Fragmented QRS (fQRS) is a convenient marker of myocardial scar evaluated by 12-lead electrocardiogram (ECG) recording. fQRS is defined as additional spikes within the QRS complex. In patients with CAD, fQRS was associated with myocardial scar detected by single photon emission tomography and was a predictor of cardiac events. fQRS was also a predictor of mortality and arrhythmic events in patients with reduced left ventricular function. The usefulness of fQRS for detecting myocardial scar and for identifying high-risk patients has been expanded to various cardiac diseases, such as cardiac sarcoidosis, arrhythmogenic right ventricular cardiomyopathy, acute coronary syndrome, Brugada syndrome, and acquired long QT syndrome. fQRS can be applied to patients with wide QRS complexes and is associated with myocardial scar and prognosis. Myocardial scar detected by fQRS is associated with subsequent ventricular dysfunction and heart failure and is a substrate for reentrant ventricular tachyarrhythmias.  相似文献

What Is a Story?     
This paper attempts to discover the criteria by which a listener accepts or rejects an item of oral narrative as being a story. The hypotheses are: (1) such criteria relate to structure rather than content, although the listener does not consciously distinguish structure from content; (2) the structure must have a certain minimal and maximal degree of complexity and be of a certain kind; (3) the criteria will hold cross-culturally. Information gathered from ethnographies and folklore literature is inadequate to confirm the hypotheses, but it does not contradict them. An experiment to test hypotheses (1) and (2) produced a certain degree of negative confirmation: listeners rejected as a story any narrative item which did not conform to the structural criteria regarded as minimal by myself; however, they also rejected items which I regarded as well structured but which were bizarre or nonsensical in content. Background

A bioartificial heart is a theoretical alternative to transplantation or mechanical left ventricular support. Native hearts decellularized with preserved architecture and vasculature may provide an acellular tissue platform for organ regeneration. We sought to develop a tissue-engineered whole-heart neoscaffold in human-sized porcine hearts.

Methods

We decellularized porcine hearts (n = 10) by coronary perfusion with ionic detergents in a modified Langendorff circuit. We confirmed decellularization by histology, transmission electron microscopy and fluorescence microscopy, quantified residual DNA by spectrophotometry, and evaluated biomechanical stability with ex-vivo left-ventricular pressure/volume studies, all compared to controls. We then mounted the decellularized porcine hearts in a bioreactor and reseeded them with murine neonatal cardiac cells and human umbilical cord derived endothelial cells (HUVEC) under simulated physiological conditions.

Results

Decellularized hearts lacked intracellular components but retained specific collagen fibers, proteoglycan, elastin and mechanical integrity; quantitative DNA analysis demonstrated a significant reduction of DNA compared to controls (82.6±3.2 ng DNA/mg tissue vs. 473.2±13.4 ng DNA/mg tissue, p<0.05). Recellularized porcine whole-heart neoscaffolds demonstrated re-endothelialization of coronary vasculature and measurable intrinsic myocardial electrical activity at 10 days, with perfused organ culture maintained for up to 3 weeks.

Conclusions

Human-sized decellularized porcine hearts provide a promising tissue-engineering platform that may lead to future clinical strategies in the treatment of heart failure.  相似文献

The mammalian ABL1 gene encodes the ubiquitously expressed nonreceptor tyrosine kinase ABL. In response to growth factors, cytokines, cell adhesion, DNA damage, oxidative stress, and other signals, ABL is activated to stimulate cell proliferation or differentiation, survival or death, retraction, or migration. ABL also regulates specialized functions such as antigen receptor signaling in lymphocytes, synapse formation in neurons, and bacterial adhesion to intestinal epithelial cells. Although discovered as the proto-oncogene from which the Abelson leukemia virus derived its Gag–v-Abl oncogene, recent results have linked ABL kinase activation to neuronal degeneration. This body of knowledge on ABL seems confusing because it does not fit the one-gene-one-function paradigm. Without question, ABL capabilities are encoded by its gene sequence and that molecular blueprint designs this kinase to be regulated by subcellular location-dependent interactions with inhibitors and substrate activators. Furthermore, ABL shuttles between the nucleus and the cytoplasm where it binds DNA and actin—two biopolymers with fundamental roles in almost all biological processes. Taken together, the cumulated results from analyses of ABL structure-function, ABL mutant mouse phenotypes, and ABL substrates suggest that this tyrosine kinase does not have its own agenda but that, instead, it has evolved to serve a variety of tissue-specific and context-dependent biological functions.  相似文献

