卵泡刺激素和表皮生长因子对小鼠精原细胞增殖的影响   总被引:2,自引:0,他引:2  
利用生殖细胞-体细胞体外无血清共培养模型研究了卵泡刺激素(FSH)和表皮生长因子(EGF)对小鼠A型精原细胞增殖的影响。精原细胞在ITS培养液(添加胰岛素、转铁蛋白和亚硒酸钠的DMEM)中培养24h后进行c-kit免疫细胞化学鉴定和EGF及其受体(EGFR)免疫细胞化学检测,72h后测定其形成集落数的情况。结果表明:ITS培养液能维持生殖细胞的活性,增殖细胞核抗原(PCNA)的表达增高。A型精原细胞呈c-kit阳性,EGF和EGFR主要表达于精原细胞。单独的FSH(1~100ng/ml)或EGF(1~10ng/ml)显著促进精原细胞集落数的增加。此外,EGF(0.1ng/ml)联合FSH(10ng/ml)具有加性效应,但更高剂量的EGF(1~10ng/ml)则降低了FSH的刺激作用。结果说明FSH可联合适量的EGF促进精原细胞的增殖。  相似文献

探讨不同波段电磁辐射对大鼠睾丸Sertoli细胞雄激素受体(androgen receptor,AR)和卵泡刺激素受体(follicle stimulating hormone receptor,FSHR)表达的影响。原代培养的Sertoli细胞分别经场强6×104 V/m的电磁脉冲(electromagnetic pulse,EMP)、平均功率密度为100 mW/cm2的S波段微波(S-band high power microwave,S-HPM)和X波段微波(X-bandhigh power microwave,X-HPM)辐射4 min。应用real-time RT-PCR和Western blot方法检测Sertoli细胞AR的表达,结果显示具有不同程度的降低(P<0.05,P<0.01);Wistar大鼠分别经S-HPM、X-HPM和EMP照射20 min,应用免疫组化方法检测睾丸组织AR的表达,发现辐射后7 d有显著降低(P<0.05,P<0.01)。三者比较,AR的表达总体呈EMP>X-HPM>S-HPM的趋势,而FSHR的表达无明显变化。Sertoli细胞AR表达的变化,可能参与了电磁辐射致...  相似文献

为评估联合子宫内膜厚度和类型对体外受精-胚胎移植(in vitro fertilization-embryo transfer,IVF-ET)临床结局的预测价值,回顾性分析1 933个IVF-ET周期.按照HCG日子宫内膜厚度分3组(组1:≤7 mm;组2:>7 mm到≤14 mm;组3:>14 mm),每组按内膜类型再分为3个亚组(组1(2、3)-A、B、C型),分析各组的妊娠率和着床率.结果显示HCG日内膜厚度是影响妊娠的独立因素;组1和组3的3个亚组妊娠率及着床率无差异;妊娠率和着床率在组2的A、B、C中依次降低,各组比较有显著差异(P<0.05).提示子宫内膜厚度和类型都是影响妊娠结局的独立因素;当内膜<7 mm或>14 mm时,联合内膜厚度和类型更好地预测临床结局;当内膜厚度适中(7~14 mm)且内膜类型有三线征时,将预示很好的临床结局.  相似文献

