Sturgeon are an ancient family (Acipenseridae) of fishes that lie close to the divergence of fish that eventually evolved into terrestrial animals and those that evolved into modern teleost species. Therefore, white sturgeon vitellogenin sequences fill a gap in the current understanding of the functional domains of this protein family. Vitellogenin cDNA was sequenced and used to investigate gene expression in white sturgeon, Acipenser transmontanus. Estrogen-induced vitellogenin mRNA was detected in the livers of both males and females and was also detected in undifferentiated gonads of estrogen-treated fish. Low levels of vitellogenin mRNA were also detected in the testis of both control and estrogen-treated males. The cDNA encoded a 186-kDa protein that was missing only six to seven of the amino-terminal amino acids. Comparisons to silver lamprey, Xenopus, and chicken vitellogenin sequences indicate that the overall structure of the yolk protein domains were highly conserved. There was considerable homology in three regions of the lipovitellin I domain. These conserved sequences are likely to be involved in vitellogenin receptor binding. The phosvitin domain of white sturgeon vitellogenin contained fewer and shorter serine repeats as predicted from yolk protein phosphate content of fish compared to Xenopus and chicken. However, the vitellogenin of white sturgeon had a lower serine content as compared with silver lamprey, indicating that an increased serine content is not strictly a function of evolutionary age. Synopsis The ovaries of four pre-spawning white sturgeon females were sampled and their oocytes incubated in the presence of eight gonadotropin preparations, 21 steroids, a prostaglandin and a catacholamine. Among the gonadotropin preparations, acetone dried pituitary gland powder from white sturgeon, common carp and chum salmon (in decreasing order of potency) were capable of inducing oocyte maturation (germinal vesicle breakdown — GVBD), while human chorionic gonadotropin, pregnant mare's serum gonadotropin, equine luteinizing hormone, bullfrog gonadotropin, and a stellate sturgeon pituitary chromatographic fraction capable of inducing testosterone production in white sturgeon testicular tissue failed to elicit any oocyte maturation response. The progesterone derivatives were the most potent steroid inducers of GVBD, followed closely by several corticosteroids. In vitro incubation of white sturgeon oocytes, in the presence of a suitable steroid (progesterone), can be used as a diagnostic tool in screening out unresponsive females for induced spawning work. The two remaining compounds, prostaglandin F2a and epinephrine, failed to cause ovulation in progesterone-matured white sturgeon oocytes.  相似文献

Synopsis Blood samples from cannulated young adult (2.5–15 kg) white sturgeon, acclimated to San Francisco Bay water (24 ppt) had plasma values of 248.8 ± 13.5 mOsm kg−1 H2O, [Na+] = 125 ± 8.0 mEq 1−1, [K+] = 2.6 ± 0.8 mEq 1−1 and [CL] = 122 ± 3.0 mEq 1−1. Freshwater acclimated sturgeon had an osmolality of 236 ± 7, [Na+] = 131.6 + 4.4, [K+] = 2.5 ± 0.7 and [CL] = 110.6 ± 3.6. Freshwater acclimated fish gradually exposed to sea water (increase of 5 ppt h−1) had higher plasma osmolalities than did the bay water acclimated fish. These young adult sturgeon are able to tolerate transfer from fresh water to sea water as well as gradual transfer from sea water to fresh water. Plasma electrolytes in transferred fish are regulated, but tend to differ from long term acclimated fish at the same salinities. There is a gradual increase in the upper salinity tolerance (abrupt transfer) of juvenile white sturgeon with weight: 5–10 ppt for 0.4–0.9 g fish, 10–15 ppt for 0.7–1.8 g fish, and 15 ppt for 4.9–50.0 g fish. The ability of juveniles to regulate plasma osmolality is limited. The young adult fish are able to tolerate higher salinities (35 ppt) than juvenile sturgeon but probably are also characterized by low activity of the necessary ion exchange mechanisms in the gills which permit rapid adjustment of blood electrolytes with graduate change in external salinity.  相似文献

Development of the digestive enzyme complement corresponded with this species' carnivorous habits and a natural dietary shift at metamorphosis from larval pelagic zooplanktivory to benthic carnivory. During the first developmental stanza when the digestive tract was differentiating and the larvae were dependent on endogenous nutritional reserves, digestive enzyme concentrations were low. Initiation of gastric secretion (pepsin and acid) was concomitant with onset of exogenous feeding. Highest amylolytic and lipolytic activities were recorded from feeding larvae prior to their metamorphosis to juveniles.  相似文献

