Metal-tolerant Datura innoxia cells synthesize large amounts of cadystin, [poly(-glutamylcysteinyl) glycines, (-EC)nG, n=2–5], a class of metal-binding polypeptides, when exposed to Cd. These polypeptides have a high affinity for Cd (II) and certain other metal ions and are thought to play a role in metal tolerance in higher plants. Cells rapidly synthesize these metal-binding polypeptides when exposed to Cd and cellular concentrations of glutathione and cysteine, precursors for the synthesis of these compounds, are initially depleted then rapidly replenished. The time-frame of de novo polypeptide, glutathione and cysteine biosynthesis suggests that this pathway is, at least initially, regulated at the enzyme level. Significant amounts of Fe are associated with Cd: polypeptide complexes isolated from D. innoxia. Exposure of cultures to Cd results in an increased Fe accumulation by the cells. All the additional Fe found in the soluble portion of cell extracts is associated with the Cd: polypeptide complexes. The physiological significance of the synthesis of these polypeptides and their precursors and its relevance to Cd tolerance and metal homeostasis are discussed.  相似文献

In the present investigation, -galactosidase was solubilized into Aerosol OT (AOT)/isooctane reverse micelles. Kinetic data for the hydrolysis of o-nitrophenyl--D-galactopyranoside (ONPG) at different pH values and molar ratios of water to AOT (Wo) were collected. It was observed that the usual kinetic model used for -galactosidase catalysis in aqueous systems failed to represent the experimental data. A bounded water model, however, showed a better correlation between enzymatic activity and Wo. In contrast to the aqueous system, controlling the water concentration in the reverse micelles allows the rate constants for the reaction between water molecules and glycosyl-enzyme complexes to be evaluated.  相似文献

Using the precursor compound 3,4:10,11-dibenzo-1,13[N,N′-bis{(3-formyl-2-hydroxy-5-methyl)benzyl}diaza]-5,9-dioxocyclopentadecane, a series of macrobicyclic heterobinuclear Ni(II)Zn(II) complexes have been synthesized from the corresponding mononuclear nickel(II) complexes via a template method by Schiff’s base condensation. Electrochemical and kinetic studies of the complexes have been carried out on the basis of macrocyclic ring size. Cyclic voltammetry and controlled electrolysis studies indicate that the nickel(II) metal ion in the heterobinuclear complexes undergo quasireversible one electron reduction and oxidation, whereas the zinc(II) metal ion does not undergo any reduction and oxidation. All the heterobinuclear Ni(II)Zn(II) complexes are ESR inactive and diamagnetic in nature. The kinetics of hydrolysis of 4-nitrophenyl phosphate explores that the catalytic activities of the complexes are found to increase with macrocyclic ring size of the complexes. As the macrocyclic ring size increases, the spectral, electrochemical and catalytic studies of the complexes show variation due to distortion in the geometry of metal centre.  相似文献

Summary When solutions of nucleoside 5-phosphates and trimetaphosphate are dried out at room temperature, nucleoside 5-polyphosphates are formed. The Mg++ ion shows a superior catalytic function in this reaction when compared with other divalent metal ions. Starting with nucleoside 5-phosphates, Mg++ and trimetaphosphate, the predominant products in the nucleoside 5-polyphosphate series pnN are p4N, p7N and P10N. Nucleoside 5-diphosphates yield p5N and p8N, nucleoside 5-triphosphates give p6N and p9N. 1. Increased production of A in a form of lipoprotein antioxidant under the action of increased oxidative stress in aging with subsequent chelation of transition metal ions by A, accumulation of toxic A–metal lipoprotein complexes, production of reactive oxygen species, and neurotoxicity are reviewed and postulated to form the temporal sequence of events in the development of Alzheimer's disease (AD).2. Since (i) A binds copper stronger than iron and other transition metals, and (ii) copper is a more efficient catalyst of oxidation than other transition metals, chelation of copper by A is proposed to be a most important part of this pathway.3. Whereas this amyloid-binds-copper (ABC) model does not remove A peptide from its central place in our current thinking of AD, it places additional factors in the center of discussion.4. Most importantly, they embrace pathological mechanisms known to develop in aging (which is the most important risk factor for AD), such as increased production of reactive oxygen species by mitochondria, that can be positioned upstream relative to the generation of A.  相似文献

