A β-glucuronidase has been isolated from pig kidney and purified 1600-fold using sodium desoxycholate precipitation, ammonium sulphate fractionation, heat treatment and chromatography on Sephadex G200, DEAE-cellulose (DE-52) and hydroxyapatite. The enzyme activity was assayed using oestrone 3-glucuronide as substrate; the final specific activity was 254 nmol oestrone/min/mg of protein. The purified enzyme showed apparent homogeneity in gel filtration and polyacrylamide gel electrophoresis. The pig kidney β-glucuronidase has a single pH optimum of 4.0–4.4 in acetate- and 5.4 in citrate-buffer; an activation energy of 16,800 cal/mol and a molecular weight of 275,000 were estimated. The KM for oestrone 3-glucuronide was 22.6 μM. The enzyme was not inhibited by N-ethylmaleimide nor by dithioerythritol, however, it was strongly inhibited by Hg2+. Oestradiol-17β 3-glucuronide and oestriol 3-glucuronide acted as competitive inhibitors, whereas oestradiol-17β 17β-glucuronide, oestriol 16α-glucuronide, testosterone 17-glucuronide and cholesteryl 3-glucuronide were uncompetitive, pregnanediol 3-glucuronide was noncompetitive, and Cortisol 21-glucuronide gave a mixed type inhibition. The synthetic β-d-glucuronides of phenolphthalein, p-nitrophenol, naphthol, 6-bromo-naphthol and methylumbelliferone all inhibited the hydrolysis of oestrone 3-glucuronide; the inhibition was of a more complex type than simple competitive inhibition.  相似文献

The inhibitory effects of a variety of estrogens on rat prostate testosterone Δ4–5α-reductase activity were measured by a specific in vitro assay. The conversion of 3H-testosterone (initial concentration 2.8 × 10?9 M) to labelled 5α-dihydrotestosterone and 5α-androstane-3α, 17β-diol was used as a measure of Δ4?5a-reductase activity. At a concentration of 1.8 × 10?6 M, estradiol was the most potent inhibitor (83.4%) of the estrogens tested. Various ester derivatives, e.g. 3-acetate, 3-phosphate, were effective inhibitors. The 17-glucuronide and 3-sulfate conjugates were less effective inhibitors. The estriol isomers exerted similar degrees of inhibition (40–60%). The 3-methoxy derivatives of estradiol and estriol were poor inhibitors. The introduction of certain groups into the steroid structure, e.g. 15α-hydroxy and 6-ketone, greatly decreased the inhibitory effect of estradiol. The nature of the oxygen function at carbon 17 did not greatly influence the inhibitory effects.  相似文献

The in vitro metabolism of [3H estradiol-17β-by the uterus was studied in non-pregnant, prenant (day 30-term) and post-parturant guinea pigs. Following incubation of tissue sections for one hour is Krebs-Ringer phosphate buffer, five major metabolites could be extracted from the medium or tissue depending upon age of gestation: estrone-3-glucuronide, estrone-3-sulfate, estradiol-3-glucuronide and estradiol-3-sulfate. Both sulfated estrogens were detected at each age of gestation studied, whereas the glucuronides, mainly of estrone, were not detected until approximately day 50. Thereafter, as term (day 65–70) was approached, their percentage contribution to total radioactivity increased at the expense of estradiol and the sulfates. Following parturition, total metabolites of estradiol rapidly decreased, particularly the glucuronides. No conjugates were detected in uteri from nonpregnant guinea pigs. In addition, no conjugates were found in the pre-partum mouse, rat and hamster or in human endometrium obtained immediately after birth. The data suggest that, in the guinea pig, a biochemical factor in the termination of normal pregnancy is the control of tissue levels of active estrogen (estradiol) by conjugation with glucuronic acid.  相似文献

