Summary In blood smears from 527 females and 457 males examined for the presence of Y chromosomes in interphase nuclei, 0.6% false-positive results and 11% false-negative results were found. There was a clear tendency for the falsenegative results to occur among those with small fluorescent or non-existing bands on the Y chromosome. The three falsepositive females all had fluorescent chromosomal variants. In a comparison between female samples with and without chromosomal variants respectively, the former showed significantly higher false Y-body counts. There was a decrease in the number of Y-bodies with increasing age. There were no significant differences between staining with 0.1% Quinacrine mustard and 0.1% and 1% Mepacrine. This study provides a] more solid basis for the use of Y chromosome detection in forensic medicine, for screening purposes etc.  相似文献

The epidemiology of Treponema paraluis-cuniculi infection in a commercial rabbit breeding facility was described using several serologic tests. The Venereal Disease Research Laboratory, rapid plasma reagin, microhemagglutination and fluorescent treponemal antibody-absorption tests were used to detect antibodies to T paraluis-cuniculi. Young adult New Zealand white rabbits, tested prior to entry into the breeding program, were nearly always free of T paraluis-cuniculi infection. In adult females, the prevalence of T paraluis-cuniculi infection increased with parity; females para 6 or greater were usually seropositive. Most adult males seroconverted within 6 months of entering the breeding program; all males were seropositive after 12 months in the breeding program. This suggested that T paraluis-cuniculi spreads mainly by horizontal transmission during breeding in adult rabbits. Of the two nontreponemal antigen tests used, the Venereal Disease Research Laboratory test was more sensitive, whereas the rapid plasma reagin test was more specific in detecting T paraluis-cuniculi infection; the fluorescent treponemal antibody-absorption test was used as the confirmatory treponemal antigen test. However, neither nontreponemal antigen test was completely satisfactory. On the other hand, the sensitivity and specificity of the microhemagglutination test compared favorably with the fluorescent treponemal antibody-absorption test. Since the microhemagglutination test combines desirable features of both a screening and verification procedure, it should be the test of choice for detection of T paraluis-cuniculi infection.  相似文献

Objectives: A prospective study to estimate benefits due to fecal occult blood tests for colorectal cancer are carried out for both males and females, under different screening frequencies. Methods: We apply the statistical method developed by Wu et al. (2007) [1] using the Minnesota colorectal cancer study group data, to make Bayesian inference for the lead time, the time of diagnosis advanced by screening for both male and female participants in a periodic screening program. The lead time is distributed as a mixture of a point mass at zero and a piecewise continuous distribution. The two parts of the mixture correspond to two aspects of the screening: the probability of no benefit, or the percentage of interval cases; and the probability distribution of the early diagnosis time. We present estimates of these two measures for males and females by simulation studies using the Minnesota study group data. We also provide the mean, mode, variance, and density curve of the program's lead time by gender. This may provide policy makers important information on the effectiveness of the FOBT screening in colorectal cancer early detection. Results: The mean lead time increases as the screening time interval decreases for both males and females. The standard error of the lead time also increases as the screening time interval decreases for both genders. Females seem get more benefit than males, in that females usually have a longer lead time than males if both take the test at the same time interval and the lead time mode for females is greater than that of males in the same screening time interval. Conclusion: According to the predictive estimation of the lead time distribution, to guarantee a 90% chance of early detection, it seems necessary for the males to take the fecal occult blood test every 9 months, while the females can take it annually.  相似文献

The population dynamics of tundra-nesting Peregrine Falcons Falco peregrinus tundrius was studied over 7 years on a 450 km2 study area in the Keewatin District of Canada's Northwest Territories. Peregrines showed strong fidelity to nest sites; none of 25 males ringed changed territories, while five of 38 females ringed were recorded changing territories. Such changes usually occurred after nesting failure. Annual turnover of territorial adults was estimated to be 22% (15% for males and 26% for females). Annual mortality of adults was estimated to be 17% (15% for males and 19% for females). If we assumed that territory vacancies, in addition to mate replacements, were indicative of mortality, then maximum annual mortality was estimated at 24% for each sex. Territories were held only by adult Peregrines. The oldest male on territory was at least 7 years old, the youngest was 2. The oldest female on territory was at least 7 years old, the youngest was 3. Territories were held by individuals of each sex for at least 6 years. One pair remained together for at least 4 years. Less than 4% of all young Peregrines produced on the study area in the first 5 years of the study were recruited into the breeding population. More male than female young were recruited despite an even sex ratio among nestlings. Peregrines did poorly in their first breeding attempts. The single young female recruited into the study population dispersed more than three times the median dispersal distance of six recruited males, suggesting that other females probably dispersed beyond the boundaries of the study area.  相似文献