摘要 目的:探讨体外受精-胚胎移植(IVF-ET)女性患者焦虑状况的影响因素及对妊娠结局的影响。方法:选择2019年3月~2021年4月期间南京鼓楼医院生殖医学科收治的192例IVF-ET女性患者。采用焦虑自评量表(SAS)评估所有患者焦虑状况。收集患者的临床资料,单因素及多因素Logistic回归分析IVF-ET女性患者焦虑的影响因素。观察不同焦虑状况患者的妊娠结局。结果:192例IVF-ET女性患者中,有37例(19.27%)患者出现焦虑状况。其中轻度焦虑15例(40.54%)、中度焦虑10例(27.03%)、重度焦虑12例(32.43%)。单因素分析结果显示,IVF-ET女性患者是否焦虑与家庭月收入、辅助生殖技术治疗失败史、文化程度、职业、不孕年限、年龄、社会支持、不孕类型、不孕原因有关(P<0.05)。多因素Logistic回归分析结果显示:不孕类型为原发不孕、不孕原因为女方因素、年龄>34岁、家庭月收入<3000元、文化程度为高中或中专及以下是导致IVF-ET女性患者发生焦虑的危险因素(P<0.05)。无焦虑患者优胚例数、2PN受精例数、获卵例数、临床妊娠例数占比均高于焦虑患者(P<0.05)。结论:IVF-ET女性患者较易产生焦虑情绪,其焦虑状况受不孕类型、不孕原因、年龄、家庭月收入、文化程度影响,且焦虑状况还会影响患者的妊娠结局,临床工作中应对存在上述影响因素的患者进行积极心理疏导。  相似文献

给成年男子(n=5)口服酪氨酸300mg/日,共6天,第四天肌注 LRH-A 150μg/人,用RIA 法测定其血清 LH、FSH 和睾酮浓度。与对照组(n=5)相比,酪氨酸对 LH 和 FSH 的基础水平和 LRH-A 诱发的 LH 峰和 FSH 峰均无明显影响。睾酮的基础水平稍有升高,而酪氨酸对 LRH-A 引起的睾酮水平升高有明显的抑制作用。结果提示,酪氨酸可不通过垂体而直接作用于睾丸以影响睾酮的分泌。  相似文献

目的:探讨OCT4基因对卵泡刺激素作用下的永生化人卵巢上皮细胞株(Moody细胞)增殖、凋亡和侵袭能力影响。方法:将不同浓度的FSH(0、25、50、100mIU/ml)作用于Moody细胞48小时,应用Western-blot技术检测OCT4表达情况。采用慢病毒介导将重组质粒OCT4稳定转染至人卵巢上皮细胞株中,应用Western-blot法鉴定OCT4蛋白表达情况。FSH以50 mIU/ml作为工作浓度,实验对象分为4组:①siCon组,转染空载体的阴性对照组;②OCT4组:稳定转染OCT4基因的Moody细胞组;③FSH+siCon组:以FSH处理的siCon组;④FSH+OCT4组:以FSH处理的OCT4组。采用MTT比色法检测各组细胞的增殖情况,流式细胞仪检测各组细胞凋亡情况,Transwell侵袭实验检测各组细胞侵袭能力的变化。结果:(1)随着FSH浓度的增加,Moody细胞中OCT4蛋白表达逐渐增高,在FSH浓度为50 mIU/ml时达最高;(2)OCT4基因成功转染至Moody细胞中,经Western-blot检测该基因在细胞中进行蛋白高表达;(3)FSH+OCT4组细胞增殖活性明显增高,同时凋亡率降低,与另外三组相比差异具有统计学意义(P<0.05);(4)在FSH作用下,转染OCT4后明显增强了细胞的侵袭能力,与另外三组相比差异具有统计学意义(P<0.05)。结论:OCT4介导了FSH对人卵巢上皮细胞增殖、凋亡、侵袭活性的调控。  相似文献

目的:探讨体外受精/单精子胞浆内注射-胚胎移植(IVF/ICSI-ET)治疗前宫腔镜治疗对妊娠结局的影响.方法:回顾分析2006年1月-2007年12月在本中心接受IVF/ICSI-ET治疗的周期2988个.实验组为在治疗周期前B超或造影检查发现有宫腔病变,或曾有IVF/ICSI-ET失败史而行宫腔镜检查的周期;对照组为未行宫腔镜检查的周期.比较两组妊娠结局的差异.结果:实验组266个周期,第一次行IVF/ICSI-ET治疗前行宫腔镜检查和治疗后妊娠率34.0%(48/141);第二次IVF/ICSI-E治疗前行宫腔镜检查和治疗后妊娠率妊娠率61.7%(21/34);大于等于三次IVF/ICSI-E治疗前行宫腔镜检查和治疗后妊娠率57.6%(15/26);冻胚复苏周期前行宫腔镜检查和治疗后妊娠率53.8%(35/65).对照组2722例,第一次IVF/ICSI-ET治疗妊娠率43.2%(783/1811);第二次IVF/ICSI-ET治疗妊娠率34.7%(139/400);大于等于三次IVF/ICSI-ET治疗妊娠率28.3%(36/127);冻胚复苏周期治疗妊娠率35.2%(135/384).两组间妊娠率在IVF/ICSI-ET治疗大于等于二次时有统计学差异(P<0.05).结论:对于曾有IVF/ICSI-ET治疗失败的患者,拟再次治疗前行宫腔镜检查和治疗对妊娠结局有积极的作用.  相似文献

