Effects of meridional migrations in shuttle from middle latitudes to regions beyond the Polar Circle, where an expedition and the organization of work shifts were being conducted, were studied. Three main types of systemic reactions of human hemodynamics and hemostasis were established. The first type was characterized by a moderate increase in the arterial blood pressure (BP) at the beginning of the shift with its subsequent normalization; by hypocoagulemia; and by an increase in the total phospholipid content, with a significant increase in the content of lysolecithins (an adaptive type). The second type was characterized by hypertension throughout the whole shift, by hypercoagulemia with significant changes in the phospholipid spectrum, and with a pronounced increase in the lysolecithin and cardiolipin fractions (a hypertensive type). The third type was characterized by a decrease in BP and an increase in heart rate (HR), by hypocoagulemia and an asthenic state associated with sleep disorders, and a decreased mental capacity (an asthenic type).  相似文献

BackgroundMeniscal tears, specifically lateral meniscal tears, have a larger than expected un-derdiagnosis rate in the presence of an ACL tear. The purpose of our study was to search for an MRI bone contusion pattern associated with MRI occult meniscal tears in patients with an ACL tear, specifically a contusion of the rim of the medial femoral condyle (RMFC). Our hypothesis was that there would be a significant association between RMFC contusions and MRI occult meniscal tears in patients with an ACL tear. We also searched for a difference between sexes with respect to the presence of the RMFC contusion in the setting of an occult meniscal tear. We also categorized the type, size, and location of these occult meniscal tears in the setting of an ACL tear.Methods This was a retrospective study that examined characteristics of occult meniscal tears and their association with a RMFC bone contusion. IRB approval was obtained. The date range of the study was June 2009 through December 2015. 6392 consecutive knee MRI reports in patients with an ACL deficient knee were reviewed. The study group included 22 patients with MRI occult meniscal tears, the control group included 110 patients. Relevant statistical values were calculated.ResultsThe most common type of occult meniscal tears were small radial and small longitudinal tears of the lateral meniscus. Occult meniscal tears were associated with an RMFC contusion in the study group (p=0.0457), particularly in males (p = 0.0003). In males with a torn ACL, the sensitivity of an RMFC contusion for an occult meniscal tear was 80%.ConclusionIn males with an ACL tear, there was a significant association between a contusion of the RMFC and an occult meniscal tear (commonly small radial or small peripheral partial-thickness longitudinal tears). Acetohydroxyacid isomeroreductase (AHAIR) is the shared second enzyme in the biosynthetic pathways leading to isoleucine and valine. AHAIR is encoded by the ilvC gene in bacteria. A 1,544-bp fragment of genomic DNA containing the ilvC gene was cloned from the cyanobacterium Synechocystis sp. strain PCC 6803, and the complete nucleotide sequence was determined. The identity of the gene was established by comparison of the nucleotide and derived peptide sequences with those of other ilvC genes. The highest degree of sequence similarity was found with the ilvC gene from Rhizobium meliloti. The isolated Synechocystis ilvC gene complemented an Escherichia coli ilvC mutant lacking AHAIR activity. The expressed Synechocystis gene encodes a protein that has a molecular mass of 35.7 kDa and that has AHAIR activity in an in vitro assay. Polyclonal antibodies raised against purified Synechocystis AHAIR produced a single band on a Western blot (immunoblot) of a Synechocystis cell extract and detected the protein in an extract of an E. coli ilvC mutant strain that was transformed with a plasmid containing the Synechocystis ilvC gene. The antibody did not react with an extract of an E. coli ilvC mutant strain that was transformed with a control plasmid lacking the Synechocystis ilvC gene or with an extract of an E. coli IlvC+ control strain.  相似文献