The study considers the lipid content of farm-raised sturgeon, Acipenser transmontanus , from northern Italy (commercial size: 24 sturgeon each weighing 5kg; 24 sturgeon each weighing 10 kg). Considered parameters included total lipids, proteins and subsample analysis as saturated and unsaturated fatty acids. The results revealed no variation with seasonal change. Lipid percentages showed the presence of highly unsaturated fatty acids. Both sturgeon sizes investigated showed uniformity in the composition of individual fatty acids of muscle tissue, a factor that can be attributed to the constancy of the culture conditions (feed and water temperature). The product is of reduced lipidic content and high nutritional and market value; as a low fat product sturgeon can be used fresh, and as a semi-fat product may be smoked or canned.  相似文献

Resistin as an adipokine identified from rodents in 2001 is involved in many biological processes. However, little is known about this gene in fish. We cloned Siberian sturgeon (Acipenser baerii) resistin cDNA of 795 base pairs, encoding 107 amino acids, which showed 38–40% identity to human and rodents. Real-time quantitative PCR showed that the resistin gene was widely distributed in tissues of Siberian sturgeon, with the highest expression in liver. After fasting for 1, 3, 6 and 10 days, the expression of the resistin gene in the liver of Siberian sturgeon decreased significantly, and after refeeding on the 10 days of fasting the resistin mRNA expression increased rapidly, suggesting that resistin may play an important role in liver in response to starvation. Taken together, these results suggest that resistin may be involved in the regulation of energy homeostasis in liver.  相似文献

Sturgeons are primitive bony fishes and their hearts have structural features found in other primitive fishes. Sturgeons have a pericardioperitoneal canal (PPC), a one-way conduit into the peritoneum. A PPC also occurs in elasmobranchs (sharks and rays) and studies with that group demonstrate that pericardial pressure and pericardial fluid loss via the PPC affect stroke volume. A study of white sturgeon (Acipenser transmontanus) heart function was conducted to test for a comparable PPC and pericardial effects. White sturgeon-elasmobranch heart-function similarities include biphasic ventricular filling, a comparable operational pericardial pressure (-0.03 kPa), and a strongly negative pressure (-0.2 to -0.6 kPa) with complete pericardial fluid withdrawal. Differences include the white sturgeon's relatively smaller atrium and ventricle but a larger conus arteriosus. Although white sturgeon heart size is also smaller, its pericardial volume is disproportionately less (2.4 to 2.7 vs. 3.5 to 5.4 ml kg(-1) in elasmobranchs), meaning it has less scope for increasing stroke volume upon PPC fluid release. These differences may reflect the phylogenetic progression from the less complex operation of the elasmobranch heart, which lacks sympathetic innervation and has a mechanically mediated (PPC) stroke volume, to the condition in the more derived bony fishes which have sympathetic and parasympathetic regulation of both stroke volume and heart rate.  相似文献

