Aphids (superfamily Aphidoidea) contain eubacterial endosymbionts localized within specialized cells (mycetocytes). The endosymbionts are essential for the survival of the aphid hosts. Sequence analyses of the 16S rRNAs from endosymbionts of 11 aphid species from seven tribes and four families have indicated that the endosymbionts are monophyletic. Furthermore, phylogenetic relationships within the symbiont clade parallel the relationships of the corresponding aphid hosts. Our findings suggest that this endocytobiotic association was established in a common ancestor of the four aphid families with subsequent diversification into the present species of aphids and their endosymbionts.  相似文献

The effects of neem (Azadirachta indica A. Juss) seed kernel extracts (NSKE) and azadirachtin on the ability of Myzus persicae (Sulz.) to transmit potato leafroll luteovirus (PLRV) was studied. Moreover, it was investigated whether treatments with these compounds would exert an effect on larval growth and mortality, and on the aphid intracellular symbionts. Endosymbiotic bacteria play an essential role in the performance of aphids, and in luteovirus transmission by aphids. NSKE and azadirachtin were offered to one-day-old M. persicae nymphs via a membrane feeding system. The neem metabolites displayed a 100% mortality at doses higher than 2560 ppm. At intermediate doses, ranging between 320 and 2560 ppm, larval growth and mortality were affected in a dose-dependent manner. The transmission of PLRV by M. persicae was inhibited by 55–90%. The endosymbiont population of the aphid was clearly affected by a treatment with neem metabolites as the release of their most abundant protein, Buchnera GroEL, into the haemocoel of the aphid was inhibited. Moreover, morphological aberrations on the bacterial endosymbionts were observed in aphids which fed on 2560 ppm of azadirachtin. At doses lower than 160 ppm of NSKE or azadirachtin, the endosymbiont population of M. persicae, and mortality, growth and feeding behaviour were similar to that of the untreated groups of aphids. However, PLRV transmission was still inhibited by 40–70%. The possible targets of the neem metabolites in the aphid are discussed.  相似文献

Embryo production in aphids is absolutely dependent on the function of symbiotic bacteria, mainly Buchnera, and the growth and development of koinobiont parasitoids in aphids requires the diversion of nutrients from aphid embryo production to the parasitoid. The implication that the bacterial symbiosis may be promoted in parasitized aphids to support the growing parasitoid was explored by analysis of the number and biomass of mycetocytes, and the aphid cells bearing Buchnera, in the pea aphid Acyrthosiphon pisum Harris (Hemiptera: Aphididae) parasitized by the wasp Aphidius ervi Haliday (Hymenoptera: Braconidae). Aphids hosting a young larval parasitoid bore more mycetocytes of greater total biomass, and embryos of lower biomass than unparasitized aphids. Furthermore, one of the three aphid clones tested, which limited teratocyte growth (giant cells of parasitoid origin having a trophic role), bore smaller mycetocytes and larger embryos, than one or both of the two aphid clones with greater susceptibility to the parasitoid. These data suggest that susceptibility of the aphid‐Buchnera symbiosis to parasitoid‐mediated manipulation may, directly or indirectly, contribute to aphid susceptibility to parasitoid exploitation.  相似文献

The emergence of resistance mechanisms to, and revocation of, many insecticides used in the control of the polyphagus aphid pest, Myzus persicae (Sulzer), has increased the pressure to develop novel approaches for the control of the pest in many crops. Kaolin-based particle films provide a physical barrier against insect pests and show considerable potential for controlling M. persicae. We conducted a series of laboratory experiments to investigate the mode of action of kaolin against aphids. The material appeared to have no direct effect on M. persicae; spraying adult aphids with aqueous kaolin suspension had no significant impact on their subsequent survival or reproduction on untreated plants. Similarly, when aphids were placed on kaolin-treated host-plants (Brassica oleracea), their performance (survival, growth rate and reproduction) was not significantly different from aphids on untreated plants. However, when M. persicae were given a choice between kaolin-treated and untreated (or water solvent-treated) leaf areas, both adults and nymphs exhibited a significant preference for non-kaolin-treated host-plant material. Rejection of kaolin-treated plant material occurred very rapidly (within 20 min) and this behavioural effect may be related to the efficacy of kaolin in controlling aphids under field conditions.  相似文献

