对北京东灵山地衣蜈蚣衣进行内生真菌分离,经过6种不同的方法消毒后,内生真菌的定殖率为55.4%~100%;从316个地衣组织块上共分离到28个真菌分类单元,其中包括1个子囊菌,22个丝孢菌,3个腔孢菌,1个毛霉和1个酵母菌。根据定殖率和分离到的内生真菌的种类两个指标综合考虑,消毒方法IV(75%乙醇1min,2%次氯酸钠3min,75%乙醇30s)为进行地衣内生真菌研究的最佳消毒方法。  相似文献

表面消毒是植物内生菌分离的关键环节,在南方红豆杉(Taxus chinensis var. mairei)内生真菌分离的过程中,使用75%乙醇、2%次氯酸钠(NaClO)代替升汞对外植体进行了表面消毒摸索。结果显示:红豆杉叶、茎、皮经预处理后,先用75%乙醇浸泡3 min,再用2%次氯酸钠浸泡2 min,然后红豆杉叶、茎、皮分别用75%乙醇浸泡3 min、5 min、7 min,可取得满意的表面消毒效果。  相似文献

When a combination of hydrogen peroxide and hypochlorite was used to surface sterilize rice seeds, a 102- to 104-fold decrease in CFU was observed during the first 15 h after inoculation of the rice rhizosphere organism Burkholderia vietnamiensis TVV75. This artifact could not be eliminated simply by rinsing the seeds, even thoroughly, with sterile distilled water. When growth resumed, a significant increase in the frequency of rifampin- and nalidixic acid-resistant mutants in the population was observed compared to the control without seeds. This phenomenon was a specific effect of hypochlorite; it was not observed with hydrogen peroxide alone. It was also not observed when the effect of hypochlorite was counteracted by sodium thiosulfate. We hypothesized that the hypochlorite used for disinfection reacted with the rice seed surface, forming a chlorine cover which was not removed by rinsing and generated mutagenic chloramines. We studied a set of rifampin- and nalidixic acid-resistant mutants obtained after seed surface sterilization. The corresponding rpoB and gyrA genes were amplified and sequenced to characterize the induced mutations. The mutations in five of seven nalidixic acid-resistant mutants and all of the rifampin-resistant mutants studied were found to correspond to single amino acid substitutions. Hypochlorite surface sterilization can thus be a source of artifacts when the initial bacterial colonization of a plant is studied.  相似文献

Decontamination of bacterial endospores such as Bacillus anthracis has traditionally required the use of harsh or caustic chemicals. The aim of this study was to evaluate the efficacy of a chlorine dioxide decontaminant in killing Bacillus anthracis spores in solution and on a human skin simulant (porcine cadaver skin), compared to that of commonly used sodium hypochlorite or soapy water decontamination procedures. In addition, the relative toxicities of these decontaminants were compared in human skin keratinocyte primary cultures. The chlorine dioxide decontaminant was similarly effective to sodium hypochlorite in reducing spore numbers of Bacillus anthracis Ames in liquid suspension after a 10 minute exposure. After five minutes, the chlorine dioxide product was significantly more efficacious. Decontamination of isolated swine skin contaminated with Bacillus anthracis Sterne with the chlorine dioxide product resulted in no viable spores sampled. The toxicity of the chlorine dioxide decontaminant was up to two orders of magnitude less than that of sodium hypochlorite in human skin keratinocyte cultures. In summary, the chlorine dioxide based decontaminant efficiently killed Bacillus anthracis spores in liquid suspension, as well as on isolated swine skin, and was less toxic than sodium hypochlorite in cultures of human skin keratinocytes.  相似文献