The effect of environmental hypercapnia on respiratory and acid-base variables was studied in white sturgeon, Acipenser transmontanus. Blood PCO2, PO2, pH, hemoglobin concentration, and plasma lactate, glucose, catecholamines and cortisol were measured first under normocapnia (water PCO2 < 0.5 Torr, 1 Torr = 133.32 Pa), then under hypercapnia (25–35 Torr) and a final return to normocapnia at 19 ± 0.5 °C. Acute (≤ 2h) hypercapnia significantly increased arterial PCO2 (8-fold increase), ventilation frequency (2-fold increase), plasma HCO3 (2.3-fold) and decreased arterial pH (to 7.15 ± 0.02). After 24 h, norepinephrine, epinephrine and cortisol, were significantly increased, and arterial pH reached its nadir (7.10 ± 0.03). During the 72- and 96-h-periods, arterial PCO2 (24 ± 4.4 Torr) and ventilatory frequency (105 ± 5 breaths min−1) stabilized, HCO3 reached its apparent maximum (23.6 ± 0.0 mmol−1), glucose decreased by 32%, and pH increased significantly to 7.31 + 0.03. The return to normocapnia completely restored arterial PCO2 (2.5 ± 0.14 Torr), HCO3 (7.4 ± 0.59 mmol · l−1), ventilation frequency (71 ± 7 breaths · min−1), and pH (7.75 ± 0.04). Overall, hypercapnia produced a respiratory acidosis, hyperventilation, a transient norepinephrine “spike”, and increased plasma catecholamines, cortisol, and arterial PO2. The respiratory acidosis was only partially compensated (35% pH restoration) 96 h after the onset of hypercapnia and resulted in a significantly decreased blood-O2 affinity (Bohr effect), as determined by construction of in vitro blood O2 equilibrium curves at 15 °C and 20 °C. The gut of adult sturgeon was examined. The oesophageal mucosa contained numerous caliciform cells, synthesizing both neutral and acidic glycoconjugates, the latter of the sialylated type. The deep tunica propria-submucosa contained lobules of multilocular adipose tissue, specially abundant during the cold season. The oesophageal tunica muscularis was made up of a large sheath of striated muscle fibres, arranged orthogonally to a thin, subserous smooth muscle layer. The siphon-shaped stomach showed a ciliated epithelium in cardiac and gastric proper gland zones, where tubular glands were present in the tunica propria. The columnar cells which composed the superficial epithelium and gastric pits were demonstrated to synthesize almost exclusively neutral glycoconjugates. Appendices pyloricae constituted a glandular body equipped with intestinal mucosa. The intestinal mucosa was organized in folds, containing numerous caliciform cells which synthesized neutral or acidic glycoconjugates, the latter either of the sialylated and sulphated type. The sulphoglycoconjugates were more abundant in the caliciform cells of the distal intestinal tracts. The tunica propria-submucosa of the spiral valve (medium intestine) contained lymphatic tissue and large lymphatic follicles. A muscularis mucosae was present only in the rectum, where in addition a peculiar granular cell type was present in the superficial tunica propria-submucosa, possibly related to defensive properties. The subserous connective tissue contained pancreatic lobules all along the stomach and intestine. The enteric nervous system showed some special aspects, the most intriguing of which was the presence of large, longitudinally oriented nerve bundles in the t. propria-submucosa of oesophagus and cardiac stomach. The nerve bundles contained, near unmyelinated nerves, some myelinated nerves, as well as neuronal bodies. Both these aspects are exceptional in vertebrates and obscure in their significance. The structural and histochemical aspects we here describe are in part different from those described for other fish. Some of these special features are possibly related with special functional roles, others require a deeper insight and different approaches to clarify them functionally.  相似文献

Green sturgeon (Acipenser medirostris) and white sturgeon (A. transmontanus) are closely related, sympatric species that inhabit the San Francisco estuary. Green sturgeon have a more marine life history but both species spawn in the Sacramento River and reside for some duration in San Francisco Bay. These sturgeons are of conservation concern, yet little is known about their dietary competition when they overlap in space and time. To examine evidence of dietary differentiation, we collected whole blood and blood plasma from 26 green sturgeon and 35 white sturgeon in San Francisco Bay. Using carbon and nitrogen stable isotope analyses, we compared their relative trophic levels and foraging locations along the freshwater to marine gradient. Sampling blood plasma and whole blood allowed comparison of dietary integration over shorter and longer time scales, respectively. Plasma and whole blood δ13C values confirmed green sturgeon had more marine dietary sources than white sturgeon. Plasma δ15N values revealed white sturgeon fed at lower trophic levels than green sturgeon recently, however, whole blood δ15N values demonstrated the two species fed at the same trophic level over longer time scales. Larger individuals of both species had higher δ13C values than smaller individuals, reflecting more marine food sources in adulthood. Length did not affect δ15N values of either species. Isotope analyses supported the more marine life history of green than white sturgeon and potentially highlight a temporary trophic differentiation of diet between species during and preceding the overlapping life stage in San Francisco Bay.  相似文献

Routine oxygen consumption rates (MO2) and swimming activity rates of juvenile white sturgeon were determined using closed respirometers at life-interval-appropriate temperatures: 10° C (0.2 g mean wet weight), 16° C (1.9 g mean wet weight), and 20° C (63.1 g mean wet weight) under normoxic (PO2 > 140 mmHg) and moderately hypoxic (PO2=80 ± 5.0 mmHg) water conditions. At all temperatures and body sizes, hypoxia significantly depressed (p < 0.05) MO2 (57% mean reduction) and swimming activity (70% mean reduction). Overall mean MO2 was 228 µg O2 g-1 wet weight h-1 (normoxia) and 99 µg O2 g-1 wet weight h-1 (hypoxia). Thus, juvenile white sturgeon appear to decrease overall energy expenditures (hypometabolism) during hypoxia via reductions in spontaneous swimming activity. This is a life style that may increase survival during widespread or prolonged environmental hypoxia.  相似文献

