The objective of the present study was to assess the impact of season on haematological and biochemical parameters of crossbred female calves. Seven crossbred female calves of 6–12 months of age were selected. The study was conducted in three different spells of 30 days each in pre-monsoon, monsoon and post-monsoon seasons. Ambient temperature (AT), relative humidity (RH) and temperature humidity index (THI) was recorded in all the three seasons. Whole blood was collected for estimation of haematological and plasma was used estimation of biochemical parameters. Highest AT and THI was recorded in pre-monsoon while highest RH was recorded in monsoon. The study revealed that season had no effect on haematological parameters such as total leucocyte, haemoglobin, packed cell volume, mean corpuscular volume, mean corpuscular haemoglobin, mean corpuscular haemoglobin concentration, while total erythrocyte count was significantly lower in monsoon compared to pre and post-monsoon seasons. Glucose and cholesterol were significantly lower in pre-monsoon and monsoon compared to post-monsoon. Glutathione peroxidase activity was significantly higher in pre-monsoon compared to monsoon and post-monsoon. No significant difference between seasons in the concentration of total protein, albumin, globulin and albumin globulin ratio was noticed. The results indicate that hot conditions have profound effect on haematological and biochemical parameters of crossbred calves.  相似文献

The behaviour of suspended particulate matter (SPM), salinity profile, dissolved nutrients, total (T.Chl-a) and size fractionated chlorophyll-a (F.Chl-a) were studied seasonally at Chilika Lagoon, east coast of India, during 2008–2009. The study showed large spatio-temporal variations among these parameters. The concentration of dissolved inorganic nitrogen and inorganic phosphate were found to be maximum during the monsoon, followed by post- and pre-monsoon, although the mean N:P ratios, which indicate the relative availability of N with respect to P, were 9.13 ± 3.09, 16.57 ± 11.53 and 5.47 ± 3.13, respectively. It was evident from the results that during pre-monsoon and postmonsoon, the lagoon exhibits nitrogen limitation. Mean T.Chl-a biomass in the lagoon showed distinct seasonality with maximum values during the pre-monsoon (23.12 ± 9.75 mg m?3) followed by monsoon and post-monsoon. Irrespective of seasons, maximum T.Chl-a was found in the northern part of the lagoon. SPM concentrations during the monsoon were relatively higher in the freshwater dominated zones compared to seawater dominated areas, indicating its riverine sources. The correlation between SPM and various dissolved nutrients (p < 0.05) suggests its influence on the physico-chemical conditions at varying levels. It is summarized that seasonal variation of SPM and nutrients contributed by rivers, wind induced re-suspension events and in situ  regeneration processes play a crucial role in the lagoon biogeochemical cycle.  相似文献

Mangrove species are broadly classified as ‘true mangroves’ and ‘mangrove associates’. We hypothesized that the leaf litter decomposition rates of true mangroves differ significantly from the mangrove associates under the same ecological and bio-climatic conditions. In order to test this hypothesis, the leaf litter decay rates of 24 true mangrove species and 10 mangrove associates along with the concomitant carbon and nitrogen dynamics of the litters were studied in the tropical mangrove forest of Sundarban by means of litter bags. The decomposition was monitored for six consecutive weeks in the pre-monsoon, monsoon and post-monsoon season. All the species in general went through a rapid decay phase in the first 2 weeks, however, the rate substantially decreased in the following 4 weeks. Most of the species studied had significant seasonal variability (p < 0.05) in the decay rate. Species-specific decay was highest throughout the monsoon and least during the post-monsoon season. The mean dry weight composition (i.e. percentage of dry weight of the leaf litters remaining at the end of weekly intervals) of the true mangroves was 10–12 % higher than the mangrove associates throughout the sampling period. The mean decay constants (K in week?1) of the true mangroves were 0.15 ± 0.05, 0.20 ± 0.06 and 0.16 ± 0.05 in the pre-monsoon, monsoon and post-monsoon season respectively. The mangrove associates had significantly higher decay constants in the respective seasons that followed the order 0.23 ± 0.09, 0.25 ± 0.06 and 0.24 ± 0.09. As a consequence, the computed mean half-life period of the true mangrove litters (32 ± 11 days) was much higher than the mangrove associates (23 ± 11 days). This showed that collectively the leaf litters of mangrove associates degraded at a much faster rate than the true mangroves throughout the annual cycle and thus our hypothesis was justified.  相似文献

