The activity of four different sialyltransferases acting on N- or O-linked chains of glycoproteins was studied in brains of 19 days-old embryos, 1 day-old newborns and adult rats. By using asialofetuin, fetuin andN-acetyllactosamine as acceptors, it has been possible to measure independently the following enzyme activities: CMP-NeuAc:Gal1-3GalNac (2–3)-sialyltransferase (EC 2.4.99.4), CMP-NeuAc:Gal1-4GlcNAc (2–3)-sialyltransferase (EC 2.4.99.6), CMP-NeuAc:Gal1-4GlcNAc (2–6)-sialyltransferase (EC 2.4.99.1) and CMP-NeuAc:NeuAc2-3Gal1-3GalNac (2–6)-sialyltransferase (EC 2.4.99.7). The specific activity of the first three enzymes which act on asialylated acceptors showed a 2.6-fold decrease in a parallel manner after ontogenic development, while the activity of NeuAc2-3Gal1-3GalNac (2–6)-sialyltransferase was four times lower in adult than in embryonic brain, showing a stronger dependence on ontogenic development. Despite the higher level of sialyltransferases able to act on glycoproteins, in fetal brain these glycoproteins do not contain a higher amount of sialic acid.Abbreviations HPLC high performance liquid chromatography - N-CAM neural cell adhesion molecule  相似文献

The more common ones are Cryptococcus laurentii, Cryptococcus albidus, Sporobolomyces pararoseus and Sporobolomyces roseus.These yeasts stem from the phyllosphere of the surrounding vegetation, and constitute a population which is characteristic of the natural zones.

The seeds of 19 sunflower species were compared on the basis of their protein contents and the relative proportions of their protein fractions. The globulin content varied from 50% to about 70% and the albumin content from 18% to 35% according to the species. The level of intermediateMr polypeptides showed a great variability (9.6 to 24.3%). Comparative studies onMr polymorphism were carried out by means of sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE) of non reduced and/or reduced samples using both mono- and bidimensional procedures. Polypeptide constituents of helianthinin were compared including both number and molecular size (cultivatedH. annuus was used as a standard). Studies focused on differences observed between the major two (Mr 38 000), (Mr 32 000) and (Mr 25 500), (21 000) polypeptides families constituting the main A, B, and C subunits. and polypeptides analyses permit to discriminate easilyH. petiolaris from the other species. Charge polymorphism was studied using isoelectric focusing (IEF) and IEF-PAGE in mono and bidimensional procedures in the presence or absence of 2-mercaptoethanol (2-ME). Only a specific 4 polypeptide enables an easy discrimination betweenH. petiolaris and all the other species. Detailed nomenclature of the , and , polypeptides constituting the different helianthinin globulin subunits is given via the results of pI andMr analyses. Monodimensional IEF patterns of the more basic albumins (pI > 8.0) appear to provide a more valuable approach to identifying specific protein markers.  相似文献

Trichodesmium is a filamentous, colonial nitrogen fixing cyanobacteria, ubiquitous in tropical and subtropical regions of the world's oceans. Trichodesmium fixes atmospheric nitrogen and can comprise a significant fraction of total primary production in oceanic surface waters. Therefore, the consumption and fate of Trichodesmium has important consequences for understanding carbon and nitrogen cycling in the open ocean. The pelagic harpacticoid copepod Macrosetella gracilis uses Trichodesmium not only as a physical substrate for juvenile development, but also as a food source. Several different types of pelagic copepods (including several species of calanoids, harpacticoids and a poecilostomatoid species) were tested for ingestion of Trichodesmium by labelling the cyanobacteria with 14C. Only the pelagic harpacticoids ingested Trichodesmium. Here we report the first grazing rates based on 14C-uptake measurements for Macrosetella gracilis (0.173 µg C copepod–1 h–1), and the first quantitative measurements of both Miracia efferata (0.402 µg C copepod–1 h–1) and Oculosetella gracilis (0.126 µg C copepod–1 h–1) ingesting this cyanobacteria. Ingestion rates of M. gracilis and M. efferata on the two different species of Trichodesmium, T. thiebautii and T. erythraeum, as well as the two different colonial morphologies of T. thiebautii, spherical-shaped (puffs) and fusiform (tufts), were also compared. Both Miracia and Macrosetella had higher ingestion rates on the puff colonies than the tuft colonies of T. thiebautii.. Both also had higher ingestion rates of T. erythraeum than T. thiebautii. Trichodesmium thiebautii contains a previously reported neurotoxin which may be an important factor in determining trophodynamic interactions. Our results suggest that pelagic harpacticoid copepods can be quantitatively important in determining the fate of Trichodesmium carbon and nitrogen.  相似文献

