To explore the possible bioactive compounds and to study the antioxidant capacity of Coscinium fenestratum (Goetgh.) Colebr (Menispermaceae), the qualitative and quantitative phytochemical screening for various secondary metabolites were evaluated. Using the GC–MS analysis, a total number of 30 phytochemical compounds were predicted with their retention time, molecular weight, molecular formula, peak area, structure and activities. The most prevailing heterocyclic compound was Bis(2,4,6- triisopropylphenyl) phosphinicazide (6.70%). The antioxidant activity was evaluated by spectrophotometric methods using the reducing power assay and the DPPH and ABTS+ scavenging assays. The activity was determined to be increased in all the test samples with the increase in the volume of the extract. C. fenestratum possess a good source of many bioactive compounds that are used to prevent diseases linked with oxidative stress. 相似文献

Salt stress, which is dominant among environmental stresses, poses challenges to global agriculture. We studied the role of exogenous application of sodium chloride (NaCl) in three arid and three semi-arid genotypes of mungbean [Vigna radiata (L.) Wilczek] by examining some physiological and biochemical stress indicators. Ten-day old seedlings were subjected to salt stress (00–250 mM) by split application along with the half strength Hoagland’s medium. The salt stress caused a decline in the fresh weight, dry weight, relative water content, photosynthetic pigments (chlorophyll and carotenoids) and glutathione content of the seedlings. On the other hand, it increased the electrolyte leakage, lipoxygenase activity, and the proline, protein and total soluble sugar contents. Osmolyte accumulation was relatively higher in the arid genotypes revealing that they are more tolerant to NaCl stress. The physiological and biochemical screening provides a basic platform for selecting the stress-tolerant genotypes in the absence of suitable salt-tolerance markers in mungbean. 相似文献

A series of copper(II) and zinc(II) complexes involving a tridentate O,N,O'-donor Schiff base derived from salicylaldehyde and beta-alanine {i.e. N-salicylidene-beta-alanine(2-), (L)}, having the composition [Cu(2)(L)(2)(H(2)O)].H(2)O (1), [Cu(L)(H(2)O)](n) (2), and [Zn(L)(H(2)O)](n) (3), have been prepared and characterized by elemental analyses, UV-visible (UV-VIS), FT-IR and ESI-MS spectra, and thermal analyses. Complexes 1 and 2 have been investigated by single crystal X-ray analysis and also by temperature dependent magnetic susceptibility measurements (294-80K). All prepared complexes have been evaluated by the antiperoxynitrite activity assay and alloxan-induced diabetes model. The significant antioxidant and antidiabetic activities have been found in the case of both copper(II) complexes 1 and 2. In spite of first two complexes, the zinc(II) complex 3, as well as the potassium salt of the ligand (KHL) showed only insignificant protective effect against the tyrosine nitration in vitro. 相似文献

We have developed a new class of PDE10A inhibitor, a pyrazolo[1,5-a]pyrimidine derivative MT-3014 (1). A previous compound introduced was deprioritized due to concerns for E/Z-isomerization and glutathione-adduct formation at the core stilbene structure. We discovered pyrazolo [1,5-a] pyrimidine as a new lead scaffold by structure-based drug design utilizing a co-crystal structure with PDE10A. The lead compound was optimized for in vitro activity, solubility, and selectivity against human ether-á-go-go related gene cardiac channel binding. We observed that MT-3014 shows excellent efficacy in rat conditioned avoidance response test and suitable pharmacokinetic properties in rats, especially high brain penetration. 相似文献