Diatoms constitute an important component of the fouling community. Although a lot of work has dealt with the fouling diatom community structure, work on the species interactions within the community is still meagre. In this regard, a study was carried out by transferring natural diatom biofilms into controlled conditions in order to understand the roles of nutrients, initial cell inoculum and seasonal variation in species composition in structuring the fouling diatom community. This community exhibited seasonal variation during the monsoon, post-monsoon and pre-monsoon periods. During each of these seasons, diatom species interactions varied depending upon the species composition. It was observed that excess nutrients favoured those species with comparatively higher growth rates, thereby suppressing the growth of other co-existing species. This competitive trait was found to be effective at an appropriate cell density ratio of the competitive and target species. Understanding such pathways will be useful for modelling the interactions between diatom species in various habitats under different resource conditions.  相似文献

Heterotrophic microorganisms are widely recognized as crucial components of ecosystems; yet information on their community structure and dynamics in benthic freshwater habitats is notably scarce. Using denaturing gradient gel electrophoresis (DGGE), we determined the composition of bacterial and fungal communities in a freshwater marsh over four seasons. DGGE revealed diverse bacterial communities in four contrasting microhabitats. The greatest compositional differences emerged between water-column and surface-associated bacteria, although communities associated with sediment also differed from those on plant litter and epiphytic biofilms. Sequences of bacterial clones derived from DGGE bands belonged to the Alphaproteobacteria (31%), Actinobacteria (19%) and Bacteriodetes (19%). Betaproteobacteria were notably absent. Fungal clones obtained from leaf litter were mainly Ascomycota , but two members of the Basidiomycota were also identified. Overall, habitat type was the most important factor explaining variation in bacterial communities among samples, whereas temporal patterns in community composition were less pronounced in spite of large seasonal variation in environmental conditions such as temperature. The observed differences among bacterial communities in different microhabitats were not caused by random variation, but rather appeared to be determined by habitat characteristics, as evidenced by largely congruent community profiles of replicate samples taken at 10–100 m distances within the marsh.  相似文献

The aim of this study was to investigate and compare the microbial community structures of eubacteria and archaea in the pit mud of Chinese Luzhou-flavor liquor from the wall (Cw) and bottom (Cb) of cellar through nested PCR–denaturing gradient gel electrophoresis (DGGE). The Shannon–Wiener index (H) calculated from the DGGE profiles showed that the community diversities of eubacteria and archaea in samples from Cb were almost higher than that from Cw. In addition, cluster analysis of the DGGE profiles revealed that some differences were found in the microbial community structure in samples from different locations. The closely relative microorganisms of all eubacterial 16S rRNA gene sequences fell into four phyla (Firmicutes, Proteobacteria, Bacteroidetes and Actinobacteria), including 12 genera and 2 uncultured eubacteria. Moreover, 37.1 % eubacteria were affiliated with Clostridium. Particularly, genus Acinetobacter was absent in all samples from Cb but present in all samples from Cw. The closely relative microorganisms of all archaeal 16S rRNA gene sequences fell into four genera, which included Methanobrevibacter, Methanoculleus, Methanobacterium and Methanosaeta, while the dominant archaea in samples from Cw and Cb were similar. Results presented in this study provide further understanding of the spatial differences in microbial community structure in the pit mud, and is of great importance for the production and quality improvement of Luzhou-flavor liquor.  相似文献

Elevated CO2 generally increases plant productivity, and has been found to alter plant community composition in many ecosystems. Because soil microbes depend on plant-derived C and are often associated with specific plant species, elevated CO2 has the potential to alter structure and functioning of soil microbial communities. We investigated soil microbial community structure of a species-rich semi-natural calcareous grassland that had been exposed to elevated CO2 (600 μL L?1) for 6 growing seasons. We analysed microbial community structure using phospholipid fatty acid (PLFA) profiles and DNA fingerprints obtained by Denaturing Gradient Gel Electrophoresis (DGGE) of 16S rDNA fragments amplified by the Polymerase Chain Reaction (PCR). PLFA profiles were not affected by CO2 enrichment and the ratio of fungal and bacterial PLFA did not change. Ordination analysis of DNA fingerprints revealed a significant relation between CO2 enrichment and variation in DNA fingerprints in summer (P=0.01), but not in spring. This variation was due to changes in low-intensity bands, while dominant bands did not differ between CO2 treatments. Diversity of the bacterial community, as assessed by number of bands in DNA fingerprints and calculation of Shannon diversity indices, was not affected by elevated CO2. Overall, only minor effects on microbial community structure were detected, corroborating earlier findings that soil carbon inputs did probably change much less than suggested by plant photosynthetic responses.  相似文献

