From a (GT)13-enriched genomic library of Oplegnathus fasciatus, 14 polymorphic microsatellite were isolated and characterized in a test population with alleles ranging from two to nine,
the observed and expected heterozygosities from 0.0000 to 1.0000, and from 0.1726 to 0.8507, respectively. Five loci deviated
from the Hardy–Weinberg equilibrium, and linkage disequilibrium between two loci was significant. Two loci were also polymorphic
in Pagrosomus major assessed for cross-species amplification. These polymorphic microsatellites will be useful for genetic diversity analysis
and molecule-assisted breeding for O. fasciatus.

Chinese alligator (Alligator sinensis) is a critically endangered species endemic to China. In this study, the extent of genetic variation in the captive alligators
of the Changxing Reserve Center was investigated using microsatellite markers derived from American alligators. Out of 22
loci employed, 21 were successfully amplified in the Chinese alligator. Sequence analysis showed loci in American alligators
had a bigger average size than that of the Chinese alligators and the longest allele of an individual locus almost always
existed in the species with longer stretch of repeat units. Eight of the 22 loci were found to be polymorphic with a total
of 26 alleles present among 32 animals scored, yielding an average of 3.25 alleles per polymorphic locus. The expected heterozygosity
(H
E) ranged at a moderate level from 0.4385 to 0.7163 in this population. Compared to that in the American alligators, a lower
level of microsatellite diversity existed in the Changxing population as revealed by about 46% fewer alleles per locus and
smaller H
E at the homologous loci. The average exclusion power and the ability to detect shared genotypes and multiple paternity were
evaluated for those markers. Results suggested that when the polymorphic loci were combined, they could be sensitive markers
in genetic diversity study and relatedness inference within the Chinese alligator populations. The level of genetic diversity
present in the current Changxing population indicated an important resource to complement reintroductions based on the individuals
from the other population. In addition, the microsatellite markers and their associated diversity characterized in this population
could be utilized to further investigate the genetic status of this species. 相似文献

We identified four new polymorphic microsatellite loci in bluefin killifish (Lucania goodei) and five loci in yellow bullheads (Ameiurus natalis). We screened 400 killifish from 20 populations and 180 bullheads from nine populations, finding a high degree of polymorphism (nine to 54 alleles per locus; average expected heterozygosity 0.678–0.976). We found no evidence for linkage disequilibrium. Three of the loci found in bluefin killifish show heterozygote deficiency; the other loci do not deviate from Hardy–Weinberg expectations. 相似文献

and expected heterozygosities per locus from 0.04 to 1.00 and from 0.28 to 0.76, respectively. Three loci significantly deviated
from Hardy–Weinberg equilibrium and no significant linkage disequilibrium was found between pairs of loci after Bonferroni
correction. Cross-species amplification of these polymorphic microsatellite loci was performed in additional two related species.
These polymorphic microsatellite loci would be useful for investigating genetic resource of E. septemfasciatus and other related species.

Achnatherum sibiricum is a threaten and toxic perennial bunchgrass mainly in the north of China. We developed 11 polymorphic microsatellite loci
from A. sibiricum by combining biotin capture method. After validating and scoring, these loci were polymorphic in a test population with the
range of alleles from four to 13 per locus. The observed and expected heterozygosities ranged from 0.1649 to 0.5624 and from
0.3071 to 0.8826, respectively. All 11 microsatellite markers were in Hardy–Weinberg equilibrium. These polymorphic microsatellites

In the study, 34 microsatellite loci were isolated from a dinucleotide-enriched genomic library of Japanese Spanish mackerel
(Scomberomorus niphonius). And 12 microsatellite loci were found to be polymorphic between 3 and 8 alleles. The number of observed and expected heterozygosity
per locus in 23 individuals ranged from 0.6087 to 1.0000 and 0.8908 to 0.9773, respectively. Two loci significantly deviated
from Hardy–Weinberg equilibrium after Bonferroni correction analysis and there was no significant linkage disequilibrium found
between pairs of loci. As a result, 12 microsatellite loci probably located on different chromosome pairs and these polymorphic
microsatellite loci should provide sufficient level of genetic diversity to investigate the fine-scale population structure
and evaluate the breeding strategy in S. niphonius.

