Changes in growth parameters and 14CO2 and [U-14C]-sucrose incorporation into the primary metabolic pools and essential oil were investigated in leaves and stems of M. spicata treated with etherel and gibberellic acid (GA). Compared to the control, GA and etherel treatments induced significant phenotypic changes and a decrease in chlorophyll content, CO2 exchange rate, and stomatal conductance. Treatment with etherel led to increased total incorporation of 14CO2 into the leaves wheras total incorporation from 14C sucrose was decreased. When 14CO2 was fed, the incorporation into the ethanol soluble fraction, sugars, organic acids, and essential oil was significantly higher in etherel treated leaves than in the control. However, [U-14C]-sucrose feeding led to decreased label incorporation in the ethanol-soluble fraction, sugars, organic acids, and essential oils compared to the control. When 14CO2 was fed to GA treated leaves, label incorporation in ethanol-insoluble fraction, sugars, and oils was significantly higher than in the control. In contrast, when [U-14C]-sucrose was fed the incorporation in the ethanol soluble fraction, sugars, organic acids, and oil was significantly lower than in the control. Hence the hormone treatment induces a differential utilization of precursors for oil biosynthesis and accumulation and differences in partitioning of label between leaf and stem. Etherel and GA influence the partitioning of primary photosynthetic metabolites and thus modify plant growth and essential oil accumulation. Changes in photosynthetic carbon metabolism during the glucosebleaching of Chlorella protothecoides cells were investigatedusing NaH14CO3 as tracer. Several hours after incubating thegreen algal cells in the glucose medium in the dark, the ratesof 14C-incorporation into glucose polymers and sucrose decreasedand the incorporation into the lipid fraction (fatty acids)greatly increased. At this stage, the rate of photosynthetic14CO2 fixation and the chlorophyll content were practicallythe same as in the starting green cells. Afterwards, the photosyntheticcapacity and chlorophyll content continued to decrease throughoutthe experimental period. Assimilate distribution in leaves of Lolium temulentum was establishedby root absorption of [14C]sucrose and after exposure to 14CO2.Age determined the amount of carbon assimilated, with more labelbeing incorporated during expansion than at maturity. Duringsenescence 14C assimilation was much lower. Ethanol-solubleextracts from various tissues of root-labelled plants containedmost of the radioactivity chiefly in basic and acidic compounds.The neutral fraction was composed predominantly of sucrose. Sucrose was comparably labelled in leaves from plants fed equalamounts of either [14C]sucrose, glucose, or fructose and onlytraces of labelled monosaccharides appeared in extracts. Radioactive sucrose was translocated rapidly from mature leaveswhereas, in the expanding leaf, carbon incorporation was directedtowards growth and the greater proportion of label present atligule formation was in ethanol-insoluble material. Induced senescence, of a mature leaf fed during expansion, produceda rapid loss from the pool of insoluble 14C. This was accompaniedby a reduction in the contents of chlorophyll and soluble proteinand an accumulation of amino acids. The onset of senescencecaused changes in leaf sugar levels which were correlated withincreased rates of respiration.  相似文献

A new method is reported for the histochemical localizationof monoterpene phenols in essential oil secretory structures.The method was adapted from a spot test originally devised forin vitro detection of phenolic compounds in organic analyses.Plant subjects were the Lamiaceae species Thymus vulgaris L.,Oreganum vulgare L. and Mentha x piperita L., which accumulateessential oil in glandular trichomes. A reagent consisting of4-nitrosophenol in conc. H2SO4was applied to sample leaves ofeach species. A positive test for phenol was indicated by theproduction of coloured indophenols. Using this method, monoterpenephenols were identified in the trichomes of T. vulgaris(thymol)and O. vulgare(carvacrol), indicated by colour changes to redand green respectively. No phenol was detected in trichomesof M. x piperita. Results were confirmed by GC-MS analysis ofleaf volatile extracts from each species, and in vitro testswith thymol and carvacrol. The method could be used in fieldsurveys for rapid identification of potential medicinal plantsand bioactive compounds. Petioles of Sinapis cotyledons cultured in 6x10–2 M sucrosein the light increased 28-fold in total carbohydrate contentover a 7-d period compared with an 8-fold increase in petiolesof cotyledons cultured on water. The starch and reducing sugarfractions were the major components of this accumulation. Labelledsucrose applied to the petiole base moved quickly up the petioleand into the main veins of the lamina. Some basipetal redistributionoccurred subsequently and after 24 h radioactivity accumulatedstrongly at the petiole base. Culture in sucrose reduced basalaccumulation and increased acropetal movement of the label. Fixation of 14CO2 by petioles remained constant when cotyledonswere cultured in water, whereas in sucrose, fixation fell by50 per cent during the first 2 d. The pattern of inhibitionof fixation matched the pattern of sucrose distribution in thepetiole. Petiolar chlorophyll content remained constant in controlsbut fell rapidly after 4 d culture in sucrose. The results are discussed in relation to the role of petiolarcarbohydrate accumulation in the regulation of CO2 fixation,primordium development, and senescence in this system.  相似文献

