In developing countries, diet during pregnancy is frequently low in both protein and zinc contents and exposure to CO is common because of environmental pollution and smoking. This study was conducted to evaluate whether zinc supplementation ameliorates fetal mortality and malformations in protein-deficient, CO-exposed mice. Pregnant mice of the CD-1 strain were maintained on 17% (reference) or 9% protein diets mixed with deficient, normal, or supplemental zinc throughout gestation. The dams in each dietary group were exposed to air (control) or 500 ppm CO in air in environmental chambers from gestation days 7–18. As compared to the control group (normal protein, normal zinc), the incidence of fetal mortality was 66.8% and 57.2% higher, respectively, and malformation incidence was 74.4% and 72.4% higher (0 and 500 ppm CO, respectively) in mice fed both deficient protein-zinc diets. However, the highest malformation rate was observed in the group with normal protein, deficient zinc (96% mortality in both 500 and 0 ppm CO, as compared to the reference group, p<0.0001). The fetal mortality rate was −3.5% (0 ppm CO) and 25.4% (500 ppm CO) lower in zinc-supplemented, protein-deficient groups compared to the control group. There was a significant negative association between fetal zinc concentrations and fetal malformations (p≤0.001). The result of this study might be relevant to populations that are exposed to CO and or consume marginal zinc and protein diets during gestation.  相似文献

Pup production and mortality of beach breeding Grey seals was studied at two contrasting types of rookery, the cliff-bound island of Ramsey, Dyfed, and the low, grassy island of Auskerry, Orkney. Thirty-five per cent of pups died on Ramsey compared with 14 % on Auskerry. Almost half of the Ramsey mortality was accounted for by animals being lost from the beaches so that they were not available for analysis. The main proximate causes of death revealed by post-mortem examination were starvation and infections. The other conditions which accounted for a small proportion of deaths were drowning, trauma, non-viability, stillbirths, atelectasis, dystokia and heart abnormality. Certain pathogens associated with disease were common to both sites, while others were specific to Ramsey or Auskerry.
The most important ultimate cause of pup deaths appeared to be failure of the mother/pup bond. The differences in levels of mortality between the sites is thought to be due to differences in the topography of the beaches. Pup survival on the narrow cliff-bound beaches, or in caves, on Ramsey was reduced either directly through pups being washed off, or indirectly by overcrowding of animals at high tide.  相似文献

The critical period for increased neonatal mortality induced by perfluorooctane sulfonate (PFOS) exposure was evaluated in the rat. Timed-pregnant Sprague-Dawley rats were treated by oral gavage with 25 mg/kg/d PFOS/K(+) on four consecutive days (gestation days (GD) 2-5, 6-9, 10-13, 14-17, or 17-20) or with 0, 25, or 50 mg/kg/d PFOS/K(+) on GD 19-20. Controls received vehicle (10 ml/kg 0.5% Tween-20) on these days. Maternal weight gain was reduced in treated animals during dosing, as were food and water consumption. Following a 4-day treatment, litter size at birth was unaffected while pup weight was similarly reduced in the three earliest PFOS groups. All PFOS groups experienced decreases in survival while controls remained near 100%. Neonatal survival decreased in groups dosed later during gestation, approaching 100% with dosing on GD 17-20. Most deaths occurred before postnatal day (PND) 4, with the majority in the first 24 hours. Maternal serum PFOS levels on GD 21 were higher in groups exhibiting higher mortality. Following a 2-day treatment, PFOS groups experienced significant pup mortality by PND 1. Neonatal mortality continued through PND 5, when survival was 98, 66, and 3% for the 0, 25, and 50 mg/kg groups, respectively. Pup weight was reduced in treated groups with surviving litters. Gross dissection and histological examination of lungs revealed differences in maturation between control and treated animals on PND 0. We conclude that exposure to PFOS late in gestation is sufficient to induce 100% pup mortality and that inhibition of lung maturation may be involved.  相似文献