为了明确饲养温度对黑豆蚜Aphis fabae 内共生菌和宿主蚜虫体型大小的影响,对在室内不同温度下饲养的黑豆蚜内共生菌胞数量和宿主蚜虫体型大小进行了观察和统计分析。结果表明,温度对同一发育时期蚜虫内共生菌胞数量的影响在不同温度范围内有所不同,1龄若蚜体内的菌胞数量除在25℃与35℃间有显著差异外,在其余各温度间没有显著差异; 其余时期的蚜虫内共生菌胞数量在高温(> 30℃)下显著低于较低温度下的菌胞数量,存在负直线相关性。温度对菌胞数量随宿主发育到产仔前的变化趋势有不同程度的影响,在较低温度(15℃、20℃和25℃)下,菌胞数量随虫体发育显著增加; 但在高温(30℃和35℃)下,蚜虫体内菌胞数逐渐增加直到3龄达到最高,然后略有下降(30℃)或显著下降(35℃)。除1龄若蚜外,蚜虫体型大小总体呈现随温度升高而降低的格局,但随其内共生菌数量增多而增大(35℃下除外)。据此认为,温度可能通过作用于蚜虫内共生菌胞数量而影响蚜虫体型的大小。  相似文献

Three instar larvae could be observed forAphelinus varipes. In the second and third instars, the colon which forms the largest part of the hindgut was composed of secretory cells. There was a reduction in the size and number of embryos produced by parasitized aphids. Fat cells of parasitized aphids also degenerated sooner than those of unparasitized aphids. The number of mycetocytes remained higher in parasitizedRhopalosiphum padi than in non-parasitized aphids. Abstract:  The effect of a genetically modified eggplant line expressing oryzacystatin on Myzus persicae (Sulzer) and Macrosiphum euphorbiae (Thomas) was examined. The transgenic eggplant reduced the net reproductive rate ( R 0), the instantaneous rate of population increase ( r ), and the finite rate of population increase ( λ ) of both aphids species compared with a control eggplant line. The mean generation time ( T ) of the aphids was unaffected by the transgenic plants. Age-specific mortality rates of M. persicae and M. euphorbiae were higher on transgenic plants. These results indicate that expression of oryzacystatin in eggplant has a negative impact on population growth and mortality rates of M. persicae and M. euphorbiae and could be a source of plant resistance for pest management of these aphids.  相似文献

Abstract.  1.  Myzus persicae sensu lato demonstrates considerable genetic variation in respect to adaptation to host plants. The subspecies M. persicae nicotianae shows a preference for tobacco, while M. persicae sensu stricto ( s. str. ) for other herbaceous plants. Given that winged colonisers of several aphid species play an important role in selecting host plants, here their role in the host specialisation observed in M. persicae was examined in choice and no-choice tests conducted outdoors, in performance studies, and in DC Electrical Penetration Graph (DC-EPG) studies.
2. In outdoor choice tests, 77% of spring migrants of M. persicae nicotianae chose tobacco, whereas equal proportions of M. persicae s. str. selected tobacco and pepper. In no-choice tests, spring migrants settled more quickly after alighting on host rather than on non host plants, and significantly more alate M. persicae s. str. (27%) than M. persicae nicotianae (2%) left tobacco after walking briefly on the leaf surface, whilst no significant differences were found on pepper. Cross-host transfers significantly reduced the fecundity of both summer and spring migrants of the two subspecies. Finally, the results of no-choice tests and DC-EPG studies showed that winged aphids distinguished their host through cues located on the plant surface or in subcutaneous tissues perceived prior to the initiation of feeding.
3. This study demonstrates the important role of winged colonisers in the evolution of host specialisation in M. persicae . The multifarious divergent selection that the two host forms experience, i.e. the selection against cross-migrants and their subsequent generations, is a crucial factor involved in the development and maintenance of host specialisation and promotes the parallel evolution of improved host-recognition ability.  相似文献

Abstract The green peach aphid ( Myzus persicae ) is becoming resistant to many of the pesticides used in the tomato ( Lycopersicon esculentum ) industry. A potential alternative method of control is host-plant resistance, which may be mediated by trichomes. Resistance to the green peach aphid was assessed for six accessions of L. hirsutum, three of L. pennellii and a L. esculentum control. Mortality, entrapment and number of aphids unaffected by trichomes were recorded 24, 48 and 72 h after placement. Using multiple regression against counts of glandular and non-glandular trichomes, the role of foliar pubescence in resistance to the aphid was established. Lycopersicon pennellii possessed the highest level of resistance after 72 h and type IV trichome density accounted for 26.5% of variance in mortality ( P  < 0.001). Low densities of type IV and high densities of type VII trichomes increased the number of aphids 'unaffected' by trichomes, a relationship accounting for 29% of variance ( P  < 0.001). Information on which types of trichomes most strongly affect resistance to insect pests will aid future breeding and L. pennellii shows potential for use in breeding programs to increase levels of host-plant resistance in L. esculentum.  相似文献