The dietary bioavailability of the isoflavone genistein is decreased in older rats compared to young adults. Since flavonoids are metabolized extensively by the UDP-glucuronosyltransferases (UGTs), we hypothesized that UGT flavonoid conjugating activity changes with age. The effect of age on flavonoid glucuronidation was determined using hepatic microsomes from male F344 rats. Kinetic models of UGT activity toward the flavonol quercetin and the isoflavone genistein were established using pooled hepatic microsomal fractions of rats at different ages, and glucuronidation rates were determined using individual samples. Intrinsic clearance (Vmax/Km) values in 4-, 18- and 28-month-old rats were 0.100, 0.078 and 0.087 ml/min/mg for quercetin-7-O-glucuronide; 0.138, 0.133 and 0.088 for quercetin-3′-O-glucuronide; and 0.075, 0.077 and 0.057 for quercetin-4′-O-glucuronide, respectively. While there were no differences in formation rates of total quercetin glucuronides in individual samples, the production of the primary metabolite, quercetin-7-O-glucuronide, at 30 μM quercetin concentration was increased from 3.4 and 3.1 nmol/min/mg at 4 and 18 months to 3.8 nmol/min/mg at 28 months, while quercetin-3′-O-glucuronide formation at 28 months declined by a similar degree (P≤.05). At 30 and 300 μM quercetin concentration, the rate of quercetin-4′-O-glucuronide formation peaked at 18 months at 0.9 nmol/min/mg. Intrinsic clearance values of genistein 7-O-glucuronide increased with age, in contrast to quercetin glucuronidation. Thus, the capacity for flavonoid glucuronidation by rat liver microsomes is dependent on age, UGT isoenzymes and flavonoid structure.  相似文献

Causal relationship between sodium and hypertension has been proposed and various changes in Na+,K+-ATPase (sodium pump) activity have been described in established primary hypertension. A number of direct vascular effects of estradiol have been reported, including its impact on the regulation of sodium pump activity and vasomotor tone. The effects of estradiol involve the activation of multiple signaling cascades, including phosphatydil inositol-3 kinase (PI3K) and p42/44 mitogen-activated protein kinase (p42/44MAPK). In addition, some of the effects of estradiol have been linked to activity of cytosolic phospholipase A2 (cPLA2). One possible cardioprotective mechanism of estradiol involves of the interaction between estradiol and the rennin-angiotensin system (RAS). Elevated circulating and tissue levels of angiotensin II (Ang II) have been implicated in the development of hypertension and heart failure. The aim of our investigation was to elucidate the signaling mechanisms employed by estradiol and Ang II in mediating sodium pump, in vascular smooth muscle cells (VSMC). The aim of our investigation was to elucidate the signaling mechanisms employed by estradiol and Ang II in mediating sodium pump activity/expression in VSMC, with particular emphasis on PI3K/cPLA2/p42/44MAPK signaling pathways. Our primary hypothesis is that estradiol stimulates sodium pump activity/expression in VSMC via PI3K/cPLA2/p42/44MAPK dependent mechanism and, that impaired estradiol-stimulated sodium pump activity/expression in hypertensive rodent models (i.e. SHR), Ang II-mediated vascular impairment of estradiol is related to a decrease ability of estradiol to stimulate the PI3K/cPLA2/p42/44MAPK signaling pathways. An important corollary to this hypothesis is that in hypertensive state (i.e. SHR rats) the decreasing in ACE enzyme activity and/or AT1 receptor expression caused by administration of estradiol is accompanying with abrogated ability of Ang II to decrease IRS-1/PI3K association, and consequent PI3K/cPLA2/p42/44MAPK activity and associated sodium pump activity/expression.A clear characterization of how Ang II attenuates estradiol signaling may lead to a better understanding of the molecular mechanism(s) underlying pathophysiological conditions such as hypertension and to understanding how certain pathophysiological situations affect sodium pump activity/expression in VSMC.  相似文献

Two flavones, luteolin 7-O-β-glucuronide and diosmetin 7-O-β-glucuronide, were isolated and identified from Chrysanthemum morifolium L. v. Ramat leaves. Identification techniques included HPLC DAD, MS, 1H and 13C NMR spectroscopy. At concentrations of 0.2 and 2.0 mM, luteolin 7-O-β-glucuronide significantly reduced the frond number and chlorophyll content of Lemna gibba plants, but did not significantly affect dry weight. At a concentration of 0.2 mM diosmetin 7-O-β-glucuronide had no significant effect on frond number, dry weight or chlorophyll concentration of L. gibba. These results indicate that an ortho-3′,4′-dihydroxy arrangement of the B-flavonoid ring in the luteolin compound is probably responsible for allelopathic activity.  相似文献

