Two strains belonging to the genus Ectothiorhodospira were isolated from enrichment cultures inoculated with sulfide-containing samples from the saltern of Trapani. Cells are motile short spirilla with internal stacks of membranes. During sulfide utilization they produce external globules of sulfur that are then completely oxidized to sulfate. These halophilic microorganisms need NaCl concentrations of 11% and 18% and a slightly alkaline pH. They are typical photoautotrophic bacteria, utilizing sulfide, sulfur and, only one of them, thiosulfate as photosynthetic electron donors; growth is stimulated by organic compounds. Neither of the two strains is capable of assimilatory sulfate reduction and neither grows in the dark. Pigments of the two strains are bacteriochlorophyll a and carotenoids of the normal spirilloxanthin series subgroup 1B. Quinones are Q8 and MK8 in a strain and Q8 and MK7 in the other one: the latter situation, with quinone side chains of different lengths, is atypical within phototrophic bacteria. For morphological, physiological and biochemical characters, at least one of these strains clearly stays apart from the six Ectothiorhodospira species described until now.List of abbreviations HPLC

Rhodobacter capsulatus strains E1F1 and B10 and Rhodobacter sphaeroides DSM 158 did not use hydroxylamine as nitrogen source for growth but metabolized it mainly through the glutamine synthetase reaction. Hydroxylamine had a high toxicity for cells growing either under phototrophic or dark-aerobic conditions. l-methionine-d,l-sulfoximine partially inhibited hydroxylamine uptake and increased the inhibition time of nitrogenase activity by this nitrogen compound. Nitric oxide was also a powerful inhibitor of nitrogenase in intact cells of R. capsulatus. Since low amounts of NO were produced from hydroxylamine, short-term inhibition of nitrogenase in the presence of this compound could be mediated in vivo by nitric oxide.Abbreviations GS

The phylogenetically related phototrophic bacteria Rhodospirillum tenue and Rhodocyclus purpureus modulate activity of their glutamine synthetases by adenylylation/deadenylylation. Evidence for covalent modification includes the inhibitory effect of Mg2+ on the activity of glutamine synthetase extracted from cells of either species grown on excess ammonia, and the lack of Mg2+ inhibition of activity of the enzyme isolated from N2-(R. tenue) or glutamine (R. purpureus)-grown cells. In addition, snake venom phosphodiesterase treatment of glutamine synthetase from either species grown on excess ammonia relieved Mg2+ inhibition of the enzyme (as measured via the -glutamyl transferase assay), and changed the cation specificity from Mn2+ to Mg2+ (in the biosynthetic assay). 相似文献

485 isolates were recorded from different localities between summer 1984 and spring 1985. These isolates comprise five different genera with eight different species of purple non-sulfur bacteria- Rhodopseudomonas was the most common genus at all sites and was represented by three species. R. acidophila, R. blastica, R. palustris. Rhodomicrobium was the second in frequency and was represented by R. vannielli only. This species was only recorded for khor sites. Rhodocyclus was represented by two species, R. gelatinosus and R. tenuis. Rhodobacter and Rhodospirillum were represented by one species each, Rhodobacter capsulatus and Rhodospirillum rubrum. Khor sites were richer in number of isolates and genera of purple non-sulphur bacteria than the main body of AHDL. Some species were more common in winter and spring while others predominate in summer and autumn. 相似文献

Abstract The sulfur cycle in a microbial mat was studied by determining viable counts of sulfate-reducing bacteria, chemolithoautotrophic sulfur bacteria and anoxygenic phototrophic bacteria. All three functional groups of sulfur bacteria revealed a maximum population density in the uppermost 5 mm of the mat: 1.1 × 108 cells of sulfate reducers cm−3 sediment, 2.0 × 109 cells of chemolithoautotrophs cm−3 sediment, and 4.0 × 107 cells of anoxygenic phototrophs cm−3 sediment. Bacterial dynamics were studied by sulfate reduction rate measurements, both under anoxic conditions (dark incubation) and oxic conditions (incubation in the light), and determination of the vertical distribution of the potential rate of thiosulfate consumption under oxic conditions. Sulfate reduction rates in the top 5 mm of the sediment were 566 nmol cm−3 d−1 in the absence of oxygen, and 123 nmol cm−3 d−1 in the presence of oxygen. In the latter case, the maximum rate was found in the 5–10-mm depth horizon (361 nmol cm−3 d−1 ). Biological consumption of amended thiosulfate was rapid and decreased with depth, while in the presence of molybdate, thiosulfate consumption decreased to 10–30% of the original rate. 相似文献

