Mangoes (var. Tommy Atkins) were exposed to ethylene and acetylene over a range of concentrations at high humidity for 24 h at 25°C, then ripened in air alone. Ripeness was assessed after 4 and 8 days by analysis of texture, colour development, soluble solids and acid contents. Ethylene in air at concentrations of 0.01 ml litre-1 and above or acetylene at 1.0 ml litre-1 were found to initiate ripening. Treatment with 0.01 ml litre-1 acetylene resulted in limited softening but had no effect on the other ripening changes analysed. Individual ripening processes responded differently to treatment: texture changes were most rapidly affected, while the rate of acidity losses was often reduced in ethylene treated fruits. Acetylene-treated fruits at concentrations of 0.01 and 0.1 ml litre-1 showed delayed ripening when compared to those treated with either 1.0 ml litre-1 acetylene or ethylene. Increased acetylene concentrations of 2.0 ml litre-1 gave a similar response to 1.0 ml litre-1, although in some instances there were indications of inhibitory effects.  相似文献

The major fluxes of carbohydrate metabolism were estimated during starch breakdown by ripening bananas (Musa cavendishii Lamb ex Paxton). Hands of bananas, untreated with ethylene, were allowed to ripen in the dark at 21° C. Production of CO2 and the contents of starch, sucrose, glucose and fructose of intact fruit were determined for a period of 10 d that included the climacteric. The detailed distribution of label was determined after supplying the following to cores of pulp from climacteric fruit: [U-14C]-, [1-14C]-, [3,4-14C]-and [6-14C]glucose, [U-14C]glycerol, 14CO2. The data obtained were used to estimate the following fluxes, values given as mol hexose · (g FW)–1 · h–1 in parenthesis: starch to hexose monophosphates (5.9) and vice versa (0.4); hexose monophosphates to sucrose (7.7); sucrose to hexose (4.7); hexose to hexose monophosphate (3.8); glycolysis (0.5–1.6); triose phosphate to hexose monophosphates (0.14); oxidative pentose-phosphate pathway (0.48); CO2 fixation in the dark (0.005). The effects of high temperatures on the clam, Chamelea gallina, generally recognised as a low tolerant bivalve species, were studied by evaluating some functional responses of the haemocytes. The animals were kept for 7days at 20, 25 and 30 degrees C and total haemocyte count (THC), phagocytosis, lysozyme activity (in both haemocyte lysate and cell-free haemolymph), activity and expression of the antioxidant enzyme superoxide dismutase (SOD) (in both haemocyte lysate and cell-free haemolymph) were chosen as biomarkers of exposure to high temperatures. The survival-in-air test was also performed. During the experiment, the clams showed differing burrowing behaviour: the animals kept at 20 and 25 degrees C burrowed completely, whereas at 30 degrees C the clams progressively emerged from the sediment and then remained on the surface. The highest temperature significantly increased THC, whereas it decreased the phagocytic activity of haemocytes. The haemocyte size frequency distribution in clams kept at 30 degrees C showed that the cell population of about 8-10microm was markedly reduced compared to clams kept at 20 and 25 degrees C. In clams maintained at 25 degrees C, lysozyme activity was significantly increased in haemocyte lysate, whereas it was markedly decreased in cell-free haemolymph. Total SOD activity significantly decreased in haemocytes from clams held at 30 degrees C whereas it increased in cell-free haemolymph from clams held at 25 degrees C and 30 degrees C. A significant decrease in haemocyte Mn-SOD and Cu/Zn-SOD activities was found with increasing temperature. In cell-free haemolymph, the highest Mn-SOD activity was recorded at 30 degrees C, whereas the Cu/Zn-SOD activity showed no significant changes in clams maintained at different temperatures. SOD isoform expression exhibited different patterns in haemocyte lysate and cell-free haemolymph. The resistance to air exposure of clams kept at 30 degrees C was shown to decrease significantly, LT(50) values fell from 6days in clams kept at 20 degrees C and 25 degrees C to 4days in those kept at 30 degrees C.  相似文献

