Using a polyclonal antibody against the mate recognition pheromone (MRP) of Brachionus rotundiformis Koshiki strain, we investigated the behavioral reproductive isolation and the similarity of MRP among geographically and temporally isolated B. rotundiformis strains. Males of the Koshiki strain did not discriminate in mating attempts among females of the Koshiki strain and those of conspecific allopatric strains from Hamana, Fiji, Thailand and Spain. Likewise, Koshiki males attempted mating with statistically indistinguishable frequency with Koshiki females and B. plicatilis strains. However, copulation was not consummated between Koshiki males and B. plicatilis females. The amount of anti-MRP binding to three allopatric B. rotundiformis strains was similar to that of the Koshiki strain, but binding to Hamana and the B. plicatilis strain was significantly lower. Four temporally separated B. rotundiformis populations were hatched from resting eggs collected from 0, 5, 10 and 15 cm depth in the sediment of Kai-ike pond in Koshiki island, Japan. Sediment age was determined using the 210Pb method, allowing us to estimate that resting eggs from 15 cm depth were produced 65 years ago. Results of mating assays and anti-MRP binding showed that no behavioral reproductive isolation exists among the four temporally isolated Koshiki strains. B. rotundiformis appears to be reproductively isolated from B. plicatilis, but heterospecific matings are still attempted between B. plicatilis and B. rotundiformis, suggesting that the MRP remains sufficiently similar to elicit circling behavior.  相似文献

Brachionus plicatilis raised in our laboratory in sea water reproduces asexually even under high crowding conditions (at least 40 individuals per ml). Amictic females were induced to produce mictic females, males and resting eggs by reducing the concentration of the sea water culture medium. Mictic females and males appeared predominantly among the progeny produced by the amictic females during 4 days following their transfer into 25% sea water. Resting eggs appeared first 5–12 days after the onset of the experiment. Following the disappearance of males, the culture consisted of amictic females.Resting eggs produced by the method described above may be preserved for at least three months at –14°C or by desiccation at room temperature. Under the appropriate experimental conditions, resting eggs hatch into amictic females. Since B. plicatilis is one of the most commonly used food sources of fish larvae in aquaculture, the methods reported here may offer an easy and versatile way of preserving rotifer culture stock to be used on demand.  相似文献

We studied the effect of different concentrations (0, 3, 6, 9 and 12 g l–1) of sodium chloride at one food level of Chlorella (1×106 cells ml–1) on competition between the rotifers B. rotundiformis and H. jenkinae, both of which were isolated from a saline lake. The population growth experiments were conducted for 3 weeks. Both the rotifer species did not survive beyond one week at a salinity of 0 g l–1. Regardless of salt concentration and the presence of a competitor, H. jenkinae reached higher densities than B. rotundiformis. When grown alone, both B. rotundiformis and H. jenkinae showed optimal peak population densities at the salinity of 6 and 9 g l–1. Since biomass wise, B. rotundiformis was larger than H. jenkinae, it showed a lower numerical abundance. Thus, the maximum peak population densities of B. rotundiformis and H. jenkinae recorded in this study were 107±3 and 203±28 ind. ml–1. The maximal rates of population increase for B. rotundiformis and H, jenkinae when grown alone were 0.264±0.003 and 0.274±0.004, respectively. Our results also indicated that B. rotundiformis and H. jenkinae coexisted better at a salinity of 6 and 9 g l–1 of sodium chloride while a salinity of 3 g l–1 favoured Hexarthra over B. rotundiformis. At 12 g l–1, both the rotifer species grown alone or together showed lower growth rates compared to those at lower salinity levels. Except 0 g l–1, in all other salinity treatments, H. jenkinae was a superior competitor to B. rotundiformis.  相似文献

