Ionizing radiation causes considerable changes in the content of sterols and squalene in the lymphoid system tissues and erythrocyte membranes which is in accordance with the concept of high radiosensitivity of haemopoietic tissue. The processes of cholesterol conversion to its oxy-derivatives are increased under the effect of radiation. The content of some lipid components in the lymphoid system tissues and erythrocyte membranes is changed depending on the dose and time after irradiation. There is a relationship between the changes in the sterol composition and in the properties of erythrocyte membranes.  相似文献

Structural characteristics and Na, K-ATPase activity of erythrocyte membranes were studied by the spin probe method in patients with an angiographically verified damage to the coronary arteries. The rise of the cholesterol/phospholipid ratio in patients with coronary heart disease was associated with the increased orderliness of the fatty acid chains of erythrocyte membrane lipids, leading to inhibition of Na, K-ATPase activity. The data point to the same line of changes in erythrocyte membranes and smooth muscle cells during the development of coronary heart disease.  相似文献

The erythrocyte intracellular pressure was calculated using a biomechanical model of the erythrocyte and atomic-force-microscopy (AFM) data. The intracellular pressure was characterized as a function of the model erythrocyte morphology. Based on numerical modeling and data of erythrocyte imaging by AFM, a method was developed to estimate the erythrocyte intracellular pressure by comparing experimental data and the results of numerical calculations. Calculations were performed for erythrocytes of dwarf domestic pigs with and without obstructive jaundice that varied in severity. The erythrocyte membrane was affected with increasing disease severity and blood bilirubin concentration, i.e., the erythrocyte volume increased on average, and substantial changes were observed for erythrocyte intracellular pressure relative to its normal value. As an example, an increase in bilirubin concentration from 5 to 96 μmol/L was associated with an increase in intracellular pressure from 0 to 2.2 kPa. An examination of the erythrocyte membrane surface by high-resolution AMF showed that the membrane is disrupted with an increase in bilirubin concentration and displays lesions and an increasing rupture length.  相似文献

Actin in the human erythrocyte forms short protofilaments which are only long enough to accommodate tropomyosin monomers (Shen, B.W., Josephs, R. and Steck, T.L. (1986) J. Cell Biol. 102, 997-1006). This interaction between actin and tropomyosin monomers is predicted to be weak, since tropomyosin polymerization parallels its affinity for F-actin. We examine the binding of human erythrocyte tropomyosin to actin in the presence and absence of spectrin and its ability to polymerize. The binding of human erythrocyte tropomyosin to F-actin is not affected appreciably by the present of spectrin. Saturating F-actin with erythrocyte tropomyosin, however, weakens the binding of spectrin dimers to actin. Although tropomyosin from human erythrocyte and rabbit cardiac muscle have similar affinity for F-actin, the polymerizability of erythrocyte tropomyosin as determined by viscosity measurements is much reduced relative to muscle tropomyosin. This unusual property of erythrocyte tropomyosin is likely due to differences in its primary structure from other known tropomyosin at the amino and carboxyl terminal regions which are responsible for its head-to-tail polymerization and cooperative binding to F-actin. Analysis of the distribution of tyrosine by 2-dimensional tryptic mapping of 125I-labelled erythrocyte tropomyosin shows that tyrosine at positions 162, 214, 221, 261 and 267 in rabbit cardiac tropomyosin are conserved in human erythrocyte tropomyosin but Tyr-60 is absent. This observation suggests that erythrocyte tropomyosin has a carboxyl terminal region similar to its muscle counterparts but its amino terminal region resembles that of platelet tropomyosin which also lacks Tyr-60.  相似文献

目的 :探讨运动对红细胞变形性和红细胞膜蛋白的影响及其相互关系。方法 :设计不同强度的训练方案 ,用激光衍射法测定红细胞变形能力 ,用SDS PAGE方法测定一定体积大鼠红细胞膜中的重要蛋白带 3蛋白 (band 3)和肌动蛋白 (actin)的含量 ,研究运动即刻和恢复后红细胞变形性及膜蛋白的变化。结果 :长期的运动训练会促进大鼠红细胞变形能力的改善和红细胞膜band 3蛋白和actin的良好发展 ,一次大强度训练会引起红细胞膜band 3蛋白和actin含量的减少 ,大鼠红细胞变形能力降低 ,一周和二周的大强度训练会提高恢复期大鼠红细胞的变形能力和红细胞膜band 3蛋白和actin含量。结论 :运动训练造成的红细胞膜蛋白含量的变化 ,导致了红细胞膜结构的改变 ,从而影响红细胞变形能力 ,可能是训练对红细胞变形能力的作用机制之一。  相似文献

