Experiments were performed to investigate the influences of dietary protein on fecundity in the guppy, Poecilia reticulata . No significant differences in total body length and standard length were found between females given food with different protein levels. A significantly higher total body weight was found both in females fed on 47% and 31% protein compared with females given 15% protein. The frequencies of females with only intrafollicular embryos or with only ova or both embryos and ova in the ovary were not significantly different between populations given different protein amounts. There was a significantly lower frequency of females without embryos and ova but with previtellogenic oocytes in the group given highest protein (47%) compared with the group fed on 31%. Significantly higher values of ovarian length, width and gonad weight were found among females given 47% protein compared with females fed on 15% protein. The ovarian height was significantly higher only in females given the highest protein amount when compared with the group fed on 31% protein. No significant difference of ovarian volume was found between groups given different protein contents. The average gonadosomatic ratio was highest in the groups fed on 47% protein. Significant correlations were found between gonad weight and total body weight of females in each group. Ovarian volume versus total body weight as well as versus gonad weight also yielded significant correlations in all groups. No significant differences of fecundity were found either between females given 47% and 31% protein or between females fed on 47% and 15% protein. A somewhat higher number of ova per individual fish was, however, observed among females given 31% when compared with females fed on 15% protein. The ovum size remained the same between groups given different protein contents. No significant correlation was found between fecundity and ovum diameter in the groups.  相似文献

Ovaries from 10- to 18-day-old chick embryos hypophysectomized by partial decapitation were cultured in vitro and their estradiol secretion was compared to that of ovaries from control embryos. The production of estradiol was not less in the decapitated than in the control embryos at 15-18 days, neither per ovary nor on the basis of ovarian weight. However, the difference was significant at 10-11 days. These results suggest that the hypophysis controls estradiol secretion by the chick embryo ovary in the early stages, but not in the later ones.  相似文献

The amounts of estradiol released into culture media by ovaries from 19-day-old hypophysectomized (decapitated) and sham-operated chick embryos were determined by RIA. Per single ovary, ovaries from decapitated embryos secreted slightly less estradiol than ovaries from sham-operated ones during a 4-h culture period (837 +/- 413 vs 935 +/- 378 pg), but this difference was not significant. On an ovarian weight basis, ovaries from decapitated embryos secreted slightly more estradiol than ovaries from sham-operated ones (1.15 vs 0.91 ng), but this difference was not significant either. It is concluded from these results that the hypophysis does not control estradiol secretion by the ovary in the 19-day-old chick embryo.  相似文献

The objective of the present experiment was to evaluate the effects of different levels of inbreeding on ovarian response and embryo production from superovulated cows. One hundred and thirteen Mantiqueira cows (a medium-size, Bos taurus native dairy cattle breed), with inbreeding coefficients ranging from 0 to 30%, were allocated into five classes of inbreeding and subjected to superovulation treatment. At induced estrus, cows were mated with Mantiqueira bulls (with minimal inbreeding). Six to eight days after mating, the cows were slaughtered, ovarian structures counted and embryos recovered by flushing the uterine horns and oviducts. Sire, season, age, weight, parity and age at first calving of donors did not significantly affect ovarian response or embryo production and quality. There were no effects of inbreeding class on number of total corpora lutea (CL) or number of CL present in the right ovary. However, the number of CL in the left ovary was reduced (P<0.05) in cows with Class 5 (>9%) of inbreeding. The number of transferable, but not the number of non-transferable embryos or the total number of embryos from cows with Class 5 of inbreeding, was lower (P<0.05) than those of cows from Classes 0 to 4 (<9%) of inbreeding. There was a quadratic decrease in the number of transferable embryos as inbreeding coefficient increased (Y=11.077+0.34X-0.0529X(2); R(2)=0.91, P<0.01), but no significant linear or quadratic effect of inbreeding on total number of embryos or number of non-transferable embryos. In conclusion, an inbreeding coefficient>9% reduced the quality of bovine embryos at the initial stage of development.  相似文献

