 0.8739, respectively.
In addition, all markers have been crossly checked in the other congeneric species. These microsatellite markers would provide

An in vitro regeneration protocol has been developed from half-seed explants of a mild (cv. New Mexico-6) and a pungent (cv. Rajpur Hirapur) chile pepper (Capsicum annuum L). Imbibed seeds were cut into two parts such that one portion contained the cotyledons and a part of the hypocotyl (part A) while the other part had the proximal part of the hypocotyl and the radicle (part B). These explants were cultured on MS medium with or without cytokinins (KIN, BA, ZEA, 2iP). Cytokinins dramatically increased both the percentage of explants forming buds as well as the number of buds per explant, and also hastened the rate of bud production. The relative efficacy of cytokinins in inducing the formation of leafy buds varied in the two cultivars. However, the best response was observed with ZEA in both cultivars. The highest percentage of bud formation was recorded after presoaking part B explants for 72 hours. The elongation growth of leafy buds was severely inhibited in the continuous presence of high concentrations of cytokinins, and frequently the buds became quite thick, ill-defined and vitreous. Within 3–5 weeks of transfer to Magenta boxes containing vermiculite and soil (1:3), 70–85% of the rooted hypocotyls developed 1–2 elongated shoots. Cannabis sativa L. (Cannabaceae) is an important medicinal plant well known for its pharmacologic and therapeutic potency. Because of allogamous nature of this species, it is difficult to maintain its potency and efficacy if grown from the seeds. Therefore, chemical profile-based screening, selection of high yielding elite clones and their propagation using biotechnological tools is the most suitable way to maintain their genetic lines. In this regard, we report a simple and efficient method for the in vitro propagation of a screened and selected high yielding drug type variety of Cannabis sativa, MX-1 using synthetic seed technology. Axillary buds of Cannabis sativa isolated from aseptic multiple shoot cultures were successfully encapsulated in calcium alginate beads. The best gel complexation was achieved using 5 % sodium alginate with 50 mM CaCl2.2H2O. Regrowth and conversion after encapsulation was evaluated both under in vitro and in vivo conditions on different planting substrates. The addition of antimicrobial substance — Plant Preservative Mixture (PPM) had a positive effect on overall plantlet development. Encapsulated explants exhibited the best regrowth and conversion frequency on Murashige and Skoog medium supplemented with thidiazuron (TDZ 0.5 μM) and PPM (0.075 %) under in vitro conditions. Under in vivo conditions, 100 % conversion of encapsulated explants was obtained on 1:1 potting mix- fertilome with coco natural growth medium, moistened with full strength MS medium without TDZ, supplemented with 3 % sucrose and 0.5 % PPM. Plantlets regenerated from the encapsulated explants were hardened off and successfully transferred to the soil. Experiment on jute crop was conducted during pre-kharif to kharif seasons (April to August) at Bidhan Chandra Krishi Viswavidyalaya (BCKV), West Bengal, India in a view to record the pest incidence on olitorius jute and to determine the weather parameters impacting on pest population in jute under West Bengal conditions. Seventeen different species of pests belonging to insects, mites and nematodes were found feeding on jute. Among them, jute semilooper (Anomis sabulifera Guen.), Bihar hairy caterpillar (Spilarctia obliqua Wlk.), grey weevil (Myllocerus discolor Bohemus), yellow mite (Polyphagotarsonemus latus Banks), stem weevil (Apion corchori Marshall), and root knot nematode, Meloidogyne incognita are causing economic damage to the crop and other pests were considered as minor pests. Correlation studies with weather parameters showed that incidence of semilooper was negatively correlated (r = ?0.795 to ?0.725) with the maximum temperature but had positive significant association with minimum temperature (r = 0.528–0.715), morning relative humidity (RH) (r = 0.579–0.857) and afternoon RH (r = 0.876). Bihar hairy caterpillar incidence also exhibited positive significant relationship with morning RH (r = 0.577) and afternoon RH (r = 0.545). Morning and afternoon RH and rainfall also had significant positive correlation with the incidence of M. discolor with r = 0.535, 0.570, and 0.700, respectively. None of the meteorological parameters had any significant influence on the incidence of A. corchori. Yellow mite incidence showed positive association with morning RH (r = 0.563–0.679) and afternoon RH (r = 0.526–0.618). Rainfall was found favourable for proliferation and incidence of M. discolor only but had a negative effect on the incidence of stem weevil and yellow mite. Thus climatic factors particularly temperature, relative humidity and rainfall played a pivotal role on occurrence and existence of different pests on jute crop. 相似文献

Taking into account the concept of the "Trojan Horse", where contaminants may have its entry into specific organs potentiated by its association with nanomaterials, the aim of this study was to analyze the joint toxic effects induced by an organic nanomaterial, fullerene (C(60)) with the metalloid arsenic (As(III)). Hepatocytes of zebrafish Danio rerio were exposed to As(III) (2.5 or 100 μM), C(60) or As+C(60) for 4h, not altering cells viability. Intracellular reactive oxygen species concentration was reduced in cells exposed only to the C(60) (1mg/L) and in the treatment of 100 μM As(III)+C(60). Co-exposure with C(60) abolished the peak of the antioxidant glutathione (GSH) registered in cells exposed to the lowest As(III) concentration (2.5 μM). A similar result was observed in terms of lipid damage (TBARS). Total antioxidant capacity was significantly higher at both As(III) concentrations co-exposed to C(60) when compared with the control group. Activity of glutathione-S-transferase omega, a limiting enzyme in the methylation pathway of As(III), was reduced in the 100 μM As(III)+C(60) treatment. Cells co-exposed to C(60) had a significantly higher accumulation of As(III), showing a "Trojan Horse" effect which did not result in higher cell toxicity. Instead, co-exposure of As(III) with C(60) showed to reduce cellular injury. 相似文献