The increased prevalence of high temperatures(HTs) around the world is a major global concern, as they dramatically affect agronomic productivity. Upon HT exposure, plants sense the temperature change and initiate cellular and metabolic responses that enable them to adapt to their new environmental conditions.Decoding the mechanisms by which plants cope with HT will facilitate the development of molecular markers to enable the production of plants with improved thermotolerance. In recent decades, genetic, physiological, molecular, and biochemical studies have revealed a number of vital cellular components and processes involved in thermoresponsive growth and the acquisition of thermotolerance in plants. This review summarizes the major mechanisms involved in plant HT responses, with a special focus on recent discoveries related to plant thermosensing, heat stress signaling, and HT-regulated gene expression networks that promote plant adaptation to elevated environmental temperatures.  相似文献

The aim of this work was to determine the effects of hypoxia on the major fluxes of carbohydrate metabolism in climacteric fruit of banana (Musa cavendishii Lamb ex Paxton). Hands of bananas, untreated with ethylene, were allowed to ripen in air at 21°C in the dark. When the climacteric began, fruit were transferred to 15 or 10% oxygen and were analysed once the climacteric peak had been reached 8–12 h later. The rates of starch breakdown, sucrose, glucose and fructose accumulation, and CO2 production were determined, as were the contents of hexose monophosphates, adenylates and pyruvate. In addition, the detailed distribution of label was determined after supplying [U-14C]-, [1-14C]-, [3,4-14C]- and [6-14C]glucose, and [U-14C]glycerol to cores of tissue under hypoxia. The data were used to estimate the major fluxes of carbohydrate metabolism. There was a reduction in the rate of respiration. The ATP/ADP ratio was unaffected but there was a significant increase in the content of AMP. In 15% oxygen only minor changes in fluxes were observed. In 10% oxygen starch breakdown was reduced and starch synthesis was not detected. The rate of sucrose synthesis decreased, as did the rate of re-entry of hexose sugars into the hexose monophosphate pool. There was a large increase in both the glycolytic flux and in the flux from triose phosphates to hexose monophosphates. It is argued that the increase in these fluxes is due to activation of pyrophosphate: fructose-6-phosphate 1-phosphotransferase, and that this enzyme has an important role in hypoxia. The effect of exogenous glucose on the major fluxes of carbohydrate metabolism in cores of climacteric fruit of banana (Musa cavendishii Lamb ex Paxton) was determined with the intention of using the effects in the application of top-down metabolic control analysis. Hands of bananas, untreated with ethylene, were allowed to ripen in the dark at 21 °C. Cores were removed from climacteric fruit and incubated in 100 or 200 mM glucose for 4 or 6 h. The rates of starch breakdown, sucrose and fructose accumulation and CO2 production were measured. The steady-state contents of hexose monophosphates, adenylates and pyruvate were determined. In addition, the detailed distribution of label was determined after supply of the following: [U-14C]-, [1-14C]-, [3,414C]and [6-14C]glucose, and [U-14C]glycerol. The data were used to estimate the major fluxes of carbohydrate metabolism. Supply of exogenous glucose led to increases in the size of the hexose-monophosphate pools. There was a small stimulation of the rate of sugar synthesis and a major increase in the rate of starch synthesis. Starch breakdown was inhibited. Respiration responded to the demand for ATP by sugar synthesis. A young Cape Gannet Sula capensis responded to high ambient temperatures in the laboratory by standing, gular fluttering with raised head and open bill, wing drooping, defaecating at a fast rate close to or onto the feet, “paddling”; in the excreta, and increased breathing rate. Standing initially caused a decline in foot temperature which rose with rising ambient temperature. Gular flutter and wing drooping assisted the bird in tolerating heat stress. Reduction in heat‐load occurred when standing on a wet surface, presumably due to evaporative heat loss (of excreta on the feet = urohidrosis) and an enhanced conduction of heat to substrate. Field observations confirmed that young Cape Gannets defaecate onto their feet at high ambient temperatures. This species appears preadapted for this behaviour since it builds a solely guano nest, defaecating onto the rim which is compacted by “paddling”;.  相似文献