Follicular dysfunction induced by autoimmunity to zona pellucida   总被引:1,自引:0,他引:1  
The mammalian zona pellucida is an extracellular matrix that occurs in growing oocytes, ovulated eggs and pre-implantation embryos, and is known to be involved in several important events during ovarian folliculogenesis and fertilization. Since the zona pellucida is formed at an early stage of oocyte growth, circulating antibodies against zona pellucida may impair ovarian function. In this article we discuss whether anti-zona antibodies cause ovarian dysfunction and infertility. The discussion is based on clinical examination and animal experiments including the following approaches: 1/ immunological method using solubilized human zona pellucida detected anti-zona antibody with a high frequency in infertile patients, especially premature ovarian failure syndrome; 2/ in vivo experiment using hamsters showed that some, but not all, animals experienced ovarian failure after immunization with hamster recombinant zona proteins; 3/ in vitro experiment using mouse isolated ovarian follicles showed significant inhibitory effects on follicular growth and oocyte development. We concluded that anti-zona antibody may be involved in causing ovarian failure.  相似文献

In the viviparous teleostZoarces viviparus (L.) embryonic post-yolk sac development in the ovary is characterized by significant increases in dry weight and nitrogen per embryo, thus indicating an extensive matrotrophic relationship. In the ovarian fluid surrounding the embryos during their intraovarian development, sources of nitrogen were shown to derive from amino acids, urea, ammonia and various cellular components. The level of urea in the ovarian fluid increased significantly from 3.68±0.25 mol urea-N·ml-1 during early post-yolk sac embryonic development to 6.14±0.44 in late development. The corresponding level of ammonia-N in the ovarian fluid increased from 0.25 to 0.45 mol·ml-1. An estimation of embryonic nitrogen loss was made by measuring urea and ammonia-N excretion in vitro by post-yolk sac embryos or larvae (i.e. seawater-acclimated embryos). Urea-N constituted an average of 65% of the total nitrogen excreted by the embryos into the ambient medium during a 5-h time-course compared to only 35% in the larvae. Urea-N was excreted at maximum rates during the first hour of the experiment, 0.54±0.09 mol N·g-1·h-1 by embryos and 0.35±0.02 by larvae, and then declined to lower levels in both embryos and larvae. A decline after 1 h was also observed for excretion rates of ammonia. In conclusion, the capacity for urea excretion by post-yolk sac embryos ofZ. viviparus may be of adaptive significance for their prolonged stay in ovario. Previous publications have produced conflicting results concerning the effect and nature of uterine horn position on embryonic weight and development in the uterus bicornis. For this investigation we used 503 embryos from 41 litters of NMRI - mice on gestational day 14. The relative position of each embryo was determined by measuring the distance from the center of each placenta to the cervix in relation to the total length of the uterine horn. After cessation of shrinkage due to fixation the umbilical cord and remaining membranes were removed under the dissecting microscope and the embryos were weighed to the nearest 0.1 mg. Analysis of potential embryonic positions along the uterine horns indicated that 10% at the extreme ovarian and cervical ends were almost devoid of implantations. A mixed nested analysis of variance was used as the appropriate model to test a position on weight effect. Not only was a position effect found but, in contrast to other studies, it is also quantitatively represented. Embryos were lighter both at the ovarian and cervical ends, the mean weight difference being in the range of less than 10% of the average embryo weight.  相似文献