【目的】为探究烟蚜茧蜂Aphidius gifuensis对烟蚜Myzus persicae寄生后烟蚜生长发育和繁殖影响及其在烟蚜防控中的作用。【方法】采用室内叶片培养皿法,测定了不同龄期烟蚜(1-4龄若蚜及成蚜)被烟蚜茧蜂寄生后烟蚜当代(F0)及后代(F1)的发育历期、寿命、产仔量。【结果】烟蚜1龄若蚜被烟蚜茧蜂寄生后,1龄若蚜历期受到显著影响,较未被寄生对照缩短了0.65 d (33.00%),而2-4龄若蚜期和成蚜历期未受显著影响;2龄若蚜被寄生后,4龄若蚜和成蚜历期均受到显著影响,较未被寄生的对照分别缩短了1.50 d (100.00%)和0.83 d (100.00%),而2和3龄若蚜期未受显著影响;3龄若蚜被寄生后,4龄若蚜和成蚜期均受到显著影响,较未被寄生对照分别缩短了1.50 d (100.00%)和0.83 d (100.00%),而3龄若蚜期未受显著影响。2龄若蚜被寄生后烟蚜的寿命和产仔量均受到显著影响,寿命较未被寄生对照缩短了8.39 d(54.00%),每雌产仔量较未被寄生对照减少了48.67头(100.00%)。此外,2-3龄若蚜被寄生后其后代(F1)1龄若蚜历期受影响最大,较未被寄生烟蚜后代1龄若蚜期缩短了0.80 d(40.00%),其F1代平均寿命较未被寄生烟蚜后代平均寿命延长了5.72 d(41.20%),但其产仔量与对照差异不显著。【结论】不同龄期烟蚜被烟蚜茧蜂寄生后,其发育历期和寿命比未被寄生的缩短,产仔量比被未寄生的少,但其F1代平均寿命比未被寄生后代烟蚜的长且其产仔量未受显著影响。当代2龄若蚜期是控制烟蚜增长的关键时期。这些结果对确定烟蚜茧蜂防治烟蚜适期及认识其机理具有重要参考意义。  相似文献

Prokaryotic symbionts of aphids, which are important to the survival of the insect, are housed in specialized cells, mycetocytes. In a study of the aphids Acyrthosiphon pisum and Megoura viciae, the mycetocytes were found to exhibit a consistent pattern of variation in both size and number, both with developmental age of the insect and between alate and apterous morphs. The number of mycetocytes ranged between 70–90 and 60–70 in 1-day old larvae of Megoura viciae and Acyrthosiphon pisum, respectively, and tended to 0 in post-reproductive individuals of both species, with a decrease of 50% between birth and the time of the final moult (days 7–9) in alates and in the late-reproductive period (days 17–18) in apterae. The volume of mycetocytes of M. viciae increased with age from 1.17 × 10−5 mm3 in 1-day old larvae to 7.63 × 10−5 mm3 and 5.09 × 10−5 mm3 in apterous and alate teneral adults (day 8), respectively, and this difference between the morphs closely mirrors the difference between the relative growth rates of apterous and alatiform larvae. It is suggested that mycetocyte loss may represent an important means by which the symbiont population is regulated. The results can also be interpreted as evidence for substantial variation in the characteristics of nutritional interactions between the aphid and its symbionts with age and morph of the aphid.  相似文献

Hairy nightshade, Solanum sarrachoides (Sendtner), is a ubiquitous weed in potato agro-ecosystems and nonagricultural lands of southeastern Idaho and the Pacific Northwest. This weed increases the complexity of the Potato leafroll virus (PLRV) (Luteoviridae: Polervirus)-potato pathosystem by serving as aphid and virus reservoir. Previous field studies showed higher densities of green peach aphid, Myzus persicae (Sulzer), and potato aphid, Macrosiphum euphorbiae (Thomas), the two most important vectors of PLRV, on S. sarrachoides compared with potato plants in the same fields. Some of the S. sarrachoides plants sampled in these surveys tested positive for PLRV. Viral infections can alter the physiology of plant hosts and aphid performance on such plants. To understand better the potential effects of S. sarrachoides on the PLRV-potato-aphid pathosystem, the life histories of M. persicae and M. euphorbiae were compared on virus-free and PLRV-infected S. sarrachoides and potato. Individual nymphs of each aphid species were held in clip cages on plants from each treatment to monitor their development, survival, and reproductive output. Nymphal survival for both aphids across plant species was higher on S. sarrachoides than on potato, and, within plant species, it was higher on PLRV-infected plants than on noninfected plants. With a few exceptions, similar patterns occurred for fecundity, reproductive periods, adult longevity, and intrinsic rate of increase. The enhanced performance of aphids on S. sarrachoides and on PLRV-infected plants could alter the vector population dynamics and thus the PLRV-disease epidemiology in fields infested with this weed.  相似文献

