Incorporation of polyamine-spermidine into the nutrient mediumat 10–6 and 10–5 M concentrations stimulates pollen-tubegrowth in vitro in Catharanthus roseus L. G. Don. MGBG, an inhibitorof spermidine biosynthesis, at 0.5 x 10–3 and 1 x 10–3M concentrations reduced the percentage of germination as wellas tube growth and at a concentration of 1.5 x 10–3 Mgermination was totally inhibited. Pollen grains incubated inthe medium containing 1.5 x 10–3 M MGBG, when transferredto a fresh medium with 10–5 M spermidine, resulted in80% germination recovery, along with considerable tube growth.Experiments with actinomycin-D indicate that stimulation ofpollen-tube growth by spermidine may involve de novo synthesisof protein. Feeding behavior of the brittle star Ophiura ophiura includesorienting posture, orienting movements, arm ‘walking’,changing the direction of ‘walking’ arm coilingand ingestion. All sequential behavior patterns were releasedor enhanced by single low-molecular-weight compounds. Stimuliwhich released ‘walking’ behavior at high concentrations(10–4 M) in all the test animals are listed in decreasingorder of sensitivity: sarcosine, glycine, urea, L-valine, L-leucine,L-methionine, L-homocysteine, L-norvaline, L-norleucine, L-threonine,L-serine, S-methyl-L-cysteine, L-proline. Threshold values forsingle amino acids were as much as 100 times different in differentindividuals and ranged from 3 x 10–9 to 3 x 10–7M for the most effective stimulus, sarcosine, and from 10–6to 10–4 M for proline. Above 10–5 M, only L-prolineregularly released a second behavior pattern, the arm coilingresponse, which temporarily inhibited the ‘walking’behavior. Behavioral thresholds for the ‘walking’behavior for L(+)-lactate and L-alanine were higher than thosefor the orienting movements. Thyoglycolic acid and ß-alaninereleased tube feet walking, which is not part of the feedingbehavior. Structure—activity comparisons were studied at estimated10–5 M concentrations. Gycline, sarcosine, L-valine, L-norvaline,L-leucine, L-isoleucine, DL-norleucine and DL-homocysteine releasedarm ‘walking’ behavior in more than 75% of all thetests. With the exceptions of S-methyl-and S-ethyl-cysteine,and glycine methylester, derivatives of amino acids were noteffective behavioral stimuli in Ophiura ophiura. L-Isomers ofvaline and leucine regularly stimulated the ‘walking’behavior while their D-isomers were effective in some testsand ineffective in others. Acetylcholine iodide, acetyl-ß-methylcholine chloride and choline phosphate chloride regularly released‘walking’ behavior at concentrations above 10–5M.  相似文献

The effect of cycloheximide (10–5 M) and cordycepin (10–4M) used as protein and RNA synthesis inhibitors, respectively,on auxin action in noncellulosic ß-glucan degradationof Avena coleoptile cell wall was investigated. Both depressedauxin-induced ßglucan degradation of the cell wallas well as auxin-induced elongation and cell wall loosening,suggesting that the process of ß-glucan degradationof the cell wall is closely associated with cell wall looseningand that auxin enhances the activity of an enzyme for ß-glucandegradation through de novo synthesis of RNA and protein butnot through activation of the enzyme in situ. Kinetic studywith the inhibitors showed that RNA metabolism involved in ß-glucandegradation was stimulated by auxin treatment of only 15 minwhile a longer lag phase (about 1 hr) existed for the synthesisof the enzyme. Gibberellic acid (GA3) at 58 µM, indoleacetic acid (IAA)at 29 µM, and (2-chloroethyl) phosphonic acid (Ethephon)at 70 µM promoted pollen tube growth in Crotalaria junceapollen suspension cultures both in water and basal medium. GA3stimulated [ l-14C]acetate incorporation into total lipids inboth media, whereas IAA enhanced incorporation in water culturesonly. On the contrary, Ethephon reduced the label in total lipidswhen supplemented in basal medium. Based on [l-14C lacetateincorporation into different phospho- and glycolipids, it isproposed that these growth regulators have a definite role inthe biosynthesis of lipid components of the membranes.  相似文献

