The influence of cadmium dust (containing lead, cadmium, copper, zinc, silicium and other elements) on acid phosphatase activity of Pisolithus arrhizus was observed by means of electron microscopy. Dust-treated mycelium showed increased activity of the enzyme, especially on the surface of the cell wall. There was an increase in abundance of autophagic vacuoles marked by a strong phosphatase reaction. An increase in the number of hyphae with diffuse enzyme activity within the cytoplasm coincided with a decrease of lifespan of the fungus, rapid changes in the mictoplasm stage, earlier closing of the dolipori and presumably the earlier autolysis of cell cytoplasm. Hyphae showing strong autolytic activity were separated from other hyphae by the material deposited within the doliporus and this whole area was devoid at that stage of acid phosphatase activity. The role of the enzyme in the mechanism of resistance to toxic elements is discussed.  相似文献

Summary Eight isolates of ectomycorrhizal fungi namely, Laccaria fraterna (EM-1083),Laccaria laccata (EM-1191), Pisolithus tinctorius (EM-1081),Pisolithus tinctorius (EM-1293), Scleroderma cepa (EM-1233),Scleroderma flavidum (EM-1235),Scleroderma verucosum, (EM-1283) and Hysterangium incarceratum (EM-1185) were grown on specially designed cocktail media prepared by adding various concentrations of different heavy metals namely Al, As, Cd, Cr, Ni and Pb. The heavy metals were selected keeping in view their relative abundance in coal ash and potential toxicity. The fungal isolates were grown on such designed cocktail media. The colony diameter was used for the measurement of the fungal growth. Total heavy metal accumulated in the mycelia was assayed by atomic absorption spectrophotometry.In relation to metal tolerance ability in general,Hysterangium incarceratum (EM-1185) showed maximum tolerance with respect to growth, Laccaria fraterna (EM-1083) and Pisolithus tinctorius (EM-1293) also showed considerable tolerance to the heavy metals tested. In relation to metal uptake in particular, Pisolithus tinctorius (EM-1293), has reported maximum uptake of Al (34642.58 ppm), Cd (302.12 ppm) and Pb (3501.96 ppm). In Laccaria fraterna (EM-1083), As (130.57 ppm) and Cr (402.38 ppm) uptake was recorded maximum; and Hysterangium incarceratum (EM-1185) has recorded maximum Ni (2648.59 ppm) uptake among the three suitable isolates documented here.  相似文献

The seed coats of Digitalis purpurea L. cv. Gloxiniaflora werecultured for indigenous fungi. Alternaria alternata (Fries)was identified as the sole fungus on seeds freshly harvestedfrom unopened capsules, whereas A. alternata, Rhizopus arrhizus(Fischer), Penicillium sp. and other fungi appeared on storedseeds. The appearance of fungi in seed cultures was seasonal,being more frequent in winter and early spring than in summerand autumn. Alternaria grew well on autoclaved seeds, on dehiscentseed coats, or on seed coats separated from the embryos of ungerminatedseeds. Rhizopus did not grow on these but grew weakly on theculture medium from viable seeds. A. alternata appears to functionas a degradation agent for the seed coat subsequent to germination.Neither fungus was found to be essential to germination of Digitalisseeds. Bioassay of the culture medium from germinating seedsshowed that a fungistat effective against both Alternaria andRhizopus is produced coincident with germination. Based on chromatographicanalysis, the fungistat appears to be a cardenolide. Accumulation of lead and zinc was studied in the moss Funariahygrometrica Hedw. collected from mine tailings. Heavy metalaccumulation in gametophytes and sporophytes was quantifiedby graphite furnace atomic absorption spectrometry (GFAAS) andinductively coupled plasma-atomic emission spectrometry (ICP-AES).Pb and Zn accumulation in the placental zone was analysed byx-ray scanning electron microscopy (SEM) and transmission electronmicroscopy (TEM) microanalysis. Spectrometry showed that whilemoss gametophytes accumulated considerable concentrations ofheavy metals, sporophytes accumulated only small concentrationsof metals. X-ray SEM and TEM showed that the two metals accumulatedin placental transfer cells on both the gametophytic and sporophyticsides. To investigate the uptake pattern for both metals undercontrolled conditions, F. hygrometrica plants collected froma non-polluted site were treated in the laboratory with separatesolutions of Pb and Zn at two concentrations (10-2and 10-4 M)for 24 or 168 h. Metal accumulation was analysed separatelyin gametophytes and sporophytes using GFAAS and ICP–AES.Each generation had a different accumulation quotient for bothmetals, and gametophytes accumulated significantly more metalthan sporophytes. Concentrations of Zn in sporophytes were alwayshigher than concentrations of Pb. To discover the differences and modes of saccharifying ability among Rhizopus arrhizus strains isolated from various habitats, we grew strains in glutinous rice media to monitor the production of glucose from starch using spectrophotometry at regular time intervals. The saccharifying capacity of R. arrhizus var. delemar was found to be weaker than that of var. arrhizus and var. tonkinensis. These data provide additional phenotypic support for recognization of this genetically distinct lineage (var. delemar) as a separate species. Clinical isolates were generally low in saccharifying abilities, while all strains from southwest China had an excellent diastatic power.  相似文献

