Background  

Bioethanol isolated from lignocellulosic biomass represents one of the most promising renewable and carbon neutral alternative liquid fuel sources. Enzymatic saccharification using cellulase has proven to be a useful method in the production of bioethanol. The filamentous fungi Acremonium cellulolyticus and Trichoderma reesei are known to be potential cellulase producers. In this study, we aimed to reveal the advantages and disadvantages of the cellulase enzymes derived from these fungi.  相似文献

Background  

To reduce the production cost of bioethanol obtained from fermentation of the sugars provided by degradation of lignocellulosic biomass (i.e., second generation bioethanol), it is necessary to screen for new enzymes endowed with more efficient biomass degrading properties. This demands the set-up of high-throughput screening methods. Several methods have been devised all using microplates in the industrial SBS format. Although this size reduction and standardization has greatly improved the screening process, the published methods comprise one or more manual steps that seriously decrease throughput. Aim

Studies that monitor high‐mountain vegetation, such as paramo grasslands in the Andes, lack non‐destructive biomass estimation methods. We aimed to develop and apply allometric models for above‐ground, below‐ground and total biomass of paramo plants.

Location

The paramo of southern Colombia between 1°09′N and 077°50′W, at 3,400 and 3,700 m a.s.l.

Methods

We established 61 1‐m2 plots at random locations, excluding disturbed, inaccessible and peat bog areas. We measured heights and basal diameters of all vascular plants in these plots and classified them into seven growth forms. Near each plot, we sampled the biomass from plants of abundant genera, after having measured their height and basal diameter. Hence, we measured the biomass of 476 plants (allometric set). For each growth form we applied power‐law functions to develop allometric models of biomass against basal diameter, height, height x basal diameter and height × basal area. The best models were selected using AICc weights. Using the observed and predicted plant biomass of the allometric set we calculated absolute percentage errors using cross‐validation. The biomass of a plot was estimated by summing the predicted biomass of all plants in a plot. Confidence limits around these sums were calculated by bootstrapping.

Results

For groups of <20 plants the biomass predictions yielded large (>15%) errors. Applying groups that resembled the 1‐m2 plots in density and composition, the errors for above‐ground and total biomass estimates were <15%. Across all plots, we obtained an above‐ground, below‐ground and total plot biomass of 329 ± 190, 743 ± 486 and 1011 ± 627 g/m2 (mean ± SD), respectively. These values were within the range of biomass estimates obtained destructively in the tropical Andes.

Conclusions

In new applications, if target vegetation samples are similar regarding growth forms and genera to our allometric set, their biomass might be predicted applying our equations, provided they contain at least 50–100 plants. In other situations, we would recommend gathering additional biomass measurements from local plants to evaluate new regression equations.  相似文献

Due to its capacity to produce large amounts of cellulases, Trichoderma reesei is increasingly been researched in various fields of white biotechnology, especially in biofuel production from lignocellulosic biomass. The commercial enzyme mixtures produced at industrial scales are not well characterized, and their proteinaceous components are poorly identified and quantified. Background  

Hydrolysis of cellulose requires the action of the cellulolytic enzymes endoglucanase, cellobiohydrolase and β-glucosidase. The expression ratios and synergetic effects of these enzymes significantly influence the extent and specific rate of cellulose degradation. In this study, using our previously developed method to optimize cellulase-expression levels in yeast, we constructed a diploid Saccharomyces cerevisiae strain optimized for expression of cellulolytic enzymes, and attempted to improve the cellulose-degradation activity and enable direct ethanol production from rice straw, one of the most abundant sources of lignocellulosic biomass.  相似文献

Questions

Can hemiparasitic Rhinanthus major originating from a local population suppress the competitive clonal grass Calamagrostis epigejos and reverse its expansion in species‐rich semi‐natural grasslands? Does sowing seeds of R. major facilitate restoration of target meadow vegetation? Is R. major more beneficial for biodiversity restoration/conservation than increased mowing intensity, a conventional measure to suppress C. epigejos?

Methods

We conducted a before‐after‐control‐impact experiment in meadow patches heavily infested by C. epigejos: eight blocks, each containing four plots with four treatment combinations: (1) traditional management, i.e. mowing once in summer, (2) mowing in summer and autumn (3) mowing in summer and seed sowing of R. major, (4) mowing in summer and autumn and seed sowing of R. major. Above‐ground biomass of C. epigejos and vegetation composition of each of the plots were monitored every year from 2013 to 2016. To assess the effects of treatments, we analysed biomass production of C. epigejos, herb layer cover and vegetation composition.