Aim:  The aim of this study was to investigate the nature and bioactivities of endolichenic fungi in three abundant lichens, Pseudocyphellaria sp., Usnea sp. and Parmotrema sp. in the lower elevation of Hakgala montane forest in Sri Lanka.
Methods and Results:  Endolichenic fungal strains, fungi that live asymptomatically in the lichen thallus, much the same way as endophytic fungi live within healthy plant tissues, were isolated from three abundant lichen species, Pseudocyphellaria sp., Usnea sp. and Parmotrema sp., at Hakgala montane forest in Sri Lanka, using the surface sterilization method. Nine endolichenic fungal strains were isolated from Parmotrema sp. and Usnea sp. separately, while 11 endolichenic fungi were recovered from the lichen Pseudocyphellaria sp. Isolation of endolichenic fungus Chrysosporium sp. 2 was common to all three lichen species. Substrate utilization patterns and antifungal activities of eight endolichenic fungal species were evaluated and the results revealed that all the test fungi were able to produce at least one enzyme to utilize the test substrates. Nigrospora sp., Chrysosporium sp. 1 and 2 and Cladosporium sp. showed antifungal activities on growth of some selected plant pathogenic fungi.
Conclusions:  Endolichenic fungal strains (29) were isolated from the lichens Parmotrema sp., Usnea sp. and Pseudocyphellaria sp. in Sri Lanka. Chrysosporium sp. 2 was common in all three lichens. Some of these endolichenic fungal strains showed antifungal activities against common plant pathogenic fungi and they are capable of utilizing the substrates by producing specific enzymes.
Significance and Impact of the Study:  The diversity and prevalence of the endolichenic fungi have not been studied extensively and this is the first report of isolation and identification of endolichenic fungi in lichens available in Sri Lanka.  相似文献

We investigated the potential of entomopathogenic hypocrealean fungi that naturally occur in or on adult mosquitoes for use as biocontrol agents of vector mosquitoes. The fungi were isolated from wild mosquitoes collected in Japan and Burkina Faso using two isolation methods (with and without surface sterilization). Detected fungal species included Beauveria bassiana sensu lato, Isaria spp., Paecilomyces spp., Lecanicillium spp., and Simplicillium spp. These isolates were used in bioassays against adult female Anopheles stephensi mosquitoes. The median survival time ranged from 5.8 to 14.9 d (control, 17.0 d). Reduced survival times were observed in the isolates from surface-sterilized mosquitoes from Japan, with the isolate B. bassiana s.l. 60-2 exhibiting the highest virulence. This study indicates that adult mosquitoes are naturally infected with various entomopathogenic hypocrealean fungi, and that some of these isolates have the potential for use as fungal pesticides to control vector mosquitoes.  相似文献

This is the first report of filamentous actinobacteria isolated from surface-sterilized root tissues of healthy wheat plants (Triticum aestivum L.). Wheat roots from a range of sites across South Australia were used as the source material for the isolation of the endophytic actinobacteria. Roots were surface-sterilized by using ethanol and sodium hypochlorite prior to the isolation of the actinobacteria. Forty-nine of these isolates were identified by using 16S ribosomal DNA (rDNA) sequencing and found to belong to a small group of actinobacterial genera including Streptomyces, Microbispora, Micromonospora, and Nocardiodes spp. Many of the Streptomyces spp. were found to be similar, on the basis of their 16S rDNA gene sequence, to Streptomyces spp. that had been isolated from potato scabs. In particular, several isolates exhibited high 16S rDNA gene sequence homology to Streptomyces caviscabies and S. setonii. None of these isolates, nor the S. caviscabies and S. setonii type strains, were found to carry the nec1 pathogenicity-associated gene or to produce the toxin thaxtomin, indicating that they were nonpathogenic. These isolates were recovered from healthy plants over a range of geographically and temporally isolated sampling events and constitute an important plant-microbe interaction.  相似文献

Eight fungal assemblages were isolated from thin branch roots (0.5–1 mm diameter) of living oak trees (Quercus robur) and their stumps 2 years after felling. The roots were serially washed or surface‐sterilized with ethanol and hypochlorite. The thin roots were inhabited by fungi that are stimulants of Armillaria rhizomorphs as found previously in thick roots (0.5–1 cm diameter). However, unlike thick roots, the densities of the fungi in serially washed roots were significantly less in stump roots than in living roots. Among the most common ‘stimulants’, the frequencies of Cylindrocarpon didymum, Mortierella gracilis, Mor. microspora var. macrocystis, Mycelium radicis atrovirens, Nectria grammicospora, Pseudogymnoascus roseus and Sporothrix schenckii were usually significantly greater in stump roots than in living roots, while the frequencies of Chrysosporium merdarium, Cy. destructans, Mor. hygrophila, Penicillium adametzii, Pe. citrinum, Pe. daleae, Pe. janczewskii and Pe. spinulosum were usually significantly less in stump roots. The frequency of Trichoderma viride was less in the stump roots. Serial washing of roots was more effective than chemical surface‐sterilization in the detection of Armillaria rhizomorph ‘stimulants’. The significance of these findings in the ecology and risk assessment of Armillaria in plantations is discussed.  相似文献