O-Benzylhydroxylamine (OBHA) is a potent inhibitor of phenylalanineammonialyase (PAL, EC 4.3.1.5 [EC] ) and phenylpropanoid metabolismas evidenced by its effects on three plant species [soybean(Glycine max (L.) Merr.), buckwheat (Fagopyrum esculentum Moench.),and mung bean (Vigna radiata L.)]. When supplied to roots, OBHA(10–5 M) did not significantly inhibit light- or dark-growthof soybean seedlings, but reduced (25%) soluble hydroxyphenoliccompound accumulation in light-grown axes. Higher concentrations(510–5 M) of OBHA caused reductions (25%) in axis freshweight of light-grown seedlings (72 h), but did not lower axisweight of dark-grown seedlings. Anthocyanin accumulation inhypocotyls of intact mung bean seedlings was reduced by 25%after 3 days light growth after treatment with OBHA (10–5M) via root feeding. Anthocyanin content of excised, etiolatedbuckwheat hypocotyls floated on solutions of OBHA (10–5M) and incubated in the light for 24 h was reduced by 40%. L-Phenylalanineand t-cinnamic acid, intermediates of phenylpropanoid metabolism,were able to partially reverse this inhibition in buckwheat.Extractable PAL activity (specific activity basis) in soybeanaxes was substantially reduced (20% in dark, 40% in light) asearly as 24 h after root feeding with OBHA (10–5 M). Reductionof PAL activity (specific activity or per axis basis) by OBHAcompared to control levels, continued throughout a time courseof 96 h. A membrane fraction from flax cells was able to incorporate[14C]galactose from UDP-D-[14C]galactose in vitro. The productsof the reaction, characterized by methylation analysis, consistedof a rß-1,4-galactan (solubilized mainly in water)and a rß-1,3- rß-1,6-galactan (solubilizedmainly in alkali). These results indicated the presence of severalgalactan synthase complexes, as did a profile of the relationshipbetween pH and activity which revealed both a maximum at pH6.5 and a shoulder at pH 8. Moreover, galactan synthase activitieswere found at two densities: 1.125 g cm–3 (Golgi membranes)and 1.07–1.08 g cm–3 (corresponding to low-densityvesicles). Partial characterization of one enzymatic system (maximaly activeat pH 8 in the presence of 5 mM MgCl2) was achieved. The effects of light, auxin and cytokinin on vessel elementformation in phloem slices of carrot root were examined. When slices of carrot cultivars, ‘Nakamura-senko-futo’and ‘Yamada-hyakunichisenk6- naga’, preculturedin the dark on modified Murashige and Skoog's medium for twodays were cultured on a medium containing 5x10–6 M 2,4-Din the dark, no vessel element formation occurred. When preculturedslices were cultured in the light with 5x10–6M 2,4-D,vessel element formation was remarkable. But when 5x10–7Mkinetin, benzyladenine or zeatin was added, vessel elementswere readily formed even in the dark. When slices were cultured in the light, a cytokinin-like substance(s)that causes vessel element formation was produced in the slices,then was released to the medium. The substance(s) was fairlystable to heat. In slices of carrot cultivars, kuroda-gosun, ‘Kintoki’and ‘Kokubu-senk6-6naga’, a different result forvessel element formation was obtained. When slices of thesecultivars were cultured on a medium containing 5x10–6M2,4-D in the dark, vessel element formation was remarkable.It seemed, therefore, that these cultivars contain enough ofa cytokinin-like substance(s) to form vessel elements. The standard synthetic culture medium (Stuart and Street, 1969)has been modified by adjustment of its initial pH to 6.4 andby the addition of gibberellic acid (0.25 mg/l) and of a mixtureof 15 L-amino acids formulated from an analysis of the conditionedmedium. The minimum effective density for the growth of sycamorecell suspensions in the standard medium is 9–15 x 103cells ml–1, for the modified synthetic medium it is 2.0x 103 cells ml–1, and for conditioned medium 1.0–1.25x 103 cells ml–1. Using either conditioned medium (Stuart and Street, 1969) orthe modified synthetic medium it is demonstrated that the growthof cultures initiated at low density is enhanced by a volatilefactor released from actively growing cell suspensions. In presenceof conditioned medium and this volatile factor cultures canbe established from stationary-phase cells at a density of 6x 102 cells ml–1. The volatile factor can be absorbedin 40 per cent w/v KOH but attempts to replace the factor byair containing carbon dioxide at concentrations up to 5 percent have so far been unsuccessful.  相似文献