The genus Enhydrobacter, first reported as a member of the family Vibrionaceae, has been placed in the family Moraxellaceae, but as a genus incertae sedis in Bergey's Manual of Systematic Bacteriology 2nd edition. During our taxonomic investigation of Enhydrobacter-like organisms, we observed that the 16S rRNA sequences of E. aerosaccus-type strain versions NCIMB 12535(T) , ATCC 27094( T) and CCUG 58314(T) were very different from the accessible data (accession no. AJ550856). Phylogenetic analysis of our 16S rRNA sequence data revealed that these organisms were located within the family Rhodospirillaceae. The genera Inquilinus, Oceanibaculum, Skermanella and Nisaea were closely related (sequence similarities were 88.3~87.0%), but Enhydrobacter could be distinguished from these genera by growth characteristics, fatty acid profiles (C(19:0) cyclo ω8c; 38.4% C(18:1) ω7c; 32.2%, and C(16:0) ; 8.9% were major components), in being non-flagellated, and differing in enzymatic activities, including trypsin and β-glucosidase. From these data, we conclude that the genus Enhydrobacter should be recognized as an independent genus of the family Rhodospirillaceae within the class Alphaproteobacteria. 相似文献

After growth in the absence of nitrogenous oxides under anaerobic phototrophic conditions, several strains of Rhodopseudomonas capsulata were shown to possess a nitrous oxide reductase activity. The enzyme responsible for this activity had a periplasmic location and resembled a nitrous oxide reductase purified from Pseudomonas perfectomarinus. Electron flow to nitrous oxide reductase was coupled to generation of a membrane potential and inhibited by rotenone but not antimycin. It is suggested that electron flow to nitrous oxide reductase branches at the level of ubiquinone from the previously characterized electron transfer components of R. capsulata. This pathway of electron transport could include cytochrome c', a component hitherto without a recognized function. R. capsulata grew under dark anaerobic conditions in the presence of malate as carbon source and nitrous oxide as electron acceptor. This confirms that nitrous oxide respiration is linked to ATP synthesis. Phototrophically and anaerobically grown cultures of nondenitrifying strains of Rhodopseudomonas sphaeroides, Rhodopseudomonas palustris, and Rhodospirillum rubrum also possessed nitrous oxide reductase activity. 相似文献

Anaerobic respiration in the Rhodospirillaceae: characterisation of pathways and evaluation of roles in redox balancing during photosynthesis 总被引:2,自引:0,他引:2
Abstract Recent discoveries relating to pathways of anaerobic electron transport in the Rhodospirillaceae are reviewed. The main emphasis is on the organism Rhodobacter capsulatus ** but comparisons are made with Rhodobacter sphaeroides ** f. sp. denitrificans and Rhodopseudomonas palustris . The known electron acceptors for anaerobic respiration in Rhodobacter capsulatus are trimethylamine- N -oxide (TMAO), dimethyl sulphoxide (DMSO), nitrate and nitrous oxide. In each case respiration generates a proton electrochemical gradient and in some cases can support growth on non-fermentable carbon sources. However, the principal objective of this review is to discuss the possibility that, apart from a role in energy conservation, anaerobic respiration in the photosynthetic bacteria may have a special function in maintaining redox balance during photosynthetic metabolism. Thus the electron acceptors mentioned above may serve as auxiliary oxidants: (a) to maintain an optimal redox poise of the photosynthetic electron transport chain; (b) to provide a sink for electrons during phototrophic growth on highly reduced carbon substrates.

Rhodobacter sulfidophilus, R. capsulatus, R. sphaeroides, Rhodospirillum rubrum, Rhodopseudomonas palustris, R. viridis and Rhodocyclus gelatinosus were found to be able to synthesize adenylylsulfate and 3-phosphoadenylylsulfate from sulfate and ATP. The presence of ATP sulfurylase was proven for the soluble protein fractions of all these species. ADP sulfurylase was not found. ATP sulfurylase was purified from R. sulfidophilus. Its molecular weight was 290,000. The enzyme is stabilized by magnesium ions and elevated salinities. The optimal pH was 8.0, activity was found between pH 6.8 and 9.4. The enzyme is inactivated at temperatures above 40°C. Kinetic studies resulted in K