将牛的卵母细胞置于添加有不同预处理颗粒细胞及含有卵泡液的培养液或不舍有卵泡液的培养液中进行体外成熟、受精及胚胎发育培养,研究了颗粒细胞、卵泡液及颗粒细胞与卵泡液交互作用对牛卵母细胞成熟、受精后卵裂率、囊胚率的影响。2178枚卵母细胞体外成熟受精后胚胎发育的对比观察结果表明:颗粒细胞、卵泡液及颗粒细胞与卵泡液交互作用对卵母细胞成熟受精后胚胎的卵裂具有显著影响(P〈0.05);颗粒细胞对囊胚的发育有显著影响(P〈0.05);卵泡液及颗粒细胞与卵泡液交互作用对囊胚的发育无显著影响(P〉0.05)。不同因素对卵裂率、囊胚率的影响表现为:颗粒细胞因素〉培养液因素〉培养液&#215;颗粒细胞交互作用。结论:TCM199培养液中添加卵泡液和单层颗粒细胞组成的培养系统用于牛卵母细胞体外成熟及胚胎发育的效果较好。共培养体系中的单层颗粒细胞用经酶消化分散处理后在培养箱中孵育10min的颗粒细胞替代时,胚胎的发育效果并不受影响。  相似文献

摘要 目的:血清25-羟维生素D[25(OH)D]、抗苗勒管激素(AMH)和抑制素B(INHB)联合检测在卵巢储备功能低下(DOR)患者体外受精-胚胎移植(IVR-ET)妊娠结局中的应用价值。方法:选择2019年8月~2021年10月我院行IVR-ET助孕的DOR患者292例,根据助孕后的妊娠结局分为妊娠组(n=158)和未妊娠组(n=134),比较两组临床资料及血清25(OH)D、AMH和INHB水平,应用受试者工作特征(ROC)曲线评价血清25(OH)D、AMH和INHB单独及联合检测对妊娠失败的预测价值。应用单因素、多因素Logisitic回归分析妊娠失败的影响因素。结果:妊娠组血清25(OH)D、AMH、INHB水平显著高于未妊娠组(P<0.05)。ROC曲线分析显示,血清25(OH)D、AMH、INHB及三者联合检验对DOR患者IVR-ET妊娠失败的预测的曲线下面积(AUC)分别为0.714(0.631~0.802)、0.801(0.731~0.852)、0.634(0.578~0.741)、0.853(0.806~0.913),三者联合检验对DOR患者IVR-ET妊娠失败预测AUC高于单独检验。单因素分析显示未妊娠组窦卵泡个数<5、获卵个数<8、卵泡刺激素(FSH)/黄体生成激素(LH)≥1.8、血清25(OH)D≤25.05 μg/L、血清AMH≤1.08 ng/mL、血清INHB≤84.28 pg/mL比例显著高于妊娠组(P<0.05)。多因素Logistic回归分析显示FSH/LH≥1.8、血清25(OH)D≤25.05 μg/L、血清AMH≤1.08 ng/mL、血清INHB≤84.28 pg/mL是DOR患者IVR-ET妊娠失败的危险因素(P<0.05)。结论:血清25(OH)D、AMH和INHB 水平较低与DOR患者IVR-ET妊娠失败有关,且联合检测上述指标水平对患者IVR-ET妊娠失败有较高的辅助预测价值。  相似文献