Polygyny is widespread among Orkney's Hen Harriers Circus cyaneus . From 1975–81, it was associated with a sex ratio estimated from resightings of colour-marked birds up to six years of age to be 29 females:ten males. The sex ratio of fledglings changed significantly from a greater proportion of females in the 1950s and early 1960s to a greater proportion of males subsequently. The mean estimated 'survival' rates (birds colour-marked in Orkney which were seen there in later years) of males and of females 0–2 years old were 14 and 29%. The mean estimated annual survival rates of males and females from 2–6 years old were 72 and 90%. It is suggested that the uneven sex ratio resulted in more frequent intra-sexual encounters and displays by females. The results of temporary removals of two females (immediately replaced) and of two males (not replaced) in spring indicates that there was a shortage of males in the population.  相似文献

The feeding ecology of the caridean shrimp Palaemon peringueyi in the temporarily open/closed Kasouga Estuary, Eastern Cape, was examined during summer and winter employing stomach fullness index (SFI) analysis. Mean SFI values for male and female shrimp during summer ranged from 0.85% to 2.36% and from 0.56% and 2.61% body dry weight, respectively. During winter, SFI values for males and females were lower, and varied between 0.23% and 1.56% and 0.36% and 1.84% body dry weight, respectively. There were no significant differences in the SFI values between males and females during the two seasons. A peak in the feeding activity for both sexes was observed during the daytime during both summer and winter. For both males and females, non-linear regression analysis best explained the change in SFI over time. The gut evacuation rate constants (k) of males and females during summer were estimated at 0.43 h?1 and 0.30 h?1, respectively. These values correspond to a daily food intake equivalent to 16.2% body dwt for males and 11.1% body dwt for females. During winter, the gut evacuation rates of the shrimp were estimated at 0.35 h?1 for males and 0.28 h?1 for females. The daily ration for adult male and female shrimp during winter was estimated at 6.1% and 6.8% body dwt, respectively. The lower estimates obtained during winter appear largely to reflect the influence of the reduced water temperatures on the metabolic activities of the shrimp.  相似文献

Summary Electrophoretic screening of glucose-6-phosphate dehydrogenase (EC 1.1.1.49, G6PD) was conducted one sample of 9,260 children born to the atomic bomb survivors in Hiroshima (Honshu) and Nagasaki (Kyushu). The prevalence of electrophoretic variants was 0.11% in males and 0.42% in females in Hiroshima, and 0.16% in males and 0.31% in females in Nagasaki. Enzymologic characteristics of 10 variants obtained from three males and seven hemizygous fathers of heterozygous females were examined. As a result, three new types of G6PD variants were identified among five variants detected in Hiroshima, and three new types among five variants in Nagasaki. All the variants except one belonged to Class 3, as defined by Yoshida et al. (1971).  相似文献

The fragile X syndrome is an X-linked mental retardation disorder caused by an expanded CGG repeat in the first exon of the fragile X mental retardation (FMR1) gene. Its frequency, X-linked inheritance, and consequences for relatives all prompt for diagnosis of this disorder on a large scale in all affected individuals. A screening for the fragile X syndrome has been conducted in a representative sample of 3,352 individuals in schools and institutes for the mentally retarded in the southwestern Netherlands, by use of a brief physical examination and the DNA test. The attitudes and reactions of (non)consenting parents/guardians were studied by (pre- and posttest) questionnaires. A total of 2,189 individuals (65%) were eligible for testing, since they had no valid diagnosis, cerebral palsy, or a previous test for the FMR1 gene mutation. Seventy percent (1,531/2,189) of the parents/guardians consented to testing. Besides 32 previously diagnosed fragile X patients, 11 new patients (9 males and 2 females) were diagnosed. Scoring of physical features was effective in preselection, especially for males (sensitivity .91 and specificity .92). Major motives to participate in the screening were the wish to obtain a diagnosis (82%), the hereditary implications (80%), and the support of research into mental retardation (81%). Thirty-four percent of the parents/guardians will seek additional diagnostic workup after exclusion of the fragile X syndrome. The prevalence of the fragile X syndrome was estimated at 1/ 6,045 for males (95% confidence interval 1/9,981-1/ 3,851). On the basis of the actual number of diagnosed cases in the Netherlands, it is estimated that >50% of the fragile X cases are undiagnosed at present.  相似文献