The physico-chemical properties of iron oxyhydroxide plaquesformed on the roots of Phragmites australis under field andlaboratory conditions were determined using electron microscopyand energy dispersive spectrometry (EDS) analysis. Plaques werepresent as an amorphous coating on roots with an uneven distribution.They were shown to be composed of iron in both the field andlaboratory, and phosphorus was found to be adsorbed onto thesurface of plaques formed in the laboratory. The uptake of copperand manganese in the presence and absence of plaque was investigatedunder two different pH regimes. Concentrations of Cu were lowerin the shoots of P. australis in the presence of plaque (565mg kg-1) than when it was absent (1400 mg kg-1), under growthconditions of higher pH (6.0). The adsorption of Cu and Mn ontothe plaque surface was not the mechanism by which plaque reducedthe uptake of other metals. Alternatively, the plaque may simplyact as a physical barrier. Under conditions of lower pH (3.5)the activity of hydrogen ions at the root surface interferedwith the movement of metals into the root and masked any potentialeffect of iron plaque. The purpose of this study was to determine whether an aqueousextract of grain sorghum dust increases macromolecular efflux from thenasal mucosa in vivo and, if so, whether this response is mediated, inpart, by substance P. Suffusion of grain sorghum dust extract on the insitu nasal mucosa of anesthetized hamsters elicits a significantincrease in clearance of fluorescein isothiocyanate-labeled dextran(FITC-dextran; mol mass, 70 kDa; P < 0.05). This response is significantly attenuated byCP-96345 and RP-67580, two selective, but structurally distinct,nonpeptide neurokinin 1 (substance P)-receptor antagonists, but not byCP-96344, the2R,3Renantiomer of CP-96345 (P < 0.05). CP-96345 has no significant effects on adenosine-induced increase in clearance of FITC-dextran from the insitu nasal mucosa. CP-96345 and RP-67580, but not CP-96344, significantly attenuate substance P-induced increases in clearance ofFITC-dextran from the in situ nasal mucosa(P < 0.05). Collectively, these datasuggest that grain sorghum dust elicits neurogenic plasma exudationfrom the in situ nasal mucosa.

In vitro growth of Acer pseudoplatanus L. (sycamore) callustissue derived from shoot-tip explants and screened on metal-enrichedmedia was studied in an attempt to identify resistance traitswhich may explain the survival of trees at metal-contaminatedsites. Copper and Cd-resistance traits were identified in celllines originating from trees at a site with a relatively recenthistory of severe contamination by these metals, and Cd- andZn-resistance were identified in cell lines originating frommature trees at a mining spoil site with a much longer historyof exposure to elevated concentrations of these metals. In Zn-resistantcell lines, co-resistance to Ni was also found, even thoughthis metal was not elevated at the study site. This is the firstreport of multiple resistance and co-resistance to metals occurringat the cellular level in trees. The mechanisms of the measuredresistance traits remain unclear, although there was evidenceof reduced Cu and Ni uptake by resistant cell lines. The phylogenetic relationships among 200 Pisolithus basidiomata collected from pine, oak, and eucalypt forests and rockrose scrubs in Morocco were investigated. Using PCR-RFLP analysis of the internal transcribed spacer (ITS) of ribosomal DNA, this collection could be divided into 5 groups, by using PCR-RFLP analysis of the internal transcribed spacer (ITS) of ribosomal DNA. The ITS of a representative basidioma of each group was sequenced, and a phylogenetic analysis was performed. The dendrogram suggests that at least five Pisolithus species are present in Morocco. Pisolithus basidiomata collected in the Pinus and Quercus forests correspond to Pisolithus arrhizus and P. species 4 as previously described by Martin and colleagues in 2002. Those collected from the eucalyptus forests, under E. gomphocephala and E. camaldulensis, correspond to P. albus and P. microcarpus. Basidiomata collected from the rockrose scrubs, under Cistus crispus, C. monspeliensis or C. salviifolius, are all identified as Pisolithus species 3. Phylogenetic analyses showed that our different Pisolithus grouped well with Pisolithus specimens from other geographical origins.  相似文献