目的:针对接受体外受精-胚胎移植(IVF-ET)的超重肥胖患者实施个体动态化体重管理,探讨该干预方法对体重控制及妊娠结局的影响。方法:选择2017年5月至2018年12月就诊于我院拟行IVF-ET的超重肥胖患者共99例纳入本研究,随机分为实验组50例与对照组49例,分别采取个体动态化体重管理及常规干预的方法对两组进行体重管理,记录并统计干预前后相关指标的组间差异。结果:两组干预前后BMI的差值差异有统计学意义(P0.05);内膜厚度、获卵数、可用胚胎数、移植胚胎数的组间比较差异无统计学意义(P0.05)。两组临床妊娠率与活产率差异有统计学意义(P0.05);出生婴儿体重组间差异无统计学意义(P0.05);干预满意度评分差异有统计学意义(P0.05)。结论:个体动态化体重管理的干预方法可有效减轻接受IVF-ET的超重肥胖患者的体重,降低体重指数,提高IVF-ET的移植成功率、活产率,值得临床推广应用。  相似文献

摘要 目的:探讨早卵泡期长效长方案联合重组黄体生成素(r-LH)对卵巢高反应人群排卵和妊娠结局的影响。方法:选入2019年1月~2023年3月在我院行早卵泡期长效长方案促排卵的卵巢高反应患者150例,根据是否添加r-LH分为对照组(未添加r-LH,n=75)和观察组(添加r-LH,n=75)。比较两组的促排卵相关指标和妊娠结局。结果:观察组Gn天数、Gn总量、HCG日血清LH水平、优质胚胎率均显著高于对照组,获卵数显著少于对照组(P<0.05)。两组降调天数、Gn启动量以及HCG日E2、P水平和子宫内膜厚度、临床妊娠率、活产率、流产率比较无差异(P>0.05)。结论:卵巢高反应患者行早卵泡期长效长方案促排卵时,添加r-LH可显著增加体内LH水平,提高优质卵泡率,对妊娠结局具有一定改善作用。  相似文献

Social and economical development is closely associated with technological innovation and a well-developed biotechnological industry. In the last few years, Brazil’s scientific production has been steadily increasing; however, the number of patents is lagging behind, with technological and translational research requiring governmental incentive and reinforcement. The Cell and Molecular Therapy Center (NUCEL) was created to develop activities in the translational research field, addressing concrete problems found in biomedical and veterinary areas and actively searching for solutions by employing a genetic engineering approach to generate cell lines over-expressing recombinant proteins to be transferred to local biotech companies, aiming at furthering the development of a national competence for local production of biopharmaceuticals of widespread use and of life-saving importance. To this end, mammalian cell engineering technologies were used to generate cell lines over-expressing several different recombinant proteins of biomedical and biotechnological interest, namely, recombinant human Amylin/IAPP for diabetes treatment, human FVIII and FIX clotting factors for hemophilia, human and bovine FSH for fertility and reproduction, and human bone repair proteins (BMPs). Expression of some of these proteins is also being sought with the baculovirus/insect cell system (BEVS) which, in many cases, is able to deliver high-yield production of recombinant proteins with biological activity comparable to that of mammalian systems, but in a much more cost-effective manner. Transfer of some of these recombinant products to local Biotech companies has been pursued by taking advantage of the São Paulo State Foundation (FAPESP) and Federal Government (FINEP, CNPq) incentives for joint Research Development and Innovation partnership projects.  相似文献

