Summary The influence of reserpine on the argentaffin and non-argentaffin argyrophile cells of the duodenum has been studied in 8 dogs. In two dogs that were given 5 mg/kg of reserpine there was a complete disappearance of both types of cells. In another dog given 5 mg/kg and in dogs given 2.5 mg/kg and 1 mg/kg reserpine, the number of argentaffin cells was reduced to almost nil while the number of non-argentaffin argyrophile cells showed a statistically significant reduction. The degree of depletion of non-argentaffin argyrophile cells was proportional to the dose of reserpine given. The findings show that the non-argentaffin argyrophile cells are closely related to 5-hydroxytryptamine and their distinction from argentaffin cells is only a quantitative one.  相似文献

Summary The individual granules of enterochromaffin cells of normal and reserpine treated guinea pigs have been studied by staining slides of the duodenum first by an argentaffin method and subsequently by an argyrophile method. Some argentaffin cells can be shown to contain not only argentaffin granules, but also granules that are purely argyrophile. The relative number of such argentaffin cells is greatly increased following administration of reserpine, as depletion of their 5-hydroxytryptamine content converts argentaffin granules into purely argyrophile ones. On the basis of this finding it is confirmed that the argyrophile granule is merely an argentaffin granule depleted of its 5-HT content, and that the argyrophile (nonargentaffin) and the argentaffin cells represent different phases of a secretory cycle.This investigation was supported by a grant from the Indian Council of Medical Research. Summary Successive staining of cells of the enterochromaffin system of the guinea pig by an argentaffin and an argyrophile method shows that, as in man and rabbit, they can be divided into (a) those in which all granules are apparently argentaffin, (b) those in which all granules are argyrophile but none are argentaffin, and (c) those in which some granules are argentaffin (as well as argyrophile) while others are purely argyrophile. Summary The distribution of argyrophile and argentaffin cells has been studied in the small intestine of five human adults. In proceeding cranio-caudally the characteristic feature of their distribution is the presence of eight to ten waves of rising and falling density. A progressive decrease in density of cells from duodenum to terminal ileum (described by previous workers) is not present.Re-examination of findings reported earlier in the small intestines of human foetuses shows that a predominant U shaped pattern of distribution is present in younger foetuses. This changes to the adult pattern by full term. The appearance of the adult pattern occurs earlier for argyrophile cells than for argentaffin cells.  相似文献

Amount of argyrophile and argentoffin endocrine cells per 1 mm2 of the large intestine mucous membrane has been counted in the turtle, pigeon, white rat and frog at various periods of their life (winter hibernation, awakening, active life). As the animal's organization becomes more complicated, the amount of the endocrine cells in the epithelium increases both in the initial and in the terminal parts of the large intestine, their amount in the terminal (rectum) part being less than in the initial one. The greatest amount of the endocrine cells is found in simmer frogs at the expense of increasing content of the elements that do not belong to the argentaffin group. Summary The relationship between the argentaffin and argyrophile cells of the human gastro-intestinal tract has been studied by staining paraffin sections first by the Masson-Hamperl method and then by the Bodian method. The investigations confirms the author's earlier observation (Singh 1963) that all argentaffin cells of the human gastro-intestinal tract are also argyrophile and that there is no evidence to support the claim of Hellweg (1952) and of Hamperl (1952) regarding the presence of non-argyrophile argentaffin cells.  相似文献

The aim of this study was to identify and quantify the argyrophil, argentaffin and insulin-immunoreactive cells (IIC) in the small intestine of the opossum Didelphis aurita. Seven adult male specimens of opossums were investigated. The animals were captured, and their blood insulin levels were determined. After euthanasia, fragments of the small intestine were processed for light microscopy and transmission electron microscopy, and submitted to histochemistry and immunohistochemistry for identification of argyrophil and argentaffin endocrine cells, and IIC. Argyrophil and argentaffin cells were identified in the intestinal villi and Liberkühn crypts, whereas IIC were present exclusively in the crypts. Ultrastructure of the IIC revealed cytoplasmic granules of different sizes and electron densities. The numbers of IIC per mm2 in the duodenum and jejunum were higher than in the ileum (p < 0.05). The animals had low levels of blood insulin (2.8 ± 0.78 μIU/ml). There was no correlation between insulin levels and the number of IIC in the small intestine. The IIC presented secretory granules, elongated and variable morphology. It is believed that insulin secretion by the IIC may influence the proliferation of cells in the Liberkühn crypts, and local glucose homeostasis, primarily in animals with low serum insulin levels, such as the opossum.  相似文献