A novel cap-independent translation has been performed where the ribosome entry is regulated by the 5-noncoding region (NCR) of L-A virus mRNA. Despite L-A virus mRNA containing neither cap structure nor a poly(A) tail, the reconstructed mRNA encoding the 5 NCR of L-A virus mRNA and a reporter gene (luciferase) was translated, in yeast lysate, 60 times more efficiently than control mRNA. The 5 NCR from L-A virus was effective in regulating the recruitment of ribosome in vitro. A possible mechanism in Saccharomyces cerevisiae is also suggested, whereby the ribosome entry is regulated by the 5 NCR of L-A virus mRNA.  相似文献

Summary The inheritance of resistance to whitebacked planthopper Sogatella furcifera (Horvath) was studied in 21 rice varieties. Reactions of F1; F2 and F3 progenies of the crosses of 21 resistant varieties with the susceptible variety TN 1 revealed that a single dominant gene governs resistance in Mushkan 41, Santhi, Siahnakidar 195, SM2-34, Tirisurkh 251, Zirijowaian 245, 18, 24A, 39, 76 S, 78, 180, 213 B, 267, 293, CI 6037-4, NP97, S39 JKW and Bansphul. In varieties 65 and 274 A, resistance is governed by one dominant and one recessive gene which segregate independently of each other. Tests for allelism with the Wbph 1 gene originally identified in N 22 revealed that the dominant gene present in all the test varieties is the same as Wbph 1. Further studies are required to determine the allelic relationships of the recessive gene found in varieties 65 and 274 A.  相似文献

Summary Individual callus cultures were initiated from 400 immature embryos of bacterial leaf spot-susceptible Sunhigh peach. Each was subjected to several selection cycles of a toxic culture filtrate produced by Xanthomonas campestris pv. pruni, the causal agent of leaf spot of peach. Progressively higher concentrations of the filtrate were used in each cycle. Two calli survived, and two plants were regenerated from each of the surviving calli. Each of the four clones was propagated in vitro and tested for whole plant resistance to X. c. pv. pruni. Results from bioassays on greenhouse-grown plants indicated that two out of the four selected clones were significantly more resistant to X. c. pv. pruni than the parental cv Sunhigh. In addition, one clone was significantly more resistant than the moderately resistant cv Redhaven.  相似文献

In this paper we analysed autotrophic sources of the carbon ( 13C) and the trophic position ( 15N) of Leporinus friderici in the influence area of Corumbá Reservoir, Brazil. We collected samples of muscles of fish from different sizes riparian vegetation, C4 grasses, zooplankton, periphyton and particulate organic carbon (POC). There were significant differences for the carbon isotope proportion found in muscles of L.friderici in the different size groups analysed. The highest values of 13C recorded for middle sized individuals is attributed to the large contribution of C4 plants in their diet. Small individuals sampled upstream also receive similar contribution from C4 plants. In contrast the same size group sampled downstream from the reservoir, has a much smaller of C4 plants. The 13C negative character of small individuals from downstream is due to the larger contribution of C3 plants (except periphyton). At larger sizes we found intermediate 13C values. The 15N proportions we found for each size group were not significantly different, however we found decreasing mean values with increasing size. The trophic level calculated from the dietary data was higher than that found with the 13C concentration in the muscle, except for small individuals, when the values were equal.  相似文献