Myriadenolide is a diterpene that we have recently isolated from the extract of Alomia myriadenia (Asteraceae). Here we show for the first time that myriadenolide has caspase-dependent cytotoxic activity against human leukemia cells from both lymphocytic (Jurkat) and monocytic (THP-1) lineages, because preincubation of Jurkat or THP-1 cells with the broad-spectrum caspase inhibitor z-VAD-fmk completely abrogated cell death. Moreover, the mitochondrial pathway is implicated as mitochondrial depolarization and caspase-9 and caspase-3 activation were observed. Interestingly, caspase-8 and cleavage of the proapoptotic member of the Bcl-2 family BID was also observed during apoptosis induced by myriadenolide, suggesting a role for the caspase-8/BID pathway. However, interference with Fas or TNFR1 signaling did not interfere with apoptosis in our experimental system. Furthermore, pretreatment of cells with the caspase-3 inhibitor DEVD-fmk completely blocked the activation of caspase-8, suggesting that the activation of the caspase-8/BID pathway is part of an amplification loop initiated by caspase-3. Taken together, our results indicate myriadenolide as a novel candidate for the treatment of hematological malignancies. 相似文献

The jaw apparatus, or lantern, of sea-urchins contains five pairs of retractor and protractor muscles which are responsible
for lantern displacement. Using intact retractor or protractor groups, the force-length relations of these muscles were compared
in two taxonomically distant species, Paracentrotus lividus and Stylocidaris affinis. The total contractile forces generated by the muscles can be resolved into vertical and horizontal components. It was found
that the vertical component of the retractors is maximal at a lantern position which is significantly lower (i.e. more protruded)
in Paracentrotus than in Stylocidaris. Total forces generated by the retractors were in both species maximal at or above the lantern `resting positions'. In Paracentrotus alone, the total force-displacement curves tended to be bimodal. It is hypothesized that the retractors of Paracentrotus contain two populations of muscle fibres, one adapted for jaw opening and one for lantern retraction. No significant differences
in the properties of the protractors of the two species could be identified. The lantern of Paracentrotus is more mobile than that of Stylocidaris and is able to exploit a wider range of food sources. This investigation has shown that the force-length relations of the
lantern muscles match their differing working conditions.

Rhinoleucophenga pallida Hendel, 1917 (type species of the genus) is redescribed based on its female holotype and a male from a nearby locality, and Rhinoleucophenga obesa (Loew, 1872) on its two syntypes, which are designated as the male lectotype and a female paralectotype. Both are valid species. A proposal is made to establish the genus Pseudophortica Sturtevant, 1918 (type species R. obesa), a junior synonym of Rhinoleucophenga, to subgenus rank and include all species of Rhinoleucophenga described or redescribed from males except R. pallida, which is unique in having a remarkable pedunculate surstylus, among other differences. The North American R. obesa is compared to its closest sibling, the South American species Rhinoleucophenga gigantea (Thomson, 1869). The occurrence of R. obesa in Brazil is also questioned, as suggested long ago by Marshall R. Wheeler. The specimens from Brazil previously identified as such most probably belong to the new species described in the present paper as Rhinoleucophenga (Pseudophortica) cantareira sp. nov. (type locality: Parque Estadual da Cantareira, City of São Paulo, State of São Paulo, Brazil). Numerous photomicrographs of their habitus and male terminalia taken with a Smartphone's rear camera and digitally stacked to create images with greater depth of focus are provided. 相似文献

A new genus and species of Euptychiina (Satyrinae), Cristalinaia vitoria Mota, Zacca & Freitas gen. et sp. nov., is described based on three specimens collected in the region of the Cristalino River, Alta Floresta, Mato Grosso, Brazil. This rare species is known only from this region, where it flies inside the dense bamboo patches typical of that area. The gypsy moth, Lymantria dispar L. (Lepidoptera, Lymantriidae), a serious defoliator of deciduous trees, is an economically important pest when population densities are high. Outbreaking populations are, however, subject to some moderating influences in the form of entomopathogens, including several species of microsporidia. In this study, we conducted laboratory experiments to investigate the transmission of an unusual Nosema sp. isolated from L. dispar in Schweinfurt, Germany; this isolate infects only the silk glands and, to a lesser extent, Malpighian tubules of the larval host. The latent period ended between 8 and 15 days after oral inoculation and spores were continuously released in the feces of infected larvae until pupation. Exclusion of feces from the rearing cages resulted in a 58% decrease in horizontal transmission. The silk of only 2 of 25 infected larvae contained microsporidian spores. When larvae were exposed to silk that was artificially contaminated with Nosema sp., 5% became infected. No evidence was found for venereal or transovum (including transovarial) transmission of this parasite. 相似文献