The production of resting eggs by the rotifer Brachionus plicatilis was tested at four salinities (9, 18, 27 and 36\%) and six concentrations of the alga Chlorella stigmatophora (0.25, 0.5, 1.0, 2.0, 4.0 and 6.0 × 106 cells ml–1). The results indicated that resting eggs were produced only at two salinities (9\% and 18\%) and that their number was affected by the amount of food provided. A model consisting of two generalized linear sub-models was built to evaluate the contribution of each of the tested food concentrations at the two salinities. The sub-models were used to distinguish between two different components of resting egg production: one related to the presence or absence of resting egg production, and the other to the number of resting eggs produced, given that production had occurred. Besides indicating the best combination of salinity and food concentration for obtaining large numbers of resting eggs, they revealed the contribution of internal population factors that were not controlled in the course of the experiment. The model identified the positive contribution of the relative number of females to males, and the negative association between high rotifer densities and the production of resting eggs. The results of the present study help in defining the optimal conditions for mass production of resting eggs, which are of potential importance in aquaculture.Deceased, September 1991.  相似文献

Pesticides have been major contributors to environmental pollution and they are now widely distributed in aquatic environments. Zooplankters are frequently used as test animals to detect aquatic contaminants because of their sensitivity and ecological importance. We investigated the effect of a 7-day exposure to four commonly used pesticides (diazinon, fenitrothion, methoprene and isoprothiolane) on reproduction of the rotifer Brachionus plicatilis. Pesticide concentrations of 3–7 times lower than the 24-h 50% lethal concentration (24-h LC50) were tested to determine the ‚no observed effect’ concentration (NOEC), ‚lowest observed effect’ concentration (LOEC), and the ‚50% effective’ concentration (EC50) on specific growth rate (r), sexual reproduction, fertilization, resting egg production, and hatchability of resting eggs. Results showed that the lowest EC50 value of r, mixis, fertilization, and resting egg production of 1.4 μM for diazinon was 63 times lower than its 24-h LC50 of 88.4 μM, while for fenitrothion it was 66 times (3.5 and 229.8 μM, respectively). For isoprothiolane, the lowest EC50 value of r, mixis, fertilization, and resting egg production of 8.9 μM was 25 times lower than its 24-h LC50 of 220.7 μM, while for methoprene was 37 times (2.7 and 100.8 μM, respectively). In all pesticides, hatching rate of resting eggs consistently gave the lowest EC50 values which is about 2–40 times lower than the lowest EC50 of r, mixis, fertilization, and resting egg production. Hatchability of resting eggs therefore is the most sensitive parameter in detecting effects of pesticides exposure in rotifer B. plicatilis.  相似文献

Clearance and ingestion rates of Brachionus plicatilis were measured using 14C-labeled Isochrysis galbana Tahiti. Experiments were conducted at 20–22 °C, 20 ppt salinity, and algal concentrations ranging from 0.13–64 mg C 1–1. Clearance rates were constant and maximal at concentrations <2 mg C 1–1, with maximum rates ranging from 3.4–6.9 µl ind.–1 hr–1. The ingestion rate varied with food concentration, and was described by a rectilinear model. The maximum ingestion rate varied considerably, and was dependent on the growth rate of the rotifers. Depending on the pre-conditions, B. plicatilis ingested about 0.5 to 2 times its body carbon per day at saturating food concentrations.  相似文献

Detection and characterization of sibling species complexes in zooplankton are critical to understanding their ecological responses and patterns of evolution. The taxon Brachionus plicatilis is a complex of at least 14 species with three major, deeply diverged clades, which are morphologically distinct. We studied morphometric differences between two species – B. plicatilis sensu stricto and B. ‘Manjavacas’– which belong to the L-(large) morphotype and often co-occur in ponds or lakes. B. plicatilis s.s. was on average 6% longer than B. ‘Manjavacas’. They differed significantly in the measurements related to lorica spines. A significant discriminant function relating spine measurements was found, however, individuals from each species showed extensive overlap. Our morphometric data provide additional evidence for the species status of B. plicatilis s.s. and B. ‘Manjavacas’. Since these are ancient species, our results support that a morphological stasis occurs in these taxa. We identified COI restriction sites for PvuII and KpnI which are diagnostic for B. ‘Manjavacas’ and B. plicatilis s.s., respectively. We conclude that morphometry is not useful in classifying the two species. At present, this can only be done reliably using molecular methods.  相似文献