Summary The relationship between the argentaffin and argyrophile cells of the human gastrointestinal tract has been studied, in foetal and adult material, by a technique involving the staining of sections first by an argentaffin method (Gomori-hexamine silver, Schmorl, Diazonium) and subsequently by an argyrophile method (Bodian). A comparison of the cells staining by the two methods shows that all argentaffin cells of the human gastrointestinal tract are also argyrophile and that there is no evidence to support the claim of Hellweg (1952) and of HamPerl (1952) regarding the presence of non-argyrophile argentaffin cells.W. The intestinal of the 3-toed sloth, Bradypus tridactylus, was studied macroscopically, with light microscope and with histochemical methods for mucosubstances. Macroscopically, the inner surface of the duodenum shows longitudinal and circular folds. There is no caecum, nor appendix. The large intestine consists of a short colon and a large rectal pouch, which has a thick wall. The mucosa of the small intestine has long leaf-shaped villi covered with columnar epithelium having a well developed striated border, and the goblet cells are scattered among the columnar cells. An association between neutral and acidic mucosubstances was detected in the goblet cells. The duodenal (Brunner's) glands are confined exclusively in the lamina propria of the duodenum. No Paneth cells were observed in the crypt lining. Argyrophil and argentaffin cells were found in the entire length of the intestine. The large intestine does not possess villi, but many goblet cells were observed in its mucosa.  相似文献

This study aimed to investigate the distribution of argyrophil, argentaffin, and insulin-immunoreactive endocrine cells in the large intestine of opossums (Didelphis aurita) and to describe the ultrastructure of the secretory granules of insulin-immunoreactive endocrine cells. Fragments of the large intestine of 10 male specimens of D. aurita were collected, processed, and subjected to staining, immunohistochemistry, and transmission electron microscopy. The argyrophil, the argentaffin, and the insulin-immunoreactive endocrine cells were sparsely distributed in the intestinal glands of the mucous layer, among other cell types of the epithelium in all regions studied. Proportionally, the argyrophil, the argentaffin, and the insulin-immunoreactive endocrine cells represented 62.75%, 36.26%, and 0.99% of the total determined endocrine cells of the large intestine, respectively. Quantitatively, there was no difference between the argyrophil and the argentaffin endocrine cells, whereas insulin-immunoreactive endocrine cells were less numerous. The insulin-immunoreactive endocrine cells were elongated or pyramidal, with rounded nuclei of irregularly contoured, and large amounts of secretory granules distributed throughout the cytoplasm. The granules have different sizes and electron densities and are classified as immature and mature, with the mature granules in predominant form in the overall granular population. In general, the granule is shown with an external electron-lucent halo and electron-dense core. The ultrastructure pattern in the granules of the insulin-immunoreactive endocrine cells was similar to that of the B cells of pancreatic islets in rats.  相似文献

