A newly isolated fungal strain used as whole‐cell biocatalyst for biodiesel production from palm oil
A strain of Aspergillus niger isolated from atmospherically exposed bread and Jatropha curcas seed was utilized as a whole‐cell biocatalyst for palm oil methanolysis to produce fatty acid methyl esters (FAME), or biodiesel. The A. niger strain had a lipase activity of 212.58 mU mL?1 after 144 h incubation at 25 °C with an initial pH value of 6.5, using 7% polypeptone (w/w on basal medium) as the nitrogen source and 3% olive oil (w/w on basal medium) as a carbon source. The A. niger cells spontaneously immobilized within polyurethane biomass support particles (BSPs) during submerged fermentation. Thereafter, the methanolysis of palm oil was achieved via a three‐step addition of methanol in the presence of BSPs‐immobilized with A. niger cells. The influence of water content, reaction temperature and enzyme concentration on reaction rate was investigated. An 8% water content and a temperature of 40 °C in the presence of 30 immobilized BSPs, resulted in an 87% FAME yield after 72 h. 相似文献

This work describes the continuous synthesis of ethyl esters via enzymatic catalysis on a packed‐bed continuous reactor, using mixtures of immobilized lipases (combi‐lipases) of Candida antarctica (CALB), Thermomyces lanuginosus (TLL), and Rhizomucor miehei (RML). The influence of the addition of glass beads to the reactor bed, evaluation of the use of different solvents, and flow rate on reaction conditions was studied. All experiments were conducted using the best combination of lipases according to the fatty acid composition of the waste oil (combi‐lipase composition: 40% of TLL, 35% of CALB, and 25% of RML) and soybean oil (combi‐lipase composition: 22.5% of TLL, 50% of CALB, and 27.5% of RML). The best general reaction conditions were found to be using tert‐butanol as solvent, and the flow rate of 0.08 mL min?1. The combi‐lipase reactors operating at steady state for over 30 days (720 h), kept conversion yields of ~50%, with average productivity of 1.94 gethyl esters h?1, regardless of the type of oil in use. © 2018 American Institute of Chemical Engineers Biotechnol. Biodiesel has gained widespread importance in recent years as an alternative, renewable liquid transportation fuel. It is derived from natural triglycerides in the presence of an alcohol and an alkali catalyst via a transesterification reaction. To date, transesterification based on the use of chemical catalysts has been predominant for biodiesel production at the industrial scale due to its high conversion efficiency at reasonable cost. Recently, biocatalytic transesterification has received considerable attention due to its favorable conversion rate and relatively simple downstream processing demands for the recovery of by-products and purification of biodiesel. Biocatalysis of the transesterification reaction using commercially purified lipase represents a major cost constraint. However, more cost-effective techniques based on the immobilization of both extracellular and intracellular lipases on support materials facilitate the reusability of the catalyst. Other variables, including the presence of alcohol, glycerol and the activity of water can profoundly affect lipase activity and stability during the reaction. This review evaluates the current status for lipase biocatalyst-mediated production of biodiesel, and identifies the key parameters affecting lipase activity and stability. Pioneer studies on reactor-based lipase conversion of triglycerides are presented. 相似文献

Enzymatic transesterification of waste cooking oil, comprising fats, oil and grease (FOG), to produce fatty acid methyl esters (FAME) i.e. biodiesel, was investigated using a novel strain of the fungus Aspergillus niger, immobilized as a whole‐cell biocatalyst. Response surface methodology (RSM), with a five‐level‐three‐factor central composite rotatable design, was used to optimize the reaction and analyze the relationship of reaction variables and their coinfluent on the response i.e. FAME yield. Independent variables that affect the transesterification reaction include temperature, feedstock water content and enzyme amount. Using RSM, a second‐order polynomial equation was derived for FAME yield using multiple regression analysis. The second‐order polynomial regression model was highly significant (P<0.001) in predicting the actual relationship between the response and the variables, where a linear relationship was apparent between observed and predicted values (R2=0.9651). In addition, the predicted determination coefficient q2 i.e. 0.7723, also proved that the model has a high predictive ability. The validation experiments, under optimized conditions, showed that the predicted value of maximum FAME yield (i.e. 91.3%) was in close agreement with the experimental value (i.e. 91.8%). 相似文献

An enzymatic process using a packed bed bioreactor with recirculation was developed for the scale-up synthesis of 2-ethylhexyl palmitate with a lipase from Candida sp. 99–125 immobilized on a fabric membrane by natural attachment to the membrane surface. Esterification was effectively performed by circulating the reaction mixture between a packed bed column and a substrate container. A maximum esterification yield of 98% was obtained. Adding molecular sieves and drying the immobilized lipase both decreased the water content at the reactor outlet and around the enzyme, which led to an increase in the rate of esterification. The long-term stability of the reactor was tested by continuing the reaction for 30 batches (over 300 h) with an average esterification yield of about 95%. This immobilized lipase bioreactor is scalable and is thus suitable for industrial production of 2-ethylhexyl palmitate. 相似文献