Pregnant Sprague-Dawley rats were used to determine the effects of the addition of 200 ppm of Cd (as CdCl2) to the diet factorially with two levels of dietary Ca (0.07% and 0.96%) on reproductive performance, concentrations of Cd, Cu, Fe, Zn, Ca and Mg in dam liver and kidney and in newborn progeny. High Cd significantly increased liver and kidney Cd, Zn and Ca and decreased liver Fe. High dietary Ca partially protected against accumulation of Cd in liver and kidney but had no effect on concentration of other elements. Number of live or stillborn pups per litter was not significantly affected by diet but high Cd significantly reduced pup birth weight. No grossly abnormal pups were noted. Concentration of Cd in bodies of newborn pups was increased approximately 8.6-fold by high Cd in the diet of dams fed the 0.07% Ca-diet and 3.8-fold by high-Cd in the diet of dams fed the 0.96% Ca diet. Pup, Zn, Cu and Fe contents were significantly decreased and Ca was significantly increased by high-Cd in the maternal diet whereas pup Mg content was unchanged. Maternal Ca intake had no effect on concentration of Zn, Cu, Fe or Ca in newborn pups. The biological importance of the alteration in maternal and fetal tissue concentration of Zn, Cu and Fe by high-Cd maternal diets is unknown.  相似文献

As part of population dynamics studies of the South American fur seal (Arctophoca australis gracilis) rookery at Punta Weather in Guafo Island (43°36'S, 74°43’W), the causes and extent of pup mortality were monitored. During four breeding seasons, daily counts of live and dead pups were carried out to determine pup production and pup mortality. Dead pups were retrieved from the rookery to perform necropsies. The mean pup production was 1,735.5 ± 336 pups and the mean pup mortality up to 12 wk old was 6.0%± 2.6%. The major causes of death were enteritis with microscopic lesions of bacteremia (28.4%), starvation (23.5%), drowning (21%), trauma (19.8%), and stillbirths (2.5%). Enteritis with microscopic lesions of bacteremia, and starvation had higher incidence during January (beginning and middle of the breeding season) while most trauma and drowning occurred during February (end of the breeding season). In the 2006–2007 breeding season there was an increase in mortality due to starvation and trauma. Most pup deaths at Guafo Island are generated by extrinsic factors; therefore, additional studies that assess the impact of environmental changes and fishing activities, are needed in order to determine the exact causes of the decline of this species along Chilean coasts.  相似文献

BACKGROUND: Gastroschisis is a rare congenital defect of the abdominal wall. Its occurrence is noted primarily in the offspring of young mothers who often smoke during pregnancy. The incidence of gastroschisis has been increasing in many countries in recent years. The etiology of gastroschisis is not known. METHODS: Pregnant mice of CD‐1 strain were maintained on 17 and 9% protein diets mixed with deficient, normal, or supplemental zinc levels throughout gestation. The dams in each protein‐zinc diet group were randomly divided in two groups. One group was exposed to air (control) and the other to 500 ppm carbon monoxide (CO) in air, in environmental chambers, from gestation days (GD) 8–18. The dams were sacrificed by carbon dioxide asphyxiation on GD 18, and data on malformations was collected. RESULTS: The rates of fetal mortality and malformations were increased by protein and zinc deficiencies. Carbon monoxide exposure also increased fetal mortality. In the low protein group, the rate of fetal mortality was inversely related to the dietary zinc level, and the rate of fetal malformations was highest in the zinc deficient group. The incidence of gastroschisis in the low protein/zinc deficient/CO exposed group was 47%, and 60% of the litters were affected. The incidence of gastroschisis in the rest of the low protein/zinc diets/air or CO groups was 0. CONCLUSION: The data indicates that gastroschisis is caused by the combination of protein‐zinc deficiencies and carbon monoxide exposure during gestation. The finding may be relevant to human populations that experience protein and zinc deficiencies during gestation, and are exposed to CO pollution, or cigarette, or marijuana smoke during pregnancy. Offspring birth mass and growth rate represent important life history traits, which influence many vital population and individual characteristics, while offspring survival is a key factor in variation in female reproductive success. For a threatened population of pinnipeds, such as New Zealand sea lions, Phocarctos hookeri, (Grey, 1844, NZ sea lions), understanding individual life history parameters and population dynamics is vital for their management and conservation. This is the first study of the behaviour of females during parturition, pup birth mass and growth, and pre-weaning survival of NZ sea lions, Enderby Island, Auckland Islands during austral summer breeding seasons, 2001/2002 to 2003/2004. Pregnant females arrived ashore 2.1 ± 0.16 days prior to giving birth. After parturition, mothers suckled their pups for 8.6 ± 0.16 days before leaving on their first foraging trip. Male pups were born significantly heavier than female (males 10.6 ± 1.4 kg, females 9.7 ± 0.9 kg). Pups lost on average 48 ± 0.14 g per day mass during the early postpartum period (between birth and mothers first foraging trip). Pup mortality did not vary by pup sex, birth mass, date of birth or any maternal characteristics however it varied significantly between years due to a bacterial infection epidemic (Pup mortality at 60 days: 2001 32%; 2002 21%; 2003 12%). The absolute growth rate per day for pups was 151 g/day over all years. Pup growth rate measured as the slope of linear line fitted to pup mass by age was consistently higher for pups with heavier birth mass, male pups and during the 2002 season. High offspring mortality and slow growth rates coupled with maternal foraging behaviour at their physiological limits may reflect a threatened species which has limited ability for population growth in an environment which is at the extreme of their historical range and impacted upon by fisheries.  相似文献

