A unique feature, frequent heterocyst germination, has been observed in a nonsporulating mutant clone (of spontaneous origin) of the blue-green alga Gloeotrichia ghosei Singh. The controlling factor seems to be the presence of ammoniacal nitrogen in the medium. In addition, such a medium supports differentiation of successive crops of new heterocysts and their germination in the name medium and in the same algal culture. Contrary to previous observations with oilier blue-green algae, ammoniacal nitrogen does not seem to inhibit heterocyst differentiation in this alga. Both the parent alga and its mutant clone grow poorly in a nitrogen-free medium, which, although they are not completely free from bacteria, may indicate that they tire poor fixers or nonfixers. However, they form a large number of heterocysts under these conditions. The general conclusion is that the heterocysts of blue-green algae show a multiplicity of structure and function. In the present case they have reproductive function leading to direct propagation of the alga. The bearing of these findings on the interrelationships of the genera Gloeotrichia and Rivularia has been discussed. It has been concluded that the distinction between them is purely artificial.  相似文献

We conducted an outdoor mesocosm experiment of factorial design consisting of three levels of nutrient supply (no nutrient addition and additions of nitrogen and phosphorus in ratios of 10:1 and 45:1) cross-classified with two levels of bluegill (Lepomis macrochirus) (presence and absence). Nutrient supply significantly affected total phosphorus (TP), total nitrogen (TN), TN: TP ratio, turbidity, Secchi depth, phytoplankton chlorophyll, filamentous blue-green algae, periphyton chlorophyll, Asplanchna and non-predatory rotifers. The presence of bluegill significantly increased TP, turbidity, diatoms, unicellular green algae, colonial blue-green algae, filamentous blue-green algae, periphyton chlorophyll, Asplanchna and non-predatory rotifers, and decreased Secchi depth, cladocerans, cyclopoid copepodids, copepod nauplii and chironomid tube densities. Nutrient supply and fish effects were not independent of each other as shown by significant nutrient × fish interaction effects for TP, Secchi depth, filamentous blue-green algae, periphyton chlorophyll, Asplanchna and non-predatory rotifers.  相似文献

The Waikato River (latitude 38°S, longitude 176°E, North Island, New Zealand) is overwhelming y dominated by diatoms (mainly Melosira species) while blue-green and green algae are of minor importance. Both laboratory and in situ nutrient enrichment experiments showed enhanced growth of natural and index blue-green and green algae by addition of phosphate and nitrate. These algae were also shown to require higher temperature and light intensity than the diatoms. On the other hand, Waikato River with its higher silica content, moderate range of temperature and running water habitat was more favourable an environment for diatoms.  相似文献

β-Glucans (BGs) are polysaccharides that are found in the cell walls of organisms such as bacteria, fungi, and some cereals. The objective of the present study was to investigate the genotoxic and antigenotoxic effects of BG extracted from the mushroom Agaricus brasiliensis (= Agaricus blazei Murrill ss. Heinemann). The mutagenic activity of BG was tested in single-cell gel electrophoresis assays with human peripheral lymphocytes. In addition, the protective effects against the cooked food mutagen 3-amino-1-methyl-5H-pyrido[4,3-b]indole (Trp-P-2) and (+/−)-anti-B[a]P-7,8-dihydrodiol-9,10-epoxide (BPDE), which is the main metabolite of B[a]P, and against ROS (H2O2)-induced DNA damage, were studied. The results showed that the compound itself was devoid of mutagenic activity, and that a significant dose-dependent protective effect against damage induced by hydrogen peroxide and Trp-P-2 occurred in the dose range 20–80 μg/ml. To investigate the prevention of Trp-P-2-induced DNA damage, a binding assay was carried out to determine whether BG inactivates the amine via direct binding. Since no such interactions were observed, it is likely that BG interacts with enzymes involved in the metabolism of the amine.  相似文献

Summary p-Nitrophenol (PNP),m-nitrophenol (MNP), 2,4-dinitrophenol (DNP) and catechol were tested for their effects on algal population in a soil and on pure cultures of two algae isolated from soil. Both PNP and MNP, even at 0.5 kg ha−1 level were toxic to the soil algae; high doses effected increase in toxicity. Inhibition of algae was relatively more with PNP compared to the other two nitrophenols. Catechol treatment up to 1.0 kg ha−1 led to a significant initial enhancement of algae with a subsequent far less toxic effect. The toxicity of the phenolic compounds towardChlorella vulgaris, a green alga andNostoc linckia, a blue-green alga, decreased in the order: MNP≧PNP>DNP>Catechol. However, algicidal or algistatic effect of the test chemicals was fairly more againstC. vulgaris, suggesting that the eukaryotic alga is highly sensitive to such soil pollutants compared to the prokaryotic alga.  相似文献