Summary Argyrophilic and argentaffin cells occur in the stomach and intestinal epithelium of the sea-squirt, Ciona intestinalis L.. These cells are characterized by their basal swelling which contains the nucleus surrounded by small secretory granules and by a filamentous cell-apex which reaches the gut lumen. The cells are scattered unevenly within the epithelium. Their number decreases rapidly towards the lower part of the intestine. The localization, size of granules and their shape are features which differentiate these cells from other secretory cells in the gut epithelium such as mucous cells. In the gastric mucous membrane of 40 rats representing 4 age groups (pubertal, mature, old and very old age) per 10 animals in each, thickness of the mucous membrane has been measured by the argyrophil method of Grimelius and the argentaffin method of Masson - Hamperl and the number of the endocrine cells has been counted. The thickness of the mucous membrane and the length of its glands, as well as the number of argyrophil cells increase from the pubertal towards the mature age. From the mature up to the old age the number of argyrophil cells rises in the antral part, and in the acid-producing zone it significantly decreases. The number of argentaffin cells in the stomach does not change from the pubertal towards the old age. In the very old age, against the background of atrophy of gastric glands, the number of both argyrophil and argentaffin cells decreases in both parts of the stomach studied. The data obtained are discussed taking into consideration functional role of various types of endocrine cells, producing biologically active peptides and monoamines.  相似文献

Histological and electron microscopic investigations of mucosal strucutre of the small intestine in gnotobiotic and conventional rats have demonstrated that mucosal index (lenear dimentions of villi/crypt dimentions ratio) is equal to 3--4 and 1.4--2.5, respectively. In gnotobiotic rats the number of mitotic figures in crypts is less, migratory-desquamative parameters of enterocytes are 5--6 days, cranio-caudal gradient of linear dimentions of villi, crypts and the number of goblet-shaped cells is not expressed. The goblet-shaped cells and cells with acidophilic granules have secretion of moderate intensity. The number of cells infiltrating epithelium of villi and crypts is greater when microbes are present in the gut lumen. A considerable difference in the structure of the connective tissue basis of the gnotobiotic and conventional rats mucosa are also connected with presence or absence of microflora in the gut lumen.  相似文献

The time, when the first villi appear, is defined. Quantitative composition, percent content, height of the villi of the mucous membrane in all parts of the small intestine during various periods of the intrauterine development are presented. In 17-week-old embryos at the level of the duodenum the first villi are noted and their formation in the middle part of the intestinal tube is revealed. By the 8th-9th weeks of development they appear at the level of the caudal part of the intestine. This demonstrates asynchronism of their formation in the cranio-caudal direction. The periods of their intensive growth in height in the first part of the intestine correspond to 8-10, 12-16, 24 weeks of the fetus age, in the second part--to 8-10, 18-20 weeks, in the third--to 10-12, 16-18, 26-28 weeks. These periods alternate with an abatement in growth intensity, which is connected with an increasing width of the villi themselves, with amount of newly formed villi and with changes of the intestinal tube diameter. The epithelium of the distal part of intestine of the lizard Lacerta vivipara has been studied by light and electron microscopy. The total number of endocrinocytes (argyrophilic cells) was found to increase from small bowel (57 +/- cell/mm2) to colon (9 +/- 69), and cloaca (99 +/- 8). Although the number of argentaffin cells increases from the small bowel to colon, cell decrease occurs from colon (42 +/- 6 cell/mm2) to cloaca (65 +/- 10 cell/mm2). On electronograms of the colon mucosal epithelium four types of endocrinocytes were identified. Type I--with secretory granules polymorphic for the size and form, with a high electron density core, and average size 206 +/- 31 nm. Type II--with secretory granules 265 +/- 20 nm in size, having spherical form and highly electronic dense contents. Type III--contains largest (350 +/- 12 nm), spherical, oval or irregularly-shaped secretory granules, with contents of various electronic density. Type IV--endocrine cells having small (176 +/- 5 nm) spherical or oval secretory granules with a highly electronic dense core. Besides, "mixed" cells were identified, whose cytoplasm contained simultaneously mucous and endocrinous granules.  相似文献