Chromatographic profiles of the conjugates of 17α-ethynylestradiol (EE2) were obtained from the urine of castrate and normal women given tritium labeled EE2 orally. Five distinct radioactive peaks were observed, with considerable quantitative variation between individuals. Glucosiduronate fractions comprised the dominant excreted radioactivity. In vitro incubation of normal human liver also produced five conjugate types. Simultaneous intravenous/oral (14C/3H) administration of EE2 demonstrated conversion to identical urinary conjugate and free steroid products, with a greater excretion of the intravenous material. Authentic 3-glucuronide of EE2 was synthesized and its position in the chromatographic system relative to the urinary conjugate peaks demonstrated.  相似文献

Homogenates of kidney from laying Japanese quail incubated in vitro with 25-hydroxy-[26,27-3H] vitamin D3 produce more 1,25-dihydroxy-[26,27-3H]vitamin D3 than do homogenates of kidney from mature nonlaying females or males maintained on the same diet and under identical conditions. Instead, the homogenates from male quail or nonlaying female quail convert 25-hydroxyvitamin D3 to 24,25-dihydroxyvitamin D3. The administration of 5 mg of estradiol to mature male quail 24 h prior to sacrifice suppressed the 25-hydroxyvitamin D3-24-hydroxylase and markedly stimulated 25-hydroxyvitamin D3-1-hydroxylase. The administration of estradiol to male quail caused hypercalcemia, which responded more slowly than did the 1-hydroxylase. As little as 0.1 mg of estradiol/quail was found effective in stimulating the 1-hydroxylase and suppressing the 24-hydroxylase. Other hormones such as follicle stimulating hormone (FSH), cortisone, testosterone, and progesterone, even at high dose levels, produced little or no change in the 25-hydroxyvitamin D3-1-hydroxylase. Testosterone did, however, suppress the 25-hydroxyvitamin D3-24-hydroxylase. The stimulation of the 25-hydroxyvitamin D3-1-hydroxylase by parathyroid hormone was of a smaller magnitude than that of the estradiol, and the effects of the two hormones were additive, suggesting that they function by a different mechanism.  相似文献

1. The fate of sulphadimethoxine (2,4-dimethoxy-6-sulphanilamidopyrimidine) was studied in man, rhesus monkey, dog, rat, guinea pig and rabbit. 2. About 20–46% of the dose (0·1g./kg.) of the drug is excreted in the urine in 24hr. in these species, except the rat, in which only 13% is excreted. 3. In man and the monkey sulphadimethoxine N1-glucuronide is the major metabolite in the urine. In the rabbit and guinea pig N4-acetylsulphadimethoxine is the main metabolite. In the dog the drug is excreted mainly unchanged. In the rat equal amounts of the unchanged drug and its N4-acetyl derivative are the main products. 4. Small amounts of sulphadimethoxine N4-glucuronide are found in the urine of all the species. Sulphadimethoxine N1-glucuronide occurs in small amounts in the urine of rat, dog and guinea pig; none is found in rabbit urine. 5. Sulphadimethoxine N4-sulphate was synthesized and found to occur in small amounts in rat urine. 6. Monkey liver homogenates fortified with UDP-glucuronic acid are able to synthesize sulphadimethoxine N1-glucuronide with the drug as substrate. Rat liver has also this ability to a slight extent, but rabbit liver is unable to do so. 7. Sulphadimethoxine N4-glucuronide is formed spontaneously when the drug is added to human urine. 8. The biliary excretion of the drug and its metabolites was examined in rats. The drug is excreted in rat bile mainly as the N1-glucuronide. The N1- and N4-glucuronides administered as such are extensively excreted in the bile by rats.  相似文献

Elimination and metabolic profiles of the O-glucuronide conjugated products of pyrilamine and their nonconjugated O-dealkylated and N-desmethyl pyrilamine products were determined after the oral administration of (14C)-pyrilamine maleate to Fischer 344 rats, B6C3F1 mice and female rhesus monkeys by stomach tube or i.v. The total cumulative urinary and fecal pyrilamine products were determined. The conjugated pyrilamine metabolites, isolated and identified were the glucuronide products of O-dealkylated pyrilamine and ring-hydroxylated pyrilamine, and the nonconjugated metabolites were predominately the N-desmethylpyrilamine and O-dealkylated pyrilamine and their ring-hydroxylated products. Statistically significant differences were observed in the percentages of the conjugated and nonconjugated metabolites of pyrilamine excreted by the three species studied.  相似文献

