目的实现对致病性大肠埃希菌(E.coli)、沙门菌(Salmonella)的同时检测,建立快速灵敏的双重PCR检测方法。方法以致病性大肠埃希菌和沙门菌毒力岛基因为研究对象,根据GenBank发表的大肠埃希菌和沙门菌毒力岛基因序列,分别设计合成了大肠埃希菌毒力岛irpl、irl)2和fyuA,沙门菌毒力岛mgtC、sseL和sopB等6对引物,以禽致病性大肠埃希菌(CVCC1565)菌株和沙门菌(ATCC9150)菌株的核酸混合物为模板,经引物特异性试验,引物组合,成功建立了快速鉴别检测致病性大肠埃希菌和沙门菌的双重PCR方法。结果特异性试验结果显示,引物irpl、irp2和fyuA仅能扩增出大肠埃希菌(CVCC1565)的特异性片段,大小分别是799、414和948bp;引物mgtC、sseL和sopB仅能扩增出沙门菌(ATCC9150)的特异性片段,大小分别是500、269和1000bp。敏感性试验结果表明大肠埃希菌和沙门菌的最低检测限分别为2.2×101CFU/mL和2.0×101CFU/mL。结论本研究建立的双重PCR方法具有特异性强、敏感性高、快速简便等特点,可用于致病性大肠埃希菌和沙门菌的联合检测与鉴别诊断。  相似文献

为了解上海地区社区获得性腹泻儿童致病菌流行病学情况,对2007年度就诊于我院的2871例社区获得性腹泻患儿进行粪便弯曲菌、志贺菌、沙门菌,霍乱弧菌、副溶血性弧菌、致腹泻大肠埃希菌、气单胞菌、耶尔森菌进行培养分离、鉴定及部分菌株的血清分型。结果发现,弯曲菌在腹泻患儿粪便中的分离率为11.1%(172/1556),4月份感染率最高(P<0.001),多见于1~4岁儿童(P<0.05);其次是志贺菌,分离率为3.2%(91/2871),其中福氏志贺菌占63.7%;第3位是致腹泻大肠埃希菌,分离率为1.0%(30/2871),其中肠致病性大肠埃希菌占70.0%,以O55/K59血清型为主。沙门菌、气单胞菌分离率不超过0.5%,未检测到霍乱弧菌、副溶血性弧菌和耶尔森菌。本次调查显示,弯曲菌、志贺菌、致腹泻大肠埃希菌是目前上海地区儿童社区获得性腹泻的3种主要致病菌。弯曲菌感染率明显高于志贺菌及致腹泻大肠埃希菌,提示临床微生物室应将弯曲菌、致腹泻大肠埃希菌作为儿童社区获得性腹泻常规监测的病原微生物。  相似文献

目的:建立妇科凝胶的微生物限度检查方法。方法:按2010?年版《中国药典(二部)》相关要求进行试验,以枯草芽孢杆菌、金黄色葡萄球菌、大肠埃希氏菌、白色念珠菌、黑曲霉为试验菌,铜绿假单胞菌和金黄色葡萄球菌为控制菌对样品进行微生物限度检查的方法学验证。结果:细菌、霉菌和酵母菌计数方法验证试验中,试验菌的回收率均大于70%;控制菌检查结果符合《药典》相关要求。结论:该方法准确可靠,适用于妇科凝胶类微生物限度的检测。  相似文献

目的 研究凝结芽胞杆菌TBC 169株对大肠埃希菌、痢疾志贺菌、伤寒沙门菌、普通变形杆菌、铜绿假单胞菌和金黄色葡萄球菌的抑制作用。方法 先将大肠埃希菌、痢疾杆菌等6种菌分别进行单独培养,测定不同培养时间内的pH和活菌数,然后将凝结芽胞杆菌TBC 169株分别和致病菌进行混合培养,再测pH和活菌数,并与单独培养时的测定情况进行比较。结果 凝结芽胞杆菌TBC 169株对大肠埃希菌、痢疾志贺菌、伤寒沙门菌等6种菌均有明显的抑制作用,尤其是对伤寒沙门菌和铜绿假单胞菌的抑制作用更强。结论 凝结芽胞杆菌TBC 169株对肠道致病菌有显著的抑制作用。  相似文献