Mangrove ecosystems are significant sources of greenhouse gases (GHG) that is attributed to microbial activity. However, it is still unknown how the sediment microbial populations affect GHG emissions in mangrove ecosystem. Since little is known about microbial populations of mangroves, the present study was aimed to understand the structure and function of microbial communities in the Indian part of the Sundarban mangrove ecosystem in relation to environmental variables and variation of GHG emissions during three seasons: pre-monsoon (March–June), monsoon (July–October) and post-monsoon (November–February).Seasonal variations of carbon dioxide (CO2), methane (CH4) and nitrous oxide (N2O) gas samples were taken from the mangrove bed. Culture methods were used to detect twelve different types of microbes such as heterotrophic (Htp), N2 fixing (Nfix), nitrifying (Ntfn), sulfur oxidizing (Soxd), Gram-negative (GMn), Gram-positive (GMp), spore forming (Sfor), denitrifying (DNtfn), anaerobic (Anrb), phosphate solubilizing (Psol), cellulose degrading (Cdeg) bacteria and actinomycetes (Actm). In the monsoon, populations of the Htp, Anrb, Psol, and Cdeg bacteria were more prevalent, whereas populations of the GMn, GMp, Ntfn, DNtfn bacteria, and Actm bacteria were more prevalent in the post-monsoon. Monsoonal CO2 and CH4 fluxes were larger than pre-monsoon and post-monsoon, resulting in increased microbial soil respiration and breakdown of soil organic carbon. Because of higher denitrification and soil temperature, N2O flux was higher in the pre-monsoon period, followed by monsoon and post-monsoon periods. A univariate statistical correlation was employed to assess the relationships between environmental variables and different microbial populations. An ANN (artificial neural network) model was proposed to evaluate the relevance of microbial population contribution to GHG emissions, and it indicated that the Htp, Anrb, and Dntfn microbial populations were most relevant for CO2, CH4, and N2O emissions. The suggested model would be used to assess the drivers behind GHG emission in the mangroves located at different parts of the world.  相似文献

Diurnally subperiodic Wuchereria bancrofti (DSPWB) filariasis is transmitted by the Downsiomyia nivea. In India, it is prevalent only in the Nicobar district of Andaman and Nicobar Islands. Knowledge of the population dynamics and bio-ecology of vectors is crucial to develop intervention measures. Entomological studies were performed for 12 months by collecting mosquitoes that landed on human baits for feeding in Teressa Island. Age composition, mosquitoes that had laid eggs at least once, finite rate of natural increase (λ) and vector survival reflecting the population dynamics of Do. nivea in relation to transmission of filariasis were estimated. A total of 3625 Do. nivea were collected. Changes in finite rate of increase (λ > 1) during favorable months indicate an increase in the vector population, suggesting the need for intensified intervention. The proportion of mosquitoes completing more than one gonotrophic cycle was higher in months when λ was 1. Do. nivea abundance and its parous densities varied with seasons. Survival of Do. nivea (indicated by the proportion of parous mosquitoes) was lower in the pre-monsoon season than during the monsoon and post-monsoon seasons. The probability of the daily survival of Do. nivea through one gonotrophic cycle was 0.75, with a declining trend as age increased. The maximum transmission of DSPWB occurred during the pre-monsoon, followed by the late monsoon, post-monsoon and monsoon seasons. Perennial transmission is evident. Therefore intervention measures must be planned throughout the year. Prospects of vector control are limited due to the cryptic breeding and diurnal biting habits. Mass chemotherapy with a single dose of an antifilarial drug is a potential option for control.  相似文献

The bacterial, archaeal, and eukaryal diversity in fecal samples from ten Koreans were analyzed and compared by using the PCR-fingerprinting method, denaturing gradient gel electrophoresis (DGGE). The bacteria all belonged to the Firmicutes and Bacteroidetes phyla, which were known to be the dominant bacterial species in the human intestine. Most of the archaeal sequences belonged to the methane-producing archaea but several halophilic archarea-related sequences were also detected unexpectedly. While a small number of eukaryal sequences were also detected upon DGGE analysis, these sequences were related to fungi and stramenopiles (Blastocystis hominis). With regard to the bacterial and archaeal DGGE analysis, all ten samples had one and two prominent bands, respectively, but many individual-specific bands were also observed. However, only five of the ten samples had small eukaryal DGGE bands and none of these bands was observed in all five samples. Unweighted pair group method and arithmetic averages clustering algorithm (UPGMA) clustering analysis revealed that the archaeal and bacterial communities in the ten samples had relatively higher relatedness (the average Dice coefficient values were 68.9 and 59.2% for archaea and bacteria, respectively) but the eukaryal community showed low relatedness (39.6%).  相似文献

