土壤中多环芳烃的微生物降解及土壤细菌种群多样性   总被引:4,自引:0,他引:4  
利用室内模拟方法,研究中、低浓度多环芳烃(PAHs)污染土壤的微生物修复效果,阐明土壤微生物(接种和土著)与PAHs降解的关系.结果表明:投加PAHs高效降解菌可以促进土壤中PAHs的降解,2周内效果显著;典型PAHs降解的难易程度依据为:菲<蒽<芘<苯并(a)芘和屈;细菌种群丰度和多样性均与PAHs降解呈负相关关系,同一处理细菌种群结构随时间变化不大.对于中、低浓度PAHs原位污染土壤,增强土著菌的活性是提高土壤PAHs降解率的有效途径之一.  相似文献

Biodegradation of UV-irradiated anthracene, pyrene,benz[a]anthracene,and dibenz[a,h]anthracene was comparedto that of the non-irradiated samples, individuallyand in synthetic mixtures with enrichment cultures.Combined treatment was repeated for individual anthraceneand for the PAH mixture with Sphingomonas sp.strain EPA 505 and Sphingomonas yanoikuyae.Enrichment culture studies were performed on the PAHmixtures in the presence of the main photoproduct ofanthracene, pure 9,10-anthracenedione. Photochemicallypretreated creosote solutions were also subjected tobiodegradation and the results were compared tothose of the non-irradiated solutions. The primaryinterest was on 16 polycyclic aromatic hydrocarbons(PAHs) listed as priority pollutants by European Union(EU) and the United States Environmental ProtectionAgency (USEPA). Irradiation accelerated thebiodegradation onset for anthracene, pyrene, andbenz[a]anthracene when they were treatedindividually. The biodegradation of irradiatedpyrene started with no lag phase andwas complete by 122 h whereas biodegradation of thenon-irradiated sample had a lag of 280 h andresulted in complete degradation by 720 h. Biodegradation ofPAHs was accelerated in synthetic mixtures, especiallyin the presence of pure 9,10-anthracenedione.In general, irradiation had no effect on the biodegradation of PAHsincubated in synthetic mixtures or with pure cultures. Undercurrent experimental conditions, the UV-irradiation invariablyreduced the biodegradation of PAHs in creosote. Based onthe results of the present and previous photochemical-biologicalstudies of PAHs, the influence of the photochemical pretreatmenton the biodegradation is highly dependent on the compoundsbeing treated and other process parameters.  相似文献

High-molecular-weight (HMW) polycyclic aromatic hydrocarbons (PAHs) are pollutants that persist in the environment due to their low solubility in water and their sequestration by soil and sediments. The addition of a water-immiscible, nonbiodegradable, and biocompatible liquid, silicone oil, to a soil slurry was studied to promote the desorption of PAHs from soil and to increase their bioavailability. First, the transfer into silicone oil of phenanthrene, pyrene, chrysene, and benzo[a]pyrene added to a sterilized soil (sandy soil with 0.65% total volatile solids) was measured for 4 days in three two-liquid-phase (TLP) slurry systems each containing 30% (w/v) soil but different volumes of silicone oil (2.5%, 7.5%, and 15% [v/v]). Except for chrysene, a high percentage of these PAHs was transferred from soil to silicone oil in the TLP slurry system containing 15% silicone oil. Rapid PAH transfer occurred during the first 8 h, probably resulting from the extraction of nonsolubilized and of poorly sorbed PAHs. This was followed by a period in which a slower but constant transfer occurred, suggesting extraction of more tightly bound PAHs. Second, a HMW PAH-degrading consortium was enriched in a TLP slurry system with a microbial population isolated from a creosote-contaminated soil. This consortium was then added to three other TLP slurry systems each containing 30% (w/v) sterilized soil that had been artificially contaminated with pyrene, chrysene, and benzo[a]pyrene, but different volumes of silicone oil (10%, 20%, and 30% [v/v]). The resulting TLP slurry bioreactors were much more efficient than the control slurry bioreactor containing the same contaminated soil but no oil phase. In the TLP slurry bioreactor containing 30% silicone oil, the rate of pyrene degradation was 19 mg L(-)(1) day(-)(1) and no pyrene was detected after 4 days. The degradation rates of chrysene and benzo[a]pyrene in the 30% TLP slurry bioreactor were, respectively, 3.5 and 0.94 mg L(-)(1) day(-)(1). Low degradation of pyrene and no significant degradation of chrysene and benzo[a]pyrene occurred in the slurry bioreactor. This is the first report in which a TLP system was combined with a slurry system to improve the biodegradation of PAHs in soil.  相似文献