目的:建立花蛇解痒胶囊微生物限度检查方法。方法:采用培养基稀释法和常规法。结果:采用常规法验证,大肠埃希菌、金黄色葡萄球菌、白色念珠菌、黑曲霉回收率均能达到70%以上,采用培养基稀释法验证枯草芽孢杆菌回收率能达到70%以上,控制菌可检出大肠埃希菌、大肠菌群、沙门菌。结论:花蛇解痒胶囊微生物限度检查,可采用培养基稀释法检查细菌数、常规法检查霉菌和酵母菌数;控制菌采用常规法和稀释法。  相似文献

目的检测通过基因工程获得的片球菌素Pediocin PA-1抑菌活性。方法采用琼脂扩散法检测片球菌素Pediocin PA-1对单核细胞增生李斯特杆菌、金黄色葡萄球菌、铜绿假单胞菌、沙门菌和大肠埃希菌O157的抑菌活性。结果片球菌素Pediocin PA-1对单核细胞增生李斯特杆菌、金黄色葡萄球菌、沙门菌、铜绿假单胞菌和大肠埃希菌O157等均有抑制作用。其中对单核细胞增生李斯特杆菌、沙门菌、大肠埃希菌和金黄色葡萄球菌的抑制作用效果明显,对铜绿假单胞菌有微弱的抑制作用。结论通过基因工程获得的片球菌素Pediocin PA-1具有抑菌活性。  相似文献

目的对花红胶囊微生物限度检查方法进行方法学验证。方法按《中国药典》(2010版)一部附录ⅧC项下的常规方法检查,采用平皿计数法,分组分别使用5种试验菌验证。结果:花红胶囊中5种试验菌的回收率均达70%以上。控制菌大肠埃希菌、大肠菌群、沙门菌经方法学验证可行性强。结论采用常规检查法对花红胶囊微生物限度检查,可控制该产品的细菌、霉菌和酵母菌的总数和控制菌。  相似文献

目的:建立一种能够快速、准确地检测流感病毒亚单位疫苗中间体样品中沙门菌污染的方法。方法:首先利用选择性增菌培养基对样品进行增菌培养,然后提取样品中的细菌基因组DNA,通过沙门茵特异性引物对基因组DNA进行PCR扩增,以琼脂糖凝胶电泳实现对目的片段的检测。结果:该PCR反应体系的扩增检测灵敏度可达1pg的沙门菌DNA,利用选择性增菌培养配合该PCR体系可在最快24h内实现对沙门菌的准确检测,测定结果与传统方法相符。结论:此方法应用于流感病毒亚单位疫苗中间体的沙门菌检查,较之传统的培养法结合生化鉴定的方法,大大缩短了检测周期,降低了结果判读的难度,在实际生产中有很好的应用前景。  相似文献

目的研究伤寒沙门菌耐药质粒pRST98在小鼠体内向大肠埃希菌的接合转移,比较质粒在体内、外接合转移的异同。方法由于伤寒沙门菌是一种只对人类致病的病原菌,因此将伤寒沙门菌的耐药质粒pRST98导入遗传背景明确的鼠伤寒沙门菌低毒株RIA中,用接合子pRST98/RIA口饲BALB/c小鼠进行体内接合转移。结果在体外伤寒沙门菌很容易将pRST98转移给大肠埃希菌E.coliK12W1485(F-)RifrLac+,该接合子又可将pRST98转移给鼠伤寒沙门菌RIA,但在不同宿主菌中耐药标志的表达有差异。未经人工感染小鼠肠道分离的大肠埃希菌耐药情况严重,口饲pRST98/RIA后出现了部分耐药标志与pRST98耐药谱相同的大肠埃希菌,但有些抗生素的耐药标记未能表达。质粒检测显示体内形成的接合子均含耐药质粒pRST98。结论伤寒沙门菌耐药质粒pRST98在动物体内、外均可转移给大肠埃希菌,但同一质粒在体内、外大肠埃希菌株中耐药标志表达有差异,即使在同一小鼠体内分离的不同接合子,pRST98/E.coli菌株耐药性亦有不同,显示耐药质粒表达的多样性和复杂性。  相似文献