Acanthamoeba rhysodes has been found to be a predominant intertidal benthic gymnamoeba in the mangrove ecosystem of Sundarbans of lower deltaic Bengal, facing the Bay. The sampling zones under study were the highest high tide regions, with characteristic mangrove litter-soil, inundated twice per month during the highest ebb of spring tide. Population abundance of this species, both in its trophic and cystic forms in the three distinct seasonal periods of pre-monsoon (March to June), monsoon (July to October), and post-monsoon (November to February) has been surveyed for over two years. These seasonal periods affect the physico-chemical parameters of the habitat substrata, including temperature, pH, and salinity. It has been found that the overall number of organisms per gram of soil attains peak value during the monsoon period. This value comes down in post-monsoon samples and is the least in pre-monsoon ones.  相似文献

Bacillus subtilis B579, which was isolated from rhizosphere of cucumber, exhibited an excellent biocontrol activity on soil-born pathogens under greenhouse conditions. It could colonize in rhizosphere of cucumber with large number of populations after inoculated in plant growth season. To reveal the effect of high level colonization of B. subtilis B579 on rhizobacteria community structure, cultivation-based analysis coupled with denaturing gradient gel electrophoresis (DGGE) analysis were used to profile the changes of rhizobacteria community structure sampling at 1 week interval. Cultivation-based and DGGE fingerprinting analysis showed significant plant-dependent and seasonal shifts in rhizobacteria populations. Only minimal and transient effects were observed at 4–9 weeks after sowing in samples of B579 treatment, without the pathogen inoculation and showed the best plant growth potential. Sequencing of dominant bands excised from the gel revealed that Streptomyces sp. was the dominate species in soils before and after sowing. Burkholderia sp. was the dominate species in bulk soil, while Bacillus sp. was dominated in rhizosphere within the growth season. Arthrobacter ramosus and Nocardioides sp. were identified as the specific species in samples treated by B579 at the maturity and flowering stages of cucumber.  相似文献

Community composition of ammonia-oxidizing archaea (AOA) and ammonia-oxidizing bacteria (AOB) in the albic soil grown with soybean and rice for different years was investigated by construction of clone libraries, denaturing gradient gel electrophoresis (DGGE), and quantitative polymerase chain reaction (q-PCR) by PCR amplification of the ammonia monooxygenase subunit A (amoA) gene. Soil samples were collected at two layers (0–5 and 20–25 cm) from a soybean field and four rice paddy fields with 1, 5, 9, and 17 years of continuous rice cultivation. Both the community structures and abundances of AOA and AOB showed detectable changes after conversion from soybean to rice paddy judged by clone library, DGGE, and q-PCR analyses. In general, the archaeal amoA gene abundance increased after conversion to rice cultivation, while bacterial amoA gene abundance decreased. The abundances of both AOA and AOB were higher in the surface layer than the bottom one in the soybean field, but a reverse trend was observed for AOB in all paddy samples regardless of the duration of paddy cultivation. Phylogenetic analysis identified nine subclusters of AOA and seven subclusters of AOB. Community composition of both AOA and AOB was correlated with available ammonium and increased pH value caused by flooding in multiple variance analysis. Community shift of AOB was also observed in different paddy fields, but the two layers did not show any detectable changes in DGGE analysis. Conversion from soybean to rice cultivation changed the community structure and abundance of AOA and AOB in albic agricultural soil, which requires that necessary cultivation practice be followed to manage the N utilization more effectively.  相似文献

Shifts in bacterioplankton community composition along the salinity gradient of the Parker River estuary and Plum Island Sound, in northeastern Massachusetts, were related to residence time and bacterial community doubling time in spring, summer, and fall seasons. Bacterial community composition was characterized with denaturing gradient gel electrophoresis (DGGE) of PCR-amplified 16S ribosomal DNA. Average community doubling time was calculated from bacterial production ([14C]leucine incorporation) and bacterial abundance (direct counts). Freshwater and marine populations advected into the estuary represented a large fraction of the bacterioplankton community in all seasons. However, a unique estuarine community formed at intermediate salinities in summer and fall, when average doubling time was much shorter than water residence time, but not in spring, when doubling time was similar to residence time. Sequencing of DNA in DGGE bands demonstrated that most bands represented single phylotypes and that matching bands from different samples represented identical phylotypes. Most river and coastal ocean bacterioplankton were members of common freshwater and marine phylogenetic clusters within the phyla Proteobacteria, Bacteroidetes, and Actinobacteria. Estuarine bacterioplankton also belonged to these phyla but were related to clones and isolates from several different environments, including marine water columns, freshwater sediments, and soil.  相似文献