In a field experiment, the transfer of polychlorinated biphenyls (PCBs) and polycyclic aromatic hydrocarbons (PAHs) from contaminated soil to maize (Zea mays L.), sunflower (Helianthus annuus), poplar (Populus nigra × P. maximowiczii) and willow (Salix × smithiana) and the distribution of PCB congeners in maize and sunflower was investigated. The former waste incinerator in Hradec Králové (Czech Republic) was chosen for the experiment. Results of plot screening showed heterogenous contamination by PCBs and PAHs. PCB soil contamination was evidently caused by Delor 106 or Aroclor 1260 stocking and PAH contamination by chemicals containing fluoranthene, benzo/b/fluoranthene, phenanthrene and pyrene. Tested plants were planted on a contaminated field site, in soil contaminated with 1530 μg/kg of total PCBs and 0.138 and 3.42 mg/kg of total PAHs. The results show that maize and sunflower roots accumulated the most PCBs from soil. These plants accumulated hexa- and heptachlorobiphenyl congeners more than tri-, tetra-, and pentachlorobiphenyl congeners. Total concentrations of PAHs in tested plants ranged from 0.096 to 1.34 mg/kg. The highest phenanthrene concentration was found in aboveground biomass of sunflower and the highest concentration of pyrene, in maize roots.  相似文献

Nonexhaustive extraction (propanol, butanol, hydroxypropyl-β-cyclodextrin [HPCD]), persulfate oxidation and biodegradability assays were employed to determine the bioavailability of polycyclic aromatic hydrocarbons (PAHs) in creosote-contaminated soil. After 16 weeks incubation, greater than 89% of three-ring compounds (acenaphthene, anthracene, fluorene, and phenanthrene) and 21% to 79% of four-ring compounds (benz[a]anthracene, chrysene, fluoranthene, and pyrene) were degraded by the indigenous microorganisms under biopile conditions. No significant decrease in five- (benzo[a]pyrene, benzo[b+k]fluoranthene) and six-ring compounds (benz[g,h,i]perylene, indeno[1,2,3-c,d]pyrene) was observed. Desorption of PAHs using propanol or butanol could not predict PAH biodegradability: low-molecular-weight PAH biodegradability was underestimated whereas high-molecular-weight PAH biodegradability was overestimated. Persulfate oxidation and HPCD extraction of creosote-contaminated soil was able to predict three- and four-ring PAH biodegradability; however, the biodegradability of five-ring PAHs was overestimated. These results demonstrate that persulfate oxidation and HPCD extraction are good predictors of PAH biodegradability for compounds with octanol-water partitioning coefficients of < 6.  相似文献

Remediation of polycyclic aromatic hydrocarbons (PAHs) contaminated alkaline saline soil with phreatophyte or "water loving plants" was investigated by spiking soil from the former lake Texcoco with 100 mg phenanthrene (Phen) kg(-1) soil, 120 mg anthracene (Ant)kg(-1) soil and 45 mg benzo(a)pyrene (BaP) kg(-1) soil and vegetating it with Athel tamarisk (Tamarix aphylla L Karst.). The growth of the Athel tamarisk was not affected by the PAHs. In soil cultivated with Athel tamarisk, the leaching of PAHs to the 32-34 cm layer decreased 2-fold compared to the uncultivated soil. The BaP concentration decreased to 39% of the initial concentration at a distance smaller than 3 cm from the roots and to 45% at a distance larger than 3cm, but 59% remained in unvegetated soil after 240 days. Dissipation of Ant and Phen decreased with depth, but not BaP. The biodegradation of PAHs was affected by their chemical properties and increased in the presence of T. aphylla, but decreased with depth.  相似文献