Oxygen consumption, heat production (HP) and core temperature (T(af)) were measured over 3 h in 20-34-day-old Muscovy duck and 12-21-day-old chicken embryos at ambient temperature (T(a)) of 37.5 degrees C and thereafter for 3 h at T(a) of 39.0 degrees C. At 37.5 degrees C T(a), HP increased with age in avian embryos of both species, following an exponential function. In muscovy duck embryos, a plateau phase occurred between D29 and D32; in chicken embryos, a similar plateau occurred between D19 and D21. T(af) rose in accordance with HP, and the relationships between T(af) and HP could be described by significant linear regressions in both species. Mostly, HP increased in embryos of both species during heat load, but by less than calculated by the van't Hoff rule; however, there was often also a decrease in HP under these conditions. Obviously, in avian embryos high T(a) causes a down-regulation of HP mediated by active thermoregulatory mechanisms. This is in agreement with data describing the influence of hyperthermia on HP in the postnatal period of birds and mammals. Because of this, the term 'second chemical thermoregulation' defined by Gelineo [C. R. Soc. Biol. (1936) 122 337] for birds and mammals should also be used for avian embryos.  相似文献

Either systemic or central administration of apomorphine produced dose-related decreases in rectal temperature at ambient temperatures (Ta) of 8 and 22 degrees C in rats. At Ta = 8 degrees C, the hypothermia was brought about by a decrease in metabolic rate (M). At Ta = 22 degrees C, the hypothermia was due to an increase in mean skin temperature, an increase in respiratory evaporative heat loss (Eres) and a decrease in M. This increased mean skin temperature was due to increased tail and foot skin temperatures. However, at Ta = 29 degrees C, apomorphine produced increased rectal temperatures due to increased M and decreased Eres. Moreover, the apomorphine-induced hypothermia or hyperthermia was antagonized by either haloperidol or 6-hydroxydopamine, but not by 5,6-dihydroxytryptamine. The data indicate that apomorphine acts on dopamine neurons within brain, with both pre- and post-synaptic sites of action, to influence body temperature.  相似文献

During ripening of bananas (Musa spp. [AAA group, Cavendish subgroup]), there is a massive conversion of starch to sucrose. Also during ripening there is a rise in respiration known as the respiratory climacteric. In this study changes in carbohydrate content, activities of starch and sucrose metabolizing enzymes, and respiration were measured to assess their potential interrelationships. Sucrose phosphate synthase activity increased dramatically during the first 4 days after initiation of ripening by ethylene treatment. Starch concentration decreased and sucrose concentration increased during this time period. Developmental changes in sucrose phosphate synthase activity were measured with limiting substrate (plus Pi) and saturating substrate concentrations. Activities were not parallel under the two assay conditions, providing tentative evidence that kinetically different forms of the enzyme may exist at different stages of ripening. Sucrose accumulation rate was most highly correlated with sucrose phosphate synthase activity assayed with limiting substrate concentrations (plus Pi). The cumulative amount of CO2 respired during ripening was positively correlated with sugar accumulation (R2 = 0.97). From this linear regression it was calculated that a constant 0.605 millimoles of CO2 was evolved per mole of sucrose formed throughout ripening. Using this quantity, the percentage of the total respiratory ATP produced which was required for the conversion of starch to sucrose was calculated assuming different models for carbon export from the amyloplast. The results suggest that sucrose biosynthesis during ripening constitutes a significant sink for respiratory ATP.  相似文献

Bananas were exposed to acetylene or ethylene at 0·01, 0·1 and 1 ml/litre, under high humidity, for 24 h at 18 °C. They were then transferred to an atmosphere of air alone for a further 4 days and during this period the respiration rate of three fruit from each treatment was measured. Ripeness was then assessed by colour score and soluble solids content. All levels of ethylene initiated ripening. Treatment with ethylene induced a climacteric rise in respiration, an increase in the soluble solids content of the pulp and degreening of the peel. All levels of acetylene, except 0·01 ml/litre, induced a climacteric rise in respiration. Fruit treated with acetylene at 1 ml/litre had a similar colour score and soluble solids content to those ripened by exposure to ethylene. Fruits treated with acetylene at 0·1 ml/litre had a lower soluble solids content and their peel remained green. Treatment with acetylene at 0·01 ml/litre failed to initiate ripening. Sensory evaluation of fruit ripened by acetylene at 1 ml/litre indicated that the acetylene treated fruit ripened slightly more slowly. When compared at the same stage of ripeness fruits from the two treatments were equally palatable.  相似文献