The complex structure of soil and the heterogeneity of resources available to microorganisms have implications for sampling regimens when the structure and diversity of microbial communities are analyzed. To assess the heterogeneity in community structure, archaeal communities, which typically contain sequences belonging to the nonthermophilic Crenarchaeota, were examined at two contrasting spatial scales by using PCR-denaturing gradient gel electrophoresis (DGGE) analysis followed by unweighted pair group method with arithmetic mean analysis of 16S rRNA- and ribosomal DNA-derived profiles. A macroscale analysis was carried out with soil cores taken at 2-m intervals along triplicate 8-m transects from both managed (improved) and natural (unimproved) grassland rhizosphere soils. A microscale analysis was carried out with a single soil core by assessing the effects of both sample size (10, 1, and 0.1 g) and distance between samples. The much reduced complexity of archaeal profiles compared to the complexity typical of the bacterial community facilitated visual comparison of profiles based on band presence and revealed different levels of heterogeneity between sets of samples. At the macroscale level, heterogeneity over the transect could not be related to grassland type. Substantial heterogeneity was observed across both improved and unimproved transects, except for one improved transect that exhibited substantial homogeneity, so that profiles for a single core were largely representative of the entire transect. At the smaller scale, the heterogeneity of the archaeal community structure varied with sample size within a single 8- by 8-cm core. The archaeal DGGE profiles for replicate 10-g soil samples were similar, while those for 1-g samples and 0.1-g samples showed greater heterogeneity. In addition, there was no relationship between the archaeal profiles and the distance between 1- or 0.1-g samples, although relationships between community structure and distance of separation may occur at a smaller scale. Our findings demonstrate the care required when workers attempt to obtain a representative picture of microbial community structure in the soil environment.  相似文献

The community structure of rhizosphere soil Archaea from three grassland types, associated with different management practices, was examined at a site in the Borders region of Scotland, by analysis of 16S rRNA gene fragments amplified from 16S rDNA and from rRNA. Denaturing gradient gel electrophoresis (DGGE) and sequence analysis of amplified products indicated high relative abundance within the archaeal community of two distinct lineages of non-thermophilic (group 1) Crenarchaeota. Grassland management practices influenced archaeal community structure, as characterized by both 16S rRNA- and 16S rDNA-derived DGGE profiles. One band dominated DGGE profiles in all three grassland types examined, and reproducible differences in the presence and intensity of bands were observed between profiles from managed and natural grassland sites. Analysis of 16S rRNA-derived amplicons from managed and natural grasslands at sites in the north of England and the north of Wales also indicated high relative abundance of non-thermophilic crenarchaeotes within the archaeal community. The band dominating the Scottish grassland site also dominated DGGE profiles from the English and Welsh sites, and similar differences were seen between profiles derived from soils subjected to different management regimes. The study indicates that grassland archaeal communities are dominated by Crenarchaeota, with closely related members of this lineage ubiquitous in distribution in UK upland pasture, and indicate that management practices influence the nature of the crenarchaeotal community.  相似文献

Denaturing gradient gel electrophoresis (DGGE) and terminal restriction fragment length polymorphism (T-RFLP) were used to characterise the changes that occurred in Bacillus cereus group strains present in the phylloplane of clover Trifolium hybridum over 4 months. These strains had previously been analysed by multiple locus sequence typing (MLST). DGGE displayed many equally intense bands which indicated many equally abundant ribotypes. The bacterial community composition was variable and the leaves sampled as little as a week apart were found to have some temporal variability, indicating that diverse phylloplane bacterial communities follow sequential patterns from time to time. The B. cereus group community clearly clustered into early, mid and late branches, possibly due to multiple successional sequences occurring during growing seasons. The functionally and phylogenetically diverse microbial communities appeared to exhibit predictable successional patterns over shorter time scales. DGGE analysis with the molecular marker rpoB gave better resolution than 16S rRNA amplicons. There were no strong similarities between the dendrograms produced by DGGE, MLST and T-RFLP and the clustering produced by the automated T-RFLP method was variable even between the three restriction enzymes used. The DGGE–MLST method emerged as a superior method to T-RFLP–MLST for rapid typing of bacterial communities.  相似文献