When solutions of [14C]glycollate, glycine, serine, glycerate,or glucose were supplied to segments of wheat leaves throughtheir cut bases in the light, most of the 14C was incorporatedinto sucrose in air but in CO2-free air less sucrose was made.The synthesis of sucrose was decreased because metabolism ofserine was partly blocked. Sucrose synthesis from glucose andglycerate in CO2-free air was decreased but to a smaller extent;relatively more CO2 was evolved and serine accumulated. Theeffects of DCMU and light of different wavelengths on metabolismby leaves of L-[U-14C]serine confirmed that simultaneous photosyntheticassimilation of carbon was necessary for the conversion of serineto sucrose. Of various products of photosynthesis fed exogenouslyto the leaves -keto acids were the most effective in promotingphotosynthesis of sucrose and release of 14CO2 from 14C-labelledserine. This suggests that in CO2-free air the metabolism ofserine may be limited by a shortage of -keto acid acceptorsfor the amino group. In CO2-free air added glucose stimulatedproduction of CO2 and sucrose from D-[U-14C]- glycerate andno competitive effects were evident even though glucose is convertedrapidly to sucrose under these conditions. In addition to asupply of keto acid, photosynthesis may also provide substratesthat can be degraded and provide energy in the cytoplasm forthe conversion of glycerate to sugar and phosphates and sucrose.  相似文献

The effect of exposure of kidney bean primary leaves to NO2and O3, alone or in combination, on the fate of 13CO2 assimilatedby photosynthesis was examined by 13C-NMR. There were more than70 peaks appearing in the 13C-NMR spectra for substances extractedfrom leaves with 80% ethanol. The 16 relatively well resolvedpeaks corresponded to signals from three sugars, two organicacids and four amino acids. These signals were used to estimatepool sizes and l3C incorporation. Exposures to NO2 and O3 increased the amounts of sucrose andfructose, but not the incorporation of the 13C label during10 min photosynthesis from 13CO2. This suggests the presenceof photo-synthetically inactive pools of sucrose and fructose.Amounts of glycine and serine, and 13C incorporation into them,were increased by the exposure to the pollutants. The incorporationof 13C into alanine was stimulated by exposure to NO2, but notby exposure to O3 alone. The present study shows that with only simple procedures ofsample preparations 13C-NMR provides information on the productsof photosynthesis in leaves stressed by the two air pollutants. The rates of CO2 incorporation into the epidermis of C. communiswere linear and were similar during the completion of opening(2 h) and closing (1 h) movements of stomata. The kinetics of14C turnover between metabolites and the rates of ‘leakage’of metabolites were determined for opening and closing movements.When stomata were opening there was a slow turnover of 14C frommalate chiefly into sugars. Upon stomatal closure 14C was initiallymainly in sugars, malate, and sugar phosphates. Thereafter,there was a slight loss of label from sugar phosphates witha corresponding increase in malate. Starch became labelled duringopening and closing movements. Rates of incorporation of CO2found in the ‘leakage’ fraction were greatest whenstomata were opening. Of the labelled compounds Most‘from the tissue, malate was the most highly labelled whetherstomata were opening or closing. Although interpretation of the turnover patterns is difficultwithout knowledge of pool sizes for the metabolites it is suggestedthat a pool of sugars exists within the guard cells, which havefairly direct and reversible access to carbon from starch andmalate. The implications of loss of malate from guard cellsduring stomatal opening and closing are discussed.  相似文献