Diapause and hatching of Brachionus plicatilis Müller resting eggs were examined through histological and optical approaches. Compound microscope observations on 1% toluidine blue-stained embryo sections suggests that the total number of nuclei in an embryo during the internal diapause period increased from 22 on Day 2 to 39 (each n = 1) on Day 6. The outer layer of embryo membrane gradually thickens from 1.2 (Day 0) to 4.0 µm (Day 8) (each n = 10).Resting eggs that have completed maturation and are in the external diapause period require light for hatching. The threshold of light (halogen lamp) intensity for hatching was estimated to be 4400 lux for 30 min. Hatching rate decreased with longer wavelength irradiation (mercury lamp). Irradiation at more than 350 nm caused 1–25% hatching, but it reached 50–60% at 250–310 nm light. The addition of hydrogen peroxide or prostaglandins (E 1, E 2 or F 2) caused resting egg hatching even in darkness. The production of peroxide in seawater caused by light as well as the oxidation of fatty acid to prostaglandins inside the embryo is a possible mechanism of resting egg hatching.  相似文献

Tolerance to low oxygen concentrations is expected in Brachionus plicatilis, a rotifer adapted to live in saline warm waters. The population dynamics of a clone of this species, isolated from an endorreic saline lake, was studied under controlled laboratory conditions. Although their growth and metabolism is extremely reduced, B. plicatilis populations are able to maintain relatively high-density populations (a mean of 35 ind ml–1) in oxygen concentrations below 1 mg 1–1, for more than one month. The effect of temperature and salinity on resting egg size of two Brachionus plicatilis (Rotifers) clones was investigated. Clones were selected according to their different behaviour in laying resting eggs: one clone ejects them, whereas they remain inside the females body in the other clone. The difference in resting eggs size between the two clones is noticeable, although the difference is not as great as that between female body size. An important temperature-salinity interaction on resting egg size has been observed. The general inverse relationship between size and temperature is only true at lower temperatures. At high temperatures size varies around the mean although could be greater than at intermediate temperatures. This is more evident at the intermediate salinity tested which is considered to be the closest to the optimum in our experiments. This pattern of variation suggests that mean size is bigger than expected, in relation to temperature and salinity, when these factors have values close to the extremes of their range, normally found in nature, and to which adaptative mechanisms can evolve. Size is bigger at the salinity — temperature low - low and high - high combinations which are the most commonly found in the temperate environments.  相似文献

Fertilized resting eggs of Australian Brachionus quadridentatus hatched 2–3 days after hydration into females with or, more frequently, without posterior lateral spines. These females then produced clones with short-spined or long-spined phenotypes. Asplanchna girodi induced females from two short-spined clones and one long-spined clone to produce daughters with significantly longer posterior lateral spines. In all clones, there were significant differences in spine development among offspring of mothers within Asplanchna and control treatments. The range of phenotypes reported in one short-spined clone is observed in the billabong and includes much of the variation described for the species, with mehleni (long-spined) phenotypes occurring with Asplanchna. In B. quadridentatus, the ecological significance of long-spined, basic phenotypes, and of the spine-development response to Asplanchna, is unclear. In laboratory cultures, females of all clones were attached to the substratum or water surface, and were safe from Asplanchna; in nature, females are epiphytic and probably rarely susceptible to Asplanchna. Most (96%) resting eggs produced in cultures and kept under culture conditions hatched after a 7-day latent period. This raises questions regarding natural conditions which might prevent hatching and allow accumulation of resting eggs in a sediment egg bank. Hatching of resting eggs in nature may be enhanced in sediments which dry and then become flooded after rains.  相似文献