Aiming to identify new sources of bioactive secondary metabolites, we isolated 82
endophytic fungi from stems and barks of the native Brazilian tree Caesalpinia
echinata Lam. (Fabaceae). We tested their ethyl acetate extracts in several in vitro
assays. The organic extracts from three isolates showed antibacterial activity
against Staphylococcus aureus and Escherichia coli [minimal inhibitory concentration
(MIC) 32-64 μg/mL]. One isolate inhibited the growth of Salmonella typhimurium (MIC
64 μg/mL) and two isolates inhibited the growth of Klebsiella oxytoca (MIC 64 μg/mL),
Candida albicans and Candida tropicalis (MIC 64-128 μg/mL). Fourteen extracts at a
concentration of 20 μg/mL showed antitumour activities against human breast cancer
and human renal cancer cells, while two isolates showed anti-tumour activities
against human melanoma cancer cells. Six extracts were able to reduce the
proliferation of human peripheral blood mononuclear cells, indicating some degree of
selective toxicity. Four isolates were able to inhibit Leishmania (Leishmania)
amazonensis and one isolate inhibited Trypanosoma cruzi by at least 40% at 20 μg/mL.
The trypanocidal extract obtained from Fusarium sp. [] culture was subjected
to bioguided fractionation, which revealed beauvericin as the compound responsible
for the observed toxicity of Fusarium sp. to T. cruzi. This depsipeptide showed a
half maximal inhibitory concentration of 1.9 μg/mL (2.43 μM) in a T. cruzi cellular

The species Naarda postpallida de Joannis, 1929 is reported for the first time from mainland China. Naarda pectinata Sugi, 1982 is synonymised with N. postpallida on the basis of material from Vietnam. This species can immediately be recognised by its dark grey forewing, white hindwing and several genital characters. The female genitalia of N. postpallida, together with those of N. lingualis Tóth and Ronkay, 2015 are presented for the first time. Naarda macrostigma sp. n. is described. Background
Snake venoms are rich sources of bioactive molecules, and several venom-derived proteins have entered clinical trials for use in ischemic disorders; however, late-stage failure of a recent drug candidate due to low in vivo efficacy demonstrated the need for new sources of fibrinogenolytic drug candidates.Methods
A 51.3 kDa thrombin-like serine protease (Russelobin) purified from the venom of Russell's Viper (Daboia russelii russelii) was subjected to extensive biochemical characterization, including N-terminal sequencing, substrate specificity, kinetic and inhibitor assays, glycosylation analysis and stability assays. Toxicity and pathology analyses were conducted in NSA mice.Results
Russelobin has extensive N-terminus identity with a beta-fibrinogenase-like serine proteinase precursor from Daboia russelii siamensis venom, a mass of 51.3 kDa and contains extensive N-linked oligosaccharides. Serine protease inhibitors and heparin significantly decreased activity, with much lower inhibition by DTT, antithrombin-III and α2-macroglobulin. Russelobin preferentially released FPA and slowly released FPB from human fibrinogen, forming a labile fibrin clot readily hydrolyzed by plasmin. The partially deglycosylated enzyme showed significantly lower activity toward fibrinogen and less resistance against neutralization by plasma α2MG and antithrombin-III. Russelobin was non-cytotoxic, non-lethal and produced no histopathologies in mice, and it demonstrated in vivo dose-dependent defibrinogenating activity.Conclusions
Russelobin is an A/B fibrinogenase with high specificity toward fibrinogen, both in vitro and in vivo. Extensive glycosylation appears to protect the molecule against endogenous protease inhibitors, prolonging its in vivo efficacy.General significance
Due to its low toxicity, stability and activity as a defibrinogenating agent, Russelobin shows high potential for cardiovascular drug development. 相似文献14.