The occurrence and the dynamics of phototrophic purple nonsulphur bacteria (PPNSB) as well as Azospirillum, Azotobacter, Clostridium, and cyanobacteria at different rice growth stages were studied in two ricefields, at Kafr-El-Shiekh and Al-Fayoum in Egypt.The PPNSB existed in the both rice fields examined, but their numbers varied according to field conditions, habitat and rice growth stage. After transplanting, the number of PPNSB increased gradually, reached its maximum at maximum tillering stage, and thereafter declined toward harvest time. Numbers of PPNSB were generally comparable with that of the heterotrophic N2-fixers namely Azospirillum, Azotobacter, Clostridium and cyanobacteria, while that of phototrophic purple and green sulphur bacteria were relatively lower.The highest PPNSB numbers were generally found in rhizosphere (103–106 per g–1 dw soil) followed by soil (103–105 per g–1 dw soil) and floodwater (10–102 per ml). Rice plants showed a positive rhizosphere effect on PPNSB, clostridia, Azotobacter and Azospirillum, negative rhizosphere effect on cyanobacteria and green sulphur bacteria, and no effect on purple sulphur bacteria. 相似文献

Sequences of the 16S rRNA gene were determined from all type strains of the recognized Ectothiorhodospira species and from a number of additional strains. For the first time, these data resolve the phylogenetic relationships of
the Ectothiorhodospiraceae in detail, confirm the established species, and improve the classification of strains of uncertain
affiliation. Two major groups that are recognized as separate genera were clearly established. The extremely halophilic species
were removed from the genus Ectothiorhodospira and reassigned to the new genus Halorhodospira gen. nov., to recognize that the most halophilic eubacteria are species of this genus. These species are Halorhodospira halophila comb. nov., Halorhodospira halochloris comb. nov., and Halorhodospira abdelmalekii comb. nov. Among the slightly halophilic Ectothiorhodospira species, the classification of strains belonging to Ectothiorhodospira mobilis and Ectothiorhodospira shaposhnikovii was improved. Several strains that were tentatively identified as Ectothiorhodospira mobilis form a separate cluster on the basis of their 16S rDNA sequences and are recognized as two new species: Ectothiorhodospira haloalkaliphila sp. nov., which includes the most alkaliphilic strains originating from strongly alkaline soda lakes, and Ectothiorhodospira marina, describing isolates from the marine environment.

Regulatory aspects of the assimilation of inorganic nitrogen compounds (ammonia, nitrate, nitrogen) were studied in 12 strains belonging to the Rhodospirillaceae. All strains possessed an ammonium transport system, as demonstrated by 14C-methylammonium uptake. This uptake showed saturation kinetics (K
m between 50–150 M), and was competitively inhibited by ammonium (K
i between 5–18 M). The ammonium transport systems were repressed by ammonium in the growth medium. The nitrogenase activity of all strains was reversibly inhibited by ammonium (switch-off). This effect was not shown under nitrogen starvation conditions with the exception of some strains of Rhodopseudomonas capsulata, the nitrogenase of which was always susceptible to switch-off by ammonium. 【目的】从海洋沉积物中富集获得硫酸盐还原菌群,改变pH值进行培养,分析pH值对硫酸盐还原性质的影响,明确菌群组成和进行硫酸盐还原功能基因预测,探究硫酸盐还原机制。【方法】分析硫酸盐还原菌群在不同pH值条件下的硫酸盐还原率,在此基础上,利用高通量测序技术和PICRUSt软件分析硫酸盐还原菌群优势菌组成及硫酸盐还原相关基因相对丰度。【结果】硫酸盐还原菌群在不同pH值培养条件下的生长和硫酸盐还原率出现显著变化(P<0.01),在pH 5.0时达到峰值,分别为0.34±0.01和96.52%±0.44%。高通量测序数据显示,pH 5.0时菌群丰富度和多样性最高,优势菌属为假单胞菌(Pseudomonas)和芽孢杆菌(Bacillus),相对丰度较高的基因为同化性硫酸盐还原相关基因。【结论】硫酸盐还原菌富集生长的最适pH 5.0,在此条件下的高硫酸盐还原率由同化性硫酸盐还原途径主导,为揭示硫酸盐还原机制提供了实验支持,并拓宽了硫酸盐还原菌实践应用方面的种质资源。 相似文献