Acidolysis of butteroil with free conjugated linoleic acid (CLA) was studied in a packed bed reactor containing an immobilized Candida antarctica (fraction B) lipase. Kinetic data were used to develop quantitative reversible rate expressions of the general Michaelis-Menten form that also incorporate a term for first-order deactivation of the enzyme. The extent of incorporation of CLA in the triacylglycerols of butteroil was characterized for reactions carried out at several temperatures (namely 45 degrees, 50 degrees, and 55 degrees C) with different weight ratios of butteroil to CLA (namely 10:1 and 2:1). At the optimum operating temperature of 50 degrees C, similar levels of incorporation of CLA (60% to 85%) were achieved at low space times (<3 h) for both 10:1 and 2:1 (w/w) ratios of butteroil to CLA. 相似文献

A lipase from Aspergillus niger immobilized by adsorption on microporous, polypropylene hollow fibers was used to effect the continuous hydrolysis of the glycerides of butter oil at 40 degrees C and pH 7.0. The effluent concentrations of 10 different free fatty acid products were measured by highperformancee liquid chromatography (HPLC). Multiresponse nonlinear regression methods were used to fit the data to a multisubstrate rate expression derived from a Ping Pong Bi Bi mechanism in which the rate-controlling step is deacylation of the lipase. Thermal deactivation of the enzyme was also included in the mathematical model of reactor performance. A postulated normal distribution of v(max) with respect to the chain length of the fatty acid (with an additive correction for the degree of unsaturation) was tested for statistical significance. The model is useful for predicting the free fatty acid profile of the lipolyzed butteroil product over a wide range of flow rates. 相似文献

A recirculating packed bed reactor (RPBR) was used for efficient production of R-mandelic acid (R-MA) by kinetic resolution of racemic R,S-mandelonitrile (R,S-MN) using the recombinant E. coli cells crosslinked with diatomite (DA)/glutaraldehyde (GA)/polyethyleneimine (PEI). The performance and productivity of RPBR were evaluated by several parameters, including cell load, substrate feeding rate, height diameter (H/D) ratio, reactor structures, and operation stability. The kinetic resolution process showed higher initial reaction rate (1.52?mM/min) and yield (100%) by recycling 100?mL of substrate solution (70?mM) through RPBR packed with 6.0?g immobilized cells at a substrate-feeding rate of 19?mL/min while the H/D ratio was 2.8. The immobilized cells were successfully applied into kinetic resolution of R,S-MN in the RPBR for 50 batches with an average productivity of 4.12?g/L/h for R-MA with >99% of enantiomeric excess. 相似文献

黑曲霉表面展示南极假丝酵母脂肪酶B(CALB)可有效应用于食品、化妆品、医药等行业。以黑曲霉Aspergillus niger SH-1为宿主细胞构建诱导型糖化酶基因启动子表面展示CALB,在较高浓度葡萄糖碳源的发酵中CALB表达会受到抑制,发酵后期菌体容易出现菌丝断裂和展示酶活力下降等问题。采用组成型3-磷酸甘油醛脱氢酶基因启动子替代诱导型糖化酶基因启动子的细胞表面展示CALB黑曲霉菌株可有效解决上述问题,该菌株不但可以利用葡萄糖,而且还能利用木糖为发酵碳源,以木糖为碳源发酵在144 h展示酶水平达到1 100.28 U/g。文中探讨了甘蔗渣水解液发酵生产黑曲霉表面展示CALB,初步达到预期的结果,为甘蔗渣的综合利用提供了新途径。 相似文献

Aims: An integrated dual reactor system for continuous production of lactic acid by Lactobacillus delbrueckii using biofilms developed on reticulated polyurethane foam (PUF) is demonstrated.
Methods and Results: Lactobacillus delbrueckii was immobilized on PUF, packed in a bioreactor and used in lactic acid fermentation. The rate of lactic acid production was significantly high with a volumetric productivity of 5 g l−1 h−1 over extended period of time. When coupled to a bioreactor, the system could be operated as dual reactor for over 1000 h continuously without augmentation of inoculum and no compromise on productivity.
Conclusions: Polyurethane foams offer an excellent support for biofilm formation.