In a Hawaii Hereditary Anemia Screening Project, 4,984 participants were tested for glucose-6-phosphate dehydrogenase (G6PD) deficiency by a filter paper blood spot fluorescence test. Abnormal samples and suspected heterozygotes were checked by quantitative G6PD assay (normal 4.5 to 14 units/g Hb). G6PD was deficient (< 1.5 units/g Hb) in 188 of 2,155 males; 7 other males had low activity (1.5 to 2.8 units/g Hb). The gene frequency, estimated from males after excluding referred and related cases, was 0.037 for Chinese, 0.134 for Filipinos, and 0.203 for Laotians. Among 2,829 females tested, family data showed 111 females were obliged to be at least heterozygous, regardless of G6PD activity, and 43 others had low G6PD activity. Most heterozygotes probably remained undetected by G6PD screening. In 28 females, activity was under 10%; in another 9 females, activity was < 1.5 units/g Hb. Since only 25 homozygotes would be predicted, this apparent excess of females with deficient activity could be due to unequal X-inactivation in some heterozygotes. DNA analysis by polymerase chain reaction amplification and special analytic procedures revealed 10 different missense mutations in 75 males. The nucleotide 835 AT and 1360 CT transitions were first detected in this Hawaiian Project; we found that the nucleotide 1360 mutation was the most common cause of G6PD deficiency in Filipinos. This is the first report of G6PD screening and analysis of molecular G6PD mutations in Filipino and Laotian populations.  相似文献

It is currently not known whether most lung cancers detected by computerized tomography (CT) screening are aggressive and likely to be fatal if left untreated, or if a sizable fraction are indolent and unlikely to cause death during the natural lifetime of the individual. We developed a longitudinal biologically-based model of the relationship between individual smoking histories and the probability for lung cancer incidence, CT screen detection, lung cancer mortality, and other-cause mortality. The longitudinal model relates these different outcomes to an underlying lung cancer disease pathway and an effective other-cause mortality pathway, which are both influenced by the individual smoking history. The longitudinal analysis provides additional information over that available if these outcomes were analyzed separately, including testing if the number of CT detected and histologically-confirmed lung cancers is consistent with the expected number of lung cancers "in the pipeline". We assume indolent nodules undergo Gompertz growth and are detectable by CT, but do not grow large enough to contribute significantly to symptom-based lung cancer incidence or mortality. Likelihood-based model calibration was done jointly to data from 6878 heavy smokers without asbestos exposure in the control (placebo) arm of the Carotene and Retinol Efficacy Trial (CARET); and to 3,642 heavy smokers with comparable smoking histories in the Pittsburgh Lung Screening Study (PLuSS), a single-arm prospective trial of low-dose spiral CT screening for diagnosis of lung cancer. Model calibration was checked using data from two other single-arm prospective CT screening trials, the New York University Lung Cancer Biomarker Center (NYU) (n=1,021), and Moffitt Cancer Center (Moffitt) cohorts (n=677). In the PLuSS cohort, we estimate that at the end of year 2, after the baseline and first annual CT exam, that 33.0 (26.9, 36.9)% of diagnosed lung cancers among females and 7.0 (4.9,11.7)% among males were overdiagnosed due to being indolent cancers. At the end of the PLuSS study, with maximum follow-up of 5.8years, we estimate that due to early detection by CT and limited follow-up, an additional 2.2 (2.0,2.4)% of all diagnosed cancers among females and 7.1 (6.7,8.0)% among males would not have been diagnosed in the absence of CT screening. We also find a higher apparent cure rate for lung cancer among CARET females than males, consistent with the larger indolent fraction of CT detected and histologically confirmed lung cancers among PLuSS females. This suggests that there are significant gender differences in the aggressiveness of lung cancer. Females may have an inherently higher proportion of indolent lung cancers than males, or aggressive lung cancers may be brought into check by the immune system more frequently among females than males.  相似文献