The Mr 245,000 calmodulin-binding protein of the dogfish erythrocyte cytoskeleton (D245) has been compared with human erythrocyte spectrin and mammalian brain fodrin [J. Levine and M. Willard (1981) J. Cell Biol. 90, 631-643]. Mammalian erythrocyte alpha-spectrin, brain alpha-fodrin, and D245 are all localized in the cell surface-associated cytoskeleton, and have similar molecular weights. Like mammalian erythrocyte spectrin, D245 was extracted from erythrocyte ghosts under low-ionic-strength conditions. However, D245 failed to bind an antibody which reacted strongly with both subunits of human erythrocyte spectrin. Unlike mammalian erythrocyte alpha- and beta-spectrin, D245 bound calmodulin in the absence of urea both in a "gel-binding" assay and in situ using azidocalmodulin [D.C. Bartelt, R.K. Carlin, G.A. Scheele, and W.D. Cohen (1982) J. Cell Biol. 95, 278-284]. Striking similarities were noted between D245 and alpha-fodrin in that both exhibited (a) comparable calcium-dependent calmodulin binding properties, (b) strong reactivity with two different anti-fodrin antibody preparations, (c) similar reactivity with antibody to brain CBP-I, now believed to be fodrin, (d) proteolytic degradation yielding an Mr 150,000 calmodulin-binding fragment, and (e) lack of reactivity with an anti-spectrin antibody. A protein with calmodulin-binding and anti-fodrin-binding properties similar to D245 was detected in cytoskeletal preparations of chicken erythrocytes. Moderate and consistent cross-reactivity of anti-fodrin with human erythrocyte alpha-spectrin was also observed. The data indicate that D245 is functionally and immunologically more closely related to alpha-fodrin than to alpha-spectrin of the mammalian erythrocyte.  相似文献

A study of human erythrocyte and brain spectrin with particular emphasis on the beta subunits revealed a structural homology but functional dissimilarity between these two molecules. Six monoclonal antibodies raised to human erythrocyte beta spectrin identify three of the four proteolytically defined domains of erythrocyte beta spectrin. Five of these monoclonal antibodies cross-react with human brain spectrin. None of a previously identified set of alpha erythrocyte spectrin monoclonal antibodies [Yurchenco et al: J Biol Chem 257:9102, 1982] reacted with brain spectrin. A domain map generated by limited tryptic digestion shows that brain spectrin is composed of proteolytically resistant domains analogous to erythrocyte spectrin, but the brain protein is more basic. The binding of brain spectrin to erythrocyte ankyrin, both in solution and on erythrocyte IOVs, yielded an association constant approximately 100 time weaker than for erythrocyte spectrin. The binding of azido-calmodulin under native conditions was specific for the erythrocyte beta subunit but was not calcium dependent. In contrast, azido-calmodulin bound only to the alpha subunit of brain spectrin in a calcium-dependent manner. The similarity of structure but modified functional characteristics of the brain and erythrocyte beta spectrins suggest that these proteins serve different cellular roles.  相似文献

Morphofunctional and biochemical properties of erythrocyte membrane were investigated in early postnatal ontogenesis in rats in norm and after prenatal immobilization stress. The transient decrease of erythrocyte membranes stability was revealed in the control rats. The ability to erythrocyte transformation and the concentration of lipid peroxidation products are increased. It has been shown by an increase of percentage discocytes and lower lipid peroxidation level that the erythrocyte membrane of the rats after prenatal stress is more stable.  相似文献

To investigate the role played by plasmatic lipids in the altered erythrocyte deformability observed in cirrhotic patients we studied 15 patients with liver cirrhosis (histologically diagnosed) and 10 healthy volunteers. Erythrocyte filtration time, plasmatic free and esterified cholesterol and phospholipids were measured in all subjects. The erythrocyte filtration time resulted to be significantly increased in cirrhotic patients (35' +/- 3, 35 M +/- SEM) when compared to control subjects (26' +/- 2, 53: M +/- SEM) (t = 2,078 p less than 0,05). This increase correlated in cirrhotic patients (but not in control subjects) with free/esterified cholesterol ratio (p less than 0,01) as well as free cholesterol/phospholipid ratio (p less than 0,001). Our results confirm that decreased erythrocyte deformability in cirrhotic patients which is accompanied by altered erythrocyte morphology is due, at least in part, to the altered lipids blood levels.  相似文献