The effects of different management practices (mainly mowing, grazing, nutrient enrichment) on floodplain vegetation were compared between the Austrian and Czech part of the Lunice River floodplain using nitrophilous vegetation with Urtica dioica. The vegetation samples situated on crosswise transects (40 in total) were used for the analysis of vegetation. Correlations between the floodplain width on these transects and the proportions (expressed as percentage of the total floodplain width) of selected characteristics (Urtica dioica and Phalaris arundinacea stands and managed areas) were not statistically significant. Differences in the proportions of selected characteristics between the two parts of the floodplain were not significant either, except differences in the proportion of managed areas and species richness (higher species richness and proportion of managed areas are in the Austrian part of the floodplain). The different environmental variables were used in ordinations (DCA and CCA) of vegetation samples. In the DCA, four groups of samples were interpreted. In the CCA changes the moisture gradient was the most important one. The transect distance (from the first transect in Austria) and the soil moisture had the closest relationship to the species data.  相似文献

Summary Genes for resistance toPuccinia graminis tritici andPuccinia recondita tritici identified in four South African wheats were:Sr6,Sr8a,Sr9e, andLr13 in W3762;Sr5,Sr8a,Sr9b,Sr12,Sr24,Lr13, andLr24 in W3760;Sr2,Sr24,SrC,Lr13, andLr24 in W3751; andSr7a,Sr23,Sr36, andLr16 in W3755. GenesSr2,Sr9e, andSr24 also conferred adult plant resistance to the predominant pathotypes ofP. graminis tritici. GenesSr7a,Sr23, andSrC, when present alone, did not confer acceptable adult plant resistance, even though low seedling reactions were associated with them when tested with the same pathotypes. We have used the lacZ reversion assay to study the mutation spectra induced by the Escherichia coli chromosomal umuDC operon and of its two plasmid-borne analogues impCAB and mucAB following exposure of cells to UV light and methyl methane-sulfonate (MMS). We have shown that the impCAB, mucAB and umuDC operons all produce a similar response to UV light which results almost exclusively in AT GC transitions. However, we found that the three operons produced different responses to alkylating agents. We found that with MMS the chromosomal umuDC operon produced almost exclusively AT GC transitions, whilst both mucAB and impCAB produced predominantly transversions. In the case of the impCAB operon the mutation spectrum contained more AT TA than GC TA transversions; this balance was reversed with mucAB. The effect of the copy number of the error-prone DNA repair operons upon the mutagenic spectra was also studied. The results obtained suggest that the copy number of the imp operon does not greatly affect the specificity of base substitutions observed. However, an increase in the copy number of the umuDC operon greatly affected the specificity of base substitution, such that virtually no transitions were produced and the spectrum was dominated by GC/AT TA transversions. It appears that the three error-prone DNA repair operons impCAB, mucAB and umuDC, despite showing strong structural and functional homologies, can display major differences in the spectrum of base changes induced during mutagenesis. We propose that the type of misincorporation/chain extension which DNA polymerase III is allowed to synthesize on a damaged DNA template is extremely sensitive to both the amount and type of error-prone repair proteins present. The modulation of these events by the different proteins can result in widely different mutagenic changes in the repaired DNA.  相似文献