We studied odor learning in Leptopilina boulardi,a specialist larval parasitoid of Drosophila melanogaster.The behavioral responses of differently experienced females to an artificial odor (Must de Cartier, Paris) were analyzed using a fourarmed airflow olfactometer. The responses of females with an oviposition experience in the presence of the perfume were compared with those of four control groups. As controls we used naive females, females with an oviposition experience in the absence of odor, females which had been previously exposed to perfume but without an oviposition experience, and females with an oviposition experience which also had been exposed to perfume but not at the same time. The results demonstrate that a specialist such as L. boulardican learn very well to respond to an artificial odor by associating this odor with a reward, i.e., an oviposition. The four control groups responded more or less in a similar way.  相似文献

The chemotaxis receptor for aspartate, Tar, generates responses by regulating the activity of an associated histidine kinase, CheA. Tar is composed of an extracellular sensory domain connected by a transmembrane sequence to a cytoplasmic signaling domain. The cytoplasmic domain fused to a leucine zipper dimerization domain forms soluble active ternary complexes with CheA and an adapter protein, CheW. The kinetics of kinase activity within these complexes compared to CheA alone indicate approximately a 50% decrease in the KM for ATP and a 100-fold increase in the Vmax. A truncated CheA construct that lacks the phosphoaccepting H-domain and the CheY/CheB-binding domain forms an activated ternary complex that is similar to the one formed by the full-length CheA protein. The Vmax of H-domain phosphorylation by this complex is enhanced approximately 60-fold, the KM for ATP decreased to 50%, and the KM for H-domain decreased to 20% of the values obtained with the same CheA construct in the absence of receptor and CheW. The kinetic data support a mechanism of CheA regulation that involves perturbation of an equilibrium between an inactive form where the H-domain is loosely bound and an active form where the H-domain is tightly associated with the CheA active site and properly positioned for phosphotransfer. The data are consistent with an asymmetric mechanism of CheA activation [Levit, M., Liu, I., Surette, M. G., and Stock, J. B. (1996) J. Biol. Chem. 271, 32057-32063] wherein only one phosphoaccepting domain of CheA at a time can interact with an active center within a CheA dimer.  相似文献

The possibility of terminating cardiac arrhythmias with electric fields of moderate intensity is a challenging problem from a fundamental point of view and an important issue for clinical applications. In an effort to understand how anatomical re-entries are affected by electric fields, we found that a weak shock, with an amplitude of an order of magnitude less than the defibrillating shock, may unpin the vortices rotating around the defects (obstacles). The unpinning results from a depolarization of the tissue near the obstacle, induced by an external electric field within a distance of order lambda approximately 1 mm. Unpinning was observed both in the FitzHugh model of excitable tissue, and in a specific Beeler-Reuter model of cardiac tissue. This theoretical observation suggests that anatomical re-entries can be transformed into functional re-entries, an effect that can be tested in experiments with cardiac muscle.  相似文献

Oligodeoxynucleotides with an internal intercalating agent have been targeted to single-stranded sequences containing hairpin structures. The oligonucleotide binds to nonadjacent single-stranded sequences on both sides of the hairpin structure in such a way as to form a three-way junction. The acridine derivative is inserted at a position that allows it to interact with the three-way junction. The melting temperature (Tm) of complexes formed between the hairpin-containing target and oligonucleotides containing one internal acridine derivative was higher than that obtained with the same target and an unmodified oligonucleotide (DeltaTm = +13 degrees C). The internal acridine provided the oligonucleotide with a higher affinity than covalent attachment to the 5' end. Oligonucleotides could also be designed to recognize a hairpin-containing single-stranded nucleic acid by formation of Watson-Crick hydrogen bonds with a single-stranded part and Hoogsteen hydrogen bonds with the stem of the hairpin. An internal acridine derivative was introduced at the junction between the two domains, the double helix domain with Watson-Crick base pairs and the triple helix domain involving Hoogsteen base triplets in the major groove of the hairpin stem. Oligonucleotides with an internal acridine or an acridine at their 5' end have similar binding affinities for the stem-loop-containing target. The bis-modified oligonucleotide containing two acridines, one at the 5' end and one at an internal site, did not exhibit a higher affinity than the oligonucleotides with only one intercalating agent. The design of oligonucleotides with an internal intercalating agent might be of interest to control gene expression through recognition of secondary structures in single-stranded targets.  相似文献