Phytoremediation was evaluated as a potential treatment for the creosote-contaminated surface soil at the McCormick and Baxter (M&B) Superfund Site in Portland, Oregon. Soil at the M&B site is contaminated with pentachlorophenol (PCP) and polyaromatic hydrocarbons (PAHs). Eight individual PAH compounds (containing four to six aromatic rings) were included in the investigation. Greenhouse and field studies were carried out using perennial ryegrass (Lolium perenne). The following three treatments were compared in both studies: T1 = planted-amended soil; T2 = unplanted-amended soil; and T3 = unplanted-unamended soil. The amendments were mineral nutrients and dolomite, which was used to raise the acidic pH of the soil. Contaminant concentrations in the soil were measured initially and at regular intervals for several months. In the greenhouse study, the concentrations of certain contaminants decreased as a function of time. Thus, PCP, fluoranthene, pyrene, chrysene, and benzo(k)fluoranthene appeared to undergo biodegradation in all of the treatments. On the other hand, certain larger molecular weight PAHs were relatively recalcitrant. These “recalcitrant PAHs” included benzo(b)fluoranthene, benzo(a)pyrene, benzo(g,h,i)perylene and indeno(1,2,3-cd)pyrene. Statistical methods were used to compare the concentrations of the more easily biodegraded contaminants in treatments T1, T2, and T3. The statistical analysis was facilitated by normalizing the contaminant concentrations relative to the sum of the recalcitrant PAHs in the same sample. Thus, ratios were created that could be compared directly to benchmark values indicative of the contaminant at the beginning of the study. In the greenhouse study, statistically significant differences between T1 and T2, across all treatment times, were obtained for fluoranthene, pyrene, and chrysene (p ≤0.05), suggesting enhanced rhizosphere biodegradation for these compounds. Significant differences between T2 and T3 were obtained for pyrene (p ≤0.03), indicating that nutrients stimulated the biodegradation of this contaminant. Although the greenhouse study was carried out with a well-mixed soil sample from the M&B site, an extremely uneven distribution of contaminants was encountered in the field study. The resulting scatter in the field data made comparisons difficult, and treatment-specific effects observed in the greenhouse study were not statistically significant in the field study. However, analysis of the normalized data from the field revealed the same time-dependent decreases in contaminant concentration as observed in the greenhouse study.  相似文献

【背景】真菌和细菌被认为在多环芳烃污染土壤生物修复过程中发挥协同作用,目前在真实土壤体系中开展真菌-细菌协同降解研究较少。【目的】研究真菌和细菌对不同种类多环芳烃降解的差异及对蒽和苯并[a]蒽的生物强化与协同作用。【方法】选用多环芳烃降解真菌和细菌各一株,在液体纯培养体系下分析它们对不同种类多环芳烃降解的差异,在土壤体系中采用放射性同位素示踪技术研究2种微生物对蒽和苯并[a]蒽的生物强化与协同作用。【结果】供试细菌鞘脂菌NS7能够很好地降解低环种类多环芳烃,以蒽作为唯一碳源时可以将其完全降解,在复合污染条件下对菲、蒽、荧蒽、芘等降解效果突出(>90%),对苯并[a]芘降解效果较差(9.76%)。相比而言,供试真菌糙皮侧耳菌对苯并[a]芘具有更好的降解效果(21.18%),对低环多环芳烃降解效果明显不如降解菌NS7。在自然土壤中,蒽和苯并[a]蒽具有明显不同的矿化效率,分别为18.61%和4.28%,在蒽污染土壤中加入鞘脂菌NS7并未显著提高蒽的矿化率(P>0.05),相比而言,苯并[a]蒽污染土壤中加入糙皮侧耳显著提高了污染物矿化效率(2.24倍),表明真菌和细菌在土壤环境...  相似文献