A putative -glucosidase clone has been isolated from a cDNA library constructed from mRNA of barley aleurones treated with gibberellin A3 (GA). The clone is 2752 bp in length and has an uninterrupted open reading frame encoding a polypeptide of 877 amino acids. A 680 amino acid region is 43% identical to human lysosomal -glucosidase and other glycosyl hydrolases. In isolated aleurones, the levels of the corresponding mRNA increase strongly after the application of GA, similar to the pattern exhibited by low-pI -amylase mRNA. High levels are also observed in the aleurone and scutellum after germination, while low levels are found in developing seeds. The genome contains a single form of this -glucosidase gene and two additional sequences that may be related genes or pseudogenes.Names are necessary to report factually on available data; however, the USDA neither guarantees nor warrants the standard of the product, and the use of the name by USDA implies no approval of the product to the exclusion of others that may also be suitable.  相似文献

A new ligand N-Nicotinoyl-N-o-hydroxythiobenzhydrazide (H2Notbh) forms complexes [Mn(Notbh)(H2O)], [M(Notbh)] [M=Ni(II) Cu(II) and Zn(II)] which were characterized by various physico-chemical techniques. All the metal complexes were observed to inhibit the growth of tumor in vitro, whereas, ligand did not. In vivo administration of these complexes resulted in prolongation of survival of tumor bearing mice. Tumor bearing mice administered with metal complexes showed reversal of tumor growth associated induction of apoptosis in lymphocytes. The paper discusses the possible mechanisms and therapeutic implication of the H2Notbh and its metal complexes in tumor regression and tumor growth associated immunosuppression.  相似文献

Summary Aqueous solutions of linear inorganic polyphosphates incubated in presence of Mg ions depolymerize to give trimetaphosphate. The presence of a nucleoside 5-phosphate has little influence upon the reaction. Drying the products obtained by incubating a linear polyphosphate with Mg ions in the presence of a nucleoside 5-phosphate yields nucleoside 5-polyphos-phates. Summary Sirofluor, a chemically defined fluorochrome from aniline blue in aqueous unbuffered solutions, complexes with isolated (1 3)--glucans, but not (1 4)--glucans, after embedding in JB-4 resin and sectioning. Recently, we demonstrated that the 36 kDa catalytic subunit of protein phosphatase 2A (PP2Ac) undergoes methylation at its C-terminal leucine in normal rat islets, human islets and isolated cells; this modification increases the catalytic activity of PP2A [Kowluru et al. Endocrinology. 137:2315–2323, 1996]. Previous studies have suggested that adenine and guanine nucleotides or glycolytic intermediates [which are critical mediators in cell function] also modulate phosphatase activity in the pancreatic cell. Therefore, we examined whether these phosphorylated molecules specifically regulate the carboxyl methylation and the catalytic activity of PP2A in cells. Micromolar concentrations of ATP, ADP, GTP or GDP each inhibited the carboxyl methylation of PP2Ac and, to a lesser degree, the catalytic activity of PP2A. Likewise, the carboxyl methylation of PP2Ac and its catalytic activity were inhibited by [mono- or di-] phosphates of glucose or fructose. Additionally, however, the carboxyl methylation of PP2Ac was significantly stimulated by divalent metal ions (Mn2+ > Mg2+ > Ca2+ > control). The nucleotide or sugar phosphate-mediated inhibition of carboxyl methylation of PP2Ac and the catalytic activity of PP2A were completely prevented by Mn2+ or Mg2+. These data indicate that divalent metal ions protect against the inhibition by purine nucleotides or sugar phosphates of the carboxyl methylation of PP2Ac perhaps permitting PP2A to function under physiologic conditions. Therefore, these data warrant caution in interpretation of extant data on the regulation of phosphatase function by purine nucleotides.  相似文献

T-lymphocyte-mediated immunosuppression has been described in several animal models and in man. In animal models, T-cell-mediated immunosuppression can hasten the development of cancers, permit the growth of tumors in immunocompetent hosts, and inhibit otherwise effective antitumor immunotherapy. Cyclophosphamide can abrogate the T-cell-mediated immunosuppression. However, inappropriately administered cyclophosphamide can adversely affect antitumor immunity. On the basis of data showing that interferon / (IFN/) and IFN selectively abrogate the T-cell-mediated dinitrofluorobenzene-specific suppressor function, we investigated the efficacy of purified murine IFN/ in manipulating tumorinduced T-cell-mediated immunosuppression in the wellcharacterized P815 mastocytoma model. In this model, generation of cytotoxicity in vitro and its inhibition by T cells correlates with antitumor immunity in vivo. We report that IFN/ selectively diminishes the generation of tumor-induced suppressor activity.  相似文献