A new phototrophic purple bacterium was isolated from a flat, laminated microbial mat in a salt marsh near Woods Hole, Mass.,
USA. The spiral-shaped bacterium was highly motile and had bipolar tufts of flagella and intracytoplasmic membranes of the
vesicular type. The major photosynthetic pigments were identified as the carotenoid tetrahydrospirilloxanthin and bacteriochlorophyll
b. The long wavelength in vivo absorption maximum of the bacteriochlorophyll was at 986 nm. The marine bacterium showed optimal
growth in the presence of 2% NaCl. It utilized a number of organic substrates as carbon and energy sources and required vitamins
and sulfide as a reduced sulfur source for growth. In the presence of sulfide, elemental sulfur globules were formed outside
the cells. Elemental sulfur was not further oxidized to sulfate. The new isolate had a unique lipid and fatty acid composition,
and according to the 16S rRNA gene sequence, it is most similar to Rhodospirillum rubrum. It is described as a new species and assigned to a new genus with the proposed name Rhodospira trueperi.

Aquatic hyphomycetes play a key role in decomposition of submerged organic matter and stream ecosystem functioning. We examined the phylogenetic relationships among various genera of aquatic hyphomycetes belonging to the Leotiomycetes (Ascomycota) using sequences of internal transcribed spacer (ITS) and large subunit (LSU) regions of rDNA generated from 42 pure cultures including 19 ex-types. These new sequence data were analyzed together with additional sequences from 36 aquatic hyphomycetes and 60 related fungi obtained from GenBank. Aquatic hyphomycetes, characterized by their tetraradiate or sigmoid conidia, were scattered in nine supported clades within the Helotiales (Leotiomycetes). Tricladium, Lemonniera, Articulospora, Anguillospora, Varicosporium, Filosporella, and Flagellospora are not monophyletic, with species from the same genus distributed among several major clades. The Gyoerffyella clade and the Hymenoscyphus clade accommodated species from eight and six different genera, respectively. Thirteen aquatic hyphomycete taxa were grouped in the Leotia-Bulgaria clade while twelve species clustered within the Hymenoscyphus clade along with several amphibious ascomycetes. Species of Filosporella and some species from four other aquatic genera were placed in the Ascocoryne-Hydrocina clade. It is evident that many aquatic hyphomycetes have relatives of terrestrial origin. Adaptation to colonize the aquatic environment has evolved independently in multiple phylogenetic lineages within the Leotiomycetes. 相似文献

In some Rhodospirillaceae, the primary light-harvesting (LH I) antenna absorbs near-infrared light around 870 nm, whereas LH II (holochrome B800-860) has a major absorption band between 850 and 860 nm (B860) and a minor absorbancy around 800 nm (B800). Results show that, unlike LH I, holochrome B800-860 (LH II) exhibits unstable light absorption properties in whole cells. This was observed in Rhodopseudomonas capsulata grown anaerobically in light in weakly buffered carbohydrate medium; cultures lost both carotenoid-dependent brown-yellow pigmentation and LH II absorbancy. The whole cell spectrophotometric changes were attributed to mild acid conditions generated during sugar metabolism. LH II absorbancy was also destroyed in both R. capsulata and Rhodopseudomonas gelatinosa when cultures growing at neutral pH were acidified to a pH value around 5.0 with HCl. In contrast, during the same time period of exposure to pH 5.0, only a 50% decrease in Rhodopseudomonas sphaeroides LH II B800 absorbancy was measured. At neutral pH, LH II absorbancy in suspensions of nongrowing Rhodopseudomonas spp. was also sensitive to O2 exposure and to incubation at 30 to 40 degrees C. During treatment with O2, the rate of LH II B800 absorption decrease in R. gelatinosa and R. sphaeroides was 60 and 40% per h, respectively, compared with their absorbancy maximum around 860 nm. Both 860-nm absorbancy and the total bacteriochlorophyll content of the cells remained unchanged. On the other hand, no significant decrease in B800 if LH II in R. capsulata occurred during O2 exposure, but a 20% absorption decay rate per h of B800 was observed in cells incubated anaerobically at 40 degrees C. These B800 LH II spectral changes Rhodopseudomonas spp. were prevented by maintaining cells at neutral pH and at 10 degrees C. The near-infrared absorption spectrum of Rhodospirillum rubrum, which does not form LH II, was not significantly influenced by these different pH, aerobic, or temperature conditions. 相似文献