Xylanase production by Aspergillus niger NRRL‐567 in solid‐state fermentation (koji fermentation) was optimized using 24 factorial design and response surface methodology. The evaluated variables were the initial moisture level and concentration of inducers [veratryl alcohol (VA), copper sulphate (CS), and lactose (LAC)], leading to the response of xylanase production. Initial moisture level and LAC were found to be the most significant variable for xylanase production (p<0.05). The highest xylanase production was observed with 3578.8 ± 65.3 IU/gds (gram dry substrate) under optimal conditions using initial moisture of 85% (v/w), pH 5.0 and inducers VA (2 mM/kg), LAC 2% (w/w), and CS (1.5 mM/kg) after 48 h of incubation time. Higher xylanase activity of 3952 ± 78.3 IU/gds was attained during scale‐up of the process in solid‐state tray fermentation under optimum conditions after 72 h of incubation time. The present study demonstrates that A. niger NRRL‐567 can efficiently be used to achieve xylanase production with an economical and environmental benefit in solid‐state tray fermentation. The developed process can be used to develop an effective process for commercially feasible bioproduction of xylanases for speciality applications, such as conversion of lignocellulosic biomass to biofuels and other value‐added products. 相似文献

A lipase from Aspergillus niger immobilized by adsorption on microporous, polypropylene hollow fibers was used to effect the hydrolysis of the glycerides of melted butterfat at 40 degrees C and pH 7.0. Mcllvane buffer was pumped through the lumen and melted butterfat was pumped courrently through the shell side of a shell-and-tube reactor. Nonlinear regression methods were employed to determine the kinetic parameters of three nested rate expressions derived from a Ping Pong Bi Bi enzymatic mechanism coupled with three nested rate expressions for the thermal deactivation of the enzyme. For the reaction conditions used in this research, a four-parameter rate expression (which includes a two-parameter deactivation rate expression and a two-parameter hydrolysis rate expression) is sufficient to model the overall release of free fatty acids from the triglycerides of butterfat as a function of space time and time elapsed after immobilization. At a space time of 3.7 h immediately after immobilization of lipase, 50% of the fatty acid residues esterified in the sn-1,3 positions of the triglycerides can be released in the hollow-fiber reactor. 相似文献

AbstractPowdered biomass of Aspergillus sp. (RBD01) was used to carry out esterification of long chain fatty acids for biodiesel production. Dry biomass of Aspergillus sp. at 20% (w/w) with respect to the fatty acid was able to completely esterify oleic acid to ethyl ester at 35°C, in 36 h, with step wise addition of alcohol. Similar conditions also resulted in yield of 64.5% and 58% of ethyl ester from stearic acid and palmitic acid, respectively. However, the presence of methanol resulted in 87.5%, 71% and 41% of methyl ester from oleic, palmitic and stearic acid. Furthermore, it was found that the efficiency of biomass decreased by only 10% with its repeated use up to four cycles. 相似文献

A simultaneous synthesis of biodiesel, as fatty acid methyl esters, and monoacylglycerols catalysed by the recombinant Rhizopus oryzae lipase immobilized by adsorption on Relizyme OD/403M is presented. The use of this 1(3)-positional specific lipase prevents the formation of glycerol as a by-product, thus avoiding its drawbacks. The synthesis was carried out in a solvent-free system and it has been studied in two different reactor systems: stirred tank and packed-bed reactor. Stirred tank reactor presented a high-initial reaction rate and achieved a 33.6% yield, which corresponds to a value of 50.4% of the maximum yield that can be achieved with a 1(3)-positional specific lipase. In packed-bed reactor there was a smaller initial reaction rate, but it was achieved a 49.1% yield, which corresponds to a 73.6% of the maximum yield. When a second batch is performed, the yield decreased only 4% when packed-bed reactor is employed whereas a drastic decrease is observed in a stirred tank operation. Therefore, packed-bed reactor showed a best performance and minor damage to the biocatalyst. 相似文献

A lipase from A spergillus niger, immobilized by adsorption on a microporous, polypropylene flat-sheet membrane, was used to effect the continous hydrolysis of the glycerides of melted butterfat at 35°C. For the reaction conditions used in this research, a pseudo-zero order rate expression can be used to model the kinetics of the overall hydrolysis of butterfat. Multiresponse nonlinear regression methods were employed to determine the kinetic parameters of a multisubstrate rate expression derived fro ma mechanism based on the general Michaëlis–Menten approach. For the multiresponse data taken at pH 7.0, the dependence of the maximum rate of release of each fatty acid residue of butterfat on its carbon chain length is accurately described by a skewed, bell-shaped (or Γ-type) distribution. Data taken at five different pH values were fit assuming a Dixon–Webb diprotic model for the pH dependence of the reaction rate. The thermal deactivation of the immobilized lipase obeyed first-order kinetics with a half-life of 19.9 days at 35°C. The multisubstrate model is useful for the prediction of the free fatty acid profile of lipolyzed butterfat, whereas the lumped-substrate model provides an estimate of the overall degree of hydrolysis as a function of the reactor space time. 相似文献

The kinetics of enzymatic hydrolysis of rice bran oil in isooctane by immobilized Candida rugosa lipase in a batch reactor showed competitive inhibition by isooctane with a dissociation constant, K1, of 0.92 M.