The effect of estriol on the positive cooperativity of [3H]estradiol binding to the partially purified calf uterine estrogen receptor was investigated using the kinetic analysis of Sasson and Notides (J. biol. Chem. 257 1982, 11540). The receptor was titrated with variable concentrations of [3H]estradiol with or without estriol; the estriol was maintained in a constant molar ratio to the [3H]estradiol concentration. A 4-fold molar excess of estriol above the [3H]estradiol concentrations inhibited the receptor's cooperative [3H]estradiol binding. In the absence of estriol, the [3H]estradiol receptor interaction was highly cooperative, the Scatchard plot was convex and the Hill coefficient was 1.61 ± 0.02a. In the presence of sufficient estriol to reduce the maximally bound [3H]estradiol to 77%, the Scatchard plot was linear and the Hill coefficient was 1.04 ± 0.04. The inhibition of the cooperative [3H]estradiol binding by estriol was not due to isotope dilution of the specifically bound [3H]estradiol by the unlabeled estriol.These data demonstrate that the cooperative binding of 3H]estradiol by the receptor that is characteristic of the equilibrium between the two states of the receptor (active and nonactive) is eliminated by the presence of estriol. This finding is consistent with the agonist/antagonist activity of estriol observed in vivo.  相似文献

The growth of the SC-115 mammary carcinoma in mice is androgen dependent. Estrogens antagonize the androgen effect. The high affinity binding of androgens and estrogens has been studied in soluble extracts of the tumor, of primary culture cells and clone MI1 cells.Results indicate that two distinct specific steroid hormone-binding sites (termed ‘receptors’) are found in all cytosol fractions. The androgen-receptor (A) binds testosterone, androstanolone, cyproterone (an anti-androgen), progesterone and estradiol, but only very weakly non-steroidal diethylstilbestrol. The estrogen-receptor (E) binds estrogenic substances such as estradiol and diethylstilbestrol, but no androgen. The apparent KD, eq for A and E receptors of [3H]androstanolone and [3H]estradiol respectively, is identical (-0.5-1 nM at 4 °C). The affinity of estradiol for the A-receptor, when measured against [3H]androstanolone binding, indicates a Ki = 17.5 nM. The concentration of binding sites is of the order of 0.1 pmole/mg protein (somewhat higher for A than for E receptor) in MI1 cell cytosol. Studies by ultracentrifugation through glycerol-Tris gradients (low salt medium) reveal the macromolecular nature of the cytosol A and E receptors (7–7.5 S). Evidence is presented of the transfer of the A and the E receptors to nuclei after incubation of tumor slices as well as of clone MI1 cells with the corresponding hormones.Experiments suggest that the two different binding sites are present on two separated macromolecular moieties. After incubation at 37 °C of tumor slices with 10–20 nM [3H]testosterone, or with 10 nM [3H]estradiol, the corresponding radioactive hormone-receptor complexes are, as expected, found in the nuclear KCl extracts. In parallel experiments, where slices are incubated with non-radioactive hormones at the same concentration and the nuclear KCl extracts subsequently treated by radioactive steroids, no available androgen-binding sites are found in the nuclei after exposure to estradiol, nor estrogen-binding sites after exposure to testosterone.Therefore, in the same cell, two receptors are present which bind androgens and estrogens with high affinity, and one given hormone (estradiol) can be specifically bound (with different affinities) by two different receptors which, however, discriminate a synthetic analog (diethylstilbestrol). The data may give some molecular background for interpreting responses to the same hormone which may differ at various concentrations, for studying effects of analogs, and for analysing the control of tumor growth by antagonistic steroids.  相似文献