Aphids are important pests of crop plants in Europe. Increasing resistance of aphids to insecticides and their side effects on the environment and non target organism's including human's stimulated research on alterative methods of aphid control, including the use of entomotoxic proteins. Lectins are carbohydrate binding proteins that are widely distributed in nature; they have been isolated from microorganisms, fungi, plants and animals. Several of these proteins were tested for their potential biocide effect on plenty of pests. A fungal lectin, namely Xerocomus Chrysenteron lectin (XCL) was previously purified and was shown to be toxic for several pests including aphids. XCL was clearly the most toxic lectin against M. persicae. In this work, bioassays using artificial diets incorporating a broad range of XCL concentrations (from 10 microg x ml(-1) to 5000 microg x ml(-1)) were developed to assess the negative effects of XCL on the biological parameters (development duration, weight and fecundity) of M. persicae a polyphagous aphid found on more than 400 host plant species and transmitting more than 100 viral diseases. A significant mortality of aphids was observed, corresponding to the LC50 and LC90 of 0, 46 and 6, 02 mg/ml respectively after 24hrs. Significant differences of M. persicae weight, development duration and fecundity (P < 0.05) was observed between the tested XCL concentrations. Conavalia ensifomris lectin (ConA) was included as lectin reference on the bioassay experiments and was shown to be less toxic and induced lower negative changes in M. persicae biological parameters when compared with XCL.  相似文献

【目的】研究筛选对桃蚜Myzus persicae有致死作用的安全微波频率和照射时长,以为探究新型物理防蚜技术,弥补化学防治上的缺陷提供参考依据。【方法】在暗箱中,应用微波发射仪分别发射1375, 2 750, 5 500和11 000 MHz 4个不同频率的微波照射桃蚜1日龄无翅成蚜,每个频率的照射时长分别为15, 30, 60和120 s;照射后在人工气候箱中饲养,分别于照射后8, 24, 48和72 h观察其生长发育及繁殖状况,统计桃蚜死亡率、繁殖力(累计产蚜量)及子代有翅蚜率。【结果】4个不同频率的微波分别在4个不同照射时长下,对桃蚜1日龄无翅成蚜的死亡率、繁殖力和子代翅型分化都有不同程度的影响。照射后72 h, 5 500 MHz微波照射时间为15 s时对桃蚜1日龄无翅成蚜的致死作用最强,死亡率达到55.00%,在照射时间为30和120 s时可抑制子代桃蚜向有翅蚜的分化。2 750 MHz微波照射30和60 s时促进桃蚜1日龄成蚜繁殖,照射30 s时繁殖力最强,而照射15和120 s时却表现为抑制繁殖,且2 750 MHz微波照射30 s能抑制子代桃蚜向有翅蚜分化。【结论】微波辐射能够影响桃蚜1日龄成蚜的存活、繁殖和子代翅型分化。本研究初步筛选出了对桃蚜1日龄无翅成蚜有致死作用的微波频率和照射时长。  相似文献

The Mi-1.2 gene in tomato confers resistance against certain clones of the potato aphid (Macrosiphum euphorbiae). This study used 2D-DIGE coupled with protein identification by MALDI-TOF-MS to compare the proteome patterns of avirulent and semivirulent potato aphids and their bacterial endosymbionts on resistant (Mi-1.2+) and susceptible (Mi-1.2−) tomato lines. Avirulent aphids had low survival on resistant plants, whereas the semivirulent clone could colonize these plants. Eighty-two protein spots showed significant quantitative differences among the four treatment groups, and of these, 48 could be assigned putative identities. Numerous structural proteins and enzymes associated with primary metabolism were more abundant in the semivirulent than in the avirulent aphid clone. Several proteins were also up-regulated in semivirulent aphids when they were transferred from susceptible to resistant plants. Nearly 25% of the differentially regulated proteins originated from aphid endosymbionts and not the aphid itself. Six were assigned to the primary endosymbiont Buchnera aphidicola, and 5 appeared to be derived from a Rickettsia-like secondary symbiont. These results indicate that symbiont expression patterns differ between aphid clones with differing levels of virulence, and are influenced by the aphids’ host plant. Potentially, symbionts may contribute to differential adaptation of aphids to host plant resistance.  相似文献