Adhesion followed by fusion of LP-X vesicles with the erythrocyte membrane is an important contribution to the erythrocyte enlargement in patients with intra or extra hepatic cholestasis. Adhesion of LP-X vesicles is demonstrated by thin section and freeze-etch electronmicroscopy. Fusion of LP-X with the erythrocyte membrane is deduced from biochemical data and freeze-etch electronmicroscopy in that the uptake of cholesterol and lecithin coincides with the increase in smoooth areas on the fracture faces of the erythrocyte membrane.  相似文献

α-Tocotrienol (α-T3) has been suggested to protect cellular membranes against free radical damage. This study was done to estimate the effect of α-T3 on free radical-induced impairment of erythrocyte deformability by comparing it to α-tocopherol (α-T). An erythrocyte suspension containing 2,2′-azobis (2-amidinopropane) dihydrochloride (AAPH) was forced to flow through microchannels with an equivalent diameter of 7 μm for measuring erythrocyte deformability. A higher concentration of AAPH caused a marked decrease in erythrocyte deformability with concomitant increase of membranous lipid peroxidation. Treatment of erythrocytes with α-T or α-T3 suppressed the impairment of erythrocyte deformability as well as membranous lipid peroxidation and they also increased erythrocyte deformability even in the absence of AAPH. In these cases, the protecting effect of α-T3 was significantly higher than that of α-T. We emphasize that higher incorporating activity of α-T3 into erythrocyte membranes seems to be the most important reason for higher protection against erythrocyte oxidation and impairment its deformability.  相似文献

Plasma and erythrocyte samples from Trygonorhina fasciata guanerius were analysed for urea content. The erythrocyte urea concentration is higher than plasma urea concentration. The difference in concentration is probably due to the presence of urea normally bound to protein within the erythrocyte. Erythrocyte urea concentrations followed environmentally induced changes in plasma urea concentrations. Naturally occurring plasma urea concentrations, exceeding 500 mMolar, are some of the highest recorded for an elasmobranch.  相似文献

The different isozymes of carbonic anhydrase (CA) have been the subject of intensive study in mammals, but there is still much to be learned about the early evolution of this enzyme in vertebrates. Erythrocyte CA plays an essential role in the respiratory processes of most vertebrates and is probably the most well studied CA isozyme. The available evidence indicates that there has been a progressive increase in the efficiency of erythrocyte CA during the early evolution of vertebrates. There also appears to be a substantial increase in erythrocyte CA activity during development in some species. At the present time, however, the selective pressures that may be influencing the properties of erythrocyte CA during vertebrate evolution and development have not been clearly determined. When the available molecular sequence information is examined, it is evident that the erythrocyte CAs of early vertebrates have active sites that are more similar to those of mammalian CA VII and II, rather than CA I. We can now also begin to examine the phylogenetic relationships between the different rbc CAs in vertebrates, but more CA sequence information is clearly required from different groups of vertebrates before we have a complete picture of the molecular evolution of erythrocyte CA.  相似文献

A study was made of a combined influence of the temperature of the medium and of sensitin concentration on the process of interaction of tannin-treated sheep erythrocytes and human serum albumin. On the basis of determination of the number of molecules bound by a single erythrocyte during hemosensitization,, depending on the mentioned conditions, it was revealed that the relative total albumin binding increased with the elevation of temperature. Elevation of temperature also led to the absolute and the relative increase in the stable and a reduction in the loose albumin binding by a single erythrocyte and the growth of sensitivity of the passive hemagglutination test. The role of chemical mechanisms in the erythrocyte albumin loading was demonstrated; this permitted to carry out erythrocyte albumin sensitization at a comparatively high temperature for the purpose of increasing the efficacy of passive hemagglutination test.  相似文献