Size selective feeding by Brachionus plicatilis was investigated with algae and bacteria (0.3–3.5 µm) and mono-disperse latex beads (0.3–3.0 µm) in short term feeding experiments. B. plicatilis demonstrated maximum clearance rate of particles with diameter 2µm, but particles with diameter down to 0.3 µm were also ingested. The clearance rate of bacteria was 15–55% of that obtained for optimal sized particles (2 µm), and was related to particle size. The relative reduction in retention of particles with diameter < 2 µm was more pronounced for latex beads than for natural food particles, suggesting other mechanisms than size to be important for the particle retention by the rotifer. This is emphasized by the fact that the clearance rates were much lower for latex beads than for natural food particles of comparable size. Efficient retention of bacteria was observed for rotifers in poor physiological condition, i.e. rotifers with low maximum clearance rate. This may reflect a strategy to optimize energy utilization by reducing locomotion costs and increasing energy intake.The results indicate that B. plicatilis has a low to medium ability to feed on bacteria. In natural ecosystems, its importance as a bacterial grazer is of limited importance. At high population densities, such as in live feed cultures, the rotifer may, however, efficiently remove bacteria from the culture.  相似文献

This study was initiated to search for species-specific and strain-specific satellite DNA sequences for which oligonucleotide primers could be designed to differentiate between various commercially important strains of the marine monogonont rotifers Brachionus rotundiformis and Brachionus plicatilis. Two unrelated, highly reiterated satellite sequences were cloned and characterized. The eight sequenced monomers from B. rotundiformis and six from B. plicatilis had low intrarepeat variability and were similar in their overall lengths, A + T compositions, and high degrees of repeated motif substructure. However, hybridizations to 19 representative strains, sequence characterizations, and GenBank searches indicated that these two satellites are morphotype-specific and population-specific, respectively, and share little homology to each other or to other characterized sequences in the database. Primer pairs designed for the B. rotundiformis satellite confirmed hybridization specificities on polymerase chain reaction and could serve as a useful molecular diagnostic tool to identify strains belonging to the SS morphotype, which are gaining widespread usage as first feeds for marine fish in commercial production. Brachionus quadridentatus and Brachionus plicatilis were examined in laboratory cultures to investigate their (1) food requirements and (2) ability to select particles on the basis of size. Growth experiments showed that the Chlorophyceans Nannochloris sp. and Chlorella vulgaris possess good nutritional value for these rotifers. However, B. quadridentatus declined in cultures with Microcystis firma, detritus of Enteromorpha sp. or Pseudomonas sp. (bacteria) as a sole source of food, thus indicating that these foods were inadequate. Utilization of latex microspheres by these two species indicate that they respond to particle size in different ways: B. quadridentatus had a higher selectivity index for 3 and 5 µm beads, while B. plicatilis had a higher selectivity index for 2 µm beads.  相似文献

We determined the effect of environmental stressors on the physiological condition of Brachionus rotundiformis. For two morphologically distinct B. rotundiformis strains: Hawaii (average lorica length = 222 m) and Langkawi strains (average lorica length 180 m), neonates hatched from resting eggs were exposed to different levels of unionized ammonia (0.7–9.8 mg l–1), viscosity (relative viscosity against natural seawater = 1–1.17) and Euplotes sp. (protozoan) contamination (1–40 cells ml–1). Increasing stress decreased fecundity and lifespan of both rotifer strains. Glucosidase and phospholipase activities were correlated with reproductive responses of both the strains exposed to unionized ammonia. When culture water viscosity was changed, the activity of esterase and phospholipase was correlated with reproductive responses of the Hawaiian strain, and glucosidase activity was correlated with those of Langkawi strain.With the protozoan contamination, esterase and glucosidase activities were correlated only with reproductive responses of the Hawaiian strain, while activity of all three enzymes was correlated to those of the Langkawi strain. Glucosidase activity proved to be a reliable indicator of stress for cultured B. rotundiformis.  相似文献