Winter tree pruning is a cultural practice known to modify vegetative growth, which is likely to affect the development of pests. However, it has been poorly addressed as a cultural control method for diminishing the population levels of the green peach aphid, Myzus persicae (Sulzer) (Homoptera: Aphididae), in peach [ Prunus persica (L.) Batsch (Rosaceae)] orchards. In this study, we conducted a 2-year, on-station experiment to evaluate how winter pruning affects peach– M. persicae interactions, by examining tree vegetative growth, aphid population dynamics, and crop yield and fruit quality. We collected data under an insect-proof shelter on adult peach trees submitted to various levels of pruning and artificially infested with aphids. Our results showed that pruning enhanced shoot growth due to the proportion of growing shoots, which increased exponentially (10–60%), whereas the growth rate of growing shoots was not affected. The degree of infestation of peach trees increased with increasing pruning intensity. This effect was mainly due to the increase of the proportion of growing shoots, on which aphids developed better than on rosettes. In turn, the higher the aphid infestation, the higher the aphid-induced shoot-tip damage, leaf curling, and leaf fall that disturbed the growth of growing shoots. However, aphids did not considerably reduce fruit quality at harvest. They did not affect fresh fruit weight, and the refractometric index (indicator of sugar content) was reduced by only 3–4%. The relevance of winter pruning as a cultural method for pest control in orchards conducted under integrated fruit production guidelines is discussed.  相似文献

Two groups of brussels sprout plants of one variety given different fertilizer treatments proved ‘resistant’ and ‘susceptible’ to both Myzus persicae and Breicoryrie brassicae, as measured by aphid mean relative growth rate. The hypothesis that leaf disc would fail to reflect differences in ‘nutritionallybased’ plant resistance was confirmed with B.brussicue but not M.persicae. Both aphids grew more slowly on discs than leaves of ‘susceptible’ plants; the reverse tendency showed on ‘resistant’ material. Total:(, amino nitrogen proved poorly correlated with aphid performance. Previously published (van Emden & Bashford, 1971) equations relating individual amino acids to aphid performance gave a better fit to the results, but still failed to account for the reduced performance of the aphids on ‘susceptible’ discs.  相似文献

Abstract:  The predatory effect of female adults of Coccinella septempunctata L. (Coleoptera: Coccinellidae) on aphid Macrosiphum euphorbiae (Thomas) was examined under laboratory conditions, in controlled environmental chambers. This study took place on single rose leaves in transparent small plastic cages at varying proportions of predator/number of aphids. Coccinella septempunctata proved to be an effective predator for the biological control of M. euphorbiae under controlled conditions in closed environments. It is proposed to use starved adult females of C. septempunctata in biological control programmes in greenhouses, as a specialized predator of M. euphorbiae in proportions near 1 : 35 predator/aphids. The percentage of aphids that may escape was correlated to the hunger satiation of the predator and the total number of aphids, and the theoretical models describing the predator's behaviour under controlled conditions were defined.  相似文献

Potato leafroll virus (PLRV; genus Polerovirus, family Luteoviridae) is a persistently transmitted circulative virus that depends on aphids for spreading. The primary vector of PLRV is the aphid Myzus persicae (Sulzer) (Homoptera: Aphididae). Solanum tuberosum L. potato cv. Kardal (Solanaceae) has a certain degree of resistance to M. persicae: young leaves seem to be resistant, whereas senescent leaves are susceptible. In this study, we investigated whether PLRV‐infection of potato plants affected aphid behaviour. We found that M. persicae's ability to differentiate headspace volatiles emitted from PLRV‐infected and non‐infected potato plants depends on the age of the leaf. In young apical leaves, no difference in aphid attraction was found between PLRV‐infected and non‐infected leaves. In fact, hardly any aphids were attracted. On the contrary, in mature leaves, headspace volatiles from virus infected leaves attracted the aphids. We also studied the effect of PLRV‐infection on probing and feeding behaviour (plant penetration) of M. persicae using the electrical penetration graph technique (DC system). Several differences were observed between plant penetration in PLRV‐infected and non‐infected plants, but only after infected plants showed visual symptoms of PLRV infection. The effects of PLRV‐infection in plants on the behaviour of M. persicae, the vector of the virus, and the implications of these effects on the transmission of the virus are thoroughly discussed.  相似文献