Surveys were undertaken at Campbell Island / Motu Ihupuku during January and February 2008, to determine the distribution and pup production of New Zealand sea lions (NZ sea lion; Phocarctos hookeri). In addition, necropsies were performed at the main breeding site of Davis Point to determine the principal causes of early mortality for NZ sea lion pups. In total, 397 pups were tagged and 186 untagged pups were found dead, giving a minimum pup production of 583 pups and a one month of age mortality estimate of 40%. This represents a higher pup production than previous estimates from Campbell Island (although survey techniques are not comparable), and equates to 21% of the total pup production for NZ sea lions in the 2007/08 season. Early pup mortality was high (40%) at Campbell Island, with trauma, starvation, and drowning in rock pools and peat mires the major causes of death. Pups were concentrated in two colonial breeding sites: Davis Point on the north shore of Perseverance Harbour (76%) and a newly recorded breeding site (Paradise Point) on the southern shore of Perseverance Harbour (21%). Non-colonial breeding or single pups occurred around the southern parts of the island from sea level to 400 m; however, these only contributed 3% of the known pup production.  相似文献

Dietary protein and zinc deficiencies known to be detrimental to the developing fetus are common in pregnant women in developing countries. Everyone in modern society is at risk of exposure to carbon monoxide (CO). This study was conducted to observe the effect of dietary protein, zinc, and exposure to CO on the fetal zinc concentrations by factorial experimentation. Pregnant mice of CD-1 strain were maintained on 17% (control) or 9% (deficient) protein diets mixed with deficient, normal (control), or supplemental zinc throughout gestation. The dams in each dietary group were exposed to air (control) or 500 ppm CO in air in environmental chambers from gestation day 8 to gestational day 18. The dams were sacrificed on d 18 and fetal zinc levels were measured by atomic absorption spectrophotometry. Carbon monoxide levels used in this study had no significant effect on fetal zinc concentration in any treatment group. When both dietary protein and zinc levels were normal, the mean fetal zinc concentrations were higher than all other dietary protein/zinc combinations (15.2±6.0 and 14.2±4.1 μg Zn/g of tissue for 0 and 500 ppm CO levels). However, when dietary protein levels were deficient, supplemental zinc increased the fetal zinc concentrations significantly (12.7±3.8 and 13.1±0.3.6 μg Zn/g of tissue, in 0 and 500 ppm CO groups) as compared to zinc-deficient groups (8.7±3.0 and 10.0±3.3 μg Zn/g of tissue in 0 and 500 ppm CO groups). The results of this study may be relevant to populations that experience both marginal zinc and protein diets during gestation.  相似文献