【目的】了解大学生宿舍空气中产超广谱β-内酰胺酶(ESBLs)大肠埃希菌的检出率和分离株之间的遗传相似性, 为预防和控制传染病的传播提供依据。【方法】用FA-1型六级筛孔撞击式空气微生物采样器采集大学生宿舍内室内空气并分离鉴定其中的产ESBLs大肠埃希菌, 采用ERIC-PCR扩增基因组DNA, 形成聚类图谱, 分析分离株的相似性。【结果】从收集到的300份空气样品中分离鉴定出194株大肠埃希菌, 30株鉴定为超广谱β-内酰胺酶分离株, 检出率达15.46%; 产ESBLs大肠埃希菌ERIC-PCR指纹图谱条带相似性在50%?100%之间。【结论】大学生宿舍空气中存在着产ESBLs大肠埃希菌污染, 应加强对空气中大肠埃希菌耐药性监测。  相似文献

AIMS: To create a fast, sensitive and inexpensive high-throughput method for the extraction of bacterial genomic DNA from selective-enrichment culture media. METHODS AND RESULTS: Lysis of bacteria was achieved using guanidinium isothiocyanate, and DNA was extracted using 96-well glass microfibre filtration plates. Extraction-PCR detected the presence of 1 cfu Yersinia ruckeri and 16 cfu Lactococcus garvieae 200 microl(-1) sample of selective-enrichment medium. CONCLUSION: An efficient method for high-throughput extraction of bacterial genomic DNA from selective-enrichment culture media was achieved. SIGNIFICANCE AND IMPACT OF THE STUDY: This method enables detection of covert bacterial infections in fish. The simultaneous extraction of large numbers of samples allows for its use in bacterial monitoring programmes and quarantine.  相似文献

A membrane-based DNA hybridization technique was used in a model system to examine the effect of irradiation treatment on the detection of bacterial contamination in foodstuffs. Although hybridization signals were reduced compared with otherwise identical unirradiated food samples, artificial contamination levels in excess of 105 cfu per test could be distinguished in 12 of the 13 foods examined following the irradiation process. In no case were viable bacteria detected following irradiation treatment.  相似文献

AIM: To determine the sensitivity of methods for detection of injured and uninjured Escherichia coli O157:H7 (E. coli O157) in raw and pasteurized milk. METHODS AND RESULTS: Raw milk, pasteurized milk with 1.5% fat content and pasteurized milk with 3.5% fat content were spiked with E. coli O157 at low levels. The samples were enriched in modified tryptone soya broth with novobiocin (mTSBn) at 37 degrees C. Aliquots of the enriched culture were analysed either by manual immunomagnetic separation (MIMS) and culturing on sorbitol MacConkey agar with or without cefixime and potassium tellurite (SMACct or SMAC), or by automated immunomagnetic separation and integrated ELISA (EiaFosstrade mark). Uninjured E. coli O157 organisms were detected in milk by both methods at 1 cfu 10 ml-1 sample). Injured organisms were detected at levels of about 4 cfu 10 ml-1 sample. Direct enrichment in mTSBn (22 h incubation) showed better sensitivity for injured cells than enrichment in buffered peptone water (BPW, 22 h incubation), or in a two-step enrichment consisting of BPW (6 h, 37 degrees C) and mTSBn (16 h, 37 degrees C), successively. CONCLUSIONS: The methods showed equal sensitivity in that they were both able to detect 1 cfu 10 ml-1 milk sample. Injured organisms can be detected and isolated at a level almost as low as this. A resuscitation step is not recommended for the detection and isolation of injured and non-injured E. coli O157 from milk. SIGNIFICANCE AND IMPACT OF THE STUDY: Due to the dilution of contamination in the bulk tank, analysis of milk for the presence of E. coli O157 requires a very sensitive method. Both methods described here are useful for such analysis.  相似文献

