Genetic differentiation among coastal and offshore common bottlenose dolphins,Tursiops truncatus,in the eastern North Pacific Ocean 下载免费PDF全文

Common bottlenose dolphins (Tursiops truncatus) are found worldwide in temperate and tropical regions, often occurring as distinct coastal and offshore ecotypes. Along the west coast of the United States, two stocks are recognized for management based on morphological and photo‐identification studies: a California coastal stock, estimated at 450–500 individuals, and a California/Oregon/Washington offshore stock of about 1,000 animals. This study is the first to analyze genetic differentiation between these stocks. We examined both the hypervariable portion of the mitochondrial DNA (mtDNA) control region and fifteen microsatellite markers for coastal (n = 64) and offshore (n = 69) dolphins. Significant genetic differentiation was found between the two stocks for mtDNA (ΦST = 0.30, P < 0.001; FST = 0.14, P < 0.001) and microsatellite loci (FST = 0.19, P < 0.001). Coastal dolphins had less genetic diversity than offshore dolphins. Further substructuring within the offshore stock was not detected. The level of genetic differentiation between the coastal and offshore dolphins is consistent with long‐term separation and reinforces recognizing them as separate stocks. These findings are particularly important for management of the smaller, less genetically diverse, coastal stock that is vulnerable to a variety of anthropogenic threats. 相似文献

Bones are frequently the only cetacean samples available in the wild and may be useful for determination of sex, physical maturity, body length, and body mass. We analyzed the postnatal morphological development and morphometric characteristics of the cleaned and dried, paired hip bone (os coxae) from 131 bottlenose dolphins, Tursiops truncatus, (61 females, 70 males; total body length 117–322 cm) ranging in age from neonates to 28 yr, salvaged from Croatian waters of the eastern Adriatic Sea. Adult dolphins exhibited sexual dimorphism in their hip bone morphology and morphometrics: fully‐formed hip bones from males were significantly longer, heavier, and more robust than those from females. Moreover, the fully formed hip bone from males featured a prominent bony tuberosity protruding dorsolaterally from the caudal half that was significantly wider and higher than in females. Morphometric measurements were used to generate classification functions for reliable sex determination (97%). In addition, hip bone length in both sexes correlated tightly with total body length, allowing one measurement to be calculated from the other. Only a single ossification developed in all hip bones although soft tissue attachments and comparative review suggests homological equivalence with the tripartite terrestrial mammal hip bone. 相似文献

Frozen sections stained with Oil-red-O and semithin (0.5 μm) plastic sections stained with toluidine blue revealed an abundance of fat globules of various sizes in all strata of the epidermis of bottlenose dolphins (Tursiops truncatus and T. gilli). The fat was rather evenly distributed but sometimes appeared as circumscribed areas of heavier concentration involving hundreds of cells (as seen in a single plane). Occasionally, there were smaller groups of epidermal cells heavily loaded with lipid. The dermis presented a unique phenomenon in the presence of abundant extracellular fat distributed among the collagen bundles as droplets of various sizes or as larger, irregularly shaped lipid particles that seemed to conform to the spaces between collagen bundles. These lipid particles were sometimes seen to be closely applied to the dermal surface of the stratum basale. Equally unusual was the presence of lipid particles of various sizes and shapes in the lumen of some of the vessels of the dermal papillae. Granular cells resembling mast cells were commonly seen in the papillary dermis and some were closely associated with lipid particles. Blood vessels of the reticular dermis tended to have collections of lipid droplets in the loose connective tissue often found adjacent to the tunica adventitia. It is postulated that the extracellular dermal lipids (probably mainly triglycerides) are broken down to free fatty acids that diffuse into the basal layer of the epidermis and are there resynthesized into triglycerides. Possible uses for the epidermal lipids are discussed. 相似文献