Cytologic specimens of neuroendocrine tumors metastatic to the liver were examined with regard to their silver staining properties after the application of argentaffin and argyrophil staining techniques (Masson, Grimelius and Sevier-Munger). In tumors with a content of serotonin (small intestine carcinoids), the presence of this substance was demonstrated cytologically as an argentaffin reaction in individual tumor cells; however, formalin fixation was a prerequisite for positive staining. Melanin in malignant melanoma cells displayed a positive argentaffin reaction, irrespective of the fixation used (air drying, formalin, Bouin's fluid or acetone-alcohol). Thus, serotonin and melanin can be distinguished in cytologic samples of neuroendocrine tumors by the use of the Masson argentaffin reaction with different fixatives. The nonargentaffin-positive neuroendocrine tumor cells were weakly stained or unreactive with the Grimelius argyrophil technique. The Sevier-Munger argyrophil technique was negative or gave a disturbing nonspecific background staining reaction that was difficult to interpret in the cytologic samples. Thus, the Grimelius method appears to be the most useful silver stain for identifying neuroendocrine tumor cells in cytologic material, irrespective of their hormone content, since both argentaffin-positive and argentaffin-negative cell samples were stained at least to some degree.  相似文献

白鲟消化道具有肉食性鱼类的典型特征,其口咽腔结构既适合捕食又适合吞食与滤食水生动物。咽后消化道可分为食道、胃后行支、胃前行支、小肠、瓣肠、直肠与肛门。幽门盲囊似一致密器官,小肠与瓣肠连接处有一特殊淋巴器官,肛门两侧有腹孔。白鲟口咽腔被覆层扁平上皮,上皮内有味蕾分布。咽后消化道组织分层为粘膜(无粘膜肌层)、粘膜下层(小肠及瓣肠前部无)、肌层与外膜。粘膜上皮为单层柱状上皮,由纤毛柱状细胞、一般柱状细胞和杯状细胞组成,其间还散在有颗粒细胞和游走细胞。食道后部与胃的一般柱状细胞为分泌粘液的细胞,肠内的一般柱状细胞为吸收细胞。胃后行支及部分前行支固有膜内有消化腺,其余各部的固有膜为致密层。小肠前中部粘膜形成蜂窝状粘膜窦,无肠腺。除食道前部肌层中有横纹肌外,其余部的肌层均为平滑肌。外膜内结缔组织有的致密有的疏松,外膜表面细胞柱状或立方形或扁平。  相似文献

The influence of 50% proximal resection of the small intestine on the intestinal endocrine system was investigated on white male rats. The quantitative changes of argyrophil and argentaffin cells correlated with their secretory activity changes. The opposite character of secretory function synchronization was revealed on the 7th and 30th days along the length of the small intestine (proximo-distal sinusoid phenomenon). The secretory function normalization of the endocrine system is carried out in the distal and proximal direction along the length of the small intestine.  相似文献

ABSTRACT Fifth instars of Triatoma infestans with established Trypanosoma cruzi infections were dissected after different periods of starvation to determine the population density and the percentage of different developmental stages of T. cruzi in the small intestine and rectum of the bugs. After a short starvation period of 20 days, the population density in the small intestine was 20% (about 60,000) of the rectal population. The population in the small intestine was strongly reduced after an additional ten days of starvation, and no flagellates could be found there 60, 90 and 120 days after the last feeding. In the rectum, this reduction went down to 1% of the initial population, but a total elimination never occurred. Usually the remaining population contained more live than dead flagellates. Starvation also resulted in an increase in the rectum in the number and percentage of drop-like forms, intermediates between sphere- and epi- or trypomastigotes, from 1% initially to about 10% after 90 days of starvation. The percentage of spheromastigotes increased from 2% at 20 days after the last feeding to about 20% after an additional 40 and 70 days. Therefore, the spheromastigotes of T. cruzi seem to be induced by stress conditions.  相似文献

Summary In this investigation glueagon has been histochemically demonstrated by the fluorescent antibody technique in the developing gastrointestinal tract of the chick embryo. The levels of monoamine oxidase activity at various stages during the development of the gastrointestinal tract has been studied. Both monoamine oxidase activity and argentaffin positive cells have been demonstrated histochemically in the epithelial cells of the embryonic stomach-complex and intestine. This similarity in the localisation suggests that monoamine oxidase activity may be related with the seratonin producing cells.  相似文献