A stratagem for the synthesis ofneoglycoproteins suitable for the selective serodiagnosis of leprosy is described in which synthetic 3,6-di-O-methyl--d-glucopyranose, the epitope of phenolic glycolipid I fromMycobacterium leprae, was used. Condensation of 8-methoxycarbonyloctanol with the acetobromo derivative of 3,6-di-O-methylglucose gave 8-methoxycarbonyloctyl 2,4-di-O-acetyl-3,6-di-O-methyl--d-glucopyranoside in 65% yield, and with absolute stereospecificity for the anomer. The deacylated product was converted to the crystalline hydrazide and coupled to bovine gamma globulin, bovine serum albumin and poly-d-lysinevia intermediate acyl azide formation to produce the 8-carbonyloctyl 3,6-di-O-methyl--d-glucopyranosyl polypeptides. Theneoglycoproteins were highly sensitive in ELISA and emulated the specificity of the native glycolipid in analysis of sera from patients throughout the spectrum of leprosy and from different geographical regions. The 8-carbonyloctyl 3,6-di-O-methyl--d-glucopyranoside-bovine serum albumin was also synthesized and shown to have about one-half the activity of the -linkedneoglycoprotein. A different synthetic approach produced the 8-carbonyloctyl 4-O-(3,6-di-O-methyl--d-glucopyranosyl)--l-rhamnopyranoside-bovine serum albumin which was also highly sensitive and specific for the serodiagnosis of leprosy. Except when cinnamic acids were provided in the culture medium, the major anthocyanin present in the two clones examined was 2. When the naturally occurring and some non-naturally occurring cinnamic acids were provided individually in the medium, 1 and 2 were minor components and the anthocyanin acylated with the supplied cinnamic acid, namely 3, 4, 5, 6, or 7 was the major anthocyanin present in the tissue. When caffeic acid was provided the major anthocyanin in the tissue was 4, thereby suggesting that the caffeic acid was methylated before its use in anthocyanin biosynthesis. Other cinnamic acids supplied had limited effects on the anthocyanins accumulated and appeared not to result in the accumulation of new anthocyanins by the tissue. Thus the tissue can use some but not all analogues of sinapic acid to acylate anthocyanins. Additional anthocyanins were detected in extracts of the wild carrot tissue cultures using mass spectrometry (both MS/MS and HPLC/MS). The additional compounds detected have also been found in cultures of black carrot, an Afghan cultivar of Daucus carota ssp. sativa and the flowers of wild carrot giving no evidence for qualitative differences in the anthocyanins synthesized by subspecies, cell cultures from subspecies, or clones from cell cultures. There are major differences in the amounts of individual anthocyanins found in cultures from different subspecies and in different clones from cell cultures. Here anthocyanins without acyl groups were usually found in the tissues and their accumulation is discussed. Summary Meiotic pairing in Triticum turgidum cv. Ma (4x) with a mean chiasmata frequency of 27.16 per cell was compared with chiasmata frequencies in its hybrids with several triticale strains, Chinese Spring wheat and its addition lines for Imperial rye chromosomes 4R and 6R. In hybrids between Ma and x Triticosecale cv. Rosner the chiasmata frequency was marginally reduced by an average of 1.25%, by 8.8% in hybrids with x Triticosecale cv. DRIRA HH and by 6.7% with DRIRA EE (lacking 90% telomeric heterochromatin from chromosome arm 7RL). In pentaploid hybrids between Ma and T. aestivum cv. Chinese Spring the reduction was an average of 10.30%, while addition lines with rye chromosome 6R reduced chiasmata frequencies by an average of 7.4% and rye addition line for 4R showed the greatest depression in chiasmata frequency in hybrids by a 25.04% reduction. Myrkdalen Lake in Western Norway was subjected to a 1.4 mdrawdown in June 1987. Plant establishment and vegetation succession on theexposed sediments of a fluvial delta plain was monitored through 2000. Theinvestigated area extended from the original Equisetumfluviatile zone to the new lake edge. The substrate was homogeneousand consisted mainly of minerogenous fluvial sediments. Vegetation data weresampled within continuous quadrats along transects perpendicular to the shore,and they dropped 93 cm in elevation. Detrended CorrespondenceAnalysis and Canonical Correspondence Analysis confirmed that time sincedrawdown and elevation of the quadrats appeared to be ofnearly equal importance in explaining succession. Plant establishment was rapidon the exposed sediments due to a seed bank and to rapid invasion of plants.Thesuccession includes both floristic change as a function of time and a spatialseparation in relation to the water level. The species succession was marked bya growth form progression: mosses and annuals non-clonal perennialsclonal perennials. After one month, the annual Subulariaaquatica and small acrocarpous mosses dominated the site. Dominancethen shifted to Deschampsia cespitosa, Juncus filiformis, Blasiapusilla and Polytrichum commune. Subsequentlythere was an increase of Carex vesicaria in the lower zoneand Calamagrostis purpurea in the upper zone,while Phalaris arundinacea was common over most of theelevational gradient. Equisetum declined where it haddominated before drawdown, but it expanded gradually towards the new shoreline.The vegetation remained dynamic after 13 years and it is not considered to beinequilibrium with the new environmental gradient yet. However, annual changesmeasured by DCA scores have slowed and two vegetation zones have developed. Themajor vegetational differences along the elevational gradient can be explainedby the height of the mean June water limit. This example of species turnover inspace and time may be a model for other successions that occur along a stronggradient.  相似文献