Summary Adventitious buds were induced from immature embryos of Picea chihuahuana, a species from the north of Mexico. The medium was Schenk and Hildebrandt supplemented with benzyladenine and kinetin, alone
or in combination with naphthaleneacetic acid and 2,4-dichlorophenoxyacetic acid. Adventitious buds were obtained through
a wide range of growth regulator concentrations, mainly from the cotyledonary region. Kinetin was more effective than benzyladenine
in shoot induction. The optimum shoot induction medium contained 23.0 μM kinetin, on which 11.2 adventitious buds per embryo were obtained. Shoot development and elongation were achieved on basal
SH medium at 50% concentration (SH 50) without growth regulators and improved with the reduction of sucrose concentration
to 10 g l−1. Differential response appeared to be associated with the provenance of the seeds, relative to the percentage of explants
that responded as well as the number of adventitious buds produced per embryo. A low percentage of shoot rooting was obtained
with a 24-h pulse on a medium with 16.1 or 26.85 μM of naphthaleneacetic acid. In the process of micropropagation of Picea chihuahuana, rooting is still the limiting factor. 相似文献

A rapid and efficient in vitro plant regeneration method was developed for Matteuccia struthiopteris (L.) Todaro (Ostrich fern). Side shoots, originating in meristems of sectioned rhizomes, were used as explant material. A
very high rate of meristem multiplication was achieved by culturing the explants in half-strength MS liquid medium supplemented
with 2.0 mg/l N-(4-Pyridyl)-N′-phenylurea (4-PU) and 0.5 mg/l thidiazuron (TDZ). Multiplication of the shoot primordia was
faster in suspension culture than on solid medium. Rhizogenesis and growth of regenerants were best achieved on hormone-free
one-quarter-strength MS solid medium amended with 0.4% agar and 1.0% activated charcoal. Regenerated plantlets continued to
grow after transfer to soil in a phytotron.

Ectomycorrhizal fungi are mutualistic symbionts of boreal forest trees and may mediate mineral weathering through their direct access to photosyntentically derived carbon. In soil, fungal mycelia (i) provide a large surface for nutrient uptake; (ii) induce intense colonization of nutrient rich substrates; (iii) cause local acidification and (iv) produce organic acids. Mechanisms of ectomyorrhizal fungi induced weathering in response to nutrient limiting growth conditions remains largely unresolved. proliferation and regeneration studies. A significant amount of sporophyte regeneration was observed on solid medium containing
10 mgl−1 6-(δ, δ-dimethylallylamino) purine (2iP). Sporophyte regeneration from callus followed an atypical pattern of development.
Leafy structures of single-cell thickness with a microrhizome were formed as sporophyte initials. Prolonged cultures of these
structures resulted in the formation of juvenile sporophytes in vitro. The use of liquid media resulted in increased biomass in culture. The present study is the first report of a successful
system for callus production and regeneration of sporophytes from leafy structures in ferns. The method can be successfully

Chili (Capsicum chinense) cv. ‘Bhut jalakia’ is used in India for extraction of oleoresin and capsaicin as it is characterized by a very high capsaicin content. The conventional method of propagation of ‘Bhut jalakia’ is through seeds, but this is beset by short viability and low germination rates. Developing a suitable regeneration protocol for ‘Bhut jalakia’ was the focus of this study; as to date, in vitro regeneration for this cultivar has not been investigated. Cotyledon and shoot tip explants were cultured on Murashige and Skoog (MS) media supplemented with different concentrations of cytokinins and auxins. In the case of cotyledon explants, MS medium supplemented with 6-benzylaminopurine (BAP) at 35 μM and kinetin (KIN) at 15 μM were found to be optimal (4.00?±?0.57) for induction of multiple shoots per explant, whereas BAP at 14.8 μM and KIN at 60 μM were best (5.00?±?0.57) for growth of shoot tip explants. Shoots developed from cotyledon explants produced the maximum (8.67?±?0.32) number of roots on MS medium supplemented with low concentration (2.6 μM) of 2-naphthaleneacetic acid (NAA). Supplementation of indole-3-butyric acid (IBA) at 5 μM was found optimal for root formation (16.67?±?2.60) for shoots derived from of shoot tip explants. One month after transfer of in vitro regenerated plantlets to various potting mixes, the highest survival rate (40%) was observed in a mixture of sand, soil, and cow dung in a ratio of 1:1:1. Thus, both shoot tip and cotyledon explants may be cultured on MS medium modified with BAP, IBA, NAA, and KIN to regenerate ‘Bhut jalakia’ chili plants within 90 d. 相似文献