One of the monoazo dyes reported by us in an earlier study (Lillie et at. 1972), amidol brown NAP, yielded an oxidation product, amidol black I, which was an excellent elastin stain. The present study revises the synthesis of amidol black I to a more convenient form giving much larger yields. Briefly, diazotized 4-nitro-2-aminophenol is azo-coupled into 2, 4-diaminophenol (amidol) and the product is oxidized to a quinoneimine with nitric. The product stains elastin black from an acid alcohol bath according to a Taenzer-Unna (1890) orcein type technic. The stain may be combined with a Van Gibson collagen stain or with an oil red O fat stain. The synthesis is presented in sufficient detail to permit its repetition in a hospital pathology laboratory.  相似文献

Sexually mature adults, embryos and larvae of the pterobranch Rhabdopleura normani from Bermuda were studied with light and electron microscopy. The sexes are separate among the zooids of a colony, but a given colony may contain females and males. In zooids of either sex the single gonad is associated with a large haemal sinus in the trunk sac and is displaced laterally (to the right or to the left). The wall of the gonad is composed of three layers: an outer metasomal peritoneum, an internal lining of germinal epithelium and an intervening genital haemal sinus. The mature gametes lie in the lumen within the gonad. The spermatozoon is characterized by an elongate nucleus, no obvious acrosome, a long mitochondrial filament in a midpiece appendix and a single flagellum with a 9+2 axoneme. Females brood 200 μm eggs and embryos in their distinctive, basally coiled tubes. The yolky eggs undergo radial cleavage and develop into ciliated, lecithotrophic, oblong larvae (400 μm in length) that are characterized by: (1) yellow coloration peppered with black pigment spots; (2) a deep ventral depression; (3) a posterior adhesive organ; (4) an anterior apical sensory organ; (5) an evenly ciliated epitdermis. The ventral depression is not invaginating endoderm, but is instead a glandular epithelium that evidently secretes the larval cocoon and the adult tube. Internally, the peritoneum of the coelomic cavities begins to split from the periphery of a large, central mass of yolky mesenchyme cells. The larva swims using cilia, but also undergoes contractions, evidently powered by the peritoneal cells, which constitute a myoepithelium. The discussion considers pterobranch affinities with other deuterostomes and with lophophorates.  相似文献

The flagellar apparatus of an undescribed species of Chrysochromulina Lackey that bears “eyelash” scales is reconstructed. The transitional region consists of two transitional plates each with an axosome, with no stellate pattern between them. Fine osmiophilic rings lie between the flagellar membrane and the outer doublets in the transitional region. The two jagella and the haptonema are inserted in a subapical depression that is lined ventrally by a spine-like projection formed by one of the parietal chloroplasts. The angles of insertion are similar to those of some other Chrysochromulina species in that both the haptonema and the right basal body lie at an extreme angle to the left basal body. The connectives of the apparatus consist of a striated distal band with a dorsal extension to the R1 and a ventral extension overlying the R2, a striated distal accessory band, an auxiliary connective from the right basal body to the adjacent ventral chloroplast, a well-developed intermediate band, two striated proximal bands, and a striated proximal accessory band. Of the microtubular roots in this Chrysochromulina species, three are associated with the left side of the cell (an R1 of 8+3; a small crystalline compound root, the R1C, associated with the R1; an R2 of three micro-tubules), and two are associated with the right basal body (an R3 of 2/2 microtubules with which the single-stranded R4 converges to form a 2/2+1 and then a 2/3 tiered arrangement). Comparisons are drawn with other species in the genus and related genera, particularly Prymne-sium.  相似文献

The spiracular structure characterized with a system of snarling dense outgrowths, has been studied in order to elucidate their reinterpreted role in respiration. The tracheal anatomy has undergone modifications to subserve a secondary function of reducing the metabolic heat in the thorax—a region of relatively high metabolic activity. Tracheation is so arranged as to enable this insect to lead successfully an amphibious existence. The rate of its oxygen consumption and its capacity to remain under water with an air bubble which functions as an efficient physical gill with temperature as an important limiting factor has been studied. Attempts have been made to analyse the anatomy of the respiratory system with reference to the insect's behaviour in its environment.  相似文献