Activities of - and ß-glucosidase, - and ß-galactosidase,-mannosidase, ß-1,3-glucanase, acid and neutral invertaseswere detected in the cytoplasmic fraction as well as in cellwalls isolated from callus cultures of cotton. Activity of ß-mannosidase,however, could not be detected in the cell walls. Transfer ofcallus to a fresh medium did not immediately influence the activitiesof -glucosidase and ß-galactosidase but increasedsignificantly ß-glucosidase, -mannosidase, acid andneutral invertases. Addition of cycloheximide (1 and 100 mgl–1) further stimulated acid and neutral invertases butnot other enzymes tested. Sodium chloride (NaCl) was effectivein extracting a-glucosidase, ß-glucosidase, ß-galactosidase,acid and neutral invertases. EDTA extracted most of the -galactosidase,-mannosidase, ß-1,3-glucanase and some -glucosidase.But, NaCl and EDTA could not extract some of the - and ß-glucosidasesand also acid and neutral invertases as evidenced from the residualand extra cellular activity. Induction of CN-insensitive respiration with low concentrationsof respiratory inhibitors was studied. If roots were treatedwith 10–3 M CN for 96 hr, the plants died, whilethose treated with 10–4 M CN showed healthy growth. O2 uptake in untreated rice and wheat roots showed a negativeresponse to 10–2 M CN to a considerable extent.On the other hand, pretreatment with 10–4 M CNfor more than 6 hr did not greatly affect respiratory rate,but made respiration insensitive to 10–2 M CN.A similar induction of CN-insensitivity was also broughtabout with 10–4 and 10–3 M H2S and 10–4 MNaN3. Effects of isovitexin (isolated from leaves of Rhynchosia minima),leucocyanidin, gallic acid, and protocatechuic acid on seedgermination and subsequent seedling growth of a crucifer Brassicacampestris, and two legumes Lens esculenta and R. minima, aswell as effects of isovitexin on rooting of onion bulbs aredescribed. Neither of the flavonoids affected seed germinationin any of the three systems studied. However, both the flavonoidspromoted seedling growth in B. campestris and L. esculenta;the promotion of root growth was more marked. At 10–5and 10–7M, isovitexin also promoted rooting of onion bulbs.In the range of 2.27 ? 10–4M to 3.28 ? 10–3M, leucocyanidinpromoted growth of both root and shoot in L. esculenta, whereasin the range 3.28 ? 10–3M to l.64?lO–3M it suppressedseedling growth. In contrast to the flavonoids, the two phenolicacids tested inhibited seed germination at 10–3M, andat lower concentrations they suppressed seedling growth. The promotive effects of the flavonoids isovitexin and leucocyanidin,especially on root growth, hold promise for the use of suchnaturally occurring plant substances in studies on the physiologyof plant growth and development. The NADP$-specific isocitrate dehydrogenase was partially purifiedfrom photosynthetically-grown Rhodospirillum rubrum. The pHoptimum is between 7.5 and 9.0 in phosphate buffer. The apparentKm is 3.1x10–5 M for isocitrate, 5.1x10–5 M forNADP$, 1.7x10–5 M for manganese, 1.5x10–4 M formagnesium, and 3.5x10–3 M for inorganic orthophosphate.Arsenate exerts a slight inhibition. The Q10 between 17.5°Cand 40°C is 1.62, and the energy of activation at 25°Cis 9.74 Kcal/mole. Glyoxylate and oxalacetate cause concertedinhibition of the enzyme activity. Various nucleotides inhibitthe activity. The kinetics of inhibition by ATP was found tobe mixed type with respect to NADP$ and isocitrate, the Ki valuesbeing 1.17x10–3 M and 1.10x10–3 M respectively.The inhibition between ATP and orthophosphate is competitivewith a Ki of 10–4M. Guttation was used as a non-destructive way to study the flowof water and mineral ions from the roots and compared with parallelmeasurements of root exudation. Guttation of the leaves of barley seedlings depends on age andon the culture solution. Best rates of guttation were obtainedwith the primary leaves of 6- to 7-day-old seedlings grown onfull mineral nutrient solution. The growing leaf tissue becomessaturated with K+ below 1.5 mM K+ in the medium, whereas K+concentration in the guttated fluid still increases furtheras K+ concentration in the medium is raised. At 3 mM K+ averagevalues of guttation were 1.4–2.4 