The subclass Chaetothyriomycetidae (Eurotiomycetes, Ascomycota) is an assemblage of ecologically diverse species, ranging from mutualistic lichenised fungi to human opportunistic pathogens. Recent contributions from molecular studies have changed our understanding of the composition of this subclass. Among others, ant-associated fungi, deep-sea fungi and bryophilous fungi were also shown to belong to this group of ascomycetes. The delimitation of orders and families within this subclass has not previously been re-assessed using a broad phylogenetic study and the phylogenetic position of some taxa such as the lichenised family Celotheliaceae or the Chaetothyrialean bryophilous fungi is still unclear. In our study, we assemble new and published sequences from 132 taxa and reconstruct phylogenetic relationships using four markers (nuLSU, nuSSU, mtSSU and RPB1). Results highlight several shortfalls in the current classification of this subclass, mainly due to un-assigned paraphyletic taxa. The family Epibryaceae is therefore described to circumscribe a previously un-assigned lineage. Celotheliales ad int. is suggested for the lineage including the lichen genus Celothelium and various plant pathogens. The delimitation of the family Trichomeriaceae is also broadened to include the genus Knufia and some anamorphic taxa. As defined here, Chaetothyriomycetidae includes four orders (Celotheliales ad int., Chaetothyriales, Pyrenulales, and Verrucariales) and ten families (Adelococcaceae, Celotheliaceae, Chaetothyriaceae, Cyphellophoraceae, Epibryaceae fam. nov., Herpotrichiellaceae, Pyrenulaceae, Requienellaceae, Trichomeriaceae, and Verrucariaceae).  相似文献

In the present study, a well-defined lichen community was screened for associated fungi for the first time. The photophilous lichen community Letharietum vulpinae was chosen because its character species, Letharia vulpina, was expected to host rather specialized fungi due to the presence of antimycotic secondary compounds. A considerable number of the associated fungi that were isolated were probably selective for lichens, because they appeared to be distantly related to fungi known from other substrates. The majority of these obligatory, lichen-associated fungi were only isolated in the course of the present study and represent hitherto unknown phylogenetic lineages. Parts of the lichen-associated fungi overlapped those colonizing rock surfaces or were closely related to endophytic fungi, but the lichen-associated and endophytic fungi still represented separate lineages.  相似文献

Aspergillus nidulans MCCC 3A00050 was isolated from a deep-sea sediment sample of the western Pacific Ocean. A systematic investigation on its chemical constituents led to the isolation of 19 compounds, including 13 steroids (113), four anthraquinones (1417), one phenolics (18), and one chromanone (19). For the first time, three diosgenins (13) were isolated from fungi, three ergosterols (46) from the genus of Aspergillus, while other 11 miscellaneous compounds (711 and 1419) from the species of Aspergillus nidulans. The isolation of diosgenins (13) and ergosterols (46) might be used as chemotaxonomic markers for the genus of Aspergillus and the species of Aspergillus nidulans, respectively.  相似文献

Summary Three seed coat sterilants were evaluated for effectiveness as surface sterilants and for effects on seed germination. Several exposure times in sodium hypochlorite, hydrogen peroxide and peracetic acid were evaluated. Seeds of the following crop plants were used: wheat (Triticum aestivum L.); sorghum [Sorghum bicolor (L.) Moench]; soybean [Glycine max (L.) Merrill]. All treatments reduced germination of wheat and soybean seed, but only the most severe peracetic acid treatments substantially reduced germination of sorghum seed. Considering both sterilization effectiveness and seed germination, a treatment with 5% sodium hypochlorite for 45 min appeared to be the most satisfactory of the treatments used.  相似文献

Field bean is a major crops in different parts of northwest Iran especially Zanjan province. Recently the bean plants were severely subjected to damping off or decline disease which caused yield losses in bean growing regions. A regional research was done from 2003 to 2005 to get general information on the causal agent of disease and its control management. Infected plants were collected from different studied areas and transferred to laboratory. Crown and plant roots were cultured in PDA as common media and PPA as selective media for Fusarium species after surface sterilization with sodium hypochlorite. Plates were incubated in standard culture room then isolated fungi were identified. Different Fusarium species were isolated, however the main pathogen isolated from plant samples and soil around the roots was F. sambucium Fuckel. The disease caused up to 50% yield losses in some fields in studied areas. Study showed the "Naz" cultivar was the main resistant race to the disease and had the most yield production in the field.  相似文献