 The aim of this study was to elucidate the effect of intraperitoneal (i.p.) instillations of granulocyte-colony-stimulating factor (G-CSF) and/or interleukin-2 (IL-2) on ascites formation and the survival time of nude mice with malignant ascites, produced by i.p. inoculation of human ovarian cancer cells. When the nude mice were treated with medium alone, ascites was observed in all mice 28 days after tumor inoculation. When the mice were treated with cis-diamminedichloroplatinum(II) (cisplatin) alone, G-CSF alone or IL-2 alone, it took 35 days for the ascites to form in all mice. When cisplatin was combined with G-CSF or IL-2, one of ten mice did not form ascites during the observation period. Surprisingly, when G-CSF and IL-2 were simultaneously administered, ascites formation was not observed in any mice. Although i.p. treatment with cisplatin alone significantly prolonged the survival time, compared to medium alone, the lytic activity of spleen cells against HRA cells was significantly suppressed. When G-CSF or IL-2 was combined with cisplatin, the suppression by cisplatin was eliminated and subsequently resulted in a prolongation of the survival time. When G-CSF was combined with IL-2, both the peritoneal and splenic macrophages/monocytes were stimulated and the splenic lytic activity was about double that following treatment with G-CSF alone on IL-2 alone, suggesting that complete inhibition of ascites formation results not only from a significant increase of the peritoneal macrophages but also from enhancement of the lytic activity. Two mice, died from dissemination of tumor in the abdominal cavity, but eight mice survived without tumor for more than 90 days. As confirmed by monitoring body weight and hematocrit, G-CSF and IL-2 seemed to have no adverse effect. From these results, we conclude that a combination therapy with G-CSF and IL-2 might be of clinical use for inhibiting large amounts of ascites, which may inhibit therapeutic effects for ovarian cancer patients. 为建立重组汉逊酵母乙肝疫苗HPLC检定方法,应用TSK-G5000PW检测系统测定汉逊酵母重组乙肝疫苗表面抗原的纯度,对不同样品处理液的配比浓度和处理时间分别进行了探讨,作者选用DTT/Tween-80作为样品处理效果优于DTT+Tween-20,1:50Tween-80与0.1mol/L等量混合为样品处理液的适宜浓度。样品处理液与等量样品混匀时间介于35s-2min时,HPLC分离效果好,结果稳定。该处理液及处理时间对CHO细胞及Merck酿酒酵母重组乙肝疫苗表面抗原的HPLC纯度测定无影响。结果表明:现有的HPLC检测系统用0.1mol/L DTT与1:50 Tween-80等量混合处理后能有效地检测不同类型重组乙肝疫苗表面抗原的纯度。  相似文献

目的:研究促卵泡激素(FSH)结合片段对卵巢上皮性细胞癌细胞株hey增殖活性的影响。方法:应用卵巢上皮性细胞癌细胞株hey作为实验对象,分别加入FSH、FSH结合片段、FSH+FSH结合片段。用四甲基偶氮唑蓝(MTT)法检测卵巢癌hey细胞的生长状况,Western blot检测cyclinD1、Akt、pAkt分子的表达。结果:FSH对卵巢癌hey细胞的生长有明显的促进作用,细胞生长活性提高21%。FSH结合片段对卵巢癌细胞的生长有明显抑制作用,对细胞平均抑制率为22%,且能下调cyclinDI的表达,在2.5×10-9Mol/L达70%。FSH结合片段对FSH有竞争抑制作用,细胞抑制率为18%,能抑制FSH上调cyclinD1的作用,抑制率为61%。FSH能上调pAkt的表达,对Akt的表达没有明显影响,在40mIU/ml的浓度时pAkt/Akt上调率达224%。FSH结合片段对Akt的表达没有明显影响,但能明显下调pAkt的表达,且呈时间依赖性(在1 5分钟时达66%)和剂量依赖性(在2.5×10-9Mol/L达31%)。FSH结合片段能抑制FSH上调pAkt的作用,抑制率为80%。结论:FSH结合片段可通过抑制FSH诱导的PI3K/Akt-cycl-inD1信号通路进而抑制上皮性卵巢癌hey细胞的增殖活性。  相似文献