We conducted a study on nest mortality of an individually marked population of European rabbits (Oryctolagus cuniculus L., 1758) living in a field enclosure. Over 4 years, we determined maternities and quantified pup mortality during the nest period of 703 pups born in subterranean breeding burrows. Overall, pup mortality occurred in 42.7% of the litters, whereas 32.4% of all born pups were affected. Mortality was highest during the first few postnatal days. In about 50% of the cases, we managed to quantify different causes of mortality such as malnutrition, flooding, cooling of the pups, infanticide, or predation. The pups’ body mass on postnatal day 1, the thermal environment and the number of litter mates were the most important predictors of nest mortality. Litter mortality risk decreased with increasing soil temperature around the subterranean nests. A comparatively higher average pup body mass lowered the nest mortality risk of a litter, whereas this effect was more pronounced when soil temperatures were low. Furthermore, mortality was lowest in medium-sized litters, most probably due to the balance between the thermal benefits of huddling with litter siblings and the costs of having them due to the lower share of milk obtained by the individual pups in larger litters. In addition, nest mortality depended on characteristics of the mother; mortality was increased in litters of low-ranking females and of mothers with lower body mass. In conclusion, our study highlights multiple causes and the effects of different environmental and social factors on nest mortality of this small mammal.  相似文献

Pre- and early post-weaning pup mortality of southern elephant seals (Mirounga leonina) at Marion Island from 1990 through 1999 ranged from 1.6% to 7.3% and averaged 3.8%. Mortality of pups after weaning before their first trip to sea accounted for only 12% of the total mortality. We found no relationship between population size and percentage pup mortality, indicating that pup survival is independent of seal density, at least at the densities of breeding seals that prevailed. Indeed, pup mortality was greatest in the smallest harems, apparently owing to a greater number of younger, less experienced mothers. Small harems were generally also found on less suitable beaches than larger harems and this could have contributed to pup injury as a cause of pup mortality on these beaches. Mother-pup separation and injury caused by beachmasters is likely to be responsible for pup mortality in the larger harems. The low rates of pup mortality observed in this study obviate it being a major population regulating agent at Marion Island.  相似文献

Human and animal studies suggest a poorer outcome in the presence of abnormal blood glucose concentration during cerebral hypoxia-ischemia. It is unknown whether this is also the case in acute severe carbon monoxide poisoning. Using Levine-prepared rats, three groups were established and exposed to CO to answer this question: (1) hyperglycemics resulting from the administration of a 50% glucose solution, (2) hypoglycemics resulting from the administration of normal saline, and (3) untreated controls. The rats inhaled 2400 ppm CO for 90 min in the absence of anesthesia. Blood glucose was raised to a mean value of 402 mg/dL just prior to CO exposure in group 1. This resulted in an increased mortality rate (i.e., 54%), and during 4 h of room air recovery an impaired ability to regain body temperature, an increased plasma lactate dehydrogenase activity, and an increased neurologic deficit as compared with group 3. Hypoglycemia, which developed during CO exposure in group 2 (mean minimum glucose after 90 min, 44 mg/dL), resulted in an increased mortality rate (i.e., 46%), and during 4 h of room air recovery an impaired ability to regain body temperature and an increased neurologic deficit as compared with group 3. Blood glucose concentration in the rats in groups 2 and 3 that died during or shortly after CO exposure was significantly depressed relative to the survivors of those groups. Plasma insulin activity was elevated during CO exposure in group 1 as compared with group 3, but fell during recovery; insulin remained low throughout CO exposure and recovery in group 2. The results demonstrate the deleterious effects of both a very high and a very low blood glucose concentration during acute CO exposure.  相似文献

Bitopertin is a glycine type 1 (GlyT1) inhibitor intended for the treatment of psychiatric disorders. The principle adverse effect in the regulatory reproductive toxicity studies was peri‐natal pup death when rat dams were treated during parturition at a dose resulting in five‐times the human therapeutic exposure (AUC). Cessation of dosing two days before parturition prevented the pup deaths. Investigatory experiments and pharmacokinetic modelling suggested that the neonatal mortality was related to transplacental passage of bitopertin leading to high systemic levels in the newborn pups. Brain levels of bitopertin in the rat fetus and neonate were two‐fold higher than in the mother. As illustrated by knock‐out mice models, GlyT1 function is essential for neonatal pup survival in rodents, but is not necessary for normal prenatal morphological development. The glycine transport systems are immature at birth in the rat, but are functionally well‐developed in the human newborn. While the relevance to humans of the neonatal mortality seen in rats following late gestational exposure is unknown, bitopertin would not be recommended for use during late pregnancy unless the anticipated benefit for the mother outweighs the potential risk to the newborn.  相似文献

