Acid phosphatase activity was present in unimbibed barley seed,but rose during incubation of embryoless half-seeds and isolatedaleurone layers, and was further increased by 10–6 M gibberellicacid (GA3). Release of total acid phosphatase activity fromhalf-seeds and aleurone layers was markedly enhanced by GA3.Inhibitor studies with cycloheximide and actinomycin D suggestedthat de novo synthesis of acid phosphatase occurred followingimbibition. Gel nitration, electrophoresis, and [14C]leucineincorporation studies revealed that a single molecular formof acid phosphatase was present in dry seed, whereas on incubationtwo further forms arose. A proportion of the three molecularforms of the enzyme was synthesized de novo. Gibberellic acidstimulated activation, but not de novo synthesis, of all threemolecular forms of acid phosphatase. Although a small amountof one of the molecular forms was secreted in the absence ofGA3, the presence of gibberellin greatly increased secretionof the same form of acid phosphatase.  相似文献

In Pisum sativum L. a third, more severe, allele at the internodelength locus le is identified and named led. Plants homozygousfor led possess shorter internodes and appear relatively lessresponsive to GA20 than comparable le (dwarf) plants. Gene ledmay act by reducing the 3ß-hydroxylation of GA20 tothe highly active GA1 more effectively than does gene le. Theresults indicate that le is a leaky mutant and therefore thatendogenous GA1 influences internode elongation in dwarf (le)plants. Oryzains, cysteine proteinases of rice seeds, are induced byGA3 in germinating rice seeds [Abe et al. (1987) Agric. Biol.Chem. 51: 1509]. The effects of GA1, GA3, GA4, GA9, and GA20on the production of oryzain and -amylase were investigatedin embryoless half- and whole-seeds of rice (cv. Nipponbare).When gibberellins (GAs) were incubated with embryoless half-seeds,GA1, GA3 and GA4 induced oryzain and -amylase, but GA9, andGA20 did not. GA9 and GAM induced oryzain and -amylase productionin whole seeds, but this production was inhibited by the simultaneousapplication of prohexadione, an inhibitor of 2ß- and3ß-hydroxylation of GAs. Prohexadione did not inhibitthe activities of oryzain and -amylase induced by GA1. Theseresults suggest that GAs possessing the 3ß-hydroxylgroup induce activities of oryzain and -amylase in rice seedsand that GA9 and GA20 have activity only after they are convertedmetabolically to active GAs, probably GA4 and GA1, respectively.GA1, was more active than GA4 in both half seeds and wholeseeds incubation. Oryzain and -amylase activities induced byGA4 were significantly inhibited in the presence of 10–4M prohexadione. Barley (Hordeum vulgare L. C.I.666) was shown to be susceptibleto the growth retardant (2-chloroethyl)-trimethylammonium chloride(CCC). The estimation of cell number in the dwarfed third leafblade indicated that a decrease in mitotic activity had occurredin treated plants. There was also a decrease in cell size intreated plants. The dwarfing action of CCC was reversed by exogenousgibberellic acid (GA3) but this was shown to be the result ofincreased cell elongation only. GA3 did not promote cell divisionin healthy or CCC-treated plants. Assay of endogenous gibberellinsshowed a significant reduction in the level of a substance correspondingto GA3 in CCC-treated plants. It is suggested that CCC-induceddwarfing of barley is largely the result of a reduction in meristematicactivity. This may be related to an effect on gibberellin biosynthesisbut is not reversed by the application of exogenous GA 3.  相似文献

The effect of PGE1 and PGE2 on the induction of acid phosphataseactivity in embryoless barley half-seeds was examined. Thesecompounds were found to promote the total amount of enzyme synthesizedas well as the amount released into the medium to nearly thesame levels obtainable when these seeds are incubated in GA3or cyclic AMP. The enzyme induction by PGE1 and PGE2 was inhibited by cycloheximide,ABA, 6-methyl purine and acetylsalicyclic acid, but not by AMO-1618.A time course for the induction of acid phosphatase activityby PGE1 and PGE2 lagged behind by 9 hr a similar time courseobtained when seeds were incubated in GA3 or cyclic AMP. The biological activity of a cell wall-derived xyloglucan nonasaccharide(XG9) was investigated using a bioassay with entire pea epicotyls(Pisum sativum cv. Progress). The xyloglucan fragment