ABSTRACT: Background Our previous work found that mouse embryos could invade malignant cancer cells. In the process of implantation, embryo trophoblast cells express matrix metalloproteinases and the invasive ability of trophoblast cells is proportional to matrix metalloproteinase-9 protein expression. So the purpose of this study is to observe the effects of mouse embryos on human ovarian cancer cells in the co-culture environment in vitro and explore the possible mechanism of matrix metalloproteinase-9. Methods Several groups of human ovarian cancer cells HO8910PM were co-cultured with mouse embryos for different time duration, after which the effects of mouse embryos on morphology and growth behavior of HO8910PM were observed under the light microscope real-time or by H.E staining. Apoptosis was detected under laser confocal microscope by Annexin V-EGFP/PI staining in situ. Invasion ability of tumor cells was studied by transwell experiments. After matrix metalloproteinase 9 (MMP -9) activity was inhibited by MMP-9 Inhibitor I, the interaction between mouse embryos and human ovarian cancer cells HO8910PM was observed. Results Mouse embryos were able to invade co-cultured human ovarian cancer cell layer which extended in the bottom of the culture dish, and gradually pushed away tumor cells to form their own growth space. The number of apoptosis tumor cells surrounding the embryo increased under laser confocal microscope. After co-cultured with mouse embryos, tumor cells invasive ability was lowered compared with the control group. After MMP-9 activity was inhibited, the interaction between mouse embryos and HO8910PM cells had no significant difference compared with the normal MMP-9 activity group. Conclusion Mouse embryos were able to invade human ovarian cancer cells in vitro and form their own growth space, promote apoptosis of human ovarian cancer cells and lower their invasive ability. The mouse embryo was still able to invade human ovarian cancer cells after MMP-9 activity was inhibited.  相似文献

The effects of applying extremely low-frequency (50 Hz) electromagnetic fields (ELF-EMF) for 24 hr and different densities (1.33-7.32 mT) were examined on healthy, freshly fertilized white leghorn chicken eggs (55-65 g). Results showed no increase in the rate of abnormalities in exposed groups, but were only significant in 4.19, 5.32 and 5.86, 6.65 mT densities. Alizarin red S and alcian blue 8GX staining showed some embryos with extra ribs, defects in ribs and vertebrae, anuria and abnormal beaks. Study of egg weight, after 9 days of incubation, showed no significant differences between control, sham-exposed and experimental groups. Analysis of crown-rump, beak-occipital length and weight of embryo, showed significant decrease in weight at 4.39 and 5.52 mT intensities, comparing with control and sham-exposed groups. These results revealed that 50 Hz electromagnetic fields can even induce developmental alterations in preincubated chick embryos and confirm that its strength could be a determinant factor for the embryonic response to extremely low frequency EMFs (window effects) prior to incubation.  相似文献

Embryos of goodeid fishes develop to term within the ovarian lumen, where they undergo considerable increase in weight due to transfer of maternal nutrients across a trophotaenial placenta. The placenta consists of an embryonic component, the trophotaeniae, and a maternal component, the ovarian lining. The latter was examined by transmission electron microscopy, scanning electron microscopy, and light microscopy in both gravid and nongravid ovaries of the viviparous goodeid fish, Ameca splendens. The single median ovary of A. splendens is a hollow structure whose lumen is divided into lateral chambers by a highly folded longitudinal ovarian septum. Germinal tissue occurs within folds of the ovarian lining that extend into each of the two lateral chambers. Matrotrophic embryonic development takes place within ovarian chambers. During gestation, the lining of the ovarian lumen is in direct apposition to body surfaces and trophotaenial epithelia of developing embryos. The ovarian lining consists of a simple cuboidal epithelium, termed the internal ovarian epithelium (IOE), overlying a well-vascularized bed of connective tissue. Cells of the IOE are apically convex. Well-developed granular and agranular endoplasmic reticula and numerous large membrane-bound vesicles with electron-dense content occupy the apical cytoplasm of IOE cells. Two functional states of the same cell type are distinguished within the IOE. Phase I cells contain few, if any, large apically situated vesicles; Phase II cells contain many. Secretory products of the IOE are presumed to be an important source of nutrients for embryonic development. Structural and functional relationships of the IOE to the trophotaenial epithelium of developing embryos are discussed in relation to maternal-embryonic nutrient transfer processes.  相似文献