The biological effects of estriol (E3) have been studied in three estrogen targets, namely, the rat uterus in vivo and in vitro, in primary human endometrial cell cultures and in MCF-7 human breast cancer cells in culture. Studies on the temporal relationships between estrogen receptor binding and biological responses in the uterus using estriol and several more long-acting estriol derivatives, namely, 17α-ethynyl estriol, estriol-3-cyclopentyl ether, and 17α-ethynyl estriol-3-cyclopentyl ether, indicate that estriol is a short-acting compound with a brief duration of action. Estriol is a poor stimulator of uterine growth and plasminogen activator activity in vivo. Chemical modifications of the estriol molecule produce long-acting derivatives that result in a prolonged input of hormone receptor complexes into the nucleus and a prolonged and marked stimulation of uterine growth. In human endometrial cells in primary tissue culture, E3 has 12% the affinity of estradiol (E2) for cytosol estrogen receptor and it is quite effective yet slightly less potent than estradiol in stimulation of progesterone receptor synthesis. Low concentrations of E3(10−10 M) stimulate growth of MCF-7 cells in vitro and dose-response curves show E3 to be only slightly less effective than E2. In these endometrial and breast cancer cell systems in vitro, there is no metabolism of E3 while E2 is metabolized to estrone.Hence, estriol is an effective estrogen in vitro. In vivo, it is short-acting, but it can be made a full estrogen agonist when given at a sufficiently high concentration or in a chemically modified form which prolongs its activity by enabling effective concentrations of the compound to be maintained in the blood and in target tissues.  相似文献

The interaction of [3H]estriol with the partially purified estrogen receptor from calf uterus shows positive cooperativity that is dependent upon receptor concentration and temperature. At a receptor concentration of 1 nM and 25°C the [3H]estriol-receptor cooperativity was low, the Hill coefficient (nH) was 1.03 ± 0.02; however, with increasing receptor concentrations the receptor's cooperativity increased until at approximately intracellular receptor concentration (20 nM) the nH = 1.20 ± 0.04. At 0°C and a receptor concentration of 10 nM the [3H]estriol-receptor interaction was highly cooperative, the Scatchard plot was convex and nH = 1.58 ±0.04 while at 30°C the Scatchard plot approached linearity and nH = 1.03 ± 0.02. In comparison, [3H]estradiol was capable of inducing, at 0 or 30°C and at a receptor concentration of 1 nM or greater, maximal receptor cooperativity, nH = 1.63.These data demonstrate: (a) the receptor's conformation and binding mechanism change in a specific manner with temperature, so that receptor analysis at 0°C does not necessarily reflect the receptor's properties at biologically relevant temperatures; (b) the dependence of the receptor's cooperativity on receptor concentration, which suggests interaction between dissociable subunits; and (c) the lower cooperativity induced by estriol, in comparison with estradiol, which indicates estriol is less efficient in shifting the receptor toward a higher affinity or the activated state of the receptor.  相似文献

We have shown that the rate of release of PGF by monolayer cultures of epithelial cells from proliferative endometrium is markedly elevated by addition of estradiol to the medium. In cultures maintained in HAM F-10 medium containing charcoal stripped calf serum, estradiol (10−8M) increased the levels of PGF several fold during the second and third days in culture. Similar responses were obtained with estradiol at 10−10M concentration.When this system was used to compare the effects of estradiol and estriol at equal concentrations (10−8M), similar elevation (10–16-fold) of PGF levels were noted during 3 consecutive days in culture. When cultures of epithelial cells derived from secretory endometrium were used for these tests, estriol was as effective as than estradiol in elevating PGF levels in the medium.When the effects of estradiol and estriol were compared using fragments of secretory endometrium in organ culture, the increases in PGF levels noted in the medium were about equal (2- to 10-fold) for the two estrogens at the same concentration (10−9−10−8M).Exposure of the tissue to either estradiol or estriol for only 1 h resulted in increases in PGF output for the following 3 days.These results clearly show that estriol is as effective as estradiol in stimulating PGF output by human endometrial tissue.  相似文献

The nature of the water-soluble products formed by incubating labelled estradiol with uterine peroxidase in the presence of H2O2 and tyrosine was examined by two-dimensional thin-layer chromatography and high voltage electrophoresis. It was shown that the steroid and amino acid were associated in a 1:2 or 1:3 ratio and evidence was provided by 3H-exchange for the interaction of tyrosine with ring A of estradiol at C-2 and C-4. The possible role of estrogen-induced peroxidase in the uterus in vivo is discussed.  相似文献