mm3 h–1 per plantwith a K+ concentration of 10–20 mM; for exuding plantsthe flow was 4.2–7.6 mm3 h–1 per plant and K+ concentration35–55 mM. Abscisic acid (ABA) at 10–6 to 10–4 M 0–2h after addition to the root medium increased volume flow ofguttation and exudation and the amount of K+ exported. Threeh after addition of ABA both volume and amount of K+ were reduced.There was an ABA-dependent increase in water permeability (Lp)of exuding roots shortly after ABA addition. Later Lp was decreasedby 35 per cent and salt export by 60 per cent suggesting aneffect of ABA on salt transport to the xylem apart from itseffect on Lp. Benzyladenine (5 x 10–8 to 10–5 M)and kinetin (5 x 10–6 M) progressively reduced volumeflow and K+ export in guttation and exudation and reduced Lp. Guttation showed a qualitatively similar response to phytohormonesas found here and elsewhere using exuding roots. The quantitatively major nonasaccharide (XG9) derived from xyloglucanby digestion with cellulase exhibits anti-auxin activity inthe pea stem segment straight-growth bioassay; the most effectiveconcentration of XG9 is c. 10–9 M. Previous work had shownthat XG9 owes its biological activity to the presence of a terminal-L-fucopyranose residue. In order to investigate to what extentthe remainder of the XG9 molecule is essential for activity,several fucose-containing compounds were tested for their abilityto mimic the anti-auxin effect of XG9. A fucose-containing pentasaccharideof xyloglucan (XG5; probable structure FucGalXylGlcGlc) was,at 10–8 M, about as effective an anti-auxin as 10–9M XG9; unlike XG9, XG5 did not diminish in effectiveness at10–7 M. The human milk trisaccharide, 2'-fucosyl-lactose[L-fucopyranosyl--(12)-D-galactopyranosyl-ß-(14)-D-glucose],whose FucGal unit is identical with that of XG9, inhibited auxin-inducedelongation over a wide range of concentrations centred on about10–8 M. 2'-Fucosyl-lactose at 10–8 M was about aseffective an anti-auxin as 10–9 M XG9. Free L-fucose andmethyl--L-fucopyranoside were unable to inhibit auxin-inducedgrowth at any concentration tested (10–10 M to 10–6M) and neither compound interfered with the inhibition causedby 10–9 M XG9 when co-incubated at concentrations up to10–4 M. The results confirm the essential r?le of an -linkedterminal fucose residue in the anti-auxin activity of XG9 andshow that the sub-terminal galactose residue may also be required.Possible reasons why high concentrations of XG9 fail to antagonizeauxin-induced growth while high concentrations of XG5 and 2'-fucosyl-lactosecontinue to do so are discussed. The regulatory role of plant growth substances in relation tosource and sink activity was studied. Leaching of electrolytesfrom flag leaf cells increased with age. Abscisic acid, however,increased the permeability of leaf cells at 10–4 M andlower concentrations. Plant growth regulators like auxin, gibberellinand cytokinin at 10–5 M inhibited the leaching of electrolytesbut slightly enhanced the process at 10–3 M. The effluxof 14C-sucrose loaded on the flag leaves was not detectablein the very early stage of panicle development but was prominentin the later stages. While auxin prevented sucrose efflux atall stages of panicle development, gibberellin and cytokininwere effective only in different degrees. All the three growthregulators enhanced the activity of -amylase in the flag leafcells but protease activity was enhanced only by gibberellin.Synthesis of polymers like starch, DNA, RNA and protein in developingrice grains was stimulated by gibberellin, cytokinin and auxinlargely in the order mentioned. To a certain extent the flowering of Lemna paucicostata 6746is evoked in continuous light by application of abscisic acid(ABA) and CCC. Moreover, the action of the combined substancesappears in two separate concentration ranges. In the range ofABA 2?10–9 M/CCC 10–7–10–6 M floweringis initiated without inhibition of vegetative growth and proceedsonly in the presence of high intensity light and sucrose. Acombination of ABA 2?10–5 M/CCC 10–3M simultaneouslycauses a strong inhibition of frond multiplication. Here theeffect can be observed also under low intensity light conditionsand without sucrose in the medium. Seedlings of cotton (Gossypium hirsutum L. cv. Acala