Calcium metabolism was studied in relation to ovarian functions and embryonic development during early and late pregnancy in the viviparous blenny Zoarces viviparus . The level of total calcium in the maternal serum decreased during pregnancy from 21·7 (± 2·6) to 8·7 (± 1·1) mg 100 ml−1. Calcium in the ovarian fluid surrounding the embryos appeared at a level which was always lower than in the maternal blood. The calcium content of the embryos increased during their development in the ovarian cavity. During early pregnancy, oestradiol-treatment increased the level of total calcium and alkkali-labile protein P (vitellogenin) in serum of the maternal organism, but did not have any effect on the level of ultrafiltrable calcium. However, in the untreated females, the post-ovulatory follicles, believed to act as calyces nutriciae in the pregnant ovary, were able to concentrate the ultrafiltrable calcium above the level in the serum. The activity of injected45Ca per unit plasma and follicular fluid volume was always higher in oestradiol-pretreated fish when compared with controls over a time course range. Embryos from females, which were not treated with oestradiol, accumulated high levels of labelled calcium and the accumulation was time-course dependent. Embryos from oestradiol-treated females showed low levels of tracer accumulation at all samplings and their appearance indicated an overall negative effect of oestradiol. During late pregnancy, labelled calcium showed a rapid turnover within 24 h post-injection in the maternal blood and ovarian fluid and a large accumulation in the embryos. Calcium influx in embryos, which were incubated with labelled calcium during 72 h in in vitro systems was largest during the initial 4·5–24h time interval.  相似文献

The reproductive organs in non-obese diabetic (NOD) mice were histopathologically studied, in order to elucidate the relationships between developmental abnormalities, such as diminished rates of implantation and viable embryos, and structural changes in the reproductive organs. NOD mice with (NOD-DM) and without (NOD-N) diabetes mellitus and ICR mice were compared. The severity of histopathological changes in the pancreas and in the liver were used as parameters which indicated the severity of diabetes itself and of the secondary metabolic disorder. NOD-DM mice exhibited uterine weight loss, accumulation of lipids in luminal and glandular epithelium, atrophies of the endometrium and myometrium and a decrease in the number of muscle cell layers. They also showed a high concentration of lipid droplets in ovarian granulosa cells, atretic follicles and atrophy and lack of lipids in ovarian stroma cells. The severity of these structural changes in the reproductive organs corresponded to those of the changes in the pancreas and the liver. The structural alterations in the ovary suggested disorder in oocyte maturation. The structural changes in the uterus appeared to be related to the decrease in the ratios of implantation and of viable embryos at post-implantation stage. The present studies suggest that the impaired structural environment together with the metabolic environment caused the abnormal development seen, for example, in the oocyte maturation, and at the implantation and post-implantation stage of diabetic mice. This study was carried out to investigate the impact of tripterygium glycosides (TGs) on ovarian function of female rats in vitro and in vivo. In vitro studies showed that TG induced cells decrease at G1 phase and inhibited cell proliferation in rat granulosa cells. In vivo, female rats were intragastrically administered with TG at the dose of 60 mg/kg/day for consecutive 50 days. TG caused a prolonged estrous cycle, and a significant reduction in ovarian index, serum E2 level, and numbers of secondary and antral follicles (p < 0.05) in these rats. A significant reduction of viable embryos was demonstrated in TG‐treated female rats after mating (p < 0.01). Further, we observed observed the reduced expression level of TGF‐β1 after TG treatment in vitro and in vivo. Moreover, the expression of Smad2 and AKT was also decreased after TG treatment. These results suggest that TG can impair ovarian function through Smads‐mediated TGF‐β1 signal pathway  相似文献