Biodegradation of a mixture of PAHs was assessed in forest soil microcosms performed either without or with bioaugmentation using individual fungi and bacterial and a fungal consortia. Respiratory activity, metabolic intermediates and extent of PAH degradation were determined. In all microcosms the low molecular weight PAH’s naphthalene, phenanthrene and anthracene, showed a rapid initial rate of removal. However, bioaugmentation did not significantly affect the biodegradation efficiency for these compounds. Significantly slower degradation rates were demonstrated for the high molecular weight PAH’s pyrene, benz[a]anthracene and benz[a]pyrene. Bioaugmentation did not improve the rate or extent of PAH degradation, except in the case of Aspergillus sp. Respiratory activity was determined by CO2 evolution and correlated roughly with the rate and timing of PAH removal. This indicated that the PAHs were being used as an energy source. The native microbiota responded rapidly to the addition of the PAHs and demonstrated the ability to degrade all of the PAHs added to the soil, indicating their ability to remediate PAH-contaminated soils.  相似文献

Accumulation of Cr(VI) in rice seeds cultivated in Cr-contaminated soil of the Sundarbans (India) is an environmental problem. Cr(VI) concentration in this soil was 6.2 ± 0.3 mg/kg, whereas total chromium was 32.04 ± 1.60 mg/kg. A Cr(VI)-removing bacterium isolated from Cr-contaminated paddy field soil of Sundarbans was identified as Staphylococcus sciuri. Enrichment culture of S. sciuri was applied to pot cultivation of rice in Cr-contaminated soil. After 8 weeks, 71 ± 3% Cr(VI) (final concentration 2.15 ± 0.01 mg/kg) and 65 ± 2% total Cr removal (end concentration 11.3 ± 0.5 mg/kg) were attained in bacterium-treated soils. Growth parameters indicated healthy development of plants cultivated in bacterium-treated soils that was not observed in control plants. Total Cr removal attained in rice seeds of plants cultivated in bacterium-treated soils compared with control rice seeds was 78 ± 4%. Total Cr concentration in test seeds was 0.72 ± 0.05 mg/kg (World Health Organization [WHO] permissible limit: 1.30 mg/kg), whereas the same in control seeds was 3.27 ± 0.16 mg/kg. Cr(VI) reduction achieved in rice seeds cultivated in bacterium-treated soil compared with control rice seeds was 95 ± 5%. Cr(VI) concentration in rice seeds cultivated in treated soil was 0.050 ± 0.003 mg/kg, whereas the same in untreated control was 0.93 ± 0.05 mg/kg. Successful paddy field soil bioremediation by any Staphylococcus species was demonstrated for the first time.  相似文献

The phytoremediation, with industrial hemp (Cannabis sativa), of a Hawaiian silty clay soil contaminated with two polycyclic aromatic hydrocarbons (PAHs), chrysene and benzo[a]pyrene, was studied. Hemp showed a very high tolerance to the contaminants. The growth rates of hemp, compared with control, in soils fortified with chrysene and benzo[a]pyrene at concentrations of each varying from 25 to 200 micrograms/g were consistently above 100%. The plants grew from seed for 45 days in soil fortified with PAHs at concentrations of 25, 50, and 75 micrograms/g. Controls were pots with contaminated soil but no plant. PAHs levels were significantly reduced in all pots (control and seeded pots), expect for one set at a high concentration of chrysene, which may be due to uneven spiking. A time course study over 28 days was done to monitor changes of microbial count and levels of chrysene. Little changes were observed for the total microbial count in the soil, and the concentration of chrysene in the soil decreased slightly in the pots containing plants. However, the chrysene levels in those pots were consistently lower than those in the pots without plants.  相似文献