Bacteriophage contamination of starter culture and raw material poses a major problem in the fermentation industry. In this study, a rapid detection of lytic phage contamination in starter culture using water-in-oil-in-water (W/O/W) emulsion microdroplets was described. A model bacteria with varying concentrations of lytic phages were encapsulated in W/O/W emulsion microdroplets using a simple needle-in-tube setup. The detection of lytic phage contamination was accomplished in 1 h using the propidium iodide labeling of the phage-infected bacteria inside the W/O/W emulsion microdroplets. Using this approach, a detection limit of 102 PFU/mL of phages was achieved quantitatively, while 104 PFU/mL of phages could be detected qualitatively based on visual comparison of the fluorescence images. Given the simplicity and sensitivity of this approach, it is anticipated that this method can be adapted to any strains of bacteria and lytic phages that are commonly used for fermentation, and has potential for a rapid detection of lytic phage contamination in the fermentation industry.  相似文献

Aims: Microbial contamination of cell culture production processes is a current concern for biopharmaceutical industries. Traditional testing methods require several days to detect contamination and may advantageously be replaced by a rapid detection method. We developed a new method combining membrane filtration to microcolonies fluorescence staining method (MFSM) and compared it to epifluorescence microscopy. Methods and Results: Both methods were used to detect bacteria in CHO cells cultures. The epifluorescence microscopy showed to be limited by filterability, media interference and nonrobustness issues, whereas MFSM enabled consistent detection of Bacillus cereus, Staphylococcus epidermidis and Propionibacterium acnes after, respectively, 8, 9 and 48 h of incubation. Thanks to the nondestructive feature of the MFSM, stained membranes could be reincubated on culture media to yield visible colonies used for identification. Conclusions: The new method described in this study showed its ability to detect microbial contaminants in cell culture samples with time‐to‐results from 2–5 times shorter than the traditional testing method. Significance and Impact of the Study: The MFSM can be used as monitoring tool for cell cultures to significantly shorten detection times of microbial contamination, while preserving the ability to identify the contaminants and their viability.  相似文献

Mycoplasma contamination in cell culture is considered as serious problem in the manufacturing of biological products. Our goal in this research is to find the best standard and rapid method with high sensitivity, specificity, accuracy and predictive values of positive and negative results for detection of mycoplasma contamination in cell cultures of the National Cell Bank of Iran. In this study, 40 cell lines suspected to mycoplasma contamination were evaluated by three different methods: microbial culture, enzymatic mycoalert® and molecular. Enzymatic evaluation was performed using the mycoalert® kit while in the molecular technique, a universal primer pair was designed based on the common and fixed 16SrRNA ribosomal sequences used. Mycoplasma contaminations in cell cultures with molecular, enzymatic and microbial culture methods were determined as 57.5, 52.5 and 40 %, respectively. These results confirmed the higher rate of sensitivity, specificity and accuracy for the molecular method in comparison with enzymatic and microbial methods. Polymerase chain reaction (PCR) assay based on fixed and common sequences in the 16SrRNA, is a useful valuable and reliable technique with high sensitivity, specificity and accuracy for detection of mycoplasma contamination in cell cultures and other biological products. The enzymatic mycoalert® method can be considered as a substitution for conventional microbial culture and DNA staining fluorochrome methods due to its higher sensitivity, specificity and speed of detection (<20 min).  相似文献

PCR-ELISA detection of Escherichia coli in milk   总被引:4,自引:0,他引:4  
AIMS: The purpose of this study was to develop a reliable molecular procedure for the detection of Escherichia coli in milk. METHODS AND RESULTS: Robust and expeditious DNA extraction and PCR techniques were evaluated using Enzyme-Linked Immunosorbent Assay (ELISA) detection of biotin-labelled amplicons to facilitate optimal detection of E. coli DNA. CONCLUSIONS: It was found that 5 E. coli colony-forming units (cfu) could be detected per PCR reaction using the PCR-ELISA system, equating to a sensitivity of detection of 100 E. coli cfu ml(-1) pasteurized milk. SIGNIFICANCE AND IMPACT OF THE STUDY: This approach should facilitate evaluation of milk contamination and enable rapid detection of E. coli mastitis, leading to correct deployment of relevant antibiotic therapy and improved animal welfare.  相似文献