Progestin-primed ovarian stimulation (PPOS) regimen was established for assisted reproduction. However, its feasibility and outcomes in polycystic ovary syndrome (PCOS) patients need further evaluation. The outcomes of infertile patients with PCOS (study group) and normal ovaries (control group with unexplained infertility and tubal factor infertility) who underwent PPOS and IVF/ICSI protocol were retrospectively studied. The baseline information, primary, and secondary outcomes of patients were collected. The dynamic changes of hormones were closely monitored. 198 PCOS patients and 374 controls were included in this study. After controlled ovarian hyperstimulation (COH), 15 oocytes were retrieved from PCOS patients on average, which was more than those from the controls (p < 0.001). The oocytes and embryos obtained from the PCOS patients exhibited better developmental potential as the number of fertilized oocytes, cleaved embryos, top-quality embryos, viable embryos, cryopreserved embryos, the rate of fertilization, and viable embryo per oocyte retrieved in PCOS patients were significantly higher than those in the controls (all p < 0.001). No significant difference between the two groups was identified regarding the primary outcome, ongoing pregnancy, and other secondary outcomes. No moderate to severe ovarian hyperstimulation syndrome (OHSS) was diagnosed in either group. With the proposed PPOS protocol, the quantity, quality, developmental potential of oocytes, and embryos obtained from PCOS patients were superior to those from controls. The protocol was efficient and safe in terms of pregnancy, obstetric, and perinatal outcomes. OHSS was effectively mitigated in the patients, with or without PCOS, who underwent COH.  相似文献

Fertilization and development in the ovarian cavity of oviparous fish, Oryzias latipes, were examined using the S-rR strain in which the sex genotype can be easily distinguished by the body color of the fish. Mature eggs were fertilized within the ovarian cavity after a sperm suspension was artificially introduced with a small bore-pipette through the urinogenital opening. Three batches of eggs ovulated within 48 hrs were fertilized and began to develop in the ovarian cavity, while eggs ovulated 72 hrs post-insemination (PI) were no longer fertilized. These observations indicate that ovulation occurs irrespective of the existence of developing embryos within the ovarian cavity. All embryos developing in the ovarian cavity were, however, retarded and ceased development before the stage of initiation of blood circulation at room temperature. These embryos developed normally and hatched after they were transferred from the ovarian cavity into regular saline 48 or 72 hrs PI. When these individuals matured sexually, their sex differentiation was found to be normal, and sex reversal was not observed.  相似文献

To examine the potential teratogenicity of electromagnetic fields (EMF; sinusoidal and rectangular) on development of chick embryos (white leghorn), 221 freshly fertilized chicken eggs (55-65 g) were exposed during first 24 hr of postlaying incubation (38 degrees +/- 0.5 degree C) to 24 different EMFs, with 50Hz repetition rate and 8.007-10.143 mT flux density. Following exposure, the exposed fertilized chicken eggs (n = 8-10) and sham-exposed fertilized chicken eggs (n = 15) were incubated simultaneously for 8 more days and unexposed control fertilized chicken eggs (n = 20) for 9 days in absence of EMFs. The embryos were removed from egg shells and studied blind. All 24 EMF exposed-groups (inside the coil with exposure) showed an increase in the percentage of developmental anomalies compared to sham-exposed (inside the coil with no exposure) and control groups (outside the coil). Further, egg's weight was evaluated on day 9. This variable did not show significant difference between control and exposed-groups. The investigation also covered the measurement of body weight, length of crown to rump, length of tip of the beak to occipital bone, heart and liver weight. Statistical comparison between sham-exposed and control values did not show significant differences, but comparison between 8.007, 8.453 and 8.713 mT exposed-groups and control groups showed significant differences; in other exposed-groups, the changes were not significant. These results revealed that 50 Hz electromagnetic fields can induce irreversible developmental alterations in 24 hr chick embryos and confirm that its strength could be a determinant factor for the embryonic response to extremely low frequency electromagnetic fields (window effects).  相似文献