Many black meristematic fungi persist on rock surfaces—hostile and exposed habitats where high doses of radiation and periods of desiccation alternate with rain and temperature extremes. To cope with these extremes, rock-inhabiting black fungi show phenotypic plasticity and produce melanin as cell wall pigments. The rather slow growth rate seems to be an additional prerequisite to oligotrophic conditions. At least some of these fungi can undergo facultative, lichen-like associations with photoautotrophs. Certain genera presenting different lifestyles are phylogenetic related among the superclass Dothideomyceta. In this paper, we focus on the genus Lichenothelia, which includes border-line lichens, that is, associations of melanised fungi with algae without forming proper lichen thalli. We provide a first phylogenetic hypothesis to show that Lichenothelia belongs to the superclass Dothideomyceta. Further, culture experiments revealed the presence of co-occurring fungi in Lichenothelia thalli. These fungi are related to plant pathogenic fungi (Mycosphaerellaceae) and to other rock-inhabiting lineages (Teratosphaeriaceae). The Lichenothelia thallus-forming fungi represent therefore consortia of different black fungal strains. Our results suggest a common link between rock-inhabiting meristematic and lichen-forming lifestyles of ascomycetous fungi.  相似文献

Effect of the physical properties of acrylic resin of overnight immersion in sodium hypochlorite solution Objectives: This study evaluated colour stability, surface roughness and flexural strength of microwave‐polymerised acrylic resin after overnight immersion in sodium hypochlorite, simulating 180 days use. Materials and methods: Forty disc‐shaped (15 mm × 4 mm) and 40 rectangular specimens (65 mm × 10 mm × 3 mm) were prepared from microwave‐polymerised acrylic resin. The specimens were immersed in 0.5, 1% sodium hypochlorite, Clorox/Calgon and distilled water. Colour measurements (ΔE) were determined by a portable colorimeter. A surface analyser was used to measure roughness (μm). The flexural strength (MPa) was measured using a three‐point bending test in a universal testing machine. Data were evaluated by one‐way anova , followed by Student–Newman–Keuls test (α = 0.05). Results: Statistical analysis found significantly higher colour changes (SNK, p < 0.001) for the 1% sodium hypochlorite, but mean ΔE value quantified by National Bureau of Standards was classified as slight. When comparing the surface roughness, no statistical significance was found among the solutions (anova , p = 0.637). The 1% sodium hypochlorite presented significantly lower flexural strength compared with the control group (SNK, p = 0.034). Conclusion: It was concluded that immersion in 1% sodium hypochlorite solutions for 8 h does influence the colour stability and flexural strength of microwave‐polymerised acrylic resin, during the simulated period of 180 days.  相似文献

Aims: To evaluate a sodium hypochlorite and hydrogen peroxide solution (Ox‐B7) as a potential decontaminant of Bacillus subtilis spore‐contaminated surface materials (porous and nonporous). Methods and Results: Test materials were contaminated with B. subtilis spores to a final concentration in the range of 5·7–6·6 log CFU cm?2. Ox‐B7 reduced spore counts by 99·999% (5 log) for both porous and nonporous surfaces within a 5‐min contact. Treatment with equivalent concentrations of only sodium hypochlorite reduced spore counts by 99% (2 log) on porous materials and by 99·99% (4 log) on nonporous materials. Hydrogen peroxide treatments reduced spores by less than 90% (<1 log) on both porous and nonporous materials when compared with untreated samples. Conclusions: A combination of sodium hypochlorite and hydrogen peroxide (Ox‐B7) effectively killed B. subtilis spores on both porous and nonporous surface materials. Significance and Impact of the Study: The combination of sodium hypochlorite and hydrogen peroxide can be used as an alternative disinfectant of spore‐contaminated surface materials, as it is more effective than when hydrogen peroxide or sodium hypochlorite are used separately.  相似文献

An ecological study was carried out on the fungal flora in the human lung of 159 autopsy cases. Fungi were isolated from 129 cases (81.1%). Filamentous fungi consisting of 918 strains were isolated from 113 cases, and the dominant genera were Aspergillus and Penicillium. Yeasts were isolated from 58 cases, and the dominant genus appeared to be Candida.With morbid anatomical study, the incidence of the fungus isolation was examined from various points of view. It was concluded that even a healthy respiratory parenchyma of the human lung cannot be assumed as aseptic. However, only a few numbers of a few genera of air-born fungi were isolated.Special stress was laid on the exposure of the respiratory parenchyma of the human lung to air-born fungi in connection with the pathogenesis of opportunistic fungus infections.  相似文献