The primary objective of this study was to identify and characterize juvenile white sturgeon (Acipenser transmontanus) habitat in the Lower Fraser River downstream of Hope, BC, Canada. A secondary objective was to estimate the juvenile white sturgeon population in the Lower Fraser River. A total of 1867 white sturgeon was captured with gill nets at 26 sites in the Lower Fraser River. The greatest numbers of sturgeon were caught in three sloughs; all but three sturgeon were captured in the June to August period. These three sloughs all had water deeper than 5 m and current that was multidirectional. Turbidity ranged greatly from 1.5 NTU (Nephelometric Units) to 67.0 NTU and the substrate of most sites was fine sediments, fine sand, silt and clay. Stomach contents were mysid shrimp (Mysidacea), midge larvae (Chironomidae) and peamouth chub (Mylocheilus caurinus). We identified three of 26 sites with appreciable numbers of juvenile sturgeon, identified water quality parameters of these sites, identified any incidental species that might be prey and also determined that between 1985 and 1993 the juvenile white sturgeon population had declined.  相似文献

Serum and cutaneous mucus antibodies were monitored in white sturgeon for 15 weeks following intraperitoneal immunization. Ten fish were immunized (50 microg) with white sturgeon iridovirus (WSIV) or white sturgeon gonad (WSGO) tissue culture cells emulsified with or without FCA. An additional group was immunized with FITC:KLH+FCA. Fish were booster immunized at 6 weeks. Fish immunized with FITC:KLH+FCA produced significant serum antibodies to FITC by 6 weeks and this response peaked at 12 weeks (average titer 31,000). Mucosal antibodies to FITC were first detected at 12 weeks and significantly elevated by 15 weeks (average titer 18). Anti-WSIV antibody titers were detected in the serum by 9 weeks in fish immunized with WSIV and WSIV+FCA, but only a small number responded to immunization. At 15 weeks, four fish immunized with WSIV produced serum antibodies (average titer 838) and one fish immunized with WSIV+FCA had a serum titer of 1600. Mucosal anti-WSIV antibody titers of 8 and 16 were observed in two fish from the WSIV group at 12 weeks while four different fish from this group responded at 15 weeks (average titer 4). Western Blot using a monoclonal antibody confirmed immunoglobulin in mucus, and specificity to WSIV was further demonstrated by immunocytochemistry using serum from fish immunized with WSIV. Specific antibody was not detected in mucus of fish immunized with WSIV+FCA, WSGO, or WSGO+FCA. Collectively, these experiments demonstrate that white sturgeon can generate a specific antibody response following immunization, and is the first report showing mucosal immunoglobulin is present in this species.  相似文献

White sturgeon, Acipenser transmontanus (Richardson), are at risk of entrainment from dredging, with young-of-the-year fish at greatest risk. To evaluate this entrainment risk, swimming performance trials were conducted in a laboratory swim tunnel with hatchery-reared juvenile white sturgeon with varying experience levels including: naïve (only tested once), tested (re-tested after being kept in no flow) and trained (re-tested after kept in flow for nearly three weeks). Individuals of various sizes (80–100 mm TL) and all experience levels were strongly rheotactic (> 80%), but endurance was highly variable among fish. Small juveniles [< 82 mm total length (TL)] had lower escape speeds (< 40 cm s−1) than medium (82–92 mm TL) and large (> 93 mm TL) naïve fish (42–45 cm s−1), all of which had lower escape speeds than trained fish (72 cm s−1). Behavior was also highly variable among fish. Overall, benthic station-holding behaviors were least frequent in small fish, intermediate in medium and large fish, and most frequent in trained large fish. Probability of entrainment of juvenile white sturgeon can be reduced by maintaining dredge head flow fields at less than 45 cm s−1 for wild-spawned fish or by rearing hatchery fish to > 93 mm TL and exposing the fish to moderate flow velocities (10–12 cm s−1) prior to their release.  相似文献

Synopsis The effect of constant incubation temperatures (between 10°C and 26°C) on the developmental rates was found to fit a similar exponential relationship in both the lake and white sturgeon embryos and larvae. Although the lake sturgeon had an overall slower rate of development than the white sturgeon, no statistically significant difference was detected in the slopes of the exponential equations describing the effect of temperature on developmental rate. The effect of these incubation temperatures on embryonic survival also did not differ between these two species. Both species exhibited optimal survival between 14–17° C and incipient mortalities occurred at 20°C. Temperatures above 20°C were lethal for white sturgeon embryos. No effect of low incubation temperature on survival was evident from this study. A comparison of these North American species with Eurasian acipenserids suggests that all the sturgeon that have been examined exhibit a similar influence of incubation temperature on developmental rate.  相似文献