SJ-2) weregrown in modified Hoagland nutrient solution with various combinationsof NaCl and CaCl2. Marking experiments and numerical analysiswere conducted to characterize the spatial and temporal patternsof cotton root growth at varied Na/Ca ratios. At 1 mol m–3Ca, 150 mol m–3 NaCl reduced overall root elongation rateto 60% of the control, while increasing Ca to 10 mol m–3at the same NaCl concentration restored the elongation rateto 80% of the control. Analysis of the spatial distributionof elongation revealed that the presence of 150 mol m–3NaCl in the medium shortened the growth zone by about 2 mm fromthe approximate 10 mm in the control and also reduced the relativeelemental elongation rate (i.e. the longitudinal strain rate,defined as the derivatives of displacement velocity of a cellularparticle with respect to position on root axis). Supply of 10mol m–3 Ca at the high salt condition restored partiallythe relative elemental elongation rate, but not the length ofthe growth zone. Compared to the control, the growth trajectoriesshowed that at 1 mol m–3 CaCl2 it took more time for acellular particle to move through the growth zone at 150 molm–3 NaCl, while at 10 mol m–3 CaCl it took lesstime and there was no difference between the NaCl treatments Key words: Gossypium hirsutum, salinity stress, root growth kinematics  相似文献

Pine (Pinus roxburghii) pollen grown in suspension cultureswas used to study the effects of growth regulators and lightconditions on germination and pollen tube growth. Indol-3-ylacetic acid, gibberellic acid, ethylene, abscisic acid and cyclicAMP (cAMP) at low concentrations (1–10 mg 1–1) promotedgermination and tube growth. Addition of 1 and 10 mg 1–1cAMP to any of the growth regulators had a promotory effect.Pollen tube growth decreased in white light as compared to thedark, and was increased in red light. Far-red light counteractedthe effect of red light. The effect of growth regulators incausing the enhanced tube growth appears to be manifested throughsubstances such as cAMP, and phytochrome seems to be involved. The effects of a number of growth-promoting and growth-inhibitingsubstances, including two fungal toxins, were studied on theextension of segments of etiolated tomato seedling hypocotyls.The bioassay was sensitive to small quantities of NaF, coumarinand 2, 4-DNP and inhibition was observed at all concentrations.2, 4-DNP or Iodoacetate stimulated growth at concentrationsbetween 1? 10–4 and 5 ? 10–6M. or 1 ? 10–6and 1 ? 10–7M. respectively. Inhibitor experiments inbuffered nutrient solution were approximately 10 per cent. moresensitive than those in deionized water. By means of paper partition chromatography small quantitiesof two fungal toxins, fusaric and alternaric acid were chromatographedand bioassayed. The effect of fussric acid (5, n-butyI picolinicacid) on hypocoty1 growth was detected at concentrations aslow as 1 ? 10–5M. Experiments with recongnized growth-promoting substances showedthat Kinetin inhibited growth at concentrations up to 1 ?10–8M.in both light and dark. IAA inhibited growth up to 1 ? 10–6M.At 1 ? 10–7 and 1 ? 10–8 only small increases occurredwith IAA and the effect of light was negligible. Gibberellicacid (GA2)stimulated growth at concentrations from 10–3to 10–7M. and significant increases up to 17 per cent.were recorded in the light. Since the light induced inhibitionwas only partly restored, the existence of some other naturallight sensitive growth substance is suggested. The value ofthe bioassay as a method for estimating natural growth-inhibitingand growth-promoting substances is discussed.  相似文献

The effect of iodotyrosine and its related compounds on theroot elongation was studied using intact roots of rice seedlingsgrown on an agar medium and in a culture solution. MIT (3-monoiodotyrosine), DIT (3,5-diiodotyrosine) markedlystimulated the growth of seminal root, but tyrosine itself wasnot active. Both iodinated amino acids doubled the length ofseminal root in comparison with the control when used at theconcentration of 10–5M, where the growth of shoot andcrown root was not affected. And above 10–5M, the developmentof lateral root and root hairs was inhibited. DBT (3,5-dibromotyrosine) also notably increased the lengthof seminal root.