Acetohydroxamic acid reacts with the enzyme-CoA form of succinyl-CoA:3-ketoacid coenzyme A transferase to give an inactive product with a rate constant of 860 M-1 min-1 at pH 8.1, 25 degrees C. The reaction is reversible in the presence of coenzyme A and has an equilibrium constant of 0.040. The product is an anhydride that is an analog of the intermediate that has been postulated in the normal catalytic pathway; it is inactive because coenzyme A does not react with the acyl group of the hydroxamic acid. The equilibrium constant for formation of the anhydride from the thil ester of enzyme and methyl 3-mercaptopropionate is 75 times larger than the equilibrium constant of 2.2 for the formation of N,O-diacetylhydroxylamine from acetohydroxamic acid and acetyl-CoA. This shows that the enzyme stabilizes the anhydride at the active site by at least -2.6 kcal mol-1. Succinomonohydroxamic acid reacts with enzyme-CoA as both a substrate and an inactivator, with relative rate constants of 25:1. The inactivation is irreversible, indicating that the enzyme provides a larger stabilization of at least -5.9 kcal mol-1 for the anhydride of an analog of the specific substrate, succinate. The results are consistent with the hypothesis that the enzyme stabilizes an anhydride that is formed at the active site during turnover of normal substrates through a stepwise reaction mechanism.  相似文献

We have carried out the third in a series of anatomical dissections of amputated congenitally deformed human limbs in an attempt to determine the etiological relationship between the bony and soft tissue anomalies. This specimen consisted of a limb with congenital tibial aplasia and an adducted foot with five toes. The arterial and nerve patterns were reminiscent of those seen previously by us. The arteries were characterized by absence of the anterior tibial artery and an incomplete plantar arch. The superficial peroneal nerve terminated at the ankle. Extra branches from the sural, deep peroneal, and medial plantar nerves supplied branches to the dorsum of the foot. There were two extra muscles in the leg. The remaining muscles were normal, with the exception that most muscles normally inserting on the plantar surface of the foot inserted instead into a common tendon sheet. The tibia was replaced by a tendinous band, and the talus and calcaneus were united by a complete synchondrosis. The contralateral limb clinically appeared to have a clubfoot. The combination of an absent anterior tibial artery and an incomplete plantar arch is consistent with our theory that a reduced number of vessels puts the embryonic limb at increased risk of congenital defects due to the reduction in the number of collateral blood routes. Some event, such as extravasation of blood or embolization, may concurrently or subsequently compromise blood flow in the remaining vessels. We have previously observed abnormal arterial patterns similar to that described above in limbs having absence of the tibia and in other patients having clubfeet. The presence of abnormal arterial patterns in a limb with absence of the tibia and a contralateral limb with clubfoot suggests that absent tibia and clubfoot may be etiologically related.  相似文献

The classical methods of mathematical physics are applied to construct an integral solution to the Chapman-Cowling-Davydov equation, which is derived from the kinetic Boltzmann equation with a collision term in the Lorentzian-gas approximation. For a particular initial distribution, the solution is obtained in an explicit form in terms of a Whittaker function. It is shown that, on long (macroscopic) time scales, the evolving distribution function with an arbitrary initial shape approaches a Maxwellian distribution. This result agrees with the accepted views regarding the overall temporal evolution of an arbitrary unsteady isolated system.  相似文献