Abstract

Annual survival by age, sex and population size and structure were estimated for bellbirds (Anthornis melanura) from a banding study undertaken on 66 ha Aorangi Island, the Poor Knights Islands, during the period 1978–86. The most parsimonious model showed that survival varied little with age and between years, and survival of males was significantly greater than that of females. Recapture probabilities of males and females were similar, but did vary with age and among years. There was a greater proportion of adult males than adult females and sub‐adult males and females in the population, and among adults the male bias in the population was greater during the breeding season than at other times of the year. The high density of bellbirds on the Poor Knights and larger size of males may explain the reduced survival of females relative to that of males.  相似文献

IntroductionAlcohol use is a global public health problem, including as a risk factor for HIV infection, but few data are available on the epidemiology of alcohol use and alcohol use disorders (AUD) among young people in sub-Saharan Africa.MethodsWe conducted a cross-sectional survey among 4 groups of young people aged 15–24 years old (secondary school students, college/university students, employees of local industries and casual labourers) in two regions (Kilimanjaro and Mwanza) of northern Tanzania. Using a multistage stratified random sampling strategy, we collected information on demographics, alcohol use, and behavioural factors. We screened severity of alcohol use using the Alcohol Use Disorder Identification Test (AUDIT) and estimated the quantity and frequency of alcohol consumption using the timeline-follow-back-calendar (TLFB) method.ResultsA total of 1954 young people were surveyed. The prevalence of reported alcohol use was higher among males (47–70% ever users and 20–45% current users) than females (24–54% ever users and 12–47% current users). Prevalence of use was substantially higher in Kilimanjaro than Mwanza region. In both regions, participants reported high exposure to alcohol advertisements, and wide alcohol availability. College students reported the highest prevalence of current alcohol use (45% among males; 26% among females) and of heavy episodic drinking (71% among males; 27% among females) followed by casual labourers. Males were more likely to have AUD (an AUDIT score ≥8) than females, with 11–28% of males screening positive for AUD. Alcohol use was associated with male gender, being in a relationship, greater disposable income, non-Muslim religion and a higher number of sexual partners.ConclusionsAlcohol use is a significant problem among young people in northern Tanzania. There is an urgent need to develop, pilot and deliver interventions to help young people delay initiation and reduce levels of harmful drinking, particularly among college students and casual labourers.  相似文献

Background

Patients infected by Plasmodium vivax or Plasmodium ovale suffer repeated clinical attacks without primaquine therapy against latent stages in liver. Primaquine causes seriously threatening acute hemolytic anemia in patients having inherited glucose-6-phosphate dehydrogenase (G6PD) deficiency. Access to safe primaquine therapy hinges upon the ability to confirm G6PD normal status. CareStart G6PD, a qualitative G6PD rapid diagnostic test (G6PD RDT) intended for use at point-of-care in impoverished rural settings where most malaria patients live, was evaluated.

Methodology/Principal Findings

This device and the standard qualitative fluorescent spot test (FST) were each compared against the quantitative spectrophotometric assay for G6PD activity as the diagnostic gold standard. The assessment occurred at meso-endemic Panenggo Ede in western Sumba Island in eastern Indonesia, where 610 residents provided venous blood. The G6PD RDT and FST qualitative assessments were performed in the field, whereas the quantitative assay was performed in a research laboratory at Jakarta. The median G6PD activity ≥5 U/gHb was 9.7 U/gHb and was considered 100% of normal activity. The prevalence of G6PD deficiency by quantitative assessment (<5 U/gHb) was 7.2%. Applying 30% of normal G6PD activity as the cut-off for qualitative testing, the sensitivity, specificity, positive predictive value, and negative predictive value for G6PD RDT versus FST among males were as follows: 100%, 98.7%, 89%, and 100% versus 91.7%, 92%, 55%, and 99%; P = 0.49, 0.001, 0.004, and 0.24, respectively. These values among females were: 83%, 92.7%, 17%, and 99.7% versus 100%, 92%, 18%, and 100%; P = 1.0, 0.89, 1.0 and 1.0, respectively.