Black cutworm (BCW) is an economically important lepidopteran insect. The control of this insect by a Bt toxin and the understanding of the interaction between the Bt toxin and its receptor molecule were the objectives of this research work. A gene coding for a Vip3A receptor molecule was identified, characterized, and cloned, from the brush border membrane vesicles (BBMV) of the BCW. The nucleotide sequence analysis of the cloned putative Vip3A-receptor gene revealed that the gene was 1.3-kb long and exhibited no homology with any gene in the gene bank. We succeeded in identifying and characterizing most of the Vip3A-receptor gene sequence; and the nucleotide sequence analysis of the cloned putative Vip3A-receptor gene (accession no. KX858809) revealed about 92% of the expected sequence was recovered, which exhibited no homology with any gene in the GenBank. 相似文献

The structure of the extracellular polysaccharide (EPS) produced by Erwinia chrysanthemi strain RA3W, a mutant strain of E. chrysanthemi RA3, has been determined using low pressure size-exclusion and anion-exchange chromatographies, high pH anion-exchange chromatography, glycosyl linkage analysis, and 1D 1H NMR spectroscopy. The polysaccharide is structurally similar, if not identical, to the family of EPS produced by such as E. chrysanthemi strains Ech9, Ech9Sm6, and SR260. A microsporidium possessing molecular and morphological characteristics of the genus Nosema was isolated from larvae of the thee-spot grass yellow butterfly, Eurema blanda arsakia. The complete rRNA gene sequences of the E. blanda isolate contained 4,428 base pairs (GenBank Accession No. EU338534). The organization of the rRNA genes is LSU rRNA-ITS-SSU rRNA-IGS-5S, which corresponds with that of Nosema species closely related to Nosema bombycis. Phylogenetic analysis based on rRNA gene sequences show that this isolate is closely related to Nosema bombycis, Nosema plutellae, Nosema spodopterae, and Nosema antheraeae. The ultrastructure of all developmental stages of this microsporidium confirmed its placement in the genus Nosema. The isolate was successfully propagated in cell lines IPLB-LD652Y (Lymantria dispar) and NTU-LY (Lymantria xylina) and, in the in vitro system, it was frequently found to develop in the nuclei of the host cells, a circumstance that seldom occurs in other Nosema species. An extra-cellular vegetative stage of this microsporidium was also observed in the culture medium after 14 days of infection. The ECMDFs might be released from disrupted host cells. 相似文献

The influence of the colonization of salt marsh sediments with Halimione portulacoides, on the composition of the sedimentary humic acids was evaluated. For this purpose, cores of colonized and non-colonized sediments from a salt marsh in Ria de Aveiro (Portugal) were collected, and the humic acids of different layers were extracted and characterized by Fourier transform infrared, synchronous molecular fluorescence ( = 60 nm) and UV-visible spectroscopies and also by elemental analysis. The infrared spectra suggest the presence of more peptide residues and carbohydrates in the sedimentary humic acids from surface and around the plant roots at the site colonized by H. portulacoides, when compared with the humic acids from the depth-equivalent sediment layers at the non-colonized site. The higher content of protein-type materials is confirmed by the lowest values of C/N ratios and the highest relative intensities of a band at exc = 280 nm in the fluorescence spectra. The lowest 280 values obtained in the UV-visible spectra, and the infrared spectra suggest a lower aromatic content of the humic acids from the colonized site. 相似文献

A water soluble glucan, PLB-2C, was isolated from the water extract of the root of Pueraria lobata (Willd) Ohwi using anion-exchange and gel permeation chromatography. Its structure was investigated by gas chromatography (GC), gas chromatography–mass spectrometry (GC–MS), infrared (IR) spectra, and nuclear magnetic resonance (NMR) spectroscopy of heteronuclear single quantum coherence (HSQC) and heteronuclear multiple bond correlation (HMBC) techniques. The results indicated that PLB-2C was a linear glucan composed of (1 → 6)-α-d-Glcp. Chain conformation study showed that the polysaccharide took random coil compact conformation. In vitro cell viability assay by MTT method, its sulfated derivative PLB-2CS which was substituted at 2-O, 3-O, 4-O positions, at 0.1, 1, and 5 mg/ml, could attenuate PC12 cell damage significantly caused by hydrogen peroxide. 相似文献