Chronic diseases pose a severe burden to modern National Health Systems. Individuals nowadays have a far more extended lifespan than in the past, but healthy living was only scantily extended. As much as longer life is desirable, it is saddened by chronic diseases and organ malfunctions. One contributor to these problems was recognized to be represented by microparticles (MPs). Our purpose is to better understand MPs, to contrast their ominous threat and possible clinical importance. For this intent we correlated MPs with thrombotic pathologies, hemophilia, malaria, diabetes, cardiovascular diseases, endothelial dysfunctions, pulmonary hypertension, ischemic stroke, pre-eclampsia, rheumatologic diseases-rheumatoid arthritis, polymyositis-dermatomyositis, angiogenesis and tumor progression-cancer; we listed the possibilities of using them to improve transfusion methods, as a marker for acute allograft rejection, in stem cell transplantation, as neuronal biomarkers, to understand gender-specific susceptibility for diseases and to improve vaccination methods and we presented some methods for the detection of MPs.  相似文献

Dipeptidyl peptidase (DPP) IV inhibitors provide a new strategy for the treatment of type 2 diabetes. Human DPP-IV gene was cloned from differentiated Caco-2 cells and expressed in Pichia pastoris. The recombinant enzyme was used in a new system for screening of DPP-IV inhibitors. By high throughput screening, a novel compound (W5188) was identified from 75,000 compounds with an IC50 of 6.5 μM. This method is highly reproducible and reliable for discovery of DPP-IV inhibitors as shown by Z′ value of 0.73 and S/N ratio of 6.89.  相似文献

光生物调节作用(photobiomodulation,PBM)是激光或单色光(laser irradiation or monochromatic light,LI)对生物系统的非损伤调节作用,包括低强度LI(low intensity LI,LIL)的PBM(uL PBM,LPBM)和中等强度LI(moderate intensity LI,MIL)的PBM(MIL PBM,MPBM).功能内稳态(function-specific homeostasis,FSH)是生物系统维持某种功能稳定发挥的负反馈状态.从FSH的角度可以将PBM分为内稳态PBM(FSH PBM,fPBM)和发展PBM(developmental PBM,dPBM).fPBM只能调节生物系统的功能,但对处于FSH的生物系统无效.dPBM可以打破FSH.LPBM属于fPBM.MPBM由内源性光敏剂所产生的活性氧(reactive oxygen species,ROS)介导,ROS低者属于fPBM,ROS高者属于dPBM.低水平u治疗(low level LI therapy,LLLT)是fPBM的临床应用.只要LI的参数选择合理,LLLT可以实现细胞康复作用.  相似文献

Basal serum levels of follicle stimulating hormone (FSH), luteinizing hormone (LH), and testosterone (T) and the responsiveness of these hormones to a challenge dose of luteinizing hormone releasing hormone (LHRH), were determined in juvenile, pubertal, and adult rhesus monkeys. The monkey gonadotrophins were analyzed using RIA reagents supplied by the World Health Organization (WHO) Special Programme of Human Reproduction. The FSH levels which were near the assay sensitivity in immature monkeys (2.4 +/- 0.8 ng/ml) showed a discernible increase in pubertal animals (6.4 +/- 1.8 ng/ml). Compared to other two age groups, the serum FSH concentration was markedly higher (16.1 +/- 1.8 ng/ml) in adults. Serum LH levels were below the detectable limits of the assay in juvenile monkeys but rose to 16.2 +/- 3.1 ng/ml in pubertal animals. When compared to pubertal animals, a two-fold increase in LH levels paralleled changes in serum LH during the three developmental stages. Response of serum gonadotrophins and T levels to a challenge dose of LHRH (2.5 micrograms; i.v.) was variable in the different age groups. The present data suggest: an asynchronous rise of FSH and LH during the pubertal period and a temporal correlation between the testicular size and FSH concentrations; the challenge dose of LHRH, which induces a significant rise in serum LH and T levels, fails to elicit an FSH response in all the three age groups; and the pubertal as compared to adult monkeys release significantly larger quantities of LH in response to exogenous LHRH.  相似文献