Pup production on Sable Island, Nova Scotia, has been increasing exponentially since the early 1960s and by 1997 Sable Island was the largest gray seal colony worldwide. Using an aerial photographic survey, as in previous years, we estimated pup production in January 2004 to determine if this exponential rate of increase had continued. A total of 33,268 pups was counted on the color positives. When corrected for the proportion pups missed on the imagery (1.106 for the 12th; 1.527 on the 13th), the proportion of pups that died prior to the survey (0.031), and the proportion of pups born before the survey (east colony 0.966, west colony 0.962), estimated total pup production was 41,500 with SE = 4,381. The 2004 estimate indicates that pup production on Sable Island has continued to increase, but suggests that the rate of increase ( r ) may have declined (0.070 compared to previous 0.128). Females from the 1998–2000 cohorts were about 16 times less likely to give birth for the first time at age 4 yr and more than twice as likely at age 6 yr compared to those in the mid-late 1980s. The new estimate of pup production and observed changes in age of primiparity provide the first indication of changes in vital rates of this population. However, additional estimates of pup production and vital rates are needed to confirm this conclusion and to investigate the underlying mechanisms.  相似文献

The extent and causes of pup mortality in the Antarctic fur seal, Arctocephalus gazella , were investigated at sites of high and low density at South Georgia. Mortality was greater at the high density site, (17—31% of annual pup production) than at the low density site (36%). The main causes of death, starvation and skull injury, occur more frequently at the high density site. Most starvation was caused by failure of the mother-pup bond to form, often caused by disturbance induced by the activities of breeding bulls. In some seasons starvation of older pups may be influenced by reduced food availability but this was usually of minor importance. Injury to the skull resulted from bites inflicted by females either accidently during birth or when pups tried to suck from females other than their mother. Trampling of young pups by bulls was probably responsible for the appreciable incidence of ruptured livers. Infectious disease and drowning played minor roles in pup mortality. Pups born late in the season suffered disproportionately greater mortality which may relate to female age and condition. Food availability (both during and prior to the breeding season) and weather are likely to account for year to year variation in pup mortality rates but the basic rate is primarily determined by breeding density. Further population increase and colonization of new beaches is expected until food resources during the summer, or more probably the winter, become limiting.  相似文献

BACKGROUND: Diethanolamine (DEA), a widely used surfactant, was administered to pregnant mice at the oral LD10 resulting in failure of pups to grow and thrive through postnatal day (PND) 3 [National Toxicology Program, 1987; York et al., Teratology 37:503-504, 1988]. The toxicity profile for DEA differs among rodent species. This study investigated DEA-induced postnatal toxicity in a second species. METHODS: Timed-mated Sprague-Dawley rats were dosed (0, 50, 125, 200, 250, or 300 mg DEA/kg/day, p.o.) on gestational days (GD) 6-19. Dams and pups were monitored for body weight, feed/water intake, clinical signs, litter size, and sex ratio. At necropsy (PND 21), maternal liver and kidney weights and number of uterine implantation sites were recorded. RESULTS: The high-dose group was terminated early due to excessive toxicity. The estimated maternal LD10 was 218 mg/kg/day. Maternal effects included decreased body weight and relative feed intake (>or=200 mg/kg/day), transiently reduced relative water intake (125 and 250 mg/kg/day), and increased absolute kidney weight (>or=125 mg/kg/day). Postimplantation loss (PND 0) and pup mortality (PND 0-4) were increased (>or=200 and >or=125 mg/kg/day, respectively). Pup body weight was reduced (>or=200 mg/kg/day) as late as PND 21. CONCLUSIONS: This study demonstrates reduced postnatal growth and survival in a second species after gestational exposure to DEA, persistence of toxic effects through the end of lactation, possibly due to long elimination half-life, and maternal and developmental toxicity no-observed-adverse-effect level (NOAELs) (50 mg/kg/day) and lowest-observed-adverse-effect level (LOAELs) (125 mg/kg/day) for oral DEA exposure during embryo/fetal development in the rat.  相似文献