We are seeing the beginnings of a process of folkloricization of various Tukanoan cultural traits. Perhaps in the future we shall see the commoditization of them. Elsewhere in the hemisphere various interest groups, including indigenous groups themselves, package and promote Indianness. The cultural forms that are retained from earlier traditions can therefore totally change in meaning. This poses problems when we talk about cultures using an organic model, because we find we cannot describe these processes in other than negative language. Both anthropologists and pro-Indian activists at times find it academically and/or politically expedient to talk of culture as enduring over time: while changing, these cultures are nonetheless seen as remaining the same in some fundamental ways. But when, as is beginning to occur in the Tukanoan case, traits are retained, cast aside or redefined as part of a self-conscious awareness and promotion of a particular kind of Indian identity as a political strategy, the meaning of these traits has often radically changed. We cannot use a quasi-biological model to account for these similarities over time.Since resemblances between earlier forms of Tukanoan culture and later forms may be superficial, conceiving of a culture in terms of traits that persist over time can be misleading. We need to think of culture change over relatively short periods of time in a more dynamic fashion, rather than as either the same or syncretized or lost. We need to see Tukanoans and others as creating and improvising, rather than possessing, culture. And we need to create and invent models and metaphors that analyze this process in nonderogatory terms.  相似文献

Speciation has been a major focus of evolutionary biology research in recent years, with many important advances. However, some of the traditional organising principles of the subject area no longer provide a satisfactory framework, such as the classification of speciation mechanisms by geographical context into allopatric, parapatric and sympatry classes. Therefore, we have asked where speciation research should be directed in the coming years. Here, we present a distillation of questions about the mechanisms of speciation, the genetic basis of speciation and the relationship between speciation and diversity. Our list of topics is not exhaustive; rather we aim to promote discussion on research priorities and on the common themes that underlie disparate speciation processes.  相似文献

The ability of dogs to use human communicative signals has been exhaustively studied. However, few studies have focused on the production of communicative signals by dogs. The current study investigated if dogs are able to communicate by using directional signals towards some desirable object in the environment and also if they show an apparent intention to manipulate their owner’s behavior in order to receive it. Some operational criteria were used to investigate referential and intentional communication: the signal should be influenced by the audience and by the recipient’s direction of visual attention; the sender should display gaze alternations between the recipient and the object and attention-getting behaviors, and, finally, the sender should persist and elaborate the communication when attempts to manipulate the recipient failed. Aiming to investigate these criteria in dogs, 29 subjects were tested using an experimental set up in which they could see a desirable but unreachable food and they needed the cooperation of their owners in order to receive it. This study found evidence of all operational criteria, especially for gaze alternation between the owner and the food, which suggested that some dogs’ communicative behaviors could be functionally referential and intentional. Nevertheless, similar to other studies about social cognition in animals, it is not possible to distinguish if the dog’s behaviors are based on simple mechanisms or on a theory of mind about their owners.  相似文献

Though bacteria are predominantly asexual, the genetic information in their genomes can be expanded and modified through mechanisms that introduce DNA from outside sources. Bacterial sex differs from that of eukaryotes in that it is unidirectional and does not involve gamete fusion or reproduction. The input of DNA during bacterial sex generates diversity in two ways--through the alteration of existing genes by recombination and through the introduction of novel sequences--and each of these processes has been shown to aid in the survival and diversification of lineages.  相似文献