Summary A blue-green algae, Anabaena N-7363, was immobilized in 2% agar gel. The hydrogen productivity of the immobilized algae was three times higher than that of free algae. The maximum hydrogen production rate by the immobilized blue-green algae was 0.52 moles h–1 g–1 (of wet gel) in the medium without nitrogen sources under illumination (10,000 lux). The oxygen evolved was then removed by a reactor containing aerobic bacteria. A photo-current of 15–20 mA was continuously produced for 7 days by the photochemical fuel cell system consisting of the immobilized Anabaena reactor, the oxygen-removing reactor and the hydrogen-oxygen fuel cell. The conversion ratio of hydrogen to current was from 80% to 100%.  相似文献

The effect of the auxins dicamba (3,6-dichloro-2-methoxybenzoic acid) and picloram (4-amino-3,5,6-trichloropicolinic acid) on callus growth and embryogenesis in Phoenix dactylifera L. was investigated. Maximum callus fresh weight was obtained in nutrient medium enriched with 200 µm picloram. Somatic embryogenesis and subsequent plant regeneration was achieved following transfer of such calli to hormone-free medium. Germination of the somatic embryos was influenced by treatment with the chemical mutagen ethylmethanesulphonate (EMS). Uptake of the labelled mutagen ([14C]EMS) by the somatic embryos increased with increased incubation time. Presence of dimethyl sulphoxide (DMSO) as a carrier agent during mutagenic treatment was necessary for efficient mutagen uptake.  相似文献

Summary Regulation of DAHP synthetase activity was investigated in autotrophically grown blue-green and green algae. Members of the class of blue-green algae possess an enzyme, the activity of which is regulated by l-tyrosine and l-phenylalanine, whereby l-tyrosine is effective in 100 fold lower concentrations. DAHP synthetases of two organisms, Anabaena and Anacystis, were shown to belong to the V-type of allosteric enzymes.In contrast to the DAHP synthetase of blue-green algae regulation of this enzyme could not be demonstrated in two green algae, Ankistrodesmus and Maesotaenium. However, Euglena gracilis, both under conditions of mixotrophic and autotrophic growth, exhibits very effective regulation of this key enzyme; again, the inhibitors are tyrosine and phenylalanine. DAHP synthetase activity of Euglena has been purified about 40 fold; during this enrichment no separation of the enzyme activity inhibited by tyrosine and that by phenylalanine could be observed.  相似文献

The Comparative Boron Nutrition of Several Green and Blue-Green Algae   总被引:1,自引:0,他引:1  
The boron requirements of filamentous and non-filamentous species of green and blue-green algae have been investigated. A primary feature of this study was the quantitative analysis of the cells for boron. This permitted expression of the degree of purification of the environment and the boron requirements of the organisms in terms of cell content. Three species of green algae were shown neither to require boron nor to absorb it in appreciable amounts. In contrast, three species of blue-green algae readily absorbed boron and nitrogen fixing species showed a marked growth response to boron when nitrate was omitted from the culture medium. The response to boron was less with nitrate in the medium, probably due to increased contamination from the nitrate salt rather than an involvement of boron in the fixation process. The boron content of the deficient blue-green algae was 1–2 μg/g, suggesting a requirement below dicot angiosperms and comparable to monocots. In agreement with other reports, the fungus Penicillium chrysogenum neither required nor absorbed boron.  相似文献

Summary Nitrogen fixation (C2H2 reduction) by blue-green algae occurring on the juvenile lava field of Heimaey, Iceland was examined both in the laboratory (potential at 20° C and 39° C) and in the field, three and a half years after the volcanic eruption.Already at this early stage of colonization representatives of unicellular and filamentous heterocystous and non-heterocystous blue-green algae were commonly observed. The predominating algae were Nostoc sp. (20° C) and Schizothrix sp. — Microcoleus chthonoplastes, (39° C), the former often in association with the protonemata-rhizoids of moss plants.The potential for nitrogen fixation was recorded at an average rate of 109.2 (20° C) and 138.1 (39° C) ng N g-1 h-1 in soil collected from localities randomly distributed over the lava field.Tests for nitrogen fixation performed in situ revealed significant fixation activities in all the eleven localities subject to examination. The activities ranged from 2.8 to 63.4 (mean 21.5) ng N g-1 h-1 and 1.9 to 17.7 (mean 7.9) ng N cm-2 h-1.All the nitrogen fixation data noted imply that blue-green algae contribute a substantial part of the nitrogen input to the lava. Further, it was found that material incubated under micro-aerophilic conditions exhibited considerably enhanced nitrogenase activity.The role of nitrogen-fixing blue-green algae in general and Nostoc muscorum in particular in being suitable as pioneering organisms preparing the bare lava for ingress of other plants is also discussed.  相似文献