Mycobacterium sp. PYR‐1, which was previously shown to mineralize several individual polycyclic aromatic hydrocarbons (PAHs), simultaneously degraded phenanthrene, anthracene, fluoranthene, pyrene and benzo[a]pyrene in a six‐component synthetic mixture. Chrysene was not degraded significantly. When provided with a complex carbon source, Mycobacterium sp. PYR‐1 degraded greater than 74% of the total PAH mixture during 6 d of incubation. Mycobacterium sp. PYR‐1 appeared to preferentially degrade phenanthrene. No significant difference in degradation rates was observed between fluoranthene and pyrene. Anthracene degradation was slightly delayed but, once initiated, proceeded at a constant rate. Benzo[a]pyrene was degraded slowly. Degradation of a crude mixture of benzene‐soluble PAHs from contaminated sediments resulted in a 47% reduction of the material in 6 d compared with that of autoclaved controls. Experiments using an environmental microcosm test system indicated that mineralization rates of individual 14C‐labeled compounds were significantly lower in the mixtures than in equivalent doses of these compounds alone. Mineralization of the complete mixture was estimated conservatively to be between 49.7 and 53.6% and was nearly 50% in 30 d of incubation when all compounds were radiolabeled. These results strengthen the argument for the potential application of Mycobacterium sp. PYR‐1 for bioremediation of PAH‐contaminated wastes.  相似文献

Aims: In order to develop effective bioremediation strategies for polyaromatic hydrocarbons (PAHs) degradation, the composition and metabolic potential of microbial communities need to be better understood, especially in highly PAH contaminated sites in which little information on the cultivation‐independent communities is available. Methods and Results: Coal‐tar‐contaminated soil was collected, which consisted of 122·5 mg g?1 total extractable PAH compounds. Biodegradation studies with this soil indicated the presence of microbial community that is capable of degrading the model PAH compounds viz naphthalene, phenanthrene and pyrene at 50 ppm each. PCR clone libraries were established from the DNA of the coal‐tar‐contaminated soil, targeting the 16S rRNA to characterize (i) the microbial communities, (ii) partial gene fragment encoding the Rieske iron sulfur center (α‐subunit) common to all PAH dioxygenase enzymes and (iii) β‐subunit of dioxygenase. Phylotypes related to Proteobacteria (Alpha‐, Epsilon‐ and Gammaproteobacteria), Acidobacteria, Actinobacteria, Firmicutes, Gemmatimonadetes and Deinococci were detected in 16S rRNA derived clone libraries. Many of the gene fragment sequences of α‐subunit and β‐subunit of dioxygenase obtained from the respective clone libraries fell into clades that are distinct from the reference dioxygenase gene sequences. Presence of consensus sequence of the Rieske type [2Fe‐2S] cluster binding site suggested that these gene fragments encode for α‐subunit of dioxygenase gene. Conclusions: Sequencing of the cloned libraries representing α‐subunit gene fragments (Rf1) and β‐subunit of dioxygenase showed the presence of hitherto unidentified dioxygenase in coal‐tar‐contaminated soil. Significance and Impact of the Study: The combination of the Rieske primers and bacterial community profiling represents a powerful tool for both assessing bioremediation potential and the exploration of novel dioxygenase genes in a contaminated environment.  相似文献

Bioaugmentation of polycyclic aromatic hydrocarbon (PAH)-contaminated soil was investigated using a mixed bacterial culture (community five) isolated from an abandoned industrial site. Community five was inoculated into contaminated soil containing a total PAH (two- to five-ring compounds) concentration of approximately 820 mg/kg soil. PAH degradation by the indigenous microbial population was restricted to the lower molecular weight compounds (naphthalene, acenaphthene, fluorene and phenanthrene) even with yeast extract addition: these compounds decreased by 14 to 37%, in soil hydrated to 50% water capacity, following 91 days of incubation at 24°C. Inoculation of community five into this PAH-contaminated soil resulted in significant decreases in the concentration of all PAHs over the incubation period: greater than 86% of naphthalene, acenaphthene, fluorene, and phenanthrene were degraded after 91 days, while anthracene, fluoranthene, and pyrene were degraded to lesser extents (51.7 to 57.6%). A lag period of 48 to 63 days was observed before the onset of benz[a]anthracene, benzo[a]pyrene, and dibenz[a,h]anthracene removal. However, significant decreases in the concentration of these compounds (32.6, 25.2, and 18.5%, respectively) were observed after 91 days. No significant decrease in the mutagenic potential of organic soil extracts (as measured by the Ames Test) was observed after incubation of the soil with the indigenous microflora; however, the Microtox toxicity of aqueous soil extracts was reduced sevenfold. In contrast, extracts from contaminated soil inoculated with community five underwent a 43% decrease in mutagenic potential and the toxicity was reduced 170-fold after 91 days incubation. These observations suggest that community five could be utilised for the detoxification of PAH-contaminated soil.  相似文献