The influence of continuous electromagnetic fields (0, 181 or 361 Gs/cm2) on the development of chick embryo (n = 144) was studied. Several parameters were determined at days 5, 10 and 15 of incubation: stage (following Hamburger and Hamilton), vertex-coccyx length (size) and weight. At 5 days of incubation, all embryos showed a similar stage. However, at days 10 and 15, the embryos exposed to 181 Gs/cm2 showed a stage significantly superior to that of the others. There were no differences between the exposed embryos and the control ones with regard to weight and stature, except at 15 days when the embryos exposed to 361 Gs/cm2 showed greater weight and stature than those of the controls.  相似文献

A nonestrogenic component of young, rapidly growing plants, 6-methoxybenzoxazolinone (6-MBOA), was examined to determine its effect on the reproductive responses of prepubertal and mature female rats. Prepubertal animals treated with a single injection of 6-MBOA or with Silastic capsules implanted for 3 days showed a significant increase in both ovarian and uterine weight. Serum luteinizing hormone was unaffected by 6-MBOA treatment for 3 days in 32-day-old animals, whereas serum follicle-stimulating hormone was elevated. Silastic capsule treatment of mature animals showed the following results. Extended treatment for 6 estrous cycles had no influence on the timing of vaginal cyclicity; despite this, 6-MBOA treatment for 2 cycles caused an increase in ovarian weight resulting from an increase in the number of corpora lutea per ovary. Animals treated for 1 cycle showed a significant increase in the number of ova shed. Uterine weight in mature animals did not increase. This study indicates that 6-MBOA has a stimulatory effect on the reproductive system of young and mature female rats. It is the first attempt to relate the effects of the compound on the endocrine system of any animal. That nonestrogenic plant compounds can trigger reproduction has important ecologic and physiologic significance.  相似文献

Significant increase in ovarian and uterine weight and stimulation of ovarian delta5-3beta-hydroxysteroid dehydrogenase (delta5-3beta-HSD) activity and elevation of serum estradiol level were observed following gold chloride (0.2 mg/kg body weight/day), s.c. administration in immature female albino rats. Moreover, normal cyclic changes of estrus were found in vaginal smears of these rats whereas the rats of other groups showed diestrus phase throughout the period of experiment. Histological study of ovary also showed Graafian follicle with ovum in rats treated with 0.2 mg/kg/day of gold proving stimulation of reproductive function, which was not found in the ovarian histological study of other groups including controls. Thus, the results suggest a significant stimulatory effect of gold chloride on female reproductive activity in immature rats. Further, since the above-mentioned changes were evident at a specific dose of gold chloride, the data may have some clinical implications on stimulation and enhancement of fertility in immature female rats.  相似文献

We investigated the effects of body fat content on mouse fecundity, ovarian hormone release, and their response to follicle stimulation hormone (FSH). 4 types of females were produced: lean (group 1), normal (group 2), slightly fat (group 3), and significantly fat (group 4). The body weights, fat content, fertility rate, embryo number produced, retarded and degenerated embryo percentage, the release of progesterone (P4), testosterone (T), and insulin-like growth factor I (IGF-I) by isolated ovaries cultured with and without FSH (1.0 IU/mL medium) were evaluated. A gradual increase in body weight and fat contents from groups 1 to 4 was observed. Group 2 had higher fertility rate than those from the other groups. Groups 2 and 3 had fewer retarded and degenerated embryos that those from groups 1 and 4. Embryo production rate was not different among the groups. P4 and T secretion was higher from group 4 than in those from groups 1–3; secretion of IGF-I of group 3 was less than that of groups 1, 2, and 4. FSH promoted ovarian T output in all groups and stimulated ovarian P4 release in groups 1, 3, and 4, but not in group 2. FSH did not affect IGF-I release in any group. Therefore, both malnutrition and overfeeding can affect body weight and fat content in female mice, reducing embryo quality or developmental capacity, but not fertility and embryo production. Excess weight or fat can have stimulatory effects on ovarian P4 and T, but inhibitory effects on ovarian IGF-I release. Both leanness and excess weight or fat can induce the stimulatory action of FSH on ovarian P4.  相似文献