Detection and identification of bacterial etiology in urine is critical for accurate diagnosis and subsequent rational treatment of urinary tract infections (UTIs). Urine culture followed by a series of biochemical reactions is currently the standard method for detecting and distinguishing microorganisms associated with UTIs. The whole procedure commonly takes more than 24 h. Here we developed a new system combining 16S rRNA gene broad-range PCR with pyrosequencing technology that allows for bacteria detection and identification in urine in 5 h. To evaluate this system for rapid diagnosis of bacteriuria, 768 urine specimens were collected from patients with suspected UTIs and were tested side-by-side using standard urine culture-based identification method and the pyrosequencing method. The results from pyrosequencing correlated well with those from traditional culture-based identification method. The overall agreement between these two methods reached 98.0% (753/768). In addition, we tested the sensitivity of pyrosequencing method and determined that urine bacterial numbers as low as 104 cfu/ml could be accurately detected and identified. In conclusion, compared with traditional biochemical method, the PCR-pyrosequencing system significantly improved the detection and identification of bacteriuria with shorter time, higher accuracy, and higher throughput, thus allowing earlier pathogen-adapted antibiotic therapy for patients.  相似文献

AIMS: To develop a procedure for direct detection of viable cells of Clavibacter michiganensis subsp. sepedonicus (Cms), the causal organism of bacterial ring rot in potato, based on AmpliDet RNA, in which amplicons generated by nucleic acid sequence based amplification (NASBA) are monitored in real time with a molecular beacon. METHODS AND RESULTS: Five methods were evaluated and fine-tuned for extraction of RNA from Cms. The most efficient non-commercial RNA extraction method included an enzymatic breakdown of the cell wall followed by a phenol extraction. AmpliDet RNA enabled detection of 10,000 molecules of purified rRNA per reaction and 100 cfu of Cms per reaction in more complex samples. Two primer pairs were tested with DNA and RNA purified from Cms. One primer pair was able to distinguish live from dead cells. CONCLUSIONS: An AmpliDet RNA was developed which enabled fast and specific detection of viable cells of Cms in complex substrates at a detection limit of 100 cfu per reaction. SIGNIFICANCE AND IMPACT OF THE STUDY: This novel AmpliDet RNA is carried out in sealed tubes, thus reducing the risk of carry-over contamination. The method will be particularly suitable for studies on the epidemiology of Cms in which viable cells should be exclusively detected.  相似文献

Chinese hamster ovary (CHO) cell cultures used to produce biopharmaceuticals are tested for mycoplasma contamination as part of the ensurance of a safe and pure product. The current U.S. Food and Drug Administration (FDA) regulatory guideline recommends using two procedures: broth/agar cultures and DNA staining of indicator cell cultures. Although these culture methods are relatively sensitive to most species, theoretically capable of detecting as few as 1-10 cfu/ml of most species, the overall procedure is lengthy (28 d), costly and less sensitive to noncultivable species. The detection of mycoplasma using the polymerase chain reaction (PCR) method has been considered an alternative method because it is relatively fast (1-2 d), inexpensive, and independent of culture conditions, however, limitations in sensitivity (limit of detection >/=1000 cfu/ml) and the risk of false positive and false negative results have prevented PCR from replacing the traditional culture methods in the industrial setting. In this report, we describe a new PCR assay for mycoplasma detection that appears to resolve these issues while being sufficiently simple and inexpensive for routine use. This assay applies readily available techniques in DNA extraction together with a modified single-step PCR using a previously characterized primer pair that is homologous to a broad spectrum of mycoplasma species known to infect mammalian cell cultures. Analysis is made easy by the detection of only a single amplification product within a narrow size range, 438-470 bp. A high sensitivity and specificity for mycoplasma detection in CHO cell production cultures is made possible through the combination of three key techniques: 8-methoxypsoralen and UV light treatment to decontaminate PCR reagents of DNA; hot-start Taq DNA polymerase to reduce nonspecific priming events; and touchdown- (TD-) PCR to increase sensitivity while also reducing nonspecific priming events. In extracts of mycoplasma DNA, the limit of detection for eight different mycoplasma species is 10 genomic copies. In CHO cell production cultures containing gentamicin, the limit of detection for a model organism, gentamicin-resistant M. hyorhinis, is 1 cfu/ml. The sensitivity and specificity of this PCR assay for mycoplasma detection in CHO cell production cultures appear similar to the currently used culture methods and thus should be considered as an alternative method by the biopharmaceutical industry.  相似文献