was foundto inhibit gibberellic acid-induced elongation of etiolatedpea epicotyls with maximum inhibition at concentrations rangingfrom 10–11 to 10–9M. Growth of etiolated epicotylsin the absence of exogenously applied GA3 was also inhibitedby XG9 in the same concentration range. A cell wall-derivedheptasaccharide (XG7) lacking the fucosyl-galactosyl-side chainshowed no inhibitory effect in the pea epicotyl bioassay withand without exogenous GA3. Furthermore, the biological activityof a synthetic pentasaccharide (XG5), containing the fucosylgalactosyl-sidechain which is necessary for the biological activity was investigatedin the same bioassay. Compared to XG9 the pentasaccharide hada similar inhibitory activity on GA3-promoted elongation aswell as on the endogenous growth in the absence of exogenouslyapplied GA3, but did not exhibit a distinct concentration optimum. Cell-free extracts were prepared from anthers of normal anddwarf rice (Oryza sativa L.), and the metabolism of radioisotope-labeledgibberellins in the extracts was analyzed by HPLC and gas chromatography-massspectrometry (GC/MS). GA12 was converted to GA15 and GA34 inthe extracts. GA20 was converted to GA1, GA8 and GA29, but GA9was converted only to GA34. The extracts of the dwarf cultivar,Waito-C (dy mutant), showed the same 3ß-hydroxylationactivity as did those of the normal cultivar, Nihonbare, indicatingthat the dy gene is not expressed in the anthers. A 24 hr preincubation UV (254 nm) irradiation of RS-ABA andGA3 solutions completely negated the inhibitory effect of ABAon GA3 induced -amylase production with barley half-seeds. Thisirradiation, however, caused no significant inhibition of GA3-induced-amylase production. Longitudinal microtubules are predominant in epidermal cellsof the 3rd internodes of dwarf pea (Pisum sativum L. cv. LittleMarvel) seedlings. In more than 50% of the cells, cortical microtubulesare running parallel to the cell axis. GA3 promotes elongation of the internodes and gives rise toa predominance of transverse microtubules. In more than 60%of the GA3-treatd cells, cortical microtubules are running transverseto the cell axis. Longitudinal microtubules in the GA3-untreated cells are resistantto low-temperature treatment, but transverse microtubules inthe GA3-treated cells are sensitive to it. Longitudinal microtubulesare present in GA3-treated epidermal cells with low frequency.They are resistant to low-temperature treatment. Longitudinal, oblique and transverse microtubules are presentwith almost the same frequency in epidermal cells of the 3rdinternodes of tall pea (cv. Early Alaska) seedlings. GA3 promoteselongation of the internodes also in tall pea seedlings, butit does not alter the direction of cortical microtubules sodistinctly as it does in dwarf pea seedlings. As in dwarf pea seedlings, longitudinal microtubules are resistantto low-temperature treatment, and transverse microtubules aresensitive to it in tall pea seedlings. A Crucial Role for Gibberellins in Stress Protection of Plants   总被引:2,自引:0,他引:2  
The hypothesis that there is an intimate relationship betweengibberellin levels and plant stress protection has been testedusing near-isogenic lines of a normal and dwarf barley (Hordeumvulgare L.). Application of paclobutrazol (a triazole), inducedstress protection in the normal line and application of GA3to the dwarf reversed the inherent stress tolerance. Reversalof the dwarf phenotype by specific gibberellins (GAs) suggeststhat the conversion of GA20 to GA1 and GA9 to GA4 has been compromised.These observations indicate that modulation of specific GAsplays a key role in stress protection. The preferential useof non-chemical technologies including enzyme regulation, phytochromeA overexpression, and breeding for induction of stress tolerancein plants are discussed. Effects of kinetin (K), gibberellin A3 (GA3), and 2-(chloroethyl)-trimethylammoniumchloride (CCC) on levels of alanine aminotransferase (GPT) andrates of protein synthesis were studied with both intact plantsand isolated leaf segments of Lolium temulentum L. In intactplants CCC stimulated and CA3 reduced GPT activity, the effectsbsing much greater in 8.h than in 16-h photoporiods. CCC showedmaximum stimulatory effects at 10–2 M and K at 5 x 105M. No effect of GA3 could be demonstrated with concentrationsup to 10–4M. Both K and CCC retarded GPT decline in leafsections, the latter without associated effects upon pigmentbreakdown. Cycloheximide was highly effective in reducing proteinsynthesis in leaf sections. A close correlation between rateof protein synthesis and GPT activity was found over an inhibitorconcentration range from 10–6 to 10–4 M. The resultsare discussed in terms of possible methods of in vivo regulationof GPT activity.  相似文献