This study describes the age structure and sex-specific growth patterns of Lahille's bottlenose dolphins (Tursiops truncatus gephyreus), a subspecies endemic to the Southwest Atlantic Ocean (SWAO). The ages of 120 animals collected in southern Brazil between 1976 and 2017 were determined. We found high frequencies of young animals, mostly males, with no sex bias among adults. A temporal change in the age structure of strandings was observed, and we conclude it is a consequence of increased bycatch rates of young dolphins after 2000. The oldest male and female were 27 and 44 years old, respectively, suggesting that females live longer than males in southern Brazil. Growth curve analysis using Gompertz and Laird-Gompertz growth models estimated asymptotic lengths of 316.5 cm for females and 351.6 cm for males, (reached around 13 and 18 years old, respectively), supporting sexual size dimorphism for the subspecies. A second growth pulse was identified for males. Our work highlights the benefit of long-term studies and contributes valuable information toward understanding the life history of Lahille's bottlenose dolphin. It will serve as baseline for future studies that seek to understand mortality patterns of bottlenose dolphins elsewhere and the effects of anthropogenic actions on the survival of these animals. 相似文献

Environmental enrichment is often used to improve well-being and reduce stereotyped behaviors in animals under human care. However, the use of objects to enrich animal environments should not be considered to be effective until its success has been scientifically demonstrated. This study was conducted at Asterix Park in France in April 2009. The study investigated the use of 21 familiar objects with a group of six bottlenose dolphins (Tursiops truncatus). The dolphin trainers introduced four different objects into the dolphin pool every day on a rotating basis. Using a focal-object sampling method, we collected and analyzed data from twenty-one 15 min sessions. The results revealed a positive correlation between interest behaviors and interactive behaviors. Some dolphins had "favorite toys". However, only 50% of objects elicited manipulative behaviors. These findings demonstrate that dolphins do not treat all objects provided to them as "toys". Behavioral changes in the animals subsequent to the introduction of objects do not necessarily indicate an enrichment effect of the objects; rather, the motivation for the dolphins' behaviors toward the objects must be investigated. The animals' behavior must be considered in light of the social context and of the animals' individual behavioral profiles. The relevance of a constructivist approach to evaluating the effectiveness of enrichment programs is discussed. 相似文献

Knowledge about the ecology of bottlenose dolphins in the Southwestern Atlantic Ocean is scarce. Increased by-catch rates over the last decade in coastal waters of southern Brazil have raised concerns about the decline in abundance of local dolphin communities. Lack of relevant data, including information on population structure and connectivity, have hampered an assessment of the conservation status of bottlenose dolphin communities in this region. Here we combined analyses of 16 microsatellite loci and mitochondrial DNA (mtDNA) control region sequences to investigate genetic diversity, structure and connectivity in 124 biopsy samples collected over six communities of photographically identified coastal bottlenose dolphins in southern Brazil, Uruguay and central Argentina. Levels of nuclear genetic diversity were remarkably low (mean values of allelic diversity and heterozygosity across all loci were 3.6 and 0.21, respectively), a result that possibly reflects the small size of local dolphin communities. On a broad geographical scale, strong and significant genetic differentiation was found between bottlenose dolphins from southern Brazil–Uruguay (SB–U) and Bahía San Antonio (BSA), Argentina (AMOVA mtDNA ΦST = 0.43; nuclear FST = 0.46), with negligible contemporary gene flow detected based on Bayesian estimates. On a finer scale, moderate but significant differentiation (AMOVA mtDNA ΦST = 0.29; nuclear FST = 0.13) and asymmetric gene flow was detected between five neighbouring communities in SB–U. Based on the results we propose that BSA and SB–U represent two distinct evolutionarily significant units, and that communities from SB–U comprise five distinct Management Units (MUs). Under this scenario, conservation efforts should prioritize the areas in southern Brazil where dolphins from three MUs overlap in their home ranges and where by-catch rates are reportedly higher. 相似文献

Between 2007 and 2009, we witnessed three aggressive interactions between harbor porpoises and bottlenose dolphins in Monterey Bay, California. This is the first time such aggression has been documented in the Pacific, and the first time a harbor porpoise was collected immediately after witnessing its death, inflicted by bottlenose dolphins. Of the bottlenose dolphins present, 92% were males either confirmed (61%) or putative (31%). Since 2005, 44 harbor porpoise deaths inflicted by bottlenose dolphins were documented in California. Aberrant behavior was rejected as a cause of aggression, based on widespread documentation of similar behaviors in other populations of free‐ranging bottlenose dolphins. The evidence for interspecies territoriality as a form of competition for prey was weak: there is little dietary overlap and there are differences in bottlenose dolphin and harbor porpoise distribution patterns in California. Object‐oriented play was plausible as a form of practice to maintain intraspecific infanticidal skills or a form of play to maintain fighting skills between male associates. Contributing factors could be high‐testosterone levels, as attacks occurred at the height of the breeding season, and/or a skewed operational sex ratio. Ultimately, we need more information about bottlenose dolphin social structure at the time of the aggression. 相似文献