Results

Both sowing R. major and an additional autumn meadow cut significantly suppressed C. epigejos. Their effects were additive and of comparable size. Both treatments also had significant but markedly different effects on community composition. Rhinanthus major facilitated directional community composition change towards the regional Brachypodio‐Molinetum meadows. In contrast, increased mowing intensity significantly decreased frequency of threatened species, which however may have also been influenced by R. major.

Conclusions

Sowing of autochthonous R. major seeds was demonstrated as an efficient tool to suppress C. epigejos and facilitate community restoration. It can be combined with an additional meadow cut to further accelerate decline of the grass. The additional cut should however be used as a short‐term practice (1–2 years) only to minimize potential negative effects of its long‐term application on some threatened plant species. The effects of R. major are comparable to those of Rhinanthus alectorolophus reported previously. As a species occurring naturally in species‐rich dry grasslands, R. major has a broader and longer‐term application potential than R. alectorolophus in ecological restoration and conservation of these communities.  相似文献

Cellular metabolism is one of the most investigated system of biological interactions. While the topological nature of individual reactions and pathways in the network is quite well understood there is still a lack of comprehension regarding the global functional behavior of the system. In the last few years flux-balance analysis (FBA) has been the most successful and widely used technique for studying metabolism at system level. This method strongly relies on the hypothesis that the organism maximizes an objective function. However only under very specific biological conditions (e.g. maximization of biomass for E. coli in reach nutrient medium) the cell seems to obey such optimization law. The production of microbial lipids has attracted considerable interest during the past decade since they can be successfully used to produce biodiesel by catalyzed transesterification with short chain alcohols. Aim

Decreasing in the diversity and distribution of native submerged plants have been widely observed in recent decades. Global underwater darkening, which is mainly caused by radiation dimming and a decrease in transparency due to, e.g. eutrophication, has emerged as a general trend that strongly hampers the growth of submerged plants in lakes by decreasing the light available for photosynthesis. However, few studies have attempted to compare the responses of native and invasive submerged plants to underwater darkening. In this study, we aimed to compare the effects of light attenuation on the growth and photosynthesis traits of native and invasive submerged plants.

Method

Through field investigations and a mesocosm experiment, the responses of functional traits of two representative native [water thyme (Hydrilla verticillata), Eurasian watermilfoil (Myriophyllum spicatum)] and one invasive [Carolina fanwort (Cabomba caroliniana)] plant species to various environmental factors, notably to underwater light attenuation, were studied.

Results

Underwater photosynthetically active radiation (PAR) exerted a substantial effect on the relative coverage and abundance of the three studied submerged plants in their natural freshwater habitats. Invasive C. caroliniana showed relatively superior growth (total biomass and relative growth rate) and photosynthesis traits (maximum quantum yield of photosystem II Fv/Fm, chlorophyll a content, chlorophyll b content and the ratio of Chl a and b contents) compared to the two native plants under low underwater PAR conditions. In contrast, under high underwater PAR conditions, C. caroliniana showed the opposite response.

Main Conclusions

Light attenuation inhibits the growth of native submerged plants but facilitates the growth of invasive plant species. Restoration of freshwater lakes by reducing deterioration from underwater darkening (for instance, by reducing of external nutrients loading) may therefore constrain the growth and spread of the invasive C. caroliniana.  相似文献

Aim

Typha latifolia causes serious problems in wet meadows by overgrowing and suppressing other native plants. To determine suitable management for T. latifolia control, we addressed the following question: What are the effects of long-term cutting at different frequencies (once or twice per year and no management) and biomass removal on cover and other characteristics of T. latifolia, and on sward productivity and plant species composition?

Malá Strana nature reserve, Jizerské hory Mountains, Czechia.

Methods

A long-term experiment arranged in a randomised block design with three blocks was established in 2005. Data were collected from five treatments: unmanaged control; cutting once a year in June without biomass removal and with biomass removal; cutting twice per year in June and August without biomass removal and with biomass removal. Percentage cover of T. latifolia and other vascular plant species was visually estimated and T. latifolia characteristics (tiller density, height, dry-matter biomass [DMB] yield and litter), sward height and DMB yield were measured during 2005–2018 at the end of June.

Results and Discussion

Regular cutting once or twice per year regardless of cut biomass removal led to reductions in tiller density, height, litter and DMB yield of T. latifolia. Biomass removal had only a slight tendency to affect T. latifolia characteristics. The higher frequency of cutting significantly decreased the mean T. latifolia cover, litter and DMB yield. Cutting once or twice per year regardless of biomass removal led to successive changes in plant species composition but had no effect on the species richness and evenness.

Conclusions

Cutting at least once per year without biomass removal seems to be sufficient to achieve a decrease in DMB yield and litter of T. latifolia plants, and thereby maintain the wet-meadow vegetation without loss of species richness and also preventing the overgrowth of shrubs and trees.  相似文献