Nucleation-dependent protein aggregation (seeding) and amyloid fibril-free formation of soluble SDS-resistant oligomers (oligomerization) by hydrophobic interaction is an in vitro model thought to propagate -amyloid (A) deposition, accumulation, and incur neurotoxicity and synaptotoxicity in Alzheimers disease (AD), and other amyloid-associated neurodegenerative diseases. However, A is a high-affinity metalloprotein that aggregates in the presence of biometals (zinc, copper, and iron), and neocortical A deposition is abolished by genetic ablation of synaptic zinc in transgenic mice. We now present in vitro evidence that trace (0.8 µM) levels of zinc, copper, and iron, present as common contaminants of laboratory buffers and culture media, are the actual initiators of the classic A1–42-mediated seeding process and A oligomerization. Replicating the experimental conditions of earlier workers, we found that the in vitro precipitation and amyloidosis of A1–40 (20 µM) initiated by A1–42 (2 µM) were abolished by chelation of trace metal contaminants. Further, metal chelation attenuated formation of soluble A oligomers from a cell-free culture medium. These data suggest that protein self-assembly and oligomerization are not spontaneous in this system as previously thought, and that there may be an obligatory role for metal ions in initiating A amyloidosis and oligomerization.  相似文献

Role of some metal ions on the conformations of peptides was examined by using a series of short alanine-based peptides with single Trp-His (W-H) interaction in different environments. Circular dichroism (CD), Trp (W) fluorescence emission, and Fourier transform infrared (FTIR) spectroscopy revealed that there is a conformational role of Cu2+ in inducing and enhancing the formation of -helix conformation. The complexation of the peptide with Cu2+ is responsible to the conformational effect because the chelation is able to stabilize peptide with an -helix conformation. The possible factors affecting the role of Cu2+ are discussed in the paper. The results in this paper are useful to understand the important structural role of Cu2+ in protein folding and the possible mechanism in some neurodegenerative diseases such as Alzheimer's disease.  相似文献

Summary A new approach to joint regression analysis, entitled Trimmed Joint Regression (TJR), is proposed in which the adjustment of the linear relative-yield pattern of the cultivars is trimmed from the residues which can be attributed to a specific interaction. The ranking of the residues to the joint regressions for each cultivar, within each trial, is analysed by the Friedman test to ascertain if they belong to the same parental distribution of the population of residues, for the different genotypes. The rejection of the null hypothesis is envisaged as the result of an organized pattern of the residues, due to a specific interaction, and the genotypes responsible for such interactions are identified. The trimming method consists of the assessment of the linear regressions, after a reassessment of data related with specific interactions. The increased accuracy which can be achieved is shown in a numerical example where the high repeatability of the method is illustrated by means of a comparison of the estimated yields for 1- and 2-year trials.  相似文献

Transient expression and electrophoretic mobility shift assay were used to investigate the cis elements and the DNA-binding proteins involved in the regulation of expression of a 22 kDa zein-like -coixin gene. A set of unidirectional deletions was generated in a 962 bp fragment of the -coixin promoter that had been previously fused to the reporter gene GUS. The constructs were assayed by transient expression in immature maize endosperm. There was no significant decrease in GUS activity as deletions progressed from –1084 to –238. However, deletion from –238 to –158, which partially deleted the O2c box, resulted in a dramatic decrease in GUS activity emphasizing the importance of the O2 box in the quantitative expression of the gene. The –238 promoter fragment interacted with Coix endosperm nuclear proteins to form 5 DNA-protein complexes, C1–C5, as detected by EMSA. The same retarded complexes were observed when the –158 promoter fragment was used in the binding reactions. Reactions with nuclear extracts isolated from Coix endosperms harvested from 6 to 35 days after pollination revealed that the 5 DNA-protein complexes that interact with the -coixin promoter are differentially assembled during seed development. Deletion analysis carried out on the –238/ATG promoter fragment showed that a 35 bp region from –86 to –51 is essential for the formation of the complexes observed. When nuclear extracts were incubated with an antiserum raised against the maize Opaque-2 protein, the formation of 4 complexes, C1, C3, C4 and C5, was prevented indicating that an Opaque-2 like protein participates in the formation of those complexes. Complex C2 was not affected by the addition of the O2 antibody, suggesting the existence of a novel nuclear factor, CBF1, that binds to the promoter and makes protein-protein associations with other proteins present in Coix endosperm nuclei.  相似文献