A mixture of organic chemicals (MOC) containing equal molar amounts of benzoic acid, hexadecane, 2,2‐dimethyl 4,n‐propyl‐benzene, phenanthrene, pyrene, and either cycloheptane or cis‐decahydronaphthalene (cis‐decalin) was applied to soil at rates of 0 to 8000 mg/kg. In a plant‐screening experiment, growth responses of four legume and five nonlegume species were determined at 10 and 25°C. The MOC applied at 2000 mg/kg reduced the growth of several species without resulting in significant seedling death. At 10°C, the growth of alpine bluegrass (Poa alpina L.) in the 1000 and 2000 mg/kg treatments of soil increased by more than 185%. In a plant growth response experiment, alpine bluegrass and alfalfa (Medicago sativa L.) were grown in soil that had been contaminated at rates of 0 and 2000 mg/kg. At 14 weeks, the shoot and root dry weights of alfalfa were 97% lower in the contaminated soil, while the shoot dry weight, root dry weight, and root length of alpine bluegrass were 135,235 and 268% higher, respectively. Except for pyrene, <23% of the compounds comprising the MOC remained in the soil after 4 weeks and <5% after 14 weeks. The disappearance of the MOC was not significantly influenced by the presence of alfalfa or alpine bluegrass.  相似文献

A microbial consortium degrading the high-molecular-weight polycyclic aromatic hydrocarbons (HMW PAHs) pyrene, chrysene, benzo[a]pyrene and perylene in a two-liquid-phase reactor was studied. The highest PAH-degrading activity was observed with silicone oil as the water-immiscible phase; 2,2,4,4,6,8, 8-heptamethylnonane, paraffin oil, hexadecane and corn oil were much less, or not efficient in improving PAH degradation by the consortium. Addition of surfactants (Triton X-100, Witconol SN70, Brij 35 and rhamnolipids) or Inipol EAP22 did not promote PAH biodegradation. Rhamnolipids had an inhibitory effect. Addition of salicylate, benzoate, 1-hydroxy-2-naphtoic acid or catechol did not increase the PAH-degrading activity of the consortium, but the addition of low-molecular-weight (LMW) PAHs such as naphthalene and phenanthrene did. In these conditions, the degradation rates were 27 mg l-1 d-1 for pyrene, 8.9 mg l-1 d-1 for chrysene, 1.8 mg l-1 d-1 for benzo[a]pyrene and 0.37 mg l-1 d-1 for perylene. Micro-organisms from the interface were slightly more effective in degrading PAHs than those from the aqueous phase.  相似文献

【目的】多环芳烃(PAHs)是一类普遍存在于环境中且具有高毒性的持久性有机污染物,高效降解菌的筛选对利用生物修复技术有效去除环境中的多环芳烃具有重要意义。研究拟从供试菌株中筛选多环芳烃高效降解菌,并分析其降解特性,为多环芳烃污染环境的微生物修复提供资源保障和科学依据。【方法】采用平板法从25株供试菌株中筛选出以菲和芘为唯一碳源和能源的高效降解菌,经16S rRNA基因序列进行初步鉴定,通过单因素实验法分析其在液体培养基中的降解特性。【结果】筛选出的3株多环芳烃高效降解菌SL-1、02173和02830经16S rRNA基因序列分析,02173和02830分别与假单胞菌属中的Pseudomonas alcaliphila和Pseudomonas corrugate同源性最近,SL-1为本课题组发表新类群Rhizobium petrolearium的模式菌株;降解实验表明,菌株SL-1 3 d内对单一多环芳烃菲(100 mg/L)和芘(50 mg/L)的降解率分别达到100%和48%,5 d后能够降解74%的芘;而其3 d内对混合PAHs中菲和芘的降解率分别为75.89%和81.98%。菌株02173和02830 3 d内对混合多环芳烃中萘(200 mg/L)、芴(50 mg/L)、菲(100 mg/L)和芘(50 mg/L)的降解率均分别超过97%。【结论】筛选出的3株PAHs降解菌SL-1、02173和02830不仅可以高效降解低分子量PAHs,还对高分子量PAHs具有很好的降解潜力。研究表明,由于共代谢作用低分子量多环芳烃可促进高分子量多环芳烃的降解,而此时低分子量多环芳烃的降解将受到抑制。  相似文献