Conclusions/Significance

The overall performance of G6PD RDT, especially 100% negative predictive value, demonstrates suitable safety for G6PD screening prior to administering hemolytic drugs like primaquine and many others. Relatively poor diagnostic performance among females due to mosaic G6PD phenotype is an inherent limitation of any current practical screening methodology.  相似文献

We studied survival of elk (Cervus elaphus) ≥1 yr old and quantified mortality sources in the Blue Mountains of Washington, 2003–2006, following a period of extensive poaching. The population was managed under a spike-only general hunting season, with limited permits for larger males and for females. We radiomarked 190 elk (82 males and 39 females >1 yr old and 65 males 11 months old), most with rumen transmitters and neck radiocollars; 60 elk only received rumen transmitters. We estimated annual survival using known fate models and explored survival differences among sex and age classes and in 2 potentially different vulnerability zones for males. We found little support for differences in survival between younger (2–3-yr old) and older (≥4-yr old) branch-antlered males or zone differences for yearling males. A model with zone differences for branch-antlered males was the second ranked model and accounted for 14% of the available model weight. From the best-supported models, we estimated annual survival for yearling males at 0.41 (95% CI: 0.29–0.53). We estimated pooled adult female survival at 0.80 (95% CI: 0.64–0.93); when an age-class effect was included, point estimates were higher for prime-aged females (2–11 yr: S = 0.81 [0.70–0.88]) than for older females (≥12 yr: S = 0.72 [0.56–0.83]), but confidence intervals broadly overlapped. Only 1 of 7 models with a female age effect on survival was among the competitive models. For branch-antlered males, survival ranged 0.80–0.85, depending on whether zone variation was modeled. We recorded 78 deaths of radiomarked elk. Human-caused deaths (n = 55) predominated among causes and most were of yearling males killed during state-sanctioned hunts (n = 28). Most subadult male deaths were from tribal hunting (n = 5), and most mature males died from natural causes (n = 6) and tribal hunting (n = 5). We detected few illegal kills (n = 4). Our results suggest that increased enforcement effectively reduced poaching, that unreported tribal harvest was not a trivial source of mortality, and that spike-only general seasons were effective in recruiting branch-antlered males. © 2011 The Wildlife Society.  相似文献

In this study, 36 males, 33 females and 15 juveniles of the common or weedy seadragon Phyllopteryx taeniolatus , a syngnathid fish endemic to the waters of southern Australia, were identified using visual implant fluorescent elastomer tags and pattern of appendages and their reproductive cycle and growth was studied from May 2001 to June 2002. Brooding males were present for a period of >6 months, from June–July to early‐January. The percentage of males that were pregnant in the studied areas peaked in November–December, near the end of the breeding season. Two pregnancies were reported for some males. Ten recruits were identified from November to June. Growth rates for young‐of‐the‐year, young of the previous year, males and females were measured. Female juvenile vundu Heterobranchus longifilis outweighed males by > 60% ( P < 0·0001) margins when aged 142 or 177 days. Not only did females grow faster ( P =0·0494) than males during the experiment, but they also outweighed them by a 38·5% margin at the start of the experiment (86 days: P =0·0109). Sexual growth dimorphism (SGD) was estimated as having arisen at 15 ± 10 days. Vundu aquaculture could be improved substantially by monosex female rearing. Because the modelled emergence of SGD corresponded precisely with the start of type II cannibalism, it was suggested that cannibalism among juvenile vundu be exerted essentially by females, and might have different impacts on faster-growing females and slower-growing males.  相似文献

Parameters of a mathematical function of growth, fit to the body weight curve of two randombred control populations of each sex of chickens from hatching through 45 weeks of age, were estimated. The logistic function was chosen from among growth formulae that express rate of gain as a function of weight at a given time and gain to be made. Two logistic parameters, growth-rate constant and age at the point of inflection, were estimated by the methods of sample quantiles and nonlinear regression from weekly mean body weights of 225 males and 281 females of the Rhode Island Red (RIR) line, and 164 males and 239 females of the White Leghorn (WL) line. Males had a larger growth-rate constant than females of the same line. The RIR line had a larger rate constant than the WL line, for each sex. Age at the point of inflection was similar for males and females in the RIR line, but smaller for males than females in the WL line. Sample quantiles yielded larger, less precise estimates of the growth-rate constant than nonlinear regression. Estimates of age at the point of inflection were usually smaller using sample quantiles.  相似文献