To study the wavelength-effect on photosynthetic carbon metabolism,14C-bicarbon-ate was added to Chlorella vulgaris 1 lh suspensionunder monochromatic blue (456 nm) and red (660 nm) light. Thelight intensities were so adjusted that the rates of 14CO2 fixationunder blue and red light were practically equal. Analysis of14C-fixation products revealed that the rates of 14CO2 incorporationinto sucrose and starch were greater under red light than underblue light, while blue light specifically enhanced 14CO2 incorporationinto alanine, aspartate, glutamate, glutamine, malate, citrate,lipid fraction and alcohol-water insoluble non-carbohydratefraction. Pretreatment of the algal cells in phosphate mediumin the dark, which was essential for the blue light enhancementof PEP carboxylase activity, was not necessary to induce theabove wavelength effects. Superimposition of monochromatic bluelight at low intensity (450 erg.cm–2.sec–1) on thered light at saturating intensity caused a significant decreasein the rate of 14CO2 incorporation into sucrose and increasein incorporation into alanine, lipid-fraction, aspartate andother related compounds, indicating that the path of carbonin photosynthesis is regulated by short wavelengdi light ofvery low intensity. A study has been made of photosynthetic 14CO2 fixation by isolated‘mature’ internodes of Nitella translucens. Experimentalconditions were similar to those used in studies of the ionicrelations of these cells. Maximum rates of photosynthesis were33–40µµmoles CO2, fixed per cm2 of surfacearea per second (equivalent to 12–15 /xmoles fixed permg chlorophyll per hour). l4CO2 fixation was inhibited to thedark level by 3(3,4,dichlorophenyl)-1, 1-dimethylurea (at 0-6µM or 10µM) and by the uncoupler carbonyl cyanide-m-chlorophenylhydrazone(SµM). The presence of imidazole or ammonium sulphate(both of which uncouple ATP production in vitro) did not resultin an inhibition of 14CO2 fixation. These results are discussedin relation to published work on solute uptake by Nitella translucens.During photosynthesis there was rapid movement of 14C-labelledorganic compounds out of the chloroplasts. 14C-labelled sucrose,ammo-acids, and sugar phosphates were found in samples of vacuolarsap.  相似文献

The pattern for primary products of CO2-fixation and the chloroplaststructure of Amaranthus retrqflexus L., a species which incorporatescarbon dioxide into C4 dicarboxylic acids as the primary productof photosynthesis, were compared in various chlorophyll containingtissues,i.e., foliage leaves, stems, cotyledons and pale-greencallus induced from stem pith. Despite some morphological differencesin these assimilatory tissues, malate and aspartate were identifiedas the major compounds labelled during a 10 sec fixation of14CO2 in all tissues. Whereas, aspartate was the major componentin C4-dicarboxylic acids formed in foliage leaves, malate predominatedas the primary product in stems, cotyledons and the pale-greencallus. Carbon dioxide enhanced chloroplast regeneration in glucose-bleachedcells of Chlorella protothecoides in the presence of CMU inthe light. Both the formation of chlorophyll and the synthesesof RNA and protein were considerably enhanced. The CO2 metabolism of algal cells during greening was investigatedusing 14C-bicarbonate as the tracer. Ammonia Induces Starch Degradation in Chlorella Cells   总被引:3,自引:0,他引:3  
When ammonia was added to cells of Chlorella which had fixed14CO2 photo synthetically, 14C which had been incorporated intostarch was greatly decreased. A similar effect was observedwhen potassium nitrate and sodium nitrite were added. The ammonia-induceddecrease in 14C-starch was observed in all species of Chlorellatested. With cells of C. vulgaris 11h, most of the radioactivityin starch was recovered in sucrose, indicating that ammoniainduces the conversion of starch into sucrose. The percent of14C recovered in sucrose differed from species to species andpractically no recovery in sucrose was observed in C. pyrenoidosa.In most species tested, the enhancing effects of blue lightand ammonia on O2 uptake as well as the ammonia effect on starchdegradation were greater in cells which had been starved inphosphate medium in the dark than in non-starved cells. In contrast,the enhancing effect of ammonia on dark CO2 fixation was muchgreater in non-starved cells. C. pyrenoidosa was unique in thatblue light did not show any effect on its O2 uptake. Levels of the activities of glutamine-dependent carbamylphosphatesynthetase, ornithine-and aspartate-transcabamylase and phosphoenolpyruvatecarboxylase were followed in greening cells of Chlorella prolothecoides.Among the enzymes examined the activity of carbamylphosphatesynthetase was extremely low, especially at the early phaseof greening. Arginine (but not ornithine or aspartate), when administeredto algal cells at the 24th hour of greening, stimulated thesyntheses of RNA, protein and chlorophyll in the subsequentperiod. It also affected the metabolic pathway of the 14CO2supplied simultaneously with arginine in the presence of CMU.Arginine produced a decreased incorporation of 14C into proteinand an increased incorporation into nucleic acid. The mechanismof the action of CO2 on chloroplast regeneration is discussed.We concluded that chloroplast regeneration in glucose-bleachedcells is limited by the synthesis of carbamylphosphate, especiallyin the early phase of greening. Purine alkaloid catabolism pathways in young, mature and agedleaves of tea (Camellia sinensis L.) were investigated by incubatingleaf sections with 14C-labelled theobromine, caffeine, theophyllineand xanthine. Incorporation of label into CO2 was determinedand methanol-soluble metabolites were analysed by high-performanceliquid chromatography-radiocounting and thin layer chro-matography.The data obtained demonstrate that theobromine is the immediateprecursor of caffeine, which accumulates in tea leaves becauseits conversion to theophylline is the rate limiting step inthe purine alkaloid catabolism pathway. The main fate of [8-14C]theophyllineincubated with mature and aged leaves, and to a lesser extentyoung leaves, is conversion to 3-methylxanthine and onto xanthinewhich is degraded to 14CO2 via the purine catabolism pathway.However, with young leaves, sizable amounts of [8-14C]-theophyllinewere salvaged for the synthesis of caffeine via a 3-methylxanthine  相似文献