After administration of [4-(14)C]cholesterol to rats, blood was obtained and incubated for 6 hr or less. Incubation resulted in a net loss of erythrocyte cholesterol and, simultaneously, in an increase of esterified cholesterol in plasma and alpha-lipoproteins. Erythrocyte labile cholesterol was shown to be the sole precursor of esterified cholesterol. However, the relation between loss and esterification was not absolute. Loss of erythrocyte cholesterol could be inhibited without affecting esterification and vice versa. A catenary turnover model is proposed, which links in vivo erythrocyte labile cholesterol and plasma esterified cholesterol. Free cholesterol also exchanged between erythrocytes and lipoproteins. The topological model, as tested by analog computer, appears to be a bicompartmental system governed by nonconstant exchange fluxes. They are exponential functions of time and vary from 0.065 to 0.020 mg/hr/g of blood. The fitting of the curves obtained by analog computer analysis to the experimental curves requires esterification as described above. Variation of the exchange fluxes would be the consequence of lipoprotein structural alterations. If this is true, the initial value of the measured flux in vitro is identical with the in vivo value, and the turnover time of erythrocyte cholesterol is 9.2 hr. Initial exchange flux is not dependent on plasma cholesterol level or on the age of the rats, but it is temperature dependent. Addition of amphotericin B to the plasma does not modify exchange fluxes, but erythrocyte cholesterol loss is increased.  相似文献

This investigation was focussed on the gravity of tissue injury caused by complete ischemia (for five min) and hypoxia (for three weeks) in the cerebral cortex (homogenate) and the erythrocyte lysate or the erythrocyte membrane of the rat in order to investigate if the changes that occur in brain tissue are reflected in the erythrocyte. To this end, glutathione (GSH), superoxide dismutase (SOD) and catalase were measured, also alterations in beta-adrenoceptor density under these two conditions were examined. It was found that in ischemia partial parallelism in changes that occur in the central nervous system (cerebral cortex) and the erythrocyte exists. The SOD activity became higher and the beta-adrenoceptor density (measured as specific (-)-[125I] iodocyanopindolol binding) was decreased in both tissues. However after the hypoxic condition we established a decrease in the number of beta-adrenoceptors in the cerebral cortex but an increase in beta-adrenoceptor density in the erythrocyte.  相似文献

Human neutrophils incubated with phorbol myristate acetate oxidized hemoglobin within the intact erythrocyte by a mechanism dependent on cell-cell contact but independent of phagocytosis. Spectrophotometric examination of the erythrocyte lysates revealed that the major component formed was methemoglobin along with small amounts of a species with spectral characteristics similar to choleglobin. Methemoglobin formation was directly related to the neutrophil concentration and the time of incubation. The addition of superoxide dismutase or catalase modestly inhibited the formation of methemoglobin, while a combination of the enzymes provided the most dramatic protection. Methemoglobin of hydroxyl radical or hypochlorous acid scavengers. Apparently, either O2.- or H2O2 alone was capable of mediating methemoglobin formation in the intact erythrocyte. Maintenance of the intraerythrocytic hemoglobin in its oxygenated state or its derivatization to carbon monoxyhemoglobin markedly inhibited methemoglobin formation. Blockade of the anion channels in the intact erythrocyte with sulfonated stilbenes inhibited O2.- but not H2O2 from oxidizing intracellular hemoglobin. It appears that neutrophil-derived O2.- and H2O2 can cross the erythrocyte membrane through the anion channel or diffuse directly into the intracellular space and react with oxyhemoglobin or deoxyhemoglobin to form a mixture of hemoglobin oxidation products within the intact cell.  相似文献

A study was made of the process of interaction between the tannin-treated sheep erythrocytes and the human serum albumin. Protein binding increased, whereas the loose/stable binding ratio decreased with the rise of the initial protein concentration. The process of stable albumin binding is described by the Langmour equation, this permitting to assess the limiting binding values -- about 6-10(5) molecules per erythrocyte. Albumin molecules were bound by erythrocyte with their greatest surface. At any concentration albumin was incapable of occupying more than 85% of the erythrocyte surface; at 90% binding level it blocked only 51--52% of the tannin-treated erythrocyte. The data obtained were regarded as a methodical basis for the preparation of erythrocytic diagnostic agents with the proteins of albumin type.  相似文献

The extent and time course of the impairments occurring in whole blood and erythrocyte cells stored under blood bank conditions were studied by monitoring the reduction of MAL-6 spin label added to the media containing whole blood or erythrocyte cells using electron spin resonance (ESR) technique. Impairments forming in the erythrocyte cells incubated for various times at 37 degrees C were also studied. Erythrocyte cells were found to undergo changes during the storage or incubation, leading to fast decay of MAL-6 spin labels signal height. The extent of the changes depends on storage or incubation time. However, the reduction in incubated or artificially aged erythrocyte (AAE) cells was faster than the reduction in whole blood (WB) and aged erythrocyte (AE) cells stored under blood bank conditions. Two exponential curves attributed to the liquid and cellular parts of a given samples were found to be described best in the reduction of MAL-6 spin label in WB, AE and AAE.  相似文献