This study examined the morphology and hatchability of Brachionus plicatilis resting eggs as a function of the aging of maternal fertilized mictic females. One-hundred twenty fertilized B. plicatilis (Australian strain) were individually cultured and monitored daily until death. All cultures were maintained at 25°C, 11 ppt, and fed the micro-algae Tetraselmis tetrathele. Resting eggs produced by the females were investigated using two parameters: egg morphology, and hatching rate. Under these culture conditions, females normally produce 1–6 (mean ± SD = 2.7 ± 1.2) resting eggs during their lifetime. However, the number of resting eggs with abnormal morphology increased as a function of maternal age. Among resting eggs with normal morphology (n = 225), 82.2% were produced during the first and second spawning, and had hatching rates of more than 60%, while the hatching rates were below 30% in resting eggs with a spawning order of >2. Thus, the quality of B. plicatilis resting eggs was negatively correlated with maternal age.  相似文献

A detailed study of the life history of the rotifer Brachionus plicatilis was done at 20 °C, 20 ppt salinity, and 90 mg C 1–1 food concentration. Rotifers were grown individually in culture plate wells (150 µl culture volume) and fed Isochrysis galbana Tahiti, Tetraselmis sp., Nannochloris atomus, or a l : 1 mixture (weight) of two of the algae. Observations were made every 2–8 hr and rotifers were sized and transferred to new food daily. A total of 19 different parameters were compared. Rotifers fed Isochrysis averaged 21 offspring per female, a 6.7 day reproductive period, a lifespan of 10.5 days and a mean length of 234 µm. After Isochrysis, the foods giving the highest growth, survival, and reproduction in decreasing order were Isochrysis + Nannochloris, Nannochloris, Isochrysis + Tetraselmis, Tetraselmis + Nannochloris, and Tetraselmis. Although the small volume culture system used in this study seems appropriate for studying life history of B. plicatilis, the results cannot always be directly applied to larger cultures.  相似文献

The rotifer Brachionus plicatilis (O.F. Muller) can be mass cultivated in large quantities and is an important live feed in aquaculture. This rotifer is commonly offered to larvae during the first 7–30 days of exogenous feeding. Variation in prey density affects larval fish feeding rates, rations, activity, evacuation time, growth rates and growth efficiencies. B. plicatilis can be supplied at the food concentrations required for meeting larval metabolic demands and yielding high survival rates. Live food may enhance the digestive processes of larval predators. A large range of genetically distinct B. plicatilis strains with a wide range of body size permit larval rearing of many fish species. Larvae are first fed on a small strain of rotifers, and as larvae increase in size, a larger strain of rotifers is introduced. Rotifers are regarded as living food capsules for transferring nutrients to fish larvae. These nutrients include highly unsaturated fatty acids (mainly 20: 5 n–3 and 22: 6 n–3) essential for survival of marine fish larvae. In addition, rotifers treated with antibiotics may promote higher survival rates. The possibility of preserving live rotifers at low temperatures or through their resting eggs has been investigated.  相似文献

Behavioural Reproductive Isolation in a Rotifer Hybrid Zone   总被引:1,自引:1,他引:0  
A hybrid zone between two Brachionus plicatilis rotifer mitochondrial DNA (mtDNA) lineages was recently described in the Iberian Peninsula between a pond (Santed 2) and a lake (Gallocanta). The patterns of mitochondrial and nuclear genetic variation observed suggested that gene flow is mainly male-mediated from the lake to the pond. Here we test two hypotheses: (a) that male-mediated gene flow occurs through assortative mating between individuals from these ponds, (b) that behavioural isolation occurs between the two mtDNA lineages. We isolated, reared and genotyped rotifer clones from resting eggs collected in the sediments of these and two other distant ponds. We devised a quick, inexpensive RFLP method to discriminate between B. plicatilis and its sibling species B. ‘Manjavaeas’ and between both mtDNA B. plicatilis lineages. Behavioural no-choice tests using new-born, virgin males and females were performed between five clones. B. ‘Manjavacas’ and B. plicatilis were reproductively isolated. B. plicatilis clones did not show evidence of reproductive isolation, regardless of their mtDNA lineage, except Santed 2 males, which discriminated strongly against Gallocanta females. These results could help to explain the discrepancies between mitochondrial and nuclear genetic variation reported in the two populations.  相似文献