This paper describes the characterization of 24 polymorphic microsatellite loci in Paphiopedilum rothschildianum, an endangered slipper orchid endemic to Sabah, Malaysia. A total of 36 clones containing microsatellite repeat motifs yielded
41 dinucleotide, 20 tetranucleotide and 30 cryptic simple repeat sequences. Thirty microsatellite loci were randomly selected
to characterize the population, of which 24 were found to be polymorphic. The expected heterozygosity ranged from 0.0813 to
0.7279 (mean 0.4533) and the observed heterozygosity ranged from 0 to 1 (mean 0.3800). The results indicated that there was
limited migration between the three subpopulations and a significant degree of genetic differentiation (FST = 0.5098) between populations implying that gene flow has been disrupted as a result of habitat fragmentation. The molecular
markers have a potential application in population management, CITES enforcement and the forensic detection of specimens in

The first set of polymorphic microsatellite markers were developed from so-iuy mullet (Mugil soiuy Basilewsky 1855). From a (GT)n-enriched genomic library, 53 microsatellites were selected for designing microsatellite primers,
of which 36 gave working primer pairs. Ten of these loci were polymorphic in a test population of 24 individuals with alleles
ranging from 3 to 9, and observed and expected heterozygosities from 0.2083 to 0.9167 and from 0.2651 to 0.8812, respectively.
No significant linkage disequilibrium between pairs of loci was found, but two loci significantly deviated from Hardy–Weinberg
equilibrium after Bonferroni correction. These polymorphic microsatellite loci should provide sufficient level of genetic
diversity to investigate the fine-scale population structure and evaluate the breeding strategy in Mullet.

Isolation and Characterization of Twenty Microsatellite Loci in Japanese Sea Cucumber (Stichopus japonicus) 总被引:5,自引:0,他引:5
Twenty microsatellite markers were first developed from the Japanese sea cucumber Stichopus japonicus using an enrichment protocol. Of the 20 microsatellite loci, 19 loci were polymorphic in the population examined. At these polymorphic loci, the number of alleles per locus varied from 2 to 15, and the observed heterozygosities ranged from 0.03 to 0.97, which is considerably higher than those previously found for allozymes. The high variability of the microsatellite markers identified in this study will make them excellent tools for genetic analyses of S. japonicus. 相似文献

in Nibea albiflora.

Nine polymorphic microsatellite loci were developed and characterized for Handeliodendron bodinieri, an endangered tree endemic to karst forest in southwest China. Each locus displayed between 4 and 11 alleles. The observed

An enrichment protocol was used to isolate and characterize microsatellite loci in Acer opalus, a Mediterranean tree species. Highly polymorphic microsatellite loci were required for paternity analyses in a population
of this species. Eight microsatellite loci were amplified and a total of 87 alleles were detected in a sample of 142 individuals
from one population, allowing the identification of each individual with a unique multilocus genotype. The paternity exclusion
probabilities varied from 0.261 for locus Aop820 to 0.806 for locus Aop450, and the parent-pair exclusion probabilities varied
from 0.433 for Aop820 to 0.940 for Aop450. The cumulative probabilities of exclusion for paternity and parentage of the eight
loci were both higher than 0.999, supporting the usefulness of these microsatellite loci for future paternity and parentage
analyses in A. opalus. Cross-species transferability was also assayed, supporting their potential use in other eight Acer species. 相似文献

ranging from 3 to 18, and expected and observed heterozygosities from 0.5772 to 0.9449 and from 0.4286 to 0.9231, respectively.
No significant linkage disequilibrium between pairs of loci was found, however, loci Jobe24 significantly deviated from Hardy–Weinberg
equilibrium after Bonferroni correction. These polymorphic microsatellite loci should provide sufficient level of genetic

Forty microsatellite markers were developed from a (CA)n enrichment library created from the DNA of southern catfish (Silurus meriaionalis). Also, the population structure of northern sheatfish (Silurus soldatovi) was examined by 24 microsatellite loci. They are polymorphic in at least one of the two geographically distant populations sampled from the Heilongjiang River and the Songhuajiang River in North China, respectively. Unbiased expected heterozygosity levels varied from 0.435 to 0.946 and number of alleles per locus varied from three to 20. The sandfly Lutzomyia longipalpis, an important vector of visceral leishmaniasis in the New World, is believed to be a species complex. In an effort to better understand population dynamics and speciation in this vector we developed a panel of dinucleotide — (CA)n— microsatellite loci using an enrichment technique. Eleven polymorphic loci that produced consistent allelic banding patterns were characterized using a laboratory population of L. longipalpis. These dinucleotide microsatellite loci were more polymorphic than trinucleotide microsatellites characterized in wild‐caught samples of two other sandfly species; the variability of these loci was unexpected because the laboratory flies were believed to be inbred. 相似文献