Tree-rings of Pinus kesiya from southern region of Manipur, Northeast India were used to develop chronologies of multiple tree-ring parameters that are: total-ring width (TRW), earlywood width (EW), latewood width (LW) and adjusted latewood (LWadj). The time span of these chronologies is 39 years (1980–2018 C.E.) and we compared their growth responses with monthly and daily climatic records. The comparison revealed a broadly consistent pattern of climate sensitivity with daily climate exhibiting higher correlation. The climate signals during pre-monsoon (March–May) were recorded in TRW and EW, whereas late-monsoon to post-monsoon climate signals were recorded in LW and LWadj. The spatial correlation analysis of tree-ring parameters and global sea surface temperature (SST) showed a positive relationship between tree growth with tropical Pacific Ocean and Indian Ocean during winter (December–February) and pre-monsoon (March–May) seasons. The LW and LWadj were also correlated with peak summer monsoon (July–August) SST over the tropical Pacific Ocean. IADFs observed in EW (E-IADF) were caused by dry and warm conditions during March–April. IADFs in LW (L-IADF) occurred due to a combination of enhanced rainfall and temperature during post-monsoon (October–November) season. Evidence of stand-specific micro-climatic conditions in the formation IADFs in this species was also found. This study showed that multiple parameters of P. kesiya provides a lucid understanding of climate response on its growth and can be considered as a proxy for studying sub-seasonal changes in past environmental conditions in longer records.  相似文献

Aims:  To explore the association of microbial community structure with the development of eutrophication in a large shallow freshwater lake, Lake Taihu.
Methods and Results:  The bacterial and archaeal assemblages in sediments of different lake areas were analysed using denaturing gradient gel electrophoresis (DGGE) of amplified 16S rDNA fragments. The bacterial DGGE profiles showed that eutrophied sites, grass-bottom areas and relatively clean sites with a eutrophic (albeit dredged) site are three respective clusters. Fifty-one dominant bacterial DGGE bands were detected and 92 corresponding clones were sequenced, most of which were affiliated with bacterial phylotypes commonly found in freshwater ecosystems. Actinobacteria were detected in the centre of the lake and not at eutrophied sites whereas the opposite was found with respect to Verrucomicrobiales . Twenty-five dominant archaeal DGGE bands were detected and 31 corresponding clones were sequenced, most of which were affiliated with freshwater archaeal phylotypes.
Conclusions:  The bacterial community structures in the sediments of different areas with similar water quality and situation tend to be similar in Taihu Lake.

We have studied the distribution of the archaeal communities in Rambla Salada (Murcia, Spain) over three different seasons and observed the influence upon them of the environmental variables, salinity, pH, oxygen and temperature. Samples were collected from three representative sites in order to gain an insight into the archaeal population of the rambla as a whole. Denaturing gradient gel electrophoresis patterns and diversity indexes indicate that the diversity of the archaeal community in Rambla Salada changed mainly according to the season. We found no significant differences between the types of sample studied: watery sediments and soils. The upwelling zone showed most diversity in its archaeal community. The overall archaeal community was composed mainly of Halobacteriales and Thermoplasmatales, accounting for 72.6 and 12.1 % of the total, respectively. Haloarcula was the most abundant genus, being present at all three sites during all three seasons. Some few Crenarchaeota were always found, mainly at low-salinity levels. Ordination canonical correspondence analysis demonstrated that salinity affected the structure of the community significantly, whilst pH, oxygen and temperature did so to a lesser extent. Most Halobacteriales correlated positively with salinity and pH, whilst Thermoplasmatales correlated negatively with both salinity and pH and positively with temperature and oxygen. The archaeal community with the highest diversity was sampled during June 2006, the season with the highest salt concentration. Catalyzed reporter deposition-fluorescence in situ hybridization showed that the percentage of archaea in Rambla Salada compared to the total number of microorganisms (as measured by DAPI) ranged from 11.1 to 16.7 %. Our research group had isolated the most abundant taxon, Haloarcula, previously in Rambla Salada using classical culture techniques, but on this occasion, using culture-independent methods, we were also able to identify some phylotypes, Halorubrum, Methanolobus, Natronomonas, Halomicrobium, Halobacterium, Halosimplex, uncultured Thermoplasmatales and uncultured Crenarchaeota, that had remained undetected during our earlier studies in this habitat.  相似文献