Summary When inoculated at high cell densities, three strains of Pseudomonas cepacia degraded the polycyclic aromatic hydrocarbons (PAHs) benzo[a]pyrene, dibenz[a,h]anthracene and coronene as sole carbon and energy sources. After 63 days incubation, there was a 20 to 30% decrease in the concentration of benzo[a]pyrene and dibenz[a,h]anthracene and a 65 to 70% decrease in coronene concentration. The three strains were also able to degrade all the PAHs simultaneously in a PAH substrate mixture containing three-, four-, five- and seven-benzene ring compounds. Furthermore, improved degradation of the five- and seven-ring PAHs was observed when low molecular weight PAHs were present.  相似文献

Polycyclic aromatic hydrocarbons (PAHs) are a class of persistent organic compounds derived from natural sources and anthropogenic processes, which have been recommended as priority pollutants. Degradation of PAHs in the environment is becoming more necessary and urgent. In the current study, strain PL2, which is capable of growing aerobically on pyrene (PYR) as the sole carbon source, was isolated from hydrocarbons-contaminated soil and then identified as Pseudomonas putida by morphological and physiological characteristics as well as 16S rDNA sequence. The strain PL2 was able to degrade 50.0% of the pyrene at 28°C within 6 days in the presence of 50 mg/L pyrene, while the strain PL2 degraded 50.0% of the pyrene within 2 days when a solution of 50 mg/L pyrene and 50 mg/L phenanthrene was used. In addition, phenanthrene was shown to increase the biodegradation efficiency of pyrene by the strain PL2. The order of degradation by the strain PL2 was pH 6.0 > pH 7.0 > pH 5.0 > pH 8.0. The degradation rate of PYR in the soil by the strain PL2 reached 70.0% at the 10th day. The dynamics of PYR degradation in soil by PL2 was fit to the first order model and the strain PL2 was shown to efficiently degrade PYR in soil. The current study showed that P. putida PL2 was a novel bacterium that could degrade pyrene and holds great promise for use in PAHs bioremediation in soil.  相似文献

Biodegradation kinetics of naphthalene, phenanthrene and pyrene were studied in sole-substrate systems, and in binary and ternary mixtures to examine substrate interactions. The experiments were conducted in aerobic batch aqueous systems inoculated with a mixed culture that had been isolated from soils contaminated with polycyclic aromatic hydrocarbons (PAHs). Monod kinetic parameters and yield coefficients for the individual compounds were estimated from substrate depletion and CO(2) evolution rate data in sole-substrate experiments. In all three binary mixture experiments, biodegradation kinetics were comparable to the sole-substrate kinetics. In the ternary mixture, biodegradation of naphthalene was inhibited and the biodegradation rates of phenanthrene and pyrene were enhanced. A multisubstrate form of the Monod kinetic model was found to adequately predict substrate interactions in the binary and ternary mixtures using only the parameters derived from sole-substrate experiments. Numerical simulations of biomass growth kinetics explain the observed range of behaviors in PAH mixtures. In general, the biodegradation rates of the more degradable and abundant compounds are reduced due to competitive inhibition, but enhanced biodegradation of the more recalcitrant PAHs occurs due to simultaneous biomass growth on multiple substrates. In PAH-contaminated environments, substrate interactions may be very large due to additive effects from the large number of compounds present.  相似文献