Summary a)Nitrogen fixation in rice fields. Nitrogen-fixing blue-green algae grow abundantly in tropical regions and are particularly common in paddy fields. Their possible role in the nitrogen accumulation of soil has been studied. The most vigorous nitrogen-fixing blue-green algae have been assessed for use as green manure in rice fields and favorable effects have been reported in India and other countries. b)Nitrogen fixation by algae in water. The planktonic blue-green algae occur abundantly at certain time of the year in sea water and lake water, and some of them are known to be nitrogen fixers. Certain Japanese species of blue-green algae can withstand high temperatures including ten nitrogen-fixing species from hot-spring waters. c)Nitrogen fixation by symbiotic blue-green algae. Certain species of blue-green algae form associations with other organisms such as fungi, liverworts, ferns and seed plants. The relationship between these two organisms is on one occasion commensal and on others symbiotic. A new and inexpensive inorganic medium (‘DS medium’) for the mass cultivation of freshwater blue-green algae (Cyanobacteria) and green algae has been developed. It consists basically of distilled or demineralized water (90%) and seawater (10%) and requires only little addition of pure purchasable chemicals (phosphate, trace elements, if necessary nitrate). No addition of macronutrients (NaCl, MgCl2 or MgSO4, KCl or K2SO4, CaCl2) and of boron is required because they are sufficiently provided by the seawater. Decalcified water may also be suitable instead of demineralized water. For the cultivation of green algae, a higher trace element concentration is recommended than for blue-green algae. Because of its low total salt concentration the DS medium is freshwater-like. It is easy to prepare and effects rapid algal growth. It may be of special value for algal mass culture in regions which are close to the sea.  相似文献

The coloration of cells of the cyanobacterium Synechococcus sp. PCC 7002 changed from normal blue-green to yellow-green when cells were grown at 15° C in a medium containing nitrate as the sole nitrogen source. This change of coloration was similar to a general response to nutrient deprivation (chlorosis). For the chlorotic cells at 15° C, the total amounts of phycobiliproteins and chlorophyll a decreased, high levels of glycogen accumulated, and growth was arithmetic rather than exponential. These changes in composition and growth occurred in cells grown at low (50 μE m–2 s–1) as well as high (250 μE m–2 s–1) light intensity. After a temperature shift-up to 38° C, chlorotic cells rapidly regained their normal blue-green coloration and normal exponential growth rate within 7 h. When cells were grown at 15° C in a medium containing urea as the reduced nitrogen source, cells grew exponentially and the symptoms of chlorosis were not observed. The decrease in photosynthetic oxygen evolution activity at low temperature was much smaller than the decrease in growth rate for cells grown on nitrate as the nitrogen source. These studies demonstrate that low-temperature-induced chlorosis of Synechococcus sp. PCC 7002 is caused by nitrogen limitation and is not the result of limited photosynthetic activity or photodamage to the photosynthetic apparatus, and that nitrogen assimilation is an important aspect of the low-temperature physiology of cyanobacteria. Electron microscopic studies of Sphagnum lindbergii (Schimp.) and S. riparium (Ångstr.) have revealed the presence of intracellular organisms such as blue-green algae, green algae, bacteria and fungi. Nitrogenase activities of these Sphagnum mosses were found to be related mainly to the presence of intracellular Nostoc filaments. The appearance of nitrogen-fixing blue-green algae within bryophytes is thus not restricted to liverworts. The association is likely to be of ecological importance as it seems to occur in very acid habitats generally lacking blue-green algae. Possible interrelations between the moss, the blue-green algae and different types of bacteria are discussed.  相似文献

The hypotheses that larval fish density may potentially affect phytoplankton abundance through regulating zooplankton community structure, and that fish effect may also depend on nutrient levels were tested experimentally in ponds with three densities of larval walleye, Stizostedion vitreum (0, 25, and 50 fish m–3), and two fertilizer types (inorganic vs organic fertilizer). A significant negative relationship between larval fish density and large zooplankton abundance was observed despite fertilizer types. Larval walleye significantly reduced the abundances of Daphnia, Bosmina, and Diaptomus but enhanced the abundance of various rotifer species (Brachionus, Polyarthra, and Keratella). When fish predation was excluded, Daphnia became dominant, but Daphnia grazing did not significantly suppress blue-green algae. Clearly, larval fish can be an important regulator for zooplankton community. Algal composition and abundance were affected more by fertilizer type than by fish density. Inorganic fertilizer with a high N:P ratio (20:1) enhanced blue-green algal blooms, while organic fertilizer with a lower N:P ratio (10:1) suppressed the abundance of blue-green algae. This result may be attributed to the high density of blue-green algae at the beginning of the experiment and the fertilizer type. Our data suggest that continuous release of nutrients from suspended organic fertilizer at a low rate may discourage the development of blue-green algae. Nutrient inputs at a low N:P ratio do not necessarily result in the dominance of blue-green algae.  相似文献