Equilibrium sedimentation studies show that the serine acetyltransferase (SAT) of Escherichia coli is a hexamer. The results of velocity sedimentation and quasi-elastic light scattering experiments suggest that the identical subunits are loosely packed and/or arranged in an ellipsoidal fashion. Chemical cross-linking studies indicate that the fundamental unit of quaternary structure is a trimer. The likelihood, therefore, is that in solution SAT exists as an open arrangement of paired trimers. Crystals of SAT have 32 symmetry, consistent with such an arrangement, and the cell density function is that expected for a hexamer. Electron microscopy with negative staining provides further evidence that SAT has an ellipsoidal subunit organization, the dimensions of the particles consistent with the proposed paired trimeric subunit arrangement. A bead model analysis supports the view that SAT has a low packing density and, furthermore, indicates that the monomers may have an ellipsoidal shape. Such a view is in keeping with the ellipsoidal subunit shape of trimeric LpxA, an acyltransferase with which SAT shares contiguous repeats of a hexapeptide motif.  相似文献

DNA from healthy Malaysian newborns was studied on gene maps after digestion with different restriction endonucleases. Of 65 newborns, two were found to be carriers of two different variants of triplicated alpha-globin loci. In variant no. 1, found in an Malay, the three alpha-globin genes are in an elongated DNA fragment on digestion with Eco RI and Bam HI. The third alpha-globin gene was found in a additional 3.7-kb fragment on digestion with Hpa I, Bgl II and Hind III. In variant no. 2, a new type of triplicated alpha-globin loci, found in a Chinese, the three alpha-globin genes reside in an elongated DNA fragment longer than that of variant no. 1 on digestion with Eco RI and Bam HI. The third alpha-globin gene was found in an additional 4.2-kb fragment on digestion with Hpa I and Hind III. Digestion of this variant DNA with Bg1 II produced an abnormal 16.7-kb fragment in addition to the normal 7.0-kb Bgl-II fragment. The locations of the restriction sites in the two types of triplicated alpha-globin loci are compatible with a mechanism of unequal crossing over following two different modes of misalignment.  相似文献

The implantable cardioverter defibrillator (ICD) has significantly improved survival in patients with an increased risk of sudden cardiac death (SCD). The wearable cardioverter defibrillator (WCD) is an alternative to the ICD in patients with a transient ICD indication or those in whom an ICD temporarily cannot be implanted. We describe here the technical details of the WCD and report three patients who were treated with a WCD in an outpatient setting. The WCD allowed the cardiac condition of two patients to improve to such an extent that permanent ICD implantation was deemed unnecessary. This new form of therapy may result in significant cost reduction, avoidance of unnecessary ICD implantation, and increased patient satisfaction.  相似文献

The influence of an artificial leg length discrepancy (= ALLD) on stride times, pelvic rotations and activity of the intrinsic lumbar back muscles (= ILBM) was investigated for 20 subjects. An ALLD was created by shoes with a raised sole. Walking with an ALLD produced an increase of the swing phase time and a decrease of the stance phase time for both feet. The influence of an ALLD on pelvic rotations in the sagittal and frontal plane and on ILBM-activity was small. Changes in pelvic rotations in the sagittal plane were too small to observe. The mean pelvic rotation angle in the frontal plane was changed 1.52 degrees when walking with an ALLD of 40 mm (6.9 degrees while standing with an ALLD of 40 mm with extended knees). Only small changes were found in activity time due to an ALLD (not in EMG-amplitude). The activity time of the ILBM around heel strike of the raised limb was increased and unilaterally shifted from toe off in the direction of heel strike with the raised limb.  相似文献

The state of aggregation found in water-soluble preparations of an hepatic membrane protein responsible for the clearance of serum asialoglycoproteins has been shown to result from the self-associating properties of a single oligomeric protein. The smallest functional unit identifiable in aqueous solution possessed an estimated molecular weight of 500,000 with each of the successive components increasing in size by an equal amount to form an oligomeric series bearing an integral ratio of 1:2:3:4:5. The tendency towards self-association was promptly and completely reversed by the addition of Triton X-100 with the concomitant appearance of a single component. Extensive treatment with sodium dodecyl sulfate permitted the identification and isolation of two subunits with estimated molecular weights of 48,000 and 40,000, respectively. Amino acid and carbohydrate analyses revealed both subunits to be glycoproteins with a closely similar, but not identical, composition.  相似文献