The presence of GA9, GA19 and GA20 was demonstrated by gas chromatography/massspectrometry (GC/MS) and the presence of GA44 strongly indicatedby GC/MS in selected ion monitoring mode (GC/SIM) in extractsof shoots of light-grown tall peas (Pisum sativum L.). Usingthe rice seedling bioassay with cv. Tan-ginbozu, the levelsof gibberellins in pea shoots were monitored from early shootgrowth through to apical senescence in a tall pea line. Levelsof activity corresponding to GA20, GA19 and GA44 remained relativelystable in the shoot despite reproductive development and apicalsenescence. The level of GA1-like activity increased to a maximumwhen the leaves had between 7 and 9 leaves expanded and decreasedonly with apical senescence. The na gene which blocks the productionof biologically active gibberellins in shoots but not in developingseed, was also operative in pod walls, with na pods containinglittle or no significant gibberellin-like activity when comparedto na pods at contact. This occurred despite the presence ofrelatively high levels of gibberellins in developing seed atthe same time. The results suggest that there is little or nosignificant leakage of biologically active gibberellins fromdeveloping seed to pods or shoots. Extracts of pods of tallpeas with Na contained low levels of gibberellin-like activitybut like developing seed, contained little or no significantGA1-like activity despite the presence of significant GA1-likeactivity in shoot extracts of tall peas. Effects of GA3 and CCC application on the sprouting of bulbilsor subterranean dormant organs of 10 species in the genus Dioscoreawere observed. Although the efficiency of both chemicals differedby species, in general GA3 inhibited and CCC promoted the sproutingof the above dormant organs. In some species, however, dilutedGA3 (0.003–0.3 µM) has a promotive and diluted CCC(3–30 µM) has an inhibitive effect on sprouting. Effects of GA3 application on shoot elongation were tested onsprouted bulbils. GA3 promoted elongation when applied directlyto the shoots and inhibited it when applied to the bulbous parts. These results suggest that GA activates two opposing reactions—sprouting-promotingand sprouting-inhibiting—in these organs. Changes in the contents of phytate (IP6) and other phosphorus(P)-compoundsin germinating seeds of a huskless barley were investigatedin the embryo with scutellum (EM), the starchy endosperm (EN),and the aleurone layer with pericarp-testa (AL). More than 80%of the total P in the AL of 1-day germinated seeds was foundin acid-soluble organic P, most of which was IP6. During germination,the IP6 in AL decreased markedly with no accumulation of lessphosphorylated myo-inositols and Pi and acid-insoluble organicP increased in the EM. The total P in the EN of 1-day germinatedseeds was about one-third that in the AL, the greater part ofwhich was found in the acid-insoluble fraction and decreasedgradually during germination. Only a small amount of IP6 couldbe detected in the EM and EN during the early stage of germination. IP6 in AL of embryoless half-seeds incubated without gibberellicacid (GA3) decreased slightly even after 6 days. Incubationwith 10 ppm GA3 remarkably stimulated the IP6 degradation. Thisstimulation was reduced, with no change in the Pi content, byabout 80–90% with 1 mM 6-methylpurine or 10 ppm cycloheximide.The addition of 0.1 M KH2PO4 caused a 4-fold increase in thePi content of AL in the presence of GA3. In addition, it suppressedthe GA3-dependent -amylase synthesis by about 20% and the GA3effect on IP6 degradation by about 50%. In light of these results, IP6 seems to be hydrolyzed completelyinto Pi and myo-inositol within the aleurone tissue, and gibberellinseems to control this process. The synthetic methyl ester of GA73 (GA73-Me) and the naturalantheridiogen of Lygodium japonicum showed almost the same activityto induce the formation of antheridia in dark-grown protonemataof L. japonicum at concentrations of 10-14 M and higher. Thus,it appears that the principal antheridiogen in L. japonicumis