The melon is a lipid‐rich structure located in the forehead of odontocetes that functions to propagate echolocation sounds into the surrounding aquatic environment. To date, the melon's ability to guide and impedance match biosonar sounds to seawater has been attributed to its unique fatty acid composition. However, the melon is also acted upon by complex facial muscles derived from the m. maxillonasolabialis. The goal of this study was to investigate the gross morphology of the melon in bottlenose dolphins (Tursiops truncatus) and to describe how it is tendinously connected to these facial muscles. Standard gross dissection (N = 8 specimens) and serial sectioning (N = 3 specimens) techniques were used to describe the melon and to identify its connections to the surrounding muscles and blubber in three orthogonal body planes. The dolphin forehead was also thin‐sectioned in three body planes (N = 3 specimens), and polarized light was used to reveal the birefringent collagen fibers within and surrounding the melon. This study identified distinct regions of the melon that vary in shape and display locally specific muscle‐tendon morphologies. These regions include the bilaterally symmetric main body and cone and the asymmetric right and left caudal melon. This study is the first to identify that each caudal melon terminates in a lipid cup that envelopes the echolocation sound generators. Facial muscles of the melon have highly organized tendon populations that traverse the melon and insert into either the surrounding blubber, the connective tissue matrix of the nasal plug, or the connective tissue sheath surrounding the sound generators. The facial muscles and tendons also lie within multiple orthogonal body planes, which suggest that the melon is capable of complex shape change. The results of this study suggest that these muscles could function to change the frequency, beam width, and directionality of the emitted sound beam in bottlenose dolphins. The marine environment provides an opportunity to examine population structure in species with high dispersal capabilities and often no obvious barriers to genetic exchange. In coastal waters of the western North Atlantic, common bottlenose dolphins, Tursiops truncatus, are a highly mobile species with a continuous distribution from New York to Florida. We examine if the highly mobile nature coupled with no obvious geographic barriers to movement in this region result in a large panmictic population. Mitochondrial control region sequences and 18 microsatellite loci indicate dolphins are partitioning the habitat both latitudinally and longitudinally. A minimum of five genetically differentiated populations were identified among 404 samples collected in the range of New Jersey to northern Florida using both genetic marker types, some inhabiting nearshore coastal waters and others utilizing inshore estuarine waters. The genetic results reject the hypothesis of a single stock of coastal bottlenose dolphins put forth after the 1987–1988 epizootic that caused a large‐scale die‐off of dolphins and suggest instead the disease vector was transferred from one population to the next as a result of seasonal migratory movements of some populations. These coastal Atlantic populations also differ significantly from bottlenose dolphin samples collected in coastal waters of the northern Gulf of Mexico, implying a long‐term barrier to movement between the two basins. 相似文献

We developed polymerase chain reaction primers for 12 dinucleotide microsatellite loci in the bottlenose dolphin, Tursiops truncatus. Seven markers were obtained after hybridization screening, and five following random genome sequencing. Orthologous positions were computed for nine markers on the bovine genome and for seven on the human genome. The markers are distributed across chromosomes and found in different types of DNA regions. All 12 loci are polymorphic for Tursiops. Five loci were also polymorphic in the related species Stenella frontalis and the more distantly related river dolphin, Inia geoffrensis, indicating these markers will be informative across the Delphinidae and other cetacean taxa. 相似文献