A program of house dust sample collection and analysis has begun in Syracuse, New York, USA, in order to determine the feasibility of a geography-based exposure assessment for urban metals. The sampling program, and the protocols it employs, is described for two different types of wipe media, Ghost Wipes and Whatman Filters. Preliminary results show that strong spatial patterns of floor dust loading (mg dust per square foot) can be observed for data aggregated at a spatial scale of about 1600 m (~2.5 km2). Floor dust metal concentrations were similar to those found in other urban environments, with some regional variation. The median floor dust Pb concentration was ~108 mg · kg?1 for this initial data set of ~264 sampled residential locations, and varied from 50 to 1100 mg Pb · kg?1.  相似文献

The green alga Kirchneriella lunaris was incubated with variousheavy metals (Cd2+, Co2+, Mn2+, Ni2+) in presence/absence ofcalcium (Ca2+). The uptake of heavy metal was affected by Ca2+.Growth rate was inhibited by all heavy metals applied. In allCa2+-containing cultures Kirchneriella exhibited higher ratesof growth than those containing heavy metal alone. Photosynthesis/respirationratio of K. lunaris cells seems to be the determinant in thiswork. Ca2+ variably abolished the effects of the heavy metalsstudied. This study examines factors affecting oral bioaccessibility of metals in household dust, in particular metal speciation, organic carbon content, and particle size, with the goal of addressing risk assessment information requirements. Investigation of copper (Cu) and zinc (Zn) speciation in two size fractions of dust (< 36 μ m and 80–150 μ m) using synchrotron X-ray absorption spectroscopy (XAS) indicates that the two metals are bound to different components of the dust: Cu is predominately associated with the organic phase of the dust, while Zn is predominately associated with the mineral fraction. Total and bioaccessible Cu, nickel (Ni), and Zn were determined (on dry weight basis) in the < 150 μ m size fraction of a set of archived indoor dust samples (n = 63) and corresponding garden soil samples (n = 66) from the City of Ottawa, Canada. The median bioaccessible Cu content is 66 μ g g?1 in dust compared to 5 μ g g?1 in soil; the median bioaccessible Ni content is 16 μ g g?1 in dust compared to 2 μ g g?1 in soil; and the median bioaccessible Zn content is 410 μ g g?1 in dust compared to 18 μ g g?1 in soil. For the same data set, the median total Cu content is 152 μ g g?1 in dust compared to 17 μ g g?1 in soil; the median total Ni content is 41 μ g g?1 in dust compared to 13 μ g g?1 in soil; and the median total Zn content is 626 μ g g?1 in dust compared to 84 μ g g?1 in soil. Organic carbon is elevated in indoor dust (median 28%) compared to soil (median 5%), and is a key factor controlling metal partitioning and therefore bioaccessibility. The results show that house dust and soil have distinct geochemical signatures and should not be treated as identical media in exposure and risk assessments. Separate measurements of the indoor and outdoor environment are essential to improve the accuracy of residential risk assessments.  相似文献