GA73-Me. GA73-Me inhibited formation of ar-chegonia in light-grownprothallia of L. japonicum at concentrations of 10-11 M andhigher and induced germination of spores in the dark in thisspecies at the same range of concentrations. GA73(free acidform) promoted growth of seedlings of dwarf rice and hypocotylsof cucumber seedlings at dosages of and above 1 and 100 ng/plant,respectively. Eight compounds related to GA73-Me, includingantheridiogens of Anemia phyllitidis and Anemia mexicana, wereactive in inducing an-theridial formation in L. japonicum, althoughtheir activities were considerably lower than that of GA73-Me. Summary Pea seedlings (cv. Alaska), were treated with two concentrations of (2-chloroethyl)trimethylammonium chloride (CCC) and choline chloride. Treatment with 1 mg/l CCC resulted in as much as a 150fold increase in endogenous gibberellin (GA) levels without there being any parallel stimulation of growth. Plants grown in 1,000 mg/l CCC were severely dwarfed but contained GA levels not significantly different from control plants grown in distilled water. CCC also retarded GA3-induced growth of pea seedlings. These effects appear to be CCC specific as the CCC analogue choline chloride affected neither the GA content of pea seedlings nor their response to GA3. The lack of correlation between endogenous GA levels and stem height suggests that in peas the predominant factor in CCC-induced inhibition of stem growth is not related to an effect of CCC on GA biosynthesis.Supported by National Research Council (Canada) grant A-5727.Supported by a Postdoctoral Fellowship from NRC Grant A-2585 to R.P. The effects of a 24 hr short day, a 24 hr long day, and a 48hr short day were analyzed with regard to flowering and stemgrowth of normal and dwarf Pharbitis nil, and were comparedto effects of these photoperiodic treatments plus applied GA3.Both short day treatments produced the same number of flowersper plant after seven cycles. The applied GA3 was effectivein overcoming the growth deficiency of the dwarf; however, theextended dark period of the 48 hr short day and applied GA3were both required to enhance a flowering response in the dwarfequal to that of the normal. Pure samples of the antheridiogen of Anemia phyllitidis (AAn) were tested for their ability to affect the growth of dwarf corn (d5) and lettuce seedlings, and to influence α-amylase production by barley half-seeds. Stimulation of dwarf corn growth and barley amylase production was, on a molar basis, from 1/2 to 1/250 that given by GA3. In lettuce, AAn had a synergistic effect with low levels of GA3; alone, AAn was inhibitory or ineffective. Therefore, in addition to having a close chemical resemblance to gibberellin, AAn induces similar, but not identical physiological responses in flowering plants as well as ferns.  相似文献

The kinetics of extension induced by GA31 in the hypocotyl ofintact seedlings of Lactuca sativa are similar in the dark andin the light, and differs fundamentally from the kinetics ofelongation in the dark without GA3. Both in continuous lightand in the dark, GA3-induced promotion starts 24 h after incubation.In the dark, even low concentrations of GA3, which do not affectthe length measured after 6 d when the extension of hypocotylalmost ceases, remove the lag period of 48 h which precedesextension, and prolong the high rate of elongation. FollowingGA3 supply the hypocotyl length in the dark and in the lightdoes not differ until 48 h; thereafter the rate of elongationin the light is less, so that the final length of the hypocotylis 40 per cent shorter than that of the dark-grown seedlingswithout GA3. IAA supplied apically to light-grown seedlings induces a weakpromotion at a concentration of 1 mg l–1 only. With anincreasing concentration of GA3 supplied simultaneously, theconcentration of IAA inducing a significant promotion decreases.A combined supply of both these regulators, however, does notrestore the light-mediated inhibition of hypocotyl elongationcompletely. The maximum decrease in hypocotyl length induced by the growthretardants AMO-1618, CCC, and B-9 supplied from the beginningin the dark does not exceed 70 per cent. Saturating doses ofGA3 supplied in combination with any one of the retardants compensateonly a fraction of the decrease. The results have been interpreted to show that native GAs arenot involved in extension growth in the dark.  相似文献