Despite the openness of the oceanic environment, limited dispersal and tight social structure often induce genetic structuring
in marine organisms, even in large animals such as cetaceans. In the bottlenose dolphin, mitochondrial and nuclear DNA analyses
have revealed the existence of genetic differentiation between pelagic (or offshore) and coastal (or nearshore) ecotypes in
the western North Atlantic, as well as between coastal populations. Because previous studies concentrated on continental margins,
we analysed the population structure of bottlenose dolphins in two of the most isolated archipelagos of the North Atlantic:
the Azores and Madeira. We analysed 112 samples collected on live animals in the two archipelagos, and nine samples collected
on stranded animals in Madeira and mainland Portugal. Genetic analyses consisted in molecular sexing, sequencing of part of
the mitochondrial hyper-variable region, and screening of ten microsatellite loci. We predicted that: (1) there is at least
one pelagic and one or more coastal populations in each archipelago; (2) populations are differentiated between and possibly
within archipelagos. Contrary to these predictions, results indicated a lack of population structure in the study area. In
addition, comparison with published sequences revealed that the samples from the Azores and Madeira were not significantly
differentiated from samples of the pelagic population of the western North Atlantic. Thus, bottlenose dolphins occurring in
the pelagic waters of the North Atlantic belong to a large oceanic population, which should be regarded as a single conservation
unit. Unlike what is known for coastal populations, oceanic bottlenose dolphins are able to maintain high levels of gene flow. 相似文献

Some populations of coastal bottlenose dolphins ( Tursiops truncatus ) comprise discrete communities, defined by patterns of social association and long-term site fidelity. We tested the hypothesis that bottlenose dolphins in Tampa Bay, Florida, form a single community. The longitudinal study of dolphins in Sarasota Bay, adjacent to Tampa Bay, allowed us to ground-truth the definition of community and test whether our approach was robust to small sample sizes of resightings. We conducted photo-identification surveys in Tampa Bay during 1988–1993, and identified 102 dolphins with 10 or more sightings. We used hierarchical cluster analysis to examine the locations and association indices of these dolphins. We used analysis of variance (ANOVA) to test for differences in mean locations and determine whether mean coefficient of association (CoA) values within a community were higher than among communities. Dolphins in Tampa Bay clustered into five putative communities differing significantly in location and CoA values. Kernel estimates of the ranges of these five communities exhibited little overlap; some communities had no overlap at all. We conclude that five discrete communities of bottlenose dolphins exist in Tampa Bay and that such fine-scale structure may be a common feature of bottlenose dolphin populations throughout the southeastern United States. 相似文献

Bottlenose dolphins (Tursiops truncatus) are marine mammals with body water needs challenged by little access to fresh water and constant exposure to salt water.
Osmoregulation has been studied in marine mammals for a century. Research assessing the effects of ingested fresh water or
seawater in dolphins, however, has been limited to few animals and sampling times. Nine 16- to 25-h studies were conducted
on eight adult dolphins to assess the hourly impact of fresh water, seawater, and seawater with protein ingestion on plasma
and urine osmolality, urine flow rate (ufr), urinary and plasma solute concentrations, and solute clearance rates. Fresh water
ingestion increased ufr. Fresh water ingestion also decreased plasma and urine osmolality, sodium and chloride urine concentrations,
and solute excretion rates. Seawater ingestion resulted in increased ufr, sodium, chloride, and potassium urine concentrations,
sodium excretion rates, and urine osmolality. Seawater with protein ingestion was associated with increased ufr, plasma osmolality,
sodium excretion, and sodium, chloride, potassium, and urea urine concentrations. In conclusion, bottlenose dolphins appear
to maintain water and plasma solute balance after ingesting fresh water or seawater by altering urine osmolality and solute
clearance. Ingestion of protein with seawater appears to further push osmoregulation limits and urine solute concentrations

Marine mammals are routinely caught in the wild in an effort to monitor their health. However, capture-associated stress could potentially bias various biochemical parameters used to monitor the health of these wild caught animals. Therefore, the effects of capture were quantified by measuring plasma adrenal steroids and arginine vasopressin (AVP) in free-ranging bottlenose dolphins (Tursiops truncatus) (n=31). Total capture and restraint times were also correlated to hormone concentrations to quantify the effects of capture. Significant, positive correlations between corticosterone and cortisol (R=0.752; P<0.0001), and between corticosterone and aldosterone (R=0.441; P=0.045) were demonstrated. Significant correlations between capture and restraint time and hormone levels were not observed. Animals restrained for less than 20 min exhibited hormone levels similar to those for animals restrained for more than 20 min. The positive correlations among the adrenal steroids suggest that release of these steroids was stimulated by adrenocorticotropin (ACTH). The lack of a correlation between cortisol and AVP indicates that AVP did not influence ACTH-induced cortisol release in this situation. The study suggests that (1) a typical hypothalamic-pituitary-adrenal axis is present in these animals, and (2) the relatively short capture and restraint times did not induce a significant neuroendocrine stress response. 相似文献