This study was done to confirm our previous observation withthe pattern of changes in electron transport composition inducedby an imbalance of the electron transport state. Contents ofphotosystem (PS) I and II complexes and their antennae and Cytb6/f complex were determined for systems of cyanobacterium SynechocystisPCC 6714 of different PS I/PS II ratios. The results indicatedthat (1) the observed changes in the PS I/PS II ratio are not-dueto regulation of the activities of the respective PS's but tochanges in their contents, (2) the molar ratio between PS IIand Cyt b6/f complexes was fairly constant when marked changesoccurred in the PS I content, and (3) the PS II and Cyt b6/fcontents per cell remained fairly constant while the PS I contentchanged markedly. These findings agree with our previous observationwith autotrophic cells of Anacystis nidulans Tx 20 and supportour argument that in cyanobacterial and red algal electron transportsystems, the content of the terminalcomponent(s), such as PSI complex, is regulated in order to maintain a balance betweenthe electron influx by PS II action to the system and the effluxby PS I action from it. Summary The biosorption of uranium, strontium and caesium by pelleted mycelium of two species of fungi,Rhizopus arrhizus andPenicillium chrysogenum and immobilizedSaccharomyces cerevisiae was evaluated in both batch and continuous flow systems where the presence of competing cations affected accumulation. The uptake mechanism for the pelleted fungal biomass differed from that of the immobilized yeast, the former being metabolism-independent biosorption of the metals while, in the presence of glucose, uptake in the latter organism was biphasic, surface biosorption being followed by energy-dependent influx. Removal of surface-bound metals was achieved by eluting with mineral acids or carbonate/bicarbonate solutions; a high degree of metal recovery was observed for uranium.  相似文献

The culture media for the in vitro pollen assay of Brassicaspecies have so far shown good percentage germination, but limitedpollen tube growth. It is reported here that by lowering sucroseconcentration from 20% to 5 or 10 % and by adding polyethyleneglycol 4000 in the culture medium, high percentage germinationand high tube growth (10 times that in the standard media) canbe achieved in three species of cultivated Brassica. This improvedmedium should be useful in fundamental and applied studies onthe pollen biology of Brassica species. Metal accumulation in the mycelia was assayed by atomic absorption spectrophotometry. Six metals, namely aluminum, cadmium, chromium, iron, lead, and nickel, were selected on the basis of their abundance in coal ash and toxicity potential for the present work. Growth of vegetative mycelium on fly ash variants and metal accumulation data indicated that Pisolithus tinctorius (EM-1290) was the most tolerant among the isolates tested for most of the metals. Since this isolate is known to be mycorrhizal with Eucalyptus, it could be used for the reclamation of coal ash over burdened sites.  相似文献

Experiments were conducted to investigate the effect of coal ash on organic acid exudation and subsequent metal uptake by ectomycorrhizal fungi. Four isolates of ectomycorrhizal fungi namely, Pisolithus tinctorius (EM-1293 and EM-1299), Scleroderma verucosum (EM-1283) and Scleroderma cepa (EM-1233) were grown on pond ash moistened with Modified Melin-Norkans medium in vitro. Exudation of formic acid, malic acid and succinic acid by these fungi were detected by HPLC. Mycelial accumulation of Al, As, Cd, Cr, Ni and Pb by these fungi was assayed by atomic absorption spectrophotometer. Relationship between organic acid exudation and metal uptake was determined using classical multivariate linear regression model. Correlation between organic acid exudation and metal uptake could be substantiated when several metals are considered collectively. The finding supports the widespread role of low molecular weight organic acid as a function of tolerance, when exposed to metals in vitro.  相似文献

The oil palm of commerce, Elaeis guineensis (tenera), derivesfrom hybridization of variety dura x pisifera. Germ plasm ofpisifera is limited, however, because of varying degrees offemale sterility. Efforts to increase the range of availablepisifera male germ plasm have thus far been limited to fertilepisiferas but it is anticipated that clonal multiplication ofembryos and explanted organs from less female-fertile, but nonethelessfertile, male, plants could play a significant role in breedingprogrammes. Callus has been produced from mature pisifera embryosand leaf explants from young plantlets using a half-strengthMurashige and Skoog medium supplemented with inositol, caseinhydrolysate, activated charcoal and varying concentrations ofauxins. After sub-culture on media of similar composition, organizedstructures can develop. Those which form in stale nutrient media(e.g. left in the same medium for up to 3 months) give riseto embryonal structures which do not readily develop furtherand can be regarded as neomorphs. If callus once formed is transferredto media with lower auxin concentrations (40–0 mg l–1)embryonal buds and well-formed shoots are produced; additionof gibberellic acid (GA3) fosters the production of well-organizedshoots accompanied by roots. N,N'-Dicyclohexylcarbodiimide (DCCD) inhibited the flash-inducedreduction of cytochrome b560, by blocking the electron flowbetween the secondary electron acceptor and cytochrome b560presumably in the vicinity of the ubiquinone pool. The stoichiometryof the reduced cytochrome b560 per reaction center bacteriochlorophylldimer was 0.77?0.12 throughout the redox potential range of150 to 390 mV at pH 7.0.