This paper reports the first study of an inclusion complex of abrassinosteroid with -cyclodextrin. The formation of inclusion complexesbetween 24-epibrassinolide and -cyclodextrin was confirmed by theirphysicochemical properties and the compounds were analysed by differentialscanning calorimetry, powder X-ray diffraction, nuclear magnetic resonancespectrometry and scanning electron microscopy. Theoretical calculations usingthe MM+ HyperChem force field showed a preference for inclusion of thesidechain of the epibrassinolide molecule into the -cyclodextrin cavity toform a 1:1 inclusion complex, although complexes involving inclusion ofthe steroidal nucleus also possess a favourable interaction energy. Rice laminainclination assay, employing IAC-103 and IAC-104 cultivars, showed an improvedactivity for the epibrassinolide-cyclodextrin complex compared to theepibrassinolide itself. The results suggest that brassinosteroid complexationwith cyclodextrins may enhance the biological activity of these plant growthregulators.  相似文献

Summary Adenosine 5-phosphoramidates form when solutions containing adenosine 5-polyphosphates pnA (n 3) or P1, P2-diadenosine 5-diphosphate and amines are allowed to dry out. Mg ions catalyze these reactions. We have studied systems containing ammonia, imidazole, glycine, ethylenediamine and histamine. The yields of adenosine 5-phosphoramidates range from 10–50 % based on the nucleotide. The amyloid -peptide (A) is a major component of insoluble amyloid deposits in Alzheimers disease, and the ability of the -peptide to exist in different conformations is dependent on residues 1–28 [-(1–28)]. However, different from humans, no A amyloid deposition has been found in aged rats brains. Studying the three-dimensional solution structure of rat A-(1–28) and the binding circumstance of Zn2+ is beneficial to a clear understanding of the potential role of Zn2+ in Alzheimer-associated neuropathogenesis and to suggest why there is no amyloid deposition in aged rats brains. Here we used nuclear magnetic resonance (NMR) spectroscopy to determine the solution structure of rat A-(1–28) and the binding constant of Zn2+ to rat A-(1–28). Our results suggest that (1) the three-dimensional solution structure of rat A-(1–28) is more stable than that of human A-(1–28) in DMSO-d6 and that a helical region from Glu16 to Val24 exists in the rat A-(1–28); (2) the affinity of Zn2+ for rat A-(1–28) is lower than that for human A-(1–28) and the NMR data suggest that Arg13, His6, and His14 residues provide the primary binding sites for Zn2+; and (3) the proper binding of Zn2+ to rat A-(1–28) can induce the peptide to change to a more stable conformation.Electronic Supplementary Material Supplementary material is available in the online version of this article at http://dx.doi.org/10.1007/s00775-004-0556-xAbbreviations A amyloid -peptide - AD Alzheimers disease - hA-(1–28) human A-(1–28) - rA-(1-28) rat A-(1–28) - REM restrained energy minimization  相似文献

The results of NMR studies using several nuclei indicate that proteins have considerable internal mobility. The most obvious is the mobility of side-chains. This mobility is general on the exterior surfaces but extends internally in a differential way. The functional value of surface mobility concerns both on and off rates of ligand binding (e.g. metal ions and parts of substrates) and protein/protein interactions. The mobility, which indicates that recognition is more in the hand-in-glove class than in the lock-in-key class, makes for a modified view of the specificity of protein interactions. Thus, fast on/off systems cannot be as selective as slower systems. Segmental mobility of proteins is considered in the context of protein secondary structure. The least mobile segments are the -sheet and the tight -turn. Mobility is always possible for, but not within, rod-like helices and in loose turns. Many examples are given and the importance of mobility in molecular machines is described. Finally, examples are given of virtually random-coil proteins, segments, and linker regions between domains and the functional value of such extremely dynamic regions of proteins is discussed.This work is based on a lecture at the EBSA Symposium, organised by the Italian Biophysical Society (S.I.B.P.A.), Tabiano Terme, September 1992  相似文献