The relationship between northern fur seal pup weight and post-weaning survival is examined. Males that survived at least two years after weaning had been significantly heavier than the mean for their cohort as pups. Among a small sample of females no significant relationship was found. Cohort survival from weaning to age two was not significantly correlated with mean pup weight. Pup weight may influence post-weaning survival less than other factors such as gender and variability of the environment. To the degree that it determines survival, pup weight may reflect the reproductive success or fitness of parents. Weight differences between tagged and untagged pups explain, in part, differences observed in their survival.  相似文献

The aim of this study was to analyze zinc (Zn), calcium (Ca) and phosphorus (P) contents in milk and the lactational performance in rats fed different Ca levels. Female Wistar rats were fed during pregnancy and lactation with experimental diets containing 20% protein and high (0.90%, HCa), normal (0.60%, NCa) or low (0.20%, LCa) Ca levels. Milk samples were collected after 15 days to determine the milk mineral composition. Pup weight was recorded from birth to weaning (litter size: 6-8 pups) to determine weight gain and calculate milk production. At delivery there were no significant differences in the body weight of the pups between the groups, but at day 15, the LCa group showed lower values than both NCa and HCa groups (p<0.05). The weight gain of the LCa group was significantly lower than of the HCa and NCa groups, between delivery and day 5 (p<0.05). This reduced rate of weight gain led to the LCa group reaching weaning weight later than the other groups. Milk production (g/pup/day) was significantly lower when dams were fed the LCa than the NCa and HCa diets (p<0.05). There were no significant differences among the groups in milk Ca, P and Zn levels and Ca/P ratio. The body mineral composition of the pups at birth did not differ between the groups; at weaning, however, both LCa and HCa groups had lower element contents than the NCa group (p<0.05). In conclusion, dams fed with a diet containing low Ca levels produced smaller volumes of milk and their pups reached weaning weights later than the other groups. As the milk mineral composition was not affected, it can be hypothesized that in dams fed low dietary Ca, the smaller milk yield might have been a way of maintaining milk quality. High Ca levels affected neither pregnancy outcome nor lactational performance.  相似文献

We surveyed pup survival in Mediterranean monk seals ( Monachus monachus ), at Cabo Blanco Peninsula (Western Sahara-Mauritania) colony from May 1993 to December 1997. This species breeds and hauls out on beaches inside two main caves. During the study period we detected a total of 93 pups that died or disappeared. The survival rate of 84 pups through the age of first moult (approximately two months) was 0.47. This value is similar to those reported for other pinnipeds breeding in caves but lower than for those breeding on open beaches. Mortality varied seasonally and appeared to increase as a result of storms, large ocean swells, and high tides. Mother-pup pair separation (and resulting pup starvation) and physical injury caused by impact against the rock walls of the cave and cliffs were established as the causes of most deaths. Beach surface area inside the caves also appeared to be a mediating factor in the effects of sea conditions. High pup mortality may be a limiting growth factor in this population, although cave dwelling protects the population from predators and human disturbance.  相似文献

OBJECTIVE: To investigate the association of dietary pattern and mortality in international data. DESIGN: Cohort study with 20 years'' follow up of mortality. SETTING: Five cohorts in Finland, the Netherlands, and Italy. SUBJECTS: Population based random sample of 3045 men aged 50-70 years in 1970. MAIN OUTCOME MEASURES: Food intake was estimated using a cross check dietary history. In this dietary survey method, the usual food consumption pattern in the 6-12 months is estimated. A healthy diet indicator was calculated for the dietary pattern, using the World Health Organisation''s guidelines for the prevention of chronic diseases. Vital status was verified after 20 years of follow up, and death rates were calculated. RESULTS: Dietary intake varied greatly in 1970 between the three countries. In Finland and the Netherlands the intake of saturated fatty acids and cholesterol was high and the intake of alcohol was low; in Italy the opposite was observed. In total 1796 men (59%) died during 20 years of follow up. The healthy diet indicator was inversely associated with mortality (P for trend < 0.05). After adjustment for age, smoking, and alcohol consumption, the relative risk in the group with the healthiest diet indicator compared with the group with the least healthy was 0.87 (95% confidence interval 0.77 to 0.98). Estimated relative risks were essentially similar within each country. CONCLUSIONS: Dietary intake of men aged 50-70 is associated with a 20 year, all cause mortality in different cultures. The healthy diet indicator is useful in evaluating the relation of mortality to dietary patterns.  相似文献