Bananas (Musa AAA Group, Cavendish Subgroup) were ripened over a range of temperatures from 15 to 35°C. The rate of ripening in both pulp and peel was accelerated with an increase in storage temperature up to about 24°C. Above this temperature the pulp softened and sweetened without the development of a fully yellow peel due to a decrease in the rate of chlorophyll breakdown. Peel carotenoid content was higher at 35°C than at 20°C.  相似文献

Changes in the plastid ultrastructure as revealed by thin-section electron-microscopy, chlorophyll a/b ratio, and the polypeptides of the thylakoid chlorophyll-protein complexes have been examined during the degreening of bananas (Musa AAA Group, Cavendish Subgroup) and plantains (Musa AAB Group, Plantain Subgroup) ripened at 20°C and 35°C. In bananas, where degreening is inhibited at temperatures above 24°C, ripening at the higher temperature results in a retention of thylakoid membranes, a relatively delayed breakdown in chlorophyll b, and a reduced dismantling of pigment-protein complexes. By contrast, in plantains, where degreening is complete within 4 days at both 20°C and 35°C, thylakoid membranes and their associated pigment-protein complexes are lost, and there is a rapid increase in chlorophyll a/b ratios at both ripening temperatures. It is suggested that the retention of thylakoid membranes is an important factor in the failure of Cavendish bananas to degreen when ripened at tropical temperatures, and that the degreening problem may be related to the comparatively high chlorophyll b content of the preclimacteric fruit.  相似文献

Six resting men were exposed to three temperatures (15.5, 21, 26.5 degrees C) for 120 min at three altitudes (sea level, 2,500 m, 5,000 m). A 60-min sea-level control at the scheduled temperature preceded the nine altitude episodes. Comparison of the base-line results at any one temperature showed no differences between rectal temperatures (Tre) or mean weighted skin temperatures (Tsk). After 120 min, Tre and Tsk not only depended on ambient temperature but also altitude. The initial rate of fall in Tre increased with altitude and equilibrium occurred earlier. At 15.5 degrees C, Tre was 0.3 degrees C lower at 5,000 m and 0.2 degrees C lower at 2,500 m than at sea level. Tsk was almost 2 degrees C higher at 15.5 degrees C at 5,000 m and 1 degrees C higher at 2,500 m than at sea level. Similar, smaller differences were observed at 21 degrees C. Mean weighted body temperature showed no change with altitude, but, since the gradient between core and shell was reduced, a shift of blood toward the periphery is implied.  相似文献

Under high temperature conditions, insects can tolerate to survive through various physiological mechanisms, which have been well documented in laboratory studies. However, it is still unclear as to whether these laboratory data can scale up to those in the field. Here we studied dynamics of heat-induced metabolites in Corythucha ciliata adults under both laboratory and field conditions to examine their significance in thermal tolerance of the species. We compared the effects of controlled thermal treatments (2 h at 33–43 °C at 2 °C intervals in the laboratory) and naturally increasing thermal conditions (10:00–14:00 at 2-h intervals (33.5–37.2 °C) on a hot summer day in a field in Shanghai, China) on water content and levels of water-soluble protein, triglycerides, mannitol, and sorbitol in the adult bodies. The results showed that water content significantly decreased and all other metabolic parameters significantly increased in response to temperature stresses with similar patterns in both the laboratory and field, although the respective threshold temperatures were different under the two conditions. The close linkage observed in the two conditions suggests that a short period of heat stress induces water loss and accumulation of thermal metabolites in C. ciliata adults. This heat-resistance provides a defense mechanism counteracting thermal damage in C. ciliata.  相似文献