A field experiment was carried out at two sites off Yucatan State, Southeast Mexico, in order to determine the feasibility of culturing the red seaweedEucheuma isiforme; this was done during May–September 1989. At both sites (Uaymitun and Dzilam) the 25 days harvest and 14 algae per line plant density growth rates (2.21% day–1 and 1.21% day–1, respectively) were significantly higher (p<0.05) than those obtained with other combinations of the two factors tested (50, 75, 100 and 125 days harvest and 9 and 14 algae per line plant density). The mean carrageenan content of the cultured algae was 35.8% and 31.4% at Uaymitun and Dzilam, respectively.  相似文献

The complete definition of the chemical structure of GD1b-ganglioside (GD1b) lactone isolated from human brain has been given by means of spectrometric and spectroscopic analyses. GD1h lactone contains a single ester linkage involving the external sialic acid carboxyl group and the C-9 hydroxyl group of the internal sialic acid unit. We assessed the effect of four different virulence (vir) gene combinations on plant transformation efficiency and transgene behaviour in rice using the pGreen/pSoup dual binary vector system. Transformation experiments were conducted using a pGreen vector containing the bar and gusA expression units with, or without, the virG542, virGN54D, virGwt or the virG/B/C genes added to the backbone. Additonal vir gene(s) significantly altered plant transformation efficiency and the integration of vector backbone sequences. However, no differences in transgene copy number, percentage of expressing lines and expression levels could be detected. Addition of virGwt was the most beneficial, doubling the overall performance of the pGreen/pSoup vector system based on transformation frequency, absence of backbone sequence integration and expression of unselected transgenes. In 39 of the plant lines, the additional vir genes were integrated into the rice genome. The contribution of super dual binary pGreen/pSoup vectors to the development of efficient rice transformation systems and to the production of plants free of selectable marker genes are discussed.  相似文献

Summary In nondiapause adults raised under a long-day photoperiod, the critical daylength for diapause induction was between 13 and 14 h although some individuals did not respond to the short-day photoperiod and went on laying eggs. In postdiapause adults in which LD 1311 induced the first diapause (L13 insects), the critical daylength for diapause reinduction was between 13 and 14 h, whereas it was between 12 and 13 h in postdiapause adults in which LD 1014 induced the first diapause (L10 insects). Under LD 1311, a small proportion of L10 insects went into the second diapause after great delay as compared with L13 insects. Under LD 1014, on the other hand, L10 insects went into the second diapause more rapidly than L13 insects. Therefore, the photoperiod which had induced the first diapause affected the photoperiodic induction of the second diapause not only in the critical daylength but also in the speed of response. We evaluated the effects of ecosystem composition and structure (species richness and diversity, cover and spatial attributes of vegetation), abiotic factors (climate and topographical features) and the condition of the bare-ground areas (evaluated using soil-surface indicators) on the performance of Stipa tenacissima [evaluated using foliar 13C, 15N, nitrogen concentration and the carbon-to-nitrogen (C:N) ratio] in 15 steppes of SE Spain. Foliar 13C values of S. tenacissima showed a low degree of variation in the studied steppes, with average values ranging from –24.1 to –22.9. Higher variation was found in the 15N values, which ranged from –5.5 to –2.4. The nitrogen concentration and the C:N ratio varied between 5.0 and 8.0 mg g–1, and between 55.4 and 85.3, respectively. The 13C values became less negative with increasing spatial aggregation of perennial vegetation, while the C:N values increased with increasing perennial vegetation cover. The 15N values became more negative with increasing infiltration in the bare-ground areas, but the nitrogen concentration was not related to any of the environmental variables measured. Our results suggest that the relative importance of ecosystem structure and soil-surface conditions in the bare ground areas was higher than that of abiotic factors as determinants of the performance of S. tenacissima. The results also show that even subtle changes in these ecosystem features may lead to modifications in plant performance in semiarid S. tenacissima steppes, and thus to modifications in the associated ecosystem functions in the mid- to long-term.  相似文献