A new method was developed to determine the mutagenic efficacy of a suspected mutagen by employing green fluorescent protein (GFP) as a direct biosensor for mutation detection. Alterations in target gene (AcGFP1) expression after mutagen [(±)-7p,8a-dihydroxy-9a,10a-epoxy-7,8,9,10-tetrahydrobenzo[a]pyrene (BPDE)] treatment were measured to detect the mutagenic efficacy of the carcinogen. In contrast to mutagen treatment of the entire plasmid or cell culture, the target AcGFP1gene devoid of the plasmid backbone was exposed to BPDE (10–500 μM) to eliminate the need for an additional fusion gene. Shuttle vectors (pAcGFP-N1) were religated to the AcGFP1 gene with BPDE adducts (0–8.59 μM) and replicated in the eukaryotic host. This approach eliminated false-negative errors in target gene expression that arose from BPDE adduct formation in the residual plasmid backbone rather than in the AcGFP1 gene. Determination of the BPDE–AcGFP1 adducts allowed the quantitative mutagenic effect of the BPDE adducts on AcGFP1 gene expression to be monitored. The results obtained with flow cytometry and confocal microscopy validate our method and demonstrate efficient and direct use of GFP as a biosensor for mutation detection.  相似文献

Marine blue-green algae, Lyngbya Lagerheimii, Microcoleus chthonoplastes, Plectonema terebrans, Agmenellum quadruplicatum, and freshwater blue-green algae, Anacystis nidulans, Anabaena variabilis, Nostoc muscorura, and Oscillatoria sp. treated with lysozyme (muramidase) formed spheroplasts but not protoplasts. The time needed for spheroplast induction varied with the species. Approximate internal osmotic pressures of the blue-green algae were determined. The mutagenicity of the commonly used glutathione S-transferase substrates 1-chloro-2,4-dinitrobenzene (CDNB) and 1,2-dichloro-4-nitrobenzene (DCNB) was investigated in the Salmonella mutagenicity assay. CDNB induced a concentration-dependent mutagenic response in Salmonella typhimurium strain TA98. Incorporation of an activation system derived from Aroclor 1254-induced rats did not influence mutagenic response. Under the same conditions DCNB failed to display mutagenic activity. The mutagenic activity of CDNB was attenuated in bacterial strains under-expressing nitroreductase or O-acetylase activity but, in contrast, it was exaggerated in an O-acetylase over-expressing strain. It is inferred that CDNB exhibits a mutagenic response following reduction of the nitro-group to the hydroxylamine, which is further acetylated to form the acetoxy derivative that presumably breaks down spontaneously to generate the nitrenium ion, the likely ultimate mutagen.  相似文献

To clarify spatial and seasonal differences in net plankton and zoobenthos in Lake Tonle Sap, Cambodia, quantitative surveys were carried out at 14 stations in the north and south basins in high- and low-water seasons during 2003–2005. In the phytoplankton communities, a diatom Aulacoseira granulata dominated throughout the lake in the high-water seasons, while blue-green algae, mostly composed of Microcystis, surpassed other algae in the low-water season when the lake water was very turbid and the Secchi disk readings were only a few centimeters. In the low-water seasons, a bloom of floating blue-green algae occurred everywhere, especially prominent in the coastal areas. Protozoans and rotifers dominated the zooplankton communities. In the open-water stations, diversity was higher in high-water seasons in phytoplankton, while it was not significantly different between seasons in zooplankton. Composition of plankton communities in Lake Tonle Sap appears to have changed little since the 1950s, at least in phytoplankton, while the phytoplankton density appears to be higher in the present study. Among the macrozoobenthos, mollusks, oligochaetes and chironomids dominated in density, and mollusks exceeded others in biomass in both basins and seasons. The total densities of macrozobenthos were not high, being fewer than 1,300 m−2 throughout the stations and seasons. Possible reasons for the low zoobenthos abundance in the lake may include high predation pressures by benthivorous fish or unfavorable unstable and flocculant substrates.  相似文献

To identify the major mutagen in pyroligneous acid (PA), 10 wood and 10 bamboo pyroligneous acids were examined using the Ames test in Salmonella typhimurium strains TA100 and TA98. Subsequently, the mutagenic dicarbonyl compounds (DCs), glyoxal, methylglyoxal (MG), and diacetyl in PA were quantified using high-performance liquid chromatography, and the mutagenic contribution ratios for each DC were calculated relative to the mutagenicity of PA. Eighteen samples were positive for mutagens and showed the strongest mutagenicity in TA100 in the absence of S9 mix. MG had the highest mutagenic contribution ratio, and its presence was strongly correlated with the specific mutagenicity of PA. These data indicate that MG is the major mutagen in PA.  相似文献