In Daucus carota cells cultivated in vitro, the ammonium ionstimulates the incorporation of radioactivity from labelledglucose and labelled pyruvate into CO2 and into the residueinsoluble in 60 per cent (v/v) ethanol. There is a higher 14CO2production from [6-14C2] glucose than from [6-14C] glucose.These results suggest a possible stimulation of glycolysis bythe ammonium ion.  相似文献

A study was made of the incorporation of 14C by intact leavesof Coffea arabica (cultivars Mundo Novo, Catuai, 1130–13,and H 6586–2) and Coffea canephora (cultivar Guarini)supplied with gas mixtures containing 14CO2 under controlledconditions. Samples of the leaves were combusted and the 14Cin the CO2 produced measured using a liquid scintillation counter.The results were used to estimate photosynthetic rates. Theeffects of changing the partial pressures of O2 and CO2 on thephotosynthetic rate were studied and estimates made of the CO2compensation point and photorespiration. The data obtained show differences between the mean net photosyntheticrates of the C. arabica cultivars (6·14 mg CO2 dm–2h–1) and the mean rate for the C. canephora cultivar (3·96mg CO2 dm–2 h–1). The cultivar of the latter speciesphotorespired more rapidly than the cultivar Catuai of C. arabica.Rates of photosynthesis in coffee measured using the 14CO2 methodwere similar to rates obtained by others using an infrared gasanalyser. The 14CO2 method proved to be reliable for photosyntheticmeasurements and the apparatus is suitable for use in fieldconditions.  相似文献

Leaves of different ages from B. calycinum were exposed to 14CO2in light during day and night. The labelling pattern on thechromatogram differed with leaf age. Young leaves had similarpatterns to those of C3 plants during both day and night. Matureleaves showed high incorporation of 14C into C4 acids, especiallyat night. In contrast, no significant difference with leaf agewas observed in the pattern of dark 14CO2 fixation products.Study of the enzyme activity and the content of titratable acidat each leaf age suggested that high incorporation of 14C inC4 acids during the night was due to the simultaneous absorptionof CO2 by both enzymes RuDPcarboxylase and PEPcarboxylase. In order to study the effects of inorganic phosphate (P1) starvationon C4plants, 3-week-old maize plants (Zea maysL cv. Brulouis)were grown in a growth chamber on a nutrient solution withoutP1 over 22 d During the first 2 weeks, Pi-starved plants grewas well as control plants The Pi concentration in the planttissue decreased rapidly with time, which suggests that normalbiomass production can be maintained at the expense of internalP1 In addition, photosynthetic CO2 assimilation measured 4-6h after dawn was not affected, but the concentration of glucose,sucrose, and starch in leaves was much higher than in the controls14CO2 pulse-chase experiments earned out on the ninth day oftreatment showed that 14CO2 assimilation was perturbed duringthis initial period, resulting in a larger flow of carbon toboth starch and sucrose At the beginning of the third week ofP1 starvation (15 d after treatment) 14C incorporation intosucrose stayed high relative to controls but this was not thecase for starch At the end of the third week of P1-deficiency,shoot growth was considerably reduced and fresh weight was onlyone-third of that of the control plants. The P1 concentrationof both the leaf and root tissues was less than 1.0 µmolg–1 FW compared to 20-25µmol g1 FW in the controls.Photosynthetic CO2 assimilation was reduced and the leaf concentrationof sucrose and starch, which had begun to decrease after theend of the second week of P1 limitation, became lower than inthe controls. These results obtained on maize plants show thatphotosynthesis and carbon partitioning between sucrose and starchwere strongly affected by P1 deficiency, similar to C3 species. The rates of 14CO2 fixation and translocation of 14C labelledassimilates were measured in field experiments at two timesof the day in two sugar-cane clones known to have differentrates of sucrose uptake in vitro but the same weight of leafper unit weight of cane. The rate of 14CO2 fixation and the velocity and rate of translocationwere significantly greater at both times in the clone with thehigher rate of sucrose uptake in vitro. The velocities of translocationwere 2.18 and 2.36 cm/min–1 for the clone with high sucroseuptake and 1.46 cm min–1 at both times in the clone withlow uptake. It is suggested that among sugar-cane clones the ability oftheir canes to store sugar may play a part in determining theirrates of photosynthesis and translocation.  相似文献