A multidisciplinary anthropological and epidemiological pilot field study of the Bayash population living in 6 villages of the eastern Croatian region of Baranya has been performed in 2005/06. The Bayash (or Boyash) belong to the Roma minority population speaking a distinct archaic dialect of the Romanian language. Since the bone mineral density values in the Roma have not been explored so far and the prevalence of osteoporosis is unknown for this ethnic minority group a screening by means of the Sahara Hologic clinical bone sonometer has been performed on 232 voluntary participants (73 males and 159 females). The prevalence of osteoporosis (T-score <-2.5) in the Bayash aged 50 and older is found to be 9.1% in males and 34.4% in females, which is substantially higher than in the general population of Croatia. The prevalence of T-values ranging from -1 to -2.5 indicating osteopenia is found to be 63.6% in males and 45.3% in females, while T-values within the normal range are found only in 27.3% males and 20.3% females. In addition to the low bone mass in older subjects, the mean estimated bone mineral density in all age groups of Bayash men and women was lower in comparison to the manufacturer's reference ranges for European population of the same age. Since body size effects could not be declined, the reference values that would be appropriate for the Roma population should be further explored. The high estimated prevalence of developed osteoporosis calls for attention and the survey should also be extended to exploring the association of low bone mineral density with particular life style and reproductive factors present in this semi-sedentary Roma population.  相似文献

We monitored seven resident (three males and four females) and six dispersing subadult Eurasian lynx from to in a population that was re-introduced to the Swiss Jura Mountains in the early 1970s. Home-range areas of the neighbouring adults were 71–281 km2, and significant core areas 34–252 km2. Males occupied significantly larger areas than females. Home-range overlap was 9% for neighbouring males and 3%) for females. Core areas of males did touch, but those of females were clearly separated. Each male's home range covered those of one or two females. Population density was 0.94 lynx/100 km2 for resident animals. Pre-dispersal mortality was estimated to be 50%. Juveniles dispersed from their mothers' home area at the age of 10 months. Of six monitored subadults. only one survived the first year of independence. Human-caused mortality (traffic accidents. illegal killing) was high. This was also the case among resident adults. This might be a threat to the long-term survival of the reintroduced population.  相似文献

We studied demographic effects of the mitochondria-targeted antioxidant SkQ1 on free-breeding Campbell dwarf hamsters (Phodopus campbelli, Thomas, 1905, Rodentia, Cricetidae) in an outdoor vivarium with seasonally varying day length and temperatures. The animals were kept in pairs from their young age. We removed litters from parental cages at their age of 25 days. Experimental hamsters received daily 50 nmol/kg SkQ1 with water by oral dosing, whereas control animals received water. SkQ1 had no effect on the lifespan of either males or females in reproductive pairs. Mortality among females was higher than among males irrespective of SkQ1 treatment, this being related to higher costs of reproduction in females. However, SkQ1 accelerated breeding in pairs in the first half of the reproductive period of a year. Although there were no statistical differences in body mass of males and females between experimental and control animals during most of their life, SkQ1-receiving males had higher body mass at the end of their life. The opposite tendency was characteristic for old females. One-year-old males and females of the experimental and control groups showed no difference in intensity of immune response to sheep red blood cells. The dermal hypersensitivity response to phytohemagglutinin (test for T-cell immunity) was significantly higher in SkQ1-treated 1- and 1.5-year-old males. This was not true for females. There was a tendency toward increased density of the neutrophil population in blood in 1-year-old SkQ1-treated males. However, experimental males showed no difference from control males in the activity of the “peroxidase-endogenous hydrogen peroxide system” of neutrophils. The background level of stress estimated by the concentration of cortisol in blood serum was significantly lower in the SkQ1-treated males during autumn adaptive adjustment of the organism. A similar trend was also observed during the January frosts, when the background level of stress was rather high. We observed no differences between cortisol concentration in experimental and control animals during the reproductive period in early spring and mid-summer. We tend to interpret the absence of geroprotective effect of SkQ1 on free-breeding dwarf hamsters by its ability to intensify breeding. We previously demonstrated the ability of SkQ1 to increase the lifespan of non-breeding females.  相似文献