Fumonisin B1 (FB1) is a Fusarium mycotoxin frequently occurring in corn in combination with deoxynivalenol (DON) and zearalenone. The aim of this study was to determine if FB1, alone and combined with DON or α-zearalenol (ZEA), zearalenone major active metabolite, can affect granulosa cell proliferation, steroid production, and gene expression in swine. Porcine granulosa cells were cultured for 2 days in serum-containing medium followed by 1 or 2 days in serum-free medium with or without added treatments. Fumonisin B1 had inhibitory effects on granulosa cell proliferation. Deoxynivalenol strongly inhibited cell growth, and no significant difference was detected in combination with FB1. α-Zearalenol showed a stimulatory effect on granulosa cell numbers even in combination with FB1. Regarding steroid production, FB1 increased progesterone production, and FB1 had no effect on estradiol production. Deoxynivalenol strongly inhibited progesterone and estradiol production, and FB1 had no significant effect on this response. α-Zearalenol increased progesterone production, and its combination with FB1 produced additive effects. α-Zearalenol had no effect on estradiol production, whereas it decreased estradiol production when co-treated with FB1. Fumonisin B1 was found to decrease CYP11A1 messenger RNA abundance, and the stimulatory effect of FB1 on progesterone production was found to be not dependent on 3-hydroxy-3-methylglutaryl-coenzyme A reductase activity suggesting that FB1 increases progesterone production through a different mechanism. The results show that these Fusarium mycotoxins can influence porcine granulosa cell proliferation and steroid production, thereby demonstrating their potential reproductive effects on swine.  相似文献

A rapid, highly sensitive method for the determination of morphine and its metabolites morphine-3-glucuronide (M3G), morphine-6-glucuronide (M6G) and normorphine has been developed using high-performance liquid chromatography–electrospray mass spectrometry, with the deuterated analogues as internal standards. The analytes were extracted automatically using end-capped C2 solid-phase extraction cartridges. Baseline separation of morphine, M3G and M6G was achieved on a LiChrospher 100 RP-18 end-capped analytical column (125×3 mm I.D., 5 μm particle size) with water–acetonitrile–tetrahydrofuran–formic acid (100:1:1:0.1, v/v) as the mobile phase. Morphine and normorphine coeluate and were separated mass spectrometrically. The mass spectrometer was operated in the selected-ion monitoring mode using m/z 272 for normorphine, m/z 286 for morphine, m/z 462 for morphine-6-glucuronide. Due to an interfering peak, M3G was measured by tandem mass spectrometry in the daughter-ion mode. The limits of quantitation achieved with this method were 1.3 pmol/ml for morphine, 1.5 pmol/ml for normorphine, 1.0 pmol/ml for M6G and 5.4 pmol/ml for M3G in serum or cerebrospinal fluid. The limits of quantitation achieved in urine were 10 pmol/ml for morphine, 20 pmol/ml for normorphine and M6G and 50 pmol/ml for M3G using a sample size of 100 μl. The method described was successfully applied to the determination of morphine and its metabolites in human serum, cerebrospinal fluid and urine in pharmacokinetic and drug interaction studies.  相似文献

The subcutaneous implantation of an estradiol pellet (10 mg) into female rats induced a hypophyseal hyperplasia with hyperprolactinaemia. Examination of neurotransmitter receptors in the hippocampus, striatum and cerebral cortex one month after the implantation revealed that estrogenization was associated with: an increased density of 3H-domperidone binding sites (D2 receptors) in the striatum and reduced numbers of 3H-serotonin high affinity sites (5-HT1 receptors) in the hippocampus and of 3H-muscimol binding sites (GABA receptors) in the hippocampus, striatum and cerebral cortex. In contrast, the characteristics of 3H-spiperone binding to 5-HT2 receptors (in the cerebral cortex) and those of 3H-flunitrazepam binding to benzodiazepine sites (in the three brain regions examined) were not significantly different in estrogenized and in control female rats. However, the enhancing effect of GABA on 3H-flunitrazepam binding was markedly reduced in brain membranes from estrogenized animals.
